El objetivo de esta tesis fue el estuóio y la caracterización de lipasas de semillas. Se seleccionó un grupo de especies de la familia de las Cucurbitaceas: zapallo (CuGurbita pepo), zapallito (CuGurbita maxima), pepino (Cucumis'satlvus) melon (Cucumis melo) y sandia (Citrolus lanatus). Esta familia se caracteriza por utilizar triacilgliceroles como fuente de carbono y energia, por 10 tanto es de esperar que durante la germinación de sus semillas aparezcan lipasas en sus tejidos de reserva. Para caracterizar las lipasas en su comportamiento en la hidrólisis endógena del aceite de reserva se estudió la variación de su composición y de su contenido en distintos estadios de la germinación. El estudio se realizó a varias temperaturas de germinación para determinar el efecto de esta variable. El efecto de la temperatura sobre las velocidades de germinación de estas especies, medidas en función del tiempo que demora una plantula en alcanzar un cierto estadio en su desarrollo, es muy notorio. Se establecieron correlaciones entre la velocidad de germinaci6n y la velocidad de consumo del aceite alas distintas temperaturas de trabajo. La composición del aceite varió muy poco durante la gerrninaci6n, aunque se observó un aumento en los <icidos grasos que 10 integran en forma minoritaria. Para poder utilizar estas lipasas, se las extrajo de los tejidos y se estudió su comportamiento en la hidr6lisis de un aceite comercial. Como los extractos obtenidos por 10s metodos utilizados comUnmente en la extracci6n de lipasas de semillas (crudos 0 polvos de acetona) presentaron actividad muy baja para todas las especies estudiadas, se optimizó un metodo diferente para lograr su extracción, y a los preparados de lipasas, obtenidos por ese metoda, se les llamó concentrados en polvo (C. P.) .. La actividad lipolitica de los C. P. se estudió en función del estadio y de la temperatura de germinación de las semillas. Las mayores actividades lipoliticas se obtuvieron en estadios de germinación altos y cuando la temperatura es la óptima para la germinación de las semillas.

Aspergillus nidulans possesses three well-characterized purine transporters encoded by the genes uapA , uapC and azgA . Expression of these genes in mycelium is induced by purines and repressed by ammonium or glutamine through the action of the pathwayspecific UaY regulator and the general GATA factor AreA respectively. Here, we describe the regulation of expression of purine transporters during conidiospore germination and the onset of mycelium development. In resting conidiospores, mRNA steady-state levels of purine transporter genes and purine uptake activities are undetectable or very low. Both mRNA steady-state levels and purine transport activities increase substantially during the isotropic growth phase of conidial germination. Both processes occur in the absence of purine induction and independently of the nitrogen source present in the medium. The transcriptional activator UaY is dispensable for the germination-induced expression of the three transporter genes. AreA, on the other hand, is essential for the expression of uapA , but not for that of azgA or uapC , during germination. Transcriptional activation of uapA , uapC and azgA during germination is also independent of the presence of a carbon source in the medium. This work establishes the presence of a novel system triggering purine transporter transcription during germination. Similar results have been found in studies on the expression of other transporters in A. nidulans , suggesting that global expression of transporters might operate as a general system for sensing solute availability.

The Asian citrus psyllid, Diaphorina citri, is currently the most important pest of citrus worldwide because it vectors ‘Candidatus Liberibacter’ spp., the etiological agents of lethal Huanglongbing (HLB). Reduction of D. citri populations is a key component of HLB management. Identifying potential D. citri attractants, such as volatiles, could be useful for behavioral management of this insect. This could, for instance, enhance catches of adults on monitoring traps. The overall aim of this study was to investigate the response of D. citri to volatiles characteristic of preferred citrus hosts. An ancillary objective was to formulate a synthetic blend of volatiles based on preferred host odors that could enhance monitoring traps. Volatile collections of two preferred citrus species, Duncan grapefruit (Citrus paradise) and sweet orange (Citrus sinensis), were performed in aeration chambers and analyzed by gas chromatography-mass spectrometry. Based on the grapefruit volatile profile, a synthetic blend consisting in d-limonene (0.62 mg), methyl N-methylanthranilate (0.19 mg), β-ocimene (0.11 mg), β-elemene (0.02 mg), and β-caryophyllene (0.02 mg) was prepared in 9.9 mL of dichloromethane and evaluated for attractiveness, as well as the individual compounds, and compared with a positive control of natural volatiles collected from Citrus sinensis. Behavior of D. citri adults was evaluated in two-choice behavioral assays comparing response to lures with different combinations and dosages of the volatiles deployed on yellow sticky traps. Individually, neither d-limonene nor methyl N-methylanthranilate affected behavior of D. citri. However, these volatiles increased catches of D. citri, as compared with solvent control, when presented as components of the synthetic grapefruit blend, as did the natural citrus odor positive control. Importantly, catch of D. citri appeared to vary with the release rate of the identified grapefruit volatile blend, which was attractive at a 0.1 mg loading dosage, but repellent at a dosage only 1 log step higher.

The citrus borer, Diploschema rotundicolle, is a Neotropical longhorn beetle that has become a serious citrus pest in southern South America. Management strategies for this insect rely on trimming off damaged shoots, which is expensive and inefficient.We studied the chemical communication system in D. rotundicolle in search of attractants for monitoring or control. GC-MS and enantioselective GC analyses of volatile extracts from field-collected adults showed that males produce (R)-3-hydroxy-2-hexanone, irregularly accompanied by minor amounts of 2,3-hexanediol (all four stereoisomers) and 2,3-hexanedione. Males emit the compounds only at night, when the adults are active. GC-EAD analyses of natural and synthetic compounds showed that bothmale and female antennae respond to the natural enantiomer (R)-3-hydroxy-2-hexanone, suggesting that itmay function as an aggregation-sex pheromone as seen in many cerambycines. The non-natural (S) enantiomer as well as the minor component 2,3-hexanediol did not trigger antennal responses. Field tests with the racemic 3-hydroxy-2-hexanone, enantiomerically pure (R)-3-hydroxy-2-hexanone, as well as a mixture of racemic 3- hydroxy-2-hexanone and 2,3-hexanediol, showed in all cases low capture levels of D. rotundicolle. However, increasing the elevation of the trap and the emission rate of dispensers enhanced field captures in traps baitedwith racemic hydroxyketone. Incidental catches of another native cerambycine, Retrachydes thoracicus, in traps baited with 3-hydroxy-2-hexanone are also reported. This is the first report of pheromone chemistry in the genus Diploschema and in the tribe Torneutini, reaffirming the pheromone parsimony well established for the Cerambycinae. Potential factors explaining the weak attraction of D. rotundicolle in the field are discussed

Sodium titanates represent a promising alternative for the construction of anodes for sodium-ion batteries. To study the electrical response of nanostructured sodium titanate samples under plausible temperature conditions, we evaluate the electrical impedance in the range of temperatures from −30 ◦C to 50 ◦C, and the effect of consecutive temperature cycling. Strong changes in electrical properties are observed during the first temperature cycle, followed by a stabilization in the consecutive cycles. This behavior is reversible when the sample is exposed to external conditions and room temperature for less than an hour. Probably a structural and surface water removal and a surface water reabsorption occur, affecting the charge carriers and with it the impedance. Structurally, no differences are detected before and after the cycles, nor when evaluating only the vacuum process. However, when the optical properties are evaluated we find non-reversible changes after the temperature cycles. The optical behavior after the temperature cycling becomes uniform, the same type of electronic defects become predominant and similar direct and indirect band gap values are obtained, regardless of the starting synthesis condition. These results have significant implications for potential technological applications of these materials.

The influence of two sample presentation and instruction delivery protocols on the results of consumer-based sensory evaluations was studied. In two independent experiments, five peach jams and four Colonia-type cheese (from the Colonia region in Uruguay) were taste-tested. In the first experiment, one group of 54 consumers used the sequential monadic (SM) and the other group of 54 consumers used the rank-rating (RR) sample presentation protocols for the jam sample evaluation. In the second experiment, one group of 54 consumers used the SM and the other group of 54 consumers used the RR sample presentation protocols for the cheese sample evaluation. All samples were evaluated using hedonic scales and CATA (check-all-that-apply) questions. Overall liking scores were found not to depend on the sample presentation protocol. Although the protocol was found to affect the choice and number of attributes listed in the CATA question used by respondents to differentiate the samples, the spatial configurations of samples derived by multiple factor analysis showed a high degree of correlation between the two protocols for most of the CATA attributes. Based on these results, the SM and RR protocols were found to be equally sensitive, for the products tested, in the conditions of this study.

<p>Natural fibres constitute interesting, low cost and environmentally friendly precursors of carbon fibres. However,<br />they are prone to fuse and degrade upon thermal treatment, affecting the properties of the products unless a stabilization step was included. Aiming to reach a better understanding of the stabilization mechanism of wool as a potential precursor for the preparation of carbon fibres, in this study we have analysed the chemical, structural and morphological transformations during its oxidative stabilization in air. Below 400 ◦C, both the structure and morphology of the fibres were preserved. The chemical composition changed with the stabilization temperature; main changes include the loss of secondary protein structure, cleavage of sulphur bridges, dehydration, oxidative crosslinking and rearrangements of N-groups through the formation of carbodiimides. Oxidation of the wool at 300 ◦C rendered materials with a fibrous morphology and high carbon yield (ca. 36 wt%), yielding carbon fibres upon carbonization at high temperatures.</p>

Tomato bacterial canker caused by Clavibacter michiganensis subsp. michiganensis is one of the most important seed-borne tomato diseases around the globe. The disease was initially reported in 1993 in Iran, and it became a rising threat for the multibillion dollar tomato industry of the country during the last decade. In this study, using phylogeographic analyses, we determined genetic diversity and geographic distribution of C. michiganensis subsp. michiganensis in Iran. Our field surveys showed that the pathogen is expanding into the southern and eastern areas of the country. Furthermore, multilocus sequence analysis and typing (MLSA/MLST) using the sequences of five housekeeping genes (atpD, gyrB, ppk, recA, and rpoB) revealed that 37 C. michiganensis subsp. michiganensis strains isolated in Iran had high genetic diversity and placed in 15 sequence types (STs), while all the available 184 worldwide C. michiganensis subsp. michiganensis sequences were placed in 43 STs. MLSA divided the worldwide C. michiganensis subsp. michiganensis strains into two phylogroups (I and II). Among the 37 strains isolated in Iran, 30 strains clustered in phylogroup I, while 7 strains clustered in phylogroup II. Phylogeographic data inferred from the allelic profile of the five housekeeping genes suggested multiple introductions of C. michiganensis subsp. michiganensis inoculum into Iran, while the geographic origin of the Iranian C. michiganensis subsp. michiganensis strains remains undetermined. Further analyses using higher numbers of strains are warranted to decipher the evolutionary history of C. michiganensis subsp. michiganensis in Iran. Additionally, stricter seed/transplant inspections are recommended to reduce the risk of pathogen expansion to areas with no history of the disease

Interest in consumer-based methodologies for sensory characterization has largely increased in the last decade. However, research on their applicability and reliability is still necessary. In this context, the aim of the present work was to compare three consumer-based methodologies and descriptive analysis for sensory characterization, considering as case study four sets of orange-flavoured powdered drinks, differing in the number of samples and the extent to which they represented the whole product category. A trained assessor panel evaluated sample sets using descriptive analysis, while groups of 100 consumers evaluated samples using one of three methodologies: CATA questions, projective mapping (PM) and polarized sensory positioning (PSP). Across the four sample sets, the three-consumer based methodologies provided similar information regarding the main similarities and differences among samples, which did not largely differ from that obtained using descriptive analysis. The main difference among the methodologies was related to the relative importance given to the sensory characteristics when evaluating differences among samples, which led to differences in the dimensionality of the sensory spaces. Sample configurations from PM and PSP required more dimensions than those from descriptive analysis and CATA questions to fully explain differences among samples. None of the consumer-based methodologies outperformed descriptive analysis in terms of ability to discriminate samples, being CATA questions the methodology that showed the lowest discrimination. Implications for the selection of consumer-based methodologies for specific applications are discussed.

Salt-reduction in processed products has been proposed as a high-impact intervention for reducing the sodium intake at population level. A major limitation for this approach is its potential negative impact on the sensory characteristics of products. The current practice in sensory and consumer science involves single sip/bite evaluations, which may not properly reflect the sensory experience that occurs during product consumption. In this context, the aim of the present work was to compare single and two bite evaluations of consumer sensory and hedonic perception of salt-reduced breads. Five studies with a total of 499 consumers were carried out, in which overall-liking scores of five salt-reduced bread samples were collected after the first and the second bite evaluation. In one of the studies consumers also answered a CATA (check-all-that-apply) question after the first and the second bite. Neither bite nor the interaction between samples and bite had a significant effect on hedonic scores. However, when hedonic scores were analysed separately for each bite, the overall liking scores from the second bite evaluation better reflected differences among samples according to their salt content in two of the five studies. The sensory characterization of the samples did not largely vary between the first and the second bite. Results suggest that consumers' perception of salt reduced bread samples did not largely vary between a single and a two bites evaluation. Further research is warranted in this regard, in particular considering more complex products.

Objective: Tostudy the influence of 2 interpretational aids of front-of-package (FOP) nutrition labels (color code and text descriptors) on attentional capture and consumers’ understanding of nutritional information. Design: A full factorial design was used to assess the influence of color code and text descriptors using visual search and eye tracking. Participants: Ten trained assessors participated in the visual search study and 54 consumers completed the eye-tracking study. Main Outcome Measures: In the visual search study, assessors were asked to indicate whether there was a label high in fat within sets of mayonnaise labels with different FOP labels. In the eye-tracking study, assessors answered a set of questions about the nutritional content of labels. Analysis: The researchers used logistic regression to evaluate the influence of interpretational aids of FOP nutrition labels on the percentage of correct answers. Analyses of variance were used to evaluate the influence of the studied variables on attentional measures and participants’ response times. Results: Response times were significantly higher for monochromatic FOP labels compared with colorcoded ones (3,225 vs 964 ms; P < .001), which suggests that color codes increase attentional capture. The highest number and duration of fixations and visits were recorded on labels that did not include color codes or text descriptors (P < .05). The lowest percentage of incorrect answers was observed when the nutrient level was indicated using color code and text descriptors (P < .05). Conclusions and Implications: The combination of color codes and text descriptors seems to be the most effective alternative to increase attentional capture and understanding of nutritional information.

Polarized projective mapping (PPM) combines the main features of projective mapping and polarized sensory positioning. The main advantage of this methodology is that it allows comparing results obtained in different sessions. The aim of this study was to evaluate the reliability of PPM, focusing on individual and global repeatability, global reproducibility and data aggregation from different sessions. A consumer study with two sample sets of powdered drinks was carried out. Consumers were randomly divided into four groups: two of the groups evaluated the same sample set in two different sessions, while the other two evaluated the samples in three sessions (in one of the sessions, they evaluated the whole sample set, whereas in the other two sessions they evaluated the sample set split into two sets). Results showed that PPM can be regarded as a repeatable and reproducible methodology. Preliminary evidence of the validity of aggregating data from different sessions was gathered.

In recent years high sodium intake has raised growing concern worldwide. A widespread reduction of salt concentration in processed foods has been claimed as one of the most effective strategies to achieve a short-term impact on global health. However, one of the major challenges in reducing salt in food products is its potential negative impact on consumer perception. For this reason, gradual salt reduction has been recommended. In this context, the aim of the present work was to present a consumer-based approach to salt reduction, using bread as case study. Two consumer studies with a total of 303 consumers were carried out. In the first study, four sequential difference thresholds were determined through paired-comparison tests, starting at a salt concentration of 2%. In the second study, 99 consumers performed a two-bite evaluation of their sensory and hedonic perception of five bread samples: a control bread containing 2% salt and four samples with reduced salt content according to the difference thresholds determined in the first study. Survival analysis was used to determine average difference thresholds, which ranged from 9.4% to 14.3% of the salt concentration of the control bread. Results showed that salt concentration significantly influenced consumer overall liking of the bread samples. However, large heterogeneity was found in consumer hedonic reaction towards salt reduction: two groups of consumers with different preference and hedonic sensitivity to salt reduction were found. Results from the present work confirm that cumulative series of small salt reductions may be a feasible strategy for reducing the sodium content of bread without affecting consumer hedonic perception and stress the importance of considering consumer perception in the design of gradual salt reduction programmes.

Several studies have shown that the inclusion of front of pack nutritional warnings is a potentially effective strategy to encourage consumers to avoid products with high content of nutrients associated with non-communicable diseases. In this context, the present work aimed at exploring consumers’ reaction towards sodium reduction in the context of the implementation of warnings in three scenarios: package evaluation, tasting and intention to re-purchase the product after tasting, using white bread as case study. A total of 171 consumers participated in the study that encompassed four parts. Firstly, consumers received two bread samples with different salt content (2.00% and 1.38% salt) under blind conditions and were asked to rate their overall liking and to indicate the one they preferred. Then, they were asked to look at four bread packages differing in graphic design and the presence of warnings. They had to rate their expected liking and to indicate the one they would purchase. Finally, consumers were asked to try the bread they had previously chosen, to assess their overall liking and to indicate their willingness to purchase it again. Salt content significantly affected consumer hedonic reaction towards the breads. Two consumer segments with different preference for the salt content in bread were identified: 58% of consumers preferred the bread with 2.00% salt while 42% preferred breads with 1.38% salt. However, when looking at the packages the majority of consumers in both groups selected bread packages that did not feature warnings. In addition, after having tried the bread, most consumers were willing to buy the bread they had previously chosen again, which is promising in terms of reaching sustainable changes towards lower salt levels. Results from the present work suggest that, in the case of bread, nutritional warnings have potential to shift consumers’ preferences to lower sodium content, even after trying the products.
