Las floraciones de cianobacterias en cuerpos de agua dulce constituyen un problema mundial, asociado a la eutrofización (enriquecimiento de nutrientes) y cambios hidrológicos de los ecosistemas. En este estudio se generó una base de datos históricos (n= 3061 para 64 ecosistemas, de 1980 a 2014), elaborada por un grupo de trabajo interinstitucional (OSE, DINAMA, IM, Universidad de la República e IIBCE), y se analizó la distribución de cianobacterias planctónicas y de cianotoxinas (microcistina, saxitoxina y cilindrospermopsina) en Uruguay. Los valores de clorofila a y nutrientes totales indicaron procesos de eutrofización en diversos ecosistemas. En dos mapas georreferenciados se visualiza la distribución de cianobacterias en el país, según indicadores cuantitativos globales, y de la microcistina (cianotoxina más frecuente). En estos mapas se destacan los embalses (Río Uruguay y Río Negro) y las playas del Río de la Plata como las zonas de mayor riesgo de exposición a cianobacterias según las categorías de la Organización Mundial de la Salud para aguas recreacionales. Además, se analizaron muestras de floraciones (espuma) y se cuantificó 20 mg L-1 de microcistinas totales y la presencia de genes que sugieren diferentes variedades de microcistinas. We used a density functional that incorporates van der Waals interactions to study hydrogen adsorption onto Pt atoms attached to carbon-vacancies on graphene layers, considering molecular and dissociated hydrogen-platinum coordination structures. PtH2 complexes adsorbed on several sites of pristine graphene were also studied for comparison. Our results indicate that both a Kubas-type dihydrogen complex and a classic hydride without Hsingle bondH bond are the preferential PtH2 systems on the vacancy site of graphene. In contrast, the Kubas complex is unstable onto pristine graphene and the hydride is obtained at all adsorption sites. Our simulations suggest that the C-vacancy decreases the reactivity of the metal decoration, allowing a non-dissociative hydrogen adsorption. The H2 molecule is oriented almost perpendicular to the outermost Csingle bondPt bond, interacting also with the graphene surface through σ-H and π-C states. This stabilization of the Kubas-type complex could play a very important role for hydrogen storage in Pt-decorated carbon adsorbents with vacancies. Sturgeon aquaculture faces challenges due to global warming, as chronic heat stress (CHS) jeopardises fish health and production sustainability. To tackle this issue, we analysed serum innate components and the cellular composition of the peritoneum in Russian sturgeons (Acipenser gueldenstaedtii) subjected to CHS and challenged with Aeromonas hydrophila. Russian sturgeons exposed to CHS exhibited a reduction in the serum levels of total protein and alkaline phosphatase activity, trending to a higher decrease in sturgeons exposed to both bacterial challenge and CHS. This sturgeon group showed a worsened health status, reduced growth and a higher mortality rate. Under tolerable temperatures, the bacterial challenge increased serum ceruloplasmin, lysozyme and serum amyloid A levels, which tended to be lower under CHS. The naive peritoneal cellular composition of the Russian sturgeon revealed a predominance of myeloid cells, with lymphoid cells present in a smaller proportion. CHS altered this distribution, increasing lymphoid while reducing myeloid cell percentages and A. hydrophila challenge reverted these alterations. Myeloid cells exhibiting the highest phagocytic potential were reduced by CHS, potentially compromising peritoneal bacterial clearance. Principal component analysis revealed that sturgeons’ integral innate response varied according to temperature exposure, evidencing CHS’s impact on the antibacterial response. Overall, this study provides a first picture of the peritoneal cellular composition of a chondrostean fish. Furthermore, it demonstrates the disruption caused by CHS on the sturgeon’s early systemic and peritoneal responses mounted against A. hydrophila. For the first time a critical analysis of the influence that four different graphene oxide reduction methods have on the electrochemical properties of the resulting reduced graphene oxides (RGOs) is reported. Starting from the same graphene oxide, chemical (CRGO), hydrothermal (hTRGO), electrochemical (ERGO), and thermal (TRGO) reduced graphene oxide were produced. The materials were fully characterized and the topography and electroactivity of the resulting glassy carbon modified electrodes were also evaluated. An oligonucleotide molecule was used as a model of DNA electrochemical biosensing. The results allow for the conclusion that TRGO produced the RGOs with the best electrochemical performance for oligonucleotide electroanalysis. A clear shift in the guanine oxidation peak potential to lower values (~0.100 V) and an almost two-fold increase in the current intensity were observed compared with the other RGOs. Switchgrass (Panicum virgatum) is a perennial grass highly valued as an energy crop resource for the production of bioethanol due to its high carbohydrate content, fast growth, and ability to grow in lands that cannot support crop or food production. In the present study, this biomass was submitted to steam explosion pretreatment in a semi-continuous pre-pilot reactor with the aim of obtaining a pretreated solid with high digestibility for enzymatic hydrolysis. Different conditions of temperature (170–200 °C) and residence time (5–15 min), leading to different severity factors (2.76–4.12) were used for steam explosion pretreatment, which were combined through a 22 central composite design. The results revealed that both variables had great influence in the process, affecting both the biomass structure and the saccharification yield, as a consequence. However, in the range of values evaluated in this study, the effect of the temperature was more prominent than the effect of the residence time. The best saccharification yield (88.3%) was obtained when using the biomass pretreated at 200 °C for 10 min. Similar result was obtained using a commercial cellulose pulp as feedstock for enzymatic hydrolysis, confirming that the best conditions for switchgrass pretreatment in the pre-pilot scale were successfully established. In Uruguay wood-impregnation plants use chromated copper arsenate (CCA) as preservative applying good manufacture practices (GMP). This study aims a retrospective evaluation of toxicologically relevant species levels in CCA exposed woodworker's urine (As-U) and an assessment of the effects of work risk factors and non-occupational sources in As-U of workers from a selected plant. From 2014 to 2016, As-U in 212 urine samples (As-U) of 73 woodworkers from six CCA impregnation plants were determined. In one of these plants, 35 workers were interviewed to obtain individual data of work tasks, lifestyles, diet, habits, etc. that may contribute to their overall exposure to Arsenic. Responses were statistically evaluated. Out of the 212 urine samples from 73 woodworkers, 96% showed lower levels of As-U than those established by health regulations (<35μgL-1). According to their work tasks 34% of 35 surveyed workers showed high exposure risk to As and 29% moderate exposure risk. Although they have lower levels of As-U owing to their personal protective equipment, As-U significantly correlate to work risk factors. Consumption of bottled water could also contribute to As-U levels as a non-occupational source. These results confirm that efforts of Uruguayan authorities to promote GMP were successful and justify the importance and frequency of As-U systematic biomonitoring for occupational risk assessment. A significant accomplishment of this work is that non-occupational sources of As-like bottled water consumption should also be considered in future studies. Humans are exposed to arsenic (As) in the environment, in both organic and inorganic forms, and it has been widely demonstrated that the inorganic arsenic species (arsenate and arsenite) are the main toxic ones being drinking water one of the main sources of exposure worldwide. Urinary arsenic level is the recommended biomarker for assessing human exposure risks. Absorbed inorganic arsenic is metabolized to mono- and dimethylated arsenic compounds (MMA and DMA) prior to its excretion in urine. In this work, a simple procedure for the determination of toxicologically relevant arsenic species in urine (sum of As(III), As(V), MMA and DMA) using Hydride Generation Atomic Absorption Spectrometry (HG-AAS) was developed in order to obtain a feasible and self-sustainable technology for analysis and epidemiological studies when other expensive techniques are not available. Sample preparation was based on the derivation of arsenic species with L-cysteine. The limit of detection was 1.5 μg L-1, linearity up to 50 μg L-1 and analytical precision was 12 % (relative standard deviation, RSD %, n=10). Trueness was evaluated using spiked urine samples as well as a reference material. The range of acceptation for recoveries was established in 85-115% by means of a t-test at the 95% significance level. Recoveries for the four As species from spiked urine samples were in the range 87.8-113.5 %. The recovery from a reference material was 100.7%. The comparison between the HG-AAS and high performance liquid chromatography-hydride generation-coupled to inductively coupled plasma mass spectrometry (HPLC-HG-ICPMS), considered as a gold standard technique, showed good agreement (R2 = 0.94) for forty urine samples. El Arsénico (As) es un elemento natural que puede estar presente en un amplio rango de concentraciones en el aire, agua, suelos, vegetales y animales, en distintos estados de oxidación (–III, 0, III, V) y en formas inorgánicas y orgánicas. Es un reconocido agente tóxico para la salud humana que se encuentra en el primer lugar de la lista de sustancias peligrosas elaborada por la Agencia para Sustancias Tóxicas y Registro de Enfermedades (ATSDR). Se ha demostrado que no todos los compuestos de As presentan el mismo grado de toxicidad, dado que depende de su forma química o especiación, siendo la toxicidad mucho mayor de las especies inorgánicas que de las especies orgánicas: As (-III) > As (III) > As (V) > MMA > DMA >> Arsenobetaína. Su presencia en el ambiente puede provenir tanto de fuentes naturales como de actividades antropogénicas. El As se encuentra naturalmente en aguas subterráneas en diferentes puntos del planeta. Su presencia en aguas destinadas al abastecimiento de la población representa una especial preocupación de salud ambiental a nivel internacional. La exposición humana a largo plazo a bajos niveles de As, puede conducir a la intoxicación crónica produciendo lesiones cutáneas y cáncer de piel que son los efectos más característicos, pudiendo desarrollar también cáncer de vejiga, riñón y pulmón. En estos momentos, el límite máximo recomendado por la Organización Mundial de la Salud (OMS) para la concentración de As en el agua potable es de 10 μg L-1. Entre las actividades antropogénicas que contribuyen a la liberación de As al ambiente se encuentran entre otros, la minería, la fundición de metales y el uso de As en fórmulas de pesticidas y conservantes de la madera. En Uruguay existen varias plantas que impregnan madera con el plaguicida denominado CCA que contiene óxidos de cromo, cobre y arsénico, por lo que existen trabajadores expuestos al As. En nuestro país es obligatorio el control biológico (biomonitoreo) de trabajadores expuestos a As, mediante el análisis de las denominadas “especies toxicológicamente relevantes” de As en orina (As(III) + As(V) + DMA (ácido dimetilarsínico) + MMA (ácido monometilarsónico). El límite establecido para la suma de estas especies en orina (As-U) es de 35 μg L-1. El objetivo general planteado para esta tesis, consistió en desarrollar herramientas analíticas viables para Uruguay, con el objetivo de evaluar las especies de arsénico en matrices ambientales y biológicas, para aplicarlas a estudios poblacionales, como alternativas a la técnica de referencia internacional (HPLC-ICP-MS), no disponible en el país y altamente costosa para estudios de rutina. En cuanto a matrices ambientales, se realizó la validación de una metodología no acoplada para determinar As (III) + As (V) en agua mediante espectrometría atómica con generación de hidruros (HG-AAS). Los resultados obtenidos para algunas muestras de agua indican que este método es adecuado para el control de las mismas, teniendo en cuenta el valor máximo recomendado por la OMS. Respecto a matrices biológicas se optimizaron y validaron dos metodologías analíticas no 11 acopladas. Una fue mediante HG-AAS, para para el biomonitoreo de especies toxicológicamente relevantes de As en orina (As-U) destinado a la evaluación del riesgo a la salud de los trabajadores expuestos a As a fin de prevenir efectos adversos o intervenciones médicas. Los resultados obtenidos muestran que este método es adecuado para la determinación de las especies: As(III) + As(V) + DMA + MMA. Otra metodología validada fue para la determinación de As total en muestras de orina mediante ET-AAS, la cual cuantifica la suma de las especies toxicológicamente relevantes y la especie orgánica arsenobetaína como prueba de la incidencia de las especies orgánicas, poco tóxicas de As, derivadas de la dieta. Por otra parte, se han optimizado las condiciones de un acople HPLC-HG-AAS utilizando una interfase construida en el laboratorio de la Facultad de Química. En cuanto a los estudios poblacionales, en primer lugar, el desarrollo analítico realizado en el transcurso de esta tesis fue aplicado a controles de biomonitoreo de As-U en trabajadores expuestos a CCA; se revisaron y evaluaron los resultados obtenidos sobre 212 muestras de 73 trabajadores durante tres años y los resultados mostraron niveles de bajo riesgo de exposición ocupacional al As. En segundo lugar, se realizó un estudio piloto en una planta de impregnación de madera con CCA en el cual paralelamente a la evaluación de los niveles de As-U, se realizó una encuesta a los trabajadores para analizar la contribución de aspectos ocupacionales y no ocupacionales que pudieran incidir en los niveles de As-U determinados, utilizando métodos estadísticos. Los resultados de As-U de los trabajadores se encontraron todos dentro de los valores de la normativa vigente. Sin embargo, se constató que a mayor exposición durante las horas de trabajo, se observan niveles más altos de As-U de los trabajadores y que también existe correlación estadística de As-U con la percepción de los trabajadores de estar expuestos al ruido y con el consumo de agua embotellada. Con respecto a los resultados de As total en orina obtenidos en estos trabajadores, se comprobó la influencia de la ingesta reciente de pescado o frutos del mar, en los niveles de arsénico total en orina por el aporte “dietario” de AsB de muy baja toxicidad. Este resultado se obtuvo en 8 de los 35 trabajadores coincidiendo con su declaración de haber consumido pescado o frutos de mar en la encuesta. Por lo anterior, resulta fundamental la indicación de las “especies toxicológicamente relevantes” para la evaluación del riesgo de exposición a arsénico como lo recomienda la ACGIH y el MSP de acuerdo a sus valores límites. Hasta el alcance de nuestro conocimiento, esta es la primera revisión de resultados de biomonitoreo de especies toxicológicamente relevantes en trabajadores expuestos a CCA realizada en el Uruguay. Esta investigación marca un antecedente importante de investigación y desarrollo para futuros estudios científicos interdisciplinarios de índole epidemiológico, destinados a evaluar por ejemplo, la susceptibilidad de la población general expuesta a arsénico a través del agua y alimentos y correlacionar los resultados de As-U con otros biomarcadores de efecto y susceptibilidad para establecer la situación de riesgo presente en la población uruguaya en torno a la exposición al As.
