The biomass industrialization valorisation of grains and cereals is considered an opportunity for the countries where agro-industrial activity is one of its main economic activities, rendering new higher-value products with a concomitant solution to waste accumulation issues. To that end, in this work we describe and characterise bioactive compounds generation from rice husk by semi-solid fermentation, obtained from 500 g of the material at room temperature and 60% humidity with mixed cultures of Phanerochaete chrysosporium and Gloeophyllum trabeum. The extract was evaluated in different situations: murine mammary tumour cells (4T1), normal cells (NIH 3T3) and in Lactobacillus acidophilus, and Trichoderma harzianum. The results exhibited that the extract inhibited 4T1 cells at concentrations higher than 20μg/mL, but did not inhibit normal cells, and displayed germicide activity after 3 days incubations. We propose that these functionalized compounds have a potential application in industry/agriculture/medicine obtained from rice husk waste. Trypanosomatid parasites and fungi cause highly prevalent diseases. Ergosterol plays a crucial role in the structure and normal function of the membrane of these microorganisms. It is absent in mammals. Many enzymes of the ergosterol biosynthesis route have been recognized as molecular targets for drugs. Currently, a variety of organic drugs that interfere with sterol biosynthesis are employed to treat fungal infections, and some of them have been proposed as potential treatments for trypanosomiasis. Coordinating these organic drugs to metal ions may positively impact the biological performance by altering relevant physicochemical properties. Additionally, their organometallic derivatization could improve the effectiveness. This comprehensive review aims to highlight the considerable promise of metal-based compounds as inhibitors of essential pathways or key enzymes of these microorganisms, with focus on the ergosterol biosynthesis pathway. Additionally, bioactive metal compounds that have demonstrated experimental efficacy in acting on enzymes within this pathway although not designed for this purpose were included. Los fármacos esteroidales son usados para un amplio rango de propósitos terapéuticos, representado uno de los más grandes sectores de la industria farmacéutica. Es conocido que las transformaciones microbianas de esteroides para la producción de fármacos son uno de los ejemplos más exitosos de procesos biotecnológicos aplicados a escala industrial. Estas permiten obtener precursores e intermediarios claves para la síntesis de fármacos mediante procesos que cumplan con los principios de la química verde y el desarrollo sustentable. En este contexto es que surge el interés de investigar la biotransformación de esteroides en productos de mayor valor agregado. En esta tesis se realizó el aislamiento de microorganismos desde efluentes de lavado de lana ricos en esteroles con el fin de encontrar biocatalizadores capaces de transformar esteroides. Como resultado, se logró recuperar 15 bacterias, 8 hongos filamentosos y 2 levaduras. La mayoría de ellos fueron identificados utilizando distintas metodologías: métodos moleculares, observación de las características macroscópicas y microscópicas de hongos filamentosos e identificación de bacterias empleando pruebas bioquímicas primarias y MALDI-TOF MS. Además, los microrganismos fueron caracterizados en función de sus capacidades catalíticas realizando la detección de actividades epoxihidrolasas, esterasas, lipasas y Baeyer-Villiger monooxigenasas en células enteras. Al mismo tiempo, se evaluó la capacidad de biotransformación del colesterol en los microorganismos aislados. Cuatro bacterias y cinco hongos filamentosos, identificados como Acinetobacter sp., Exiguobacterium sp., Tsukamurella sp., Streptomyces sp., Fusarium sp., Trichoderma sp., Trichoderma koningiopsis y las dos cepas de Mucor circinelloides mostraron la capacidad de transformar el colesterol, obteniéndose los productos 7β-hidroxicolesterol, colesterona y 5,6-epoxicolesterol. Por otra parte, las bacterias capaces de transformar el colesterol fueron evaluadas en la biotransformación de los ácidos litocólico y cólico. De esta forma se logró determinar que Tsukamurella sp. es un biocatalizador novedoso para la obtención de los ácidos 3-cetolitocólico y 3-cetocólico. Fueron estudiadas distintas condiciones de las reacciones de biotransformación de xxi esteroides con esta bacteria, incluyendo el escalado de la oxidación del ácido litocólico a un fermentador de 5 L. Al mismo tiempo, se logró identificar la secuencia de los genes que codifican para varias enzimas relacionadas a la biotransformación de sustratos esteroidales en la secuencia genómica de Tsukamurella sp. La producción de biodiésel mediante la reacción de transesterificación (único proceso tecnológicamente maduro a la fecha para ese propósito) genera simultáneamente como subproducto glicerol en relación aproximadamente 9:1 en masa. La evolución positiva que en los últimos años ha presentado la producción de este biocombustible a nivel global ha desencadenado la introducción de volúmenes inmanejables para el mercado del glicerol, presionando fuertemente a la baja su precio. Este hecho -de no encontrarse una soluciónamenaza impactar en la sustentabilidad ambiental y económica del biodiésel. La búsqueda de alternativas a esta sobreproducción de glicerol ha sido un desafío atendido por la comunidad científica internacional desarrollándose múltiples estrategias (desde la búsqueda de nuevas aplicaciones que permitan su uso directo, el uso enproductos masivos que lo demanden como insumo, así como la búsqueda de síntesis de productos que impliquen agregado de valor a este co-producto). El presente trabajo se inscribe dentro de esta última alternativa proponiendo la valorización del glicerol a través de su biotransformación en 1,2-propanodiol enantioméricamente puro y su posterior derivatización obteniéndose sintones quirales con aplicación en química fina. Recurriendo a ingeniería metabólica en E. coli se handiseñado, construido y evaluado un conjunto de biocatalizadores con ese objetivo. A partir de la expresión de la metilglioxal sintasa (mgs) de E. coli,una metilglioxal reductasa (Gre2p) y una aldehído reductasa(Ypr1p ) ambas de S. cerevisiae,se ha logrado obtener (R)-1,2-propanodiol enantioméricamente puro (con un e.e. > 99 %, en concentración de 0,45 g/L)a partir de glicerol. The goal of this work was to include an interdisciplinary research-oriented laboratory experiment taken from medicinal chemistry into the undergraduate biochemistry or organic studies. Phenazine 5,10-dioxides with selective cytotoxicity under hypoxia and with relevance in solid tumor treatment were selected to simulate different metabolism states. In this laboratory experiment, metabolism of 7-bromo-2-hydroxyphenazine-5,10-dioxide under simulated hypoxia and normoxia was studied using rat liver cytosol. Differential hypoxic and normoxic metabolism were monitored by thin-layer chromatography. Specific enzyme inhibitors, dicoumarol, ketoconazole, and menadione, were employed to identify the enzymatic system involved. This experiment introduced students to the concept of drug metabolism, activation, and mechanism of action. Furthermore, basic biochemical techniques of enzyme isolation and study were also discussed. Epilachna paenulata (Coleoptera: Coccinellidae) is a South American ladybird beetle that feeds on plants of the Cucurbitaceae family. The defensive chemistry of E. paenulata has been characterized as a mixture of systemic piperidine, homotropane, and pyrrolidine alkaloids. Whole body extracts of adult beetles contain four major alkaloids: 2- (2′-oxopropyl)-6-methylpiperidine (1); 1-(6-methyl-2,3,4, 5-tetrahydro-pyridin-2-yl)-propan-2-one (2); 1-methyl-9- azabicyclo[3.3.1]nonan-3-one (3); and 1-(2″-hydroxyethyl)- 2-(12′-aminotridecyl)-pyrrolidine (4). Comparative studies of the defensive chemistry of eggs, larvae, pupae, and adults showed differences in alkaloid composition and concentration among life stages. While adults contained mainly the homotropane 1-methyl-9-azabicyclo[3.3.1]nonan-3-one (3), eggs showed the highest concentration of the piperidine 2- (2′-oxopropyl)-6-methylpiperidine (1). We studied the origin of this alkaloid in the eggs by feeding newly emerged, virgin adult beetles with [2-13C]-labeled acetate, and by performing crosses between 13C-fed and unlabeled males and females. GC-MS analysis of alkaloids from 13C-fed males and females showed high incorporation of 13C into the alkaloids, as evidenced from a 20–30% increase of isotopic peaks in diagnostic fragment ions, confirming the expected endogenous origin of these alkaloids. In addition, analyses of eggs from different crosses showed that labeled alkaloids from both parents are incorporated into eggs, indicating that E. paenulata males transfer alkaloids to the females at mating. Biparental endowment of chemical defenses into eggs has been shown previously in insects that acquire defensive compounds from dietary sources. To our knowledge, this is the first report of biparental egg endowment of endogenous defenses. In the search for a pharmacological answer to treat Chagas disease, eight metal complexes with two bioactive bisphosphonates, alendronate (Ale) and pamidronate (Pam), were described. Complexes of the formula [M 2 II(Ale) 4(H 2O) 2]·2H 2O, with M = Cu, Co, Mn, Ni, and ([CuPam]·H 2O) n as well as [M II(Pam) 2(H 2O) 2]·3H 2O, with M = Co, Mn and Ni, were synthesized and fully characterized. Crystal structure of [Cu 2 II(Ale) 4(H 2O) 2]·2H 2O, [Co II(Pam) 2(H 2O) 2] and [Ni II(Pam) 2(H 2O) 2] were solved by X-ray single crystal diffraction methods and the structures of [M 2 II(Ale) 4(H 2O) 2] ·2H 2O complexes M = Co, Mn and Ni were studied by X-ray powder diffraction methods. All obtained complexes were active against the amastigote form of Trypanosoma cruzi (T. cruzi), etiological agent of Chagas disease. Most of them were more active than the corresponding free ligands showing no toxicity for mammalian cells. The main mechanism of the antiparasitic action of bisphosphonates, inhibition of parasitic farnesyl diphosphate synthase (TcFPPS), remains in the obtained metal complexes and an increase in the inhibiting enzyme levels was observed upon coordination. Observed enzymatic inhibition was selective for TcFPPS as the metal complexes showed no or little inhibition of human FPPS. Additionally, metal complexation might improve the bioavailability of the complexes through the hindrance of the phosphonate group's ionization at physiological pH and, eventually, through the ability of plasma proteins to work as complex transporters. The title compounds, [Ni(S2O3)(C12H8N2)2]�0.92H2O�1.4- CH4O and [Ni(S2O3)(C10H8N2)2]�2H2O�0.55CH4O, are monomeric, containing nickel(II) in a distorted octahedral coordination environment provided by the four N atoms of two bidentate bipy or phen groups and one S and one O atom from a chelating thiosulfate anion. The crystals are highly unstable outside their mother liquors and are stabilized in solution by a not fully determined number of water and methanol solvate molecules. The phenanthroline structure includes two independent moieties related by a non-crystallographic inversion center.
