Dendrimers are branched nanomolecules, with a three dimensional structure, very low polydispersity and high functionality. Poly(amidoamine) (PAMAM) dendrimers are the most investigated class of dendrimers. In this study, PAMAM G4 dendrimer conjugated with HYNIC (hydrazinonicotinamide), an efficient bifunctional chelator, was characterized. Structure of the derivatized dendrimer was confirmed by (1)H-NMR and (13)C-NMR spectra and MALDI-TOF mass spectrometry. HYNIC-dendrimer was labeled with technetium-99m testing three different co-ligands (tricine, nicotinic acid and ethylenediaminodiacetic acid). The radiolabeled complexes were characterized by reverse phase HPLC, as well as their stabilities. Radiolabeling yield was about 99% with all co-ligands and complexes were found stable for 24 h. Biodistribution studies were performed administrating tricine-(99m)Tc-HYNIC-dendrimer, nicotinic acid-(99m)Tc-HYNICdendrimer and EDDA-(99m)Tc-HYNIC-dendrimer to normal mice; results showed blood clearance with hepatic and renal depuration in all cases. In this sense, labeling of PAMAM G4 dendrimer with technetium-99m using HYNIC could be obtained in high yield in a simple method and with high specific activity. Rice paddy soils are an important biogenic source of methane, a relevant greenhouse gas. Rice rotation with upland crops and pastures affects hydrolytic and fermentative bacteria and methanogenic archaea by sequential exposition to oxic and anoxic conditions. There are several ways to reduce methane emissions from rice paddy soils. Here we examine the effect of intensification in rice rotations with upland crops upon the microbial community structure and methanogenesis when dicarboxylic acids exudated by rice root were amended in microcosms assays. Soils under rotations rice-pasture, rice-soybean and continuous rice, with the last two more intensive systems recently installed in the field experiment, were sampled. Amplicon sequencing of 16S rRNA gene analysis showed that the rotation system was a main driver of the microbial community structure, explaining 33%, 38% and 55% of the differences in the structure of Bacteria, Archaea and methanogens, respectively. The density of mcrA gene copies was significantly higher in continuous rice soil than in other soils, and the relative abundance of methanogenic archaea was 33% for rice monoculture, whereas represented 15% and 14% for the rice-pasture and rice-soybean soils, respectively. The incubation of soils with tartrate and succinate confirmed functional differences among soils since rice monoculture showed similar or higher methane production and significantly less acetate and propionate accumulation than other soils. The T-RFLP analysis of mcrA gene in soils and incubated soils revealed that Rice Cluster I increased after incubation in all rotational soils, whereas Methanosarcinaceae and Methanobacteriaceae remained relevant after incubation. Furthermore, a more stable community of methanogenic archaea was established in soil under continuous rice, since T-RF profiles of soils and incubated soils with dicarboxylic acids grouped together with 62% similarity, while communities from other rotational soils experimented a great shift after incubation with these substrates. Altogether, these results showed that the conversion of rice-pasture rotational fields to a more intensive rice crop production strongly impacts the methanogenic community structure and their potential for methane emission. The production of several oxygenated derivatives of limonene was carried out by using orange oil and molecular oxygen as the sole initial reactants under solvent-free conditions. Palladium–copper supported on carbon and hydrotalcite-like compounds displayed good catalytic properties to accelerate autoxidation reactions of limonene yielding a mixture of the different oxygenated derivatives, mainly 1,2- and 8,9-epoxylimonene, 1,2,8,9-diepoxylimonene, carveol and carvone. Palladium supported on copper–magnesium–aluminium hydrotalcite displayed the best properties due to its higher catalytic activity and lifetime. The pelletized form of these catalysts can be easily recovered and reused. A reactor design combining recirculation and oxygen bubbling through a catalytic bed of the pelletized catalyst produced the highest reaction rate. Vacuum distillation of the final product mixture allowed easy separation of the 1,2/8,9-limonene epoxides. The other required compounds, mainly carveol and carvone, were not completely separated and remained together with the other oxygenated products, such as the di-epoxylimonene isomers. The goal of the present work was to study the effects of acid treatment on the foaming properties of a soybean protein isolate (SPI) and its fractions, glycinin (11S) and β-conglycinin (7S). The structural characteristics, interfacial properties, foaming capacity and stability of the treated proteins were studied. Results from surface hydrophobicity and differential scanning calorimetry (DSC) showed that the acid treatment caused the complete denaturation of 11S and a partial denaturation of 7S. This protein unfolding affected their interfacial properties, which led to an improvement in the foaming properties of both protein fractions and isolate. Treated 7S showed the best behavior in the rearrangement process, probably due to its smaller size and its modified structural characteristics. All treated proteins showed stronger interfacial films. The foams of treated proteins were destabilized mostly due to gravitational drainage rather than Ostwald ripening. Electronic, lipophilic and steric descriptors included in QSAR-2D and -3D are analyzed for a set of ortho- and para-naphthoquinones that have proved to be powerful oxidative agents with potent trypanocidal activities specially against Leptomonas seymouri and Trypanosoma cruzi. Electronic properties are calculated by means of semiempirical (PM3), ab initio (HF/3-21G) and density functional theory (B3LYP/6-31 + G&lowast;) methodologies. Three different electronic states, neutral quinones, hydroquinones and semiquinones, are studied to investigate if any one of them are statistically related with the biological activities. The best correlations were obtained at the B3LYP level of theory because it includes electronic correlation. The QSAR-2D indicates that the best trypanocidal growth inhibitors are molecules in the semiquinone electronic state, with the following properties: (a) high negative value of EHOMO, (b) high negative charge in the oxygen atoms of the carbonyl groups, (c) high positive charge in the carbon atom of one of carbonyl moieties and (d) high electronegativity (χ). In a complementary way, the QSAR-3D indicates that the electrostatic field correlates with trypanocidal activity and the presence of bulk moieties would increase activity. The idea of comparing the three electronic states may prove to be of most importance in the general strategy to the design of new trypanocidal drugs. In fact, the experimental results showed that semiquinone is the one really statistically relevant indicating a clear connection between biochemical and theoretical aspects. Finally, we demonstrated that to be a good anti-trypanosomatid compound, the molecule must be a good electron acceptor to reach easily the essential semiquinone state. We expect that the present results motivate new experimental as well as theoretical investigations that confirm our findings. Consumers have a limited capacity to process all the information they face when deciding their food choices and usually rely on effortless and intuitive thoughts. According to the cognitive-experiential self theory human behavior is determined by the interaction of the two information processing systems: rational and intuitive. Consumers differ in their preference for these two thinking styles, which leads to differences in how they make their decisions. The aim of the present work was to evaluate the influence of rational and intuitive thinking styles on consumer choices and information processing when evaluating yogurt labels. Seventy-one consumers completed a choice-conjoint task with yogurt labels. Three 2-level variables were considered: fat and sugar content (low vs. high), label background (Background A vs. Background B), and traffic light system (present vs. absent). Consumers were presented with 8 choice sets composed of two labels on a computer screen, and were asked to indicate which one they would consume. Their eye movements were recorded during the task using an eye-tracker. After completing the conjoint task, consumers completed the Rational Experiential Inventory (REI). Latent class cluster analysis identified two consumer segments with different preference for analytic-rational or intuitive-experiential thinking styles. Consumers who predominantly relied on analytical-rational thinking engaged on a greater information search and a more thoughtful analysis of nutritional information for making their choices than consumers who preferred intuitive-experiential thinking. Besides, thinking style had a strong influence on the part-worth utilities of the evaluated variables. Results from the present work provide preliminary evidence about the influence of thinking style on consumer food choices, which can have potential implications for the design of communication strategies aimed at changing dietary patterns. Labels can largely influence functional foods consumers' purchase decisions. The aim of the work was to study consumers' attention to functional food labels and to evaluate differences between regular and functional products probiotic milk as case study. Four labels were designed considering two types of product (regular milk vs. probiotic milk) and two label backgrounds. Sixty consumers were asked to look at the labels while their eye movements were recorded using an eye-tracker and to complete a word association task. Then, they had to complete eight flicker change detection tasks, involving four different changes on key aspects of the labels (brand, type of product, type of microorganism and health claim) for each label background. Visual processing of the labels was not largely affected by the type of product and label design. Health claims were not comprehensively processed, probably due to the high information density of this area. Besides, consumers' health-related associations were generated by graphic design and not by the functional aspect of the products, suggesting that graphic design plays a key role in shaping health-related associations. Recommendations for the design of functional food labels are discussed. En este trabajo, describimos el hallazgo de una nueva mutación (–47C>A) en la región promotora del gen del receptor de las lipoproteínas de baja densidad (rLDL), principal encargado de este fenotipo. Esta mutación se identificó en una familia uruguaya con un fenotipo típico de HF, en la cual aparecen individuos heterocigotos y homocigotos para la mutación debido a la existencia de consanguinidad. El cambio nucleotídico ocurre en un sitio conservado y funcionalmente relevante del promotor; región en la que anteriormente se han descrito diversas mutaciones que provocan descensos drásticos en la actividad transcripcional del gen. Ultrasonication is known to induce cavitation and can thus accelerate extraction, dissolution, digestion, and other processes, while the intense mixing caused by the propagation of ultrasound waves enhances analyte transfer to the extraction medium. Consequently, ultrasound-assisted extraction (UAE) is viewed as a simple procedure well suited for the extraction and subsequent determination of trace elements in food. Herein, we use different techniques to characterize several processes involved in probe- and bath-based UAE, apply them to the determination of trace metals (Cd, Cu, Ni, Pb, and Zn) in globe artichoke leaves and soybean seeds, and closely monitor the extraction rates of selected analytes. The developed UAE-based method is found to comply with the criteria of Green Chemistry and is concluded to be a reliable, simple, and cheap alternative to standard food analysis techniques. Consumer‐based methods for sensory product characterization continue to gain popularity. Considering that past research is mostly limited to cases where samples are sensorially simple or markedly different, the aim of this study was to compare descriptive analysis (DA) by trained assessors with product characterizations from consumers using rate‐all‐that‐apply (RATA) questions using samples that can be regarded as quite similar or quite complex. The results from three studies showed that sample differentiation was higher in DA than RATA and that information about sample similarities and differences tended to be dissimilar. The exception was Study 1 where samples were least complex. The discrepancy between the two types of panels was least evident for appearance and color attributes and basic tastes (sweet, sour, and salty), and in these instances, the correlation between attribute intensities was clearly linear (R² = 0.46). The discrepancy was highest for complex sensory terms and terms relating to specific odors/flavors. While past research suggests that it may be possible to improve the agreement between the two methods by providing consumers with short training (samples, terms, and rating scale), practitioners should consider that training of consumers may reduce the ecological validity of their responses. Practical applications Characterization of the properties of products is a cornerstone activity for sensory professionals, and to achieve this task, several methods are available. This study contributes insight regarding the choice of descriptive analysis (DA) with trained assessors or rate‐all‐that‐apply (RATA) questions with consumers. If the products are not sensorially complex, then product characterization by consumers provide the same insights as DA. However, this study shows that when samples are sensorially complex and/or similar, RATA is less discriminative than DA and provides information about sample similarities and differences that is not the same as that generated by DA. The calculated binding energies and free energy corrections suggest that the addition of azomethine ylide to perfect graphene is not spontaneous (�G � 0). However, the presence of Stones–Wales defects significantly increases reactivity: the binding energy between SW-defective graphene and the azomethine ylide is 0.83 eV, close to that determined for a (5,5) SWCNT. The electronic properties of the sheet are not modified by the 1,3 cycloaddition. We analysed specific antibody (Ab) and circulating antigen (CAg) profiles along experimental mouse infection using as control a group of mice immunized with intact but dead parasites. Results from this experiment showed an early major CAg peak followed by a larger Ab peak which partially overlaps with other minor CAg peaks. These results suggest that CAg may be a marker of early mouse infection. In order to study the relevance of these findings in humans we similarly analysed by ELISA 148 sera provided by retrospective post-surgical follow-up of 19 patients. Available records showed that 14 patients developed new cysts one to ten years after surgery while no new disease was observed in the other five. Some of the former patients showed CAg as early as two months after surgery while no CAg was observed in the other five patients at any time. In addition, a collection of 38 sera obtained before surgery were similarly tested and five of them showed only CAg, while 18 showed only Ab and 12 sera showed Ab&CAg. These results in humans are consistent with the findings in the mouse experimental model and suggest that CAg may be an early marker of hydatid infection, thus being relevant for post-surgical follow-up. Antimicrobial peptides (AMPs) play an essential role in plant defense against invading pathogens. Due to their biological properties, these molecules have been considered useful for drug development, as novel agents in disease therapeutics, applicable to both agriculture and medicine. New technologies of massive sequencing open opportunities to discover novel AMP encoding genes in wild plant species. This work aimed to identify cysteine-rich AMPs from Peltophorum dubium, a legume tree from South America. We performed whole-transcriptome sequencing of P. dubium seedlings followed by de novo transcriptome assembly, uncovering 78 AMP transcripts classified into five families: hevein-like, lipid-transfer proteins (LTPs), alpha hairpinins, defensins, and snakin/GASA (Giberellic Acid Stimulated in Arabidopsis) peptides. No transcripts with similarity to cyclotide or thionin genes were identified. Genomic DNA analysis by PCR confirmed the presence of 18 genes encoding six putative defensins and 12 snakin/GASA peptides and allowed the characterization of their exon-intron structure. The present work demonstrates that AMP prediction from a wild species is possible using RNA sequencing and de novo transcriptome assembly, regarding a starting point for studies focused on AMP gene evolution and expression. Moreover, this study allowed the detection of strong AMP candidates for drug development and novel biotechnological products.
