A summary of the resource.
After reversible immobilization of neutral beta-galactosidase from Kluyveromyces lactis on to thiolsulfinate/thiolsulfonate supports, more than 80 % of the activity was retained. Blocking the remaining reactive groups with glutathione increased the thermal stability of the derivatives almost two-fold. These derivatives achieved a high degree of conversion (85-90 %) of lactose (50g / l) in saline solution, whey, whey permeates, and skimmed milk, either batchwise or in packed beds. They remained fully active upon storage for 10 months in activity buffer at 4 deg C, and when treated with sanitizing agents.

A summary of the resource.
The behaviour of wheat β-amylase from crude extracts in chromatography on agarose gels substituted with different ligand types was investigated. The enzyme displayed high salt promoted adsorption onto thiophilic gels provided with sulfone-thioether and 2-thiopyridine ligands. Quantitative recovery of the enzyme was easily accomplished by elution with buffer in the absence of Na2SO4. The 3-(2-pyridylthio)-2-hydroxypropylagarose (PyS-gel) also allowed elimination of pigments present in the wheat extract. These pigments showed no adsorption onto the gel, thus regeneration is easily achieved, allowing its re-use. The enzyme also displayed strong salt-dependent adsorption onto adsorbents provided with pyridyldisulfide moieties, but in this case enzyme binding was due to its thiol content since elution was achieved mainly through reduction with DTT. When the enzyme was chromatographed on a series of hydrophobic alkyl ligands in the presence of 0.5 M sodium sulphate, it was partially adsorbed on pentylagarose and quantitatively adsorbed on hexyl-agarose, elution being easily performed by sodium sulphate-free buffer. This behaviour was markedly different from that towards phenyl-Sepharose, to which the enzyme was strongly adsorbed and which required much more drastic elution conditions.

Abstract
A summary of the resource.
An ab initio Hartree-Fock and second-order Møller-Plesset study of the radical cation obtained by ionization of the formaldehyde-HF hydrogen-bonded dimer is reported. The results are compared with those obtained previously for the radical cations obtained from the formaldehyde-water complex. An exploration of the [H2CO · HF]+· potential energy surface was done using the 3-21G basis set. The whole of the structures found were then refined using the 6-31G(d,p) basis set and — for the most interesting structures — 6-31+G(d,p) and 6-311++G(2d,2p). In all cases the calculations were performed using the unrestricted formalism, checking always that spin contamination remained below normal thresholds. Although most of the structures found correspond closely to those found in the case of water, the HF acceptor is not able, in general, to abstract the proton from formaldehyde. Thus, contrary to the case of water, the most stable CH2O · HF radical-cation complex has both the charge and the unpaired electron located on the formaldehyde fragment. However, HF behaves in the same fashion as H2O in the sense that it is able to stabilize the hydroxymethylene radical cation with respect to the formaldehyde radical cation.

A summary of the resource.
The levels of superoxide dismutase (SOD) were determined in detergent-soluble, somatic and excretion–secretion (E–S) preparations from adult Fasciola hepatica using the xanthine oxidase system and visualized in substrate gels. Compared to detergent-soluble and somatic extracts, E–S products showed the highest SOD activity (88 ·5 U/mg), indicating active release to the medium in which parasites were maintained. SOD specific activity was also detected at high levels in E–S products from 3-week-old and 5-week-old immature migrating flukes (25 and 143 U/mg, respectively). In all preparations except for the somatic extract, the activity was characterized as cyanide-sensitive CuZn SOD. Differences in SOD isoenzyme profiles between the extracts were observed in native polyacrylamide gel electrophoresis: the somatic and detergent-soluble extracts exhibited 1 band of activity while the E–S products from immature and adults flukes contained 2 and 3 migrating bands, respectively. SOD was purified from the detergent-soluble extract and E–S products of adult worms by a combination of ultrafiltration, gel filtration on Sephacryl S-200 HR and ion-exchange chromatography on QAE Sephadex A-50. The SOD from detergent-soluble extract showed, by SDS–PAGE analysis, 1 band of 16 kDa apparent molecular weight. The SOD from E–S products showed 2 bands of 16 and 60 kDa apparent molecular weight. N-terminal sequence analysis of the 16 kDa band from the detergent-soluble preparation showed some similarity with Schistosoma mansoni cytoplasmic SOD. These enzymes may have a potential role in the evasion of the oxidative burst killing mechanism by immune cells.

Abstract
A summary of the resource.
A series of compounds bearing an endocyclic –N–O– moiety with potential antimalarial activity based on simple derivatives of the tropolone purpurogallin was prepared by means of a hetero Diels–Alder reaction using nitrosobenzene as a dienophile. The rationale behind the design of these compounds is presented, together with the synthetic route to derivatives bearing aromatic and aliphatic esters of the C4′-position hydroxyl group of the purpurogallin framework, as well as biological data obtained from in vitro assays against Plasmodium falciparum and Trypanosoma cruzi. Several of the new compounds have activities in the 3–9 μM range, and provide leads for the development of a novel class of antiparasitic drugs with improved biological and pharmacological properties.

Abstract
A summary of the resource.
Ligand exchange is one of the possible synthetic routes to obtain99mTc coordination compounds. However, the success of this route depends on the availability of good precursors. The objective of this work is the preparation of the complex [99mTc(tu)6]3+ (tu = thiourea), as a potential precursor for99mTc(III) coordination compounds. The preparation was successfully performed in acidic conditions, the excess of tu serving as reducing agent. At pH values higher than 3, the compound becomes unstable and on addition of polydentate ligands new Tc(III) complexes are formed. With edta, the complex99mTc(III)-edta was obtained in high yield.

Efflux pumps can make a significant contribution to the capacity of bacteria to resist the action of antibiotics. Certain efflux pumps also recognize antimicrobial agents that are present in their respective hosts and their ability to export toxic agents could enhance bacterial survival during infection prior to appearance of cellular or humoral host defensive systems. This review is concerned with the principal efflux pumps possessed by two closely related strict human pathogens, Neisseria gonorrhoeae and Neisseria meningitidis. Specific emphasis is placed on the organization of the structural genes encoding the mtr and far efflux pumps, the substrates (often host- derived) recognized by these pumps, and the cis- and trans -acting transcriptional factors that regulate efflux pump gene expression in gonococci and meningococci. The overriding theme of this review is that the efflux pumps possessed by these pathogens likely contribute to their pathogenic mechanisms by providing a means to escape a number of antimicrobial compounds that bathe mucosal surfaces

Abstract
A summary of the resource.
AM1 calculations were performed for the absorption of H2O and CO2 molecules on the surface of model ZnO crystals. The absorption of isolated molecules of each species and the co-absorption of both compounds simultaneously were considered. It was found that the absorption of H2O near a site where CO; is already absorbed favors the process of sintering, in agreement with the experimental findings. This is explained by the formation of Zn(OH)CO3H bound to the surface, a more mobile species than the ZnO unit itself. The roundening of the grains observed in atmospheres containing dry CO2 but suppressed when H2O is present, is also explained by these calculations. After absorption of CO2, the rupture of one bond - so that diffusion of the ZnCO3 species on the surface is allowed - requires much less energy than the breaking of two bonds, necessary for ZnO migration. These facts explain why the speed of surface transport does not decrease in CO2 atmospheres while sintering is indeed slowed down.

Here we describe the identification of Toxoplasma gondii circulating antigens in sera of BALB/c mice experimentally infected with either the virulent RH strain, or the cystogenic WTD1 strain or with an isolate from a human patient. The circulating antigens were identified by immunoblot in tachyzoite (RH strain) and in tissue cyst (ME-49 strain) crude antigens, using antibodies produced by immunisation of BALB/c mice with homologous sera from infected animals. The most relevant tachyzoite antigen identified are in the following four clusters of 109–94, 67–57, 35–31 and 28–21 kDa. Tissue cyst-specific circulating antigens, like the 18 kDa one, were detected in sera from mice infected with the cystogenic strains. These immune sera, after depletion of tachyzoite specific antibodies, recognised three tissue cysts antigens with Mr of 120, 79 and 48 kDa, and a cluster of antigens in the range of 68–53 kDa. We produced monoclonal antibodies by fusion of myeloma cells with lymphocytes from the mouse immunised with circulating antigens from the RH strain. One of the clones (3A11/H12) obtained, secretes IgG1 and recognises a peptide epitope from a tachyzoite 67 kDa protein. This parasite protein also binds irrelevant mouse IgG1 as well as immunoglobulins from other species. The reactivity with non-specific antibodies was inhibited by preincubation with 2% normal mouse and goat serum, while the reaction with the monoclonal antibody 3A11/H12 was not. Furthermore, a biotinylated F(ab′)2 of an irrelevant mouse IgG1 did not show any reactivity while the F(ab′)2 of the monoclonal antibody 3A11/H12 reacts specifically with the 67 kDa antigen suggesting that this circulating antigen is a putative Fc binding protein.

A summary of the resource.
A method termed "PERFEXT", based on sequential perfusion and detergent extraction of lymphoid and non-lymphoid organs, has been developed for the quantitative measurement of cytokines produced at a local level in a given tissue. In vivo treatment of mice with Staphylococcus enterotoxin B (SEB) or lipopolysaccharide (LPS) served as the model systems. Interleukin-2 (IL2) and interferon-gamma (IFN gamma) levels were monitored by ELISA analysis of extracted samples. After local footpad (FP) injection with SEB, spleen and serum IL2 levels peaked at 2-4 h, while IL2 levels peaked at around 4-8 h in both FP and popliteal lymph nodes. SEB injection resulted in increased IFN gamma levels both in the FP and the draining lymph node. The detection of cytokines in the intestine allows for the application of the method at mucosal sites as well, provided enzyme inhibitors are present during the extraction procedure. After FP injection with LPS, IFN gamma production was significantly increased in the draining lymph node and was detectable in the FP, whereas IL2 was undetectable in any organ examined. IL2 and IFN gamma could also be detected at the site of elicitation of a delayed-type hypersensitivity reaction following local FP challenge. Local cytokine production correlated with the swelling response, whereas cytokine production in the spleen did not. IL2 peaked early, followed by a late increase in IFN gamma production, corresponding to the maximum swelling. This simple method should prove useful for analysing the production of other cytokines in vivo in distinct anatomical compartments.

<p>This study compared the possibilities and limitations of 99mTc-labeled synthetic peptides derived from two human antimicrobial peptides, namely, ubiquicidin (UBI) and lactoferrin (hLF), for the scintigraphic detection of bacterial and fungal infections in mice and rabbits. The rationale of our approach was that selected peptides accumulate in infected areas but not in sterile inflammatory lesions, because they bind preferentially to microorganisms. 99mTc-labeled human neutrophil peptides (defensins), ciprofloxacin, and human polyclonal IgG were included as control agents.</p>
<p>METHODS: 99mTc-labeled peptides and control agents were injected intravenously into animals that had been injected intramuscularly 18 h earlier with multidrug-resistant Staphylococcus aureus, Klebsiella pneumoniae, or fluconazole-resistant Candida albicans. Sterile inflammatory sites were induced by the injection of heat-killed microorganisms or lipopolysaccharide (LPS) into the thigh muscle. Up to 4 h after injection, the accumulation of 99mTc-labeled compounds in the infected/inflamed thigh muscles was determined using scintigraphic techniques and radioactivity counts in dissected tissues.</p>
<p>RESULTS: Scintigraphy revealed that 99mTc-labeled peptides UBI 29-41, UBI 18-35, UBI 31-38, hLF 1-11, and defensins, which showed preferential in vitro binding to microorganisms in a former study, accumulated at a significantly higher rate (P < 0.01) in bacterial and C. albicans infections in mice and rabbits than in inflamed tissues induced by heat-killed microorganisms or by LPS. No significant difference in the accumulation of 99mTc-labeled ciprofloxacin was observed between infected and sterile inflamed thigh muscles in mice.</p>
<p>CONCLUSION: 99mTc-labeled antimicrobial peptides UBI 29-41, UBI 18-35, UBI 31-38, hLF 1-11, and defensins accumulate significantly in tissues infected with gram-positive and gram-negative bacteria and C. albicans. Significantly lower (P < 0.01) accumulation of these peptides occurs in sterile inflamed tissues. These data indicate that the peptides preferentially tag microorganisms at the site of infection, which is in agreement with their preferential binding to the microorganisms in vitro and in vivo. 99mTc-labeled ciprofloxacin does not distinguish between infections and sterile inflammatory lesions, which implies that its specificity for the detection of bacterial infections is not warranted.</p>

The geometries and binding energies of CO2 adsorbates on Pt(100) and Pt(111) cluster surfaces were calculated by means of an improved version of the extended Hückel molecular orbital method. The polarization of the surface by an applied electric potential and coadsorption of H atoms were included in the model. For simulated applied potentials in the range −1.0 to 1.0 V, CO2 coordination geometries (side-on, formate) involving two adsorbate atoms bonded to the surface are favored, regardless the surface topology and the presence of coadsorbed H atoms. In agreement with experiment, larger binding energies are always calculated for the Pt(100) cluster surface.

Abstract
A summary of the resource.
La Enfermedad de Chagas es una enfermedad no resuelta, causada por el parasite Trypanosoma cruzi. Existe desde hace miles de anos y afecta a millones de personas, mayoritariamente en America Latina. Hasta el momenta no existe un farmaco adecuado para tratar est a enfermedad. Con el objetivo de buscar terapias apropiadas para est a problematica, en este trabajo se disenaron, sintetizaron y caracterizaron ochenta y cinco nuevos compuestos. Se determinaron sus capacidades tripanosomicidas in vitro obteniendose mas de veinte derivados con destacada actividad anti-Trypanosoma cruzi. Se estudi6 la toxicidad de los mismos frente a celulas de mamffero determinandose la selectividad de los derivados biol6gicamente activos. Se realizaron estudios de mutagenicidad in vitro. Se realizaron estudios in vivo con tres de los derivados mas relevantes. Se identific6 un derivado con potencialidad a farmaco que posee varias ventajas. Desde el punta de vista qufmico: sencillez sintetica, producto escalable y de producci6n econ6mica. Desde el punta de vista biol6gico: activo en diferentes estadfos y cepas del parasito, baja toxicidad inespedfica, ausencia de mutagenicidad, actividad in vivo similar a la del farmaco de referencia y ausencia de genotoxicidad in vivo. Ademas, posee un mecanismo de acci6n multiple, interfiriendo en la biosfntesis de esteroles de membrana, e inhibiendo levemente las enzimas cruzipaina y tripanoti6n sintetasa.

Exposure to arsenic (As) is considered a serious environmental public health problem in many countries, but in Uruguay has only been considered in occupational health. The aim of this work is to present the advances of the analytical development of feasible methodologies for the determination of toxicologically relevant species of As in both occupationally exposed and non- exposed populations, as an alternative for more expensive techniques such as HPLC-HG-ICPMS. These analytical methodologies are useful to have the scientific evidence of population exposure to low doses of arsenic in Uruguay in order to prevent long-term health effects and establish a basis for the development of new scientific research

<p>Se estudian varios medios de cultivo para Trichomonas vaginalis, con fines didácticos: Boiron, C.P.L.M., Kupferberg y Loeffler. De todos ellos, el medio Boiron dio resultados mas constantes. El medio C.P.L.M. propició la reproducción del protozoario en forma menos notoria y regular. Los medios restantes dieron resultados negativos.</p>
<p>En todos los casos, la técnica de toma de la muestra, el lapso transcurrido entre la estracción y la siembra, la magnitud del inóculo, las condiciones de anaembiosis parcial del cultivo y la frecuencia y densidad de los repiques, son condicionantes a tener en cuenta.</p>
<p>El cultivo a 30º permite una vitalidad el protozoario algo más prolongada que a 37ºC.</p>
<p>Particularmente en el medio C.P.L.M., se aprecian las formas de división de <em>T. vaginalis</em>, incluyendo los elemtneos redondeados conteniendo dos células hijas.</p>
<p>El pasaje recíproco del flagelado entre estos dos medios es posible sin inconvenientes.</p>
<p>El medio C.P.L.M. no es tan apropiado como meterial de partida para la confección de frotis teñidos, pues el agar que contiene crea un fondo poco propicio para el contraste.</p>
<p>Ninguno de los dos medios son económicos ni de fácil preparación. Se continuarán los estudios para resolver estos aspectos.</p>

Abstract

Supermarkets have become the most important provider of food products worldwide. However, empirical evidence about how consumers make their food purchase decisions in this environment is still scarce. The present field study aimed to: i) explore how people make their in-store food purchases, and ii) identify the information they search for when making those purchases. Consumers (n = 144) were intercepted when entering the facilities of three supermarkets in two Uruguayan cities. They were asked to wear a mobile eye-tracker while they made their purchases as they normally do. The great majority of the consumers bought at least one food product or beverage (92%) and, on average, examined products from 2.8 sections. In total, they investigated 37 categories within 13 self-service sections, corresponding to 26 categories of ultra-processed products. For 67% of the products, consumers went straight to the product they seemed to be looking for, grabbed it and put it in their shopping basket or cart, without making any comparison among products. A limited information search was observed. On average, consumers spent 22 s examining products within self-service sections and only 6.9 s were elapsed from the moment they grabbed a product until they put it in the shopping cart. These results provide empirical evidence of the habitual nature of supermarket food purchases in a context characterized by wide availability of ultra-processed products. Taken together, the findings suggest that policies and interventions aimed at reducing purchases of ultra-processed products should disrupt habitual decisions at the point of purchase. In this sense, policies targeted at introducing salient changes on food packages hold potential to disrupt food purchases and encourage consumers to establish new and more healthful food purchase habits.
