A summary of the resource.
Three independent studies were conducted in Chile, Spain and Uruguay to assess the effects of the addition of enzymes on the shelf life of brown pan bread. Four batches of brown bread were prepared in each country: a control batch with no added enzymes, a second batch with amylase, a third batch with xylanase and a fourth batch with a 1:1 mixture of amylase and xylanase. Three sensory texture parameters were evaluated (softness, size of the soft area and cohesiveness) and a consumer study was conducted to determine the acceptability of the samples. In each country, the doses of each enzyme and enzyme mixtures (1:1) added to doughs have to be adapted to obtain good dough handling characteristics and minimise adhesiveness and stickiness of the corresponding flour in the traditional formulation. In all three countries the addition of the enzyme mixture gave the best results, achieving a longer shelf life, while the addition of xylanase accelerated bread staling.

Abstract
A summary of the resource.
The objective of the research was to test the hamster for a model of transmission of congenital toxoplasmosis. A non-invasive method for the diagnosis of pregnancy in hamsters was designed, with a specificity and a sensitivity of 70.2 and 94.7%, respectively (n = 168). Of 32 females with a chronic toxoplasma infection, 3 transmitted Toxoplasma congenitally during their first pregnancy, but not during the subsequent pregnancy. Congenital transmission rates of infections initiated during pregnancy with 2 stages of 2 strains of Toxoplasma were in the range of 33 to 100% of the 76 females inoculated. Only 1 of 17 females transmitted the parasite exclusively via milk. It was concluded that the hamster is a promising species for a model of transmission of congenital toxoplasmosis.

As a part of our systematic study of foldamer structural elements, we analyze and quantify the conformational behavior of two model compounds based on a frequently used class of aromatic oligoamide building blocks. Combining computational and NMR approaches, we investigate ortho-fluoro- and ortho-chloro-N-methylbenzamide. Our results indicate that the -F substituent in an ortho position can be used to fine-tune the rigidity of the oligomer backbone. It provides a measurably attenuated but still considerably strong hydrogen bond (H-bond) to the peptide group proton when compared to the -OCH3 substituent in the same position. On the other hand, the ortho-Cl substituent does not impose significant restrictions on the flexibility of the backbone. Its effect on the final shape of an oligomer is likely governed by its size rather than by noncovalent intramolecular interactions. Furthermore, the effect of solvent on the conformational preferences of these building blocks has been quantified. The number of intramolecularly H-bonded conformations decreases significantly when going from nonprotic to protic environments. This study will facilitate rational design of novel arylamide foldamers.

Preliminary findings have suggested that 99mTc-glucarate has tumor-seeking properties. The purpose of this study was to explore the potential of this tracer to evaluate malignant head and neck tumors by means of SPECT/CT software fusion imaging. Methods: Eleven male patients with advanced head and neck carcinoma were included in the study: 9 with locally advanced disease and 2 with clinical suspicion of local relapse. Scanning started 3–6 h after the injection of 1,110 MBq of 99mTc-glucarate. Planar and SPECT images of the head, neck, and thorax were acquired. Three-dimensional images were also coregistered with CT. Results: We found 99mTc-glucarate uptake in all suspected lesions. SPECT/CT fusion imaging was helpful in all cases for topographically localizing the tracer foci. Conclusion: 99mTcglucarate can be considered a potential tracer for the evaluation of patients with head and neck tumors.

A summary of the resource.
The use of oxidoreductases as biocatalysts for the production of a wide number of chiral building blocks is presently a mature (bio-)technology. In this context some industrial applications are currently performed by means of those enzymatic approaches, and new examples are expected to be realized. Moreover, oxidoreductases provide an interesting academic platform to undertake fundamental research in enzymology, to acquire a better understanding on catalytic mechanisms, and to facilitate the development of new biocatalytic applications. Within this area, a wide number of oxidoreductases from genus Candida spp. have been characterized and used as biocatalysts. These enzymes are rather diverse, and are able to carry out many useful reactions, like highly (enantio)selective keto-reductions, (de)racemizations and stereoinversions, and promiscuous catalytic imine reductions. In addition, some Candida spp. dehydrogenases are very useful for regenerating the cofactors, with the aid of sacrificial substrates. Addressing those features, the present paper aims to give an overview of these enzymes, by focusing on practical applications that these biocatalysts can provide. Furthermore, when possible, academic insights on the enzymatic performances will be discussed as well.

Las glutation transferasas (GSTs) son una superfamilia de protefnas multifuncionales que cumplen funciones esenciales en la detoxificacion de compuestos tanto endogenos como exogenos. Las GSTs pueden ser c1asificadas en cuatro familias, siendo las citosolicas las estudiadas en mayor profundidad. En parasites helmintos se identificaron varias GSTs citosolicas con roles clave para su sobrevida en el hospedero. Asimismo, no solo la actividad detoxificante es importante sino que las GSTs, particularmente las de c1ase Sigma, estan implicadas en procesos de inmunomodulacion a traves de su actividad prostaglandina sintasa. En este sentido, es de nuestro interes caracterizar este grupo de enzimas en el parasito Echinococcus granulosus. Particularmente, este trabajo describe la identificaci6n y c1asificacion de • una nueva GST de E. granulosus (EgGST2), 105 avances en la caracterizacion cinetica de EgGSTl tipificada previamente y por ultimo, 105 estudios de expresion de las tres EgGSTs identificadas al momento. A partir del transcriptoma de larva de E. granulosus publicado se obtuvo la secuencia de EgGST2. En la Seccion I se muestra la tipificacion de EgGST2. Los estudios de funcion catalftica ubican a EgGST2 indudablemente como miembro de la c1ase Sigma. Por otro lade, se presenta el trabajo orientado a la produccion de la protefna recombinante enzimaticamente activa y finalmente se muestra que la protefna se expresa en protoescolex de E. granulosus. La enzima EgGSTl fue previa mente estudiada por nuestro grupo de trabajo donde se demostro que pertenece a la c1ase Mu. Por otro lado, se realizaron estudios bioqufmicos de la enzima recombinante, donde se analizo el panel de actividades que presenta asf como sus parametros enzimaticos. En la Seccion 1\ se presenta la profundizacion de la caracterizacion cinetica de EgGST1. En este contexto, se realizaron estudios de velocidades iniciales y de inhibicion por producto, cuyos resultados muestran que EgGSTl sigue un mecanisme secuencial aleatorio concordante con las GSTs de mamffero de c1ase Mu descritas. Por otro lado, se determine la constante de disociacion para cada uno de 105 sustratos por analisis de union en equilibrio. Finalmente, experimentos de pH permitieron acceder al pKa del tiolato del GSH en el complejo binario enzima - GSH, siendo este resultado acorde con 10 reportado para las GST que presentan una Tyr catalftica. En ultimo lugar, en la Seccion III, se exponen 105 estudios de expresion de las tres GST identificadas al momento en E. granulosus. Primero, se presenta el analisis bioinformatico de las regiones 5' flanqueantes de 105 tres genes donde se identificaron elementos relacionados con la regulacion de enzimas detoxificantes. Por otro lado, se observo que las tres isoformas presentan una expresion diferencial a nivel del ARN • mensajero cuando 105 protoescolex son expuestos a condiciones de estres oxidativo 0 xenobioticos in vitro. Finalmente, se estudio la expresion de las EgGSTs en el contexto de la infeccion secunda ria murina temprana. EI conjunto de estes resultados pretende contribuir al entendimiento de la interrelacion hospedero - parasite a partir del entendimiento del rol biologico de este versatil grupo de enzimas detoxificantes.

The reaction of [ReOCl3(PPh3)2] with 1,2-bis(2,3-dimercaptobenzamido) ethane (H4-1) in the presence of Na2CO3 in methanol under anaerobic conditions affords the dinuclear ReV oxido complex [PPh4]2[ReO(1)]2 containing two distorted square-pyramidal {ReVOS4} units bridged by the ligand strands in a double-stranded fashion. The coordination geometry around the metal centers is similar to the one observed for [ReO(bdt)2]–. The ReS4 planes are arranged in a coplanar fashion and are not twisted around the metal–metal vector, which prevents the complex to adopt a helical structure. Luminescence studies show the presence of emission bands, which are assigned to singlet-singlet transitions exhibiting very fast decays (ca. 10 ns). Theoretical Density Functional (DFT) studies on geometry and electronic properties were performed employing the hybrid B3LYP and PBE1PBE functionals. While the general trends observed in the experi- Introduction Oxido complexes of ReV have received considerable attention in the past few years. Particular interests have been aroused due to their reactivity, their photochemical and spectroscopic properties and their role in catalytic processes and oxygen atom transfer (OAT) reactions.[1–9] Furthermore, due to the nuclear decay of 186Re, rhenium complexes found very promising radiopharmaceutical applications.[ 10–13] Rhenium oxido complexes containing dithiolato ligands in particular, have been studied with different purposes. Natural Bond Orbitals (NBO) analysis indicates the presence of polarized Re�O and Re–S bonds, both of them polarized toward the non-metal. The calculation show that the molecular orbitals of the ReV are doubly degenerated, the occupied 5d orbital of rhenium lying beneath occupied sulfur- based MOs due to the rigid geometry imposed by the C– C backbone of the bis(benzene-o-dithiolato) ligands. The origin of all absorption bands is ascribed to a ligand-to-metal charge transfer (LMCT), in which occupied sulfur-based orbitals and unoccupied rhenium-centered orbitals are involved.

Molecular modeling is becoming a common component of inorganic chemistry courses. This usage is a consequence of the growing inclusion of computational techniques in all research fields. Some articles have appeared in this Journal that present molecular modeling exercises designed to show the bonding and acid–base properties of compounds (1–3). We devised a computational experiment to examine the hydration of simple cations and show the geometrical and thermochemical parameters of the hydrated ions. Most undergraduate students have only a limited exposure to the application of molecular modeling to inorganic chemistry problems. This exercise offers a good first approach to the theoretical study of interesting systems. The exercise is divided in two parts. In the first part, a structural study is performed for the species Mn+(H2O) where M = Li, Na, K, Be, Mg, Ca and n = 1 or 2. The influence of the charge and size of the cation on the M−O equilibrium distance is discussed, as well as the distortions on the water molecule caused by the cation. In the second part, the ΔhydH298° for the hydration reaction of the six metals is calculated. Finally, the performance of the different computational models is discussed.

The thermodynamics of protonation of trans-[ReVO2L2]+ (L = aliphatic di and polyamines) complexes in aqueous solution was studied by using experimental and theoretical approaches. The complexes containing diamines undergo protonation on the oxo ligands, whereas those containing polydentate amines protonate on uncoordinated amino groups. The protonation reactions were studied experimentally by microcalorimetric techniques, in all cases exothermic with |DH◦| ranging from 7 to 50 kJ/mol. For complexes containing diamines, the exothermicity was in concordance with the basicities in some cases, while in others no systematic behaviour was found. For complexes with polydentate amines, the enthalpy dominates with a modest influence on the entropy. Theoretically, Density Functional Theory (DFT) methods were employed in the gas phase, and bulk solvent effects were treated by means of the Polarizable Continuum Model. Direct solute–solvent effects were considered adding explicit water molecules. The enthalpy change calculated in the gas phase was in marked disagreement with the experimental results due to the relevancy of solvation/desolvation processes. The explicit inclusion of water molecules led to a good improvement. A discrete-continuum model was also employed, for which DG◦ was overestimated in all cases. Further investigations, both experimental and theoretical are necessary to get a more complete picture of the proton transfer reactions of these complexes. The experimental values herein determined constitute the first step to construct a set of data to which it is possible to benchmark new theoretical approaches to compute the thermodynamics of proton transfer reactions of metal complexes in aqueous solution

Biotransformation of 1,8-cineole by a strain of Aspergillus terreus isolated from Eucalyptus leaves was investigated. This strain produced four oxygenated compounds identified as 2-exo-hydroxy-1,8-cineole, 2-endohydroxy- 1,8-cineole, 3-exo-hydroxy-1,8-cineole and 3-endo-hydroxy-1,8-cineole, with good bioconversion percentage and high stereoselectivity

1. Resumen El objetivo de este Doctorado fue estudiar la nucleación heterogénea, coalescencia y obtención del compuesto laminar BiI3 a escala nano, así como la incidencia de esta última escala en las propiedades físicas y en las aplicaciones tecnológicas de los materiales obtenidos. Se eligió el BiI3 porque se tenía experiencia luego de haber realizado una Maestría en el tema, por ser detector de radiación X –su aplicación más directa-, y por ser un compuesto laminar, lo que hizo pensar que sus planos podrían curvarse y formar nanotubos o nanorods. Para ello se plantearon dos caminos: uno de ellos fue sintetizar las nanopartículas para luego tomarlas como material de partida en futuras nucleaciones, por ejemplo por deposición física de vapor. Este camino fue el que se desarrolló más ampliamente en este Doctorado. El otro camino fue partir de material micrométrico y por deposición física de vapor lograr nucleaciones nanométricas utilizando vacíos mayores y temperaturas menores que las realizadas en trabajos anteriores. Si bien se participó en el desarrollo de un equipo para llevar a cabo estos estudios, no fue posible profundizar en estos aspectos en el transcurso del Doctorado. Para sintetizar nanopartículas de BiI3 se estudiaron tres métodos: solución, hidrotérmico y microondas. En solución se lograron obtener nanopartículas de BiI3 con morfologías y tamaños variados. Las partículas más pequeñas sintetizadas por este método tuvieron un tamaño promedio de 20 nm. También se vio que dicha morfología depende de la temperatura de síntesis. Se observó además cómo estas partículas coalescen cuando incide sobre ellas el haz de electrones del microscopio electrónico de transmisión (TEM). Realizando síntesis en microondas no se obtuvo BiI3. Por el método hidrotérmico se estudió la incidencia del tiempo de síntesis en el tamaño de las partículas, y se encontró que no depende de éste en las condiciones utilizadas. Por el método hidrotérmico se sintetizó BiI3 pero también mezclas de BiI3 y BSI. Se lograron encontrar las condiciones de síntesis adecuadas para obtener solamente BiI3, como ser productos de partida, solvente y temperatura. Se lograron obtener nanopartículas de un tamaño promedio de 20 nm, con morfología hexagonal. Se observó además que las partículas sintetizadas se modifican y dan lugar a otras de diferente morfología al incidir sobre ellas el haz de electrones del TEM. Se realizaron estudios por espectroscopía UV-Vis e infrarrojo medio de las nanopartículas sintetizadas por los métodos de solución e hidrotérmico. Las nanopartículas de BiI3 obtenidas se utilizaron como producto de partida para realizar nucleaciones por deposición física de vapor (PVD). Se obtuvieron nucleaciones con partículas de un tamaño promedio de 100 nm, pero muy separadas entre ellas. También se realizaron nucleaciones suspendiendo las partículas en diferentes solventes, se depositaron sobre sustratos de vidrio por el método de spin coating, y a las mejores nucleaciones se les realizó annealing. Además, las nanopartículas se utilizaron como producto de partida para crecer films por PVD

Chagas disease represents a serious public health problem in South America. The first line of treatment is Nifurtimox and Benznidazole which generate toxic effects in treated patients. We have recently shown that a number of 5-nitrofuranes possess activity against Trypanosoma cruzi through oxidative stress and inhibition of parasite ergosterol biosynthesis, specifically at the level of squalene epoxidase. Here, we identify new 5-nitrofuranes and the thia-analogues with excellent effects on the viability of T. cruzi and adequate parasite/mammal selectivity indexes. Analysis of the free sterols from parasite incubated, during 120 h, with the compounds showed that some of them accumulated squalene suggesting the squalene epoxidase activity inhibition of the parasite. Nifurtimox was able to accumulate squalene only at lower incubation times. Due to this fact some derivatives were also tested as antifungal agents. Quantitative structure–activity relationship studies were also performed showing relevant features for further new derivatives design. Taken together, the results obtained in the present work point to a more general effect of 5-nitrofuranes and 5-nitrothiophenes in trypanosomatids, opening potential therapeutic possibilities of them for these infectious diseases.

A convenient two-step procedure for the synthesis of primary (Z)-allyl amines, (Z)-homoallyl amines [(Z)-but-3-enylamines], and (Z)-pent-4-enylamines using the Wittig reaction was achieved. The use of nonstabilized ylides from triphenylphosphonium salt, potassium salt, and apolar solvent produced (Z=E)- geometric isomer ratios generally greater than 1.6. The amine moiety was masked using a phtalimide group that was removed successfully in the last step of the process in two different conditions, NH2NH2=EtOH=rt or CH3NH2=EtOH=rt. However, in some cases, reduction of the C¼C double bond in the deprotection with hydrazine was concomitantly observed.

Los mayores obstáculos para la toma de decisiones en bioequivalencia promedio están referidos al tamaño de la variabilidad residual. Se ha visto que en ella incide mucho las características de los individuos reclutados para el ensayo. El presente trabajo propone una metodología que estudia la varianza residual según el género de los voluntarios y construye intervalos de confianza para el cociente de medias Test / Referencia (T/R), de los parámetros de biodisponibilidad, tanto en hombres como en mujeres. Asimismo se analizan las varianzas totales producidas por los medicamentos T y R, a los efectos de complementar la toma de decisiones. Las anteriores pautas fueron aplicadas a 3 estudios de bioequivalencia llevados a cabo con fármacos sustratos de la isoenzima CYP3A4 (itraconazol, alprazolam, claritromicina), algunos de los cuales son también sustratos del transportador de eflujo P-glicoproteína. Las mujeres presentaron menor exposición a los fármacos sustratos de CYP3A4 que los hombres. Los medicamentos T y R, conteniendo tales fármacos, mostraron biodisponibilidades relativas dependientes del sexo de los voluntarios, principalmente a causa de probables diferencias de disolución, originadas por la diferente fisiología del tracto digestivo que presentan hombres y mujeres. Como conclusión de este trabajo se entiende que la toma de decisiones en bioequivalencia debería tener en cuenta los resultados obtenidos en cada género, para apoyar tanto que T y R sean bioequivalentes como que sean bioinequivalentes

The cestode Echinococcus granulosus, the agent of hydatidosis/echinococcosis, is remarkably well adapted to its definitive host. However, the molecular mechanisms underlying the successful establishment of larval worms (protoscoleces) in the dog duodenum are unknown. With the aim of identifying molecules participating in the E. granulosus-dog cross-talk, we surveyed the transcriptomes of protoscoleces and protoscoleces treated with pepsin at pH 2. This analysis identified a multigene family of secreted monodomain Kunitz proteins associated mostly with pepsin/H+-treated worms, suggesting that they play a role at the onset of infection. We present the relevant molecular features of eight members of the E. granulosus Kunitz family (EgKU-1 – EgKU-8). Although diverse, the family includes three pairs of close paralogs (EgKU-1/ EgKU-4; EgKU-3/EgKU-8; EgKU-6/EgKU-7), which would be the products of recent gene duplications. In addition, we describe the purification of EgKU-1 and EgKU-8 from larval worms, and provide data indicating that some members of the family (notably, EgKU-3 and EgKU-8) are secreted by protoscoleces. Detailed kinetic studies with native EgKU-1 and EgKU-8 highlighted their functional diversity. Like most monodomain Kunitz proteins, EgKU-8 behaved as a slow, tight-binding inhibitor of serine proteases, with global inhibition constants (KI *) versus trypsins in the picomolar range. In sharp contrast, EgKU-1 did not inhibit any of the assayed peptidases. Interestingly, molecular modeling revealed structural elements associated with activity in Kunitz cation-channel blockers. We propose that this family of inhibitors has the potential to act at the E. granulosus-dog interface and interfere with host physiological processes at the initial stages of infection
