The results of a study on the use of Hansch�s series design, cluster methodology, for the generation of new benzo[1,2-c]1,2, 5-oxadiazole N-oxide derivatives as antitrypanosomal compounds are described. In vitro activity of these compounds was tested against Tulahuen 2 strain of Trypanosoma cruzi. Clearly, the Hansch methodology allowed identifying two cluster-substituents suitable for further structural modifications. The most effective drugs, derivatives 11, 18, and 21, with 50% inhibitory concentration (IC50) of the same order as that of the reference drug, represent an excellent structural point of chemical modifications for the design of future drugs. Preliminary results from the study of the mechanism of action of these benzofuroxans point to perturbation of the mitochondrial electron chain, inhibiting parasite respiration. This study is based upon a recently established method for quantification of the antioxidant capacity of natural samples via a HPLC separation and a hyphenated selective detection (post-column derivatization with cupric reducing antioxidant capacity reagent) technique. This protocol demonstrated the main improvements to transform the quantitative protocol into a high-speed qualitative automated assay to screen samples for their potential total antioxidant capacity, typically performed via manual mixing of the sample and derivatisation and measured on a 96 well plate reader/bench top UV–Vis spectrophotometer. This approach with automated mixing is a more informative alternative for total antioxidant capacity as the antioxidant peaks are profiled for each sample within four minutes. This antioxidant profile may be used for routine analysis of raw materials and/or a guide for targeted approaches for structure elucidation for laboratories interested in early drug discovery, natural product research and the search of alternative antioxidant additives in consumer goods/therapeutics. Plant-herbivore interactions have been extensively studied in tomato plants and their most common pests. Tomato plant chemical defenses, both constitutive and inducible, play a role in mediating these interactions. Damaged tomato plants alter their volatile profiles, affecting herbivore preferences between undamaged and damaged plants. However, previous studies on tomato volatiles and herbivore preferences have yielded conflicting results, both in the volatile chemistry itself as well as in the attraction/repellent herbivore response. This study revisits the volatile-mediated interactions between tomato plants and two of their main herbivores: the leafminer Tuta absoluta and the whitefly Trialeurodes vaporariorum. Tomato plant volatiles were analyzed before and after damage by each of these herbivores, and the preference for oviposition (T. absoluta) and settling (T. vaporariorum) on undamaged and damaged plants was assessed both after conspecific and heterospecific damage. We found that both insects consistently preferred damaged plants over undamaged plants. The emission of herbivore-induced plant volatiles (HIPVs) increased after T. absoluta damage but decreased after T. vaporariorum damage. While some of our findings are in line with previous reports, T. absoluta preferred to oviposit on plants damaged by conspecifics, which differs from earlier studies. A comparison of HIPVs emitted after damage by T. absoluta and T. vaporariorum revealed differences in up- or down-regulation, as well as significant variations in specific compounds (12 for T. absoluta and 26 for T. vaporariorum damaged-plants). Only two compounds, β-caryophyllene and tetradecane, significantly varied because of damage by either herbivore, in line with the overall variation of the HIPV blend. Differences in HIPVs and herbivore preferences may be attributed to the distinct feeding habits of both herbivores, which activate different defensive pathways in plants. The plant’s challenge in simultaneously activating both defensive pathways may explain the preference for heterospecific damaged plants found in this study, which are also in line with our own observations in greenhouses. Four novel alkali−manganese(II) trifluoroacetates were synthesized, and their potential as self-fluorinating precursors to layered perovskites A2MnF4 (A = K, Rb, and Cs) was demonstrated. Na2Mn(tfa)4, K4Mn2(tfa)8, Rb4Mn2(tfa)8·0.23H2O, and Cs3Mn2(tfa)7(tfaH) (tfa = CF3COO− and tfaH = CF3COOH) were grown as single crystals, and their crystal structures solved using X-ray diffraction. Chemically pure K4Mn2(tfa)8, Rb4 Mn2(tfa)8·0.23H2O, and Cs3Mn2(tfa)7(tfaH) were also prepared in polycrystalline form as confirmed by thermal analysis and powder X-ray diffraction. Thermolysis of these powders yielded the isostructural series K2MnF4, Rb2MnF4, and Cs2MnF4 at low temperatures (≈200−300 °C). Trifluoromethyl groups belonging to the trifluoroacetato ligands served as the fluorine source, thereby eliminating the need for external fluorinating agents. K2MnF4 and Rb2MnF4 were obtained as single-phase powders, whereas Cs2MnF4 crystallized along with CsMnF3. Access to polycrystalline Cs2MnF4 coupled to Rietveld analysis enabled elucidation of the crystal structure of this ternary fluoride, which had remained elusive. This study investigated the in vitro bioaccessibility of aluminum, copper, iron, manganese, lead, selenium, and zinc in three important species of farmed insects: the yellow mealworm (Tenebrio molitor), the house cricket (Acheta domesticus) and the migratory locust (Locusta migratoria). Results show that all three insect species constitute excellent sources of essential elements (Fe, Cu and Zn) for the human diet, contributing to the recommended dietary allowance, i.e., 10%, 50%, and 92%, respectively. A higher accumulation of Se (≥1.4 mg Se/ kg) was observed with increasing exposure concentration in A. domesticus, showing the possibility of using insects as a supplements for this element. The presence of Al and Fe nanoparticles was confirmed in all three species using single particle-inductively coupled plasma-mass spectrometry and transmission electron microscopy. The results also indicate that Fe bioaccessibility declines with increasing Fe-nanoparticle concentration. These findings contribute to increase the nutritional and toxicological insights of farmed insects. This is the first study to employ a reaction flow (RF) HPLC column of a short 5 cm length to identify antioxidants in a variety of complex samples represented by tea, utilising simultaneous multiplexed detection techniques. The detection responses included: underivatised UV at 280 nm, underivatised fluorescence detection (FLD) and a post column derivatisation (PCD) colorimetric response for antioxidants via the ferric reducing antioxidant power (FRAP) assay. Both similarities and differences in the chromatograms obtained using the three detection modes highlight the power of multiplexed detection. The main findings from the bioactive profiling include: the Fruit of the Forest tea contains the lowest level of antioxidants and is clearly distinct from the other three teas which are based on Camellia sinensis. The antioxidants detected in the green tea and the two black teas are similar. The green tea contains a higher concentration of the two main antioxidants that eluted at 4.0 and 5.7 min. The simplicity of sample analysis and chemical profiling using RF columns and selective detection was the focus of this study via the analysis of antioxidants in tea using the FRAP reagent. The main advantage of RF-PCD is the ability to split flows for simultaneous detection with minimal post column dead volume contributions, making peak matching between detectors much easier. To date, few studies exploit the power of RF-PCD multiplexed detection to differentiate and identify bioactive compounds in various complex samples. Applications of the approach developed in this study may include antioxidant screening and profiling of natural products, foods and beverages. In the search for new therapeutic tools against tuberculosis and to further address the therapeutic potential of pyridine-2-thiol 1-oxide (Hmpo) metal complexes, two new octahedral [M(III)(mpo)3] complexes, with M = Ga or Bi, were synthesized and characterized in the solid state and in solution. Attempts to crystallize [Ga(III)(mpo)3] in CH2Cl2 led to single crystals of the reaction product [GaCl(mpo)2], where the gallium(III) ion is in a square basis pyramidal environment, trans-coordinated at the basis to two pyridine-2-thiolato 1-oxide anions acting as bidentate ligands through their oxygen and sulfur atoms. The biological activity of the new [M(III)(mpo)3] complexes together with that of the previously reported Fe(III) analogous compound and the pyridine-2-thiol 1-oxide sodium salt (Na mpo) was evaluated on Mycobacterium tuberculosis. The compounds showed excellent activity, both in the standard strain H37Rv ATCC 27294 (pan-susceptible) and in five clinical isolates that are resistant to the standard first-line anti-tuberculosis drugs isoniazid and rifampicin. These pyridine-2-thiol 1-oxide derivatives are promising compounds for the treatment of resistant tuberculosis. Synopsis<br />Introduction<br /><br />The use of bioadhesive hydrogels for skin care presents important advantages such as long residence times on the application site and reduced product administration frequency.<br />Object<br /><br />The aim of the present work was to develop bioadhesive hydrogels for skin application, using caffeine as a model active ingredient.<br />Methods<br /><br />Eight hydrogels were formulated using binary combinations of a primary polymer (carbomer homopolymer type C (Carbopol® 980) or kappa carrageenan potassium salt (Gelcarin® GP-812 NF)) and a secondary polymer (carbomer copolymer type B (Pemulen™ TR-1), xanthan gum or guar gum). Hydrogels were characterized by means of physico-chemical (dynamic rheological measurements, spreadability and adhesion measurements) and sensory methods (projective mapping in combination with a check-all-that-apply (CATA) question). Caffeine hydrogels were formulated using two of the most promising formulations regarding adhesion properties and sensory characteristics. In vitro active ingredient release studies were carried out.<br />Results<br /><br />Hydrogel formulations showed a prevalently elastic rheological behaviour. Complex viscosity of carbomer homopolymer type C hydrogels was higher than that of the kappa carrageenan hydrogels. Besides, complex viscosity values were dependent on the secondary polymer present in the formulation. Significant differences among hydrogels were found in detachment force, work of adhesion and spreading diameter results. Association of projective mapping with CATA allowed to determine similarities and dissimilarities among samples. Cluster analysis associated the samples in two groups. Two hydrogels were selected to study the release of caffeine. Both hydrogels presented similar release profiles which were well described by the Higuchi model. Caffeine release was exclusively controlled by a diffusive process.<br />Conclusion<br /><br />Physico-chemical and sensory techniques enabled the identification of bioadhesive hydrogel formulations with positive characteristics for cosmetic applications. Formulations which combined carbomer homopolymer type C with xanthan gum or with carbomer copolymer type B were the most promising for bioadhesive skin products. Caffeine release profiles of selected formulations were not statistically different. Both hydrogels gradually released the active ingredient, reaching approximately 80% within the first 5 h, and their profiles were well described by the Higuchi model. This work demonstrates the improvement of bioavailability when the ABZS is directly administered, compared with the ABZ administration, both orally given.
