Four new palladium(II) complexes with the formula Pd(L)2, where L are quinoxaline-2-carbonitrile N1,N4- dioxide derivatives, were synthesized as a contribution to the chemistry and pharmacology of metal compounds with this class of pharmacologically interesting bioreductive prodrugs. Compounds were characterized by elemental, conductometric and thermogravimetric analyses, fast atom bombardment mass spectrometry (FAB-MS) and electronic, Fourier transform infrared (FTIR) and 1H-nuclear magnetic resonance spectroscopies. The complexes were subjected to cytotoxic evaluation on V79 cells in hypoxic and aerobic conditions. In addition, a preliminary study on interaction with plasmid DNA in normoxia was performed. Complexes showed different in vitro biological behavior depending on the nature of the substituent on the quinoxaline ring. Pd(L1)2 and Pd(L2)2, where L1 is 3-aminoquinoxaline-2-carbonitrile N1,N4-dioxide and L2 is 3-amino-6(7)-methylquinoxaline-2-carbonitrile N1,N4-dioxide, showed non selective cytotoxicity, being cytotoxic either in hypoxic or in aerobic conditions. On the other hand, Pd(L3)2, where L3 is 3-amino-6(7)-chloroquinoxaline-2-carbonitrile N1,N4-dioxide, resulted in vitro more potent cytotoxin in hypoxia (P = 5.0 lM) than the corresponding free ligand (P = 9.0 lM) and tirapazamine (P = 30.0 lM), the first bioreductive cytotoxic drug introduced into clinical trials. In addition, it showed a very good selective cytotoxicity in hypoxic conditions, being non-cytotoxic in normoxia. Its hypoxic cytotoxicity relationship value, HCR, was of the same order than those of other hypoxia selective cytotoxins (i.e., Mitomycine C, Misonidazole and the N-oxide RB90740). Interaction of the complexes with plasmid DNA in normoxia showed dose dependent ability to relax the negative supercoiled forms via different mechanisms. Pd(L2)2 introduced a scission event in supercoiled DNA yielding the circular relaxed form. Meanwhile, both Pd(L1)2 and Pd(L3)2 produced the loss of negative supercoils rendering a family of topoisomers with reduced electrophoretic mobility. Pd(L3)2 showed a more marked effect than Pd(L1)2. Indeed, for the highest doses assayed, Pd(L3)2 was even able to introduce positive supercoils on the plasmid DNA. The inositol phosphates are ubiquitous metabolites in eukaryotes, of which the most abundant are inositol hexakisphosphate (InsP6) and inositol 1,3,4,5,6-pentakisphosphate [Ins(1,3,4,5,6)P5)]. These two compounds, poorly understood functionally, have complicated complexation and solid formation behaviours with multivalent cations. For InsP6, we have previously described this chemistry and its biological implications (Veiga et al. in J Inorg Biochem 100:1800, 2006; Torres et al. in J Inorg Biochem 99:828, 2005). We now cover similar ground for Ins(1,3,4,5,6)P5, describing its interactions in solution with Na?, K?, Mg2?, Ca2?, Cu2?, Fe2? and Fe3?, and its solid-formation equilibria with Ca2? and Mg2?. Ins(1,3,4,5,6)P5 forms soluble complexes of 1:1 stoichiometry with all multivalent cations studied. The affinity for Fe3? is similar to that of InsP6 and inositol 1,2,3-trisphosphate, indicating that the 1,2,3- trisphosphate motif, which Ins(1,3,4,5,6)P5 lacks, is not absolutely necessary for high-affinity Fe3? complexation by inositol phosphates, even if it is necessary for their prevention of the Fenton reaction. With excess Ca2? and Mg2?, Ins(1,3,4,5,6)P5 also forms the polymetallic complexes [M4(H2L)] [where L is fully deprotonated Ins(1,3,4,5,6)P5]. However, unlike InsP6, Ins(1,3,4,5,6)P5 is predicted not to be fully associated with Mg2? under simulated cytosolic/ nuclear conditions. The neutral Mg2? and Ca2? complexes have significant windows of solubility, but they precipitate as [Mg4(H2L)]�23H2O or [Ca4(H2L)]�16H2O whenever they exceed 135 and 56 lM in concentration, respectively. The cellular ligands of this biologically important ‘‘chelatable’’, ‘‘labile’’ or ‘‘transit’’ iron are not known. Proposed ligands are problematic, because they are saturated by magnesium under cellular conditions and/or because they are not ‘‘safe’’, i.e. they allow iron to catalyse hydroxyl radical formation. Among small cellular molecules, certain inositol phosphates (InsPs) excel at complexing Fe3? in such a ‘‘safe’’ manner in vitro. However, we previously calculated that the most abundant InsP, inositol hexakisphosphate, cannot interact with Fe3? in the presence of cellular concentrations of Mg2?. In this work, we study the metal complexation behaviour of inositol 1,2,3-trisphosphate [Ins(1,2,3)P3], a cellular constituent of unknown function and the simplest InsP to display high-affinity, ‘‘safe’’, iron complexation. We report thermodynamic constants for the interaction of Ins(1,2,3)P3 with Na?, K?, Mg2?, Ca2?, Cu2?, Fe2? and Fe3?. Our calculations indicate that Ins(1,2,3)P3 can be expected to complex all available Fe3? in a quantitative, 1:1 reaction, both in cytosol/nucleus and in acidic compartments, in which an important labile iron subpool is thought to exist. In addition, we calculate that the fluorescent iron probe calcein would strip Fe3? from Ins(1,2,3)P3 under cellular conditions, and hence labile iron detected using this probe may include iron bound to Ins(1,2,3)P3. Therefore Ins(1,2,3)P3 is the first viable proposal for a transit iron ligand. Among small cellular molecules, certain inositol phosphates (InsPs) excel at complexing Fe3? in such a ‘‘safe’’ manner in vitro. However, we previously calculated that the most abundant InsP, inositol hexakisphosphate, cannot interact with Fe3? in the presence of cellular concentrations of Mg2?. In this work, we study the metal complexation behaviour of inositol 1,2,3-trisphosphate [Ins(1,2,3)P3], a cellular constituent of unknown function and the simplest InsP to display high-affinity, ‘‘safe’’, iron complexation. We report thermodynamic constants for the interaction of Ins(1,2,3)P3 with Na?, K?, Mg2?, Ca2?, Cu2?, Fe2? and Fe3?. Our calculations indicate that Ins(1,2,3)P3 can be expected to complex all available Fe3? in a quantitative, 1:1 reaction, both in cytosol/nucleus and in acidic compartments, in which an important labile iron subpool is thought to exist. In addition, we calculate that the fluorescent iron probe calcein would strip Fe3? from Ins(1,2,3)P3 under cellular conditions, and hence labile iron detected using this probe may include iron bound to Ins(1,2,3)P3. Therefore Ins(1,2,3)P3 is the first viable proposal for a transit iron ligand. It is generally accepted that the blood lead level (BLL) is the best exposure index, as there is an excellent correlation between the actual exposure of the individual and the concentration of lead in blood. In Uruguay, lead contamination becomes a matter of public concern in 2001, giving rise to a sensitization in the population, which in many cases brought about a change in hygienic and dietary habits of the children. In 2004, after the leaded gasoline phasing out process in Uruguay was completed, we studied non-exposed children to correlate BLL with variables such as age, sex, area of residence, and available environmental lead data and compared these results with those from our similar screening studies 10years ago. The main result of this comparison is that BLL from children in our country had a significant decrease between 1994 and 2004. Aureobasidium pullulans was the microorganism most frequently recovered from the surface of apple fruit (cv. Red Delicious) stored in commercial cold chambers for 6 months. In the present work, 10 isolates of Aureobasidium pullulans were assayed to determine if they could control blue and grey mold disease of apple during cold storage. Although nine of 10 isolates, significantly reduced the percentage of decayed wounds when compared to the control, one of them, designated isolate ApB, showed the highest levels of protection. ApB was able to grow in a wide range of temperatures lower than 358C, which is an important human health safety factor. ApB was resistant to thiabendazole, iprodione and imazalil, the most commonly commercially applied fungicides in postharvest treatment of apples in Uruguay. Regarding the mechanisms of action of the selected biocontrol agent, lytic enzymes did not seem to play a central role. ApB depleted iron from nutrient media, which may be an important aspect of its ability to inhibit Botrytis cinerea. Further experiments are needed, however, to determine if the depletion of iron is caused by the production of siderophores, by the immobilization of iron in an insoluble pigment, or a combination of both. In the search for new therapeutic tools against parasitic diseases caused by the Kinetoplastids Leishmania spp. and Trypanosoma cruzi, a novel gold(I) triphenylphosphine complex with the bioactive coligand pyridine- 2-thiol N-oxide (mpo) was synthesized and characterized by using analytical and conductometric measurements, electrospray ionization-mass spectrometry (ESI) and electronic, FTIR and 1H and 31P NMR spectroscopies. A dinuclear structure is suggested for the complex. At a 1 microM concentration the complex induced in vitro after 30 min a potent leishmanicidal effect (LD50) against promastigotes of Leishmania (L.) mexicana while on Leishmania (V.) braziliensis with the same concentration only a leishmanistatic effect (IC75) was observed 48 h after treatment. Similar differential susceptibilities were also found when testing the ligand mpo, but at a higher dose (5 lM). In addition, the compound showed growth inhibitory effect on Dm28cT. cruzi epimastigotes in culture (IC50 0.09 lM), being even more active than the anti-trypanosomal reference drug Nifurtimox (IC50 6 lM). DNA interaction studies showed that this biomolecule does not constitute a main target for the mpo complex currently tested. Instead, the significant potentiation of the antiproliferative effect against both Leishmania species and T. cruzi could be associated to the inhibition of NADH fumarate reductase, a kinetoplastid parasite-specific enzyme absent in the host. Furthermore, due to its low unspecific cytotoxicity on mammalian cells (J774 macrophages), the new gold complex showed a selective anti-parasite activity. It constitutes a promising new potent chemotherapeutic alternative to be evaluated in vivo in experimental models of leishmaniasis and Chagas´ disease. Eight new platinum(II) complexes with 3-(5-nitrofuryl)acroleine thiosemicarbazones showing antitrypanosomal activity were synthesized, characterized and in vitro evaluated. Most of the complexes showed IC50 values in the micromolar range against two different strains of Trypanosoma cruzi, causative agent of Chagas disease (American Trypanosomiasis). In addition, most of the newly developed complexes, together with the analogous platinum 5-nitrofuraldehyde containing thiosemicarbazones previously reported, resulted more active than the reference trypanocidal drug nifurtimox on the infective trypomastigote form of the parasite. Their capacity to produce free radicals that could lead to parasite death was evaluated by ESR experiments in the parasite and by respiration measurements. Two catalysts prepared separately by iron pillaring of two different-sized particle fractions of a dried, ground and sieved clay mineral were tested for catalytic performance in photo-Fenton discoloration of methylene blue aqueous solutions. Two different-sized particle fractions of the mineral were selected prior to pillaring, ranging below 250 mm and within the range of 250– 450 mm, respectively. The resulting solids were characterized by thermogravimetric analysis, nitrogen adsorption isotherms and ultimate analysis at different preparation stages. Significant differences were found in catalyst textural parameters according to the starting mineral particle size, a higher specific surface area, and specific pore volume, was found for the catalyst obtained from the smaller-sized particle fraction of the mineral. Both catalysts showed activity in the discoloration of aqueous dye solution. Differences in catalyst performance were correlated with textural parameters. Activated carbons obtained from rice husk exhibit low specific surface areas when physical activation is applied due to its high silica content. The purpose of this work was to improve textural parameters of powdered activated carbons obtained from rice husk. To avoid the negative influence of the raw material ash content, a leaching step was included in the preparation process. Hydrofluoric acid, in two concentrations (25 and 50 wt %), was used as leaching agent and applied in different stages of the process. Physical activation using water vapor as activating agent was applied. Specific surface area and porosity were evaluated from nitrogen adsorption data. When a leaching step was included, specific surface area values between 700 and 1200 m2/g were obtained. These values are higher than that corresponding to the activated carbon prepared from rice husk not treated with acid (290 m2/g). Pore size distribution depends on the step sequence of the preparation process and on the HF concentration. The synthesis and spectroscopic characterization (UV–Vis, IR, EPR) of a new copper complex with a betablocker propranolol (1-(isopropylamino)-3-(1-naphthyloxy)-2-propanol) are presented. Besides, the Xray crystal structure of [Cu(propranolol)2]�2H2O is determined, showing two different copper ions, one coordinated through two S propranolol isomers and the other through two R isomers. The effect on the heart contraction force and on the heart rate plus the block of response to adrenaline of the complex and the free ligand, were studied. The effect of [Cu(propranolol)2]�2H2O on contractility was very similar to that of the free propranolol while the reduction on the heart rate is approximately 30% of the reduction obtained with the free ligand. This is an encouraging result since the search of new beta-blocker drugs that have lesser effect on heart rate is one of the important topics in cardiac pharmacology. The block of the response to adrenaline is at least similar for both ligand and complex. In this work we present studies on applicability of transition metal additives as sintering and electrical conductivity aids for cerium gadolinium oxide electrolyte. The nanosized Ce0.85Gd0.15O1.925 powder obtained by coprecipitation method was modified with Cr3+, Fe3+, Ni2+ or Cu2+ ions. Using high-intensity high-resolution X-ray powder diffraction data we have determined that Cr, Fe and Ni ions do not incorporate into the cerium gadolinium oxide surface or bulk when sintered at 1300 ◦C, but react with Gd ions to form Cr0.9Gd0.1O, GdFeO3 and GdNiO3 phases, while Cu incorporates in the material up to 0.7 mol% with a significant fraction of remaining material showing poorly crystalline CuO phase. The nanosized Ce0.85Gd0.15O1.925 material shows already improved sintering properties than previous reports but full sintering is not achieved below 1300 ◦C, however Cr, Fe and mainly Cu impregnation allows full sintering at 1300 ◦C. 0.5 mol% Ni impregnated material sintered at 1500 ◦C shows enhanced grain boundary conductivity that probably indicates that Ni incorporates into Ce0.84Gd0.15O1.925 above 1300 ◦C. The global results indicate, however, that optimization of ceria microstructure is at least of equal importance for sinterability and grain boundary conductivity than impregnation of the material with transition metal ions.
