The development of strategies to reduce sugar intake of children and adolescents has been deemed necessary to cope with the global obesity epidemic. In this context, the aims of the present work were to study attitudes towards sugar reduction in three dairy products of children and adolescents of two income levels and to assess if these attitudes were modulated by the inclusion of the traffic light system on labels. A total of 646 children and adolescents from three primary/secondary schools in Montevideo (Uruguay) participated in the study. A total of twelve labels were designed following a full factorial design with three variables: dairy product (3 levels: yogurt, chocolate-flavoured milk and vanilla milk dessert), sugar reduction claim (2 levels: absent and present "20% less sugar") and traffic light system (2 levels: absent and present). Children had to evaluate their expected liking of the labels and to answer a check-all-that-apply question composed of terms related to tastiness, healthfulness and fun. Sugar claim significantly increased children and adolescents' expected liking scores (p=0.01), suggesting they had a positive attitude towards sugar reduction in the evaluated dairy products. However, the influence of sugar reduction claim on hedonic expectations and healthfulness perception was modulated by the inclusion of the traffic-light system on labels (p=0.01). This front-of-pack nutrition labelling scheme increased their unhealthfulness perception of products with high sugar content and could potentially discourage their consumption. Age and income modulated the influence of these strategies on hedonic and healthfulness perception (p<0.05).

Reducing the added sugar content of sugar‐sweetened beverages is one of the most cost‐effective strategies to achieve short‐term changes in added sugar intake. The present study aimed at evaluating children and adults' sensory and hedonic perception of added sugar reduction in grape nectar. For this purpose, two studies were carried out. In the first study, five sequential difference thresholds for added sugar in grape nectar were determined with school‐aged children and adults using paired comparisons. In the second study 105 children (6–12 years old) and 100 adults evaluated six samples with different added sugar content. They were asked to rate their overall liking and to answer a check‐all‐that‐apply question. Results confirmed the feasibility of reducing the added sugar concentration of grape nectar without affecting the sensory and hedonic perception of both children and adults. Children were less able to detect changes in the sensory characteristics of the added sugar reduced nectar samples than adults but showed higher hedonic sensitivity to added sugar reduction. Large heterogeneity in hedonic reaction toward added sugar‐reduced nectar samples was found in both age groups.

The aim of the present work was to explore children’s reaction to sugar reduction in the context of the imple mentation of nutritional warnings in Uruguay. The study was composed of two sessions involving three evalu ation conditions: tasting without package information, package evaluation without tasting, and tasting with package information. A total of 122 children, ages ranging between 6 and 13 years old (47% girls) were involved in the study. In the first session, children’s hedonic and emotional response to a regular and a sugar-reduced chocolate dairy dessert (without other sweeteners) was evaluated. In the second session, children first evalu ated their expected liking, emotional associations and choice of packages differing in the presence of warning labels for excessive sugar content and cartoon character (2x2 design). Finally, they tasted the selected sample in the presence of the package and evaluated their liking, emotional associations, and intention to re-taste it. Although sugar reduction led to a significant reduction in overall liking, the dessert with 40% sugar reduction showed a mean overall score of 6.5 in a 9-point hedonic scale and was described using positive emoji ( , , ). When the desserts were tasted with package information, no significant differences in the expected overall liking of the regular and sugar-reduced dessert were found. Regarding the effect of packaging elements, the presence of a warning label highlighting excessive content of sugar did not have a significant effect on children’s choice. Instead, children’s choices were defined by the presence of a cartoon character. Results from the present work provide additional evidence about the feasibility of reducing the sugar content and sweetness of dairy products targeted at children and stress the need to regulate the use of cartoon characters on products with an unfavorable nutritional profile.Methodological recommendations for sensory and consumer research with children are also discussed.

The study aimed to assess the application of the Approach avoidance task (AAT) with children to measure implicit motivational tendencies towards foods differing in sweetness and calorie content and to explore the relationship between approach bias and explicit measurements of expected liking, attitudes, and hunger state and their relation to paired-preference tasks. The simplicity and game-like procedure of the AAT, where participants use a joystick to pull or push pictures, seems particularly suitable to measure implicit motivational biases in children. However, to our knowledge, this approach has not been used with children in a food related context.<br />Children aged 9–11 participated in the study (n = 114). Their implicit bias towards pictures of snacks was measured via AAT. The test instruction was based on pushing or pulling the joystick according to picture category, food vs. non-food: food (18 snack pictures varying in sweetness and calorie) vs. non-food (18 pictures visually similar to the respective food stimuli). Further, children rated their expected liking of the snack pictures, answered an attitude questionnaire related to health and sugar consumption, and completed two paired preference tests tasting real samples under blind condition and choosing between a sugar and no-sugar added chocolate milk take-home pack.<br />The percentage of non-valid AAT responses was relatively high, leading to low testing power. There was a significant difference in approach bias between food pictures and non-food pictures; approach bias was positive for food and slightly negative for non-food. Within food pictures, no significant effect of sweetness nor calorie was found. Nevertheless, children’s approach biases were linked to their expected liking ratings, which revealed a clear preference towards high sweetness and high calorie snacks. Individual differences in children’s approach bias to pictures differing in sweetness and calorie content were related to their hunger state but not to their attitudes or preference towards chocolate milk, indicating relevance for situational food choices. In the present study, questionnaire-based measurements (affective and cognitive attitude towards sugar, sugar craving and using food as reward) were however associated with children’s preference towards chocolate milk (blind and/or informed). Higher scores in the measured attitude subscales craving for sweet food, using food as reward, affective attitude towards sweet food and cognitive attitude towards sweet food were associated with higher odds to choose the sugar added chocolate milk. Methodological considerations and recommendations with regards to the use of approachavoidance testing with children are critically discussed.

BACKGROUND: Piezodorus guildinii (Westwood, Hemiptera: Pentatomidae) causes economic losses in soybean crops by diminishing productivity, impacting grain size, oil content, and germination power. RNA interference (RNAi), a conserved gene regulation process initiated by double-stranded RNA (dsRNA), may be used as a safe technique in integrated pest management strategies. Conjugating dsRNA with cationic polymers, which protect dsRNA from degradation has shown to enhance this response. This study aims to assess the impact of dsRNA administration on the viability of P. guildinii and evaluate dsRNAchitosan nanocomplexes as a pest control strategy. RESULTS: Ten target genes were selected, dsRNAs were designed and synthesized. In injection assays, significant mortality exceeding 76% was observed, while mortality from ingestion also showed significant differences, reaching 49%. The dsRNA targeting Srp54k gene was selected and a chitosan-dsRNA nanocomplex was synthesized. These nanocomplexes demonstrated enhanced stability under RNase treatment in vitro, and, in in vivo assays, a cumulative corrected mortality rate of 66% was reported. Consistent with these findings, RT-qPCR analysis revealed that gene expression levels in treated insects were significantly reduced compared to control at 24, 48, and 72 h post treatment for targeted genes. CONCLUSION: In this work, we evaluated the effect of dsRNA administration by injection and ingestion on the viability of P. guildinii and determined its molecular silencing effects. Additionally, we synthesized and characterized chitosan-dsRNA nanoparticles, which demonstrated improved efficacy in terms of stability. These findings are promising for the development of new integrated management strategies for stink bugs control.

The objective of this research was to obtain and characterize chitosan (CS) based matrices conceived as folic acid (FA) dried reservoir that could find application as food coating upon rehydration. An Argentinean and high molecular weight (MW) chitosan (AC) was used and contrasted with a commercial type one (CC). FTIR and XRD were applied for the polysaccharide-vitamin dried mixed systems characterization. Shifts in bands of FA and AC or CC were observed in the mixed systems, confirming the interaction among the components, for example, the bands at 1382 and 1391 cm− 1 (AC and FA, respectively) shifted to 1387 cm–1 in the in AC-FA mixed systems. XRD data showed that the FA is in its protonated form as expected and is maintained in the different mixed systems obtained using either with CC or AC, demonstrating the robustness and applicability of the material to properly conserve the active ingredient. The ultrastructure and topography were studied by SEM and AFM, respectively. FA incrustations in a self-associated form were observed in the topography images. Variations in topography observed for the AC-FA and CC-FA mixed systems could be attributed to specific interactions between the components and the molecular features of each CS (i.e. the mean particle size was 63 ± 3 and 9 ± 1 nm and, roughness gave values of 0.47 and 0.41 nm, respectively). Vitamin bioaccessibility remained unchanged compared to the control (88.36% ±10.35%), being equal to 91.08% ± 5.02% for AC-FA and 71.56% ±14.08% for CC-FA, after submitted samples to an in vitro digestion process. In sum, mixed AC-FA systems obtained are promising materials for a possible additional application in coating matrices inspired by biopolymers to improve food preservation, maintaining the bioaccessibility of vitamins.

The aim of the present work is to prepare and characterize a functionalized latex with chloromethyl groups on the surface and to perform the covalent coupling of anti-human serum albumin (a-HSA) IgG protein. The chloromethyl-styrene latex (CMS) was synthesized by means of a core-shell emulsion polymerization in a batch reactor. The monodisperse-obtained latex was characterized by determining the diameter (TEM and PCS), the surface charge density (conductometric and potentiometric titration), the amount of chloromethyl groups on the surface (hydrolysis reaction), and the stability vs electrolyte concentration (turbidity measurements). Electrokinetic characterization was also performed (electrophoretic mobility versus pH and ionic strength). IgG was chemically bound to the latex particles under different sensitization and block-stabilization conditions. Colloidal stability of complexes was studied to select an immunolatex suitable for the development of latex immunoassays. The final part of this work consists of a study of the immunoreactivity of the IgG-latex complexes at different pH and ionic strength, in particular under physiological conditions. The results show that chemically bound IgG to chloromethyl latex provides an IgG-latex complex suitable for application in immunodiagnosis tests.

Background: Prenylated flavanones represent a structurally diverse class of natural compounds with significant biological potential. Among them, chromene flavanones (CFs) constitute a rare and specialized subgroup with promising therapeutic applications. These molecules have gained attention due to their potential to inhibit xanthine oxidase (XO), a key enzyme involved in oxidative stress-related disorders such as gout and hyperuricemia. Their distinctive structural features, combined with notable bioactivity, make them compelling candidates for further pharmacological exploration. Given their potential relevance, this study focuses on the in vitro and in silico evaluation of three CFs isolated from Dalea boliviana Britton [Fabacea], assessing their capacity to inhibit XO and elucidating key structure–activity relationships (SARs) that contribute to their biological effectiveness. Purpose: This study aims to investigate the in vitro and in silico interactions of the chromene flavanones, namely, (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno- (7,8:2″,3″)-flavanone (1), (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno-(7,8: 2″,3″)-3′-prenylflavanone (2), and obovatin (3), obtained from D. boliviana, with XO, in order to explore their potential as XO inhibitors and their potential therapeutic applications for hyperuricemic diseases. Material and Methods: XO inhibition by the three chromene flavanones was measured spectroscopically. The relationships between their structures and inhibitory activities were evaluated. Moreover, molecular docking studies were performed to propose the binding modes of the most active natural compounds. Results and discussion: Compounds 1 and 2 exhibited potent inhibition, with IC50 values in the nanomolar range (0.5 ± 0.01 nM and 1.7 ± 0.46 nM, respectively), demonstrating significantly higher activity than allopurinol (AL), the reference inhibitor (IC50 = 247 ± 4 nM). In contrast, compound 3 displayed only weak inhibition. SAR analysis revealed that the presence of a chromene moiety in the A-ring, combined with hydroxyl and prenyl groups in the B-ring, played a crucial role in enhancing inhibitory activity. Molecular docking studies confirmed the strong binding affinities of compounds 1 and 2 within the active site of XO (PDB ID: 3NVY), with binding energies of −6.1687 kcal/mol and −6.7820 kcal/mol, respectively. Key stabilizing interactions involved π–π interactions with Phe914 and hydrogen bonding with residues such as Leu873 and Leu1014. These findings highlight the structural features essential for potent XO inhibition and suggest that chromene flavanones represent a valuable scaffold for the development of novel inhibitors. Further molecular dynamics simulations could provide deeper insights into their stability and interaction dynamics, aiding in the rational design of more effective XO inhibitors. Conclusion: Our findings lead us to propose these chromene flavanones as lead compounds for the design and development of novel XO inhibitors for treating diseases in which exacerbated activity of this enzyme is involved.

Background: Efflux transporters overexpression has been proposed as one of the responsible mechanism for refractory epilepsy by preventing access of the antiepileptic drug to the brain. In this work we investigated whether phenytoin (PHT), could induce efflux transporters overexpression, at different biological barriers and to evaluate the implication it could have on its pharmacokinetics and therapeutic/toxic response. Methods: Forty-two adult females Sprague Dawley divided in five groups were treated with oral doses of 25, 50 and 75 mg/kg/6 h of PHT for 3 days and two additionally groups were treated with intraperitoneal ( ip ) doses of 25 mg/kg/6 h or 100 mg/kg/24 h. At day 4 PHT plasma concentrations were measured and, obtained several organs, brain, parotid gland, liver and duodenum in which were analyzed for the Pgp expression. At day 4 PHT plasma concentrations were measured and several tissues: brain, parotid gland, liver and duodenum were obtained in order to analyze Pgp expression. In order to evaluate the oral bioavailability of PHT, two groups were administered with oral or intraperitoneal doses of 100 mg/kg and plasma level were measured. Results: An induction of the expression of efflux transporter mediated by phenytoin in a concentration- and-time dependent manner was found when increasing oral and ip doses of phenytoin, One week after the interruption of ip treatment a basal expression of transporters was recovered. Conclusions: Overexpression of efflux transporters can be mediated by inducer agents like PHT in a local- concentration dependent manner, and it is reversible once the substance is removed from the body. The recovery of basal Pgp expression could allow the design of dosing schedules that optimize anticonvulsant therapy

Sturgeon aquaculture faces challenges due to global warming, as chronic heat stress (CHS) jeopardises fish health and production sustainability. To tackle this issue, we analysed serum innate components and the cellular composition of the peritoneum in Russian sturgeons (Acipenser gueldenstaedtii) subjected to CHS and challenged with Aeromonas hydrophila. Russian sturgeons exposed to CHS exhibited a reduction in the serum levels of total protein and alkaline phosphatase activity, trending to a higher decrease in sturgeons exposed to both bacterial challenge and CHS. This sturgeon group showed a worsened health status, reduced growth and a higher mortality rate. Under tolerable temperatures, the bacterial challenge increased serum ceruloplasmin, lysozyme and serum amyloid A levels, which tended to be lower under CHS. The naive peritoneal cellular composition of the Russian sturgeon revealed a predominance of myeloid cells, with lymphoid cells present in a smaller proportion. CHS altered this distribution, increasing lymphoid while reducing myeloid cell percentages and A. hydrophila challenge reverted these alterations. Myeloid cells exhibiting the highest phagocytic potential were reduced by CHS, potentially compromising peritoneal bacterial clearance. Principal component analysis revealed that sturgeons’ integral innate response varied according to temperature exposure, evidencing CHS’s impact on the antibacterial response. Overall, this study provides a first picture of the peritoneal cellular composition of a chondrostean fish. Furthermore, it demonstrates the disruption caused by CHS on the sturgeon’s early systemic and peritoneal responses mounted against A. hydrophila.

A diurnal variation study of carbamazepine (CBZ) and its metabolite carbamazepine-10,11-epoxide (EPOX) was carried out on six healthy volunteers, measuring drug levels in plasma and saliva throughout 34 hours. Four hundred milligrams of CBZ were given to subjects at 8:00 am or 8:00 pm, in a two-way crossover design. Drug and metabolite concentration in plasma and saliva stimulated with parafilm (after 24 hours post-dose) were higher during the night. The EPOX/CBZ metabolic ratio did not show any change between dark and light periods. The non-stimulated saliva/plasma concentration ratio (S/P) was higher during the day, but this quotient did not change when stimulated saliva was considered. Diurnal variation in the body blood flow seems to be a reasonable explanation for those differences. Factors such as protein binding or metabolic pathway are discarded as sources of carbamazepine chronopharmacokinetic variations.

Las floraciones de cianobacterias en cuerpos de agua dulce constituyen un problema mundial, asociado a la eutrofización (enriquecimiento de nutrientes) y cambios hidrológicos de los ecosistemas. En este estudio se generó una base de datos históricos (n= 3061 para 64 ecosistemas, de 1980 a 2014), elaborada por un grupo de trabajo interinstitucional (OSE, DINAMA, IM, Universidad de la República e IIBCE), y se analizó la distribución de cianobacterias planctónicas y de cianotoxinas (microcistina, saxitoxina y cilindrospermopsina) en Uruguay. Los valores de clorofila a y nutrientes totales indicaron procesos de eutrofización en diversos ecosistemas. En dos mapas georreferenciados se visualiza la distribución de cianobacterias en el país, según indicadores cuantitativos globales, y de la microcistina (cianotoxina más frecuente). En estos mapas se destacan los embalses (Río Uruguay y Río Negro) y las playas del Río de la Plata como las zonas de mayor riesgo de exposición a cianobacterias según las categorías de la Organización Mundial de la Salud para aguas recreacionales. Además, se analizaron muestras de floraciones (espuma) y se cuantificó 20 mg L-1 de microcistinas totales y la presencia de genes que sugieren diferentes variedades de microcistinas. Se generó información básica que podrá ser útil para programas de monitoreo nacionales e investigación

<p><em>Cissampelos pareira</em>L. (Menispermaceae) is a widely used medicinal plant, also used to treat bites of venomous animals, particularly snakes. Ophidian accidents are a serious public health problem in Argentina where the Bothrops genus of snakes is responsible for 97% of these accidents, peculiarly B. diporus (“yarará chica”) responsible for 80% of them. In the northeast of the country (Corrientes Province), C. pareira is commonly used against the venom of B. diporus; its use is described in almost all-ethnobotanical literature of the countries where the plant grows. In vitro and in vivo antivenom activities of C. pareira extracts from plants collected at two different phytogeographic regions of Corrientes (Argentina) were evaluated against B. diporus venom. The seasonal influence on the chemical composition of extracts was also studied to determine the associated range of variability and its influence on the antivenom activity. Besides, an evaluation of the chemical composition of the volatile extract was performed to assess its chemical stability. These results support the ethnopharmacological use of this species. Moreover, the presented data demonstrate that certain flavonoids may mitigate some of the venom-induced local tissue damages.</p>

La cisteína endopeptidasa de T. cruzi llamada cruzipaína, también conocida como cruzaína, es la enzima responsable de la mayor actividad proteolítica en todos los estadios del ciclo de T. cruzi. En las últimas décadas, estudios independientes la validan como un blanco prometedor contra dicho parásito. Numerosas son las publicaciones en las que se describen varias clases de nuevos inhibidores irreversibles o reversibles de esta enzima. Si bien, en la actualidad en el mercado no existe algún fármaco que posea dicho mecanismo, la vinilsulfona K777 se encuentra en etapa de estudios clínicos avanzados.
