Ejemplifica simultáneamente varios conceptos que se vinculan con la cinética química y la catálisis con la química de los lípidos y su metabolismo. Se estudia la cantidad de ácidos grasos producidos al hacer reaccionar durante 1hora a 37 grC una cantidad constante de aceite de girasol con una cantidad constante de lipasa pancreática de cerdo |b a diferentes pH. Se determina así el pH óptimo de trabajo de la mencionada lipasa.

Reducing the concentration of added sugar in processed foods is one of the most realistic strategies to reduce the intake of this nutrient in the short-term. In order to be effective, gradual sugar reduction strategies need to determine the maximum sugar reduction that can be unnoticed by consumers. In this context, the present work aimed at providing recommendations for gradual sugar reduction in chocolate-flavoured milk by determining difference thresholds for added sugar and evaluating consumers' sensory and hedonic perception of reduced-sugar products. Five studies were conducted with 50 consumers to determine five sequential difference thresholds. In each study consumers completed six paired-comparison tests. Each pair was composed of a reference chocolate-flavoured milk and a sample that was reduced in added sugar from the reference. Difference thresholds, corresponding to the smallest reduction in sugar concentration that is noticed by consumers, were determined using survival analysis. Then, a study was carried to with 100 consumers to evaluate their sensory and hedonic perception of chocolate-flavoured milk samples with different added sugar concentrations. Results suggested that sequential sugar reductions can be set at 6.7% without affecting consumers' sensory and hedonic perception. Sugar reduction in chocolate-flavoured milk without affecting consumers' perception seems feasible and easy to implement. The approach of the present work could be extended to design recommendations for gradual reduction of the added sugar concentration of other industrialized products, contributing to the development of more healthful products that meet current nutritional recommendations.

<p>CATA (check-all-that-apply) questions enjoy high popularity for sensory product characterization with consumers. Amidst extensive methodological characterization, the meaning of term citation frequency has received modest attention only,<br />possibly hindering further uptake. The present research performed a more detailed exploration of the relationship between CATA responses and perceived intensity of sensory attributes. In addition, the following question was addressed:<br />How many consumers are necessary to interpret differences in CATA citation proportions as a proxy of differences in perceived intensity? The present research includes new empirical data from four consumer studies conducted in New Zealand and China under central location test conditions and a re-analysis of<br />one consumer study. When dealing with samples with clear differences, the percentage of consumers selecting a sensory term for describing samples tended to be linearly correlated to average intensity ratings. A median of 15 consumers was<br />needed for CATA to show good linear correlations with average intensity ratings, and the minimum number of consumers needed to reach the threshold for a good correlation was generally lower than 30.</p>

Abstract
A summary of the resource.
The antiproliferative effects of four series of VIVO- and VVO-based compounds containing 8-hydroxyquinoline ligands on the bacterium Mycolicibacterium smegmatis (M. smeg) were investigated. The effects on M. smeg were compared to the antiproliferative effects on the protozoan parasite Trypanosoma cruzi (T. cruzi), the causative agent for Chagas disease. In this study, we investigate the speciation of these compounds under physiological conditions as well as the antiproliferative effects on the bacterium M. smeg. We find that the complexes are more stable the less H2O is present, and that the stability increases in lipid-like environments.<br />Only one heteroleptic complex and two homoleptic complexes were found to show similar antiproliferative effects on M. smeg as reported for T. cruzi so the responses generally observed by M.smeg. is less than observed by the pathogen. In summary, we find that M. smeg is more sensitive to the detailed structure of the V-complex but overall these complexes are less effective against M. smeg compared to T. cruzi.

Abstract
A summary of the resource.
Diseases caused by several (sub)species of parasites from the genus Trypanosoma are a major concern for public and animal health. Trypanosomes have a digenetic life cycle that involves an insect host that acts as the vector for transmission to different mammals, some of which are natural reservoirs. The zoonotic nature of the disease together with the capacity of the parasites to evade the host immune response or become dormant hampers pathogen eradication and immunoprophylactic control. A handful of drugs are available for the treatment, although none of them has been specifically designed toward these pathogens. The drugs are efficacious when applied at very early stages of the infections, but display high toxicity, require a long or assisted administration, present a limited efficacy against late stages of the diseases and are prone to develop resistance. Here we review the state-of-the-art for the drugs used to treat trypanosomiasis that causes African sleeping sickness and Chagas’ disease in humans. The state-of-the-art on drug uptake, mode of action, and resistance is here reviewed. The prospects for developing new molecules that circumvent the current challenges for treating trypanosomiasis are also discussed.

Abstract
A summary of the resource.
This work investigated the effect of temperature on the reaction efficiency of soybean oil transesterification in supercritical ethanol in a continuous catalyst-free process under different water concentrations and at varying substrate flow rates. Experiments were performed in the temperature range from 250 to 325 °C, at 20 MPa, with an oil to ethanol molar ratio of 1:40. Results showed that temperature and substrate flow rates strongly affected the reaction conversion to fatty acid ethyl esters, decomposition, and trans-isomerization of unsaturated fatty acids, mainly for C18:2 and C18:3. It is shown that the synthesis of esters was favored by the addition of water to the reaction medium and the degradation phenomenon decreased as water concentration increased from 0 to 10 wt %.<br /><br />

A summary of the resource.
While phenolic compounds can impact the sensory characteristics of quality wines, the chemical modifications induced by yeasts on phenolic compounds have been studied only to a limited extent. Forty-nine non-Saccharomyces yeast strains, belonging to different genera, were screened in a synthetic must supplemented with anthocyanins from Tannat grapes, for their effects on color and fermentation capability. Total anthocyanins, color intensity, hue and total polyphenol index were significantly affected (p < 0.001) by the different yeasts studied. The effects on these parameters were highly diverse depending on the yeast species studied according to their capacities of improving color indexes of the red grape model wines. Six strains belonging to the species Metschnikowia pulcherrima, Hanseniaspora guilliermondii, H. opuntiae, H. vineae and H. clermontiae, were selected in order to evaluate the production of anthocyanin-derived pigments, and significant variations in the production of four compounds were found between them and compared to Saccharomyces cerevisiae. The formation of vitisin A, vitisin B, malvidine-3-glucoside-4-vinylphenol and malvidin-3-glucoside-4-vinylguaiacol are reported for the first time for species of genera Hanseniaspora and Metschnikowia.

There are recent experimental reports on the preparation of edge-modified graphene quantum dots (GQDs) and its loading into conductive polymers for energy applications. Recently, we have studied the influence of the graphene quantum dots (GQDs) addition to polyaniline (PANI) conductive polymer in the electronic transport, finding important modifications in the structural and electrical properties respect to the pristine polymer. In this work, we present a mixed approach, based in first-principles molecular dynamics with time-dependent density functional calculations, for the description of the electronic dynamics, particularly for the determination of the optical properties. We found that edge modification of the GQDs favors the PANI/GQDs interaction, promoting very interesting electronic properties. Particularly, when hydroxyl chemical functional groups are present at the edges of the GQD a very good electron–hole separation during the photon excitation is obtained, demonstrating its potential use for optical applications in general and solar cells in particular

A summary of the resource.
In order to maintain the capacity to treat infections it is necessary to adopt strategies for the development of new drugs. The use of metal ions in medicine for their antimicrobial effects was prevalent for many years before the advent of antibiotics. During the second half of the twentieth century, active metal complexes with different chemical, physicochemical and biochemical properties than the constituents reborn by the threat posed by microbial multidrug resistance. In this review, essential transition metal drugs with Cu(II), Zn(II) and Fe(III), showing known chemical structure, antimicrobial activities better than the free ligands and reported in the last ten years, are presented. The scientific information realizes many metal complexes having antimicrobial activity, which covers Gram negative and Gram positive bacteria, mycobacteria, yeast and fungi and many of them would be good candidates for further pharmaceutical studies. For these reasons the strategy of metal coordination is a potent tool to get numerous and diverse new antimicrob

La fusariosis en la espiga es una de las enfermedades más destructivas en granos de trigo y cebada, causante de grandes pérdidas económicas e inconvenientes en términos de seguridad alimentaria, debido a la presencia de micotoxinas que afectan tanto animales como humanos. Varias especies del género Fusarium han sido reconocidos como agentes causales de esta enfermedad, siendo las especies del Complejo Fusarium graminearum (FGSC) las que aparecen con mayor frecuencia. El FGSC está compuesto por más de 16 especies filogenéticamente diferentes, que producen tricotecenos del tipo B (nivalenol, deoxinivalenol y derivados acetilados) y que se pueden agrupar en tres quimiotipos diferentes, dependiendo de su potencial para producir micotoxinas. En nuestro país se monitorea de forma rutinaria una sola de dichas micotoxinas: el deoxinivalenol, a pesar de que las restantes posibles toxinas tienen toxicidad similar. El presente trabajo de tesis estudia la diversidad, potencial micotoxigénico y sensibilidad a fungicidas de hongos causantes de fusariosis de la espiga de trigo en las diferentes zonas de producción de Uruguay. En primer lugar, se determinaron de forma simultánea las especies y los quimiotipos de los distintos aislamientos mediante un ensayo denominado “multilocus genotyping” (MLGT), encontrándose predominancia de la especie F. graminearum y del quimiotipo 15ADON. Además se encontraron aislamientos de las especies F. cortaderiae, F. asiaticum y F. brasilicum, del quimiotipo NIV, no reportados hasta el momento en Uruguay. Para poder diferenciar de forma rápida y sencilla, las especies del FGSC predominantes en el país, se desarrolló un método de PCR-RFLP, utilizando primers específicos para el gen del factor de elongación de la transcripción 1 alfa (TEF-1!). Los aislamientos encontrados fueron caracterizados de acuerdo a su sensibilidad al fungicida tebuconazol, determinándose una sensibilidad significativamente menor en los aislamientos de quimiotipo NIV frente al quimiotipo 15ADON. Paralelamente se desarrolló un método de PCR en tiempo real, para la cuantificación de hongos productores de tricotecenos y en particular hongos productores de nivalenol y zearalenona. Asimismo se puso a punto un método para la determinación de micotoxinas (deoxinivalenol, nivalenol, 15acetildeoxynivalenol, 3acetildeoxynivalenol) en granos contaminados, por cromatografía líquida acoplada a un espectrómetro de masa como detector.

Abstract
A summary of the resource.
A novel method for the determination of trace cadmium in honey is presented. It consists of a preconcentration step based on dispersive liquid-liquid microextraction (DLLME) and measurement by flame AAS. Samples are dissolved in water. A mixture of carbon tetrachloride (extraction solvent) and acetonitrile (dispersion solvent) containing ammoniun pyrrolidine dithiocarbamate (APDC) as complexing agent is injected rapidly into the dissolved sample by means of a syringe pump. After centrifugation, an aliquot of the organic phase is injected in the nebuliser of an atomic absorption spectrometer by means of a lab-made microsampling device. Response (integrated absorbance) was linear with concentrations up to 12 μg L−1. Precision (sr(%)) at the 5 μg L−1 level was < 3.1%. Detection and quantification limits were 1.6 and 5.5 μg per 100 g of honey respectively. Six commercial samples were spiked at the 10 μg per 100 g level and analysed by the proposed method and by electrothermal atomic absorption spectrometry. The results did not differ at the 5% significance level, with recoveries for the DLLME/FAAS method ranging from 82 to 125%. The proposed method requires simple sample preparation and does not use expensive equipment, thus it could be used as screening method for the content of cadmium in honey.

A summary of the resource.
Eukaryotes using trans-splicing for transcript processing incorporate a taxon-specific sequence tag (the spliced leader, SL) to a proportion (either all or a fraction) of their mRNAs. This feature may be exploited for the preparation of full-length-enriched cDNA libraries from these organisms (a diverse group including euglenozoa and dinoflagellates, aswell asmembers from five metazoan phyla: Cnidaria, Rotifera,Nematoda, Platyhelminths and Chordata). The strategy has indeed been widely used to construct cDNA libraries for the generation of ESTs, mainly from parasitic euglenozoa and helminths. We describe a set of optimised protocols to prepare directional SL-cDNA libraries; the method involves PCR-amplification of SL-cDNA and its subsequent cloning in a plasmid vector under a specific orientation. It uses small amounts of total RNA as starting material and may be applied to a variety of samples. The approach permits the selective cloning of mRNAs tagged with a particular SL from mixtures including large amounts of non-trans-spliced mRNAs. Thus, it allows exclusion of host contamination when isolating SL-cDNAs from parasitic organisms, and has other potential applications, such as the characterisation of the trans-spliced transcriptome from organisms in mixed pools of species.

Objectives Since KPC-2-producing Klebsiella pneumoniae are associated with successful dissemination of a major clone, defined as sequence type 258 (ST258), the aim of this study was to perform whole-genome sequencing (WGS) of the first colistin-resistant K. pneumoniae strain (Kpn666) carrying blaKPC-2 identified in Uruguay in 2011 in order to identify genomic and phylogenetic traits. Methods WGS of strain Kpn666 isolated from an asymptomatic urinary tract infection was performed using Illumina MiSeq, and de novo assembly was performed using SPADES v.3.11. Contigs were re-ordered using the ST258 reference genome NJST258_1 (GenBank CP006923) and were oriented with the MAUVE Contig Mover. Twenty complete genomes of K. pneumoniae identified as ST258 using the Pasteur MLST site were downloaded from GenBank (May 2017). A maximum-likelihood tree was created using MEGA7 based on core single nucleotide polymorphisms (SNPs) from whole-genome alignment obtained with SNP sites (https://github.com/sanger-pathogens/snp-sites). Results WGS analysis revealed a genome of 5 448 179 bp (5232 CDS, 108 RNAs). Phylogenetic analysis identified that Kpn666 belonged to clade I lineage of ST258. Further studies also identified IncR, IncFIB(K) and IncFII(K) plasmid replicons and 11 transferable associated antimicrobial resistance genes (ARGs) comprising four drug classes. The mgrB gene involved in colistin resistance was shown to be disrupted by insertion of an IS5-like element. Conclusions The first isolate of KPC-2-producing K. pneumoniae

Honey mineral content depends on the type and the surroundings of the flower from where it originates and on the production process. The presence of heavy metals represents a risk to the consumer’s health. Lead is usually determined by atomic spectrometry where a stage of acidic digestion is mandatory. In this work, hydride generation was chosen as the sample introduction technique; plumbane was directly generated in honey and determined by microwave induced plasma emission spectrometry (HG MIP OES). Different cleaning procedures, use of arsenic as internal standard, matrix match calibration and oxygen flow injection in the plasma were evaluated for controlling the frothing in the reaction chamber, abatement of the memory effect, and minimising gas-phase interferences. Limits of detection (3 s) and quantification (10 s) were 0.024 mg kg 1 and 0.081 mg kg 1; linear range was within 8.1–56 μg kg 1 and precision of repeatability 13 %. Trueness was assessed at three concentration levels with a reference sample (recoveries between 107 % and 122 %) and with spiked samples at 0.10 mg kg 1 level (recoveries between 82 % and 116 %). Direct HG MIP OES is a fast and confident screening method for lead monitoring in honey.

The visual appearance and olfactory cues of liquid soap can trigger powerful expectations and associations in consumers' mind, which largely influence purchase decisions and shape product experience. In this context, the aims of the present work were: (a) to study the effect of appearance (color and transparency) and fragrance on conceptual associations and expectations of liquid hand soap and (b) to evaluate if preference<br />patterns moderate such effect. Eight samples of liquid hand soap were obtained following a full factorial design with three 2-level variables: fragrance (herbal vs. fruity), color (green vs. orange), and transparency (transparent vs. pearly).<br />Samples were evaluated by 104 female participants, who rated their liking and used a<br />check-all-that-apply question to describe the samples in terms of sensory characteristics,<br />expected functional aspects of the product, and conceptual associations. Results showed that appearance and transparency did not influence consumer liking. However, fragrance had a significant effect on consumer liking, expectations and conceptual associations of liquid hand soap. The effect of fragrance on expectations and<br />conceptual associations depended on consumer liking of the fragrance. In addition, pearly appearance did not influence liking but had a significant effect on consumer expectations regarding functional aspects of the liquid hand soap.

A summary of the resource.
Se analizaron bebidas de arroz sometidas a diferentes tratamientos térmicos y a UAPH. Se midió su tamaño de partícula y se correlacionó con medidas de estabilidad; se observó que las muestras procesadas por UAPH tienen una estabilidad significativamente mayor. Se estudió la interacción entre micronutrientes (Ca++) y las proteínas de la bebida y se halló que al aumentar la concentración de Ca++ la estabilidad de la bebida disminuye hasta alcanzar 80 ppm de Ca++, a partir de la cual se mantiene constante. Los recuentos de mesófilos aerobios totales, esporas, Bacillus Cereus y Enterobacteriaceaes para muestras tratadas por UAPH (temperatura de entrada 85 ºC, 300 MPa temperatura en válvula 135 ºC) y UAT (142 ºC 6 seg) muestran una supervivencia menor a 1 UFC/mL en ambos tratamientos para todos los recuentos. Se incubaron las muestras UAPH y UAT a 30 ºC durante dos semanas sin desarrollo microbiano o alteración alguna. El proceso de ultra alta presión de homogeneización (UAPH) demostró ser una tecnología válida para estabilizar y preservar la leche de arroz al reducir el tamaño de las partículas por debajo del micrómetro, favoreciendo la estabilidad fisicoquímica de la bebida y generando, concomitantemente, una reducción de la flora alterante.

The aetiological agent of cystic hydatid disease, the platyhelminth parasite Echinococcus granulosus, undergoes a series of metamorphic events during its complex life cycle. One of its developmental stages, the protoscolex, shows a remarkable degree of heterogeneous morphogenesis, being able to develop either into the vesicular or strobilar direction. Another level of complexity is added by the existence of genotypes or strains that differ in the range of intermediate hosts where they can develop and form fertile cysts. These features make E. granulosus an interesting model for developmental studies. Hence, we focused on the study of the regulation of gene expression by microRNAs (miRNAs), one of the key mechanisms that control development in metazoans and plants and which has not been analysed in E. granulosus yet. In this study, we cloned 38 distinct miRNAs, including four candidate new miRNAs that seem to be specific to Echinococcus spp. Thirty-four cloned sequences were orthologous to miRNAs already described in other organisms and were grouped in 16 metazoan miRNA families, some of them known for their role in the development of other organisms. The expression of some of the cloned miRNAs differs according to the parasite life cycle stage analysed, showing differential developmental expression. We did not detect differences in the expression of the analysed miRNAs between protoscoleces of two parasite genotypes. This work sets the scene for the study of gene regulation mediated by miRNAs in E. granulosus and provides a new approach to study the molecules involved in its developmental plasticity and intermediate host specificity. Understanding the developmental processes of E. granulosus may help to find new strategies for the control of cystic hydatid disease, caused by the metacestode stage of the parasite.

Background Uruguay, a South American country of 3.4 million inhabitants that has already banned tobacco advertising, prohibited such terms as light, mild and low-tar and required graphic warnings covering 80% of cigarette packs, is considering the imposition of plain, standardised packaging.
Methods We conducted an experimental choice-based conjoint analysis of the impact of alternative cigarette package designs on the risk perceptions of 180 adult current Uruguayan smokers. We compared plain packaging, with a standardised brand description and the dark brown background colour required on Australian cigarette packages, to two controls: the current package design with distinctive brand elements and colours; and a modified package design, with distinctive brand elements and the dark brown background colour. Graphic warnings were also varied.
Results Plain packaging significantly reduced the probability of perceiving the stimulus cigarettes as less harmful in comparison to the current package design (OR 0.398, 95% CI 0.333 to 0.476, p<0.001) and the modified package design (OR 0.729, 95% CI 0.626 to 0.849, p<0.001).
Conclusions Plain packaging enhanced the perceived risk of cigarette products even in a highly regulated setting such as Uruguay. Both the elimination of distinctive brand elements and the use of Australia’s dark brown background colour contributed to the observed effect.

Abstract

Diabetes pathogenesis encompasses oxidative stress, inflammation, insulin malfunctioning and partial or total insulin secretion impairment, which leads to a constant hyperglycemia. Polyphenols are known to possess bioactive properties, being Tannat grape skin a natural and sustainable source of these compounds. The present study aimed to find out the bioaccessibility of health-promoting molecules composing a multifunctional extract from Tannat grape skin obtained under hydro-alcoholic-acid conditions. The identification of phenolic compounds in the samples was performed by ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS). Subsequently, the samples were in vitro digested mimicking the human oral gastrointestinal conditions and the bioactivity of the digest (antioxidant, anti-inflammatory and modulation of glucose metabolism) was assessed. E�ect on glucose metabolism was estimated by measuring carbohydrases activity and the functionality of glucose transporters of small intestine cells in presence and absence of the digested extract. Flavonoids, phenolic acids and phenolic alcohols were the major phenol compounds detected in the extract. The bioaccessible compounds protected the intestinal cells and macrophages against the induced formation of reactive oxygen species (ROS) and nitric oxide (NO). In addition, glucose transporters were inhibited by the digested extract. In conclusion, the bioaccessible compounds of the extract, including phenols, modulated key biochemical events involved in the pathogenesis of diabetes such as oxidative stress, inflammation and glucose absorption. The extract was e�ective under prevention with co-administration conditions supporting its potential for either reducing the risk or treating this disease.
