A summary of the resource.
Bioavailability and bioequivalence evaluations of drug products carried out using the experimental maximum concentration (Cmax) and the experimental time to reach Cmax (Tmax), are not advisable for slow-release formulations and for trials performed with saliva as biologic fluid. When slow-release curves are considered the drug concentration profiles usually show multiple peaks, making it difficult to compute a Cmax, Tmax value. The saliva profile throughout time shows a high variability observed as more than one peak in the saliva concentration versus time curves. In both cases, even if there is a major peak, when the statistical analysis of the data is performed, an important variability in Cmax results in high values in the residual variance of the ANOVA test. Consequently, the power of the bioequivalence test decreases and sometimes it is not possible to conclude on bioequivalence. The average concentration (Cav), the average maximum concentration (Cmax, av) and Cmax,av/Cav x 100 (%Cmax,av) are proposed in this paper as possible parameters in order to evaluate the profile of the concentration-time curves, as they reduce the residual variability in bioequivalence studies.

In case a Cmax value is computed, important variability in this parameter results in high values in the residual variance of the ANOVA test. A 30–33 kDa electroeluted fraction of T.gondii tachyzoites improved discrimination between acute and chronic phase sera when used instead of the whole tachyzoite extract in an avidity-ELISA. In order to identify the components of these fractions, crude tachyzoite antigen was fractionated by anionic exchange chromatography. The 30–33 kDa antigen cluster eluted in the not-bound fraction could account for a large proportion of the antibody response against the 30–33 kDa electroeluted fraction. According to the N-terminal sequence data, this antigen fraction is composed mainly of SAG1 and another protein with high homology to chitin binding proteins from plants.

A primary cause of food waste in households is a lack of food capabilities. Yet, the antecedents of food capabilities and its relationship with causes of food waste across different situational contexts remain poorly understood. We apply self‐efficacy theory in combination with convenience orientation and good provider identity in a mixed‐methods study. Findings reveal that the sequential structure of self‐efficacy explains how capabilities contribute to food waste avoidance. A good provider identity, however, increases food waste, and also turns out to further convenience orientation. Food capabilities, in turn, reduce convenience orientation. Qualitative insights shed light on consumer's own accounts of their abilities. The findings contribute to a theory‐based understanding of how consumers manage the complex issue of food waste. Findings imply that public policy and marketing should aim to strengthen consumers' self‐efficacy beliefs and redefine the good provider identity, in particular in social contexts, to reverse its effect on waste.

A summary of the resource.
This study is an interdisciplinary research into Uruguayan ayahuasca users belonging to one neoshamanic and one Santo Daime group. The study involved the chemical analysis of ayahuasca samples, an ethnographic description of the two traditions and rituals, and the application of psychometric scales to measure personality differences, and the acute psychological effects during an ayahuasca ritual. Personality measurements showed lower scores for Santo Daime in Neuroticism-Anxiety, Dependence, Low Self-Esteem, Anger and Restlessness. These differences may be related to the presence of participants under treatment in the neo-shamanic group and/or to the protective effects of a church religion such as Santo Daime. Regarding acute effects, the neoshamanic group showed higher scores in Somesthesia and Perception, which can be related to the high-arousal ritual setting. Chemical analysis for the ayahuasca samples showed a typical composition of alkaloids. No adulterants were found. The sample from the neo-shamanic group displayed a higher β-carbolines:DMT ratio compared to the Santo Daime sample, which could be related to the higher effects observed for Somesthesia for the neo-shamanic group. Significant positive correlations between some personality traits and acute effects were found only in the neoshamanic group, which may be related to the more individualistic approach of this tradition.

Abstract
A summary of the resource.
Aza and oxo Diels–Alder reactions using enantiopure cis-cyclohexadienediols were studied. These dienediols were obtained from the biotransformation of monosubstituted arenes using bacterial dioxygenases (toluene and benzoate dioxygenases). Ethyl glyoxylate and its N-tosyl imine were used as dienophiles to afford the corresponding hetero Diels–Alder bicyclic adducts with excellent regio- and stereoselectivities. Quantum chemical calculations at the B3LYP/6-31+G(d,p) level of theory were performed to rationalize the observed selectivities especially the stereochemical aspects of the cycloadditions. The synthetic importance of these adducts is highlighted for the preparation of enantiopure 2,2,3,4,5,6-hexasubstituted piperidine and tetrahydropyran from toluene

Azospirillum sp promotes the growth of many important crop plants. We demonstrated lectin binding activity in outer-membrane protein extracts of A. brasilense Sp7 by hemagglutination assays. The lectin specifically recognised the exopolysaccharide (EPS) produced by aggregated cells. Affinity chromatography using EPS-Sepharose was used to identify a 67 kDa outer-membrane lectin (OML) that recognised a binding region in the extracellular polysaccharide. Results show the specific recognition and binding between EPS and OML. The potential relationship between cell-to-cell aggregation and the OML–EPS interaction is discussed.

<p>Resumen<br />Se estudió la capacidad inhibitoria para tripsina y elastasa de extractos de vegetales autóctonos: flores de Achyrocline satureioidehojas (marcela), hojas de Baccharis trimera (carqueja), frutos de Eugenia uniflora (pitanga) y frutos Schinus molle (anacahuita). Los extractos, que presentaron diferentes propiedades, se obtuvieron utilizando agua o etanol como solventes en distintas condiciones de tiempo y temperatura. La acción inhibitoriapara elastasa fue baja (< 30%) o casi nula en los extractos ensayados. Para la tripsina, todos los extractos presentaron acción inhibitoria, observándose para un mismo material vegetal importantes diferencias según el solvente utilizado, independientemente de la temperatura de extracción. Los extractos de S. molle fueron los de mayor actividad inhibitoria para tripsina. Al extracto acuoso de este vegetal, obtenido a 100 °C, se le determinó IC 50y tipo de inhibición.</p>
The inhibitory capacity of trypsin and elastase of extracts prepared from Achyrocline atureioidehojas flowers, Baccharis trimera leaves, Eugenia uniflora fruits and Schinus molle fruits was studied. The extracts showed differences for the same plant material depending on using water or ethanol as solvents, and were obtained under different time and temperature conditions. The elastase inhibitory action was low (< 30%) or almost absent in the extracts tested. For trypsin, all extracts showed inhibitory action and significant differences depending on solvent used, regardless of the extraction temperature. Extracts of S. molle showed the most potent inhibitory activity for trypsin. The aqueous extract of this plant obtained at 100 °C was selected to determine IC 50 and inhibition type.

A pesar de los grandes avances en la quimioterapia de las enfermedades infecciosas estas son aún una de las principales causas de muerte en el mundo. Aunque parece existir una gran gama de fármacos antibacterianos y antifúngicos de uso clínico, la aparición de microorganismos resistentes los hace a veces ineficaces. Entre los patógenos más problemáticos clínicamente relevantes en la actualidad el Staphylococcus aureus meticilino resistente (MRSA) se ubica como una de las bacterias más difíciles de tratar. La importancia de las enfermedades de plantas en la agricultura moderna es un hecho ampliamente documentado y reconocido. Las enfermedades tienen el potencial de destruir enteramente las cosechas o reducir en forma crónica el rendimiento de la mayoría de los cultivos, obligan a tomar medidas de combate que aumentan los costos de producción, y afectan la calidad y la durabilidad de los productos cosechados. Alrededor de dos tercios de las enfermedades de las plantas son causadas por hongos. La utilización intensiva de compuestos antimicrobianos en la agricultura industrial favorece la aparición de resistencias. Esto, unido a cambios frecuentes en los límites de aplicación, límites de residuos en el ambiente y en el producto, lleva a que este sea un sector altamente dinámico con una rápida rotación de los compuestos activos. Los hongos comparten con las plantas y los animales ciertos agentes infecciosos tales como bacterias y hongos patógenos y por lo tanto tienen el potencial de producir metabolitos con actividad farmacológica de uso en humanos y cultivares. Los “microhongos” son una fuente ya establecida como importante de compuestos antimicrobianos. Sin embargo, para los “macrohongos” (aquellos cuyo cuerpo de fructificación es visible a simple vista, la gran mayoría perteneciente al filo Basidiomycota) sólo unos pocos compuestos aislados se encuentran en la etapa de ensayos clínicos y de campo. 2 El presente trabajo apuntó a la enorme diversidad metabólica de los microorganismos, utilizando basidiomicetes como fuente original. Para eso se abordaron dos estrategias diferentes, el trabajo con los micelios y caldos extracelulares de cultivos fúngicos y el trabajo con los cuerpos de fructificación. A partir de cultivos de diecinueve hongos basidiomicetes se prepararon cincuenta y tres extractos que posteriormente fueron sometidos a ensayos biológicos. Cuarenta por ciento de los extractos presentaron actividad frente a los patógenos humanos evaluados y diecisiete por ciento de los extractos presentaron actividad frente a los fitopatógenos evaluados. Para la cepa más prometedora, se estudió la influencia de variables en las condiciones de cultivo sobre la producción de metabolitos de interés para obtener cantidades de extracto suficientes para avanzar en el aislamiento de los compuestos responsables de la actividad antimicrobiana. Considerando que los resultados obtenidos no fueron los esperados se recorrió el camino inverso, aislando compuestos con actividad antimicrobiana a partir de cuerpos de fructificación de Gymnopilus junonius y posteriormente induciendo la producción de los mismos en cultivos fúngicos de dicho hongo. El agregado de agentes epigenéticos influyó positivamente en la riqueza química de los extractos, no incidiendo en la producción de bisnoryangonina, compuesto aislado con promisoria actividad antimicrobiana frente a S. aureus meticilino resistente.

Las enzimas son biocatalizadores eficientes que se investigan para su uso a escala industrial. Según Business Communication Company (BCC) Research, se prevé que el mercado global de enzimas aumente de $ 5.01 mil millones en 2016 a $ 6.32 mil millones en 2021, con tendencias del mercado que predice un cambio hacia una mayor producción de enzimas para las industrias textiles, del papel, el cuero y las biorrefinerías (Chapman, Ismail y Dinu, 2018). Se estima que tales proyecciones de mercado son en gran medida impulsadas por el desarrollo de procesos en la producción enzimática de biocombustibles que han crecido en últimas décadas. Este interés está motivado por el enorme potencial que tiene la biomasa lignocelulósica para la generación de energía y bio-productos y la reducción de la dependencia de materias primas no renovables, lo que conduce a una reducción de las emisiones de gases de efecto invernadero (Silva, Vaz y Filho, 2018). Es en este contexto que surge el concepto de biorrefinería, en la que se busca agregar valor a la biomasa lignocelulósica. Sin embargo, la etapa de hidrólisis enzimática de la lignocelulosa está en desarrollo ya que existen dificultades operacionales, como resultado el proceso general es muy costoso y se refleja en el precio del producto, lo que reduce la competitividad en el mercado. Por lo tanto, aún se necesita mucha investigación para mejorar la viabilidad operacional y financiera de este proceso. Una de las vías es mejorar la etapa de hidrólisis enzimática a través de la eficiencia de la enzima y la reducción del costo de producción de ésta (Kumar y Sani, 2017). En particular, dentro del sistema de enzimas degradadoras del material lignocelulósico, las xilanasas y celulasas tienen una importancia significativa en esta y otras industrias. En la presente tesis se contribuyó a la valorización de residuos lignocelulósicos mediante la aplicación de enzimas xilanasa y celulasa producidas a partir de microorganismos nativos. De acuerdo a los objetivos propuestos en esta tesis, se realizaron aislamientos de microorganismos a partir de diferentes partes (ramas, corteza y hojas) de un árbol de Eucalyptus globulus. Sobre todos los microorganismos aislados (bacterias y hongos) se realizó un cribado cualitativo de actividades xilanasa y celulasa. Luego de seleccionar los microorganismos que se mostraron positivos a estas actividades se llevó a cabo un análisis cuantitativo de las actividades xilanasa y celulasa producidas en los microorganismos aislados. De esta forma se seleccionaron los microorganismos que presentaron una mayor actividad enzimática. Estos microorganismos fueron identificados mediante observación macroscópica y microscópica y análisis moleculares. Posteriormente, se realizó un estudio de la variación de la producción de las actividades xilanasa y celulasa en cada microorganismo seleccionado a través del tiempo de incubación y utilizando diferentes medios de cultivo. Con los resultados obtenidos se profundizó en el estudio de la actividad xilanasa producida por una cepa de Bacillus, una de Pseudozyma y una de Penicillium. Para ello se realizó la purificación parcial de las enzimas a partir de los cultivos de los microorganismos. Las xilanasas parcialmente purificadas se caracterizaron en relación a su temperatura óptima, pH óptimo y en cuanto a los productos de hidrólisis del xilano de haya. Por último se analizó la hidrólisis de corteza deslignificada de Eucalyptus dunnii mediante las xilanasas parcialmente purificadas producidas por Bacillus sp. EBV 12-22 y Pseudozyma sp. EBV 97-87. El material vegetal resultante se estudió mediante microscopía electrónica de barrido. También se realizó el análisis químico del mismo y del sobrenadante con el fin de evaluar los productos de biotransformación obtenidos a partir de la hidrólisis enzimática y alcanzar de esta forma datos para una posible aplicación de las enzimas. A partir de los resultados obtenidos en este estudio se procedió a evaluar la aplicación de la enzima xilanasa producida por la levadura Pseudozyma sp. EBV 97-87. Para ello se realizó la hidrólisis enzimática de la corteza deslignificada y la xilosa obtenida como principal producto de hidrólisis se utilizó para la producción de xilitol mediante la fermentación por una levadura, Wickerhamomyces anomalus Z1. Por otro lado el material remanente de la hidrolisis con xilanasa se utilizó para la estudio de la producción de etanol B2G. Los resultados obtenidos en la presente tesis permiten contribuir al desarrollo de conocimiento, técnicas biotecnológicas y de análisis para ser aplicados sobre diferentes biomasas lignocelulósicas en búsqueda de su valorización mediante la obtención de productos de mayor valor agregado.

Introduction: Baccharis dracunculifolia DC. (Asteraceae), called alecrim-do-campo, vassoura, or vassourinha, is the precursor of green propolis that is used in traditional medicine for treating conditions such as dyspepsia, fatigue, inflammation, mild fever, and high blood pressure. This study aimed to summarise botanical, chemical, pharmacologic, and toxicologic reports about B. dracunculifolia and contribute new information on its anatomy and essential oil composition. Methods: This study reviewed international databases, querying keywords related to traditional use, phyto chemicals, biological activities (excluding animal studies), nanotechnology, clinical trials, and toxicity. New contributions regarding micromorphology of leaves and stems were analysed by light and scanning electron microscopy, and crystals were observed by polarised microscopy, energy dispersive X-ray, and Raman troscopies. Essential oils and hydrolates were generated from distinct sites and analysed using gas graphy/mass spectrometry. Results: Botanical, chemical, pharmacologic, and toxicologic studies of B. dracunculifolia are reviewed herein along with this plant’s use in nano- and biotechnological applications. Major compounds present in the essential oils and volatile extracts of B. dracunculifolia include monoterpenes and sesquiterpenes, while the organic tracts are rich in phenolics (flavonoids, caffeoylquinic, and prenylated acids) and terpenoids (diterpenoids and triterpenoids). Conclusions: Previously reported chemical and pharmacologic studies of B. dracunculifolia provided solid formation about the pharmacologic effects of this medicinal plant and the green propolis produced from it. New information reported here on the micromorphology and phytochemistry (essential oil and hydrolate tion) of B. dracunculifolia can be used to facilitate the identification of this plant in comparison to other related species and for quality control.

<p>Baccharis dracunculifolia DC. (Asteraceae) is an aromatic plant species widely distributed across and along South American countries where it grows in different environments, even being considered, in some cases, as a hard to control weed. Its essential oil is highly appreciated in the fragrance/perfumery industry by its woody, foral, and green notes; mainly attributed to (E)-nerolidol and other sesquiterpene alcohols. Recent research has demonstrated a wide range of applications for the essential oil besides its aromatic properties. Although B. dracunculifolia cannot be considered as a medicinal plant for most of the indigenous communities in South America (except for Brazil), modern pharmacological research has demonstrated its high potential for the development of new therapeutic drugs, mainly with antiulcer, anti-infammatory, antiparasitic and anticancer effect. The special literature regarding B. dracunculifolia is profuse and thus, this chapter attempts to summarize the principal knowledge about this species.</p>
