Background: All-trans retinoic acid (atRA) plays an essential role in the regulation of gene expression, cell growth and differentiation and is also important for normal cardiovascular development but may in turn be involved in cardiovascular diseases, i.e. atherosclerosis and restenosis. The cellular atRA levels are under strict control involving several cytochromes P450 isoforms (CYPs). CYP26 may be the most important regulator of atRA catabolism in vascular cells. The present study describes the molecular cloning, characterization and function of atRA-induced expression of a spliced variant of the CYP26B1 gene. Methodology/Principal Findings: The coding region of the spliced CYP26B1 lacking exon 2 was amplified from cDNA synthesized from atRA-treated human aortic smooth muscle cells and sequenced. Both the spliced variant and full length CYP26B1 was found to be expressed in cultured human endothelial and smooth muscle cells, and in normal and atherosclerotic vessel. atRA induced both variants of CYP26B1 in cultured vascular cells. Furthermore, the levels of spliced mRNA transcript were 4.5 times higher in the atherosclerotic lesion compared to normal arteries and the expression in the lesions was increased 20-fold upon atRA treatment. The spliced CYP26B1 still has the capability to degrade atRA, but at an initial rate one-third that of the corresponding full length enzyme. Transfection of COS-1 and THP-1 cells with the CYP26B1 spliced variant indicated either an increase or a decrease in the catabolism of atRA, probably depending on the expression of other atRA catabolizing enzymes in the cells. Conclusions/Significance: Vascular cells express the spliced variant of CYP26B1 lacking exon 2 and it is also increased in atherosclerotic lesions. The spliced variant displays a slower and reduced degradation of atRA as compared to the fulllength enzyme. The vibrational spectroscopic behavior of a series of 16 palladium(II) complexes with 8 bioactive nitrofuran containing thiosemicarbazones as ligands has been studied in the solid state. The IR and Raman spectra of these complexes and the free nitrofuran thiosemicarbazone ligands were recorded and analyzed. Experimental spectra were satisfactorily described by density functional theory (DFT) calculations. The combination of experimental and theoretical methods allowed us to perform the characterization of the main vibrations that show the mode of coordination of the thiosemicarbazone moiety to palladium even though these vibration bands are located in spectral regions showing a complicated pattern due to the presence of vibrations of the nitrofuran moiety and combination modes involving furan vibrations. A characteristic vibrational spectroscopic pattern has been defined for Pd(II) 5-nitrofuryl thiosemicarbazone complexes. This systematic knowledge may be useful for the analysis of the spectroscopic behavior of other coordination compounds holding the 5-nitrofuran thiosemicarbazone moiety. The interaction between a complex porphyrin-like system formed by an iron atom and multivacant graphene layer and O2, CO and CO2 molecules is studied, using Density Functional Theory (DFT) calculations. The multivacancy graphene system used for this study, consists in the removal of a 1,4-dimethybenzene-like moiety, in a 6 × 6 supercell. This removal and the structural optimization subsequently performed, yield to a biaxial vacancy, where the location of an iron atom embedded in it, lead to a system with resemblance to iron-porphyrin systems. This similar structure could be used to form complexes where gas molecules are allowed to interact with these iron-octavacant graphene systems. In the search for a pharmacological answer to treat Chagas disease, eight metal complexes with two bioactive bisphosphonates, alendronate (Ale) and pamidronate (Pam), were described. Complexes of the formula [M 2 II(Ale) 4(H 2O) 2]·2H 2O, with M = Cu, Co, Mn, Ni, and ([CuPam]·H 2O) n as well as [M II(Pam) 2(H 2O) 2]·3H 2O, with M = Co, Mn and Ni, were synthesized and fully characterized. Crystal structure of [Cu 2 II(Ale) 4(H 2O) 2]·2H 2O, [Co II(Pam) 2(H 2O) 2] and [Ni II(Pam) 2(H 2O) 2] were solved by X-ray single crystal diffraction methods and the structures of [M 2 II(Ale) 4(H 2O) 2] ·2H 2O complexes M = Co, Mn and Ni were studied by X-ray powder diffraction methods. All obtained complexes were active against the amastigote form of Trypanosoma cruzi (T. cruzi), etiological agent of Chagas disease. Most of them were more active than the corresponding free ligands showing no toxicity for mammalian cells. The main mechanism of the antiparasitic action of bisphosphonates, inhibition of parasitic farnesyl diphosphate synthase (TcFPPS), remains in the obtained metal complexes and an increase in the inhibiting enzyme levels was observed upon coordination. Observed enzymatic inhibition was selective for TcFPPS as the metal complexes showed no or little inhibition of human FPPS. Additionally, metal complexation might improve the bioavailability of the complexes through the hindrance of the phosphonate group's ionization at physiological pH and, eventually, through the ability of plasma proteins to work as complex transporters. Nowadays a rational design of solid oxide fuel cells (SOFCs) cathodes is possible thanks to first-principles calculations based on density functional theory (DFT). We study the effect of Cu-doping in the bulk properties for the perovskite Ba0.5Sr0.5Fe1-xCuxO3-δ (with x = 0, 0.25 and 0.50) and correlate the results with previous experimental characterization. Bulk properties such as geometric structure, charge analysis, thermodynamic stability, vacancy formation energy, oxygen diffusion and electronic structure where studied in detail to provide an explanation of the oxygen reduction reaction (ORR) activity enhancement with Cu-doping. The results obtained here, using GGA+U, demonstrate the first-principles approach gives useful information that allows the prediction and explanation of experimental characterizations. A first principles simulation of the crystal structure of potassium peroxocarbonate is presented, using density functional methods (both local and semilocal) for the calculations. An experimental crystal structure with a seemingly inconsistent disposition of the peroxide bonds was used as initial input. Both geometry optimizations of the molecular structure and optimization of the cell size were performed. While cell parameters and heavier atom positions determined at the GGA level are very close to the experimental ones, there are important discrepancies in the positioning of the hydrogen atoms. As a result of these calculations, it was shown that the assignment of the peroxydic hydrogens and the peroxydic bond in the experimental structure was incorrect. A more accurate structure is presented and geometrical as well as cell parameters described. It is also shown that LDA is not accurate enough to describe this type of ionic crystals, because of overbinding of the ions, leading to incorrect cell parameters and volume. Our methodology was validated using DFT methods with several basis sets. The relative affinity of the recently synthesized receptor bambus[6]uril (BU[6]) towards halides in encapsulation processes was theoretically studied by means of M06-2X and B3LYP calculations. The gas-phase results do not follow the affinity experimentally observed in solution, as the largest encapsulation energy was obtained for fluoride, and the smallest one for iodide. Nevertheless, this finding is in agreement with the gas phase hydrogen bond energies determined for the CH4⋯X– complexes, X = F–, Cl–, Br–, I–. The structural changes experienced by the receptor when the halide is inside decrease as we move from fluoride to iodide. Thus, the relative affinity is due to a balance between the hydrogen-bond energies between the methine groups inside the cavity and the anions (CH⋯X–) and the deformation of the receptor induced by the anion. When solvents effects are considered, the affinity of BU[6] for halides is in line with experimental evidence. In effect, the difference between the computed for iodide and bromide is 1 kcal/mol, just 0.5 kcal/mol smaller than the experimental value. The effect of the counter ion was evaluated using Na+, but inclusion of the latter worsens the agreement with experiment. Finally, we discuss methodological problems observed for the BU[6]·X– complexes. Este trabajo describe la optimización de un método espectrofotométrico clásico (“método Hornsey”) para determinar hierro hemo en carnes. Este método extrae hierro hemo de la carne mediante agitación con acetona ácida, maceración en la oscuridad y determinación espectrofotométrica a 640nm sobre el filtrado. Con el método optimizado se analizaron distintos cortes de carne vacuna para evaluar su valor nutricional en cuanto a niveles de hierro biodisponible. A partir de una optimización racional, se obtuvieron las condiciones experimentales adecuadas para realizar un método analítico, considerado de referencia, de forma sistemática y protocolizada en los laboratorios para evaluar el contenido de hierro hemo en carnes. Chagas disease is a neglected tropical disease caused by the protozoan parasite Trypanosoma cruzi. It is estimated that 6 million people are infected in Latin America. Current treatment is not effective due to the severe side effects and the limited efficacy towards the chronic phase of the disease. Considering the growing need for specific anti-Trypanosoma cruzi drugs, organometallic Pt and Pd based compounds were previously synthesized. Although the Pt-based compound effects on T. cruzi death have been reported, no mechanism of action has been proposed for the Pd-based analogous compound. In this work, we determined excellent to very good values of IC50 and SI. To analyze the compound mode of action, we measured Pd uptake and its association to the macromolecules of the parasite by electrothermal atomic absorption spectrometry. We found a poor uptake, which reaches only 16% after 24 h of incubation using 10× IC50, being the scarce incorporated metal preferentially associated to DNA. However, this compound has a trypanocidal effect, leading to morphological changes such as shortening of the parasite cell body and inducing necrosis after 24 h of treatment. Furthermore, this compound impairs the parasite development in the host both at the trypomastigote infection process and the intracellular amastigotes replication. In conclusion, our findings support that Pd-dppf-mpo compound constitutes a promising anti-T. cruzi compound effective against the chronic phase of the disease. Soybean glycinin [11S] was modified by an acidic pH treatment to improve its emulsifying properties. Glycinin was obtained by isoelectric precipitation (11Sn) and then treated with acid (11St). The oil–water interfacial tension and rheology were measured. The rate constants of adsorption (ka) and rearrangement (kr) of proteins at the interface and the dilational (E), elastic (Ed), and viscous modulus (Ev) of the protein interfacial film were determined. Particle size distribution (PSD), interfacial protein concentration (Γ), and the creaming destabilization rate constants ks (for the smaller droplets) and kl (for the larger droplets) were analyzed in oil‐in‐water emulsions (25% v/v of oil and 75% v/v of 1 mg/mL protein solution in 10 mM sodium phosphate buffers, pH 7.0 and 2.5, respectively). Compared to the native protein, the acid treatment caused irreversible denaturation of 11S and significantly increased Г, ka, E, Ed, and Ev (P ≤ 0.05), resulting in a greater rate of protein adsorption to the interface and a stronger interfacial film. PSD showed a bimodal distribution with peaks above and below 4 μm. Smaller droplets moved toward smaller diameters for 11St. It also showed lower values of creaming destabilization constants ks (d ≤ 4 μm) and kl (d ≥ 4 μm) than 11Sn. In conclusion, acid treatment of 11S enhances the creaming stability of its emulsions by improving the interfacial properties and reducing the droplet size. Chia proteins were extracted by solubilisation at pH 10 or 12 and precipitated at pH 4.5. Isolates were named as CPI10 and CPI12, according to their extraction pH, 10 or 12, respectively. The surface properties of both isolates were studied at neutral conditions. Foams were formed by air bubbling and both the formation and destabilization processes were analysed by conductimetry. The extraction pH significantly affected the interfacial properties of chia proteins. The higher surface hydrophobicity in CPI10 led to more flexible proteins with improved foaming properties. Foams formed by CPI10 were more stable than those by CPI12 due to the formation of a thicker interfacial film, which meant a greater ability to retard liquid drainage. Freshly-made coarse emulsions stabilized with CPI12 showed a lower mean droplet size and a significantly lower degree of overall destabilization than those stabilized with CPI10. The inhibitory effects of Cannabis flower extracts, obtained by supercritical carbon dioxide (scCO2) with and without modifier, on various human tumour cells and non-tumour cells were evaluated. Different techniques were used to optimize the polarity interaction between solute and solvent before extraction. An increase in the cannabinoid content of interest (CBD/THC) was evaluated by decarboxylation of the flowers at 110 and 140 °C. Extractions with pure scCO2 were conducted in a single step at temperatures of 50 and 70 °C and pressures of 22 and 40 MPa, varying the time of prior decarboxylation by 0.5 or 2.0 h. Sequential extractions were conducted in a first step at 35 °C and 10 MPa and then in a second step at 70 °C and 40 MPa, with and without modifier. Contents of the cannabinoids were evaluated using HPLC. Antitumor activity of the extracts was evaluated using the MTT assay. The highest yields and highest cannabinoid contents in the extracts were obtained with high solvent density values. Extracts with high concentrations of neutral cannabinoids showed high antitumor activity for cervical cancer cell lines. Here we present a new analytical method where immunoconcentration of the analyte is coupled to quantitative matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) analysis allowing in minutes the identification and highly sensitive quantitation of microcystins (MCs) as model targets. The key element is a site-specific in vivo biotinylated nanobody of broad cross-reactivity with microcystins. The single biotin moiety at the C-terminus and the small size of the nanobody (15 kDa) enable its oriented and tightly packed immobilization on magnetic beads, providing a highly efficient capture of the toxin. The binding capacity of the bioadsorbent is partially loaded with an easily synthesized internal standard for MS quantitation. After capture, the beads are directly dispensed on the MALDI-TOF MS target enabling the identification and sensitive quantitation of the microcystin (MC) congeners. Since salts and contaminants are removed during the concentration step, no cleanup or other sample treatments are needed. The method was validated with a large number of water and serum samples with excellent precision and recovery at quantitation limits of 0.025 μg/L of MC. Effect of a combined power ultrasound (US) and chlorine disinfection treatment on quality of fresh-cut ready-to-eat lettuce (cv. Vera) was evaluated. Fresh-cut lettuce was disinfected using 100 mg L−1 NaOCl solution alone and in combination with ultrasound (130 W, 42 kHz) for different times (0–30 min). Lettuce samples (80 g) were then packaged in polypropylene (PP) film and stored at 5 °C for 8 d. Application of US improved microbial reduction on processing day: an additional reduction in the range of 0.5–2.5 log cfu g−1 was obtained with respect to NaOCl treatment alone. Combined US-NaOCl application did not cause visual changes to lettuce neither on processing day nor when samples were stored under refrigerated conditions in passive modified atmosphere. Confocal images evidenced that US application could affect lettuce surface wax layer, removing its natural defense against microorganisms. This resulted in higher microbial growth throughout storage in samples treated with combined US-NaOCl treatment. US appears as a promising technique for fresh-cut produce disinfection, but further studies are needed to optimize US treatment to allow important microbial reductions throughout all storage time without compromising lettuce shelf life during refrigerated storage. We report the effect of lithium titanate nanotubes (LiTNT) as active fillers for lithium-ion solid polymer electrolytes for lithium batteries. The interaction of LiTNT with polyacrylonitrile host material and enhancement on lithium perchlorate ionic-pair dissociation was clearly evidenced by our vibrational spectroscopy and confocal Raman microscopy studies. In strong correlation with the structural characterization, the active role of LiTNT fillers was evidenced by means of impedance spectroscopy analysis revealing the presence of two contributions to the ionic transport, one due to the bulk and the other possibly mediated by the LiTNT surface in the nanocomposites. A four-order-magnitude enhancement on the lithium-ion bulk conductivity was observed for 15% LiTNT loadings with respect to unloaded samples showing an increment up to ~ 7.1 × 10−4 S.cm−1. In addition, we also evidence that the LiTNT surface-mediated contribution to the lithium-ion transport yielded conductivities in the same order of magnitude. Polarized sensory positioning (PSP) is a novel methodology for sensory characterization which is based on the comparison among samples and a set of reference products, called poles. Its main advantage over other holistic methodologies is that results from different sessions can be compared. This is particularly useful when working with samples with persistent flavours, when evaluating a large number of samples over a long period of time, or during quality control. Although the key step of this methodology is the selection of the products to be considered as poles, no studies have been published addressing this issue. In this context, the aim of the present work was to contribute to bridging this gap by studying the influence of the poles on results from sensory characterization of two widely different products (make-up foundations and orange-flavoured powdered drinks) using Polarized Sensory Positioning with consumers. Results from both studies provided preliminary evidence about the stability of sensory spaces obtained with different set of poles (RV coefficients higher than 0.908). However, differences in conclusions regarding similarities and differences among samples were identified between the sets of poles in both studies, suggesting the need to carefully select stable poles for PSP tasks.
