Hanseniaspora species can be isolated from grapes and grape musts, but after the initiation of spontaneous fermentation, they are displaced by Saccharomyces cerevisiae. Hanseniaspora vineae is particularly valuable since this species improves the flavour of wines and has an increased capacity to ferment relative to other apiculate yeasts. Genomic, transcriptomic, and metabolomic studies in H. vineae have enhanced our understanding of its potential utility within the wine industry. Here, we compared gene sequences of 12 glycolytic and fermentation pathway enzymes from five sequenced Hanseniaspora species and S. cerevisiae with the corresponding enzymes encoded within the two sequenced H. vineae genomes. Increased levels of protein similarity were observed for enzymes of H. vineae and S. cerevisiae, relative to the remaining Hanseniaspora species. Key di�erences between H. vineae and H. uvarum pyruvate kinase enzymes might explain observed di�erences in fermentative capacity. Further, the presence of eight putative alcohol dehydrogenases, invertase activity, and sulfite tolerance are distinctive characteristics of H. vineae, compared to other Hanseniaspora species. The definition of two clear technological groups within the Hanseniaspora genus is discussed within the slow and fast evolution concept framework previously discovered in these apiculate yeasts.

Owing to their reproducibility, stability, and cost-effective production, the recombinant variable domains of heavy-chain-only antibodies (VHHs) are becoming a salient option as immunoassay reagents. Recently, there have been several reports describing their application to the detection of small molecules (haptens). However, lacking the heavy-light chain interface of conventional antibodies, VHHs are not particularly apt to bind small analytes and failures are not uncommon. Here we describe the construction of a VHH phage display library against the cyanobacterial hepatotoxin microcystin LR and its selection using competitive panning and two novel panning strategies. The outcome of each strategy was evaluated by a large-scale screening using in vivo biotinylated nanobodies. The three methods selected for different nonoverlapping subsets of VHHs, allowing one to optimize the immunodetection of the toxin. The best results were obtained by promoting the isolation of VHHs with the slowest koff (off-rate selection). Among these, the biotinylated nanobody A2.3 performed in ELISA with excellent recovery and high sensitivity, IC50 = 0.28 μg/L, with a limit of detection that is well below the most rigorous guidelines for the toxin. While it may be case-specific, these results highlight the importance of exploring different panning strategies to optimize the selection of antihapten nanobodies.

The estimation of equivalent sweet concentrations of high-intensity sweeteners is one of the first steps for the substitution of sucrose during the development of low-calorie products. The aim of this work was to compare two sensory methodologies (paired comparison and magnitude estimation) and two data analysis approaches (logistic regression and survival analysis) to estimate equivalent sweet concentration of high-intensity sweeteners, using orange/pomegranate juice as case study. Three low-calorie sweeteners were considered: sucralose, stevia and monk fruit extract. For each sweetener, a series of six orange/pomegranate juice samples with different sweetener concentration were prepared. Three groups of 100 consumers participated in the study. Each group was randomly divided into two groups of 50, each of which evaluated samples using one of the two methodologies (paired comparison and magnitude estimation). Paired comparison and magnitude estimation provided similar estimations for the three high-intensity sweeteners which were in agreement with published data, regardless of the statistical approach used for data analysis. However, logistic regression and survival analysis differed in the accuracy of the estimations.

Abstract
A summary of the resource.
Eight chocolate milk desserts with different formulation were evaluated by two groups of consumers. Fifty consumers evaluated the samples and indicated their overall liking and answered a CATA question. Meanwhile, 40 consumers elicited up to four words to describe the desserts and completed a projective mapping task. Projective mapping and the check-all-that-apply question provided very similar sensory profiles for the evaluated milk desserts. Differences in the sensory characteristics of the samples were explained by differences in their formulations, which suggest the validity of the sensory profiles given by consumers. Projective mapping and the CATA question consisted on valuable tools to understand their perception of the sensory and hedonic characteristics of the desserts. These methodologies could consist on useful and interesting complimentary techniques to trained assessors’ data, being CATA question easier to understand and less time consuming for consumers.

Excessive sugar consumption has been identified as the main dietary determinants of obesity among children and adolescents. Sugar sweetened beverages are one of the main sources of added sugar in the diet. Therefore, gradual sugar reduction in these products can contribute to decreasing sugar intake and encourage children to slowly develop preference for products with lower sugar concentration. However, specific recommendations on how to implement gradual sugar reduction programs are still lacking. The aim of the present study was to compare two sugar reduction strategies (stepwise vs. gradual) on children’s sensory and hedonic perception of grape nectar. One hundred and seventeen 6–12 years old children participated in a two-part study: a 9-week study and a final liking test at the end of 11 weeks. In the 9-week study, children were divided in two sugar reduction strategies: stepwise and gradual. Children’s sensory and hedonic perception was evaluated using 9-point hedonic scales and check-all-that-apply questions. There were no major differences between the gradual and stepwise reduction groups in children’s sensory and hedonic perception. However, results showed that the gradual reduction strategy led to smaller changes in children’s sensory and hedonic perception than the stepwise strategy. In the final test, children in the stepwise group gave significantly higher liking scores to the samples than those in the gradual sugar reduction strategy. Comparing the overall liking between the first time the children tasted the grape nectars and the final test, a significant difference was observed for the sample with lower sugar content only for the stepwise group, which was higher in the 11th week. Although differences between the sugar reduction strategies were small, the gradual reduction strategy seems to be recommended over the step-wise strategy.

Temporal Check-All-That-Apply (TCATA) has been recently introduced as a method for temporal sensory product characterization. This method requires assessors to select all the terms they consider applicable at each moment of the evaluation, and to de-select terms when they are no longer applicable. In the present work a variant of TCATA, TCATA Fading, is presented and compared to TCATA. In TCATA Fading selected terms gradually and automatically become unselected over a predefined period of time. Eight studies were conducted with different product categories in three different countries. In Studies 1–3, TCATA and TCATA Fading were compared with trained assessors using within-subject experimental designs on the same set of products. In Studies 4–6, TCATA and TCATA Fading were compared with consumers following between-subject experimental designs. Comparison was performed in terms of citation proportion of the terms, significant differences among samples detected, dynamic sample profiles, and task perceptions. Across the eight studies, results suggested that automatic de-selection of attributes in a TCATA task can improve discrimination and provide a more accurate description of the dynamics of sensory characteristics of products than asking consumers to de-select attributes when they are no longer applicable. The present research represents a first attempt at implementing and evaluating TCATA Fading, and suggests that it is a useful variant of the TCATA method. Many avenues for methodological refinement are identified.

<p>Product-focused emotion research has been in ascendency for more than a decade, but methodological research into questionnaires, the most popular tool for measuring emotional associations is still needed. The present research was situated in this context and pertained to the valence × arousal circumplex-inspired emotion questionnaire (CEQ) which includes 12 pairs of emotion words. Variants of the CEQ that pertained to response options (single vs. multiple) and layout (circular vs. list) were compared in four consumer studies with a total of 1857 participants. In Studies 1 to 3, the CEQ variant that featured a circular layout and single response format<br />(C&S) was compared to a list variant with multiple response format (L&M) (i.e., a check-all-that-apply question).<br />Selection of CEQ emotion word pairs and degree of sample discrimination was contingent on CEQ variant, meaning that conclusions about sample similarities and differences depended on the CEQ variant that was used.<br />This was also the result in Study 4 where two circular CEQ variants were compared, with single and multiple response formats, respectively. A further important finding was that stimuli format – tasted samples in Studies 1–2 and written stimuli in Studies 3–4 – as well as degree of sample differences influenced the extent to which the results from the CEQ variants differed. Our recommendations at this stage are to be aware that conclusions about sample profiles and discrimination is CEQ variant dependent and avoid directly comparing results from studies that use different CEQ variants. Implementing the CEQ as a multi-response variant is supported since it improves<br />sample discrimination relative to single response data, particularly for pairs of emotion words that primarily<br />relate to the dimension of arousal.</p>

Nitrite ion competition has been measured for the base hydrolysis reaction of rans-Co(N-H3)4(NH2CH3)Xn+ ions (X = Cl−, Br−, NO3− and SO42−) in 1.0 M NaNO2 at 25°C. Both O- and N- bonded Co(NH3)4(NH2CH3)NO22+ are formed. Subsequently the Co(NH3)4(NH2CH3)ONO2+ isomer rearranges in OH− to give the thermodynamically more stable Co(NH3)4(NH2CH3)NO22+ ion. The total NO2− captured shows a slight dependence on the overall charge of the complex and on the nature of X (R = 2.2, 2.1, 3.6 and 1.2 ± 0.5% for X = Cl−, Br−, NO3−, NO3−, and SO42−, respectively). These results differ from those observed with complexes of the type Co(NH3)5Xn+ and Co(NH2CH3)5Xn+ where X includes a variety of anions.

Single-domain antibodies (sdAbs) found in camelids lack a light chain, and their antigen-binding site sits completely in the heavy-chain variable domain (VHH). Their simplicity, thermostability, and ease in expression have made VHHs highly attractive. Although this has been successfully exploited for macromolecular antigens, their application to the detection of small molecules is still limited to a very few reports, mostly describing low-affinity VHHs. Using triclocarban (TCC) as a model hapten, we found that conventional antibodies, IgG1 fraction, reacted with free TCC with a higher relative affinity (IC50 51.0 ng/mL) than did the sdAbs (IgG2 and IgG3, 497 and 370 ng/mL, respectively). A VHH library was prepared, and by elution of phage with limiting concentrations of TCC and competitive selection of binders, we were able to isolate high-affinity clones, KD 0.98�1.37 nM (SPR), which allowed development of a competitive assay for TCC with an IC50 = 3.5 ng/mL (11 nM). This represents a 100-fold improvement with regard to the performance of the sdAb serum fraction, and it is 100-fold better than the IC50 attained with other antihapten VHHs reported thus far. Despite the modest overall antihapten sdAbs response in llamas, a small subpopulation of high-affinity VHHs is generated that can be isolated by careful design of the selection process

Los aceites de pescado son la principal fuente de EPA y DHA en la dieta. Estos acidos grasos de la famnia w3 tienen efectos importantes sobre el metabolismo y se recomienda aumentar su ingesta. Se estudiaron las fuentes de EPA y DHA disponibles para el consumidor: filetes de pescado y aceite de pescado encapsulado. Los pescados habitual mente consumidos en Montevideo son de especies consideradas magras (clasificaci6n correspondiente al mes de julio), y se requieren entre 100 y 400 gramos de filetes crudos para consumir 1 .gramo de EPA + DHA. Tambien se estudi6 la composici6n de aceites de pescado comerciales y de tipo comestible. Una alternativa a la inclusi6n de pescado en la dieta son los concentrados de w3. Para lograr un alto % de estos acidos grasos en el preparado, es necesario romper el enlace ester de los trigliceridos y fraccionar entonces 10s acidos grasos libres 0 sus derivados. El metodo elegido para esto fue la formaci6n de aductos de urea. La caracteristica de los compuestos de inclusi6n con urea es que s610 aquellos compuestos que encajan en ellumen de los cristales de urea pueden formarlos. Los polienos de aceites de pescado, al tener 5 6 6 insaturaciones de configuraci6n cis e interrumpidas por un metileno, tienen una mayor dificultad para formar los aductos que los acidos grasos saturados. Se estudi6 la reacci6n de complejaci6n con urea como forma de preparar concentrados de w3. La temperatura de reacci6n y la cantidad de solvente utilizada no son factores tan determinantes como la relaci6n urea:acidos graso (peso:peso de acidos grasos complejeables) para lograr un alto % de EPA + DHA en la FNCU. Variando las condiciones de reacci6n se logra recuperar selectivamente el DHA. El interes en los acidos grasos w3 ha dado lugar a la revisi6n de las tecnicas de anaIisis de composici6n de aceites de pescado. Se optimiz6 el uso del equipo cromatografico disponible. Un aspecto importante de la quimica de los aceites de pescado para uso alimentarlo es la prevenci6n de su deterioro oxidativo. Se estudi6 la estabilidad oxidativa del aceite de Higado de Merluza por medida del periodo de inducci6n en un equipo Rancimat. Se estabiliz6 el aceite con varios antioxidantes, siendo el mas eficiente el TBHQ a 200 ppm y a 100 ppm.

A summary of the resource.
Hydatid disease, which affects humans and various livestock species, is caused by the larval stages of the cestode parasite Echinococcus granulosus. The disease involves the growth of a fluid-filled cyst in the host's internal organs. The hydatid cyst wall is generally thought to preclude the access of host immune cells to the cyst, while allowing passage of macromolecules. Thus, evasion of humoral effector mechanisms, in particular the complement system, may be crucial to parasite survival. Evasion of complement might involve inhibition of the C3b deposition step, which can in principle be achieved through parasite-derived or host-derived and parasite-bound regulatory molecules. In this work, extracts from bovine hydatid cysts were analyzed for the presence of factor H-like function, namely the cofactor activity for the breakdown of amidated bovine C3 by bovine factor I. Both this activity and antigenically detectable bovine factor H were found in most extracts. The cofactor activity was retained by an anti-bovine factor H Ab column; as no factor H-cross-reactive parasite molecules were detected in the active extracts, the activity was indeed due to host factor H. The pattern of extraction of host factor H from the parasite tissue indicated that at least part of it was selectively bound to the cyst wall, and that a strong charge component was involved in the binding. Our results indicate that the selective concentration of host factor H from plasma/interstitial fluid may contribute to complement evasion by E. granulosus.

A summary of the resource.
Ralstonia solanacearum is the causative agent of bacterial wilt of potato. Ralstonia solanacearum strain UY031 belongs to the American phylotype IIB, sequevar 1, also classified as race 3 biovar 2. Here we report the completely sequenced genome of this strain, the first complete genome for phylotype IIB, sequevar 1, and the fourth for the R. solanacearum species complex. In addition to standard genome annotation, we have carried out a curated annotation of type III effector genes, an important pathogenicity-related class of genes for this organism. We identified 60 effector genes, and observed that this effector repertoire is distinct when compared to those from other phylotype IIB strains. Eleven of the effectors appear to be nonfunctional due to disruptive mutations. We also report a methylome analysis of this genome, the first for a R. solanacearum strain. This analysis helped us note the presence of a toxin gene within a region of probable phage origin, raising the hypothesis that this gene may play a role in this strain’s virulence

Helminths use an alternative mitochondrial electron transport chain (ETC) under hypoxic conditions, such as those found in the gastrointestinal tract. In this alternative ETC, fumarate is the final electron acceptor and rhodoquinone (RQ) serves as an electron carrier. RQ receives electrons from reduced nicotinamide adenine dinucleotide through complex I and donates electrons to fumarate through complex II. In this latter reaction, complex II functions in the opposite direction to the conventional ETC (i.e., as fumarate reductase instead of succinate dehydrogenase). Studies in Ascaris suum indicate that this is possible due to changes in complex II, involving alternative succinate dehydrogenase (SDH) subunits SDHA and SDHD, derived from duplicated genes. We analyzed helminth genomes and found that distinct lineages have different gene duplications of complex II subunits (SDHA, SDHB, SDHC, and SDHD). Similarly, we found lineage-specific duplications in genes encoding complex I subunits that interact with quinones (NDUF2 and NDUF7). The phylogenetic analysis of ETC subunits revealed a complex history with independent evolutionary events involving gene duplications and losses. Our results indicated that there is not a common evolutionary event related to ETC subunit genes linked to RQ. The free-living nematode Caenorhabditis elegans uses RQ and has two genes encoding SDHA (sdha-1 and sdha-2) and two genes encoding NDUF2 (nduf2-1 and nduf2-2). sdha-1 and nduf2-1 are essential genes and have a similar expression pattern during C. elegans lifecycle. Using knockout strains, we found that sdha-2 and nduf2-2 are not essential, even in hypoxia. Yet, sdha-2 and nduf2-2 expression is increased in the early embryo and in dauer larvae, stages where there is low oxygen tension. Strikingly, sdha-1 and sdha-2 as well as nduf2-1 and nduf2-2 showed inverted expression profiles during the C. elegans life cycle. Finally, we found that sdha-2 and nduf2-2 knockout mutant strain progeny is affected. Our results indicate that different complex I and II subunit gene duplications provide increased fitness to worms.
