A summary of the resource.
<p>A fast and efficient automated method using a sequential injection analysis (SIA) system, based on the Griess, reaction was<br />developed for the determination of nitrate and nitrite in infant formulas and milk powder. The system enables to mix a measured<br />amount of sample (previously constituted in the liquid form and deproteinized) with the chromogenic reagent to produce a<br />colored substance whose absorbance was recorded. For nitrate determination, an on-line prereduction step was added by passing<br />the sample through a Cd minicolumn. The system was controlled from a PC by means of a user-friendly program. Figures of merit<br />include linearity (r2 > 0.999 for both analytes), limits of detection (0.32mg kg−1 NO3-N, and 0.05 mg kg−1 NO2-N), and precision<br />(sr%) 0.8–3.0. Results were statistically in good agreement with those obtained with the reference ISO-IDF method. The sampling<br />frequency was 30 hour−1 (nitrate) and 80 hour−1 (nitrite) when performed separately.</p>

This analytical note presents a simple and economic analytical method for quantifying toxicologically relevant arsenic species in urine—the sum of As (III), As (V), monomethylarsonic acid, and dimethylarsinic acid—by hydride generation microwave-induced plasma atomic emission spectrometry (HG-MIP-AES). The accuracy of the method was in the range of 95%–97%; the intra-assay precision was within the range of 1.8%–6.2% expressed as the relative standard deviation, and the interassay precision was within 6.9%–15%. The limits of detection and quantification were 1.8 and 5.4 μg L− 1 , respectively. The validated method resulted very helpful to assess the health risk of As exposure through drinking water and was successfully applied in urine samples collected from wood impregnation plant workers. The methodology can be postulated as a viable alternative to expensive techniques, such as liquid chromatography (HPLC) in combination with inductively coupled plasma mass spectrometry (ICP-MS), for biomonitoring studies.

A summary of the resource.
Uruguay is among the top eight exporters of rice in the world. Assessing the levels of Arsenic (As) in rice grain is a concern for human health. To ensure food safety, the laboratories need simple and rapid validated analytical methodologies to provide fast response, to allow the commercializa- tion of the products. A simple ultrasound-assisted method for sample preparation and determination of total arsenic using Hydride Generation Atomic Absorption Spectrometry was validated. The figures of merit were adequate for control purposes in terms of precision and trueness. Commercial samples of Uru- guayan rice were analyzed and the results did not differ significantly with those obtained using ICP-MS. The total arsenic concentrations ranged from 0.10 to 0.24 mg kg −1 . These values were below the maximum limit established in the region. The proposed methodology is simple, economic and does not require dras- tic conditions of sample preparation or expensive techniques.

African trypanosomiasis is a major problem for human and animal health in endemic countries, where it threatens millions of people and affects economic development. New drugs are needed to overcome the toxicity, administration, low efficacy, and resistance issues of the current chemotherapy. Robust, simple, and economical high‐throughput, whole‐cell‐based assays are required to accelerate the identification of novel chemical entities. With this aim, we generated a bioluminescent cell line of the bloodstream stage of Trypanosoma brucei brucei and established a screening assay. Trypanosomes were stably transfected to constitutively express a thermostable red‐shifted luciferase. The growth phenotype and drug sensitivity of the reporter cell line were essentially identical to that of the parental cell line. The endogenous luciferase activity, measured by a simple bioluminescence assay, proved to be proportional to parasite number and metabolic status. The assay, optimized to detect highly potent compounds in a 96‐well‐plate format, was validated by screening a small compound library (inter‐assay values for Z' factor and coefficient variation were 0.77 and 5.8%, respectively). With a hit‐confirmation ratio of ~97%, the assay was potent enough to identify several hits with EC50 ≤ 10 μM. Preliminary tests indicated that the assay can be scaled up to a 384‐well‐plate format without compromising its robustness. In summary, we have generated reporter trypanosomes and a simple, robust, and affordable bioluminescence screening assay with great potential to speed up the early‐phase drug discovery against African trypanosomes.

This chapter introduces a simple and robust in vitro viability assay to screen bioactive small molecules (e.g., natural, synthetic) against the monomorphic and infective (bloodstream) form of Trypanosoma brucei brucei. The assay relies on a bioluminescent transgenic parasite harboring a genetically encoded copy of a thermostable redshifted firefly luciferase from Photinus pyralis. The major advantages of the assay are simplicity and cost efficiency, along with excellent quality parameters. The bioassay allows estimating parasite numbers and viability (and metabolic state) as a function of bioluminescence (BL) signal. Parasites are grown in the presence of the molecules of interest in a 96-well microplate, and 24 h later, BL is determined with a simple protocol lacking washing steps, using cost-efficient reagents with a reasonable readout time for high-throughput applications.

Anecdotal reports and open-label case studies in humans indicated that the psychedelic alkaloid ibogaine exerts profound antiaddictive effects. Ample preclinical evidence demonstrated the efficacy of ibogaine, and its main metabolite, noribogaine, in substance-use-disorder rodent models. In contrast to addiction research, depression-relevant effects of ibogaine or noribogaine in rodents have not been previously examined. We have recently reported that the acute ibogaine administration induced a long-term increase of brain-derived neurotrophic factor mRNA levels in the rat prefrontal cortex, which led us to hypothesize that ibogaine may elicit antidepressant-like effects in rats. Accordingly, we characterized behavioral effects (dose- and time-dependence) induced by the acute ibogaine and noribogaine administration in rats using the forced swim test (FST, 20 and 40 mg/kg i.p., single injection for each dose). We also examined the correlation between plasma and brain concentrations of ibogaine and noribogaine and the elicited behavioral response. We found that ibogaine and noribogaine induced a dose- and time-dependent antidepressant-like effect without significant changes of animal locomotor activity. Noribogaine's FST effect was short-lived (30 min) and correlated with high brain concentrations (estimated >8 μM of free drug), while the ibogaine's antidepressant-like effect was significant at 3 h. At this time point, both ibogaine and noribogaine were present in rat brain at concentrations that cannot produce the same behavioral outcome on their own (ibogaine ∼0.5 μM, noribogaine ∼2.5 μM). Our data suggests a polypharmacological mechanism underpinning the antidepressant-like effects of ibogaine and noribogaine.

The local crystal structure of Y3NbO7 was probed using neutron total scattering. Five different structural models were fit to the experimental pair distribution function of this material up to 15 Å. Fits of defect fluorite (Fm3m), pyrochlore (Fd3m), and orthorhombic C weberite models (C2221) were attempted based on prior structural investigations of Y3NbO7 and related fluorite-based materials. Orthorhombic P weberite (P2221) and monoclinic weberite models (P1121) were derived via symmetry reduction of the orthorhombic C weberite structure. All models except monoclinic weberite failed to reproduce the experimental pair distribution function. Misfits were clearly visible for peaks arising from atom pairs within the first coordination shell (defect fluorite and pyrochlore) and beyond (orthorhombic C and P weberite). By contrast, monoclinic weberite (γ ≈ 91.8◦) provided an excellent fit, with no major misfits observed. This unit cell features distorted YO8 dodecahedra, YO7 monocapped octahedra, and NbO6 octahedra as building blocks. Comparison between the topologies of the cationic and anionic substructures in monoclinic weberite and its parent orthorhombic C structure revealed displacements of the metal atoms along the [100] direction and a less compact spatial distribution of oxygen atoms along the a axis. The monoclinic weberite model presented herein may serve as a starting point to revisit structural modeling of fluorite-based materials with challenging local structures

A summary of the resource.
In this work we present the synthesis, structural, morphological, optical and electrochemical characterization of sodium titanate nanotubes (NaNT) obtained by hydrothermal means. According to our detailed structural study, we found that a Na2Ti2O5·H2O layered crystalline phase with [TiO5] pyramidal square base coordination, utilized as a building block of the nanotubes, explains satisfactorily the experimental results. This is a very important aspect, since the bibliography does not always provide structural information about titanate nanotubes. With respect to the formation mechanism we highlight the crucial role of the post treatment washing in the final crystalline structure of the nanotubes. Simulated X-rays powder diffraction patterns (XRPD), based on real size nanotubes models, were refined with experimental data. Building blocks for the NaNT model were inferred from computational simulations, by means of Density Functional Theory. The obtained tube's model characteristic dimensions were also consistent with SAXS and TEM analysis. In addition, our SAXS analysis suggested that the presence of humidity in the cavities of titanate nanotubes is progressively lost at the 80–100 °C temperature range, in good agreement with thermogravimetric analysis. Electrochemical impedance spectroscopy analysis for dried samples in the temperature range, studied under cooling conditions, revealed a single ionic transport mechanism, probably associated to the in-between [TiO5] layers ion transport. However, a secondary process was observed for non-dried samples, during heating conditions, that could be associated to the ionic transport along the water filled tube's cavity, consistent with SAXS analysis. In summary, this work shows an integral approach, through theory and experiment, in order to understand the effect of the structure on the physical and electronic properties of the system.

Carquejol and its acetate are monoterpenoids based on the rare o-menthane skeleton and distinctive components of the essential oil from Baccharis trimera. Carquejol was characterized by using Fourier Transform infrared (FT-IR) and Raman (FT-Raman), Ultraviolet–Visible (UV–Visible), Electronic Circular Dichroism (ECD), Mass, Hydrogen and Carbon Nuclear Magnetic Resonance (1H and 13C NMR) and 2D 1H1H gCOSY, 1H13CgHSQC, 1H13CgHMBC spectroscopies. Due to the chirality of this monoterpenoid, six different structures were analysed, of which only four showed higher populations and minimal energies. The natural bond orbital (NBO), atoms in molecules (AIM), Merz-Kollman (MK) charges, molecular electrostatic potentials (MEP) and frontier orbitals studies were performed in order to evaluate their structural, electronic, topological and vibrational properties. All calculations were performed by using the hybrid B3LYP method and the 6-31G* and 6-311++G** basis sets. The comparison of the experimental ECD spectra with the corresponding theoretical ones confirm the (4S,5R) configuration assigned to carquejol. The force fields for the most stable configurations were computed by using those two levels of theory and the complete vibrational assignments for the two conformations of carquejol are reported. The different orientations and directions of the dipole moments of the two structures and the proximity in the nucleophilic indexes with those reported for other terpenes could justify in part the potential biological properties reported for carquejol. The MEP surfaces for both structures reveal that the nucleophilic and electrophilic sites of higher reactivity are principally centred on the OH groups.

The reported anti-Prelog selectivity in Raphanus sativus hairy roots prompted us to search for similar activity in other R. sativus systems, such as in vitro germinated sprouts, edible roots, and its associated endophytic microorganisms. Two test substrates were used in these studies: acetophenone and rac-ethyl 2-ethyl-3-oxobutanoate. Endophytes were isolated under substrate selective pressure and individual strains were evaluated for their ability to reduce both substrates. Among the isolates, two bacteria and a fungus having the potential to be used as biocatalysts were found: Bacillus megaterium which reduces acetophenone with excellent anti-Prelog selectivity, and Pseudomonas sp. and Penicillium chrysogenum with enantioselectivity and stereoselectivity for the reduction of rac-ethyl 2-ethyl-3-oxobutanoate, respectively.

The polymer precursor formation and the growth mechanism of (La0.5Ba0.5)(Mn0.5Fe0.5)O3 ceramic nanoparticles have been studied. First, we focused on the influence of isolated metals (La, Ba, Mn, Fe) on the polymer precursor formation by means of Raman, FT-IR, scanning electron microscopy and differential scanning calorimetry, showing that the presence of metal ions, especially iron, increases the oxidation rate of the polymer precursor, while the presence of barium leads to a higher degree of polymerization, preventing partial oxidation of the polymer at low temperatures and allowing the presence of nitrates at the combustion stage. Nevertheless, when all metals are present, the polymer precursor showed a largely homogeneous microstructure with a global average influence from all cations. Finally,we studied the microstructure evolution of nanoparticles obtained after calcination above 700 °C. SAXS and TEM analysis suggests that the formation of square-shaped nanoparticles below 900 °C and coalescence leads to the formation of larger-sized and round-shaped nanoparticles at 900 °C.

The present work deals with a new methodology to produce a sustainable carbon material through a simple synthetic route, and its performance as Ni-based electrocatalyst support. Such porous carbon was easily synthesized from the raw Kraft black liquor, a by-product rich in lignin, by chemical polymerization in alkaline medium with a nickel source and subsequent activation by CO2. Nickel nanoparticles were homogeneously distributed in the samples, as revealed by STEM images. XPS analyses and BET method indicated the presence of Ni species in the synthesized material and a micro-mesoporous structure, respectively. The electrochemical characterization in NaOH and NaOH+ ethanol solutions showed that the as-prepared material has the relevant potential to be applied as electrocatalyst for Ethanol Oxidation Reaction (EOR) in alkaline medium

Yucca schidigera Roezl. (yucca) is one of the major industrial sources of steroid saponins used as animal and human food additives. This work describes a new, systematic and reproducible three-step method by medium and high-pressure liquid chromatography (under RP, NP and RP conditions), for the isolation and purification of three groups of saponins, which were further purified in six sub-fractions, and finally into twelve individual steroid saponins previously reported in Y. schidigera. In accordance to the increasing applications of yucca extracts, further analytical, biological and physicochemical studies are still required. The presented method is applicable to the preparation of steroids saponins previously reported in commercial extracts of Y. schidigera, both as highly purified mixtures of defined composition, including twelve pure components.

Doping with atom impurities is currently an attractive mechanism to enhance the optical absorption capacity of TiO2 and other inorganic oxide semiconductors. In this work, the electronic and optical properties of carbondoped TiO2(B) (100) ultrathin surfaces were investigated using density functional theory (DFT) calculations, with the aim of evaluating their potential as photoelectrode for dye-sensitized solar cells (DSSCs). Carbon impurities were introduced onto oxygen sites located in the proximity of the surface and catechol was employed as a sensitizer model, focusing our study mainly on the evolution of the semiconductor’s electronic structure. The reduction of the band gap energy resulting from impurity states within the valence band of C-doped TiO2(B) was confirmed. Moreover, our findings suggest that C doping not only induces a stronger interaction between the catechol molecule and the surface but also, could increase the light absorption capacity of the material. Thus, carbon-doped TiO2(B) could act as a promising semiconductor for DSSCs and other photochemical systems.

The coloration in citrus and other plants is<br />due the accumulation of carotenoids, which are<br />isoprenoid pigments of 40 carbon atoms (C40)<br />and its derivatives. The enzymatic function of<br />CCD4a has been associated previously with the<br />metabolism of carotenoids in Chrysanthemum<br />morifolium [1]. The flowers with white petals<br />showed a high expression of CCD4a in contrast<br />with those with yellow petals that showed a low<br />expression. The interpretation was that the<br />synthetized carotenoids are degraded by CCD4a<br />to produce colorless petals [1]. In saffron stigmas<br />(Crocus sativus) CCD4a and CCD4b were<br />associated with the transformation of zeaxanthin<br />in the apocarotenoids responsible by the color,<br />taste and aroma of this so appreciated spice

Semiempirical, ab initio and DFT methods were applied to the study of proton transfer in the ground and some of the excited states of 3-hydroxyflavone, 3-hydroxychromone and the γ-pyrone ring contained in both molecules. Geometry optimizations and reaction-path potential energy curves for the normal and proton-transferred isomers were performed employing AM1 and B3LYP methodologies (in the last case both with the 3-21G and 6-31G(d,p) basis sets). Electronic transitions were calculated employing the ZINDO-S/CI method, while the reaction path for the proton transfer in the lowest singlet and triplet excited states were calculated employing the CIS/6-31G(d,p) method on the γ-pyrone ring. The singlet–singlet excitation in 3HF is calculated at 302 nm (experimental 325 nm) while the fluorescence band is calculated at 526 nm (experimental 525 nm). The data calculated for 3HC and the γ-pyrone explain the intervention of the triplet states in the dynamics of ESIPT in these molecules.

A large body of data exists about the interaction between curved π systems and C60 or C70. However, little is known about the interaction with fullerenes smaller than C60 or larger than C70. To fill that gap, we studied, by means of density functional theory (M06-2X), the interaction between corannulene, pentaindenocorannulene, C60H28 buckycatcher and the following fullerenes: C44, C50, C80, C90, C100, C180 and C240. For fullerenes smaller than C60, their high reactivity facilitated the covalent addition to the hosts assayed. Yet, the reaction energies determined for the covalent addition were comparable to those calculated for the formation of supramolecular complexes. Thus, the receptor may host a fullerene and at least have another one attached. As expected, for fullerenes larger than C70, supramolecular complexes were preferred over covalent assemblies. The binding energies with bowls increased with the size of the fullerenes in a non-monotonic fashion since they depended on the shape of the fullerene. Indeed, for one C80 isomer, it is possible to find a region which forms a complex with corannulene that is stronger than C60@corannulene, while another region exists whose interaction with corannulene is weaker. As the size of the fullerene becomes larger, ball–socket interactions are weakened, and CH–π interactions become important, accounting for the large interaction determined for corannulene and graphene. Finally, for the buckycatcher, the maximum encapsulation energy among the fullerenes assayed was displayed by C90. The fullerenes C80, C90 and C100 formed complexes with the buckycatcher which are stronger than in C60@buckycatcher. Copyright © 2014 John Wiley & Sons, Ltd.

This study describes the microbial community richness, -dynamics, and -organization of four full-scale anaerobic digesters during a time-course study of 45 days. The microbial community was analyzed using a Bacteria- and Archaea-targeting 16S rRNA gene-based Terminal-Restriction Fragment Length Polymorphism approach. Clustering analysis separated meso- and thermophilic reactors for both archaeal and bacterial communities. Regardless of the operating temperature, each installation possessed a distinct community profile. For both microbial domains, about 8 dominant terminal-restriction fragments could be observed, with a minimum of 4 and a maximum of 14. The bacterial community organization (a coefficient which describes the specific degree of evenness) showed a factor 2 more variation in the mesophilic reactors, compared with the thermophilic ones. The archaeal community structure of the mesophilic UASB reactor was found to be more stable. The community composition was highly dynamic for Bacteria and Archaea, with a rate of change between 20–50% per 15 days. This study illustrated that microbial communities in full-scale anaerobic digesters are unique to the installation and that community properties are dynamic. Converging complex microbial processes such as anaerobic digestion which rely on a multitude of microbial teams apparently can be highly dynamic.

The Tn antigen (GalNAc-O-Ser/Thr) is one of the most specific human cancer-associated structures. In the present study we characterize the biochemical and functional properties of the Myrsine coriacea lectin (McL). We show that McL is an unusual high molecular weight highly glycosylated protein, which displays a strong Tn binding activity. The lectin exhibits in vitro inhibition of proliferation in the six cancer cell lines evaluated, in a dose-dependent manner (the strongest activity being against HT-29 and HeLa cells), whereas it does not exhibit toxicity against normal lymphocytes. McL could be exploited in the design of potential new tools for the diagnosis or treatment of cancer

A summary of the resource.
Background: The cestode Echinococcus granulosus - the agent of cystic echinococcosis, a zoonosis affecting humans and domestic animals worldwide - is an excellent model for the study of host-parasite cross-talk that interfaces with two mammalian hosts. To develop the molecular analysis of these interactions, we carried out an EST survey of E. granulosus larval stages. We report the salient features of this study with a focus on genes reflecting physiological adaptations of different parasite stages. Methodology/Principal Findings: We generated ,10,000 ESTs from two sets of full-length enriched libraries (derived from oligo-capped and trans-spliced cDNAs) prepared with three parasite materials: hydatid cyst wall, larval worms (protoscoleces), and pepsin/H+-activated protoscoleces. The ESTs were clustered into 2700 distinct gene products. In the context of the biology of E. granulosus, our analyses reveal: (i) a diverse group of abundant long non-protein coding transcripts showing homology to a middle repetitive element (EgBRep) that could either be active molecular species or represent precursors of small RNAs (like piRNAs); (ii) an up-regulation of fermentative pathways in the tissue of the cyst wall; (iii) highly expressed thiol- and selenol-dependent antioxidant enzyme targets of thioredoxin glutathione reductase, the functional hub of redox metabolism in parasitic flatworms; (iv) candidate apomucins for the external layer of the tissuedwelling hydatid cyst, a mucin-rich structure that is critical for survival in the intermediate host; (v) a set of tetraspanins, a protein family that appears to have expanded in the cestode lineage; and (vi) a set of platyhelminth-specific gene products that may offer targets for novel pan-platyhelminth drug development. Conclusions/Significance: This survey has greatly increased the quality and the quantity of the molecular information on E. granulosus and constitutes a valuable resource for gene prediction on the parasite genome and for further genomic and proteomic analyses focused on cestodes and platyhelminths
