Suitable molecular size and shape is also needed to complement the electrostatic effect, as clearly evidenced by graphical analysis of output docked conformations. Docking energies values are reasonably well correlated with those obtained by Molecular Dynamics as well as with the experimental Ki values, confirming once again the validity of this type of scoring methods to rapidly assess ligand–receptor affinities. The larval stage of the parasite Echinococcus granulosus causes hydatid disease. The hydatid cyst is potentially capable of activating host complement, since it is a large, persistent, carbohydrate-rich structure, coated with host immunoglobulins, and localized in the host's internal organs. Nonetheless, in vitro studies have suggested that the cyst surface, the hydatid cyst wall (HCW), is a poor complement activator. In this study, we assessed the occurrence of in vivo complement activation on the hydatid cyst by measuring the levels of two complement activation products, C3d and complexes bearing a C9 activation neoepitope (TCC/MAC), in extracts from HCW of human origin. Low amounts of C3d and TCC/MAC were found in HCW in comparison with their levels in normal human plasma and activated human sera, suggesting that in vivo complement activation on HCW is efficiently down-regulated. This regulation may contribute to limit host inflammation which has been observed to correlate with parasite degeneration and death. Opal phytolith, geochemical and sedimentological data from two cores taken in Peña Lagoon, a small water body on the Atlantic shore of Uruguay, are presented in this paper. The choice of this specific sampling location aimed to obtain high resolution records to infer late Holocene environmental and climatic changes in southeastern Uruguay, in order to improve previous models for early and middle Holocene. The combined record of two cores extends to 2500 14C BP, with a sedimentation rate ranging between 0.6 and 2 mm y−1. A multiproxy approach allowed the identification of changes in dynamics, particularly organic matter and sediment input variability from the watershed. With this information, differences in the amount and extent of environmental signals were evaluated and a comprehensive climate change model was generated, especially for the last 1500 years. Three main climatic events were distinguished. The oldest, which lasted until 700 AD, was characterized by temperate and humid conditions. The second extended until 1200 AD and warmer and wetter conditions were inferred, but also a colder and drier pulse was observed in between. This second event was assigned to the Warm Medieval Period (WMP). During the third event, assigned to the Little Ice Age (LIA), which extended until the present, a high variability in climate was observed, but also three dry/cold phases reaching their maxima by 1300, 1600 and 1900 AD were inferred. During the “El Niño”- Southern Oscillation (ENSO) event 2009–2010, trophic state uni (TSI’s) and multimetric (TRIX’s) indexes were determined in the north coast of the Río de la Plata (RdlP). The relationships of such indexes with the hydrological, physical and chemical parameters were explored to identify climatic variability or anthropogenic effects on the eutrophication process. Eleven oceanographic sampling campaigns at 25 stations were conducted along the north coast of RdlP from 2009 to 2011. Temperature, salinity and oxygen saturation, water transparency (Secchi depth), chlorophyll (Chl a) content and total nutrient concentrations (total nitrogen and phosphorous) were determined. Trophic indexes (TSI Chl a, TSI TP, TRIX NP-Voll. and TRIX NP-Mvdeo.) were quantified and the RdlP flow was measured. RdlP flow showed significant differences between ENSO phases, with higher values during “El Niño” (EN), negative and significant correlations with salinity, Secchi depth and positive with total nutrients. The trophic status indexes showed spatial and temporal variability and associations with flow, physical and chemical parameters. Trophic state indexes showed spatial and temporal heterogeneity and were strongly associated to environmental conditions (hydrological and seasonal oscillations) during the study period but also to the anthropogenic factors affecting the north coast of RdlP. The system has been classified as mesotrophic to hypertrophic, depending on the selected indicator. According to these results, we suggest the use of a multimetric trophic state indicator, i.e., the TRIX index, as explains better the causes and effects of the eutrophication processes. To implement the TRIX index, (TRIX NP-Mvdeo.) we suggest researchers to consider a large number of local records (Chl a, nutrients, oxygen) at seasonal scales. This index is adapted to the environmental conditions of the study zone and complemented with other community structure indexes, and should be used for eutrophication assessment programs in the north coast of RdlP. Winemaking industry produces large amounts of by-products (grape pomace or marc) every year, making necessary a sustainable management of such waste. Among red wines, Tannat variety possesses a unique polyphenolic profile. Consequently, the study of the bioactive properties of its by-product and the extraction optimization of its bioactive compounds is of great interest to state its health promoting potential. In this study, different extraction processes were tested: maceration with ethanol (95%), ultrasound-assisted ethanolic extraction optimization with surface methodology, ultrasound-assisted aqueous extraction, and hydro-alcoholic-acid extraction (EHAA). Using this last extraction procedure, higher polyphenolic content (11.459 ± 1.048 g GAE/100 g of dry extract), monomeric anthocyanins (2.030 ± 0.085 g Cyd/100 g of dry extract), antioxidant capacity (0.474 ± 0.036 mg/mL for ABTS and 0.715 ± 0.063 µmol TE/mg of dry extract for ORAC-FL), α-glucosidase (IC50 888.5 ± 79.3 µg/mL), and pancreatic lipase inhibition capacity (IC50 2431.0 ± 79.9 µg/mL) were found on Tannat grape skin. Considering as a whole, these results may imply a great antidiabetic and antiobesity potential. Moreover, EHAA showed to have anti-inflammatory capacity (IC50 587 µg/mL) on RAW 264.7 cells. In summary, EHAA possesses great potential as a functional ingredient for prevention and/or treatment of chronic diseases. Herein, we report the assessment of six DFT methods (BP86, TPSS, B3LYP, X3LYP, PBE1PBE, BH&HLYP) in describing the nature of the chemical bonds, structural, and electronic properties of the closed-shell oxocomplex of Re(V) [ReO(bdt)2]� and the open-shell oxocomplex of V(IV) [VO(bdt)2]2� (bdt2� = 1,2-benzenedithiolato). Several basis sets were employed, and different effective core potentials were taken as variables into account. PBE1PBE is found to be the best overall performer in describing M@S bonds. It is followed by two groups composed in turn of BP86, TPSS, and BH&HLYP, and another one including B3LYP and X3LYP. For the M@O bonds, the performance has a direct dependency on the Hartree–Fock exchange. On going from the pure GGA (BP86) and m-GGA (TPSS) to BH&HLYP, a sensible improvement is obtained for the Re@O bond distances. However, for the V@O bond the opposite is true, being BH&HLYP the worst choice. The M@S and M@O chemical bonds have been analyzed in terms of the natural bond orbital theory. All sulfur atoms participate in r bonds strongly polarized toward the non-metal, in which the hybridization at the metal has mainly d character and at the non-metal, p character. For the Re@O bonds, interesting features were found. This work shows that PBE1PBE in combination with the MIDI! basis set and the Stuttgart energy consistent pseudopotentials for metals is the best methodology for the study of rhenium and vanadium complexes with thiolato ligands. We have tested our hypothesis performing comparative calculations for the [Re6S8X6]4� (X� = Cl�, Br�, I�) hexanuclear clusters. The results obtained at the PBE1PBE/STMIDI significantly improve those published previously and the computational cost is small. Therefore, we recommend this methodology to study large of transition metal complexes containing thiolato ligands. In order to assess the utility of mtDNA typing for forensic purposes in Uruguay, a population sample of 120 maternally unrelated individuals were amplified and directly sequenced for the HV1 and for the HV2 segments in the control region of the human mtDNA, following previous international recommendations (1). Blood and saliva samples were taken from 120 unrelated individuals and deposited into FTA cards (FTA card, Whatman WB 12-0205). The DNA was extracted in a 1.2 mm punch according to the manufacturer recommendations. DNA amplification and sequencing was performed according to previously published paper (2). All statistical analysis were calculated using the Arlequin Software ver. 2.000 (3). Results are shown in Table 1. In this population sample, 107 different mtDNA sequences were observed out of 120 individuals. Of these, 97 sequences were observed once (80.8%), 8 sequences were observed twice (13.3%), one sequence was observed three times (2.5%), and another four times (3.3%). The variation of mtDNA HV1 and HV2 regions was confined to 146 positions, of which 99 was observed in the HV1 and 47 in the HV2. Sequence variation consisted of 120 transitions, 21 transversions, and 11 indels. The genetic diversity value for combined HV1 and HV2 regions was 0.998 (±0.002) in this Uruguayan population sample, and the nucleotide diversity was 0.068 (±0.035). Process optimization for producing diacylglycerol-enriched oils has received great attention due the importance of these products for healthier and more balanced diets. This work reports experimental data of liquid-liquid phase equilibrium of hydrolyzed olive oil (which comprises on its composition triacylglycerols, diacylglycerols, monoacylglycerols and free fatty acids) with ethanol and water, and subsequent analysis of oxidative stability of the oil obtained from the liquid phases. Liquid-liquid equilibrium experimental data obtained were modeled using the UNIQUAC thermodynamic model, which was well fitted to experimental data with a root mean square deviation of 1.76 wt%. The results obtained demonstrated the potentiality of liquid-liquid extraction as a suitable procedure for olive oil enrichment in diacylglycerol, after a hydrolysis procedure. It was also observed that liquid-liquid phase separation has a significant effect on the content of important antioxidants present in the olive oil, what affects the oxidative stability of the samples submitted to the liquid phase extraction. From the results obtained in this study, it has been also noticed that the acylglycerols and free fatty acids profile in of the oil samples presented a major rule on their stabilities. Oil shales from Mangrullo Formation (Uruguay) was assessed as a potential fuel for energy production. The assessment is based on a comprehensive characterization of the material from a thermal perspective. Co-combustion of this type of fuel with biomass waste can enhance the combustion performance and decrease hazardous gas emissions. The low heating value (3.2 MJ kg−1) of the Uruguayan oil shale indicates that it could be mixed with biomass to obtain better results. Stratified sampling of drill cores was used to obtain a representative sample. The morphological and structural characteristics of the oil shale were studied by X-ray diffraction and scanning electron microscopy. Chemical composition (hydrocarbons and minerals) of oil shales was investigated by X-ray diffraction, X-ray fluorescence, proximate and ultimate analysis and Fourier transform infrared spectroscopy. Rock Eval analysis was performed to measure richness and maturity of the Uruguayan oil shale, the results of the TOC content (8.93%) and the hydrogen index (525 mg HC/ g TOC) indicated that it constitutes an excellent source rock. The kinetic of the combustion and thermal properties of oil shale were studied. The activation energy of the different reaction stages was calculated using the Flynn-Wall-Ozawa model, then we obtained the pre-exponencial factor and the reaction order optimizing the theoretical model using the experimental data. Results indicated that the combustion of the oil shale exhibits multiple reaction stages with activations energies that varies between 152.2 and 316.4 kJ mol−1. This cross-sectional study examines the association between blood lead level (BLL) and two OS markers-urinary F2-8α isoprostane or isoprostane (a marker of lipid peroxidation) and 8-hydroxy-2-deoxy-Guanosine or 8-OH-dG (a marker of DNA damage) in 211 children, aged 5-8 years, from Montevideo, Uruguay. The role of dietary intakes of vitamin C and zinc in modifying the relation between BLL and OS was also examined. The mean (SD) BLL of the study children was 4.7 (2.2) µg/dL, with 30.2% children having BLL ≥5 µg/dL, the current reference level set by the US Centre for Disease Control for identifying, monitoring and management of children with elevated BLL. In covariate-adjusted analysis, there was a weak positive association between BLL and urinary isoprostane (adjusted for specific gravity) [β=0.09, p<0.1]. No association was found between children's BLL and urinary 8-OH-dG. Interactions between dietary intakes of vitamin C or zinc and BLL on OS biomarkers were not consistent. However, when BLL and vitamin C or BLL and zinc were modeled together, BLL was independently associated with isoprostane concentration [β=0.10, p<0.05] but vitamin C or zinc intake was not. These findings suggest that there may be a potential adverse effect of BLL on OS in children with low-level Pb exposure. There is a need to study the effects of Pb on other OS measures, as well as the role of OS in mediating low-level Pb toxicity on functional outcomes. The negative effect of lead exposure on children’s intelligence is well-documented. Less is known about the impact of lead on the use of executive functions to self-regulate behavior. We measured blood lead level (BLL) in a sample of first grade children from Montevideo, Uruguay (n = 206, age 6.7 ± 0.5 years, 59.7% boys). Behavior was assessed with teacher versions of the Conners Rating Scale (CRS) and the Behavior Rating Inventory of Executive Functions (BRIEF). Mean BLL was 4.2 ± 2.1 μg/dL; 10% had mild-to-severe ratings of Attentional Deficit with Hyperactivity Disorder (ADHD) (T score > 65). In negative binomial regression, BLL was not associated with CRS sub-scales, but was associated with a poorer ability to inhibit inappropriate behaviors, prevalence ratio (PR) [95% CI]: 1.01 [1.00, 1.03] as measured by the BRIEF. In covariate-adjusted models, the association with BLL was attenuated. When stratified by sex, the covariate-adjusted association between BLL, hyperactivity, poorer inhitibion, emotional control, and behavioral regulation was marginally significant for girls but not boys. In summary, among children with low lead-exposure, we found some, but nonetheless modest, evidence of a relationship between higher BLL and child behavior. If confirmed by larger studies and other objective measures of behavior, such links could have implications for learning and social interaction, particularly among girls.
