Coca-paste (CP) is a drug of abuse that so far has not been extensively characterized. CP is an intermediate product of the cocaine alkaloid extraction process from coca leaves, hence it has a high content of cocaine base mixed with other chemical substances (impurities) and it is probably adulterated when it reaches the consumers. Despite its high prevalence and distribution through South America, little is known about its effects on the central nervous system. In the present study, a chemical analysis of CP samples from different police seizures was performed to determine the cocaine base content and the presence and content of impurities and adulterants. Some CP representative samples were selected to study the effects on the locomotor activity induced after acute systemic administration in rats as a measure of its stimulant action. The behavioral response was compared to equivalent doses of cocaine. As expected, cocaine was the main component in most of the CP samples assayed. Caffeine was the only active adulterant detected. Interestingly, several CP samples elicited a higher stimulant effect compared to that observed after cocaine when administered at equivalent doses of cocaine base. Combined treatment of cocaine and caffeine, as surrogate of different CP samples mimicked their stimulant effect. We demonstrated that cocaine and caffeine are the main components responsible for the CP-induced stimulant action while the contribution of the impurities was imperceptible.

<p>Se reportan laws variaciones del colesterol total y del HDL y no HDL colesterol, así como el Índice de Castelli, en un estudio transversal realizado en intervalos de tiempo similares en mujeres embarazadas normales. Los datos provenientes de 210 mujeres embarazadas normales en diferentes estadios, se compararon con los de una población de referencia de 30 mujeres normales no grávidas con edades similares. El colesterol HDL fue aislado con ácido fosfotúngstico/cloruro de magnesio, el colesterol se determinó enzimáticamente y el Índice de Castelli así como el no HDL colesterol fueron calculados en cada caso. Nuestro objetivo es estudiar los cambios lipídicos en esta población de embarazadas con el propósito de establecer los valores de referencia e investigar si la gestación podría estar relacionada a situaciones de riesgo cardiovascular. El HDL colesterol decreció significativamente entre las semanas 24a y 28a, al tiempo que el Índice de Castelli alcanzó el máximo valor de riesgo. Desde este punto de vista se puede deducir que este proceso puede estar relacionado con el comienzo de la resistencia a la insulina y el mayor riesgo de aparición de diabetes gestacional.</p>

A summary of the resource.
The results of computer models for colloidal aggregation, that consider both Brownian motion and gravitational drift experienced by the colloidal particles and clusters, are extended to include concentrations spanning three orders of magnitude. In previous publications and for a high colloidal concentration, it was obtained that the aggregation crosses over from diffusion-limited colloidal aggregation (DLCA) to another regime with a higher cluster fractal dimension and a speeding up followed by a slowing down of the aggregation rate. In the present work we show, as the concentration is decreased, that we can still cross over to a similar regime during the course of the aggregation, as long as the height of the sample is increased accordingly. Among the differences between the mentioned new regimes for a high and a low colloidal concentration, the cluster fractal dimension is higher for the high concentration case and lowers its value as the concentration is decreased, presumably reaching for low enough concentrations a fixed value above the DLCA value. It is also obtained the fractal dimension of the sediments, arising from the settling clusters that reach the bottom and continue a 2D-like diffusive motion and aggregation, on the floor of the container. For these clusters we now see two and sometimes three regimes, depending on concentration and sedimentation strength, with their corresponding fractal dimensions. The first two coming from the crossover already mentioned, that took place in the bulk of the sample before the cluster deposition, while the third arises from the two-dimensional aggregation on the floor of the container. For these bottom clusters we also obtain their dynamical behavior and aggregation rate.

Purpose The current research aimed to examine the prevalence of Instagram posts featuring ultra-processed products targeted at adolescents in Uruguay and hence investigate the frequency of such posts among a vulnerable consumer segment in a country that cannot be classified as WEIRD (i.e. Western, educated, industrialized, rich and democratic). Design/methodology/approach The study relied on a cross-sectional content analysis. A total of 2,014 Instagram posts promoting ultraprocessed products or brands commercializing such products, generated by 118 Instagram accounts between August 15th, 2020, and February 15th, 2021, were analyzed. Nine indicators of food marketing targeted at adolescents were selected to identify posts targeted at this age segment. Inductive coding was used to describe the content of the posts. Descriptive statistics and generalized linear models were used to analyze the data. Findings In total, 17.6% of the posts were identified as targeted at adolescents. Graphic design and adolescent language were the most prevalent indicators of marketing targeted at adolescents, followed by explicit references to adolescents or young adults and memes. Posts identified as targeted at adolescents mainly promoted snacks and discretionary foods. Differences in the content of posts identified as targeted and not targeted at adolescents were observed. Research limitations/implications The analysis was restricted to one social media platform in one country during a limited period of time, which limits the generalizability of the findings to other media platforms, samples and settings. Social implications Results stress the need to implement digital food marketing regulations to reduce exposure of adolescents to the deleterious effects of stemming from marketing of unhealthy foods and provide empirical evidence to inform their development. A group of Gram-negative denitrifying bacteria has been isolated from a denitrifying reactor treating landfill leachate. The new isolates produced both oxidase and catalase and showed growth on acetate, butyrate, n-caproate, i-butyrate, i-valerate, propionate, n-valerate, lactate, alanine, benzoate, phenylalanine and ethanol. No growth was observed on sugars. The bacteria could perform anoxic reduction of nitrate, nitrite and nitrous oxide to nitrogen, coupled to the oxidation of the same substrates as those used under aerobic conditions, except for aromatic compounds. They were very efficient denitrifiers, as estimated from the specific rate of N2 gas production. All the strains showed the same 16S rDNA restriction profile and one of them, designated 23310T, was selected for phylogenetic analysis. The organism clustered within the family Comamonadaceae, being related to Comamonas terrigena (95.8% sequence similarity). On the basis of the phylogenetic analysis, physiological characterization and the ability to efficiently reduce nitrate to N2, it is proposed that the bacterium be assigned to a new species, Comamonas nitrativorans. The type strain is 23310T (= DSM 13191T = NCCB 100007T = CCT 7062T).

Freshwater harmful algal blooms produce a broad array of bioactive compounds, with variable polarity. Acute exposure to cyanotoxins can impact the liver, nervous system, gastrointestinal tract, skin, and immune function. Increasing evidence suggests chronic effects from low-level exposures of cyanotoxins and other associated bioactive metabolites of cyanobacterial origin. These sundry compounds persist in drinking and recreational waters and challenge resource managers in detection and removal. A systematic approach to assess the developmental toxicity of cyanobacterial metabolite standards was employed utilizing a robust and high throughput developmental Danio rerio embryo platform that incorporated a neurobehavioral endpoint, photomotor response. Subsequently, we applied the platform to cyanobacterial bloom surface water samples taken from temperate recreational beaches and tropical lake subsistence drinking water sources as a model approach. Dechorionated Danio rerio embryos were statically immersed beginning at four to six hours post fertilization at environmentally relevant concentrations, and then assessed at 24 h and 5 days for morbidity, morphological changes, and photomotor response. At least one assessed endpoint deviated significantly for exposed embryos for 22 out of 25 metabolites examined. Notably, the alkaloid lyngbyatoxin–a resulted in profound, dose-dependent morbidity and mortality beginning at 5 μg/L. In addition, hydrophobic components of extracts from beach monitoring resulted in potent morbidity and mortality despite only trace cyanotoxins detected. The hydrophilic extracts with several order of magnitude higher concentrations of microcystins resulted in no morbidity or mortality. Developmental photomotor response was consistently altered in environmental bloom samples, independent of the presence or concentration of toxins detected in extracts. While limited with respect to more polar compounds, this novel screening approach complements specific fingerprinting of acutely toxic metabolites with robust assessment of developmental toxicity, critical for chronic exposure scenarios.

The effect of pure Zn2+ and combined Zn2+ and Mn2+ supplements on the growth and ligninolytic activity of wood-rotting basidiomycetes was studied. Based on measurements of minimum inhibitory concentration and minimum decolourisation-inhibiting concentration available from cultures of Inocutis sp., Trametes versicolor and Gloeophyllum trabeum, and a comparison of such results with those obtained in previous work for P. chrysosporium and P. atropurpurascens, metal supplements were shown to inhibit fungal physiology at concentrations within the study range. Average minimum inhibitory concentration values were consistently lower for ZnSO4 than for ZnSO4/MnSO4 (2 mM Zn2+), demonstrating the antagonistic effect of the tested cations. Furthermore, because the ligninolytic system of P. atropurpurascens had shown a high tolerance to metals, its ligninolytic and germinative behaviour was compared with that of P. chrysosporium. Conidial germination was inhibited at 1 mM of Zn2+ in the two strains, whereas extracellular activity was inhibited at concentrations above 2 mM.<br /><br />

A combination of three-dimensional quantitative structure-activity relationship (3D-QSAR), and molecular modelling methods were used to understand the potent inhibitory NAD(P)H:quinone oxidoreductase 1 (NQO1) activity of a set of 52 heterocyclic quinones. Molecular docking results indicated that some favourable interactions of key amino acid residues at the binding site of NQO1 with these quinones would be responsible for an improvement of the NQO1 activity of these compounds. The main interactions involved are hydrogen bond of the amino group of residue Tyr128, π-stacking interactions with Phe106 and Phe178, and electrostatic interactions with flavin adenine dinucleotide (FADH) cofactor. Three models were prepared by 3D-QSAR analysis. The models derived from Model I and Model III, shown leave-one-out cross-validation correlation coefficients (q2LOO ) of .75 and .73 as well as conventional correlation coefficients (R2 ) of .93 and .95, respectively. In addition, the external predictive abilities of these models were evaluated using a test set, producing the predicted correlation coefficients (r2pred ) of .76 and .74, respectively. The good concordance between the docking results and 3D-QSAR contour maps provides helpful information about a rational modification of new molecules based in quinone scaffold, in order to design more potent NQO1 inhibitors, which would exhibit highly potent antitumor activity.

Echinococcus granulosus is the parasite responsible for cystic echinococcosis (CE), an important worldwide‐distributed zoonosis. New effective vaccines against CE could potentially have great economic and health benefits. Here, we describe an innovative vaccine design scheme starting from an antigenic fraction enriched in tegumental antigens from the protoscolex stage (termed PSEx) already known to induce protection against CE. We first used mass spectrometry to characterize the protein composition of PSEx followed by Gene Ontology analysis to study the potential Biological Processes, Molecular Functions, and Cellular Localizations of the identified proteins. Following, antigenicity predictions and determination of conservancy degree against other organisms were determined. Thus, nine novel proteins were identified as potential vaccine candidates. Furthermore, linear B cell epitopes free of posttranslational modifications were predicted in the whole PSEx proteome through colocalization of in silico predicted epitopes within peptide fragments identified by matrix‐assisted laser desorption/ionization‐TOF/TOF. Resulting peptides were termed “clean linear B cell epitopes,” and through BLASTp scanning against all nonhelminth proteins, those with 100% identity against any other protein were discarded. Then, the secondary structure was predicted for peptides and their corresponding proteins. Peptides with highly similar secondary structure respect to their parental protein were selected, and those potentially toxic and/or allergenic were discarded. Finally, the selected clean linear B cell epitopes were mapped within their corresponding 3D‐modeled protein to analyze their possible antibody accessibilities, resulting in 14 putative peptide vaccine candidates. We propose nine novel proteins and 14 peptides to be further tested as vaccine candidates against CE.

This research explores the hydrochar production from grape pomace via hydrothermal carbonization and the determination of the kinetic parameters of its combustion process. The hydrochar was obtained by hydrothermal carbonization at 220 ◦C for 240 min with self-generated pressure. The process indicated a mass yield of 65.21 %, and the hydrochar exhibited an HHV of 26.60 MJ/kg, showing its potential as an efficient biofuel. Thermokinetic analysis of hydrochar combustion was conducted using multiple complementary methods: isoconversional analysis, mathematical deconvolution, combined kinetic analysis of each step, and optimization of parameters through nonlinear regression. The findings indicate that the overall process starts at ~152 ◦C and involves three apparent overlapping steps. This research analyzes the thermal and kinetic behavior of the combustion of hydrochar obtained industrially from green waste biomass. The combustion kinetic analysis was studied using thermogravimetry and applying complementary methods such as Friedman’s isoconversional analysis, mathematical deconvolution, combined kinetic analysis, and optimization by nonlinear regression of the kinetic curves. The process involves two overlapping steps: the organic compounds devolatilization and the oxidation of fixed carbon. For steps 1 and 2, the apparent activation energies were 178.5 and 183.1 kJ mol− 1, while the apparent ln(A∕s−1) were 31.2 and 30.0, respectively. The truncated Šesták–Berggren model was used as a surrogate for distributed reactivity, resulting in n1 = 1.37 and m1 = −0.54 for step 1, and n2 = 3.31 and m2 = 0.496 for step 2. In step 1, the m < 0 serves as a surrogate for an activation energy distribution, while in step 2, the value of n > 1 would substitute a gamma distribution of frequency factors. Finally, the obtained kinetic model allows successful predictions of thermal programs outside the range of the analysis.

The physico-chemical properties of some 5-nitro-2-furaldehyde semicarbazones (nitrofurazones) and thiophene analogues were compared with their in vitro and in vivo trypanocidal activity against Trypanosoma cruzi (Tulahuen strain). 3D-QSAR models were obtained by applying the SIMCA methodology to the electrostatic and steric fields (CoMFA fields) of the molecules. Nitrofurazones bearing N4 substituents. which cover a range of 14-17 A from the nitro group with a thickness of about 6 A when considering the extended conformer. produced complete survival in infected mice. The in vitro model allows larger N4 substituents than the SURVival model, but they must not bear positive centres in the region 15-16 A from the nitro group. Moreover, the in vitro model is in agreement with the active site of trypanothione reductase (TR). Abstract
A summary of the resource.
Neuroscience has the potential to allow us to more fully understand eating behavior by providing information that is inaccessible by conventional methods. However, it cannot be expected to solve all the problems sensory and consumer researchers face when trying to understand consumer food-related decisions. In the present work, we provide a commentary on the opinion paper by Niedziela & Ambroze. We urge sensory and consumer researchers to get familiar with neuroscience methods, their potential and limitations, as well as to engage in more interdisciplinary research to validate the application of neuroscience tools in the field.

Even beer being the most consumed alcoholic beverage around the world, there is not enough information generated for craft beers produced in Latin America, for either volatile profiles or physicochemical studies. In this work, the chemical and volatile components of ten commercial Blond Ale and nine Indian Pale Ale (IPA) beers from the Uruguayan market were studied using GC–MS. Principal component analysis applied to the data allowed differentiation among the two groups of samples while the volatile compounds and physicochemical parameters responsible for these differences were identified. The physicochemical properties revealed a great diversity between all beer samples even within the same beer style. The main significant differences were ob tained for alcohol, polyphenols, bitterness, colour, and pH. Most Blond Ale beer samples were differentiated from IPA ones by raw fermentation aroma compounds such as 1-pentanol, 1-hexanol, hexanoic and isobutyric acids, 4- vinyl guaiacol, and 5,5-dimethyl-2(5H)-furanone. This is the first work that contributes to the knowledge of Uruguayan craft beers. The study also showed the ability of most of the Uruguayan microbreweries to brew Blond Ale and IPA craft beer styles that meet international standards for physicochemical quality.

A summary of the resource.
Con el objetivo de desarrollar potenciales radiofarmacos de 99mTc para imagenes de zonas de hipoxia, se han preparado varios complejos conteniendo el farmac6foro paranitroaromatico. Dicho farmac6foro en condiciones de hipoxia es capaz de sufrir una reducci6n irreversible, transformandose en grupos citot6xicos como el aminobencilo y aminobenceno, que quedarian atrapados dentro de la celula. Como sistema de uni6n del farmac6foro al centro metalico se emplearon los complejos mixtos "3+1" de Tc(V) y los complejos tricarbonilicos de Tc(I). Los complejos mixtos "3+1" fueron preparados empleando cantidades equimoleculares de un ligando tridentado aminoditi61ico y un coligando ti61ico monodentado conteniendo el farmacoforo nitroaromatico. La estructura de estos complejos es del tipo TcO-[(S-(CH2Hs)-NR-(C2Hs)-S + S-R]. EI estudio estructural de los complejos mixtos, realizado empleando los complejos analogos de Re estable, verific6 que en todos los casos se formaron las estructuras esperadas. Los estudios de voltamperometria indicaron que la reducci6n del grupo nitro y el core monooxo ocurre a potenciales que se pueden alcanzar en medios biol6gicos. EI ensayo de citotoxicidad revel6 la importancia de la presencia del grupo dialquilamina en la estructura del ligando tridentado para la citotoxicidad del complejo final. Los complejos de 99mTc fueron preparados por sustituci6n empleando el 99mTc-glucoheptonato como precursor, obteniendo en todos los casos una pureza radioquimica superior al 90%, con una estabilidad de por 10 menos 6 hs. Los estudios de biodistribuci6n en ratones CD1 normales, realizados entre 5 min. y 24 horas post-inyecci6n, mostraron el perfil esperado para compuestos lipofilicos, incluyendo eliminaci6n mayorotaria por via hepatobiliar. La potencialidad para imagenes oncol6gicas fue evaluada en ratones portadores de sarcomas inducidos entre 30 min. y 24 horas post-adm. La captaci6n fue moderada (1.2 %/g a los 30 min.), pero la relaci6n tumor/musculo (TIM) alcanz6 valores entre 1.5 y 2 en el periodo estudiado. Imagenes centellograticas muestran similares resultados. Los complejos tricarbonilicos de 99mTc fueron obtenidos mediante sustituci6n a partir del precursor fac-99mTc-[(COh(H20hr, preparado por reducci6n del pertecneciato en atm6sfera de CO. Para esta etapa se emplearon dos ligandos, uno de los cuales actuaria como tridentado a traves de un sistema donor del tipo NNO EL segundo ligando actuaria como bidentado dejando una posici6n de coordinaci6n libre, este ultimo tam bien se va a emplear conjuntamente con un coligando monodentado adecuado como el CN, para formar un complejo mixto "2+1" tricarbonilico. Para el caso del ligando tridentado se obtuvo una especie con una pureza radioquimica superior al 90%, la que result6 estable por 10 menos 6 hs. Los estudios quimicos permitieron corroborar la formaci6n de la estructura propuesta. Los estudios "in vitro" revelaron que el complejo presenta muy baja citotoxicidad. La voltamperometria muestra que la reducci6n del grupo nitro ocurre a potenciales alcanzables en los medios biol6gicos. Los estudios de biodistribuci6n en animales normales dieron los resultados esperados para un complejo de lipofilicidad intermedia, presentado una eliminaci6n por via hepatobiliar y urinaria y una rapida depuraci6n de sangre y tejidos blandos. Los estudios en animales portadores de tumores revelaron una muy baja captaci6n (0,2%/g a los 30 min.), pero una relaci6n TIM favorable debido a la rapida eliminaci6n. EI complejo tricarbonilico con el ligando bidentado di6 lugar a la formaci6n de varias especies, las cuales pudieron ser restringidas mediante el agregado de un coligando monodentado adecuado como el CN. Los estudios estructurales con Re estable no permitieron la elucidaci6n de la estructura formada pero los datos obtenidos indican la ausencia del grupo nitro en los complejos finales. Los complejos obtenidos no presentaron propiedades 6ptimas para ser utilizados como radiofarmacos de diagn6stico de hipoxia, una de las posibles formas de continuaci6n de este trabajo seria la de realizar la uni6n del mismo farmac6foro al metal a traves de otros sistemas de marcaci6n. Pero dado los resultados similares obtenidos para las estrategias empleadas parece mas adecuado intentar un cambio de farmac6foro para aumentar la selectividad.

<p>En esta Tesis de Doctorado se trabajó sobre diferentes fotosensibilizadores para su uso en celdas solares sensibilizadas con pigmentos (DSSC). Estos dispositivos son celdas electroquímicas que imitan el proceso natural de fotosíntesis, utilizando pigmentos orgánicos que captan la luz incidente y provocan una separación de cargas, seguida por un flujo de electrones, representando una forma alternativa de generación de energía fotovoltaica a bajo costo.<br />En particular, en este trabajo de investigación se estudiaron tres pigmentos naturales diferentes: las antocianinas provenientes de la flor del ceibo, la proteína ficocianina proveniente del alga Spirulina spp y la proteína ficoeritrina proveniente de algas rojas antárticas.<br />Se llevaron adelante procesos de extracción y purificación de los pigmentos a través de cromatografía de exclusión molecular y fase reversa con columnas C18, así como también su caracterización por espectrofotometría UV-visible, espectroscopía de fluorescencia, voltamperometría cíclica y estudios de estabilidad frente a distintos parámetros. Además, se realizaron medidas en microbalanza de cuarzo para determinar la adsorción de los pigmentos al semiconductor de la celda.<br />Por último, se armaron celdas con los distintos fotosensibilizadores y en diferentes condiciones, evaluando su funcionamiento a través de perfiles J vs E y estudios de espectroscopía de impedancia electroquímica para determinar parámetros fotovoltaicos, utilizando un simulador solar.<br />Destacar que varios de estos estudios fueron posibles gracias a la cooperación con el Instituto Tecnológico de Chascomús (Argentina), el Karlsruher Institute für Technologie (Alemania) y la École Politechnique Fédérale de Lausanne (Suiza).</p>
