ZrCr1-xNiMox (x = 0.0, 0.3 and 0.6 corresponding to 0, 13 and 25% w/w Mo content respectively) alloys with initial particle sizes of 38–63 μm and 63–125 μm were studied. Alloys were characterized by X-ray diffraction, revealing that molybdenum substitution by chromium produces the segregation of ZrxNiy phases, ZrCr2 and ZrMo2 Laves phases. Electrodes with 13% of molybdenum depicted the highest discharge capacity of 300 mA h g−1. In all cases, the smallest initial particle size exhibited the highest hydrogen storage capacity. On the other hand, the increase of nickel content in the composition of the secondary phase (Zr7Ni10 comparing to Zr9Ni11) leds to an improvement in high rate discharge ability behavior. Hydrogen evolution reaction analysis showed that electrodes with 13% of molybdenum described the largest current intensities. The reaction rds was related to the type of secondary phase dominant in each sample, whereas self-discharge behavior was associated to the presence of molybdenum phases. Thereby, the alloy with 13% weight in molybdenum was the one which improves electrocatalytic performance for hydrogen storage purposes. Silver nanoparticles have particular properties that contribute to their very promising applications, novel in various fields of science, such as the development of biosensors, the diagnosis and treatment of cancer, the controlled release of drugs and the antimicrobial potential. The biological synthesis of nanoparticles is of great interest over other physical and chemical methods because the use of toxic chemicals and drastic reaction conditions are avoided. The extracellular biosynthesis using fungi could also make downstream processing much easier than the intracellular biosynthesis. One of the main applications of silver nanoparticles is their antimicrobial activity. Several studies have demonstrated the bactericidal properties of silver nanoparticles are different from silver ions, and that they are strongly influenced by their shape, size, concentration and colloidal state. In the present work, the ability of fungal strains from Uruguay to synthesize silver nanoparticles was studied. Eight fungi were able to synthesize nanoparticles. An extensive physicochemical characterization of the nanoparticles was carried out including ultraviolet-visible spectroscopy, transmission electron microscopy, dynamic light scattering, zeta-potential and gel electrophoretic mobility. According to the characterization and colloidal stability results, nanoparticles from three fungi were selected for antimicrobial activity assays. All nanoparticles were able to inhibit Escherichia coli growth, demonstrating their potential as effective antibacterial agent for use in biomedical applications. The free-living nematode Caenorhabditis elegans is the simplest animal model organism to work with. Substantial knowledge and tools have accumulated over 50 years of C. elegans research. The use of C. elegans relating to parasitic nematodes from a basic biology standpoint or an applied perspective has increased in recent years. The wealth of information gained on the model organism, the use of the powerful approaches and technologies that have advanced C. elegans research to parasitic nematodes and the enormous success of the omics fields have contributed to bridge the divide between C. elegans and parasite nematode researchers. We review key fields, such as genomics, drug discovery and genetics, where C. elegans and nematode parasite research have convened. In the present work, we report the enhancement of lithium-ion dissociation and transport in poly(acrylonitrile) host promoted by the addition of hydrogen titanate nanotube fillers for solid polymer electrolytes. We show experimental and theoretical evidence of lithium perchlorate dissociation due to the presence of the acidic hydrogen titanate nanotubes embedded in the polymer matrix. We performed confocal Raman microscopy analysis to reveal the presence of lithium perchlorate dissociation at the interface of polymer and nanotube fillers. The large affinity of perchlorate anions at the hydrogen titanate nanotube surface, as envisaged from the ab initio molecular dynamics simulations, could be responsible for the enhancement of more than 2 orders of magnitude in the lithium conductivity, reaching &sim;4 × 10–4 S·cm−1 for a certain amount of nanotube fillers addition. A new mononuclear complex of zinc(II), [Zn(HL)2]∙2DMF (H2L = (E)-N′-((E)-(hydroxyimino)butan-2-ylidene)salicyloylhydrazide, DMF = N,N-dimethylformamide), was prepared and characterized. Single-crystal X-ray crystallography revealed a six-coordinate zinc(II) surrounded by nitrogen of the oxime function and oxygen and distal nitrogens of the acylhydrazone group. This entity also exists in solution as demonstrated by 1H-NMR and potentiometric titrations. The computational analysis showed that the molecular orbitals involved in the main electronic transitions of the complex species in solution are centered on the ligand with negligible contribution of the metal ion. The photophysical properties of the complex were evaluated in solution and in the solid state. Luminescence studies showed that the solid has a strong emission at 550 nm with a large Stokes shift with respect to absorption. The solid state fluorescence emission is ascribed to ligand-centered and/or ligand-to-ligand charge transfer transitions, following the DFT results in solution. A comparison with a previously reported mononuclear [Zn(HL)2] allowed the investigation of the influence of DMF molecules in the structural packing and the luminescence properties. A combination of three-dimensional quantitative structure-activity relationship (3D-QSAR), and molecular modelling methods were used to understand the potent inhibitory NAD(P)H:quinone oxidoreductase 1 (NQO1) activity of a set of 52 heterocyclic quinones. Molecular docking results indicated that some favourable interactions of key amino acid residues at the binding site of NQO1 with these quinones would be responsible for an improvement of the NQO1 activity of these compounds. The main interactions involved are hydrogen bond of the amino group of residue Tyr128, π-stacking interactions with Phe106 and Phe178, and electrostatic interactions with flavin adenine dinucleotide (FADH) cofactor. Three models were prepared by 3D-QSAR analysis. The models derived from Model I and Model III, shown leave-one-out cross-validation correlation coefficients (q2LOO ) of .75 and .73 as well as conventional correlation coefficients (R2 ) of .93 and .95, respectively. In addition, the external predictive abilities of these models were evaluated using a test set, producing the predicted correlation coefficients (r2pred ) of .76 and .74, respectively. The good concordance between the docking results and 3D-QSAR contour maps provides helpful information about a rational modification of new molecules based in quinone scaffold, in order to design more potent NQO1 inhibitors, which would exhibit highly potent antitumor activity. PknG, una Ser/Thr-quinasa de proteínas, es un factor crucial para la sobrevid a de esta bacteria en el hospedero. Con el fin de elucidar las vías de señalización med iadas por PknG, recientemente estudiamos su interactoma e identificamos nuevos su stratos de esta quinasa. En el presente trabajo nos centramos en una de las proteí nas fosforiladas por PknG: la proteína FhaA. Esta proteína tiene una estructura q ue se asemeja a las proteínas de andamiaje que participan en vías de señalización de eucariotas, con un dominio N- terminal de función desconocida y un dominio C-term inal FHA que reconoce específicamente de treoninas fosforiladas, ambos unidos por una secue ncia de aproximadamente 300 residuos sin estructura definid a. Como primer paso hacia la elucidación del rol de esta proteína nos propusimos la caracterización a nivel molecular de los complejos de señalización formados in vivo por FhaA, así como sus cambios dinámicos frente a diferentes estímulos que simulen el ambiente del hospedero. La estrategia experimental utilizada se basa en la sobre-expresión de FhaA conteniendo una etiqueta (Strep-tag) en Mycobacterium smegmatis en combinación con el entrecruzamiento de proteínas in vivo , purificación por afinidad y espectrometría de masa para obtener una instantánea de los complejos de señalización. Nuestros resultados nos permitieron i dentificar 56 interactores de FhaA, entre los que se incluye el único interactor previa mente reportado [1]. Los nuevos interactores identificados nos permiten postular qu e esta proteína cumple un rol tanto en la síntesis de pared como en la regulació n de la división celular en la micobacteria. El diseño y construcción de estructuras supramoleculares polinucleares de iones lantánidos es un campo de investigación de gran interés. Ellas presentan interesantes arquitecturas y propiedades, ya sea magnéticas, ópticas, eléctricas, etc. Los lantánidos (Ln) en su estado de oxidación +3, son iones ideales para el diseño de estas estructuras poliméricas, debido a sus altos índices de coordinación que permiten construir fascinantes estructuras térmicamente estables. El futuro de este tipo de complejos depende de continuar la búsqueda de nuevos compuestos y del cuidadoso diseño y control de la arquitectura final generada. La selección del ligando utilizado es crucial para el diseño de estos compuestos. Un tipo de ligandos de amplio estudio por formar complejos de gran estabilidad y ofrecer diversos modos de coordinación son los ligandos policarboxílicos. En particular el ácido iminodiacético (H2ida) ha mostrado una sorprendente versatilidad gracias a su gran flexibilidad y diversos modos de coordinación. Varias estructuras polinucleares se han reportado con este ligando, siendo relevante la serie [Ln(Hida)3]·xH2O. La particularidad de estos compuestos es la formación de estructuras tridimensionales que contienen canales de unos 11 Å de diámetro. Es de destacar que en estas estructuras se observa la capacidad de este ligando de pasar de ser un ligando tridentado ONO, a un ligando ditópico, que puede coordinar centros metálicos alejados. La protonación del átomo de N es la responsable de estos cambios. En esta misma línea, otras estructuras porosas han sido reportadas para complejos heteropolinucleares conteniendo iones lantánidos, metales de transición 3d y el ligando ida. Un ejemplo de estos son los complejos isoestructurales del tipo [Ln2Cu3(ida)6]·xH2O. Con estos antecedentes, el objetivo de este trabajo es sintetizar nuevas series de complejos polinucleares conteniendo iones lantánidos y ligandos aminodicarboxílicos N-sustituidos derivados del iminodiacetato y caracterizarlos del punto de vista estructural y de sus propiedades. Los ligandos aquí empleados son el ácido N-hidroxietiliminodiacético (H2hida), el ácido N-benciliminodiacético (H2bzlida), el ácido N-metiliminodiacético (H2mida) y el ácido Nmetiliminodipropiónico (H2midp). Con el fin de determinar las mejores condiciones de síntesis se realizaron estudios potenciométricos en solución acuosa. A partir de estos fue posible determinar las especies principales en solución y el rango de pH en el cual pueden ser aislados al estado sólido. Luego se pasó a la etapa de síntesis, donde se obtuvieron nuevos complejos polinucleares con todos los ligandos: [Ln(bzlida)(Hbzlida)]·H2O (Ln = La, Nd), [Ln2(bzlida)3]·3H2O (L n= Nd, Sm, Eu, Gd), [Ho(hida)(H2O)2](CF3SO3)·xH2O, [Sm(Hmida)2(H2O)2]·ClO4·H2O·1/6NaClO4 y [Ln2(Hmidp)3(H2O)3](ClO4)3·3H2O (Ln = Nd, Sm, Eu, Gd, Tm, Dy, Ho). Los complejos fueron caracterizados completamente incluyendo su elucidación estructural mediante difracción de rayos X. Finalmente se evaluaron las propiedades ópticas y magnéticas de una selección de los compuestos obtenidos. Esto permitió relacionar las estructuras obtenidas con las propiedades al estado sólido. El estudio de sus propiedades ópticas permitió determinar que es posible una Resumen 2 transferencia de energía desde los ligandos a los centros metálicos, aunque la eficiencia de la transferencia es baja, siendo los compuestos de Eu los que arrojan los mejores resultados. Al evaluar las propiedades magnéticas se pudo racionalizar el origen del acoplamiento antiferromagnético en el compuesto [Gd2(bzlida)3]·3H2O. Glutathione transferases (GSTs) perform several catalytic and non-catalytic roles in the defense against toxicities of electrophile compounds and oxidative stress, and therefore are involved in stress-response and cell detoxification. Previously, we have provided evidence indicating that EgGST2 and EgGST3, two phylogenetically distant Echinococcus granulosus GSTs, can naturally form a heterodimeric structure (EgGST2-3). In the present work, the recombinant heterodimer GST (rEgGST2-3) is characterized. Hence, rEgGST2-3 was able to conjugate GSH to three substrates: 1-chloro-2,4-dinitrobenzene (CDNB, general substrate for GSTs), 1,2-dichloro-4-nitrobenzene (specific substrate for mammalian Mu class) and trans,trans-deca-2,4-dienal (reactive carbonyl). The canonical activity was considerably reduced by all the conventional inhibitors (cybacron blue, triphenylthin chloride and bromosulfophthalein) and by other inhibitors (ellagic acid, alizarin and chenodeoxycholic acid). Besides this, rEgGST2-3 activity was inhibited by a number of anthelmintic drugs, where the halogenated phenolic drugs (mainly bithionol and hexachlorophene) acted as stronger inhibitors, suggesting they may bind to the EgGST2-3. Moreover, rEgGST2-3 exhibited glutathione-peroxidase activity, and its specific constant (kcat/KM) was calculated. Finally, rEgGST2-3 displayed the ability to bind non-substrate molecules, particularly anthelmintic drugs, suggesting that ligandin activity may have potential to act as a passive protection parasite mechanism. Overall, the rEgGST2-3 behavior was shown to be both complementary and redundant to that reported for rEgGST1, another characterized GST from E. granulosus. It would be appropriate that different enzymes in the same organism do not have exactly the same functional properties to develop a better adaptation to life in the host. Dr. William B. Coley (1862–1936), a bone- sarcoma surgeon, was a pioneer in treating his patients with both live bacterial-based and mixture of heat-killed bacteria known as “Coley’s toxins.” Unfortunately, Coley was forced to stop his work which interrupted this field for about half a century. Currently, several species of bacteria are being developed against cancer. The bacterial species, their genetic background and their infectious behavior within the tumor microenvironment are thought to be relevant factors in determining their anti-tumor effectiveness in vivo. In this perspective article we will update the most promising results achieved using bacterial therapy (alone or combined with other strategies) in clinically-relevant animal models of cancer and critically discuss the impact of the bacterial variants, route of administration and mechanisms of bacteria-cancer-cell interaction. We will also discuss strategies to apply this information using modern mouse models, molecular biology, genetics and imaging for future bacterial therapy of cancer patients. Back to the Controversial Future The use of microorganisms, in particular live bacteria, for prophylactic vaccination and cancer therapy have been used in humans for long periods in the past and have been a matter of controversy (Payette and Davis, 2001; Hoption Cann et al., 2003). Dr. William B. Coley in the 19th century at the New York Hospital, later to become the Memorial Sloan Kettering Cancer Center (McCarthy, 2006; Hoffman, 2016a), observed and reported spontaneous tumor regression in patients with streptococcal infections (principally erysipelas, known to be caused by Streptococcus pyogenes). This is the first study to employ a reaction flow (RF) HPLC column of a short 5 cm length to identify antioxidants in a variety of complex samples represented by tea, utilising simultaneous multiplexed detection techniques. The detection responses included: underivatised UV at 280 nm, underivatised fluorescence detection (FLD) and a post column derivatisation (PCD) colorimetric response for antioxidants via the ferric reducing antioxidant power (FRAP) assay. Both similarities and differences in the chromatograms obtained using the three detection modes highlight the power of multiplexed detection. The main findings from the bioactive profiling include: the Fruit of the Forest tea contains the lowest level of antioxidants and is clearly distinct from the other three teas which are based on Camellia sinensis. The antioxidants detected in the green tea and the two black teas are similar. The green tea contains a higher concentration of the two main antioxidants that eluted at 4.0 and 5.7 min. The simplicity of sample analysis and chemical profiling using RF columns and selective detection was the focus of this study via the analysis of antioxidants in tea using the FRAP reagent. The main advantage of RF-PCD is the ability to split flows for simultaneous detection with minimal post column dead volume contributions, making peak matching between detectors much easier. To date, few studies exploit the power of RF-PCD multiplexed detection to differentiate and identify bioactive compounds in various complex samples. Applications of the approach developed in this study may include antioxidant screening and profiling of natural products, foods and beverages.
