Abstract
A summary of the resource.
Several strains of endophytic bacteria and yeast were isolated from Daucus carota, Erythrina crista-galli, Curcubita maxima and Fortunella margarita, and tested for the reduction of two model �-alkyl-�- ketoesters. The yeast strains resulted better biocatalysts for the reduction of the tested substrates with the exception of Enterobacter agglomerans C8 strain, isolated from Erythrina crista-galli, that provided the best results for the preparation of syn (2R,3S) �-alkyl �-hydroxyesters. Among the isolated yeast strains, the Pichia sp. strain isolated from D. carota, was the best biocatalyst for production of anti (2S,3S) �-alkyl �-hydroxyesters. Our results show that the microbial endophytic community provides an interesting niche on the search for novel biocatalysts.

Las Solamlceas Json una de [as familias botAnicas mas ricas en metabolitos L .)e las partes aereas de Solanum amygdalifolium Steud. fueron aisiac.ios c :c.icntilicac.ios soiasonina y un nuevo glic6sido, [(22S, 23R, 25S)-3P, 15a, 23en-2G-ona J-3-0-{ u-L-rhamp-( I ~2)-[a-L-rhamp-( I~)l-P-D-glcp}, que hemos denominado folillmina I\. Se estudi6 la actividad biol6gica de los glieoalcaloides de la papa, a-solanina y aachaconina, asi como los encontrados en S. commersonii contra el Mido de la papa Macrosiphum euphorbiae analizandose los efectos sobre la alimentaci6n (feeding), la supervivencia y la reproducci6n. La tomatina y la mezcla demissina y 65-demissina resultaron las mas activas, disminuyendo el feeding, aumentando la mortalidad apreeiablemente y produciendo una disminllei6n significativa de la reproducei6n. La a-ehaconina produjo un descenso marcado en la reproducci6n aunque no alter6 los otros paramatros vitales del afido, mientras que la a-solanina y la mezcla de cOlrunersonina y 65-commersonina no mostraron actividad significativa. Estos resultados muestran que los compueslOs encontrados en especies de Solanum silvestres ejercen efectos deletereos signifieativos sobre el afido, de la papa y permiten una apro:-:imaci6n a la rclaci6n estructura-actividad de estos compuestos. Esto facilitara el uso racional de gennoplasma silvestre en el mejoramiento de la resistencia natural de la papa a pestes

Investigaci6n y desarrollo de nuevas N-acilhidrazonas bioactivas 11 Resumen El agrupamiento N-acilhidrazona ha sido descrito como un fannacOforo con capacidad de proveer varias actividades farmaco16gicas a traves de su interacci6n con diferentes dianas. A su vez, los heterociclos derivados de furoxano son reconocidos por su actividad bio16gica frente a distintos blancos terapeuticos. En el presente trabajo de tesis doctoral se ha realizado el disefio y sintesis quimica de nuevos compuestos hibridos que incluyen el agrupamiento N-acilhidrazona unido a heterociclos derivados del sistema metilfuroxano, fenilfuroxano y benzofuroxano. Los nuevos compuestos han sido desarrollados empleando procedimientos sinteticos sencillos y eficientes y la caracterizaci6n estructural de los mismos se ha realizado mediante tecnicas espectrosc6picas habituales. Estos compuestos hibridos se han evaluado en diferentes modelos bio16gicos y a traves de ellos ha sido posible identificar nuevos lfderes con elevados indices de selectividad frente a Mycobacterium tuberculosis, Trypanosoma cruzi y Leishmania amazonensis. Los estudios fannaco16gicos han permitido identificar nuevos compuestos con actividades analgesicas y anti-inflamatorias que actuan principalmente a traves de su capacidad de inhibir la via NF-KB y la liberaci6n de Interleuquina-8. Los compuestos que en este trabajo se identificaron con mejor perfil bio16gico, presentan en general, baja citotoxicidad y no son mutagenicos. Esto los ubica como nuevos prototipos para la realizaci6n de futuras optimizaciones moleculares.

Este trabajo plantea el desarrollo de una familia de complejos "mixtos" de 99mTc y el estudio de sus propiedades quimicas y biol6gicas, como base para el disefio racional de nuevos Radiotarmacos. Los complejos estudiados se obtuvieron por acci6n simultanea de un ligando aminoti61ico tridentado y un coligando monoti6lico sobre el "core" (TCO)3+. Los ligandos N,N bis-(2- mercaptoetil)- N',N' -diisopropiletilendiamina, N N',N'-dietiletilendiamina y N -(2- mercaptoetil)- N',N' -diisopropiletilendiamina fueron sintetizados localmente, mientras que los coligandos tiofenol, tiocresol, p-clorotiofenol y bencilmercaptano fueron obtenidos comercialmente. Los estudios quimicos se desarrollaron en escala "carrier", utilizando Re estable como modelo. Los complejos de Re fueron preparados por metodo de sustituci6n, empleando trans-oxotriclorobistrifenilfosfmo Re(V) como precursor y caracterizados por HPLC, espectroscopia UV -vis e IR, aruilisis elemental y cristalografia de rayos X. Se obtuvieron 3 tipos diferentes de complejos "mixtos": "SNS + S", "SNN + S" Y "SN + S + S", dependiendo del ligando empleado. La geometria de los mismos fue bipiramide trigonal distorsionada, piramide de base cuadrada y piramide de base cuadrada con distorsi6n trigonal, respectivamente. Todos los complejos se encontraron en forma "syn". La ¢rdida de 3 Momos de hidr6geno durante la complejaci6n permiti6 couwensar la carga positiva del centro metaIico, obteniendose complejos neutros. Los correspondientes complejos en escala "trazadora" fueron preparados con la fmalidad de evaluar su comportamiento biol6gico. La marcaci6n con 99mTc fue realizada a partir del 99mTc-glucoheptonato. EI intercambio de ligandos se produjo con rendirniento superior al 85% y dio lugar a compuestos con pureza radioquimica mayor del 90%. La identidad de los mismos fue establecida por comparaci6n con los complejos "mixtos" obtenidos en escala "carrier", por tecnicas de HPLC. La biodistribuci6n en ratones demostr6 que los complejos estudiados son capaces de atravesar la barrera hematoencefaIica intacta, ya que todos ellos fueron selectivamente captados y retenidos por el cerebro. El intercambio de los respectivos coligandos por glutati6n "in vivo" podria ser responsable del comportamiento biol6gico observado. Aunque es improbable que estos nuevos compuestos sustituyan a los radiofannacos actualmente empleados en Medicina Nuclear para la evaluaci6n de la perfusi6n cerebral, la experiencia adquirida mediante su estudio es de gran importancia y esta siendo aplicada en el diseiio de otro tipo de radiofarmacos basados en el concepto de complejos "mixtos".

A summary of the resource.
The aim of the present study was to evaluate the suitability of saliva as a biological fluid in relative bioavailability (RBA) studies, with the focus on the statistical design and data variability. A randomized, openlabel, four periods and two sequences (4 9 2) crossover RBA study in saliva of two phenytoin (PHT) 100 mg immediate-release capsules was performed. PHT is a narrow therapeutic index drug that has been widely used for epilepsy treatment for many years. Published information regarding its bioavailability is available, but plasma assessed. This study was designed and performed using saliva as the biological fluid and the simplest conditions that produce coherent results with previously published plasma studies. Pharmacokinetic parameters (Cmax, Tmax, AUC0-t, AUC0-inf, Cmax/AUC0-t, Ke, and t1/2) for each volunteer at each period were calculated. Four different BE calculations were performed: individual bioequivalence, by the method of moments, and three average bioequivalence with data averaged over the two administrations and with data of periods 1–2 and 3–4. ANOVA calculation showed no significant subject-by-formulation interaction, period and sequence effects. The intra-subject variabilities were at least 20-fold lower than the inter-subject ones for Cmax, AUC0-t and AUC0-inf. In all four BE calculations, the 90% CIs for the T/R ratios of studied pharmacokinetics parameters fell within the 80–125% range proposed by most regulatory agencies

We investigated the accumulation and toxicity of microcystin-LR (MCLR) in the digestive gland of the freshwater clam Diplodon chilensis patagonicus. Treated clams were fed with a toxic strain of Microcystis aeruginosa (NPJB1) during 6 weeks and control clams received the non-toxic strain NPDC1. Filtration rate was estimated for both groups. Toxic effects were evaluated through the hepatosomatic index (HSI) and different oxidative stress biomarkers, lipid peroxidation (content of thiobarbituric reactive substances–TBARS), protein oxidation (carbonyl groups) and reduced glutathione (GSH) levels, and enzymatic activities of superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST). The extractable MCLR measured by ELISA in digestive gland extracts showed little or no change during the first 3 weeks and increased significantly at weeks 5 and 6. HSI was reduced by 30% in treated clams at weeks 5 and 6. No significant oxidative damage to lipids or proteins was. All the antioxidant defense parameters analyzed were significantly increased at week 5 or 6. GSH increased in treated clams at week 5, reaching 62% increase at week 6. SOD, CAT and GST activities were significantly increased in treated clams by 50%, 66% and 60%, respectively, at the end of the experiment. D. chilensis patagonicus can be exposed to prolonged cyanobacterial blooms accumulating significant quantities of MCLR, which could be a risk for mammals and birds, which feed on this species and, in a lesser extent, to humans

The synthesis of some natural and synthetic biflavonoids was performed in good overall yields starting from readily available materials via high yielding aldol and Ullmann condensations. Some of these compounds, especially bichalcones, display an interesting activity against fungi, higher than that of the corresponding monomers.

A summary of the resource.
The synthesis of fused heterocycles such as thiazolidinyl-oxazolidine 3 is described starting from Tris 3 HCl. The mercaptomethyl bisoxazolidine 8 was found to convert to the corresponding thiazolidinyloxazolidine 3 and the spiroheterocycle 4 by a ring-chain-ring tautomerism, depending on the electronic nature of the ring substituents as well as the reaction conditions. This equilibration pathway is absent in the hydroxymethyl bisoxazolidines 2. Computational studies confirm that both kinetic and thermodynamic control features play a role in the product distribution.

A summary of the resource.
Disulfide-bond formation is a major post-translational modification and is essential for protein folding, stability, and function. This is especially true for secreted proteins, many of which possess great potential for biotechnological applications. Focusing on the use of Escherichia coli for the production of this class of proteins, we describe the mechanisms that maintain redox compartmentalization in the cell, with an emphasis on those that promote the formation and isomerization of disulfide bonds in the bacterial periplasm, while presenting parallel pathways in the eukaryotic endoplasmic reticulum. Based on these concepts, we review the use of E. coli as a cell factory for the production of heterologous disulfide-containing proteins using either peri- or cytoplasmic expression and, in particular, how these compartments can be tuned to improve the yield of correctly folded recombinant proteins. Finally, we describe a few examples of the production of small disulfide- rich proteins (protease inhibitors) to illustrate how soluble, active, and fully oxidized recombinants may be successfully obtained upon peri- or cytoplasmic expression in E. coli.

La Enfermedad de Chagas es una enfermedad no resuelta, causada por el parasite Trypanosoma cruzi. Existe desde hace miles de anos y afecta a millones de personas, mayoritariamente en America Latina. Hasta el momenta no existe un farmaco adecuado para tratar est a enfermedad. Con el objetivo de buscar terapias apropiadas para est a problematica, en este trabajo se disenaron, sintetizaron y caracterizaron ochenta y cinco nuevos compuestos. Se determinaron sus capacidades tripanosomicidas in vitro obteniendose mas de veinte derivados con destacada actividad anti-Trypanosoma cruzi. Se estudi6 la toxicidad de los mismos frente a celulas de mamffero determinandose la selectividad de los derivados biol6gicamente activos. Se realizaron estudios de mutagenicidad in vitro. Se realizaron estudios in vivo con tres de los derivados mas relevantes. Se identific6 un derivado con potencialidad a farmaco que posee varias ventajas. Desde el punta de vista qufmico: sencillez sintetica, producto escalable y de producci6n econ6mica. Desde el punta de vista biol6gico: activo en diferentes estadfos y cepas del parasito, baja toxicidad inespedfica, ausencia de mutagenicidad, actividad in vivo similar a la del farmaco de referencia y ausencia de genotoxicidad in vivo. Ademas, posee un mecanismo de acci6n multiple, interfiriendo en la biosfntesis de esteroles de membrana, e inhibiendo levemente las enzimas cruzipaina y tripanoti6n sintetasa.

A summary of the resource.
Five new copper and zinc heteroleptic complexes with saccharin and aminoacids with general stoichiometry Na2[M(sac)2(aa)2]⋅nH2O (M denotes Cu or Zn, sac the saccharinate ion, and aa the aminoacids) were synthesized and characterized by elemental and thermogravimetric analysis, conductimetric measurements and IR, Raman and UV–vis spectroscopies. In all the complexes, copper and zinc ions coordinated with the aminoacids through the terminal amine and carboxylate residues and with saccharin through the heterocyclic nitrogen atom. Besides, the superoxide dismutase-like activity of the heteroleptic copper complexes was evaluated and compared with the homoleptic copper amino acid complexes with the aim to observe the influence of the saccharin coordination.

Although beef tallow (BT) has been considered a hard low-trans fat convenient to be used in several bakery applications, it has some undesirable characteristics like fatty acid composition, crystallization behavior, graininess formation and poor plastic range. This work studied the modification of BT by blending at different percentages with high oleic sunflower oil (HOSFO) followed by the enzyme-catalyzed interesterification of the blends. The reduction in the solid fat content achieved by the simple blending was enhanced by the interesterification process, as a result of the increase in the concentration of the diunsaturated monosaturated type triacylglycerols. Interesterification strongly impacted too on the crystallization behavior of the blends, since products showed more homogeneous and regular crystals than the starting mixture. Results show that lipase catalyzed interesterification of BT with HOSFO offers a useful tool for the design of fats with adjustable physicochemical properties, improved with respect to that of the starting fats.

<p>Controlled syntheses give<br />unique block oligomers with alternating<br />flexible ethylene glycol and rigid<br />perylenetetracarboxylic diimide (PDI)<br />units. The number of rigid units vary<br />from n=1 to 10. PDI units were stitched<br />together by using efficient phosphoramidite<br />chemistry. The resulting<br />oligomers undergo folding in most solvents,<br />including chloroform. In their<br />ground state, these folded oligomers<br />were characterized by using Fourier<br />transform ion cyclotron resonance<br />mass spectrometry (FTICR-MS), NMR<br />spectroscopy, and electronic absorption<br />spectroscopy. FTICR-MS revealed the<br />exact masses of these sequence-controlled<br />oligomers, which confirmed the<br />chemical composition and validated<br />the synthetic strategy. The NMR neighboring<br />ring-current effect (NRE) indicates<br />the formation of cofacial p<br />stacks; the stacked aromatic rings have<br />nearly coaxial alignment akin to a<br />nanosoleniod. Nanosolenoidal shielding<br />in p stacks causes all aromatic protons<br />to shift upfield, whereas NOE in a<br />cyclic hetero-chromophoric dimer supports<br />a rotated, cofacial p-stacking orientation<br />separated by about 3.5 �.<br />Electron–phonon coupling is much<br />stronger than excitonic coupling in<br />these self-folded PDI oligomers; thus,<br />Franck–Condon factors dictate the observed<br />spectral features in visible spectra.<br />The absorbance spectrum exhibits<br />weak hypochromism due to p stacking<br />with increasing stacking units n. Finally,<br />ab initio calculations support the experimental<br />observations, indicating 3.5 �<br />cofacial spacing in which one molecule<br />is rotated 308 from the eclipsed orientation<br />and higher oligomers can adopt,<br />without a compensating energy penalty,<br />either the right/left-handed helices or<br />the 1,3-eclipsed structures. Both theory<br />and experiments validate the nano-psolenoids<br />and their novel photophysical<br />properties.</p>

The antioxidant capacity of propolis from the southern region of Uruguay was evaluated using in vitro as well as cellular assays. Free radical scavenging capacity was assessed by ORAC, obtaining values significantly higher than those of other natural products (8000 μmol Trolox equiv/g propolis). ORAC values correlated well with total polyphenol content (determined by Folin�Ciocalteu method) and UV absorption. Total polyphenol content (150 mg gallic acid equiv/g propolis) and flavonoids (45 mg quercetin equiv/g propolis) were similar to values reported for southern Brazilian (group 3) and Argentinean propolis. Flavonoid composition determined by RP-HPLC indicates a strong poplar-tree origin. Samples high in polyphenols efficiently inhibit low-density lipoprotein lipoperoxidation and tyrosine nitration. In addition, Uruguayan propolis was found to induce the expression of endothelial nitric oxide synthase and inhibit endothelial NADPHoxidase, suggesting a potential cardiovascular benefit by increasing nitric oxide bioavailability in the endothelium.

In this study, a preparation of saponins (QB-90U) extracted from leaves of Quillaja brasiliensis collected in Uruguay was evaluated as a vaccine adjuvant by comparison with alum and the well known saponinbased adjuvant, Quil A. The haemolytic activity and cellular toxicity of the saponin preparations were also evaluated. QB-90U was only slightly haemolytic and showed a low cytotoxicity when compared to Quil A. The adjuvant properties of QB-90U were assayed by sub-cutaneous immunization of mice with a preparation of inactivated bovine herpesvirus 5 (BoHV-5) either with no adjuvant or adjuvanted with QB-90U, Quil A or alum. Serum levels of anti-BoHV-5 IgG, IgG1, IgG2a, IgG2b and also IgG3 were significantly increased by QB-90U and were of the same order as those elicited by Quil A. Furthermore, high titres of neutralizing antibodies were found to be present in the serum of immunized animals from both groups. The cellular response induced by QB-90U did also reproduce the one elicited by Quil A. In fact, a robust DTH response was observed in mice immunized with both saponin preparations; as well as increased splenocytes levels of Th1-type cytokines, namely IFN-� and IL-2. Taken together, the above results confirm and extend our previous observation regarding the similarity of the responses elicited by Quil A and the saponin preparation from Q. brasiliensis (Fleck et al., 2006) and indicate that QB-90U is worth of further studies as a safe and potent vaccine adjuvant.
