Eight chocolate milk desserts with different formulation were evaluated by two groups of consumers. Fifty consumers evaluated the samples and indicated their overall liking and answered a CATA question. Meanwhile, 40 consumers elicited up to four words to describe the desserts and completed a projective mapping task. Projective mapping and the check-all-that-apply question provided very similar sensory profiles for the evaluated milk desserts. Differences in the sensory characteristics of the samples were explained by differences in their formulations, which suggest the validity of the sensory profiles given by consumers. Projective mapping and the CATA question consisted on valuable tools to understand their perception of the sensory and hedonic characteristics of the desserts. Eleven samples of strawberry yogurt prepared with different red color concentrations using Ponceau 4R (E-124) were evaluated by instrumental and sensory methods. Color intensity evaluation was carried out by a panel of eight assessors specifically trained to measure strawberry color in yogurt. Color acceptability was measured with 120 regular and frequent consumers of yogurt. Color was measured with a Minolta Chroma Meter CR-200b, obtaining parameters L*, a* and b*. Principal component analysis was performed on the instrumental variables. Check-all-that-apply (CATA) questions could be a simple alternative to get an insight on consumer perception of a food product. In the present work, CATA questions were used in the development of chocolate milk desserts. Nine chocolate milk desserts were formulated with different sugar and cacao concentration. A consumer study was performed with 70 people, who were asked to score their overall liking and to answer a CATA question that included 18 sensory and hedonic terms. Besides, the samples were evaluated by a trained assessors' panel. Highly significant differences were found in the frequencies in which CATA terms were used for describing the nine desserts, suggesting that this methodology was able to detect differences in consumer perception of the desserts. Survival analysis methodology was used to estimate the shelf life of alfajor (a chocolate-coated individually wrapped cake) at 20 and 35C by using results obtained from consumers when asked if they would accept or reject samples with different storage times. Sensory acceptability (measured by consumers), off-flavor (measured by a trained panel) and moisture content were linearly related to time. These correlations were used to estimate values at the shelf-life times calculated for 25 and 50% rejection probability. Survival analysis provided the following shelf-life estimation: 74 days at 20C and 33 days at 35C for a 25% of rejection, 87 days at 20C and 39 days at 35C for a 50% of rejection. An alfajor stored at 20C having an acceptability value below 4.9 (1–9 hedonic scale) and off-flavor intensity above 5.3 (0–10 scale) would be rejected by 25% of the consumers. Taking into account the impact of texture on consumers’ liking of milk desserts, it is important for dairy companies to understand how consumers describe it. In this context, the aim of the present study was to identify consumers’ texture vocabulary of milk desserts using free listing and a check-all-that-apply question. A free listing task was carried out with 66 consumers, who were asked to list all the positive and negative texture characteristics of milk desserts. Moreover, 65 consumers tried 8 commercial milk dessert samples and answer a check-all-that-apply question with 27 texture terms. Both methodologies enabled the identification of the texture characteristics more relevant for consumers and the texture vocabulary they usually use to describe this type of product. Creaminess and smoothness were the most relevant characteristics, followed by attributes related to thickness. For each choice set, consumers had to indicate which yogurt would normally buy. A questionnaire on consumer attitude towards health and nutrition was answered by participants after the choice task. Brand, price and health claims highly significantly affected consumer choice of functional yogurts. The relative importance of brand was similar to that of type of yogurt. Therefore, results suggested that non-sensory factors such as brand and price could have an important impact on consumer perception of functional foods. The impact of brand, price and health claims was affected by consumer interest in health issues. The purpose of the study was to investigate the factors upon which the first order rate constant depends in order to assess the impact on the body-drug concentration when it changes throughout a treatment. A reasoning for linking the first order rate constants with several factors in a multiplicative way was proposed out taking into account kinetic and thermodynamic aspects. The saliva profile throughout time shows a high variability observed as more than one peak in the saliva concentration versus time curves. In both cases, even if there is a major peak, when the statistical analysis of the data is performed, an important variability in Cmax results in high values in the residual variance of the ANOVA test. Consequently, the power of the bioequivalence test decreases and sometimes it is not possible to conclude on bioequivalence. The average concentration (Cav), the average maximum concentration (Cmax, av) and Cmax,av/Cav x 100 (%Cmax,av) are proposed in this paper as possible parameters in order to evaluate the profile of the concentration-time curves, as they reduce the residual variability in bioequivalence studies. Glutathione transferases (GSTs) are believed to be a major detoxification system in helminths. We describe the expression and functional analysis of EgGST, a cytosolic GST from Echinococcus granulosus, related to the Mu-class of mammalian enzymes. EgGST was produced as an enzymatically active dimeric protein (rEgGST), with highest specific activity towards the standard substrate 1-chloro- 2,4-dinitrobenzene (CDNB; 2.5�mol min−1 mg−1), followed by ethacrynic acid. Interestingly, rEgGST displayed glutathione peroxidase activity (towards cumene hydroperoxide), and conjugated reactive carbonyls (trans-2-nonenal and trans,trans-2,4-decadienal), indicating that it may intercept damaging products of lipid peroxidation. In addition, classical GST inhibitors (cybacron blue, triphenylthin chloride and ellagic acid) and a number of anthelmintic drugs (mainly, hexachlorophene and rafoxanide) were found to interfere with glutathione-conjugation to CDNB; suggesting that they may bind to EgGST. Considered globally, the functional properties of rEgGST are similar to those of putative orthologs from Echinococcus multilcularis and Taenia solium, the other medically important cestodes. Interestingly, our results also indicate that differences exist between these closely related cestode GSTs, which probably reflect specific biological functions of the molecules in each parasitic organism. Confirming previous studies obtained with less than 3% of the total genes recently described, we found that A- and U-ending triplets predominate but translational selection increases the frequency of a subset of codons in highly expressed genes. However, some new results come from the analysis of the complete sequence. First, there is more variation in GC3 than previously described; second, the effect of natural selection acting at the level of translation has been analysed with real expression data at 4 different stages and third, we found that highly expressed proteins increment the frequency of energetically less expensive amino acids. This paper reports the production of monoterpenes, which elicit a floral aroma in wine, by strains of the yeast Saccharomyces cerevisiae. Terpenes, which are typical components of the essential oils of flowers and fruits, are also present as free and glycosylated conjugates amongst the secondary metabolites of certain wine grape varieties of Vitis vinifera. Hence, when these compounds are present in wine they are considered to originate from grape and not fermentation. However, the biosynthesis of monoterpenes by S. cerevisiae in the absence of grape derived precursors is shown here to be of de novo origin in wine yeast strains. Higher concentration of assimilable nitrogen increased accumulation of linalool and citronellol. Microaerobic compared with anaerobic conditions favored terpene accumulation in the ferment. The amount of linalool produced by some strains of S. cerevisiae could be of sensory importance in wine production. These unexpected results are discussed in relation to the known sterol biosynthetic pathway and to an alternative pathway for terpene biosynthesis not previously described in yeast. Although the traditional wine industry of Uruguay is virtually unknown in the international wine market, wine sales are now increasing in some of the most important markets of Europe and North America. Because of strong domestic consumption and a quiescent R&D situation in recent decades, the pioneer vision of high quality that was bom in the 1880s was not realised. The strategy of promoting the typical grape Tannat (Harriague), best known for making high‐quality Madiran reds in southwest France, is starting to result in successful competitive reds which are only found in Uruguay. This paper presents a summary of the history of the Uruguayan wine industry, beginning with the first Tannat vines planted in the 1870s by the Frenchman Harriague, who with a British family developed the most technically advanced winery of those days, including a bacteriological laboratory. With the programme of training for winemakers and the recently started R&D projects in the University of the Republic, it is hoped that the Uruguayan wine industry, based on quality wines at a good price, will continue opening markets and reach its target of 300 000 cases by the year 2000. The country could occupy an interesting position within the quality wines of the Mew World in the near future. The capacity of yeast to modify anthocyanin concentration during red wine fermentation was studied with the objective of classifying yeast strains according to their capacity for color removal during vinification. A model red grape juice medium was used to better understand yeast-anthocyanin interactions. An anthocyanin extract of Vitis vinifera cv. Tannat was prepared from grape skins and diluted in a white must in order to obtain a red grape juice without solids. Anthocyanin removal was confirmed to be higher for compounds with higher polarity. Acylated anthocyanins (acetyl and p-coumaryl compounds) were differentially removed, and the percentage of removal for each compound was determined. Results showed no correlation between color intensity and total anthocyanin concentration after fermentation, possibly because of the presence of anthocyanin derivatives formed during the process. HPLC-MS analysis allowed identification of anthocyanin derivatives, while the quantification of several derivatives was performed by HPLC-DAD. The sum of these derivatives showed a direct correlation with the color intensity obtained with each strain, thus explaining the color variability observed. MLF led to a significant decrease (P < 0.05) in secondary descriptors such as ‘berry fruit’ and ‘fresh vegetative’, as well as a decrease in related tertiary descriptors such as ‘blackcurrant’, ‘apricot’, ‘cut green grass’ and ‘green pepper’. The main differences in the chemical composition of Tannat wine aromas following MLF were (1) an increase of the lactates (mainly ethyl lactate) over the sensory threshold; (2) a significant decrease in ethyl esters and acetates, depending on the bacteria strain utilised (the most significant change was found in hexyl acetate), and (3) a small increase of 4-vinylguaiacol and 4-vinylphenol with one of the MLF strains (this change was below the threshold of human perception). We also observed an increase of compounds related to α-ketoglutarate metabolism, namely: ethyl 4-hidroxibutanoate and γ-butyro-lactone. Differences in chemical composition due to MLF are discussed in relation to variation in sensory profiles. This study investigated the degradation of proteins and amino acids by Caloramator proteoclasticus, an anaerobic thermophilic (55 °C) fermentative bacterium isolated from an anaerobic bioreactor. Experiments were performed in the presence and absence of Methanobacterium thermoformicicum Z245, a methanogen that can use both hydrogen and formate for growth. Higher production rates and yields of the principal fermentation products from gelatin were observed in methanogenic coculture. The specific proteolytic activity in coculture tripled the value obtained in pure culture. C. proteoclasticus fermented glutamate to acetate, formate, hydrogen and alanine. In methanogenic coculture, a shift towards higher amounts of acetate and hydrogen with no alanine production was observed. Extracts of glutamate-grown cells possessed high activities of β-methylaspartase, a key enzyme of the mesaconate pathway leading to acetate. The presence of two enzymes (alanine-α-ketoglutarate aminotransferase and NADH-dependent alanine dehydrogenase) usually involved in the biosynthesis of alanine from pyruvate was also detected. The fermentation of amino acids known to be oxidatively deaminated (leucine and valine) was improved in the presence of both methanogenesis and glycine, a known electron acceptor in the Stickland reaction. Culture conditions seem to be very important in the way C. proteoclasticus disposes of reducing equivalents formed during the degradation of amino acids.
