El cociente de concentraciones tejido/plasma de un fármaco depende de la fracción del gasto cardíaco que irriga cada órgano del cuerpo, como asimismo de la fracción libre de sustancia en el plasma sanguíneo. La concentración salival de droga cambia en el transcurso de una continua salivación. Al principio el valor de concentración representa el nivel de droga libre tisular, pero consecutivamente se transforma en el nivel plasmático libre del fármaco. En 7 pacientes ambulatorios y 22 hospitalizados que recibían como tratamiento antiepiléptico Fenitoína o Carbamacepina, se extrajeron muestras salivales estimuladas que se recogieron en dos fracciones en distintas horas del día. El cociente 1ra. fracción / 2da. fracción salival de concentración de droga (Sl/S2) fue mayor durante la mañana (07:00-09:00) que en horario vespertino (19:00-21:00), respondiendo al más pronunciado tono simpático matutino. Pacientes cuyas convulsiones no estaban controladas mostraron, algunas horas posteriores a la crisis, cocientes Sl/S2 inferiores que pacientes asintomáticos durante el mismo período del día. Nuestros resultados sugieren que el cociente Sl/S2 de concentraciones salivales de fármaco podría estar ligado con el tono simpático de los individuos, teniendo en cuenta su influencia sobre la regulación del aparato circulatorio. UaY is the specific ZnII2Cys6 transcriptional activator of the purine utilisation pathway in Aspergillus nidulans. Previous work has determined the consensus binding sequence by EMSA and foot-printing. We determine here that it binds as a dimer to its cognate CGG-N6-CCG sites. We identify the uaY109 mutation, which has been shown to affect differentially the expression of a number of UaY-regulated genes, as a F112I substitution in the DNA-binding motif dimerisation domain. We isolated back mutants, revertants carrying different residues at the same position (I112N and I112M) and intragenic suppressors mapping in the first loop of the Zn cluster (N75T and N75K).We have analysed the original mutant and its revertants by growth tests and by their effects on the mRNA steady states of five UaY-regulated genes. We have determined the effect of the different mutations on UaY dimerisation, on the apparent Kdiss of the UaY DNA-binding domain to appropriate DNA sequences and on the methylation interference pattern. We have attempted to rationalise these phenotypes by modelling the UaY DNA binding domain on the structure of the highly similar Ppr1p. However, modelling of the wild-type and mutant proteins provides only a partial explanation for the observed phenotypes. The effect of pure Zn2+ and combined Zn2+ and Mn2+ supplements on the growth and ligninolytic activity of wood-rotting basidiomycetes was studied. Based on measurements of minimum inhibitory concentration and minimum decolourisation-inhibiting concentration available from cultures of Inocutis sp., Trametes versicolor and Gloeophyllum trabeum, and a comparison of such results with those obtained in previous work for P. chrysosporium and P. atropurpurascens, metal supplements were shown to inhibit fungal physiology at concentrations within the study range. Average minimum inhibitory concentration values were consistently lower for ZnSO4 than for ZnSO4/MnSO4 (2 mM Zn2+), demonstrating the antagonistic effect of the tested cations. Furthermore, because the ligninolytic system of P. atropurpurascens had shown a high tolerance to metals, its ligninolytic and germinative behaviour was compared with that of P. chrysosporium. Conidial germination was inhibited at 1 mM of Zn2+ in the two strains, whereas extracellular activity was inhibited at concentrations above 2 mM. Glycosidases represent excellent green chemistry alternatives as catalysts for the synthesis of glycosides, and in particular their stereoselectivity allows the production of anomerically pure glycosides, in only one reaction step using mild reaction conditions. Here, we report the enzymatic synthesis and structural characterization of 3-aminopropyl-1-O-β-D-galactopyranoside. Optimal reaction conditions for the transgalactosylation reaction were 100 mM lactose, 500 mM 3-amino-1-propanol and 24 h of incubation at 50 °C with 6 U/mL of β-galactosidase from Aspergillus oryzae. The fact that the synthesis of 1-propyl-2-O-β-D-galactopyranoside using 1-amino-2-propanol as acceptor was not achieved, and that N-glycoside formation was not observed, confirms the selectivity of β-galactosidase for the synthesis of O-glycosides, and particularly for primary alcohols. The synthesized galactosides were evaluated for their ability to interact with bovine spleen galectin-1 (Gal-1) by using the hemagglutination inhibition assay; results demonstrated that 3-aminopropyl-1-O-β-D-galactopyranoside may be considered as a functionalized galactose moiety more than an efficient Gal-1 inhibitor. The proposed approach constitutes a promising tool for the generation of glycopolymers and glyconanoparticles with potential applications in the development of biosensors as well as construction blocks in chemical synthesis. The derivatives were characterized by kinetics parameters, co-solvent (ethanol and acetone) and temperature stability. Hydrophilization achieved important co-solvent stabilization in all cases. One of the most remarkable results obtained was a 25-fold increase in the half-life of the A. oryzae Glut90-Pal-Pam derivative in 50% (v/v) acetone. Stabilization achieved in very drastic co-solvent concentrations is directly related to the hydrophilization of the nano-environment. The KM values show that the hydrophilic shell appears to behave as an open structure and may create a “partition effect” that protects the enzymes from denaturation. The effect of increasing concentration of co-solvents on the stability of both soluble and immobilized E. coli �-galactosidase was studied. The enzyme was immobilized on to glutaraldehyde-agarose and the co-solvents tested were: N,N-dimethylformamide (DMF), ethanol, acetone, and dioxane (6–36% v/v). Deactivation kinetics were analyzed according to the two-step deactivation model proposed by Henley and Sadana. A multi-temperature study revealed that the immobilized derivative is considerably more stable than the soluble enzyme, even at high temperature where the half life of the soluble enzyme is less than 1 h. Enzyme immobilization did achieve thermal and solvent stabilization at low concentrations, but this task proved more difficult with increasing co-solvent concentrations. In the case of ethanol and acetone, with the increase in co-solvent concentration, the immobilized derivative became less stable than the soluble enzyme. Immobilization may impose certain constraints on the protein structure which have the effect of sensitizing it to denaturation at high co-solvent concentrations. Complementary stabilization strategies are, therefore, being studied. Although redox state is a well-known key process parameter in microbial activity, its impact on wine volatile aroma compounds produced during fermentation has not been studied in detail. In this study we report the effect of reductive and microaerobic conditions on wine aroma compound production using different initial amounts of yeast assimilable nitrogen (YAN: 180 and 400 mg N/l) in a simil grape must defined medium and two S. cerevisiae strains commonly used in wine-making. In batch fermentation culture conditions, reductive conditions were obtained using flasks plugged with Muller valves filled with sulphuric acid; while microaerobic conditions were attained with defined cotton plugs. It was found that significant differences in redox potential were obtained using the different plugs, and with variation of over 100 mV during the main fermentation period. Significant differences in the final concentration of higher alcohols, esters and fatty acids were attributed to differences in the redox state in the medium in both strains. A consistent increase in esters and medium chain fatty acids, as well as a decrease of higher alcohols and isoacids, was seen under reductive fermentation conditions. Interestingly, 1-propanol, d-butyrolactone and ethyl lactate concentrations, showed no significant variation under the different redox conditions. A better understanding of the influence of redox state of the fermentation medium on the composition of volatile compounds in wine could enable improvement of vinification management. Tannat is a red variety of Vitis vinifera that has becomethe major variety for the production of premiumred wines in Uruguay. Due to its small cultivation around theworld, research on the viticulture and enology of this variety is still necessary to improvewine quality. In this context, the aimof the presentworkwas to characterize the aroma profile of Uruguayan Tannat wines using chemical and sensory methodologies. The volatile composition of ten Uruguayan Tannat wines, sold in the international market, was studied by gas-chromatography (GC–MS). Sixty two volatile compounds were identified using GC–MS, being alcohols and esters the most abundant compounds. Only few volatile compounds were found at concentrations higher than their odor threshold in all samples. Sensory characterization of wine aroma was characterized by a panel of wine professionals using projective mapping. Red fruits, fruity, dry fruits, and woody were the main descriptors used for describing similarities and differences in the aroma profile of the wines. Projective mapping sorted samples into four main groups. Partial least square regression (PLSR) enabled to explain many of the most important sensory descriptors (woody, earthy, phenolic, sulfur, chemical and microbiological) through volatile composition. As a Medical Geology research issue, an environmental arsenic risk assessment study in the most important sedimentary aquifer in southern Uruguay is presented. The Raigón Aquifer System is the most exploited in Uruguay. It has a surface extent of about 1,800 square kilometres and 10,000 inhabitants in San Jose Department, where it was studied. Agriculture and cattle breeding are the main economic activities and this aquifer is the basic support. The groundwater sampling was done on 37 water samples of PRENADER (Natural Resources Management and Irrigation Development Program) wells. Outcropping sediments of Raigón Formation and the overlying Libertad Formation were also sampled in the Kiyú region. The analyses were performed by inductively coupled plasma-optical emission spectrometry (ICP-OES) and inductively coupled plasma-mass spectrometry (ICP-MS). The results showed 80% samples with arsenic levels exceeding the 10 μg/l of WHO as limit for waters, and 11% exceeds the 20 μg/l limit of uruguayan regulation. The median, maximum and minimum water arsenic concentrations determined have been 14.24, 24.19 and 1.44 μg/l, respectively. On the other hand, nine sediment samples of Raigón and Libertad Formations in Kiyú region were analysed and yielded median, maximum and minimum arsenic concentrations of 5.03, 9.82 and 1.18 ppm, respectively. This issue leads to the supposition that the population, as well as industrial and agricultural activities, are consuming water with arsenic concentrations over the national and international maximum recommended limit. Como una aproximación a la Geología Médica en Uruguay se presentan los resultados de un estudio sobre el estado del Sistema Acuífero Raigón en relación a la presencia de arsénico, en el entendido que es el acuífero más explotado en el país. En particular, el área cubierta por este trabajo es de unos 1800 km2 en un territorio con una población de unos 10,000 habitantes en el departamento de San José. La colecta de aguas del acuífero se realizó en perforaciones de la base de datos de PRENADER (Proyecto de Manejo de Recursos Naturales y Desarrollo del Riego) obteniéndose 37 muestras, y sobre 9 muestras de sedimentos tanto de la Formación Raigón como la suprayacente Libertad en afloramientos de la región de Kiyú. Los análisis fueron realizados mediante ICP-MS. Un 80% de las muestras de aguas analizadas mostraron contenidos mayores a los límites de la OMS (10 μg/l) y un 11% de las muestras superaron el límite de la OSE (20 μg/l). El valor medio, máximo y mínimo determinado en el caso del agua subterránea fue de 14.24, 24.19 and 1.44 μg/l, respectivamente. Por otro lado, las nueve muestras de sedimentos analizados mostraron concentraciones media, máxima y mínima de arsénico de 5.03, 9.82 and 1.18 ppm, respectivamente, descartándose en principio un origen geogénico. The development of a solid-phase biocatalyst based on the reversible covalent immobilization of laccase onto thiol-reactive supports (thiolsulfinate-agarose [TSI-agarose]) was performed. To achieve this goal, laccase-producing strains isolated from Eucalyptus globulus were screened and white rot fungus Trametes villosa was selected as the best strain for enzyme production. Reduction of disulfide bonds and introduction of “de novo” thiol groups in partially purified laccase were assessed to perform its reversible covalent immobilization onto thiol-reactive supports (TSI-agarose). Only the thiolation process dramatically improved the immobilization yield, from 0% for the native and reduced enzyme to 60% for the thiolated enzyme. Mild conditions for the immobilization process (pH 7.5 and 4°C) allowed the achievement of nearly 100% of coupling efficiency when low loads were applied. The kinetic parameters, pH, and thermal stabilities for the immobilized biocatalyst were similar to those for the native enzyme. After the first use and three consecutives reuses, the insoluble derivative kept more than 80% of its initial capacity for decolorizing Remazol Brilliant Blue R, showing its suitability for color removal from textile industrial effluents. The possibility of reusing the support was demonstrated by the reversibility of enzyme-support binding. A thiol-β-cyclodextrin was synthesized by a simple and environmentally friendly three-step method comprising epoxy activation of β-cyclodextrin, thiosulfate-mediated oxirane opening, and further reduction of the S-alkyl thiosulfate to a thiol group. The final step was optimized by using thiopropyl-agarose, a solid phase reducing agent with many advantages over soluble ones. β-Cyclodextrin thiolation was confirmed by titration with a thiol-reactive reagent, NMR studies, and MALDI-TOF/TOF. Thiolated cyclodextrin had an average value of one thiol group per molecule. Thiol-β-cyclodextrin proved to be an excellent agent for controlling polyphenol oxidase activity. Homology models of CYP26B1 (cytochrome P450RAI2) and CYP26B1 spliced variant were derived using the crystal structure of cyanobacterial CYP120A1 as template for the model building. The quality of the homology models generated were carefully evaluated, and the natural substrate all-trans-retinoic acid (atRA), several tetralone-derived retinoic acid metabolizing blocking agents (RAMBAs), and a well-known potent inhibitor of CYP26B1 (R115866) were docked into the homology model of full-length cytochrome P450 26B1. The results show that in the model of the full-length CYP26B1, the protein is capable of distinguishing between the natural substrate (atRA), R115866, and the tetralone derivatives. The spliced variant of CYP26B1 model displays a reduced affinity for atRA compared to the full-length enzyme, in accordance with recently described experimental information. Bacterial adenosine 5’-diphosphate-ribosylating toxins are encoded by several human pathogens, such as Pseudomonas aeruginosa (exotoxin A (ETA)), Corynebacterium diphtheriae (diphtheria toxin (DT)), and Vibrio cholerae (cholix toxin (CT)). The toxins modify eukaryotic elongation factor 2, an essential human enzyme in protein synthesis, thereby causing cell death. Targeting external virulence factors, such as the above toxins, is a promising alternative for developing new antibiotics, while at the same time avoiding drug resistance. This study aims to establish a reliable computational methodology to find a “silver bullet” able to target all three toxins. Herein, we have undertaken a detailed analysis of the active sites of ETA, DT, and CT, followed by the determination of the most appropriate selection of the size of the docking sphere. Thereafter, we tested two different approaches for normalizing the docking scores and used these to verify the best target (toxin) for each ligand. The results indicate that the methodology is suitable for identifying selective as well as multitoxin inhibitors, further validating the robustness of inverse docking for target-fishing experiments. Title A name given to the resource Structural insights into human microsomal epoxide hydrolase by combined homology modeling, molecular dynamics simulations and molecular docking calculations Abstract A summary of the resource. A new homology model of human microsomal epoxide hydrolase was derived based on multiple templates. The model obtained was fully evaluated, including MD simulations and ensemble-based docking, showing that the quality of the structure is better than that of only previously known model. Particularly, a catalytic triad was clearly identified, in agreement with the experimental information available. Analysis of intermediates in the enzymatic mechanism led to the identification of key residues for substrate binding, stereoselectivity, and intermediate stabilization during the reaction. In particular, we have confirmed the role of the oxyanion hole and the conserved motif (HGXP) in epoxide hydrolases, in excellent agreement with known experimental and computational data on similar systems. The model obtained is the first one that fully agrees with all the experimental observations on the system.
