A modified and miniaturized SweEt/QuEChERS method for pesticide residue analysis applied to vine leaves is presented. The deep-frozen plant material was cryogenically processed. A 2 g analytical portion was hydrated for 30 min and extracted with acidified ethyl acetate after buffering with NaHCO3 and adding Na2SO4. A dispersive solid-phase (d-SPE) cleanup step with primary–secondary amine (PSA) was performed. The pesticide residues were determined using GC-MS/MS. The whole procedure was validated for 54–59 pesticides at 0.01, 0.02, 0.1, and 0.2 mg/kg in fresh vine leaves (Vitis vinifera). The key method performance parameters investigated were specificity, linearity, trueness, within laboratory repeatability and reproducibility, limit of detection, limit of quantitation, and matrix effects. Recoveries for the 59 pesticides tested ranged from 60 to 110%, and the RSDs were lower than 20% for the majority of the pesticides studied. The inclusion of front of pack (FOP) nutrition labels is one of the strategies that has been proposed to encourage people to make healthful food choices, helping to cope with the increasing burden of overweight and obesity among adults and children. The aim of this study was to evaluate the effect of different FOP labels on Brazilian children and parents’ healthfulness perception. Children aged 6–12 years (n = 318) and parents (n = 278) with different socio-economic status rated their perceived healthfulness of eight food products targeted at children using a 7-point scale (1 = not healthful and 7 = very healthful). Participants were randomly allocated into one of the three groups of FOP nutrition labels: Daily Guideline Amounts (GDA), Traffic Light System (TLS) and warning system. Data were analyzed using analysis of variance. For parents, products with the warning system were rated significantly less healthful than those containing the GDA, whereas the TLS did not significantly differ from the other two systems. Age and socio-economic status influenced the effect of FOP labels on children’s perceived healthfulness. Only 9–12 years old children from middle/high socio-economic status were influenced by FOP labels: the warning system and TLS reduced healthfulness perception of frosted corn flakes compared to the GDA system. These results suggest that directive and semi-directive FOP have the potential to modify healthfulness perception of products targeted at children and stress the need to study the modulating effect of individual variables such as age and socio-economic status on the perception of FOP labels. The impacts of the modern, agrochemicals based agriculture that threatens the overall systems sustainability, need to be monitored and evaluated. Seeking for agroecosystems monitors, the present article focus in the occurrence and abundance of aquatic macroinvertebrates, that have been frequently used as bioindicators of water quality due to their relationship with land use. Some of these organisms are on the top of the food chain, where bioaccumulation and biomagnification processes can be observed, and they can turn into secondary pollution sources of systems and terrestrial organisms as well. Odonate nymphs, which belong to the functional group of predators, were selected for this study. A methodology to determine 73 pesticide residues in odonate nymphs by LC-MS/MS and GC-MS/MS was developed. A QuEChERS sample preparation strategy was adapted. As it is complex to obtain samples especially in disturbed ecosystems, the method was minimized to a sample size of 200 mg of fresh nymphs. The method was validated and good recoveries (71–120%) with RSDs below 20% for the majority of the studied pesticides at least at two of the assayed levels 1, 10 and 50 µg kg−1 were obtained. For 32 analytes the limit of quantitation was 1 µg kg−1 and 10 µg kg−1 for the others. The lineal range was observed between 1–100 µg kg−1 in matrix-matched and solvent calibration curves for most of the assessed pesticides. LC-MS/MS matrix effects were evaluated, 40% of the analytes presented low or no signal suppression. Only flufenoxuron presented high matrix effects. The obtained methodology is adequate for pesticide multiresidue analysis in aquatic macroinvertebrates (odonates) aiming to contribute to the ecological state evaluation of freshwater ecosystems. La metodología propuesta se fundamenta en el estudio del estado del arte del aprendizaje de física moderna en la enseñanza secundaria de Uruguay así como del análisis de diferentes fuentes de información: programas de cursos de física moderna de nivel terciario universitario y de formación docente y sus libros de texto más comunes, la opinión de informantes claves, programas de física y química de bachillerato de secundaria y metodologías de enseñanza con un marco conceptual coherente con el constructivismo. La propuesta metodológica incorpora experimentos que requieren responder preguntas desafiantes para estudiantes secundarios. Varios de los experimentos aquí desarrollados fueron implementados en dos cursos de tercero de bachillerato de secundaria, uno de opción científico-matemático y el otro de opción ciencias-biológicas. Los ejecutores de las propuestas fueron los docentes de estos cursos, en el científico-matemático un profesor de Química y en la opción ciencias-biológicas un profesor de Física. El análisis de los resultados de su implementación en el aula muestra que los estudiantes se ven atraídos y motivados por el desafío de responder preguntas a través de la experimentación y aprender con y del otro. Los estudiantes fueron capaces de medir cuidadosamente y analizar los datos obtenidos utilizando planillas de cálculo. Es importante destacar que, la introducción de Tecnologías de la Información y Comunicación (PC y teléfonos inteligentes, entre otros dispositivos) para realizar los experimentos propuestos, permite disponer de un laboratorio simple, económico y al alcance de todos. Permitiendo realizar experimentos con distintos grados de Metodología para el aprendizaje de Física Moderna para Educación Secundaria con Química 8 José Di Laccio (2018) sofisticación, que los estudiantes lo pueden realizar en el aula, un parque, en sus propias casas o en el lugar que lo deseen y en el momento que lo prefieran. Varios de los proyectos desarrollados en el contexto de la presente tesis fueron publicados en diversas revistas. Las propuestas educativas desarrolladas aquí, han tenido buena acogida en la comunidad educativa regional, en su presentación en diferentes congresos de enseñanza de las ciencias. El estudio de los contaminantes en los alimentos y el medioambiente es uno de los campos que mayor desarrollo ha experimentado en los últimos tiempos tanto en aspectos regulatorios como analíticos. La diversificación de las actividades económicas y humanas tendientes hacia una sociedad de consumo, ha generado una variación en las características y clases de contaminantes orgánicos que atentan contra la calidad y sustentabilidad de los recursos. El presente proyecto de tesis tuvo por objetivo el desarrollo y evaluación de métodos y estrategias analíticas cualitativas y cuantitativas para la determinación de compuestos orgánicos de relevancia sanitaria y/o ambiental presentes a bajas concentraciones en diferentes matrices como materias primas farmacéuticas, alimentos y muestras ambientales. Se han realizado aportes al análisis de compuestos orgánicos trazas, en matrices de diverso origen y complejidad. La presente tesis ha sido dividida a efectos prácticos en tres capítulos dependiendo el tipo de matriz de trabajo: materias primas farmacéuticas, alimentos y medio ambiente. En el primer capítulo, se destacan procedimientos analíticos originales para la determinación de residuos de pesticidas en materias primas farmacéuticas de origen natural, para los cuales los métodos oficiales o son obsoletos conceptualmente o simplemente no existen. Se desarrollaron dos metodologías basadas en la técnica MSPD para el análisis multirresiduo de pesticidas en lanolina y propóleos, matrices de compleja composición química donde la etapa de purificación es crítica para una buena determinación. El segundo capítulo aborda el análisis de agroquímicos en alimentos. En primera instancia, se comprobó la posibilidad de determinación de reguladores de crecimiento en vegetales empleando tratamientos de muestra multirresiduo habituales (métodos Luke, QuEChERS y acetato de etilo) para el análisis de con determinación por sistemas LC-MS/MS. Por otro lado se realizó una evaluación preliminar a la contribución del consumo de yerba mate a la ingesta diaria de pesticidas por la población uruguaya, a partir de un dispositivo específico para evaluar la extracción de pesticidas en condiciones de ingesta típica del mate. RESUMEN 13 Se comprobó que la extractabilidad de residuos y por lo tanto la ingesta de de pesticidas se puede relacionar con las propiedades fisicoquímicas de los pesticidas en estudio. En el tercer capítulo, se describe la principal vía de transformación ambiental del antibiótico amoxicilina monitoreada en aguas residuales y aguas de río. Posteriormente se expone la posibilidad de evaluación de contaminantes emergente en aguas empleando diversos arreglos instrumentales. Son expuestas las fortalezas y debilidades de diversas estrategias analíticas especialmente enfocadas hacia instrumentos QTOF en experimentos dedicados a confirmación de identidad y estrategias de screening automatizado combinado con motores de búsqueda basados en bases de datos de masas exactas. Se ha observado que las estrategias basadas en detección asistida por software generalmente necesitan una etapa adicional de confirmación. Un instrumento QTOF en modo MS/MS permitió confirmar o rechazar la identidad de la mayoría de los hallazgos de screening de contaminantes emergentes en muestras reales a pesar de que existen limitaciones de sensibilidad analítica y efecto matriz. Se compararon además estrategias cuantitativas y cualitativas basada en el análisis por inyección directa de muestras acuosas por LC-MS/MS empleando instrumentos QLIT para un grupo representativo de contaminantes emergentes operando en modo SRM y en modo IDA. La mayor sensibilidad de los instrumentos QLIT-MS/MS ofrece la posibilidad de realizar análisis cuantitativo de variedad de contaminantes por inyección directa de la muestra, reduciendo el efecto de la matriz y aumentando de la confiabilidad de los resultados. Durante esta tesis, se ha logrado cubrir los distintos aspectos del análisis de trazas contemplando desde estrategias de preparación de muestra, expansión del alcance de métodos preexistentes y caracterización de contaminantes antropogénicos, empleando distintas técnicas instrumentales y de tratamientos de datos que permiten la realización de experimentos útiles tanto en cuantificación, identificación preliminar, confirmación y elucidación estructural de contaminantes orgánicos. Our group has previously developed immunoassays for noncompetitive detection of small molecules based on the use of phage borne anti-immunocomplex peptides. Recently, we substituted the phage particles by biotinylated synthetic anti-immunocomplex peptides complexed with streptavidin and named these constructs nanopeptamers. In this work, we report the results of combining AlphaLisa, a commercial luminescent oxygen channeling bead system, with nanopeptamers for the development of a noncompetitive homogeneous assay for the detection of small molecules. The signal generation of AlphaLisa assays relies on acceptor-donor bead proximity induced by the presence of the analyte (a macromolecule) simultaneously bound by antibodies immobilized on the surface of these beads. In the developed assay, termed as nanoAlphaLisa, bead proximity is sustained by the presence of a small model molecule (atrazine, MW = 215) using an antiatrazine antibody captured on the acceptor bead and an atrazine nanopeptamer on the donor bead. Atrazine is one of the most used pesticides worldwide, and its monitoring in water has relevant human health implications. NanoAlphaLisa allowed the homogeneous detection of atrazine down to 0.3 ng/mL in undiluted water samples in 1 h, which is 10-fold below the accepted limit in drinking water. NanoAlphaLisa has the intrinsic advantages for automation and high-throughput, simple, and fast homogeneous detection of target analytes that AlphaLisa assay provides. Using the flexible ligand oxydiacetic acid (H2oda), ten new heteropolynuclear compounds with general formula [Ln2Zn3(oda)6]·xH2O (Ln = lanthanide ion, oda = oxydiacetate) were synthesized and structurally characterized. They were prepared by the direct reaction of oda in aqueous solution with stoichiometric amounts of Zn and Ln salts, followed by slow organic solvent diffusion. Two different structures were obtained: the larger lanthanides Pr, Nd, Eu and Tb gave compounds belonging to the hexagonal system, space group P6/mcc; whereas, the smaller ones, Ho, Er, Yb and Y formed cubic systems, space group Fd[3 with combining macron]c. In the case of Dy, the two polymorphs were obtained by slight variations in the synthetic conditions. The phase change observed is discussed in comparison with previously reported [Ln2M3(oda)6]·xH2O compounds (M = bivalent metal ion). Luminescence studies were also carried out. Compounds containing Eu and Tb showed the most intense luminescence, also bearing relatively long lifetimes for the excited state deactivation. The luminescence profiles of both Dy systems are independent of the hexagonal or cubic arrangements, due to the similar environment of the Ln having the same point symmetry. Phage borne peptides isolated from phage libraries have proven to be valuable reagents for the development of small-molecule immunoassays. However, the large size, low diffusion rate, and biological nature of the phage particles create some limitations to their use and require secondary reagents for its detection. In this work, we explore the use of the Nano luciferase (NanoLuc) as a fusion partner to generate recombinant tracers for immunoassay development. The imidaclothiz peptidomimetic C2-15 that specifically binds to the anti-imidaclothiz monoclonal antibody (mAb) 1E7 was fused to NanoLuc, both at the N terminus (C2-15-NanoLuc) and C terminus (NanoLuc-C2-15). NanoLuc-C2-15 showed better performance than C2-15-NanoLuc and was adopted to develop a bioluminescent enzyme immunoassay (BLEIA) and a bioluminescence lateral flow immunoassay (BLLFIA) for imidaclothiz. The luminescence signal of NanoLuc-C2-15 rapidly reaches high intensity with slow attenuation, which enabled one to capture the BLLFIA readout by using a smartphone without an external light source. The IC50 of the BLEIA and BLLFIA were 3.3 ± 0.2 and 6.4 ± 0.4 ng mL-1, respectively. Both immunoassays exhibited good accuracy for the detection of imidaclothiz in environmental and agricultural samples. We investigated the occurrence of cycloadditions on dimensional boron nitride. Our results indicated that the [2 + 2] cycloaddition of benzynes, ethyne, and maleic anhydride is allowed both from thermodynamical and kinetic stand points. In fact, our results indicated that the reaction energy associated with the formation of the [2 + 2] cycloaddition product is more exothermic than that computed for graphene. The activation energy determined for the [2 + 2] path is 15.9 kcal/mol, suggesting the feasibility of the process. Furthermore, we found that the activation energy for the retro reaction is 30.6 kcal/mol. This value is large enough to guarantee the stability of the functional groups but not too high to impede their detachment upon heating. In addition to these findings, we found that cycloadditions can be used to reduce the band gap of boron nitride nanosheets. In light of these results, we propose the use of cycloaddition reactions to attain the temperature-controlled functionalization of boron nitride sheets. The goal of our study was to improve Tannat grape and wine composition by achieving vine balance in high-capacity conditions. The current Uruguayan grape-growing paradigm accepts unpredictable periods of water deficit or excess in non-irrigated vineyards, only applying herbicides to eliminate weed competition (H). We used an under-trellis cover crop (UTCC) to limit vine water availability and to reduce vine growth rate and final canopy size. However, to avoid excessive vine water stress due to UTCC competition, irrigation was used as needed during water deficit periods. The experiment was conducted over three consecutive growing seasons from 2011 to 2013, in two experimental vineyards located in southern Uruguay (34° S 56° W). Treatments were: UTCC (full cover of the vineyard soil with tall fescue) versus conventional alleyway tall fescue with a 1.0 m-wide weed-free strip under the trellis. Deficit drip irrigation was provided at mid-day stem water potential (SWP) thresholds of −0.9 MPa early in the season and −1.1 MPa later. Shoot growth rate, SWP, berry size and berry composition (Brix, organic acids, total anthocyanins) were monitored over the season as well as final yield, cluster and pruning weights and wine sensory attributes. UTCC regulated vine vegetative growth and final canopy size and reduced bunch rot incidence as well as increasing fruit Brix and anthocyanin concentration in grapes and wines. Wines from UTCC treatments had an increased fruity aroma and overall aroma intensity levels and had distinctive sensory characteristics that exceeded those of H wines during overall palatability tests (liking tests). Nanoparticulate immunostimulating complexes (ISCOMs or ISCOMATRIX) formulated with these, are able to stimulate strong humoral and cellular immune responses. Recently, we formulated novel ISCOMs replacing QuilA® by QB-90 (IQB-90), a Quillaja brasiliensis leaf-extracted saponin fraction, and reported that IQB-90 improved antigen uptake, and induced systemic and mucosal antibody production, and T-cell responses. However, its mechanism of action remains unclear. In this study we provide a deeper insight into the immune stimulatory properties of QB-90 and ISCOMATRIX-like based on this fraction (IMXQB-90). We show herein that, when used as a viral vaccine adjuvant, QB-90 promotes an “immunocompetent environment”. In addition, QB-90 and IMXQB-90 induce immune-cells recruitment at draining-lymph nodes and spleen. Subsequently, we prove that QB-90 or IMXQB-90 stimulated dendritic cells secret IL-1β by mechanisms involving Caspase-1/11 and MyD88 pathways, implying canonical inflammasome activation. Finally, both formulations induce a change in the expression of cytokines and chemokines coding genes, many of which are up-regulated. Findings reported here provide important insights into the molecular and cellular mechanisms underlying the adjuvant activity of Q. brasiliensis leaf-saponins and its respective nanoparticles. Saponin-based adjuvants are promising adjuvants that enhance both humoral and T-cell-mediated immunity. One of the most used natural products as vaccine adjuvants are Quillaja saponaria bark saponins and its fraction named Quil A®. Despite that, its use has been restricted for human use due to safety issues. As an alternative, our group has been studying the congener species Quillaja brasiliensis saponins and its performance as vaccine adjuvants, which have shown to trigger humoral and cellular immune responses comparable to Quil A® but with milder side effects. Here, we studied a semi purified aqueous extract (AE) and a previously little characterized saponin-enriched fraction (QB-80) from Q. brasiliensis as vaccine adjuvants and an inactivated virus (bovine viral diarrhea virus, BVDV) antigen co-formulated in experimental vaccines in mice model. For the first time, we show the spectra pattern of the Q. brasiliensis saponins by MALDI-TOF, a novel and cost-effective method that could be used to characterize different batches during saponins production. Both AE and QB-80 exhibited noteworthy chemical similarities to Quil A®. In addition, the haemolytic activity and toxicity were assessed, showing that both AE and QB-80 were less toxic than Quil A®. When subcutaneously inoculated in mice, both fractions promoted long-term strong antibody responses encompassing specific IgG1 and IgG2a, enhanced the avidity of IgG antibodies, induced a robust DTH reaction and significantly increased IFN-ɣ production in T CD4+ and T CD8+ cells. Furthermore, we have proven herein that AE has the potential to promote dose-sparing, substantially reducing the dose of antigen required for the BVDV vaccines and still eliciting a mixed Th1/Th2 strong immune response. Based on these results, and considering that AE is a raw extract, easier and cheaper to produce than commercially available saponins, this product can be considered as candidate to be escalated from experimental to industrial uses.
