Abstract
A summary of the resource.
Objetivo. Identificar los argumentos de la industria alimentaria en contra del etiquetado frontal de advertencias nutricionales en Uruguay. Métodos. Se realizó un análisis de contenido de los comentarios de la industria alimentaria recibidos durante la consulta pública implementada durante el proceso de diseño del decreto que reglamenta la inclusión de rotulación nutricional frontal basada en advertencias en Uruguay mediante una codificación inductiva. Resultados. La mayoría de los comentarios manifestaron la preocupación por la elevada prevalencia de obesidad y enfermedades no transmisibles en el país y destacaron el compromiso con la implementación de acciones que contribuyan a combatir esta problemática de salud. Sin embargo, 81% de los comentarios planteó desacuerdo con la propuesta de decreto elevada a consulta pública y realizó críticas generales sobre su validez. El contenido de los comentarios se relacionó con siete dimensiones: falta de justificación de la medida, violación de compromisos internacionales, críticas al sistema de advertencias, discriminación a alimentos industrializados envasados, críticas al modelo de perfil de nutrientes, otras críticas a aspectos específicos del proyecto de decreto y efectos económicos negativos. En cada dimensión se discute la pertinencia y consistencia técnica y normativa de los contenidos vertidos por los representantes de la industria. Conclusiones. Bajo un aparente acuerdo con la necesidad de adoptar medidas, se evidenció, sin embargo, una fuerte oposición a la política y en particular al sistema de advertencias por parte de la industria alimentaria. Los resultados del análisis aportan insumos para facilitar el proceso de implementación de regulaciones similares en otros países.

Background: Aptamers represent an emerging class of oligonucleotides that have the ability to bind ligands with high affinity. Sgc8-c aptamer recognizes PTK7, a member of the catalytically defective receptor protein tyrosine kinase family that is upregulated in various cancers, including hemato-oncological malignancies. Herein, an Sgc8-c-NOTA-radiolabeled probe was prepared for theranostic purpose. Materials and Methods: In this work, an Sgc8-c-radiolabeled probe against PTK7 was prepared, and biological evaluations—pharmacokinetic studies, biodistribution analysis, and in vivo molecular imaging—were performed. To obtain the radiolabeled probe, a modified 5′-amino-derivative of the Sgc8-c aptamer was bound to the metal chelator NOTA, and subsequently labeled with 67Ga with high yield and radiochemical purity. The precursor, Sgc8-c-NOTA, the radio probe Sgc8-c-NOTA-67Ga, and its nonradioactive complex, Sgc8-c-NOTA-69/71Ga, were purified by reverse-phase high-performance liquid chromatography and characterized by electrospray ionization mass spectrometry. The binding ability of Sgc8-c-NOTA-67Ga was studied in vitro against purified PTK7 receptor. In addition, the binding was also evidenced against the hemato-oncological A20 cell line, derived from B lymphocytes, and the corresponding A20-green fluorescent protein (GFP)-transfected cells. The proof of concept was performed on A20-GFP tumor-bearing mice, in which the biodistribution of the radiolabeled probe was evaluated through imaging, using X-ray, fluorescence, and γ modalities. The specific uptake of the probe was confirmed by blocking with the Sgc8-c aptamer in an in vivo competition assay. Results: The biodistribution results showed considerable uptake in tumor since 2 h, with highest at 48 h postinjection. However, the blood and muscle ID/g (injected dose per gram of tissue) activities were decreasing with time and tumor/no-target ratios increasing to 20 at 24 h postinjection. These results are consistent with the in vivo images. Conclusions: This study supports the utility of Sgc8-c-NOTA radiolabeled as a theranostic agent.

Human milk (HM) constitutes the first immunological barrier and the main source of nutrients and bioactive components for newborns. Immune factors comprise up to 10% of the protein content in HM, where antibodies are the major components (mainly IgA, IgG, and IgM). In addition, antibacterial enzymes such as lysozyme and immunoregulatory factors such as soluble cluster of differentiation 14 (sCD14) and transforming growth factor β2 (TGF-β2) are also present and play important roles in the protection of the infant's health. Donor milk processed in HM banks by Holder pasteurization (HoP; 62.5°C, 30 min) is a safe and valuable resource for preterm newborns that are hospitalized, but is reduced in major immunological components due to thermal inactivation. We hypothesized that high hydrostatic pressure (HHP) and high-pressure homogenization (HPH) are 2 processes that can be used on HM to reduce total bacteria counts while retaining immunological components. We studied the effects of HHP (400, 450, and 500 MPa for 5 min applied at 20°C) and HPH (200, 250, and 300 MPa, milk inlet temperature of 20°C) applied to mature HM, on microbiological and immunological markers (IgA, IgG, IgM, sCD14, and TGF-β2), and compared them with those of traditional HoP in HM samples from healthy donors. The HHP processing between 400 and 500 MPa at 20°C reduced counts of coliform and total aerobic bacteria to undetectable levels (<1.0 log cfu/mL) while achieving approximately 100% of immunological component retention. In particular, comparing median percentages of retention of immunological components for 450 MPa versus HoP, we found 101.5 versus 50.5% for IgA, 89.5 versus 26.0% for IgM, 104.5 versus 75.5% for IgG, 125.0 versus 72.5% for lysozyme, 50.6 versus 0.1% for sCD14, and 88.5 versus 61.1% for TGF-β2, respectively. Regarding HPH processing, at a pressure of 250 MPa and inlet temperature of 20°C, the process showed good potential to reduce coliforms to undetectable levels and total aerobic bacteria to levels slightly above those obtained by HoP. The median percentages of retention of immunological markers for HPH versus HoP were 71.5 versus 52.0%, 71.0 versus 27.0%, 104.0 versus 66.5%, and 30.9 versus 0.2%, for IgA, IgM, IgG, and sCD14, respectively; results did not significantly differ for lysozyme and TGF-β2. The HPH at 300 MPa produced higher inactivation of immunological components, similar to values achieved with HoP.

MicroRNAs (miRNAs) regulate the levels of translation of messenger RNAs (mRNAs). At present, the major parameter that can explain the selection of the target mRNA and the efficiency of translation repression is the base pairing between the ‘seed’ region of the miRNA and its counterpart mRNA1. Here we use R1ρ relaxation-dispersion nuclear magnetic resonance2 and molecular simulations3 to reveal a dynamic switch—based on the rearrangement of a single base pair in the miRNA–mRNA duplex—that elongates a weak five-base-pair seed to a complete seven-base-pair seed. This switch also causes coaxial stacking of the seed and supplementary helix fitting into human Argonaute 2 protein (Ago2), reminiscent of an active state in prokaryotic Ago4,5. Stabilizing this transient state leads to enhanced repression of the target mRNA in cells, revealing the importance of this miRNA–mRNA structure. Our observations tie together previous findings regarding the stepwise miRNA targeting process from an initial ‘screening’ state to an ‘active’ state, and unveil the role of the RNA duplex beyond the seed in Ago2.

Flavin cofactors, like flavin adenine dinucleotide (FAD), are important electron shuttles in living systems. They catalyze a wide range of one- or two-electron redox reactions. Experimental investigations include UV-vis as well as infrared spectroscopy. FAD in aqueous solution exhibits a significantly shorter excited state lifetime than its analog, the flavin mononucleotide. This finding is explained by the presence of a "stacked" FAD conformation, in which isoalloxazine and adenine moieties form a π-complex. Stacking of the isoalloxazine and adenine rings should have an influence on the frequency of the vibrational modes. Density functional theory (DFT) studies of the closed form of FAD in microsolvation (explicit water) were used to reproduce the experimental infrared spectra, substantiating the prevalence of the stacked geometry of FAD in aqueous surroundings. It could be shown that the existence of the closed structure in FAD can be narrowed down to the presence of only a single water molecule between the third hydroxyl group (of the ribityl chain) and the N7 in the adenine ring of FAD.

Nutritional warnings are intended to enable citizens to make informed choice by clearly identifying food products with excessive content of nutrients associated with noncommunicable diseases. The efficacy of this public policy is expected to improve if accompanied by communication campaigns that raise awareness of the existence of nutritional warnings, as well as encourage citizens to take them into account in decision making. Because ordinary citizens have been shown to generate significantly more creative and valuable ideas than advanced users and professional developers, the aim of the present work was to obtain qualitative, citizen co-created insights for the design of a communication campaign. An online study was conducted with 518 Uruguayan citizens, recruited using a Facebook advertisement. Participants were asked to answer a series of open-ended questions about how they would encourage other people to use the warnings for making their food choices, as well as the key contents of a communication campaign. Responses were analyzed using content analysis. Results showed that, according to the participants’ accounts, an effective public awareness campaign aimed at promoting the use of nutritional warnings in decision making should include three main concepts: (a) position warnings as a cue to action for improving eating habits by enabling informed choices; (b) emphasize the benefits of using the warnings for avoiding consumption of unhealthy food and, consequently, achieving a healthier diet and an improvement in health status and quality of life; and (c) increase the perceived susceptibility and severity of the negative consequences of consumption of foods with excessive content of sugar, fat, and sodium. A communication campaign based on these key concepts could contribute to increasing the efficacy of nutritional warnings

Elucidation of the oxidation mechanism of naturally emitted reduced sulfur compounds, especially dimethyl sulfide, plays a central role in understanding background acid precipitation in the natural environment. Most frequently, theoretical studies of the addition and H-elimination reactions of dimethyl sulfide with hydroxyl radicals are studied considering the presence of oxygen that further reacts with the radicals formed in the initial steps. Although the reaction of intermediate species with additional hydroxyl radicals has been considered as part of the global mechanism of oxidation, little if any attention has been dedicated to the possibility of reactions of the initial radicals with a second •OH molecule. In this work, we performed a computational study using quantum-chemical methods, of the mechanism of H-abstraction from dimethyl sulfide under normal atmospheric conditions and in reaction chambers at different O2 partial pressure, including complete absence of oxygen. Additionally, important rate coefficients were computed using canonical and variational transition state theory. The rate coefficient for abstraction affords a 4.72 × 10–12 cm3 molecule–1 s–1 value, very close to the most recent experimental one (4.13 × 10–12 cm3 molecule–1 s–1). According to our best results, the initial methyl thiomethyl radical was obtained at −25.2 kcal/mol (experimentally −22.4 kcal/mol), and four important paths were identified on the potential energy surface. From the interplay of thermochemical and kinetic arguments, it was possible to demonstrate that the preferred product of the reaction of dimethyl sulfide with two hydroxyl radicals is actually dimethyl sulfoxide.

Electronic structure calculations have been performed to determine the thermochemistry and kinetics of the reaction between OH and the radicals of the S enantiomer of the herbicide Metolachlor, 2-chloro-N-(2-methyl-6-ethylphenyl)-N(2-methoxy-1-methylethyl) acetamide (MC), produced by photoinduced breaking of the C–Cl bond. Both density functional and ab initio composite methods were employed to calculate the structure of reactants, intermediates, transition states and products, in gas phase and in aqueous solution. The expected relative abundance of each product was calculated and compared to the experimentally observed concentrations. It is shown that a combination of thermodynamic and kinetic characteristics interplay to produce the expected theoretical abundances, which turn out to be in agreement with the experimentally observed distribution of products.

A simple version of a composite scheme is described, based on a combination of density functional geometry and frequencies evaluation, valence energies obtained using the CCSD(T)-f12 method extrapolated to the complete basis set limit, and core-valence correlation corrections employing the MP2 method. The procedure was applied to the 38 reactions in Truhlar’s HTBH38/08 and NHTBH38/08 databases. Mean unsigned deviation (MUD) for the complete set of 68 independent barriers is 0.43 kcal mol-1, compared to 1.37 kcal/mol for G4 and 1.69 kcal/mol for the dispersioncorrected M06-2X method. Its accuracy is also better that that of other calculations using composite methods of similar cost. The MUD of the new scheme on the barriers in the DBH24/08 subset (12 out of the 38 reactions in both other sets) is 0.31 kcal mol-1, better than that obtained at the expensive CCSD(T,full)/aug-cc-pCV(T+d)Z level (0.46 kcal mol-1) and comparable to the most exact (and costly) Wn calculations (MUD=0.14 kcal mol-1). The maximum unsigned deviation (MaxUD) of the new method for all the reactions studied is 1.71 kcal/mol. G4 and M06-2X, on the other side, exhibit MaxUDs of 6.7 and 8.4 kcal/mol respectively

A primary cause of food waste in households is a lack of food capabilities. Yet, the antecedents of food capabilities and its relationship with causes of food waste across different situational contexts remain poorly understood. We apply self‐efficacy theory in combination with convenience orientation and good provider identity in a mixed‐methods study. Findings reveal that the sequential structure of self‐efficacy explains how capabilities contribute to food waste avoidance. A good provider identity, however, increases food waste, and also turns out to further convenience orientation. Food capabilities, in turn, reduce convenience orientation. Qualitative insights shed light on consumer's own accounts of their abilities. The findings contribute to a theory‐based understanding of how consumers manage the complex issue of food waste. Findings imply that public policy and marketing should aim to strengthen consumers' self‐efficacy beliefs and redefine the good provider identity, in particular in social contexts, to reverse its effect on waste.

Abstract
A summary of the resource.
The aim of the present work was to explore the effect of the inclusion of nutritional warnings on consumer associations with labels of ultra-processed products. The study was conducted in Uruguay at two points in time, before and after the compulsory implementation of nutritional warnings. A total of 163 participants evaluated three labels of ultra-processed products using a word association task. In the first evaluation (T1), performed 9 months before the entry into force of nutritional warnings, labels were presented without warnings. Nine months later (15 days after the entry into force of the policy), participants evaluated the labels with warnings again (T2). Data were analyzed using content analysis. At T1, when labels were presented without nutritional warnings, responses to the word association task were related to a description of the product category, expected sensory characteristics of the products, brand names, liking and eating occasions. However, when the labels were presented with the warnings at T2, a significant increase in the frequency of mention of responses was observed within the categories Excessive content of sugar/fat/sodium, Not healthy, Diseases and negative health conditions, Rejection to consume, and Unexpected information. Results from the present work suggest that immediately after their implementation, nutritional warnings cause a salience bias that make excessive nutrient content and its negative health consequences more salient in consumers’ minds, especially in the case of products with a particular health-related connotation.

Oxyfluoride glass and glass‐ceramics are being studied due to the possibility of their utilization in many applications, such as thermoluminescent dosimeters, optical fibers, fluorescent concentrators, and temperature sensors. In order to improve the properties of glass, they are often crystallized. For this crystallization to be effective, knowledge and control of the structure of the starting material is required. Much work was done on silicate glass; however, a great interest exists in obtaining these kinds of materials in other matrices such as germanates, phosphates, and borates. Here, we present a structural study of some oxyfluoride borate glass with different concentrations of PbF2 by means of Nuclear Magnetic Resonance (NMR), Raman, and Fourier Transform Infrared (FTIR) spectroscopies. We also analyzed glass‐crystallization using Differential Scanning Calorimetry (DSC). The crystallization study was complemented with the use of Transmission Electron Microscopy (TEM) and Selected Area Electron Diffraction (SAED), for the purpose of identifying possible mechanisms of crystallization. Our results indicate, from the structural point of view, that the glass present groups with one or two BO4 species, such as diborate or pentaborate. The small variation in the BO4 fraction with the increase in PbF2 concentration would be an indication of the formation of F−BO3/2 species. Oxyfluoride borate glass containing BaO as a cation modifier and 30% PbF2 represents a good starting point for obtaining glass‐ceramics with a narrow size distribution of BaF2 nanocrystals. We also propose that the possible mechanism for oxyfluoride borate glass crystallization started with a separation of phases.

The antimicrobial properties of silver nanoparticles (AgNPs) have made them ubiquitous in a number of real-world industrial applications; however, the antimicrobial mode of action of biogenic AgNPs is not entirely understood. The use of Raman spectroscopy can provide molecular fingerprint information on various chemical and biochemical components in complex systems like microbial cultures, without the need for any complex sample pre-treatment. Consequently, the antimicrobial mechanism of AgNPs can be inferred through morphological and compositional changes of microbial cells that are monitored via changes in Raman band profiles. Here we show the synthesis of biogenic AgNPs using the extracellular cell-free filtrates of Penicillium expansum. The antimicrobial activity of the Penicillium expansum synthesized silver nanoparticles (hereafter PeNPs) was evaluated and the interactions between the nanoparticles and Escherichia coli were studied using Transmission Electron Microscopy (TEM) and Environmental Scanning Electron Microscopy (ESEM), showing the attachment of PeNPs to the surface of the bacteria and rupture of the bacterial cell membrane. Importantly, we show how Confocal Raman Microscopy can be used as an innovative approach to study the antimicrobial mechanisms, the results of which confirm that the PeNPs induce damage to bacterial and fungal cells, resulting in critical changes to polysaccharides, lipids, proteins and nucleic acids.

Anecdotal reports and open-label case studies in humans indicated that the psychedelic alkaloid ibogaine exerts profound antiaddictive effects. Ample preclinical evidence demonstrated the efficacy of ibogaine, and its main metabolite, noribogaine, in substance-use-disorder rodent models. In contrast to addiction research, depression-relevant effects of ibogaine or noribogaine in rodents have not been previously examined. We have recently reported that the acute ibogaine administration induced a long-term increase of brain-derived neurotrophic factor mRNA levels in the rat prefrontal cortex, which led us to hypothesize that ibogaine may elicit antidepressant-like effects in rats. Accordingly, we characterized behavioral effects (dose- and time-dependence) induced by the acute ibogaine and noribogaine administration in rats using the forced swim test (FST, 20 and 40 mg/kg i.p., single injection for each dose). We also examined the correlation between plasma and brain concentrations of ibogaine and noribogaine and the elicited behavioral response. We found that ibogaine and noribogaine induced a dose- and time-dependent antidepressant-like effect without significant changes of animal locomotor activity. Noribogaine's FST effect was short-lived (30 min) and correlated with high brain concentrations (estimated >8 μM of free drug), while the ibogaine's antidepressant-like effect was significant at 3 h. At this time point, both ibogaine and noribogaine were present in rat brain at concentrations that cannot produce the same behavioral outcome on their own (ibogaine ∼0.5 μM, noribogaine ∼2.5 μM). Our data suggests a polypharmacological mechanism underpinning the antidepressant-like effects of ibogaine and noribogaine.

Bismuth sulpho iodide (BiSI) belongs to the family of chalcohalides, which present several attractive electro-optic properties. In particular, BiSI is a semiconductor which could be used in X and gamma ray detection due to a band gap of 1.6 eV, density of 6.4 g cm−3, and absorption coefficient for 60 keV radiation of 5.6 cm2 g−1. This work presents a facile synthesis under solvothermal conditions of a nanocomposite consisting of BiSI nanorods and amorphous carbon structures. Furthermore, it studies its ionising radiation detection properties at room temperature, when prototype detectors were built from pellets. The construction conditions of pellets were also studied, varying the applied pressure and heat treatment to the nanocomposite. Dark current density and response to different exposure rates of a 241Am source were measured for the prototype detectors built. It was found that heat treatment of pellets considerably improves detectors performance. Dark current density was one order of magnitude lower than for the pellets without heat treatment, and its response to the 241Am source, linear, with a signal to noise ratio of 7 for 20 V. Finally, the resistivity for the heat treated detector was in the order of 1011 Ω cm, comparable to other materials studied for this application.

Abstract

There is an urgent need to develop new drugs against Chagas’ disease. In addition, the mechanisms of action of existing drugs have not been completely worked out at the molecular level. High throughput approaches have been demonstrated to be powerful tools not only for understanding the basic biology of Trypanosoma cruzi, but also for the identification of drug targets such as proteins or pathways that are essential for parasite infection and survival within the mammalian host. Here, we have applied these tools towards the discovery of the effects of two organometallic compounds with trypanocidal activity, Pd–dppf–mpo and Pt–dppf–mpo, on the transcriptome and proteome of T. cruzi epimastigotes. These approaches have not yet been reported for any other prospective metal-based anti T. cruzi drug. We found differentially expressed transcripts and proteins in treated parasites. Pd–dppf–mpo treatment resulted in more modulated transcripts (2327 of 10 785 identified transcripts) than Pt–dppf–mpo treatment (201 of 10 773 identified transcripts) suggesting a mechanism of action for Pd–dppf–mpo at the transcriptome level. Similar numbers of differentially expressed proteins (342 and 411 for Pd–dppf–mpo and Pt–dppf–mpo respectively) were also observed. We further functionally categorized differentially expressed transcripts and identified cellular processes and pathways significantly impacted by treatment with the compounds. Transcripts involved in DNA binding, protein metabolism, transmembrane transport, oxidative defense, and the ergosterol pathways were found to be modulated by the presence of the compounds. Our transcriptomic dataset also contained previously validated essential genes. These data allowed us to hypothesize a multimodal mechanism of action for the trypanocidal activity of Pd–dppf–mpo and Pt–dppf–mpo, and a differential contribution of the metal moiety of each compound.

Background The protective effect of breastfeeding on celiac disease (CD) onset is controversial. We studied a wide range of milk components in milk produced by celiac mothers following long-term gluten-free diet (GFD) in comparison to milk produced by healthy mothers. Methods Breast-milk samples from celiac (n = 33) and healthy (n = 41) mothers were obtained during the first year of lactation. A panel of bioactive components was analyzed by enzyme-linked immunosorbent assay in the aqueous fraction. We studied molecules involved in defenses, immunoregulation, and strengthening of the gut–epithelial barrier. Results During late lactation (from 6 to 12 months after delivery), the content of total immunoglobulin A (IgA) and IgM was significantly lower in the milk produced by celiac patients. Nevertheless, gliadin (GFD)-specific IgA relative contribution was higher in this group, in contrast to tetanus toxoid-specific antibodies. The balance between pro-inflammatory and anti-inflammatory molecules was different. While interleukin-6, tumor necrosis factor-α, and monocyte chemoattractant protein-1 were most frequently found in samples from celiac mothers, soluble Toll-like receptor-2 prevalence was lower. Conclusions We describe differences between the innate and adaptive immune profile of milk produced by celiac and healthy mothers. These results might explain previous controversial reports about breastfeeding and CD protection.

Dimethyl sulfide (DMS) is the main organosulfur compound in the atmosphere. Oxidation by OH radicals to form the methyl thiomethyl species (MTMr) has been studied under normal atmospheric conditions and in reaction chambers at different O2 partial pressure, including complete absence of oxygen. Scarce attention has been devoted however to the possibility of further reaction of OH with MTMr. We present here a computational study using DFT, CCSD(T) and composite methods, on the properties of two stable intermediates never fully investigated before, methanesulfenyl methanol (MSMOH) and S-methyl-methanesulfenic acid (SMMSA), arising from addition of a second OH radical to MTMr.

Selenium is a trace element for most organisms; its deficiency and excess are detrimental. Selenium beneficial effects are mainly due to the role of the 21st genetically encoded amino acid selenocysteine (Sec). Selenium also exerts Sec-independent beneficial effects. Its harmful effects are thought to be mainly due to non-specific incorporation in protein synthesis. Yet the selenium response in animals is poorly understood. In Caenorhabditis elegans, Sec is genetically incorporated into a single selenoprotein. Similar to mammals, a 20-fold excess of the optimal selenium requirement is harmful. Sodium selenite (Na2SeO3) excess causes development retardation, impaired growth, and neurodegeneration of motor neurons. To study the organismal response to selenium we performed a genetic screen for C. elegans mutants that are resistant to selenite. We isolated non-sense and missense egl-9/EGLN mutants that confer robust resistance to selenium. In contrast, hif-1/HIF null mutant was highly sensitive to selenium, establishing a role for this transcription factor in the selenium response. We showed that EGL-9 regulates HIF-1 activity through VHL-1, and identified CYSL-1 as a key sensor that transduces the selenium signal. Finally, we showed that the key enzymes involved in sulfide and sulfite stress (sulfide quinone oxidoreductase and sulfite oxidase) are not required for selenium resistance. In contrast, knockout strains in the persulfide dioxygenase ETHE-1 and the sulfurtransferase MPST-7 affect the organismal response to selenium. In sum, our results identified a transcriptional pathway as well as enzymes possibly involved in the organismal selenium response.

Treatment for Chagas disease, a parasitosis caused by Trypanosoma cruzi, has always been based on two drugs, nifurtimox and benznidazole, despite the toxic side effects described after prolonged prescription. In this work, we study a new prospective antitrypanosomal drug based on vanadium, here named VIVO(5Brsal)(aminophen). We found a good IC50 value, (3.76 ± 0.08) μM, on CL Brener epimastigotes. The analysis of cell death mechanism allowed us to rule out the implication of a mechanism based on early apoptosis or necrosis. Recovery assays revealed a trypanostatic effect, accompanied by cell shape and motility alterations. An uptake mostly associated with the insoluble fraction of the parasites was deduced through vanadium determinations. Concordantly, no drastic changes of the parasite transcriptome were detected after 6 h of treatment. Instead, proteomic analysis uncovered the modulation of proteins involved in different processes such as energy and redox metabolism, transport systems, detoxifying pathways, ribosomal protein synthesis, and proteasome protein degradation. Overall, the results here presented lead us to propose that VIVO(5Brsal)(aminophen) exerts a trypanostatic effect on T. cruzi affecting parasite insoluble proteins.

The QuEChERS methods, CEN 15662 and AOAC 2007.01, and different clean-up variations were applied to the determination of pesticide residues in Peumus boldus Molina leaves, using LC and GC coupled to single and tandem mass spectrometry. Boldo belongs to the class of medicinal and aromatic plants. Because of its complex chemical composition including terpene peroxides, alkaloids and phenolic acids at concentrations typically 100-fold higher than that of a possible contaminant, it is a very challenging matrix for pesticide residue determination. Thin layer chromatography was used to evaluate the efficacy of removal of matrix co-extractives using different clean-up sorbents, including neutral alumina, graphitized carbon black (GCB), octadecyl reverse phase silica gel (RP-C18) and pH modification in the extraction and clean-up steps. The most promising method was a combination of GCB, RP-C18 and primary-secondary amine (PSA) within the QuEChERS template. The final optimized method was validated according to the SANTE guidelines using LC–MS/MS and GC–MS/MS. The method scope includes 84 representative pesticide residues with recoveries ranging from 70 to 119% and within laboratory relative standard deviations below 15%. The method was initially validated for 25 representative pesticides using GC-MSD, making it applicable in those laboratories equipped only with a single mass detector. It is important to emphasize that the method complies with the requirements of international pharmacopoeias and food regulatory agencies, since herbal teas are found in both categories, as herbal remedies and foods.

The aim of this study was to concentrate whey protein by ultrafiltration process, evaluating the pressure at 1–3 bar and temperature of 10–20℃. In the conditions that show the more protein concentration were evaluated the interfacial and emulsifying properties at pH 5.7 and 7.0. The whey concentrate at 10℃ and 1.5 bar showed the higher protein value 36% (w/w), with soluble protein of 33.82% (solubility of 93.94%) for pH 5.7 and 34% (solubility of 94.4%) for pH 7.0, respectively. The whey concentrate powder present particle size distribution between 0.4-110 um. The whey at pH 5.7 and 7.0 was not observed significant differences in the resistance parameters of the oil/water layer interface. The interfacial film formed by the proteins presented an essentially elastic behavior in both pH, and in pH 5.7 the emulsion was more stable with lower diameter droplets. The concentrate whey showed techno-functional properties (emulsification and solubility), which allow the use as ingredients in products of industrial interest in food products such as mayonnaise, ice cream, sauces, and others.

A summary of the resource.
Pachyrhizus ahipa is an Andean leguminous plant which has purple-pigmented edible roots. The content and distribution of the phenolic compounds in the skins and flesh of P. ahipa roots as well as their antioxidant capacity were studied. Total phenols, flavonoids, tannins and anthocyanins were quantified and the effect of the extraction solvent on the maximum absorbance wavelength (λvis-max) was measured. The ATR-FTIR spectrum and stability of the purified anthocyanin extracts were analyzed. Individual anthocyanins were partially characterized using high-performance liquid chromatography-mass spectrometry and absorbance spectra. Ahipa skins had much more phenols (3.25 mg gallic acid equivalents/g), flavonoids (3.0 mg catechin equivalents/g) and tannins (4.6 mg tannic acid equivalents/g) than the flesh, which was reflected in a higher antioxidant capacity. Changing the extraction solvent from acidified methanol to acidified water shifted the λvis-max 16 nm lower, suggesting copigmentation reactions. Anthocyanin in the skins was >8.7 content in the flesh, and the chromatographic analysis showed at least 6 different anthocyanins with identical retention times for skins and flesh, including delphinidin, pelargonidin and cyanidin as aglycons. The absorbance spectra showed that two of them are acylated structures. Pigments were stable for at least 180 min at pH 1 and 25 °C, but they were affected by increased pH and temperature. These anthocyanins may be a potential source of natural pigments.
