Clostridium sordelli veterinary vaccine is made by cultivating the microorganism in large industrial bioreactors and purifying the toxin from culture supernatants. The harvesting time is currently established as a function of bacterial growth, but this time is not always associated with the maximum concentration of toxin present in the supernatant. As a result, it is simple to determine the total amount of toxin produced from batch to batch. Furthermore, the toxin concentration is currently measured using in vivo LD50 methods, and results can be obtained only after 72 hours, when the bioreactor has already been stopped. Moreover, the method is time-consuming and necessitates the use of a large number of experimental animals. The development and application of a latex agglutination reagent for semi-quantification of Clostridium sordelli lethal toxin (TcsL) in industrial bioreactors are described. The reagent was developed and characterised in our laboratory, with a detection limit of 8ng of toxin per ml of culture supernatant, and then validated in actual industrial conditions. The use of such a speedy (i.e., in minutes) and simple reagent will allow for real-time monitoring of the culture, standardising the optimal end-point for harvesting in terms of toxin quantity. As a result, the efficiency of TcsL industrial production may be improved immediately. The demand for modern materials, especially glasses, used in different applications, such as radiation sensors and spectral converters, requires a detailed study of their properties. The incorporation of fluoride compounds in borate glasses and their crystallization at the nanometric scale allows the properties of these materials to be further enhanced. Although many works showed improvements in some of these properties, some critical aspects, such as the crystallization mechanism and the role of the fluorine phase, need more investigation. We worked with xNaF (100 - x)BaO.2B2O3 glasses with x = 0, 5, 10, 15, 20, 25, 30, and 35% (in mol) to increase the knowledge in this field. The structural modifications and the thermally stimulated luminescence of the glasses were studied, and their crystallization was analyzed by thermal analysis and X-ray diffraction. A continuous trap distribution was found, which was responsible for its very good luminescent signal, especially in glasses with 20% NaF. By selecting a suitable amount of NaF, it is possible to obtain nanocrystals of BaF2. These promising results we reached show the applicability of these materials. There is great interest in the study of the optical properties of materials such as glasses, due to their possible technological applications and from a fundamental point of view. For this reason, an attempt has been made to advance the understanding of the existence of optical band gap in this type of material. In recent times, several ways of analyzing the dependence of the absorption coefficient with energy have been proposed. Using lead, strontium, and barium borate glasses as a model, this dependence has been analyzed. For this, we considered the glass structure and the corresponding crystal structure, and the value of the optical band gap energy has been discussed considering the theory of Mott and Davis and comparing with values of crystals and glasses of similar composition. Here, we used a correction method, that takes into account intrinsic defects, to obtain the values of optical band gap energies in the framework of a non-direct transition. We also include DFT simulations of the corresponding crystalline compounds and the band gap was evaluated. The absorption coefficient obtained from simulations shows the same trend that the experimental one. Here, we report a joint experimental and theoretical study using molecular dynamics calculations focusing on short- and long-range structure correlations with ionic transport near the glass transition for lithium-ion polyacrylonitrile- based electrolytes using DMSO as plasticizer. The presence of subtle modifications of the polymeric structural features near the glass transition (Tg) is rarely reported in the literature and could be relevant to study free volume in polymer-based electrolytes. Our in-situ small angle X-ray scattering (SAXS) analysis at different temperatures evidence an increment of free volume and partial loss of long-range coherence between polyacrylonitrile neighbor chains just above Tg in strong correlation with an Arrhenius behavior crossover observed for the ionic transport in these polyacrylonitrile-based electrolytes. Advancing the understanding of lithium-ion conductivity Arrhenius behavior for polymeric electrolytes with Tg slightly above room temperature could be of great relevance due to the overlap with the operating temperature range of lithium-ion batteries. The impact of the crystal size of the template zeolite on the textural properties of resulting Zeolite Templated Carbons was studied using nano- (20 nm) and micron-sized (5 μm) zeolite beta crystals. ZTCs were obtained for both nano- and micron-sized zeolite beta crystals, yet for the micron-sized sample the presence of supermicropores and small mesopores was detected, indicating a higher degree of structural defects during the templating process and hence in the final ZTC. The insights gained from nitrogen physisorption were confirmed by TEM, TGA and XPS. Despite, long range structural order, evidenced by X-ray powder diffraction (XRD) for both, nano- and micron-sized ZTCs was inferred. These results suggests that XRD patterns allow to evidence the well-ordered part of the sample but might not be the method of choice for characterizing the structural quality of ZTCs. Simulated XRD powder patterns allowed to propose a graphene-based nanoribbon structure grown in the zeolite pores as a satisfactory model for ZTCs in beta zeolite. Combining metallomics, proteomics, and transcriptomics allows the identification of multiple molecular targets and several parasitic metabolic pathways affected by V(IV), Pt(II), and Pd(II) potential antiparasitic drugs. Specifically, metallomics studies with Pd(II) and Pt(II) analogous compounds show higher parasite uptake of the Pt(II) than Pd(II), and both accumulate similarly in the parasite DNA fraction. Global warming has a strong impact on polar regions. Particularly, the Antarctic Peninsula and nearby islands have experienced a marked warming trend in the past 50 years. Therefore, higher methane (CH4) emissions from this area could be expected in the future. Since mitigation of these emissions can be carried out by microbial oxidation, understanding this biological process is crucial since to our knowledge,no related studies have been performed in this area before. In this work, the aerobic CH4 oxidation potential of five freshwater lake sediments of Fildes Peninsula (King George Island, South Shetland Islands) was determined with values from 0.07 to 10 mmol CH4 gdw-1 day-1 and revealed up to 100-fold increase in temperature gradients (5, 10, 15, and 20�C). The structure and diversity of the bacterial community in the sediments were analyzed by next-generation sequencing (Illumina MiSeq) of 16S rRNA and pmoA genes. A total of 4,836 ASVs were identified being Proteobacteria, Actinobacteriota, Acidobacteriota, and Bacteroidota the most abundant phyla. The analysis of the pmoA gene identified 200 ASVs of methanotrophs, being Methylobacter Clade 2 (Type I, family Methylococcaceae) the main responsible of the aerobic CH4 oxidation. Moreover, both approaches revealed the presence of methanotrophs of the classes Gammaproteobacteria (families Methylococcaceae and Crenotrichaceae), Alphaproteobacteria (family Methylocystaceae), Verrucomicrobia (family  Methylacidiphilaceae), and the candidate phylum of anaerobic methanotrophs Methylomirabilota. In addition, bacterial phospholipid fatty acids (PLFA) biomarkers were studied as a proxy for aerobic methane-oxidizing bacteria and confirmed these results. Methanotrophic bacterial diversity was significantly correlated with pH. In conclusion, our findings suggest that aerobic methanotrophs could mitigate in situ CH4 emissionsin a future scenario with higher temperatures in this climate-sensitive area. This study provides new insights into the diversity of methanotrophs, as well as the influence of temperature on the CH4 oxidation potential in sediments of freshwater lakes in polar regions of the southern hemisphere. Classic acetyl thioester protection/deprotection methodologies are widely used in organic synthesis, but deprotection step usually requires harsh conditions not suitable for labile substrates. In this work, a new method for thioester deprotection using a thiotransesterification approach is described. Firstly, thioglycolic acid (TGA) was identified as a good deprotecting reagent in solution. In order to develop a thiol polymer-supported reagent, TGA was anchored to a PEG-based resin through an amide bond (TG-NCOSH). Both homogeneous and heterogeneous approaches were conveniently carried out at room temperature, in aqueous buffer at pH 8. The mild conditions were suitable for alkyl and phenyl thioesters. Moreover labile thioesters containing thiazolidine and oxazolidine scaffolds, bearing amine, ester and acetal functionalities were also deprotected. The polymer-supported TGA gave better deprotection yields compared to TGA in solution, yields ranging from 61 to 90%. The feasibility of the recovery and reuse of TG-NCO-SH reagent was explored, showing it can be reused at least five times without lossing the activity. Rice (Oryza sativa) is one of the most frequently produced cereals in the world. Rice bran (RB) is obtained as a by-product in the rice milling process. A part of the RB is used for oil extraction, obtaining defatted rice bran as a second by-product. The aim of this work was to analyze the emulsifying properties of soluble proteins fraction present in defatted rice bran concentrate (DRBC) in acidic and neutral conditions. Fine emulsions (prepared by high-speed and ultrasound homogenization) stabilized with soluble proteins obtained from DRBC showed a mean particle size lower than coarse emulsions (prepared by only high-speed homogenization) and a significantly lower degree of overall destabilization. Coarse emulsions showed sigmoidal destabilization profiles at pH 4.5 and 7.0, related to the existence of two populations with different particle sizes. In fine emulsions, both pH provided lower particle sizes and greater stability. After 24 h of quiescent storage, the coarse emulsions showed an increase of particle size that was not observed in fine emulsions. Both types of emulsions showed a Newtonian-type behavior. Fine emulsions showed higher viscosity values and higher lightness than coarse emulsions, consistent with the reduction of particle size and increase in the number of particles. The soluble proteins obtained from a waste of the rice industry can be used to obtain stable fine oil-in-water emulsions in acidic and neutral conditions. This result is of interest since it could be used as a food ingredient, increasing the added value of this important by-product. Alpeorujo is a semi-solid residue from the olive oil industry that accounts for around 80–85% of the total processed olives; it contains phenolic compounds that could be used as natural preservatives in the food industry. The objective of this study was to optimize supercritical fluid extraction (SFE) of antioxidant compounds from alperujo using a Box-Behnken Design and response surface methodology. Antioxidant (ABTS and ORAC assays) and antimicrobial (MIC) potential of SFE extracts from Arbequina and Coratina were compared for extracts obtained in the optimal conditions to maximize the antioxidant activity (200 bar, 60 ◦C and 10% ethanol as modifier). Extracts from Coratina presented a significantly higher (p < 0.05) TPC (1487–2073 mg GAE/kg) and tocopherol content (345–454 ppm), although, in general, a correlation between these values and antioxidant activity of extracts was not observed. Sturgeons are chondrostean fish critically endangered due to anthropogenic loss and degradation of natural habitat and overfishing for meat and caviar production. Consequently, sturgeon aquaculture has extensively developed lately, being Russian sturgeon (Acipenser gueldenstaedtii) the second most important species reared for caviar production. However, Russian sturgeon aquaculture in subtropical countries, such as Uruguay, confronts difficulties because fish have to endure excessive summertime warm temperatures, which weaken their innate defences facilitating opportunistic infections. To address this problem, we look for identifying putative acute phase proteins (APPs), which might be robust serum biomarkers of both infection and chronic thermal stress, applied to monitoring Russian sturgeon health status in farms. We focused on the C-Reactive Protein/Serum Amyloid P (CRP/SAP) pentraxin since the pentraxin family includes well-known APPs, better characterised in mammals than fish. We identified A. gueldenstaedtii CRP/SAP (AgCRP/SAP), as a member of the universal CRP/SAP pentraxin sub-family, and studied AgCRP/SAP involvement in sturgeon response to bacterial challenge and chronic thermal stress, in comparison with A. gueldenstaedtii Serum Amyloid A (AgSAA), a previously described positive APP. Results showed that AgCRP/SAP is a constitutive serum component that remained constant upon Aeromonas hydrophila challenge and chronic thermal stress. Contrastingly, serum AgSAA was subjected to regulation by bacterial and thermal stress challenges, showing a 50-fold increase and 3-fold decline in serum levels, respectively. Overall, results highlight the potential value of AgSAA, but not of AgCRP/SAP, as a biomarker of bacterial infection and the need to continue searching for robust chronic thermal stress biomarkers in sturgeons. Herein, we studied the adsorption of 15 organic and three polar molecules onto pristine and fluorinated graphene. Two levels of fluorination were selected, CF fluorographene for which all carbon atoms are fluorinated and C4F fluorinated graphene, which maximizes the number of Clar sextets. Five different density functionals were employed: M06-L, M06-2X, PBE-D2 and PBE-D3BJ. In contrast with recent experimental investigations, we found that the adsorption energies for organic molecules onto perfect graphene are significantly lower than those determined for fluorographene. This situation is exacerbated when enthalpic and free energy corrections are included. However, a different scenario was found for polar molecules such as H2O, H2O2 and HF. Partial fluorination following a C4F pattern leads to significantly lower adsorption energies for these three molecules, and the superhydrophilicity of C4F is maintained after thermodynamic corrections are included. In fact, even the removal of a single fluorine atom results in a dramatic change in the adsorption energy of water on fluorographene. We discuss the performance of the density functionals assayed and the importance of using finite and infinite models. This work demonstrates that it is possible to adjust the adsorption energy of a given polar molecule onto graphene by controlling the fluorination level and in doing so, that partially fluorinated graphene models may open new avenues for the separation of molecules with different polarity. Using first-principles calculations we predict that the electronic properties of graphene buffer layer (BL) on silicon carbide (SiC) can be modified by defect-induced itinerant states. Band structures and effective masses of single vacancy (SV), divacancy (DV) and Stone-Wales (SW) defective BL were calculated. Structural reconstruction at the vicinity of defects as well as spin polarization of silicon dangling bonds in the top bilayer SiC and nearby carbons from the BL, give rise to energetically degenerate magnetic states displaying electronic properties completely apart. This is particularly true for the SW and SV defective models where as many as three different magnetic states, turned out to be degenerate, either displaying a semiconductor nature or becoming half-metallic or even metallic. This is in contrast with previously reported results for the perfect BL (PBL) which suggested that most stable degenerate magnetic configurations share a semiconductor nature. On the other hand, for the DV system, the introduction of two vacancies in the BL causes loss of magnetism whereas a band gap of 0.54 eV is opened. Hole effective masses decay upon removal of one and two carbon atoms in the SV and DV systems where an increase in mobility is conceivable, provided current direction is carefully selected. Baccharis dracunculifolia DC. and Baccharis microdonta DC. (Asteraceae) are woody species morphologically similar growing in Uruguay, where not taxonomists people often confuse them in field conditions. As the essential oil of B. dracunculifolia (‘vassoura’ oil) is highly prized by the flavor and fragrance industry, the correct differentiation of the two species is a key factor in exploiting them profitably and reasonably. To differentiate both Baccharis species, in this work their volatile expression profiles were studied as an alternative tool to determine authenticity and quality. Volatile organic compounds (VOCs) were monthly extracted during an entire year from aerial parts of wild populations by simultaneous distillation extraction (SDE), and studied by gas chromatography/mass spectrometry (GC/MS; identification) and conventional gas chromatography (GC-FID; component abundances determination). Enantioselective gas chromatography/mass spectrometry (Es-GC/MS) was applied in the search of parameters able to ensure genuineness of each species extract. Qualitative VOCs profiles were found to be similar for both species, being β-pinene, limonene, spathulenol, caryophyllene oxide, and viridiflorol the main components. However, the abundance of those VOCs were two to ten times higher in B. dracunculifolia than in B. microdonta during the year of study. These Baccharis spp. showed species-specific patterns of VOCs expression according to the seasonality, and interestingly, oxygenated compounds (transpinocarveol and myrtenal) increased their abundances at full-flowering stages. The enantiomeric distribution of selected monoterpenes (α- and β-pinenes, limonene, linalool, terpinen-4-ol, and α-terpineol) presented differential values for both Baccharis spp., meaning that Es-GC might be a useful tool for differentiating chemically both species in Uruguay for genuineness determination purposes. The novel methodologies, based on the use of a microwave- assisted protocol (MW) or a sealed vessel conventionally heated reactor (SVR) are faster, more efficient and render higher yields (average increase in yield = 11% and 16%, respectively). For the solid state char- acterization, infrared spectra of the obtained compounds were analyzed with the aid of computational tools for signal assignation. In addition, a new crystal structure for the E isomer of compound 2d was obtained and a thorough analysis of the intra and intermolecular interactions was performed by Hir- shfeld surface analysis. On the other hand, the hybrids were characterized in DMSO-d 6 solution by 1 H and 13 C NMR experiments. All of them showed a set of minor signals neither ascribable to the products nor described in the literature. Variable temperature 1 H NMR experiments revealed that a single inter- conversion equilibrium was operative in the solution, having thermodynamic parameters compatible to those predicted for a Z/E isomerization process by a DFT model. The most stable configuration ( E ) was further confirmed by DFT, calculating the 13 C NMR chemical shifts for both configurations. The compu- tational model shows that the E configuration is stabilized through the formation of a non-conventional C-H ···O = C intramolecular H-bond between the methyl and carbonyl groups, adopting a conformation less sterically strained. These findings allow to gain insights into the E / Z isomerization equilibria displayed by thiosemicarbazones in solution, a phenomenon usually under described in literature, which seems to be promoted by a balance between weak intramolecular interactions that modulate the relative isomeric stability in solution. Despite being a preventable and curable disease, Tuberculosis (TB) is the world's top infectious killer. Development of new drugs is urgently needed. In this work, the synthesis and characterization of new silver(I) complexes, that include N′ -[(E)-(pyridine-2-ylmethylene)pyrazine-2-carbohydrazide, HPCPH, as main ligand and substituted aryl-phosphines as auxiliary ligands, is reported. HPCPH was synthesized from pyrazinoic acid, the active metabolite of the first-line antimycobacterial drug pyrazinamide. Complexes [Ag(HPCPH)(PPh3)2]OTf (1), [Ag(HPCPH)((P(p-tolyl)3)2]OTf (2) and [Ag(HPCPH)(P(p-anisyl)3)2]OTf (3) were characterized in solid state and in solution by elemental analysis and FTIR and NMR spectroscopies (OTf––triflate). Crystal structures of (1,2) were determined by XRD. The Ag atom is coordinated to azomethine and pyridine nitrogen atoms of HPCPH ligand and to the phosphorous atom of each aryl-phosphine co-ligand. Although HPCPH did not show activity, the Ag(I) compounds demonstrated activity against Mycobacterium tuberculosis (MTB), H37Rv strain, and multidrug resistant clinical isolates (MDR-TB). Globally, results showed that the compounds are not only effective against the sensitive strain, but are more potent against MDR-TB than antimycobacterial drugs used in therapy. The compounds showed low to moderate selectivity index values (SI) towards the bacteria, using MRC-5 cells (ATCC CCL-171) as mammalian cell model. Interaction with DNA was explored to get insight into the potential mechanism of action against the pathogen. No significant interaction was detected, allowing to discard this biomolecule as a potential molecular target. Compound 1 was identified as a hit compound (MIC90 2.23 μM; SI 4.4) to develop further chemical modifications in the search for new drugs. One of the biggest problems worldwide is the pollution of natural water bodies by dyes coming from effluents used in the textile industry. In the quest for novel effluent treatment alternatives, the aim of this work was to immobilize Fe(III) complexes in molecularly imprinted polymers (MIPs) to produce efficient Fenton-like heterogeneous catalysts for the green oxidative degradation of the methyl orange (MO) dye pollutant. Different metal complexes bearing commercial and low-cost ligands were assayed and their catalytic activity levels towards the discoloration of MO by H2O2 were assessed. The best candidates were Fe(III)-BMPA (BMPA = di-(2-picolyl)amine) and Fe(III)-NTP (NTP = 3,30,3”-nitrilotripropionic acid), displaying above 70% MO degradation in 3 h. Fe(III)-BMPA caused the oxidative degradation through two first-order stages, related to the formation of BMPAFe- OOH and the generation of reactive oxygen species. Only the first of these stages was detected for Fe(III)-NTP. Both complexes were then employed to imprint catalytic cavities into MIPs. The polymers showed catalytic profiles that were highly dependent on the crosslinking agent employed, with N,N-methylenebisacrylamide (MBAA) being the crosslinker that rendered polymers with optimal oxidative performance (>95% conversion). The obtained ion-imprinted polymers constitute cheap and robust solid matrices, with the potential to be coupled to dye-containing effluent treatment systems with synchronous H2O2 injection.
