BACKGROUND: The aims of the present study were to study the influence of information about the source of two functional ingredients (antioxidants and fibre) on consumer perception of functional milk dessert concepts and to evaluate whether this influence depends on consumer attitudes towards health and hedonic characteristics of foods. RESULTS: Scores for expected liking decreased when information about the source of fibre or antioxidants was incorporated, which could be explained considering that when consumers knew the source of the functional ingredient they imagine the flavour of the source in the milk concept. This decrease in expected liking led to a decrease in consumer willingness to purchase, showing the importance of hedonic expectations on the acceptance of functional foods. Consumers with different attitudes towards health and hedonic characteristics of foods reacted differently towards information about the source of functional ingredients. CONCLUSION: Providing information about the source of functional ingredients would not be recommended as it might create negative taste expectations to consumers and a concomitant decrease in their willingness to purchase. These show that consumers would not compromise on the taste of milk desserts for health. Thus, the development of functional foods that taste worse than conventional ones may be a risky option. Consumers’ perceptions of conventional (regular plain yogurt, low-calorie and low-fat yogurt) and functional yogurts (enriched with fibre and antioxidants) were studied using word association and hard laddering. Both methodologies provided similar results, being consumers’ perceptions of these products mainly related to health, nutrition, sensory characteristics and pleasure. However, both methodologies provided different information. Word association provided information about attributes that were relevant for consumers’ acceptance of a product, common defects of products in the marketplace, motives behind consumers’ choice and attributes that might limit consumers’ interest in the product. On the other hand, hard laddering allowed the identification of more consequences and values related to product attributes, and the links between them, which could be useful for understanding consumers’ choices. Besides, word association was more sensitive than hard laddering in determining differences between groups of consumers with different attitudes towards health and nutrition. Results suggested that word association might be an interesting and useful qualitative technique for product development. As the markets for functional foods increase, studying and understanding consumers’ perception of functional foods is essential to develop products that have a good consumer acceptance. The objective of this work was to provide a first insight into Uruguayan consumers’ perception of functional foods. A survey with 200 consumers was performed. The participants were not familiar with the term “functional food,” as only 37% related the term with the real concept behind it. However, the participants showed a positive attitude toward the functional foods concept. Preferred areas of action of functional foods at a generic level were not reflected in the ratings of perceived healthiness and willingness to try yogurt with different enrichments, probably because they did not associate the components with their health effect. These results suggest that the use of health claims would be essential to ensure the success of functional foods. In order to assess the influence of nutritional knowledge on perceived healthiness and willingness to try functional foods, 104 consumers filled out a Nutritional Knowledge Questionnaire and answered a conjoint task. Participants had to evaluate 16 concepts consisting of combinations of carrier products (yogurt, milk desserts, pan bread and mayonnaise) and nutritional modifications (regular product, lowfat, enriched with antioxidants, and enriched with fibre). Three groups of consumerswere identified with different level of nutritional knowledge. Highly significant differences were found in the healthiness evaluations of the clusters, which mainly depended on nutritional knowledge related to the links of diet and diseases. Highly significant differences in willingness to try functional foods were also found between the clusters. Whereas consumers with the lowest nutritional knowledge were not interested in consuming functional foods, the addition of fibre or antioxidants to healthy products increased the willingness of consumers with the highest level of nutritional knowledge to try the evaluated functional foods. These results suggested that lack of nutritional knowledge might limit the acceptance of functional foods and thus the use of health claims might be necessary to assure that consumers are aware of their health benefits. The aims of the present work were to study motives underlying Uruguayan consumers’ food choice behaviour and to study the consumption frequency of some selected food items. A modification of the Food Choice Questionnaire and a food frequency questionnaire was administered to a group of 200 Uruguayan consumers. Feeling good and safety, sensory appeal and health and nutrient content were rated as the most important factors, while familiarity was rated as the least important. Using hierarchical cluster analysis, three clusters with different choice patterns were identified. Frequency of consumption of fruits, vegetables, milk and dairy products, and whole cereals, increased as the importance attributed to health and nutrition increased; consumption of fatty foods decreased. The aims of the present work were to study the crystallization of lactose in dulce de leche and to evaluate the influence of temperature on this process. Samples of approximately 1 g were placed in plastic containers and stored at –5, 5, 20 and 35°C for 25 days. Lactose crystal and lactose crystal agglomerate numbers and size were measured by using an optical microscope. Results suggested that stirring and placing a small sample of dulce de leche in a small container induced lactose crystallization. Lower storage temperatures resulted in an increase of crystal and agglomerate number but in a decrease of crystal and agglomerate size. The effect of temperature on crystal and agglomerate growth rates could be attributed to the effect of temperature on viscosity, as lower temperatures resulted in decreased crystal mobility in dulce de leche and therefore reduced crystal and agglomerate growth rate. The objective of the present work was to study the influence of both passive and active modified atmosphere packaging on sensory shelf life of butterhead lettuce leaves, stored at 5 and 10 �C. Results showed that the efficacy of active modified atmosphere with respect to passive modified atmosphere depended on the storage temperature considered. When stored at 10 �C, despite being subjected to different package atmosphere composition, lettuce leaves in active and passive modified atmosphere showed the same sensory deterioration rate and sensory shelf life. On the contrary, when stored at 5 �C, lettuce leaves in active modified atmosphere packages showed lower deterioration rate and higher sensory shelf life than those in passive modified atmosphere. Sensory shelf life of lettuce in modified atmosphere packages was estimated using criteria determined according to consumers’ rejection to purchase percentage, which consisted on an improvement over more arbitrary criteria presented in most studies. In this work CD4-knockout mice were used as a model to analyse the role of CD4+ T cells in the antibody response against Echinococcus granulosus immunization or experimental infection. Results obtained with mice immunized with protoscolex antigens indicated that these contain T-independent antigens. After infection, CD4-knockout mice and C57Bl/6 mice showed similar titres of specific antibodies indicating that T-independent antibody production was quantitatively important in early infection. We have also identified an antigenic fraction from protoscoleces (E4+) which induces CD4 T cell independent antibody response in early stages of infection. In conclusion, the results presented here directly support the existence of T-independent immunogens in E. granulosus protoscoleces and suggest that T-independent antibody response may be quantitatively important in early infection. Solanum commersonii is a wild tuber-bearing species native to Uruguay with high potential for use in potato breeding programs. Little is known about the genetic diversity within this wild species and the relationship with the resistance to the bacterial pathogen Ralstonia solanacearum. We studied 30 S. commersonii clonal accessions, 20 of which were collected from geographically diVerent areas across the country, while the other ten were grown from seeds from a single plant. Resistance against R. solanacearum was tested and diVerent levels of resistance were found, ranging from delayed wilting to asymptomatic reactions. The genetic variation and the relationships among individuals in this germplasm collection were studied by diVerent molecular markers: Random AmpliWed Polymorphic DNA (RAPD), AmpliWed Fragment Length Polymorphism (AFLP) and Microsatellites or Simple Sequence Repeats (SSR). AFLP markers generated the largest number of total and polymorphic fragments per assay unit while SSR revealed the highest frequency of polymorphic bands (100%), followed by AFLP (96.2%) and RAPD (89.4%). In contrast, when comparing the number of diVerent genetic proWles generated, the SSR markers exhibited the lowest discriminatory power. The clustering pattern obtained with the three marker systems showed a similar distribution of the S. commersonii germplasm revealing a high correlation between the three methods employed. All three dendrograms grouped most of the accessions into two main clusters, containing the same accessions regardless of the marker type. Bacterial wilt resistant accessions were present in both clusters. Accessions originated from diVerent seeds of the same plant were grouped within one of the major clusters, and diVered in the response to R. solanacearum revealing segregation of resistance. Dinuclear class II CoIII/FeII mixed-valence complexes of type [LnCoIII(m-NC)FeII(CN)3L2]m� (where Ln represents a pentadentate macrocycle and L2 a bpy ligand or two cyanides) have electronic properties that make them possible sensitizers in DSSC (dye sensitized solar cells). For this purpose the new complex Na2[{trans-L14COOCoIII(m-NC)}FeII(CN)5] has been prepared and characterized by the usual methods and its sensitizer properties compared with those of the already known [{trans-L15CoIII(m-NC)}FeII(CN)3(bipy)eq,ax](ClO4). The complex [{trans- L14COOCoIII(m-NC)}FeII(CN)5]2� has been designed for the actuation of an electron injection from the cobalt centre on MMCT, while the [{trans-LnCoIII(m-NC)}FeII(CN)3(bipy)]+ structure can produce a tuned injection from the iron centre via an MLCT electronic state, as described for similar systems. The characterization on solid oxide semiconductor supports has been carried out for these two complexes and the results have been compared with the behaviour observed in aqueous solution and in solvents of varying polarities. Their use in DSSC has been checked and, while a sensitizer response is observed for [{trans-L14COOCoIII(m-NC)}FeII(CN)5]2�, complex [{trans-L15CoIII(m-NC)}FeII(CN)3(bipy)eq,ax]+ does not produce any electrical current on illumination. The low efficiency of the built DSSC can be easily related both with the very low value of the extinction coefficient of the MMCT band responsible for the electron injection, and with the small driving force for the reduction of the complex with the standard I2/I3� system used after electron injection. One of the tools of biodiversity prospecting is searching for chemical information. In this project, we propose the botanical, chemical and biological biodiversity prospecting of the gallery forest of the Uruguay River Basin near the Guaviyú stream. A series of over a hundred furoxans, alkylnitrates and related compounds were studied as growth inhibitors of the two major kinetoplastids of Latin America, Trypanosoma cruziand Leishmania spp., in in vitro assays. The most active compounds showed 50% inhibitory doses of the same order of that of Nifurtimox and Miltefosine, reference drugs used to treat Chagas Disease and Leishmaniasis respectively. Among the studied compounds derivative 4, presenting excellent inhibitory activity against the tryposmastigote and amastigote forms of T. cruzi, has emerged as a lead compound. Mechanism of action seems to involve mitochondrial dehydrogenases as a distinct effect with respect to Nifurtimox. Excreted metabolites, studied by NMR, showed a significant decrease in succinate, confirming the observed effect on the mitochrondrial dehydrogenases. La dihidroxilación enzimática de compuestos aromáticos es una metodología de amplio uso en síntesis orgánica, produciendo ciclohexadienodioles quirales que actúan como sintones en la preparación de una variada gama de compuestos, principalmente polioxigenados. El trabajo desarrollado a lo largo de esta tesis se enmarca en un proyecto que utiliza los cisciclohexadienodioles para la síntesis de estructuras oligo-tetrahidrofuránicas análogas a las encontradas en acetogeninas anonáceas. Estructuralmente, estos productos naturales contienen un núcleo central formado por uno, dos y hasta tres anillos THF 2,5- disustituidos adyacentes. A su vez estas estructuras presentan un amplio espectro de actividad biológica (antihelmíntica, antimalárica, antibiótica, inmunosupresora, leishmanicida y antitumoral, entre otras). La metodología estudiada se basa en el uso de ciclohexadienodioles quirales para la preparación de anillos tetrahidrofuránicos (monómeros) mediante ciclaciones electrófilas, como haloeterificaciones y ciclaciones mediadas por Pd. Estos monómeros son usados para elaborar estructuras más complejas (bis-THF) usando la misma reacción de haloeterificación, a través de rutas sintéticas lineales y rutas convergentes. Se trata de desarrollar una estrategia iterativa y eficiente que usa reactivos simples. El análisis retrosintético del presente trabajo propone la síntesis de los sistemas centrales de acetogeninas con uno o dos anillos THF a partir de cis-bromociclohexadienodiol obtenido por biotransformación de bromobenceno. Los anillos cis-THF 2,3,5 trisustituídos se estudiaron a partir del sistema 3-butenilcarbinol acetilado en la posición alílica, el cual fue obtenido a partir de bromobenceno en 7 pasos con un 37 % de rendimiento. Mediante la reacción de Tsuji-Trost, se obtuvieron los anillos cis con buena diasteroselectividad. Con el objetivo de obtener los anillos trans-THF, se realizó un estudio exhaustivo de la reacción de haloeterificación en sistemas 3-butenilcarbinol, sintetizados a partir de bromobenceno con un 52% de rendimiento en 5 pasos de reacción. El estudio se realizó variando diferentes parámetros de reacción como ser: fuente de halógeno, disolvente, temperatura, dependencia con la base, relación de reactivos y agentes coordinantes. En todos los casos se obtuvo preponderantemente un anillo trans-THF, siendo las mejores condiciones de reacción el uso de I2 y K2CO3 en tolueno a temperatura ambiente, obteniéndose el anillo con un 79%. Asimismo, se logró desarrollar una metodología de síntesis de anillos trans-THF con 100% de diasteroselectividad a partir del sistema 3-butenilcarbinol, en 6 pasos de síntesis y 47% de rendimiento global; este trabajo fue publicado en una revista con referato. Luego de obtenido el primer anillo se procedió a la formación de los sistemas bis-THF mediante el alargamiento de la cadena lateral y posterior ciclación usando el método de haloeterificación optimizado. Para ello, se estudió la funcionalización de la cadena lateral en los anillos obtenidos, utilizando dos estrategias para la formación de la misma: a) reacción de Grignard, y b) apertura de epóxidos. Los estudios de adición de Grignard se realizaron sobre los anillos trans-THF con la función aldehído en su cadena lateral, obteniéndose una mezcla epimérica de alcoholes con una selectividad 7:3. Sobre la mezcla se ensayó la reacción de haloeterificación formándose un conjunto de sistemas bis-THF con baja selectividad. Los estudios de apertura de epóxidos en la cadena lateral de los anillos trans-THF no produjeron el producto de adición deseado sino productos de apertura mediada por halógenos. En el área de actividad biológica, se realizaron estudios de la actividad insecticida y de inhibición del crecimiento sobre líneas celulares tumorales, obteniéndose resultados prometedores para algunos de los anillos estudiados. Los mismos permitirán continuar los estudios de actividad de nuevas estructuras THF, modificando las mismas de una manera racional.
