Suitable conditions of temperature and humidity are required to maintain wheat grains quality, but during processing and storage, the grains can be exposed to adverse environmental conditions and presence of infectious fungi. Fusarium graminearum, the main causal agent of Fusarium head blight on wheat, affects crop yields and grain quality by alteration of their biochemical components and mycotoxin contamination, which reduces the possibilities of wheat end use and compromises food safety. Lipid degradation by hydrolytic, oxidative and microbial deterioration is the predominant cause of the loss of sensory acceptability, nutritional value and baking quality. The aim of this research was to determine the influence of adverse environmental conditions –as the increasing moisture – on lipid patterns of whole wheat flours contaminated with F. graminearum in relation to the infection degree. In vitro cultures of F. graminearum were carried out on wheat grains under different degrees of relative humidity (11, 50, 75 and 100%) throughout 45 days of incubation at 28 °C. The fungal biomass measured by q-PCR increased proportionally with the humidity. A decrease in the signals of saturated (palmitic and estearic) and unsaturated (oleic, linoleic and linolenic) fatty acids, analyzed as fatty acid methyl esters (FAMEs) by GC–MS, was observed in relation with the humidity and infection degree. The degradation rate of the lipids was high during the first 15 days of incubation, reaching the fatty acids content, values around 20–40% of those found in the control. From that moment on, the rate of degradation was slower or even null. It was observed that in all treatments, the linolenic acid reached the highest degradation ratio in comparison with the other fatty acids, which may be caused by the action of lipoxygenases. The lipase activity and the content of deoxynivalenol were also determinate on the flours. The lipase activity increased until day 25 of incubation reaching twice the initial value. The deoxynivalenol content also increased along incubation while fatty acids decreased. Our results demonstrated that the magnitude in the signal of fatty acids in whole wheat flours varied in relation to the degree of humidity and fungal infection of the grains from which they were obtained. Otherwise, lipids and their oxidation products are related with the pathogenesis and production of mycotoxins. These observations highlight the importance of an adequate manipulation of wheat grains on the processing chain to prevent quality changes and mycotoxins contamination.

Kefir granules are a complex microbial consortia with numerous properties, among which their antifungal action has been reported. This study characterized the antifungal activity of water kefir granules and two microorganisms isolated from them, identified as Saccharomyces cerevisiae and Geotrichum candidum, against aflatoxigenic Aspergillus flavus. Experiments were carried out in vitro and under minisilo conditions designed to simulate the aerobic phase of commercial corn storage. In dual culture plate assays, A. flavus was inhibited in close contact with both antagonists, and mycelial weakening with lower conidia production was observed. Cell-free supernatants of both antagonists inhibited conidial germination by 92% (G. candidum) and 65% (S. cerevisiae). Non-contact assays between A. flavus and antagonists indicated that volatile organic compounds (VOCs) also contributed to inhibition, reducing mycelial growth by 29.1% (G. candidum) and 37.9% (S. cerevisiae). In corn minisilo trials water kefir granules and both antagonists showed strong antifungal activity by day 4, with qPCR of A. flavus DNA and aflatoxin measurements indicating near complete inhibition. As mixed consortia, water-kefir granules inherently include lactic acid bacteria whose rapid acidification and diffusible metabolites complement antifungal compounds produced by other components of the consortia, supporting a multi-mechanistic mode of action. Given the compositional heterogeneity of kefir granules, strain-level identification and verification of safety and regulatory status for the intended use are essential prior to food or feed applications. The demonstrated antifungal efficacy of kefirderived microorganisms in mini-silos underscores their scale-up potential for postharvest protection of corn.

A summary of the resource.
For a fourth approach of quinoxaline N,N0- dioxides as anti-trypanosomatid agents against T. cruzi and Leishmania, we found extremely active derivatives. The present study allows us to state the correct requirements for obtaining optimal in vitro anti-T. cruzi activity. Derivatives possessing electron-withdrawing substituents in the 2-, 3-, 6-, and 7-positions were the most active compounds. With regard to these features and taking into account their mammal cytotoxicity, some trifluoromethylquinoxaline N,N0-dioxides have been proposed as candidates for further clinical studies. Consequently, mutagenicity and in vivo analyses were performed with the most promising derivatives. In addition, with regard to the mechanism of action studies, it was demonstrated that mitochondrial dehydrogenases are involved in the anti-T. cruzi activity of the most active derivatives

A summary of the resource.
Internal dosimetry intercomparisons are essential for the verification of applied models and the consistency of results'. To that aim, the First Regional Intercomparison was organised in 2005, and that results led to the Second Regional Intercomparison Exercise in 2013, which was organised in the frame of the RLA 9/066 and coordinated by Autoridad Regulatoria Nuclear of Argentina. Four simulated cases covering intakes of (131)I, (137)Cs and Tritium were proposed. Ninteen centres from thirteen different countries participated in this exercise. This paper analyses the participants' results in this second exercise in order to test their skills and acquired knowledge, particularly in the application of the IDEAS Guidelines. It is important to highlight the increased number of countries that participated in this exercise compared with the first one and, furthermore, the improvement in the overall performance. The impact of the International Atomic Energy Agency (IAEA) Projects since 2003 has led to a significant enhancement of internal dosimetry capabilities that strengthen the radiation protection of workers.

The laminated layer is the unique mucin-based extracellular matrix that protects Echinococcus larvae, and thus to an important extent, shapes host–parasite relationships in the larval echinococcoses. In 2011, we published twin reviews summarizing what was known about this structure. Since then, important advances have been made. Complete genomes and some RNAseq data are now available for E. multilocularis and E. granulosus, leading to the inference that the E. multilocularis LL is probably formed by a single type of mucin backbone, while a second apomucin subfamily additionally contributes to the E. granulosus LL. Previously suspected differences between E. granulosus and E. multilocularis in mucin glycan size have been confirmed and pinned down to the virtual absence of Galβ1–3 chains in E. multilocularis. The LL carbohydrates from both species have been found to interact selectively with the Kupffer cell receptor expressed in rodent liver macrophages, highlighting the ancestral adaptations to rodents as intermediate hosts and to the liver as infection site. Finally, LL particles have been shown to possess carbohydrate-independent mechanisms profoundly conditioning non-liver-specific dendritic cells and macrophages. These advances are discussed in an integrated way, and in the context of the newly determined phylogeny of Echinococcus and its taenid relatives.<br /><br />

The laminated layer (LL) is the massive carbohydrate-rich structure that protects Echinococcus larvae, which cause cystic echinococcosis (hydatid disease) and alveolar echinococcosis. Increased understanding of the biochemistry of the LL is allowing a more informed analysis of its immunology. The LL not only protects the parasite against host attack but also shapes the overall immune response against it. Because of its dense glycosylation, it probably contains few T-cell epitopes, being important instead in T-cell independent antibody responses. Crucially, it is decoded in non-inflammatory fashion by innate immunity, surely contributing to the strong immune-regulation observed in Echinococcus infections. Defining the active LL molecular motifs and corresponding host innate receptors is a feasible and promising goal in the field of helminth-derived immune-regulatory molecules.

Metazoan parasites of mammals (helminths) belong to highly divergent animal groups and yet induce a stereotypical host response: Th2-type immunity. It has long been debated whether this response benefits the host or the parasite. We review the current literature and suggest that Th2 immunity is an evolutionarily appropriate response to metazoan invaders both in terms of controlling parasites and repairing the damage they inflict. However, successful parasites induce regulatory responses, which become superimposed with, and control, Th2 responses. Beyond helminth infection, this superimposition of response profiles may be the norm: both Th1 and Th2 responses coexist with regulatory responses or, on the contrary, with the inflammatory Th17 responses. Thus, typical responses to helminth infections may differ from Th2- dominated allergic reactions in featuring not only a stronger regulatory component but also a weaker Th17 component. The similarity of immune response profiles to phylogenetically distinct helminths probably arises from mammalian evolution having hard-wired diverse worm molecules, plus tissue-damage signals, to the beneficial Th2 response, and from the convergent evolution of different helminths to elicit regulatory responses. We speculate that initiation of both Th2 and regulatory responses involves combinatorial signaling, whereby TLR-mediated signals are modulated by signals from other innate receptors, including lectins.

The in vitro nematocide activity of seventeen 6,7-diarylpteridines has been tested using three different experimental models, Caenorhabditis elegans, Nippostrongylus brasiliensis and Heligmosomoides polygyrus. The method of evaluation of inhibition in the secretion of acetylcholinesterase by H. polygyrus seems to be the most indicated to avoid false positives. The in vivo activities, against Trichinella spiralis, of the most in vitro active pteridines have been assayed. All pteridine derivatives bearing 6,7-di-p-bromophenyl substituents have shown in vitro nematocide activities in the three experimental models used. Amongst all the pteridines tested in vivo, only 2,4-pteridinedithione derivatives exhibited moderate activity.

Introducción. Los liposomas son sistemas supramoleculares autoensamblables preparados ad hoc,compuestos de fosfolípidos y colesterol, diseñados para transporte de fármacos o radionucleidos. El 99mTc es el radionucleido más empleado por sus propiedades físicas apropiadas para la adquisición de imágenes y estudios en pacientes en el área de medicina nuclear (emisor gamma puro con E = 140 KeV , t1/2 = 6 horas). Objetivo. Evaluar la potencialidad de liposomas convencionales marcados con 99mTc como agente de diagnóstico de procesos tumorales. Método. La composición estudiada es: fosfatidilcolina, dioleilfosfatidiletanolamina y colesterol (1.1:0.4:1 relación molar). Se utilizó como agente reductor SnF2, en diferentes cantidades para optimizar el marcado. La pureza radioquímica y eficiencia de marcado se evaluaron con sistemas cromatográficos ITLC‐SG FM NaCl 0,9%, ITLC‐SG FM Piridina:ácido acético:agua (3:5:1.5 v/v). Se adquirieron imágenes a 1 h post inyección de los liposomas en ratones sanos y portadores de tumor mamario espontáneo. Resultados. El liposoma marcado mostró estabilidad durante 24 h, siendo la cantidad óptima de reductor 138 μg. La mejor captación en tumor fue a 1 h post administración del radiofármaco en los estudios centellográficos.

With the objective to develop a potential 99mTc radiopharmaceutical for imaging the androgen receptor (AR) in prostate cancer, four ligands bearing the same pharmacophore derived from the AR antagonist flutamide were prepared, labeled with 99mTc, and their structures corroborated via comparison with the corresponding stable rhenium analogs. All complexes were obtained with high radiochemical purity. Three of the complexes were highly stable, and, due to their favorable physicochemical properties, were further evaluated using AR-positive and AR-negative cells in culture. All complexes exhibited considerable uptake in AR-positive cells, which could be blocked by an excess of flutamide. The efflux from the cells was moderate. They also showed significantly lower uptakes in AR-negative cells, indicating interactions with the AR receptor. However, the binding affinities were considerably reduced by the coordination to 99mTc, and the complex that exhibited the best biological behavior did not show sufficient specificity towards AR-positive cells.

A summary of the resource.
The extension of relative absorptiometric methods to derivative spectrophotometry is proposed as a way of improving precision an accuracy for extremely high absorbance values. The concept was developed by applying the transmittance-ratio method to second-derivative measurements of two model systems where the total absorbance is kept very high |b leading to a significant enhancement of precision and accuracy. The variation of the signal to noise ratio and of the total error as a function af absolute and relative absorbance was studied. Some of the factors affecting the analytical behaviour of this method are discussed.

Serratia marcescens can be a plant growth promoting bacteria (PGPB) and an opportunistic human andplant pathogen. We have identified and characterized strains of related species of Serratia and evaluatedtheir biological control of damping-off of tomato seeds caused by Pythium cryptoirregulare. Serratia ure-ilytica, S. bockelmannii and S. nevei were identified by phylogenetic analysis of partial gyrB gene sequenceand average nucleotide identity (ANI). Tomato seeds inoculated with S. ureilytica ILBB 145 showed highergermination percentage and reduced damping-off in greenhouse experiment resembling a commercialoperation, and volatiles produced by this strain caused the nearly complete inhibition in vitro of P. cryp-toirregulare. Analysis of volatile organic compounds (VOCs) showed that ILBB 145 produced dimethyldisulfide (DMDS), which can partially account for this inhibition. Serratia bockelmannii ILBB 162 perfor-mance against damping-off was intermediate and the inhibition of P. cryptoirregulare in vitro was lowerand explained by volatile and diffusible metabolites. Both strains augmented DMDS production in thepresence of P. cryptoirregulare, suggesting this compound may play a role in the context of interspecificcompetition. Serratia nevei ILBB 219 showed the lowest inhibition of P. cryptoirregulare in vitro, no DMDSproduction, and no biocontrol in planta. Draft genomes of the three strains were annotated and individ-ual genes and biosynthesis gene clusters were identified in relation with the observed phenotypes. Wereport S. ureilytica – a low risk species- with activity as a biological control agent and DMDS produced bythis bacterial species putatively involved in seed and seedling protection against P. cryptoirregulare.

El propóleos y el orujo de uvas son productos reconocidos por contener antioxidantes. Un extracto elaborado en base a ellos podría ser de gran aplicabilidad en las industrias alimenticia (alimentos funcionales, nutracéutico), farmacéutica (fitofármacos), y/o de aditivos preservantes de la oxidación. El trabajo de investigación presentado en esta Tesis de Doctorado fue basado en la necesidad de generar conocimiento en cuanto a las características fisicoquímicas, farmacológicas y biológicas de esos dos productos naturales uruguayos. Con el mismo se aportó nuevo conocimiento científico en la aplicación de metodología in silico al estudio de mezclas complejas de fenoles en propóleos y orujos de uvas que se complementó con las de laboratorio húmedo o in vitro. Los fenoles, moléculas antioxidantes detectadas en los propóleos y orujos de uvas, atrapan radicales libres modulando el estrés oxidativo celular. Presentan en su estructura grupos aromáticos hidroxilados e intervienen en los mecanismos redox de vegetales y animales, promoviendo un descenso de la cantidad de especies reactivas de oxigeno (ROS). En los propóleos se han identificado flavonoides (como flavonas y flavanonas), ácidos aromáticos y sus ésteres, aldehídos aromáticos, cumarinas y triglicéridos fenólicos. Los orujos de uvas contienen estilbenos, antocianinas, taninos, catequinas, flavanoles, flavonoles y flavonas. El perfil estructural es complementario de tal manera, que un “cóctel polifenólico” elaborado a partir de propóleos y orujos de uvas uruguayos, contendrá un amplio rango de estructuras fenólicas que sirvan para desarrollar un extracto patronizado en cuanto a sus capacidades como antioxidante y antiinflamatorio. Dado que es característica en los productos naturales la alta variabilidad en sus contenidos de fenoles, cualquier potencial aplicabilidad industrial implica la necesidad de analizarlos cualitativa y cuantitativamente así como su funcionalidad. Para lograrlo, se realizaron muestreos, se optimizaron los procesos extractivos, se analizaron los contenidos fenólicos totales, la capacidad antioxidante por los métodos DPPH y ABTS, se realizó la dilucidación de sus huellas dactilares de protón por resonancia magnética nuclear (RMN 1H), se estudió la reactividad de los grupos OH y la energía de disociación del enlace O-H de los polifenoles como formadores del radical libre mediante química cuántica DFT (M06-2X/6-311+G(3df,2p) y G4. By means of G4 theory and M06-2X calculations we investigated the thermochemistry and C–OH bond ruptures of quercetin, fistein, lutelin, caffeic acid, pinocembrin and p-coumaric acid. Notwithstanding the fact that we observed a good agreement for the prediction of the weakest O–H bond, the mean absolute deviation (MAD) between the bond dissociation energies (BDE) computed at the G4 and M06-2X/6-311+G(3df,2p) levels was 5.0 kcal/mol. With regard to the ΔHof,0 computed at the M06-2X/6-311+G(3df,2p) level, the MAD was 10.0 kcal/mol. Therefore, thanks to error cancellation the error found for the O–H BDE was smaller. Intramolecular hydrogen bond is one of the key factors stabilizing the oxygen radicals formed when the CO–H bonds are broken. Finally, we propose the following enthalpies of formation at 0 K: −222.0, −174.5, −180.2, −134.4, −109.7 and −91.9 kcal/mol for quercetin, fistein, lutelin, caffeic acid, pinocembrin and p-coumaric acid, respectively.

A summary of the resource.
Salto district, a key horticultural area in Uruguay, has crucial water sources such as the Guarani Aquifer System, the Salto-Arapey Aquifer System (SAAS), and the San Antonio Creek (SAC). Agricultural and livestock activities have increased in recent years, leading to demographic growth and housing expansion. Concerns about water quality arise due to limited studies on environmental exposure to harmful chemicals and microbiological factors from agriculture, inadequate wastewater treatment, and septic tank seepage in rural areas. The present study collected hydrochemical background and new sampling data for hydrogeochemical and water quality evaluation (28 parameters) from SAAS (73 wells) and SAC (6 sites). Results indicated groundwater from SAAS is mainly sodium bicarbonate type, RAS index shows low saline waters and microbiological contamination were detected in half of the sampled wells, As levels (4.0–49.3 μgL−1), total phosphorus (P) (50–178.5 mg/L), and nitrate (NO− 3 ) (7.0–168.5 mg/L). SAC hydrochemistry revealed high levels of P (77.5–163.5 μg/L), which could be one of the factors that promote the appearance of signs of eutrophication towards the mouth of the Uruguay River. Spatial distribution of total arsenic (As) in SAAS and water quality evaluation of SAC assess future human health risks. These results represent a challenge to foster Uruguayan water education and raise public awareness about the importance of using safe water for human consumption and irrigation.
