CRISPR-Cas systems in prokaryotic cells provide an adaptive immunity against invading nucleic acids. For example, phage infection leads to addition of new immunity (spacer acquisition) and DNA cleavage (interference) in the bacterial model species Streptococcus thermophilus, which primarily relies on Cas9-containing CRISPR-Cas systems. Phages can counteract this defense system through mutations in the targeted protospacers or by encoding anti-CRISPR proteins (ACRs) that block Cas9 interference activity. Here, we show that S. thermophilus can block ACR-containing phages when the CRISPR immunity specifically targets the acr gene. This in turn selects for phage mutants carrying a deletion within the acr gene. Remarkably, a truncated acrIIA allele, found in a wild-type virulent streptococcal phage, does not block the interference activity of Cas9 but still prevents the acquisition of new immunities, thereby providing an example of an ACR specifically inhibiting spacer acquisition.

We used a density functional that incorporates van der Waals interactions to study hydrogen adsorption onto Pt atoms attached to carbon-vacancies on graphene layers, considering molecular and dissociated hydrogen-platinum coordination structures. PtH2 complexes adsorbed on several sites of pristine graphene were also studied for comparison. Our results indicate that both a Kubas-type dihydrogen complex and a classic hydride without Hsingle bondH bond are the preferential PtH2 systems on the vacancy site of graphene. In contrast, the Kubas complex is unstable onto pristine graphene and the hydride is obtained at all adsorption sites. Our simulations suggest that the C-vacancy decreases the reactivity of the metal decoration, allowing a non-dissociative hydrogen adsorption. The H2 molecule is oriented almost perpendicular to the outermost Csingle bondPt bond, interacting also with the graphene surface through σ-H and π-C states. This stabilization of the Kubas-type complex could play a very important role for hydrogen storage in Pt-decorated carbon adsorbents with vacancies.

A summary of the resource.
A new mononuclear complex of zinc(II), [Zn(HL)2]∙2DMF (H2L = (E)-N′-((E)-(hydroxyimino)butan-2-ylidene)salicyloylhydrazide, DMF = N,N-dimethylformamide), was prepared and characterized. Single-crystal X-ray crystallography revealed a six-coordinate zinc(II) surrounded by nitrogen of the oxime function and oxygen and distal nitrogens of the acylhydrazone group. This entity also exists in solution as demonstrated by 1H-NMR and potentiometric titrations. The computational analysis showed that the molecular orbitals involved in the main electronic transitions of the complex species in solution are centered on the ligand with negligible contribution of the metal ion. The photophysical properties of the complex were evaluated in solution and in the solid state. Luminescence studies showed that the solid has a strong emission at 550 nm with a large Stokes shift with respect to absorption. The solid state fluorescence emission is ascribed to ligand-centered and/or ligand-to-ligand charge transfer transitions, following the DFT results in solution. A comparison with a previously reported mononuclear [Zn(HL)2] allowed the investigation of the influence of DMF molecules in the structural packing and the luminescence properties.

Los a-alquil-~-hidroxiesteres quirales son sintones de particular yersatilidad en estrategias sinteticas ya que presentan dos estereocentros en una molecula con muchas posibilidades de derivatizaci6n. La sintesis biocatalitica de los mismos por reducci6n del a-alquil-~-cetoester precursor resulta un metodo muy conveniente para su obtenci6n. Sin embargo no se cuenta aun con biocatalizadores que permitan acceder en forma general y econ6mica a todos los is6meros 6pticos. La mayoria de los biocatalizadores reportados siguen la regia de Prelog, permitiendo acceder s610 a los is6meros con configuraci6n 3S. El objetivo del presente trabajo consisti6 en la identificaci6n de reductasas con actiyidad anti Prelog y el desarrollo de cepas recombinantes que expresaran las mismas de modo de completar el espectro de biocatalizadores disponibles para la sintesis de a-alquil-~-hidroxiesteres quirales. Para lograr identificar nuevas enzimas que permitieran construir estos biocatalizadores, se desarro1l6 un metoda rflpido de screening para la identificaci6n de genes correspondientes a enzimas con actividad reductasa. Se utiliz6 como modelo la cepa Paucimonas lemoignei, la cual expresa una ~-cetoester reductasa con actividad anti Prelog, aunque no se conoce el gen que la codifica. Utilizando este metodo fueron identificados genes que codifican para posibles ~-cetoester reductasas, uno de los cuales fue clonado y expresado en diyersas cepas de E. coli. Las cepas recombinantes generadas fueron utilizadas en diversos ensayos de biotransformaci6n con el objetivo de caracterizar su especificidad de sustrato y su diastero y enantioselectividad. Uno de los biocatalizadores desarrollados present6 actividad ~-cetoester reductasa sobre todos los a-alquil-B-cetoesteres utilizados como sustratos, exhibiendo actiyidad anti Prelog en la reducci6n de a-alil y a-propargil acetoacetato de etilo. Esta cepa constituye el primer biocatalizador de celula entera para la sintesis de ami (2R, 3R) a-alquil-~-hidroxiesteres, ampliando el repertorio de biocatalizadores disponibles para la sintesis de ahidroxiesteres quirales. El metodo desarrollado se podnl utilizar en el futuro para la busqueda de nuevas ~-cetoester reductasas de interes para biocataJ.isis.

In the present work, we developed robust processes for the preparation of new antitrypanosomal benzofuroxans, E and Z isomers of 5-arylethenylbenzo[1,2- c]1,2,5-oxadiazole N 1-oxide 1– 6, in multigram batch through Wittig−Boden conditions as the key synthetic step. In these conditions, the generation of the benzofurazans, as secondary byproduct, was minimized.

A summary of the resource.
Standard enthalpies of formation of several thionitroso (XNS) and thiazyl (XSN) isomers, with X = H, F, Cl, Br, OH, SH, NH2, CH3, CF3, and SiF3, were determined using coupled cluster (CC) theory with Dunning's correlation consistent basis sets cc-pVXZ and aug-cc-pV(X+d)Z, G3 and CBS-QB3 model chemistries, as well as the B3LYP DFT method. The results support the idea that the electronegativity of the bonding atom in the substituent is correlated, albeit not perfectly, with the relative stability of the XNS over the XSN isomer. A detailed study was performed on the parent isomers HNS and HSN. They exhibit a singlet 1A‘ ground state (as all the other molecules) at 5.4 and 9.4 kcal/mol below their first excited 3A‘ ‘ state, respectively (CCSD(T)/CBS calculations). The enthalpies of formation of the isomers at the CCSD(T)/CBS limit, including core valence correlation, and spin−orbit splitting, are ΔfH°298(HNS) = 55.3 ± 1 kcal/mol and ΔfH°298(HSN) = 75.4 ± 1 kcal/mol. The activation energy at 0 K for the HNS → HSN isomerization was determined as 63.0 and 64.2 kcal/mol at the CCSD(T)/CBS and B3LYP/6-311+G(3df,2p) levels of theory, respectively. Additional calculations were performed for the cationic isomeric pairs XNS+/XSN+ with the general result that ionization increases the relative stability of the thionitroso isomer with respect to the thiazyl isomer. In the cases of ClSN and BrSN, for which the thiazyl isomer is the most stable neutral species, the thionitroso isomer is more stable for the cations. The average deviation of the B3LYP/6-311+G(3df,2p) ionization potentials with respect to the G3 model chemistry is only 0.1 eV.

Ab initio second-order Møller-Plesset (MP2), complete active space multiconfigurational SCF (CASSCF), multireference second-order perturbation theory (CASPT2) and multireference configuration interaction (MR-SDCI) calculations have been performed on the structure of O4 with D3h symmetry. The results have been compared with the hypervalent CO3 and SO3 molecules. It was found that the three molecules present two stable structures with symmetry C2v and D3h respectively. While most of the methods predict the former to be the most stable structure for CO3 — although CASPT2 and Davidson's correct MR-SDCI gave the opposite result — all methods predict the D3h structure as the global minimum for O4 and SO3. At the theoretical level employed here O4 is stable towards decomposition into O3(1A1) and O(3P) by more than 40 kcal mol−1, for which reason it is suggested that it should be observable among the products of the reaction of oxygen atoms with ozone molecules.

A summary of the resource.
An ab initio Hartree-Fock and second-order Møller-Plesset study of the radical cation obtained by ionization of the formaldehyde-HF hydrogen-bonded dimer is reported. The results are compared with those obtained previously for the radical cations obtained from the formaldehyde-water complex. An exploration of the [H2CO · HF]+· potential energy surface was done using the 3-21G basis set. The whole of the structures found were then refined using the 6-31G(d,p) basis set and — for the most interesting structures — 6-31+G(d,p) and 6-311++G(2d,2p). In all cases the calculations were performed using the unrestricted formalism, checking always that spin contamination remained below normal thresholds. Although most of the structures found correspond closely to those found in the case of water, the HF acceptor is not able, in general, to abstract the proton from formaldehyde. Thus, contrary to the case of water, the most stable CH2O · HF radical-cation complex has both the charge and the unpaired electron located on the formaldehyde fragment. However, HF behaves in the same fashion as H2O in the sense that it is able to stabilize the hydroxymethylene radical cation with respect to the formaldehyde radical cation.

A summary of the resource.
We present a Density Functional Theory (DFT) based study of the structural and magnetic properties of the (001) surface of the semiconducting oxide ZnFe2O4 (spinel structure). The calculations were performed using the DFT based ab initio plane wave and pseudopotential method as implemented in the Quantum Espresso code. The all electron Full-potential linearized-augmented-plane-wave method (FP-LAPW) was also employed to check the reproducibility of the plane wave method. In both calculations the DFT+U methodology was employed and different (001) surface terminations of ZnFe2O4 were studied. We find that the surface terminated in Zn is the stable one. For all the (001) surface terminations our calculations predict that the Zn-Fe cationic inversion (anti-sites), which are defects in bulk ZnFe2O4, becomes stable and an integral part of the surface. Also, a ferrimagnetic behavior is predicted for the case of anti-sites in the superficial layer. Our results for different properties of the surface of ZnFe2O4 are compared with those obtained in bulk samples and those reported in the literature.

Trifluoroacetonitrile (CF3CN) is a decomposition product from the promising dielectric heptafluoro isobutyronitrile (C4F7N) under partial discharge process, so can be used to detect failures in gas-insulated switchgear. In this work, we study through density functional theory (DFT) calculations the adsorption of CF3CN on pristine aluminium nitride nanosheets (AlNNS) and defective Al-vacancy (vAl) and N-vacancy (vN) AlNNS, in order to explore the potential of these nanomaterials as sensors for this gas. Our energetic analysis shows negligible adsorption energy for the pristine surface but strong interactions can be observed between the molecule and the monovacancy-defected surfaces, with adsorption energy values estimated to be about − 2.2 and − 3.1 eV for CF3CN/vN-AlNNS and CF3CN/vAl-AlNNS, respectively, which suggests chemisorption processes. Furthermore, our geometric and bonding studies revealed the formation of new N—Al interactions between gas and monovacancy-defected surfaces, with serious distortions for the vAl-AlNNS nanosheet. The electronic density of states and band structure analysis reveals moderate band gap variations after gas adsorption. In contrast, significant changes in the work function and the magnetic moment of defective AlNNS were computed after gas exposition, for all optimized adsorption configurations. Therefore, based on our results, vacancy-defected AlNNS could be suggested as possible material for CF3CN gas sensing.

Abstract
A summary of the resource.
The structure of the title compound, C32H51NO6, was determined from 62-year-old crystals at room temperature and refined with 100 K data in a monoclinic (C2) space group. This compound with a triterpenoid structure, now confirmed by this study, played an important role in the determination of the structure of lanosterol. The mol­ecules pack in linear O—H⋯O hydrogen-bonded chains along the short axis (b), while parallel chains display weak van der Waals inter­actions that explain the needle-shaped crystal morphology. The structure exhibits disorder of the flexible methyl­heptane chain at one end of the main mol­ecule with a small void around it. Crystals of the compounds were resistant to data collection for decades with the available cameras and Mo Kα radiation single-crystal diffractometer in our laboratory until a new instrument with Cu Kα radiation operating at 100 K allowed the structure to be solved and refined.

There is still an international need to develop broad-spectrum sunscreen products with an adequate UVB/UVA balance, while the approved filters available in the UVA are scarce. Currently, one of the few UVA filters approved in the United States and Europe is tert-butylmethoxydibenzoylmethane (BMDM, avobenzone). However, this compound is unstable from aphotochemical point of view and cannot be used in combination with certain sunscreens. In this paper, we investigate the photochemical behavior of a set of dibenzoylmethanes and chalcones. In particular, we carry out their absorption and emission spectra, evaluate their photochemical degradation, and study their generation of free radicals and singlet oxygen photoproduction. Two compounds resultedin having the basic properties of UVA filters (2′-hydroxy-4-methoxychalcone and 2′-hydroxy-4-methoxydibenzoylmethane). Further studies are proposed, such asformulating the compoundsinto emulsions or other commoncosmetic presentations, as well as combining them with broadly-used UVB filters. We have also considered the need to establish its toxicological profile.

A summary of the resource.
Leaves of Palicourea marcgravii were extracted successively with hexane, ethyl acetate and ethanol in order to evaluate their acaricidal activity on larvae and adult stages of the cattle tick Rhipicephalus (Boophilus) microplus. The ethyl acetate extract showed the highest bioactivity of the tested extracts, which contained 0.12% monofluoroacetic acid. On engorged female, the ethyl acetate extract showed a lethal concentration 50% – LC50 = 30.08mgml−1, inhibitory concentration 50% – IC50 = 5.79mgml−1 and lethal time 50% – LT50 = 4.72 days; 100% reproduction was controlled at concentrations of 50mgml−1 and on larvae the ethyl acetate extract showed a LC50 = 2.46mgml−1. No alkaloids were detected in any of the extracts. This is the first report on the acaricidal activity of P. marcgravii extracts against R. microplus as well as the acaricidal properties of a plant species containing monofluoroacetic acid.

A summary of the resource.
An unsupplemented peach jam and four peach jam samples prepared by supplementing the former with increasing amounts of Aloysia citriodora aqueous extract were subjected to sensory testing. A trained panel of eight assessors initially identified (reaching consensus) the relevant sensory attributes and then rated the jam samples on an individual basis. The jam samples were later evaluated for overall liking and willingness to purchase by a group of 95 interested consumers, who also provided their individual appraisal of the sensory features of the various samples by responding to an open-ended question. Overall, consumers appeared not to accept the sensory modifications introduced in traditional peach jam by the addition of A. citriodora extract, even though they were expressly acquainted with its health benefits. The degree of acceptance of the extract supplement decreased significantly with increasing amounts of extract in the jam. The groups identified to have the least and the greatest tolerance to the sensory modifications introduced in the supplemented jams differed in gender distribution, with male consumers having a greater tolerance than females.

A summary of the resource.
A new type of organic–inorganic hybrid precursor to mixed-halide optical materials is described by using Ba5(CF2ClCOO)10·7H2O as an example. This heterohaloacetate belongs to the family of extended inorganic hybrids in which metal connectivity in three dimensions is achieved via metal–organooxygen–metal bridges. Thermolysis in the solid state and in solution yield crystalline BaFCl at temperatures below 300 °C. Chlorodifluoromethyl groups act as chlorine and fluorine source, making the organic moiety a de facto single-source precursor for the anionic portion of the target mixed halide. Solution thermolysis in the presence of Yb3+–Er3+ sensitizing–activator pairs yields rare-earth-doped BaFCl nanocrystals capable of NIR-to-visible photon upconversion. Analyses of the composition, morphology, structure, and luminescence of these nanocrystals demonstrate that heterohalocetates serve as dual halogen sources for mixed-halide optical materials. Findings presented in this article enlarge the synthetic toolbox to access mixed halides at temperatures compatible with solution synthesis and processing.
