AM1 calculations were performed for the absorption of H2O and CO2 molecules on the surface of model ZnO crystals. The absorption of isolated molecules of each species and the co-absorption of both compounds simultaneously were considered. It was found that the absorption of H2O near a site where CO; is already absorbed favors the process of sintering, in agreement with the experimental findings. This is explained by the formation of Zn(OH)CO3H bound to the surface, a more mobile species than the ZnO unit itself. The roundening of the grains observed in atmospheres containing dry CO2 but suppressed when H2O is present, is also explained by these calculations. After absorption of CO2, the rupture of one bond - so that diffusion of the ZnCO3 species on the surface is allowed - requires much less energy than the breaking of two bonds, necessary for ZnO migration. These facts explain why the speed of surface transport does not decrease in CO2 atmospheres while sintering is indeed slowed down.

The behavior of the stretching vibrations of the carboxylate groups in twenty one Cu(II) complexes of amino acids (aa) of general composition Cu(aa)2 or Cu(aa)2.nH2O is analyzed on the basis of previously reported vibrational spectroscopic data and on the structural characteristics of these complexes. Some general trends are pointed out and discussed. In all cases, the spectroscopic results are consistent with the monodentate binding of the COO- groups with the metal center. Brief comments on the spectroscopic behavior of the carboxylate groups in a series of polymeric Cu(II) complexes of dipeptides are also presented

Pseudomonas fluorescens CFBP2392 has been recognized as a potential biocontrol agent due to its ability to suppress damping-off and root rot disease. This isolate has antibacterial activity in vitro as many other strains from the Pseudomonas fluorescens complex. In this work, the antibacterial and antifungal activity of the strain were explored. Dual culture assays evidenced the antifungal activity of the strain against different phytopathogens: Alternaria sp., Pythium ultimun, Fusarium oxysporum, and Rhizoctonia solani. Purification of an antifungal fraction was performed by preparative HPLC from the chemical extraction of growth media. The fraction showed altered R. solani growth and ultrastructure. Transmission electron microscopy revealed the purified compound induced hypertrophied mitochondria, membranous vesicles, and a higher number of vacuoles in R. salani cytoplasm. In addition, co-cultivation of P. fluorescens CFBP2392 with R. solani resulted in an enlarged and deformed cell wall. To gain genomic insights on this inhibition, the complete genome of P. fluorescens CFBP2392 was obtained with Oxford Nanopore technology. Different biosynthetic gene clusters (BGCs) involved in specialized metabolites production including a lokisin-like and a koreenceine-like cluster were identified. In accordance with the putative BGCs identified, sequence phylogeny analysis of the MacB transporter in the lokisin-like cluster further supports the similarity with other transporters from the amphisin family. Our results give insights into the cellular effects of the purified microbial metabolite in R. solani ultrastructure and provide a genomic background to further explore the specialized metabolite potential.

Conformational diseases are a group of diverse disorders that have been associated with misfolding of specific proteins, leading to their aggregation in particular cell tissues. Despite their relevance, the mechanisms involved in neurodegenerative processes remains poorly understood. Mutations in Cu,Zn superoxide dismutase (SOD1) are implicated in death of motor neurons in amyotrophic lateral sclerosis. Among others, the SOD1G93A mutation is known to weaken the structure and this could lead to conformational variations of the protein. As an approach to understand the tissue-specific propensity of protein aggregation, we developed an experimental procedure allowing rapid extraction of variants of human SOD1 (hSOD1) produced in different tissues. Using an antibody-based affinity chromatography procedure enzymatically active hSOD was extracted, indicating preservation of its native conformation. Analysis of the eluted fractions of hSOD extracted from the brain and liver of transgenic hSODG93A rats provided evidence about heterodimers rSOD–hSODG93A formation in both extracts. Moreover, when characterized by 2-DE and MALDI-TOF/TOF MS, the extracted hSODG93A showed a complex profile suggesting the existence of various covalent modifications of the enzyme in both tissues. Thus, this method should allow following post-translational modifications of hSOD1 produced in various tissues.

A spirostanic saponin was isolated from the ethanolic extract of the aerial parts of Solanum laxum Steud. The compound, named luciamin, was characterised by NMR spectroscopy, mass spectrometry and chemical methods, as (22R, 25S)-spirost-5-en-3 b, 15 a- diol 3-O-{b-d-glucopyranosyl (1!2)-b-d-glucopyranosyl-(1!4)-[a-l-rhamnopyranosyl-(1!2)]-b-d-galactopyranoside}. The com- pound was tested against the aphid Schizaphis graminum by incorporation in arti®cial diets. It showed a deterrent (toxic) activity against the insect and is the ®rst spirostane glycoside reported to have this activity.

Abstract
A summary of the resource.
It is estimated that 2.5 million cats enter animal shelters in the United States every year and as few as 20% leave the shelter alive. Of those adopted, the greatest risk to post-adoption human animal bond is unrealistic expectations set by the adopter. The ASPCA®’s Meet Your Match® Feline-ality™ adoption program was developed to provide adopters with an accurate assessment of an adult cat’s future behavior in the home. However, the original Feline-ality™ required a three-day hold time to collect cat behaviors on a data card, which was challenging for some shelters. This research involved creating a survey to determine in-home feline behavior post adoption and explored the predictive ability of the in-shelter assessment without the data card. Our results show that the original Feline-ality™ assessment and our modified version were predictive of feline behavior post adoption. Our modified version also decreased hold time for cats to one day. Shelters interested in increasing cat adoptions, decreasing length of stay and improving the adoption experience can now implement the modified version for future feline adoption success.

By means of density functional calculations, we studied the performance of 2 fullerene receptors that contain 3 corannulene or triphenylene pincers. For the first time, we found a receptor that has a small tendency to form intramolecular stacks between the 3 corannulene pincers. After comparing their ability to trap fullerenes, with that of buckycatcher, buckycatcher-II, and the Venus flytrap, we conclude that a record-breaking association constant is expected for the receptor with 3 corannulene pincers. Although triphenylene is less effective than the corannulene units in forming inclusion complexes with C60, the triphenylene-based host is also expected to have a high association constant. Finally, we discuss the theoretical problems found in reproducing the experimental free energy changes in solution for buckycatcher, buckycatcher-II, and the Venus flytrap.

A new method is described for the quantification of 2,4-dihydroxy-7-methoxy-1,4-benzoxazin-3-one (DIMBOA) and 2,4-dihydroxy-1,4-benzoxazin-3-one (DIBOA), the main hydroxamic acids in wheat and rye extracts, respectively, in cereal extracts based on densitometry of scanned thin layer chromatographic plates. The method allows the simultaneous quantification of up to five samples, and is linear between 0.5–7 μg and 10–30 μg for DIMBOA and between 0.5–3.0 μg and 10–30 μg for DIBOA. Quantification of DIMBOA by this method generates a linear correlation with results obtained following analysis by high performance liquid chromatography. The possibility of applying this methodology to mixtures of DIBOA and DIMBOA is discussed.

<p>Phage display library technology is a common method to produce human antibodies. In this technique, the immunoglobulin variable regions are displayed in a bacteriophage in a way that each filamentous virus displays the product of a single antibody gene on its surface. From the collection of different phages, it is possible to isolate the virus<br />that recognizes specific targets. The most common form in which to display antibody variable regions in the phage is the single chain variable fragment format (scFv), which requires assembly of the heavy and light immunoglobulin variable<br />regions in a single gene.<br />In this work, we describe a simple and efficient method for the assembly of immunoglobulin heavy and light chain variable regions in a scFv format. This procedure involves a two-step reaction: (1) DNA amplification to produce the single<br />strand form of the heavy or light chain gene required for the fusion; and (2) mixture of both single strand products followed by an assembly reaction to construct a complete scFv gene. Using this method, we produced 6-fold more scFv<br />encoding DNA than the commonly used splicing by overlap extension PCR (SOE -PCR) approach. The scFv gene produced by this method also proved to be efficient in generating a diverse scFv phage display library. From this scFv library, we<br />obtained phages that bound several non-related antigens, including recombinant proteins and rotavirus particles</p>

A rapid and efficient methodology to prepare 2,4'-bi-1,3-azoles as scaffolds for biologically active marine natural products is described. Hantzsch reaction and oxidative cyclodehydration of β-hydroxyamides or thioamides were used to construct the azole rings. The obtained biheterocycles displayed no citotoxicity on HCT-15 cell line.

<p>A new synthetic route for the quorum sensing signal Autoinducer-2 (AI-2) is described and used for the preparation of [4-13C]-AI-2 starting from [1-13C]-bromoacetic acid. The key step in this process was the enantioselective reduction of an intermediate ketone. This synthesis provides, selectively, both enantiomers of the labelled or unlabelled parent compound, (R) or (S)-4,5-dihydroxypentane-2,3-dione (DPD) and was used for an improved synthesis of [1-13C]-AI-2.</p>

Perceived freshness is a key determinant of consumers’ food choices but remains an imprecisely defined concept. A comprehensive and consumer-centric definition of freshness appears to be lacking, and the present research was situated in this knowledge gap, seeking, in particular, to explore the complexity of freshness in consumers’ minds. People from the USA (n = 2092) took part in an online survey and completed a text highlighting task. Herein, participants read a text that described different aspects of freshness and the technologies used to prolong freshness during storage. While reading, they used highlighting functions within the software to indicate text content that they liked/disliked or agreed/disagreed with. The combined results from text highlighting and responses to the open-ended question “Considering the fruit you eat, why is freshness important to you?” confirmed that freshness is a complex construct with multiple dimensions and suggested that the complexity of freshness extends from food in general to specific product categories (in the case of fruit and apples). Further, the findings showed that freshness is desired by consumers because fruit are perceived as healthier and better tasting. The findings revealed negative attitudes towards stored fruit among participants, but also indicated some acceptance of the fact that some storage was unavoidable. Results provide useful insights to the development for communication strategies to increase consumer acceptance of stored apples and fruits in general.

The coloration of Citrus fruits is related with the concentration of carotenoids, isoprenoid pigments of 40 carbon atoms (C40). Rodrigo et al. and Ma et al. reported a CCD4-type citrus dioxygenase responsible for the generation of C30 apocarotenoids providing a reddish-orange pigmentation to the peel of many mandarins and oranges. Among them, CCD4b was the first case described of a dioxygenase that cleaves carotenoids C40 in the double bond 7', 8' or 7, 8, generating β-citraurin or 8-β-apocarotenal. Here we report the three-dimensional structures of CCD4a and CCD4b, modeled by sequence homology (2BIW) and validated by molecular dynamics (MD). Docking calculations were performed in CCD4a and CCD4b structures with thousands of rotated initial carotenoid conformations and all the possible poses in the active site were found. The interaction energy was measured by means of ASE scoring, Amber99 refinement and London ΔG rescoring. For the case of CCD4a model, the results showed London ΔG score of -19, -17 and -15 kcal/mol for zeaxanthin, β-cryptoxanthin and β-carotene, respectively. The same sequence in the estimated interaction strength for the three ligands was obtained using MD. The interaction energy of CCD4b indicated that, in agreement with experimental data, zeaxanthin and β-cryptoxanthin could be cleaved by the enzyme, β- and α-carotene have chances to be oxidized and lycopene has not good interaction energy to be predicted as substrate. These findings will be discussed considering the potential in vivo substrates and products, and the physiological role in Citrus fruits. Communicated by Ramaswamy H. Sarma.

A summary of the resource.
The aim of this work was to evaluate the potential of lufenuron, a benzylphenylurea with ability to interfere with the formation of insect exoskeleton, as a therapeutic drug for larval echinococcosis (hydatid disease). For this purpose lufenuron, alone or in combination with albendazole, was administered to CD1 mice bearing Echinococcus granulosus hydatid cysts in the peritoneal cavity. Neither of the drugs alone was able to exert parasiticidal effects. However, in combination with albendazole, lufenuron reduced the growth of cysts (30–40% in cyst diameter respect to control, p < 0.05). This effect was associated with ultrastructural alterations of the hydatid cyst wall and a reduction of the content of myo-inositol-hexakisphosphate, the major component of the electron dense granules of the laminated layer. Overall, this work provides evidence that lufenuron could represent a useful compound for the use in chemotherapy against larval echinococcosis, by enhancing albendazole parasiticidal activity.

Actinomycetes are generally recognized as a diverse group of gram-positive, myceliumforming, soil bacteria that play an important role in mineralization processes and plant health, being Streptomyces the most well-known genus from this group. Although plant pathogenicity is a rare attribute in this genus, some species have significant impact worldwide due to their ability to cause important crop diseases such as potato common scab (CS). In this work, an integrative approach was applied to investigate the pathogenic potential of Streptomyces spp. isolates obtained from a local collection of actinomycetes isolated from potato fields. Secretion of phytotoxic compounds was verified in most pathogenic strains from our collection (27 out of 29), and we followed metabolomic analysis to investigate those phytotoxins. We first evaluated the
production of the known phytotoxins thaxtomin A (TXT) and desmethylmensacarcin (DMSN) in phytotoxic Streptomyces spp. by HPLC analysis, resulting in 17 TXT and 6 DMSN producers. In addition, NMR-based metabolomic models were able to classify strains according to their phytotoxicity, and metabolomic data was also used to infer chemotaxonomy within pathogenic species. A correlation between phylogeny and the production of distinct phytotoxins was found, supporting the idea that there are “species
specific” metabolites produced by this genus. The recently discovered polyketide DMSN was associated unequivocally with S. niveiscabiei strains and was not produced by other species in the growth conditions employed. Two S. niveiscabiei and two S. puniciscabiei
phytotoxic strains that did not produce TXT nor DMSN suggest the production of other kind of metabolites involved in phytotoxicity, and allowed the prioritization of these strains for further chemical studies. Indeed, we found two S. niveiscabiei strains whose supernatants were not phytotoxic in the radish assay, suggesting other pathogenic mechanisms involved. We believe our work will be useful to help understand relations between metabolites and phylogenetic clades within actinomycetes.

he non-persistent BK strain was examined for its ability to induce sterile immunity in Wistar rats. Groups of 2–9 Wistar rats were inoculated subcutaneously with 5 × 104 BK strain tachyzoites per rat. Two months later, 46 rats were dosed by gavage with 2 × 101 cysts of the C, ME-49, Prugniaud, C-56, Elg, M-7741 or M3 strains. Another 26 rats were inoculated with 103 oocysts of the ME49, M7741, Bear or Hopa–Hopa strains of Toxoplasma gondii. After 2 months, the rats were euthanized and their brains screened microscopically for toxoplasma tissue cysts and bioassayed in mice if negative. As judged by bioassay, the BK strain of Toxoplasma induced statistically significant protection against reinfection only when rats were challenged with cysts of the C and Prugniaud strains or with oocysts of the ME49 strain. Nonetheless, cysts were detected microscopically only in 23% of brains of immunized rats challenged with oocysts of the Bear and Hopa–Hopa strains of Toxoplasma and none of those challenged with tissue cysts of any strain. Tissue cysts were detected in 43 and 48% of non-immunized control rats infected with tissue cysts and oocysts, respectively. The overall infection in control rats (microscopy and bioassay) was 70 and 72% for rats inoculated with cysts and oocysts, respectively. These results are consistent with the divergent results obtained by other investigators with regard to protection after challenge with different complete strains (cyst and oocysts forming) of the parasite, of rats immunized with incomplete strains.

The activity of human transglutaminase 2 (TG2), which forms protein cross-links between glutamine and lysine residues, is controlled by an allosteric disulfide bond. However, the mechanism by which this bond is formed, like many systems regulated by oxidative cysteine modifications, was not clear. A new study from Khosla and colleagues shows that TG2 is oxidatively inactivated by the protein disulfide isomerase ERp57, providing the first example of a defined and reversible protein-controlled redox switch and pointing to new strategies to inhibit undesirable TG2 activity in pathological states.

Cold tolerance at germination and seedling stage is one of the most seeked traits in Southern Brazilian rice (Oryza sativa L.) cultivars, thus, the availability of improved methods for germplasm assessment is of high importance for breeding programs. The aim of this study was to validate an optimized Average Tolerance Index (ATI) for rice germplasm assessment under cold at germination and early seedling stage. A diverse panel composed by 124 rice accessions was assayed. Four genotypes with known performance under cold were used as controls (two tolerant and two sensitive). The genotypes were sown in roll papers, and evaluated at 13 °C for 28 days (cold stress) and 25 °C for seven days (control). Four traits were measured, germination percentage, coleoptile length, root length and shoot length, which were converted in terms of relative perfor‑ mance (cold/control). A PCA analysis was carried out. The ATI was calculated as a linear combination of all trait’s relative performance, weighted by the importance of each trait in explaining the genetic variability in the set, through including the PC1 vectors as trait coefcients. K means was applied for genotype classifcation. The panel showed expressive genetic variability for performance under low temperature. The ATI successfully distinguished the controls regarding their cold tolerance and allowed the formation of four classes of genotypes, sensitive, moderately sensitive, moderately tolerant and tolerant. The assessment of rice germplasm for cold tolerance at germination and seedling stage through ATI is a viable alternative for the identifcation of tolerant genotypes

Evidence for low-dose health effects of Arsenic (As) in humans is still controversial and presents a major public health issue in several countries worldwide. It is not clear yet, whether there is a lower safe threshold for arsenic in drinking water among other possible sources such as food, below which, exposures are not harmful. In Uruguay, safe drinking water is supplied to 94% of the population by a state company (OSE) and As levels in workplaces and food are officially regulated. This paper aims to present and discuss the issues regarding arsenic exposure risks to the environment and human population, which are being addressed in a multidisciplinary manner in Uruguay since 2007. An overview is given on both the background and the current situation, presenting reports and research studies conducted on these problems by various academic, state, and private institutions that deal with regulations, surveillance, and health care. Scientific research on geogenic As levels in groundwater indicates As levels above those recommended by the WHO for drinking water (10 μg L-1) in different Uruguayan aquifers. There is a lack of baseline studies concerning Uruguayan residents that are exposed to As in drinking water over time. Furthermore, there is a need for data on environmental chemical exposure that could be associated with disease or death in the country. In addition, only a few As risk exposure assessment studies in children, adults, and workers using biomarkers in urine are available. Furthermore, this paper presents As levels in a rice growing region and the spatial distribution of groundwater arsenic data compared to a national cancer atlas database as ongoing research advances. Multidisciplinary research projects and local future actions are also described. This contribution constitutes a first attempt to develop a feasible health risk assessment of low-dose arsenic exposure in this Latin-American country

The design and the execution of a multi-generation approach to pyrrolizidine alkaloids is summarized. A historical perspective and the various solutions to the problems of regio-, stereo-, and enantiocontrol in the preparation of these ubiquitous natural products are provided. The details of the fourth generation approach based on the resolution of ß-oxo esters of prochiral alcohols by yeast are disclosed.
