A recent study on the Raman spectrum of the cysteine zwitterion and anion, and the 2:1 (Cys)2Zn complex was reanalyzed employing B3LYP/6-311??G (3df,2pd) calculations in a simulated water environment. The spectra were rediscussed in light of the apparent incorrect structure determined in the original paper for this complex. The complex turns out to be tetrahedral and tetracoordinated instead of octahedral hexacoordinated, as initially proposed. The calculated Raman spectrum of the complex agrees very well with the experimental data, showing that both the geometrical and electronic structures are well represented. Three metal–ligand bands are found, two of them involving mostly the symmetrical and asymmetrical stretching of the Zn–N and Zn–S bonds. They were measured at 334 and 296 cm-1 and calculated at 319 and 249 cm-1, respectively. The third band involves the stretching of Zn–S bonds but also skeletal vibrations of the ligand. This band, measured at 399 cm-1 and calculated at 444 cm-1, has been previously assigned incorrectly to a Zn–O bond which does not actually exists since the CO2 -1 fragments are located away from the Zn ion. The structure of the lowest electronic states of HCOO· in C2v and Cs symmetries were optimized employing density functional theory (DFT) methods with extended basis sets including up to f- (on C and O) and d- (on H) polarization functions. Generalized gradient functionals (BLYP) and adiabatically connected functionals (B3LYP and B3PW91) were employed for studying HCOO·, as well as the isomer HOCO· (trans), the dissociation limit H·+CO2, and the transition state for the decomposition. At the best DFT levels employed, the ground state of HCOO· is 2A1 (in C2v) with equal C[BOND]O bond lengths, while the low-lying 2B2 state is only about 4 kJ/mol above (without inclusion of zero-point energies). The broken-symmetry 2A′ state (with unequal C[BOND]O bond lengths, i.e., Cs symmetry) is predicted to be about 13 kJ/mol above the 2A1 state and to be a transition state for the isomerization HCOO· (2A1)[RIGHTWARDS ARROW]HOCO· (2A′), with the trans-HOCO· isomer about 55 kJ/mol more stable. These facts agree closely with the most recent CASPT2/ANO calculations on this system. Therefore, it is concluded that some DFT models can be used safely for the study of larger radicals of the same type (despite several drawbacks discussed at length in this study). B3PW91, using several basis sets, is subsequently applied to the study of the possible reaction mechanisms of acetyloxyl radical, which exhibits a much more complicated path than formyloxyl, due to the presence of the methyl group. The optimum structures of isomers with coplanar or perpendicular CH and CO bonds were obtained for CH3COO· and two saddle points identified on the path of decomposition into CH3· and CO2. On the other side, saddle points for isomerization into CH3OCO· and CH2COOH· were also located, and the decomposition of the former to CH3O·+CO investigated. Finally, the structure of the benzoyloxyl radical (C6H5COO·) and its possible decomposition products were investigated along the same lines. Subject The topic of the resource RADICALES LIBRES Kieninger, Martina 1998 Abstract A summary of the resource. A new “non-terrestrial” molecule present in the envelope of the carbon star IRC + 10216 was described for the first time in 1986. Recently, this molecule was identified as the free radical MgNC, the first Mg-containing molecule in space. We present here the first density functional study performed on this radical, as well as on its isomer MgCN and the transition state connecting these species. It is shown that the optimum geometry obtained at the Becke3LYP/6-311+G(3df) level leads to the most exact rotational constants Be and Bo calculated up to now. It is also shown that the energy differences between the three species are completely in agreement with the best ab initio calculations available. Furthermore, it is shown that the popular MP2 method fails for this system in the same way that has been demonstrated for other radicals. High-level ab initio (MP2/6-311++G(2d,2p) geometry, Gaussian-2, MP4(SDTQ) and QCISD(T) binding energies) and density-functional (Becke3LYP/6-311++G(2df,2pd)) calculations have been performed on the charge-transfer complex between water and carbon dioxide. The complex appears to have two equivalent non-planar minima of Cs symmetry. Minima are separated by transition states with C1 symmetry, whereas the totally planar structure with C2v symmetry is a second-order transition state. All the critical points lie at approximately the same energy (less than 0.05 Kj mol−1 difference). Therefore, the experimentally observable structure should be planar. The best equilibrium intermolecular distance for this complex calculated at the MP2/6-311++G(2d,2p) level is 2.800 Å. Our best estimate of the observable intermolecular distance (corrected for anharmonicity) is 2.84 Å, in agreement with the experimentally derived value of 2.836 Å. Our best estimate of the binding energy at the QCISD(T) level, taking into account the variation of the distance owing to anharmonicity and the use of more sophisticated theoretical treatments, is −12.0 ± 0.2 kJ mol−1. Our best estimate of the barrier to internal rotation, also at the MP2/6-311++G(2d,2p) level, is 4.0 kJ mol−1, outside the error limits of the experimental determination (3.64 ± 0.04 kJ mol−1). Density functional theory at the level employed here gives an equilibrium intermolecular distance that is too large (2.857 Å), a binding energy that is too small (8.1 kJ mol−1), attributable neither to geometry nor to the basis set, and also a barrier to internal rotation that is slightly too small (3.39 kJ mol−1). The overall picture is, however, reasonably good. An analysis of static dipole polarizability of one atomic anion (F - ) and three diatomic molecules (H 2 , N 2 and HF) has been performed at the density functional theory (DFT) level. Dunning correlation-consistent basis sets, ranging from the smallest cc-pVDZ to the aug-cc-pV5Z were employed systematically for all the molecules. Additionally, special types of basis sets, derived specifically for the calculation of polarizabilities in an ab initio environment, were employed in some cases. Calculations of the polarizabilities were performed at the DFF level employing the BLYP, B3LYP and B3PW91 exchange-correlation potentials, and the accuracy of the results was judged against experimental data on the one hand, and the polarizabilities calculated at the Hartree-Fock (HF), MP2, MP4(SDTQ), QCISD and CCSD levels on the other. In all the cases studied, it was observed that the quality of the results followed the order (from worst to best) HF < MP2 < BLYP < B3LYP < B3PW91 MP4 QCISD CCSD, demonstrating that DFT (B3PW91 in particular) is a convergent alternative to highly correlated post-Hartree-Fock methods. Reduction of disulfide bonds in Kluyveromyces lactis β-galactosidase was assessed using solid phase reducing agents synthesized on three different supports (agarose, Toyopearl® and Eupergit®), provided with mercaptohydroxypropyl–ether groups. These agents are insoluble supports provided with thiol moieties as the reducing groups. The reduction performed on solid phase allowed a three-fold increase in the enzyme’s initial SH group content; the same result was achieved with dithiothreitol in solution. However, 10-fold less μmoles of SH groups per mg of protein were needed to perform the reduction of β-galactosidase with these solid phase reducing agents than the amount of SH groups required for reduction in solution. After the reduction process, the remaining content of SH groups on the solid phases was determined and found to be 60% for thiopropyl–agarose and thiopropyl–Toyopearl and 40% for thiopropyl–Eupergit. The reduced enzyme was reversibly immobilized on to agarose activated with thiol-reactive structures, thiolsulfinate (TSI) moieties, achieving high immobilization yields: more than 60% (for the enzyme reduced with dithiotreitol or thiopropyl–agarose) and more than 40% (for the enzyme reduced with thiopropyl–Toyopearl or thiopropyl–Eupergit). Half life at 45 °C of the resulting derivatives varied from 90 min (enzyme reduced with dithiothreitol) to 30 min (enzyme reduced with thiopropyl–Toyopearl and thiopropyl–Eupergit). The percentage of lactolysis at 25 °C was similar for all the derivatives, 90% after 2 h of reaction. A99Tc-ADP complex was prepared when KTcO4 was reduced in aquous medium by SnCl2, Na2S2O4, NaBH4 or Zn in the presence of ADP in excess. The resulting solution was studied by chromatography and spectrophotometry. Electrochemical reduction and substitution on [TcIII(tu)6]3+ were investigated as alternative synthetic routes. The anionic Tc-ADP complex was isolated as a solid. Cerimetric titrations confirmed the oxidation state +3 for the central atom. IR and1H-NMR data showed that the purine base is bonded to the Tc central atom but not the ribose moiety. No oxo groups seemed to be directly bonded to the Tc atom. The complex is rather stable in neutral solutions. However, it decomposes to pertechnetate and TcO2 at extreme pH values. Kinetics of the hydrolysis of BNPP (bis(4-nitrophenyl)phosphate) mediated by lanthanide – samarium (III) and ytterbium (III) – alone and in the presence of various alfa amino acids has been systematically studied at 37.0 �C and I = 0.15Min NaClO4, in the pH interval of 7–9. The rate of BNPP cleavage is sensitive to metal ion concentration, pH, and ligand to metal molar ratio. Hydrolysis follows Michaelis–Menten-type saturation kinetics. For both metals, high pH values markedly increase the observed activity. Besides, potentiometric titrations of all these systems under identical conditions allowed us to identify the active coordination compounds towards hydrolysis. The results show that complexes with phosphodiesterolytic activity are monomeric cationic species such as [Ln(aa)3(OH)]2+ or [Ln(aa)2(OH)2]+. Since phosphodiesterolytic activity is evident above pH 7 and it is increased with increasing pH, hydrolytic reactions of the metals are competitive processes that could lead to their precipitation as Ln(OH)3(s). In this sense, ligand excess (for example, ligand to metal molar ratio equal to 30) was employed. Furthermore, due to its more extended hydrolysis, ytterbium shows, in general, less activity than samarium under the studied conditions. In general, a good phosphodiesterolytic activity is observed for these complexes under similar conditions to the physiological ones. Amino acids could be easily derivatized without changing their coordinating ability, leading to lanthanide complexes possibly capable of efficiently hydrolyzing the phosphodiester linkages of nucleic acids. Subject The topic of the resource CATIONES BIBLIOGRAFIA NACIONAL QUIMICA 2005 Abstract A summary of the resource. myo-Inositol hexakisphosphate (InsP6) is an ubiquitous and abundant molecule in the cytosol and nucleus of eukaryotic cells whose biological functions are incompletely known. A major hurdle for studying the biology of InsP6 has been a deficiency of a full understanding of the chemistry of its interaction with divalent and trivalent cations. This deficiency has limited our appreciation of how it remains in solution within cells, and the likely degree to which it might interact in vivo with physiologically important cations such as Ca2+ and Fe3+.We report here the initial part of the description of the InsP6-multivalent cation chemistry, including its solution equilibria studied by high resolution potentiometry and (for the Fe(III)/Fe(II) couple) cyclic voltammetry. InsP6 forms anionic complexes of high affinities and 1:1 stoichiometry with Mg(II), Ca(II), Mn(II), Fe(II), Co(II), Ni(II), Cu(II), Zn(II) and Cd(II). Of particular importance is the observation that, in the exceptional case of Mg(II), InsP6 forms the species [Mg5(H2L)] (L representing fully deprotonated InsP6); this soluble neutral species is predicted to be the predominant form of InsP6 under nuclear or cytosolic conditions in animal cells. Contrary to previous suggestions, InsP6 is predicted not to interact with cytosolic calcium even when calcium is increased during signalling events. In vitro, InsP6 also forms high affinity 1:1 complexes with Fe(III) and Al(III). However, our data predict that in the biological context of excess free Mg(II), neither Fe(III) nor Fe(II) are complexed by InsP6. A biotype of clary sage (Salvia sclarea L.) growing in Uruguay was evaluated for its suitability to be cultivated in Uruguay. Essential oils from the inﬂorescences and leaves of plant harvested at full ﬂowering and early seed ripeness stages were obtained by steam distillation and characterized by GC and GC–MS (quadrupole). Twenty-seven components (91–98% of the total composition) were identiﬁed. The oil was characterized by a high content of linalool (8–22%), linalyl acetate (39–48%) and sesquiterpenes, with germacrene D (8–20%) and (E)-β-caryophyllene (3–5%) as the main components. The enantiomeric distribution of -pinene, sabinene, β-pinene, limonene, linalool, linalyl acetate and germacrene D was studied by multidimensional HRGC–HRGC. The development stage did not inﬂuence the oil composition or the enantiomeric distribution of the studied components of the clary sage biotype over the period of evaluation. The essential oil of aerial parts of Lippia alba (Mill.) N. E. Brown cultivated in a small-scale experimental area was analysed by GC and GC–MS (quadrupole). Twenty-seven components were identified, corresponding to 93% of the total oil. The enantiomeric distribution of α-pinene, sabinene, limonene and linalool was studied by multidimensional GC–GC. Existe creciente interés en el desarrollo de extractos antioxidantes como ingredientes funcionales. El objetivo del presente trabajo fue evaluar el perfil sensorial, el contenido de polifenoles y la capacidad de secuestrar radicales DPPH de extractos de tres plantas nativas uruguayas: Achyrocline satureoides, Baccharis trimera y Mikania guaco. Tanto el solvente como la planta nativa tuvieron un efecto significativo en el contenido de polifenoles, la capacidad de secuestrar radicales DPPH y el perfil sensorial de los extractos. Los extractos acuosos fueron los más eficientes secuestrantes de radicales DPPH y presentaron el mayor contenido de polifenoles y la menor intensidad de amargor, astringencia y sabor característico. El amargor, astringencia y sabor caracterı´stico de los extractos podrían limitar su aplicación como ingredientes funcionales. Estos resultados sugieren que durante la optimización de las condiciones de extracción de compuestos antioxidantes de plantas, además de la influencia de la capacidad antioxidante y el rendimiento, deben tenerse en cuenta las características sensoriales de los extractos. A study was carried out to identify consumers’ previous expectations of chocolate milk desserts enriched with antioxidants and to determine if these expectations affected product perception. Seventy-five consumers participated in the study and were asked to complete a word association task before the evaluation. Then, consumers tried six milk desserts with different polyphenolic concentration, scored their overall liking and willingness to purchase and provided up to four words to describe each of the samples. Cluster analysis performed on consumer-elicited terms in the word association task allowed the identification of three consumer segments with different expectations and motivations toward chocolate milk desserts enriched with antioxidants. These groups also differed in their evaluation of the desserts when tasting them, showing different overall liking, willingness to purchase and sensory description of the samples. This suggests that consumers’ prior expectations and motivations significantly affected their response when tasting the desserts.
