A bioavailability study of one of these W/O emulsions was carried out choosing a conventional tablet commercialized in Uruguay as reference. A single-dose study was carried out in eight healthy volunteers in a crossover randomized test. Saliva and plasma samples were taken and analyzed by HPLC. Saliva/plasma ratios enable the observation of absorption inputs sometimes up to 48 hours after administraion. Plasma concentrations show sustained drug levels for the emulsion during the period studied. No significant differences were found for C max,T max and AUC 0-48 h. Similar bioavailability is concluded for both dosage forms. According to the hydrolysis performance "in vitro" and lipophylicity |b two Furosemide (F) prodrugs were chosen from a series of acyloxymethylesters of F synthesized previously: PI (acetyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate) and P4 (pivaloyloxymethyl-4-chloro-N-furfuryl-5-sulfamoylanthranilate). The bioavailability studies were assayed over two groups of eight male Wistar rats as a randomized two-way crossover and balanced design: group 1) a solution of PI in propylenglycol/ethyl acetate vs an aqueous solution of F |b and group 2) PI vs.P4 solutions in oleic acid (PI,P4).These assays showed a better absorption performance of PI and P4 than F |b while the two prodrugs showed a similar bioavailability. The oleic acid seems to be responsible for the delay in the recovery of 50 of the total amount of F excreted in urine (T50). When the monitoring is done in plasma after the administration of PI |b PI was not detected as circulating prodrug. The analytical determinations of F in urine and plasma were done by high performance liquid chromatography (HPLC). From the urinary excretion data |b a slope that indicates a slow elimination was found with a half-life of 12 hours approximately. Biocarbon monoliths were obtained from Eucalyptus grandis and the influence of wood anisotropy on the electrical and electrochemical performance as supercapacitor electrodes was studied. They were produced from wood pieces cut along the transversal and longitudinal direction of the tree trunk, followed by pyrolysis and, for some of them, also by activation with CO2. Monoliths with drilled channels were also obtained. All the monoliths were characterized by SEM, nitrogen adsorption/desorption isotherms, electrical conductivity measurements and electrochemical measurements, the latter in 2M aqueous H2SO4 electrolyte. Electrical conductivity and specific capacitance are higher for the transversal carbon monoliths than for the longitudinal ones. The electrical conductivity reaches values up to 27 S cm−1 for the transversal monolith. The specific capacitance reaches values up to 260 F g−1 for the transversal monolith that was activated and drilled. However, the highest volumetric capacitance, of 90 F cm−3, is found for the longitudinal monolith that was activated and non-drilled. The energy density and power density, both referred to gravimetric and volumetric magnitudes, reach values as high as 36 Wh kg−1 and 12 Wh L−1, and 2181 W kg−1 and 783 W L−1, respectively. Habiendo nacido en una época en donde los antibióticos están ampliamente disponibles y son de fácil acceso, es fácil asumir que antes de la introducción de los mismos a la clínica, cualquier persona con una infección eventualmente perecería por esa causa. Sin embargo, siempre ha habido sobrevivientes incluso de las infecciones más letales, gracias a la eficiencia de la respuesta del sistema inmune innato. De todas maneras, la tasa de mortalidad en la era pre-antibióticos era muchísimo más alta que hoy en día. Sin embargo, la continua generación de resistencia bacteriana hace que hoy en día no se pueda dar por sentado el tratamiento de cualquier infección mediante el uso de agentes antimicrobianos. La emergente generación de resistencia a los principios activos disponibles se enfrenta mediante políticas de racionalización del uso de antibióticos y mediante la continua búsqueda de nuevos compuestos con actividad antimicrobiana. En el presente trabajo, se describen los esfuerzos realizados para la síntesis de análogos simplificados del antibiótico higromicina A (HA). El mismo es un compuesto aislado a partir de Streptomyces hygroscopicus, y fue muy estudiado en la década del 90 debido a la excelente actividad que presenta in vivo frente al microorganismo responsable de la disentería porcina (B. hyodysentereae). Su estructura modular compuesta por tres subunidades lo hace un compuesto ideal para el estudio sistemático de la relación estructura-actividad. Es así que un grupo de la empresa Pfizer ha sintetizado diversos análogos, y uno de ellos en particular (CP-111906), que es un derivado de la molécula original, presentó la misma potencia que el antibiótico natural, pero tiene una complejidad estructural disminuida. En la presente tesis doctoral nos propusimos sintetizar nuevos análogos de HA, aplicando reacciones contempladas dentro del concepto “click chemistry”. Particularmente, se propuso sintetizar una serie de aminociclitoles y azidoinositoles a partir del bromociclohexadiendodiol enantioméricamente puro, obtenido a su vez a partir de una biotransformación de bromobenceno. Los aminociclitoles o azidoinositoles fueron posteriormente acoplados a los residuos aromáticos correspondientes, mediante un enlace amida o un anillo triazólico. Con los análogos sintetizados se pretende evaluar los efectos del reemplazo bioisostérico amida-triazol, el efecto de sustituir el metilenodioxo por un isopropilidenodioxo, y el efecto de cambiar la configuración absoluta del carbono que soporta el grupo funcional nitrogenado. Ninguno de estos efectos ha sido estudiado previamente. Fishes of the perciform suborder Notothenioidei afford an excellent opportunity for studying the evolution and functional importance of diverse types of biochemical adaptation to temperature. Antarctic notothenioids have evolved numerous biochemical adaptations to stably cold waters, including antifreeze glycoproteins, which inhibit growth of ice crystals, and enzymatic proteins with cold-adapted specific activities (kcat values) and substrate binding abilities (Km values), which support metabolism at low temperatures. Antarctic notothenioids also exhibit the loss of certain biochemical traits that are ubiquitous in other fishes, including the heat-shock response (HSR) and, in members of the family Channichthyidae, hemoglobins and myoglobins. Tolerance of warm temperatures is also truncated in stenothermal Antarctic notothenioids. In contrast to Antarctic notothenioids, notothenioid species found in South American and New Zealand waters have biochemistries more reflective of cold-temperate environments. Some of the contemporary non-Antarctic notothenioids likely derive from ancestral species that evolved in the Antarctic and later “escaped” to lower latitude waters when the Antarctic Polar Front temporarily shifted northward during the late Miocene. Studies of cold-temperate notothenioids may enable the timing of critical events in the evolution of Antarctic notothenioids to be determined, notably the chronology of acquisition and amplification of antifreeze glycoprotein genes and the loss of the HSR. Genomic studies may reveal how the gene regulatory networks involved in acclimation to temperature differ between stenotherms like the Antarctic notothenioids and more eurythermal species like cold-temperate notothenioids. Glutathione transferases (GSTs) are believed to be a major detoxification system in helminths. We describe the expression and functional analysis of EgGST, a cytosolic GST from Echinococcus granulosus, related to the Mu-class of mammalian enzymes. EgGST was produced as an enzymatically active dimeric protein (rEgGST), with highest specific activity towards the standard substrate 1-chloro- 2,4-dinitrobenzene (CDNB; 2.5�mol min−1 mg−1), followed by ethacrynic acid. Interestingly, rEgGST displayed glutathione peroxidase activity (towards cumene hydroperoxide), and conjugated reactive carbonyls (trans-2-nonenal and trans,trans-2,4-decadienal), indicating that it may intercept damaging products of lipid peroxidation. In addition, classical GST inhibitors (cybacron blue, triphenylthin chloride and ellagic acid) and a number of anthelmintic drugs (mainly, hexachlorophene and rafoxanide) were found to interfere with glutathione-conjugation to CDNB; suggesting that they may bind to EgGST. Considered globally, the functional properties of rEgGST are similar to those of putative orthologs from Echinococcus multilcularis and Taenia solium, the other medically important cestodes. Interestingly, our results also indicate that differences exist between these closely related cestode GSTs, which probably reflect specific biological functions of the molecules in each parasitic organism. Global warming is causing serious problems, especially, in warm regions, where musts with excess sugars and high pH produce wines with decreased freshness and unstable evolution. This study aimed to determine biocompatibility between yeast species, the capacity for microbiological acidification, and the aromatic profile produced in ternary fermentations in which Lachancea thermotolerans has been co-inoculated with Hanseniaspora vineae, Torulaspora delbrueckii, or Metschnikowia pulcherrima, and the fermentation process is subsequently completed with sequential inoculation of Saccharomyces cerevisiae. For this purpose, different cell culture media and instruments were used such as infrared spectroscopy, enzymatic autoanalyzer, chromatograph coupled with a flame ionization detector, spectrophotometric analysis, among others. The behavior of these yeasts was evaluated alone and in co-inoculation, always finishing the fermentation with sequential inoculation of S. cerevisiae, at a stable temperature of 16°C and with a low level of sulfites (25 mg/L) in white must. Significant results were obtained in terms of biocompatibility using population counts (CFU/ml) in differential plating media that permitted monitoring. Quantification of the five species was studied. Concerning acidification by L. thermotolerans in co-inoculations, we showed some metabolic interactions, such as the inhibition of acidification when H. vineae/L. thermotolerans were used, generating just over 0.13 g/L of lactic acid and, conversely, a synergistic effect when M. pulcherrima/L. thermotolerans were used, achieving 3.2 g/L of lactic acid and a reduction in pH of up to 0.33. A diminution in alcohol content higher than 0.6% v/v was observed in co-inoculation with the L. thermotolerans/M. pulcherrima yeasts, with total sugar consumption and very slow completion of fermentation in the inoculations with H. vineae and T. delbrueckii. The aromatic composition of the wines obtained was analyzed and a sensory evaluation conducted, and it was found that both L. thermotolerans and co-inoculations retained more aromatic esters over time and had a lower evolution toward the yellow tones typical of oxidation and that the best sensory evaluation was that of the Lt + Mp co-inoculation. Lachancea thermotolerans and co-inoculations produced wines with low levels of volatile acidity (<0.4 g/L). This work shows that good consortia strategies with binary and ternary fermentations of yeast strains can be a powerful bio-tool for producing more complex wines. Kefir granules are a complex microbial consortia with numerous properties, among which their antifungal action has been reported. This study characterized the antifungal activity of water kefir granules and two microorganisms isolated from them, identified as Saccharomyces cerevisiae and Geotrichum candidum, against aflatoxigenic Aspergillus flavus. Experiments were carried out in vitro and under minisilo conditions designed to simulate the aerobic phase of commercial corn storage. In dual culture plate assays, A. flavus was inhibited in close contact with both antagonists, and mycelial weakening with lower conidia production was observed. Cell-free supernatants of both antagonists inhibited conidial germination by 92% (G. candidum) and 65% (S. cerevisiae). Non-contact assays between A. flavus and antagonists indicated that volatile organic compounds (VOCs) also contributed to inhibition, reducing mycelial growth by 29.1% (G. candidum) and 37.9% (S. cerevisiae). In corn minisilo trials water kefir granules and both antagonists showed strong antifungal activity by day 4, with qPCR of A. flavus DNA and aflatoxin measurements indicating near complete inhibition. As mixed consortia, water-kefir granules inherently include lactic acid bacteria whose rapid acidification and diffusible metabolites complement antifungal compounds produced by other components of the consortia, supporting a multi-mechanistic mode of action. Given the compositional heterogeneity of kefir granules, strain-level identification and verification of safety and regulatory status for the intended use are essential prior to food or feed applications. The demonstrated antifungal efficacy of kefirderived microorganisms in mini-silos underscores their scale-up potential for postharvest protection of corn. The capacity of microorganisms from water kefir (WK) to control Aspergillus flavus growth during the aerobic phase of ensiled sorghum grains was determined. Sorghum inoculated with A. flavus was treated with filter-sterilized and non-sterilized water kefir, ensiled, and incubated 7 days at 25 °C. A. flavus growth was quantified by qPCR after incubation. Mold growth was inhibited in the presence of water kefir while no inhibition was observed when filter-sterilized water kefir was applied, demonstrating the relevant role of the microorganisms in the kefir water in the biocontrol process. Fungal and bacterial diversity in treated sorghum mini-silos was analyzed by high-throughput sequencing. Firmicutes was the predominant bacterial phyla and Lactobacillus represented the most abundant genus, while Ascomycota was the predominant fungal phyla with Saccharomyces and Pichia as the major genera. Bacterial and yeast counts before and after incubation indicated that the microbial community obtained from WK was able to grow in the sorghum mini-silos in the presence of A. flavus. Results of the present work indicate that the use of a mixed inoculum of microorganisms present in WK may represent an alternative management practice to avoid the growth of A. flavus in ensiled sorghum grains and the concomitant contamination with aflatoxins. El glicerol es un subproducto inevitable de la producción de biodiésel. Una gran cantidad de este producto es generado durante la transesterificación de grasas y aceites con alcohol. El exceso de oferta y la limitada demanda de este producto afectan directamente los costos de producción del biodiésel. Synthetic dyes in watercourses resulting from industrial effluent discharges cause serious ecological impacts, besides carcinogenic and mutagenic effects on human health. Thus, it is important to develop effective methods to remove the dyes from industrial wastewaters, and also to carry out adequate toxicity studies to establish their safety. Azo dyes are the main class of industrial dyes and important environmental contaminants. We have examined the decolorization of two azo dyes (Acid Red 88 and Acid Black 172) by a native Trametes villosa laccase immobilized on thiolsulfinate-agarose as well as the effect of different redox mediators in the reactions. The method was effective for the decolorization of both dyes. The immobilization method did not affect the capacity of the biocatalyst for dye degradation. Therefore, the insoluble enzyme removed 97% of the color of AR88 and 92% of AB172 in 24 hours at 22 °C in the presence of the selected redox mediators, vanillin (0.1 mM) and violuric acid (1.0 mM), respectively. In addition, the immobilized enzyme kept 78% of its initial capacity for decolorization of AR88 after three cycles of use. The ecotoxicological evaluation of the solutions showed a great variation depending on the biological systems used. In the phytotoxicity test, the decolorization products were not toxic to plants, whereas in Daphnia magna and Microtox® bioassays an acute residual toxicity was found. The last outcome shows the importance of using a battery of bioassays to determine the remaining ecotoxicity of the treated effluents before their discharge into the aquatic environment. Despite adverse conditions in Antarctica for life, organic materials can be successfully colonized by extremophilic microorganisms. This work describes the isolation of filamentous fungi from terrestrial and maritime habitats of King George Island, Antarctica, and the screening of their ability to produce antimicrobial compounds. A total of 44 samples were collected during the Antarctic expeditions from soil, sediments, terrestrial and marine waters and pieces of wood that had been brought into Antarctica. One hundred thirty-seven psychrotrophic fungi were isolated. Thirty-three strains selected from different samples and with different morphological characteristics were used in molecular identification and antimicrobial activity assays. The ability of all isolations to inhibit growth of different bacteria in vitro was tested using the agar plug diffusion method. Most of the fungi identified were Ascomycota with only a few Zygomycota and no Basidiomycota isolated. Twenty-six cultures corresponded to known species. Penicillium was the most represented genus of identified fungal isolations (24 %) followed by species of Cadophora, Mortierella, Pseudogymnoascus and Neurospora. This is the first report of the presence of Aspergillus pseudodeflectus in this region. Of the isolates under study, eighteen were able to inhibit growth of at least one of the bacterial strains used. In recent years, interest in non-Saccharomyces yeasts for the innovation and development of different and alternative beer styles has been increasing, especially for the microbrewing industry. This work studied the biodiversity of non-Saccharomyces yeasts based on isolates from grapes of different Uruguayan vineyards, craft breweries and raw materials, with the aim of selecting autochthonous non-Saccharomyces yeasts with a brewing attitude. Brewing tests were performed on synthetic wort developed for this purpose, and the evolution of alcoholic fermentation was monitored by measuring glucose, maltose, maltotriose consumption, ethanol and glycerol production and final sensory analysis. A total of two hundred seventy-one yeast strains belonging to different genera were evaluated according to these parameters. After evaluating alcoholic fermentation performance, a native yeast strain belonging to the species Starmerella meliponinorum was selected due to its high maltotriose consumption and glycerol production, making it a very promising brewing yeast, especially for production of low carbohydrate beers. The statistical evaluations of the pharmacokinetics parameters showed no significant differences |b either for the extent of absorption (AUC 0-48) or for the rate of absorption (as indicated by Cmax and Tmax) between both formulations. Significant differences were found between salivary concentrations 20 min.after the ingestion between both formulations. This was in agreement with the in vitro dissolution profiles.The mean elimination half-life for carbamazepine was found to be 28 hours+-11. Both formulations were bioequivalent according to Westlake's confidence interval of 20. It may be concluded from these results that saliva is useful for bioequivalence studies of carbamazepine using an appropriate sampling schedule. Several oral ivermectin (IVM) formulations for use in sheep are available in the pharmaceutical veterinary market in different countries. All of them are indicated at the same dose rate to treat the gastrointestinal nematodes. However, there is a lack of information on the relative systemic exposure (plasma bioavailability) and clinical efficacy among oral formulations routinely used in sheep. The main goal of the work reported here was to perform a pharmaco-parasitological assessment of three different IVM oral formulations in lambs infected with multiple resistant gastrointestinal nematodes. The comparative drug systemic exposure (IVM plasma concentrations) and nematodicidal efficacies (clinical efficacy) in lambs were determined for a reference (RF) and two different test (T1, T2) IVM oral formulations. One hundred and fifty six (n= 156) healthy Corriedale lambs, naturally infected with multiple resistant gastrointestinal nematodes were allocated into four experimental groups (n=39). Animals in each group received treatment (200 μg/kg) with either the RF, one of the test IVM formulations or were kept as untreated control. Blood samples were collected over 15 days post-treatment (n=8). The IVM plasma concentrations were measured by high performance liquid chromatography with fluorescence detection. The faecal nematode egg count reduction test (FECRT) (n=39) and evaluation of the clinical efficacy were performed at day 14 post-treatment (n=6), where a predominance of IVM highly resistant nematodes was observed. Los mayores obstáculos para la toma de decisiones en bioequivalencia promedio están referidos al tamaño de la variabilidad residual. Se ha visto que en ella incide mucho las características de los individuos reclutados para el ensayo. El presente trabajo propone una metodología que estudia la varianza residual según el género de los voluntarios y construye intervalos de confianza para el cociente de medias Test / Referencia (T/R), de los parámetros de biodisponibilidad, tanto en hombres como en mujeres. Asimismo se analizan las varianzas totales producidas por los medicamentos T y R, a los efectos de complementar la toma de decisiones. Las anteriores pautas fueron aplicadas a 3 estudios de bioequivalencia llevados a cabo con fármacos sustratos de la isoenzima CYP3A4 (itraconazol, alprazolam, claritromicina), algunos de los cuales son también sustratos del transportador de eflujo P-glicoproteína. Las mujeres presentaron menor exposición a los fármacos sustratos de CYP3A4 que los hombres. Los medicamentos T y R, conteniendo tales fármacos, mostraron biodisponibilidades relativas dependientes del sexo de los voluntarios, principalmente a causa de probables diferencias de disolución, originadas por la diferente fisiología del tracto digestivo que presentan hombres y mujeres. Microorganisms offer an alternative green and scalable technology for the synthesis of value added products. Fungi secrete high quantities of bioactive substances, which play dual-functional roles as both reducing and stabilizing agents in the synthesis of colloidal metal nanoparticles such as silver nanoparticles, which display potent antimicrobial properties that can be harnessed for a number of industrial applications. The aim of this work was the production of silver nanoparticles using the extracellular cell free extracts of Phanerochaete chrysosporium, and to evaluate their activity as antimicrobial and antibiofilm agents. The 45–nm diameter silver nanoparticles synthesized using this methodology possessed a high negative surface charge close to −30 mV and showed colloidal stability from pH 3–9 and under conditions of high ionic strength ([NaCl] = 10–500 mM). A combination of environmental SEM, TEM, and confocal Raman microscopy was used to study the nanoparticle-E. coli interactions to gain a first insight into their antimicrobial mechanisms. Raman data demonstrate a significant decrease in the fatty acid content of E. coli cells, which suggests a loss of the cell membrane integrity after exposure to the PchNPs, which is also commensurate with ESEM and TEM images.
