A summary of the resource.
The volumes of activation for the spontaneous, base-, and acid- catalysed path of the hydrolysis reaction of a series of trans-[Co(MeNH2)(NH3)4X](3-n)+ ions (X = Cl-, Br-, (ONO2)- (OSO3)2-) have been determined in order to establish analogies with the dissociative trends found in previous work with the spontaneous hydrolysis of neutral ligands from the same cores. While for the base catalysed path a significant decrease in the activation volume is found on going from the {Co(NH3)5} to the trans- {Co(MeNH2)(NH3)4} inert skeleton (i.e. 9.8, 12.5, 4.0 and 9.1 cm3 mol-1 for the chloro, bromo, nitrato and sulfato derivatives), no significant changes are observed for the same complexes in the spontaneous reaction. The trends are rationalized in terms of the important changes occurring in electrostriction factors for the DCB and Id intimate mechanisms operating and the important increase in the degree of dissociativeness due to the presence of a trans-methylamino ligand. For the acid catalysed path the differences are much more difficult to assess, specially taking into account the limited information available as well as the inherent errors involved in the rate constant determination.

Lonar Lake is a unique saline and alkaline ecosystem formed by meteor impact in the Deccan basalts in India around 52,000 years ago. To investigate the role of methylotrophy in the cycling of carbon in this unusual environment, stable-isotope probing (SIP) was carried out using the one-carbon compounds methane, methanol and methylamine. Denaturing gradient gel electrophoresis fingerprinting analyses performed with heavy (13)C-labelled DNA retrieved from sediment microcosms confirmed the enrichment and labelling of active methylotrophic communities. Clone libraries were constructed using PCR primers targeting 16S rRNA genes and functional genes. Methylomicrobium, Methylophaga and Bacillus spp. were identified as the predominant active methylotrophs in methane, methanol and methylamine SIP microcosms, respectively. Absence of mauA gene amplification in the methylamine SIP heavy fraction also indicated that methylamine metabolism in Lonar Lake sediments may not be mediated by the methylamine dehydrogenase enzyme pathway. Many gene sequences retrieved in this study were not affiliated with extant methanotrophs or methylotrophs. These sequences may represent hitherto uncharacterized novel methylotrophs or heterotrophic organisms that may have been cross-feeding on methylotrophic metabolites or biomass. This study represents an essential first step towards understanding the relevance of methylotrophy in the soda lake sediments of an unusual impact crater structure

La industria del cuero se encuentra entre las seis industrias mas importantes del pais por su nivel ocupacional, incidencia en el PBI nacional y su capacidad exportadora. El volumen de efluentes producidos por el sector se estima en 2:960.000 m3 por afio y se caracteriza por su elevada carga organica e importante nivel de toxicidad debido a su contenido en sulfuro y cromo. En el Departamento de Bioingenieria se ha desarrollado un proceso de depilado enziIruitico de pieles ovinas (con extensi6n futura a pieles bovinas) que permite disminuir la carga organica y t6xica de estos efluentes al recuperar el pelo y eliminar el uso de sulfuros en los bafios de depilado. El objetivo de esta tesis es profundizar en el conocirniento de tres aspectos de este proceso: I) el rnicroorganismo, II) las enzimas presentes en el extracto depilante y III) el proceso de su producci6n. La cepa seleccionada como buena productora de enzimas depilantes se identificó como perteneciente a la especie B. subtilis segUn estudios morfo16gicos, culturales y fisio16gicos realizados. Se estudió la incidencia en la producción de proteasas de los componentes del medio de cultivo: caldo nutritivo, peptona, triptona, extracto de carne, extracto de levadura, glucosa, ~Cl, Mg2+, Ca2+, Fe2+, Mn2+ y Zn2+. Los resultados obtenidos mostraron que el caldo nutritivo es un medio apropiado para sustentar el crecirniento y la producción de proteasas de este Bacillus, que la actividad proteolitica producida aument6 con la adici6n al medio de cultivo de extracto de levadura (3 g/l), glucosa (5 g/l), Ca2+(3 mM), Mn2+ (0.8 mM) y disminuy6 con ~Cl (4 g/l) y Fe2+ (0.1 mM), que la actividad del extracto crudo producido se mantiene al suplantar el caldo nutritivo por dos de sus componentes la triptona y el extracto de carne En la busqueda de un medio de cultivo econórnico que permita su utilización a nivel industrial, se 10gr6 una producci6n de proteasas del orden de la obtenida con caldo nutritivo en un medio que contiene peptona y glucosa en cultivo por lote alimentado donde la concentraci6n de glucosa en el medio de cultivo varió entre - 0-2.5g/l. Del estudio de los mecanismos regulatorios que intervienen en la producción de proteasas se vió que: I) la sintesis de parte 0 el total de las proteasas es inducible y esta sujeta a represi6n catab6lica por glucosa; II) la producci6n de proteasas sigue una cinetica de formaci6n de producto mixta; III) las enzimas contenidas en el extracto crudo estan invoiucradas en por 10 menos dos procesos, proteasas ligadas a la hidr6lisis de fuentes de carbono y nitr6geno complejas y proteasas vinculadas a la esporulaci6n. Mediante SDS-PAGE y electroforesis de actividad se caracterizaron las proteasas presentes en el extracto crudo, 1 serino proteasa y 3 proteasas cuyas caracteristicas se asirnilan al de las metalo proteasas. El PM relativo de las metalo proteasas es de 151, 106 Y 39 kDa, su rango de actividad se extiende entre pH 5 Y 10 y su actividad se vio inhibida parcialmente con PMSF y totalmente con EDT A. La serino proteasa tiene un PM relativo de 80 kDa, su rango de actividad se limita a pH 7-8, se inhibe parcialmente con pCMB y totalmente con PMSF y EDTA y su actividad depende del ion Ca 2+.

Aims: To combine molecular and cultivation techniques to characterize the methanotrophic community in the soil–water interface (SWI) and rhizospheric soil from flooded rice fields in Uruguay, a temperate region in South America. Methods and Results: A novel type I, related to the genus Methylococcus, and three type II methanotrophs were isolated from the highest positive dilution steps from the most probable number (MPN) counts. Potential methane oxidation activities measured in slurried samples were higher in the rhizospheric soil compared to the SWI and were stimulated by N-fertilization. PmoA (particulate methane monooxygenase) clone libraries were constructed for both rice microsites. SWI clones clustered in six groups related to cultivated and uncultivated members from different ecosystems of the genera Methylobacter, Methylomonas, Methylococcus and a novel type I sublineage while cultivation and T-RFLP (terminal restriction fragment length polymorphism) analysis confirmed the presence of type II methanotrophs. Conclusions: Cultivation techniques, cloning analysis and T-RFLP fingerprinting of the pmoA gene revealed a diverse methanotrophic community in the rice rhizospheric soil and SWI. Significance and Impact of the Study: This study reports, for the first time, the analysis of the methanotrophic diversity in rice SWI and this diversity may be exploited in reducing methane emissions.

BACKGROUND: We sought to evaluate, in patients on a liver transplantation waiting list, potential biomarkers of the base calcineurin pathway activity with use of a new model of nonstimulated peripheral blood mononuclear cells (PBMC) and ex vivo response to tacrolimus (TAC). METHODS: The calcineurin pathway activity was explored ex vivo in stimulated and nonstimulated PBMC from 19 patients. The inhibition of NFAT1 translocation to PBMC nuclei, expression of intracellular IL-2, and membrane CD25 in different T-cell subsets were measured by multiparametric flow cytometry before and after exposure to TAC. We also studied the influence on the individual response of polymorphisms in 3 key genes of the calcineurin pathway: PPIA, PPP3CA, and IL2RA. RESULTS: All pharmacodynamics profiles closely fitted an I/Imax sigmoid model. Interindividual variability was higher in nonstimulated than in stimulated conditions, as well as in the presence of TAC. IL-2+CD8+ cells at TAC Imax showed the highest interindividual variability, suggesting its usefulness as a biomarker of individual TAC effects integrating many different sources of regulation and variability. Moreover, in the absence of TAC, patients with end-stage liver disease exhibited lower NFAT1 translocation and T-cell activation than healthy volunteers from a previous study under similar conditions. Multivariate statistical analysis showed strong and significant associations between TAC pharmacodynamic parameters and 2 polymorphisms in the gene-coding cyclophilin A (rs8177826 and rs6850). CONCLUSIONS: We show the feasibility of using nonstimulated PBMCs to explore the calcineurin pathway under more physiologic conditions and point toward potential biomarkers for TAC pharmacodynamic monitoring.

American trypanosomiasis or Chagas disease, caused by the protist parasite Trypanosoma cruzi (T. cruzi), is a major health concern in Latin America. In the search for new bioactive compounds, eight Pd(II) and Pt(II) complexes of thiosemicarbazones derived from 1-indanones (HL) were evaluated as potential anti-T. cruzi compounds. Their unspecific cytotoxicity was determined on human erythrocytes. Two physicochemical features, lipophilicity and redox behavior, that could be potentially relevant for the biological activity of these complexes, were determined. Crystal structure of [Pd(HL1)(L1)]Cl·CH3OH, where HL1 = 1-indanone thiosemicarbazone, was solved by X-ray diffraction methods. Five of the eight metal complexes showed activity against T. cruzi with IC50 values in the low micromolar range and showed significantly higher activity than the corresponding free ligands. Four of them resulted more active against the parasite than the reference antitrypanosomal drug Nifurtimox. Anti-T. cruzi activity and selectivity towards the parasite were both higher for the Pd(II) compounds than for the Pt(II) analogues, showing the effect of the metal center selection on the biological behavior. Among both physicochemical features tested for this series of compounds, lipophilicity and redox behavior, only the former seemed to show correlation with the antiproliferative effects observed. Metal coordination improved bioactivity but lead to an increase of mammalian cytotoxicity. Nevertheless, some of the metal complexes tested in this work still show suitable selectivity indexes and deserve further developments.<br /><br />

Divalproex sodium extended-release dosage form (divalproex-ER) has been promoted as innovative formulation for the treatment of epilepsy and manic disorders, and for migraine headache prevention, with the advantage of being dosing once a day. Due to a significant decreasing in the peak-trough fluctuation of plasma valproic acid levels, in comparison with the twice-daily dosing of conventional delayed-release formulations (divalproex-DR), concentration-dependent side effects would be prevented. However the main constraint for divalproex-ER usage is the need to be administered in a higher daily dose, because of its lower bioavailability, in order to prevent eventual breakthrough seizures when patients are switched from the twicedaily divalproex DR regimen. Taking into account free plasma drug levels, divalproex ER/DR relative bioavailability could be assessed as low as 75% in fasting condition. In order to overcome the need of increase divalproex-ER daily dose, maintenance of the twice-daily regimen is suggested. Divalproex-ER administered every 12 hours not only increases steady state trough concentration to a higher value in comparison with divalproex-DR, avoiding inefficacy of the treatment, but also achieves the safest manner to treat patients with valproic acid because of reaching practically a plateau profile of drug levels

Backgrounds: Sleep restriction is considered a stressful condition itself, causing a wide variety of physiological alterations, from cognitive and hormonal to immunological status. In addition, it is established that stress in mother rats can modify milk ejection, milk composition, and maternal care of the pups. Also, sleep disturbances during the early stages of motherhood are a common feature of all studied species. In this context, while the impacts of sleep disruption in non-lactating animals were extensively investigated, its repercussions during the initial phases of motherhood have been poorly explored. Therefore, we wonder if maternal behavior, milk ejection and its macronutrient composition would be disrupted when mother rats are subjected to an additional acute or chronic sleep restriction to the already existing sleep disturbances. Methods: Lactating rats were implanted with unilateral electrodes for polysomnographic recordings and for deep brain electrical stimulation into mesopontine waking-promoting area (for sleep deprivation). During the early postpartum period (postpartum day 5–9), mother rats were randomly assigned into one of three groups: chronic sleep restriction group (CSR; 6 h of sleep deprivation/day for five consecutive days), acute sleep restriction group (ASR; 6 h of sleep deprivation only for one day), or undisturbed group (control group). Active maternal behaviors (retrievals of the pups into the nest, mouthing, lickings [corporal and anogenital] and sniffing the pups) and passive maternal behaviors (kyphotic and supine nursing postures) were evaluated during a 30 min period without sleep restriction immediately after the sleep restriction or control period. The litter weight gain was assessed every day, and on the last experimental session mothers were milked for posterior macronutrients analysis (protein, carbohydrates and fat).Results: When compared to control group, CSR decreased the amount of milk ejected in the middle days of the sleep restriction period, while ASR did not affect this parameter. Moreover, ASR reduced milk protein content compared to control and CSR groups. Finally, compared to the control group, CSR reduced active maternal behaviors towards the end of the treatment days. Conclusions: We demonstrated that not only acute but also chronic sleep restriction impacts on the postpartum period, each one affecting different aspects of maternal behavior and lactation. Our results suggest the existence of a homeostatic recovery mechanism in breastfeeding during CSR, possibly ensuring the survival of the litter, while the decline in active maternal behaviors appears to be cumulative.

The use of Cannabis for medical purposes is rapidly expanding and is usually employed as a self-medication for the treatment of insomnia disorder. However, the effect on sleep seems to depend on multiple factors such as composition of the Cannabis, dosage and route of administration. Vaporization is the recommended route for the administration of Cannabis for medical purposes; however, there is no published research about the effects of vaporized Cannabis on sleep, neither in laboratory animals, nor in humans. Because previous reports suggested that low doses of THC have sedating effects, the aim of the present study was to characterize in rats, the acute effects on sleep induced by the administration of low doses of THC by means of vaporization of a specific type of Cannabis (THC 11.5% and negligible amounts of other cannabinoids). For this purpose, polysomnographic recordings in chronically prepared rats were performed during 6 h in the light and dark phases. Animals were treated with 0 (control), 40, 80 and 200 mg of Cannabis immediately before the beginning of recordings; the THC plasma concentrations with these doses were low (up to 6.7 ng/mL with 200 mg). A quantitative EEG analyses by means of the spectral power and coherence estimations was also performed for the highest Cannabis dose. Compared to control, 200 mg of Cannabis increased NREM sleep time during the light phase, but only during the first hour of recording. Interestingly, no changes on sleep were observed during the dark (active) phase or with lower doses of Cannabis. Cannabis 200 mg also produced EEG power reductions in different cortices, mainly for high frequency bands during W and REM sleep, but only during the light phase. On the contrary, a reduction in the sleep spindles intra-hemispheric coherence was observed during NREM sleep, but only during the dark phase. In conclusion, administration of low doses of THC by vaporization of a specific type of Cannabis produced a small increment of NREM sleep, but only during the light (resting) phase. This was accompanied by subtle modifications of high frequency bands power (during the light phase) and spindle coherence (during the dark phase), which are associated with cognitive processing. Our results reassure the importance of exploring the sleep-promoting properties of Cannabis.

A summary of the resource.
1. The electrokinetic and thermostability properties of phosphoglucose isomerase (PGI) in five species of tropical fish (Ostariophysi)—<em>Leporinus friderici, L. silvestrii, Schizodon nasuttus, Hypostomus sp</em> and <em>Pimelodus maculatus</em>—have been studied in order to study the adaptative nature of protein heterogeneity found in ectotherms.
<p>2. Unlike most diploid fishes, the PGI of these species seemed to be encoded by three, four or rive loci.</p>
<p>3. The subunits encoded by these loci occurred at different levels in the different tissues and organs analyzed.</p>
<p>4. Genetic variants at the three PGI loci were detected in two species of the family Anostomidae (<em>L. friderici</em> and <em>S. nasuttus</em>). In the family Pimelodidae, <em>P. maculatus</em> showed a genetic variant as the Pgi-1 locus.</p>
<p>5. The product of these loci could be separated in three PGI regions based on their electrophoretic mobility, tissue distribution and thermostability properties.</p>
<p>6. The more anodal region (including isozymes and allozymes) was more thermolabile than the less anodal one, which is predominant in a single tissue-skeletal muscle.</p>

Abstract
A summary of the resource.
1. Electrophoretic analyses of lactate dehydrogenase isozyme patterns of three species of temperate fish belonging to family Sciaenidae (order Perciformes) indicates that at least three LDH loci--Ldh-A, Ldh-B and Ldh-C are active. 2. The subunits encoded by these loci occurred at different levels in the different tissues and organs analyzed. 3. The products of these loci show different response to changes in temperature. The results were obtained by electrophoretic analyses. 4. Thermostability of skeletal muscle, heart and eye lactate dehydrogenase incubated at 60 degrees C for different periods of time were examined. 5. Relative activities of LDH isozymes were compared by Klebe's method to determine pattern of divergence of duplicated gene expression in the three species studied.

Abstract
A summary of the resource.
The lipid profile of milk from grazing goats supplemented with vegetable oils was evaluated. Twentyseven Saanen goats consuming pasture were grouped and supplemented with 3 concentrates: without added oil (control, C) and with added sunflower (SFO) or soybean oil (SBO) until 6% ether extract (diet basis). Fat content and sensory profile of milk were not modified. Vaccenic acid increased for SBO and SFO (1.5% vs. 2.6% and 2.7% respectively; p < 0.01), and cis-9, trans-11 conjugated linoleic acid from 0.6% to 0.8% for treated groups (p < 0.01). Oil supplementation resulted effective to decrease the saturated/unsaturated ratio of grazing goat’s milk.

Abstract
A summary of the resource.
CdTe single crystal, were grown by the Modified Markov Method. We have introduced an insulating element between the furnace and the growth chamber, trying to minimize the radiation effects in the crystal. Numerical simulation of the heat transfer phenomena predict a reduction of the axial temperature gradient in the growth chamber, confirming by numerical experimental measurements, leading to a more uniform growth. An improvement in the surface morphology and in the crystal quality as well was achieved, evaluated by the combination of X-ray rocking curves, etch pit density determination and low temperature photoluminescence.
