The quinazoline derivatives (3-chloro-4-fluorophenyl)quinazoline-4,6-diamine (2) and (3-bromophenyl)quinazoline-4,6-diamine (3) were labelled with 99mTc using the “4 + 1” mixed-ligand system [Tc(NS3)(CN-R)] and the tricarbonyl moiety fac-[Tc(CO)3]+. In the “4 + 1” approach the technetium(III) is stabilized by a monodentate isocyanide bearing a quinazoline fragment (L11, L22) and by the tetradentate tripodal ligand tris(2-mercaptoethyl)-amine (NS3). In the “4 + 1” approach, 99mTc-labelling was performed in a two-step procedure, the complexes [Tc(NS3)(L11)] (7a) and [Tc(NS3)(L22)] (8a) being obtained in about 50–70% yield. In the tricarbonyl approach, the fac-[Tc(CO)3]+ unit is anchored by two different monoanionic chelators bearing the quinazoline derivatives (3-chloro-4-fluorophenyl)quinazoline-4,6-diamine (2) and (3-bromophenyl)quinazoline-4,6-diamine (3). Both chelators have a N2O donor atom set, but one contains a pyrazolyl ring (L55H) and the other contains a pyridine unit (L66H). In both cases the conjugation of the quinazoline to the chelator was done through the secondary amine of the potentially tridentate and monoanionic chelators, the corresponding 99mTc-complexes (10a, 11a) being obtained in quantitative yield. The identities of the 99mTc-labelled quinazolines (7a, 8a, 10a, 11a) were confirmed by comparison with the HPLC profiles of the analogous Re compounds (7, 8, 10, 11). All these Re complexes were characterized by NMR and IR spectroscopy, elemental analysis and in some cases by MS and X-ray diffraction analysis. In vitro studies indicate that the quinazoline fragments, after conjugation to the cyano group (L11, L22) or to the pyrazolyl containing chelator (L55H), as well as the corresponding Re complexes (7, 8, 10) inhibit significantly the EGFR autophosphorylation and also inhibit A431 cell growth. These two effects were also found for the pyridine-containing chelator (L66H) and corresponding Re complex (11), although to a lesser extent.

Selective reduction of native disulfide bonds in immunoglobulins is one of the best methods for introducing reactive groups on to the protein surface. Additionally, the thiol groups so generated may allow oriented conjugation at a specific site of the immunoglobulin. Solid-phase reducing agents have many advantages over soluble ones (including ease of separation of excess reagent from reduced protein by filtration, and the potential for regeneration and multiple reuse). In this work we report a comparative study of the reduction of rabbit IgG and its F(ab′)2 fragments, with mercaptohydroxypropylether-agarose (thiopropyl-agarose), a solid phase reducing agent, and dithiothreitol. The effect of different parameters on the process, such as the amount of reducing agent, incubation period, and temperature, was assessed by titration of thiol groups and SDS-PAGE analysis. Optimized reduction with thiopropyl-agarose introduced six thiol groups in the F(ab′)2 fragment (mol/mol). Native IgG was less reactive, probably due to steric effects, as only an average of three thiol groups were introduced. However, by increasing reaction temperature from 22 to 37°C, six thiol groups could be introduced in native IgG (mol/mol). Reduction with dithiothreitol also introduced six thiol groups in F(ab′)2 fragments (mol/mol) but led to higher thiol content for the whole IgG. These results demonstrated that thiopropyl-agarose can be a very useful tool for exercising more precise control over the reduction treatment, and for selecting which disulfide bridges are to be broken. After 6 h incubation with reducing agent containing 8 and 16 μmoles SH per mg of protein, the resulting reduced IgG retained the same biological activity as the native immunoglobulin. The controlled modification of native disulfides achieved with thiopropyl-agarose will be useful for the development of soluble and insoluble immunoglobulin conjugates.

Nitroalkenes derivatives of free as well as esterified unsaturated fatty acids are present in human plasma and tissue, representing novel pluripotent cell signaling mediators. Lipid nitration occurs in response to pro-inflammatory stimuli as an adaptative mechanism to downregulate inflammatory responses. This chapter first discusses the generation of nitroalkenes during macrophage activation following chemical and biological characterization. In particular, it describes procedures for (a) synthesizing and characterizing esterified (cholesteryl-nitrolinoleate, CLNO2) as well as free (nitroarachidonate, AANO2) nitroalkenes, (b) determining nitration of cholesteryl linoleic acid during macrophage activation by inflammatory stimuli, (c) examining the modulatory effects of nitroalkenes on the expression of inducible enzymes by activated macrophages, and (d) discussing the signaling pathways involved in nitroalkene-mediated anti-inflammatory actions.

We present a detailed conformational study of a simplified synthetic analog of the bis-oxazole oxane fragment found in the cytostatic agents phorboxazole A and B based on results from NMR spectroscopy and molecular modeling simulations. Complete 1H and 13C resonance assignments for the bis-oxazole oxane system were carried out through the use of COSY, HSQC, HMBC, TOCSY, and HSQC-TOCSY experiments, and its conformational preferences in solution were investigated by analysis of 3JHH coupling constants and NOE enhancements obtained from 1D and 2D NOESY experiments. In order to solve inconsistencies from our preliminary structural studies, simulated annealing studies were performed to thoroughly sample the phase space available to the molecule. Our results reveal that the six-membered oxane ring, which constitutes the most important moiety regarding the three-dimensional (3D) structure and flexibility of the analog, exists in rapid equilibrium between its two accessible chair conformers in an approximate ratio of 70:30. The information gathered from these studies will be of critical importance in our efforts to prepare novel compounds with phorboxazole-like structure and activity. This work exploits the combination of the lectin affinity chromatography (LAC) with an ultra-sensitive immunochromatographic assay to differentiate several types of erythropoietin (EPO). The chromatographic behaviours of different commercial types of recombinant human EPO (rhEPO), EPO analogues (Aranesp) and urine human EPO (uhEPO) from healthy individuals on eight lectin-Sepharose columns, have been worked out. Results show that when using wheat germ agglutinin (WGA)-Sepharose columns, a careful desorption regime starting with very low concentration (2 mM) of the competitive sugar N-acetylglucosamine (GlcNAc) makes it possible to efficiently distinguish endogenous EPO from recombinant EPO and EPO analogues.

A summary of the resource.
The objective was to test immune protection against the formation of Toxoplasma gondii tissue cysts in rats. It has been previously shown that 50 T. gondii tissue cysts of strain Me49 are not pathogenic for CF-1 mice, whereas 1 T. gondii tissue cyst of strain M-7741, can be lethal for mice 11–13 days after subcutaneous or oral administration. In the present study, ten rats were fed T. gondii oocysts of strain Me49 and after a further 30 days they were each orally challenged with T. gondii oocysts of strain M-7741. Thirty days after this, they were euthanased and brain and muscle samples inoculated subcutaneously or orally dosed, respectively, to mice for bioassay. None of the mice died, whereas all the mice that were inoculated with brain homogenates or were fed muscle samples from four non-immunized rats that had been inoculated with T. gondii oocysts of strain M-7741, died. These results encourage further research towards achieving vaccinal protection against the formation of T. gondii tissue cysts in meat animals and people.

Essential oils of Baccharis uncinella DC and B. dracunculifolia DC collected from different wild populations growing in Bolivia, southern Brazil and Uruguay were analysed using gas chromatography and gas chromatography–mass spectrometry techniques, to check for chemical variability. Cluster analysis led to the identification of two chemotypes for B. dracunculifolia, related to aromadendrane- or cadinane-derived components. As the timing of sampling of the plant material may have an influence on the composition of the essential oil isolated, the seasonal influence at the moment of harvesting was studied for two populations of each species. The results suggested that the flowering season was optimal both species and this supported the practice of harvesting at the full bloom stage. Copyright © 2008 John Wiley & Sons, Ltd.

A summary of the resource.
In this work we report a conformational study of [ReVO2(pn)]2+ (pn = 1,2-propylenediamine) in aqueous solution using experimental and theoretical approaches. The studies in aqueous solution were carried out by 1H and 13C NMR spectroscopy and applying the Altona’s equation. Theoretically, the studies were performed by means of the Density Functional Theory (DFT) and the Gauge-Including Atomic Orbitals (GIAO) method. All 12 spatial dispositions arising from the relative orientation of the CH3 substituents were studied. The results are consistent with the predominance of complexes with λλ ligand conformation. The computation analysis shows that only the λλ conformers exhibit a 1H-signal pattern in agreement with the experimental evidence. We proposed that only two main species can predominate in an aqueous solution of {ReVO2[(R)-pn]2}+. They should have a λλ conformation of the (R)-pn ligands and the energy difference (PCM (Polarizable Continuum Model) methodology) between them is only 1.5 kJ mol−1.

Metazoan parasites of mammals (helminths) belong to highly divergent animal groups and yet induce a stereotypical host response: Th2-type immunity. It has long been debated whether this response benefits the host or the parasite. We review the current literature and suggest that Th2 immunity is an evolutionarily appropriate response to metazoan invaders both in terms of controlling parasites and repairing the damage they inflict. However, successful parasites induce regulatory responses, which become superimposed with, and control, Th2 responses. Beyond helminth infection, this superimposition of response profiles may be the norm: both Th1 and Th2 responses coexist with regulatory responses or, on the contrary, with the inflammatory Th17 responses. Thus, typical responses to helminth infections may differ from Th2- dominated allergic reactions in featuring not only a stronger regulatory component but also a weaker Th17 component. The similarity of immune response profiles to phylogenetically distinct helminths probably arises from mammalian evolution having hard-wired diverse worm molecules, plus tissue-damage signals, to the beneficial Th2 response, and from the convergent evolution of different helminths to elicit regulatory responses. We speculate that initiation of both Th2 and regulatory responses involves combinatorial signaling, whereby TLR-mediated signals are modulated by signals from other innate receptors, including lectins.

Heterometallic compounds containing La(III), bivalent cationsM(M = Co, Ni, Ca), and 2,20-oxydiacetate (oda) as connecting ligand have been prepared and characterized. The complexes can be formulated as [La2M3(oda)6(H2O)6] Æ 12H2O. The structure of [La2Co3(oda) 6(H2O)6] shows the presence of the La(III) coordinated by six carboxy and three ether oxygen atoms, and the Co(II) cation bonded to four carboxy oxygens and two molecules of water. An open 3D framework is observed, containing large hexagonal channels. The chemical systems were also investigated in solution (25.0 �C, I = 0.5 M Me4NCl) by potentiometry. The same kind of polynuclear species have been found in aqueous solution.

A new complex, [Cu2(sulfameter)4]3 Æ 2.5H2O (sulfameter = 4-amino-N-(5-methoxy-2-pyrimidinyl)benzenesulfonamide), has been synthesized. Its structure has been determined by single-crystal X-ray diffraction and its spectroscopic properties (EPR, IR, Raman, UV–Vis) have been analyzed. The structure presented three different dimeric units in the unit cell and the EPR spectra, characteristic of antiferromagnetically coupled dimers, revealed two magnetically different dimeric environments

A summary of the resource.
The aroma profile of five premium red wines has been studied by sensory descriptive analysis, quantitative gas chromatography-olfactometry (GC-O), and chemical quantitative analysis. The most relevant findings have been confirmed by sensory analysis. Forty-five odorants, including the most intense, were identified. At least 37 odorants can be found at concentrations above their odor threshold. A satisfactory agreement between GC-O and quantitative data was obtained in most cases. Isobutyl- 2-methoxypyrazine, (E)-whiskey lactone, and guaiacol were responsible for the veggie, woody, and toasted characters of the wines, respectively. The sweet-caramel notes are related to the presence of at least five compounds with flowery and sweet notes. The phenolic character can be similarly related to the presence of 12 volatile phenols. The berry fruit note of these wines is related to the additive effect of nine fruity esters. Ethanol exerts a strong suppression effect on fruitiness, whereas norisoprenoids and dimethyl sulfide enhance fruity notes.

The structure, magnetism, transport and thermal expansion of the perovskite oxide LaNi0.5Fe0.5O3 were studied over a wide range of temperatures. Neutron time-of-flight data have shown that this compound undergoes a first-order phase transition between ∼275 and ∼310 K. The structure transforms from orthorhombic (Pbnm) at low temperatures to rhombohedral (R3¯c) above room temperature. This phase transition is the cause for the previously observed co-existence of phases at room temperature. The main structural modification associated with the phase transition is the change of tilting pattern of the octahedra from a+b−b− at low temperatures to a−a−a− at higher. Magnetic data strongly suggests that a spin-glass magnetic state exists in the sample below 83 K consistent with the absence of magnetic ordering peaks in the neutron data collected at 30 K. At high temperatures the sample behaves as a small polaron electronic conductor with two regions of slightly different activation energies of 0.07 and 0.05 eV above and below 553 K, respectively. The dilatometric data show an average thermal expansion coefficient of 14.7×10−6 K−1 which makes this material compatible with frequently used electrolytes in solid oxide fuel cells.

A summary of the resource.
New heteroallyl-containing 5-nitrofuranes were synthesized as potential anti-Trypanosoma cruzi agents with a dual mechanism of action, oxidative stress and inhibition of membrane sterol biosynthesis. Some of the derivatives were found to have high and selective activity against the proliferative stages of the parasite, with IC50 values against the clinically relevant intracellular amastigote forms in the low micromolar to sub-micromolar range. Oxidative stress was verified measuring cyanide dependent respiration. Inhibition of the de novo sterol biosynthesis at the level of squalene epoxidase was confirmed, using high-resolution gas–liquid chromatography coupled to mass spectrometry, by the disappearance of the parasite’s mature sterols and the concomitant accumulation of squalene. The in vitro activities of these novel compounds were superior to that of nifurtimox, a nitrofuran currently used in the treatment of human Chagas’ disease, and terbinafine, a commercially available allylamine-based squalene epoxidase inhibitor. The results support further in vivo studies of some of these nitrofuran derivatives.

A summary of the resource.
A simple isocratic reverse-phase HPLC method for the determination of six antichagasic phenylethenylbenzofuroxans and its major synthetic secondary products, the corresponding geometric isomers and the benzofurazans, was developed and validated for use in the analysis of pre-clinical studies. Separation was achieved on a reverse-phase Supelco LC-18 column using either methanol–acetonitrile–water or acetonitrile–water, in different proportions, as mobile phase. The compounds were eluted isocratically at a flow rate of either 0.8 or 1.0 mL min−1. The compounds were analyzed with UV detection at 210 and 300 nm. The validation characteristics included linearity, accuracy, precision, specificity, limit of detection and quantification and robustness. Validation acceptance criteria were met in all cases. This method was used successfully for the quality assessment of the drugs production in the scale-up procedures.

A summary of the resource.
The synthesis of a ligand containing a nitrobenzyl group as bioreductive pharmacophore and the preparation of the corresponding technetium and rhenium complexes are presented. 99mTc labelling was performed in high yield (>90%) by ligand substitution using fac-[99mTc(CO)3(H2O)3]+ as precursor. The structure of the technetium complex was established by chromatographic comparison with the analogous rhenium compound which was fully characterized by elemental analysis, spectroscopic methods and X-ray crystallography. Reduction potential of the rhenium complex was in the characteristic range for bioreductive compounds. Biodistribution in normal mice was characterized by fast blood and soft tissue depuration and combined excretion via the hepatobiliary and urinary systems. Tumour uptake was low, probably due to low lipophilicity but tumour/muscle ratios were favourable as a consequence of high excretion

Highly variable drugs have been defined as drugs with a residual variability of more than 30% in terms of the ANOVA coefficient of variation. Different approaches have been proposed during the last years to deal with this problem but the topic remains controversial. Itraconazole, a highly variable drug, has low bioavailability with a high CYP3A4 presystemic biotransformation. Also, it has a very poor aqueous solubility which is very dependent on the pH of the dissolution medium. The pregnane X receptor (PXR) has been shown to mediate the genomic effects of progesterone and estradiol in the expression of the cytochrome P-450 gene family, which plays an important role in the metabolism of hormones and xenobiotics. During the menstrual cycle both hormone concentrations vary, providing a rationale for the more variable CYP3A4 activity in women. The analysis of the data of an itraconazole bioequivalence study involving 24 healthy volunteers (12 men and 12 women) carried out by other investigators enables us to conclude that women have less oral bioavailability and more variable AUC than men. Low bioavailability seems to be related with the higher stomach pH observed in women and variability with the aforementioned menstrual cycle incidence on both pH and CYP3A4 expression. The lower variability observed in men made it possible to discriminate differences in AUC’s variability displayed by each brand.

Divalproex sodium extended-release dosage form (divalproex-ER) has been promoted as innovative formulation for the treatment of epilepsy and manic disorders, and for migraine headache prevention, with the advantage of being dosing once a day. Due to a significant decreasing in the peak-trough fluctuation of plasma valproic acid levels, in comparison with the twice-daily dosing of conventional delayed-release formulations (divalproex-DR), concentration-dependent side effects would be prevented. However the main constraint for divalproex-ER usage is the need to be administered in a higher daily dose, because of its lower bioavailability, in order to prevent eventual breakthrough seizures when patients are switched from the twicedaily divalproex DR regimen. Taking into account free plasma drug levels, divalproex ER/DR relative bioavailability could be assessed as low as 75% in fasting condition. In order to overcome the need of increase divalproex-ER daily dose, maintenance of the twice-daily regimen is suggested. Divalproex-ER administered every 12 hours not only increases steady state trough concentration to a higher value in comparison with divalproex-DR, avoiding inefficacy of the treatment, but also achieves the safest manner to treat patients with valproic acid because of reaching practically a plateau profile of drug levels

<strong>ABSTRACT</strong><br /><br />Sandiness, one of the most common defects of dulce de leche, is caused by lactose crystallization. In order to study consumer reaction to the presence of different levels of this defect, survival analysis statistics was applied to the consumer acceptance/rejection data of the samples. Limits for this defect were estimated by working with 10% and 25% consumer rejection probabilities. The consumers were also asked to score the sample sandiness according to their perception, using a 9-point scale. Cluster analysis and correspondence analysis performed showed the heterogeneity of the consumer responses toward sandiness in dulce de leche.<br /><br />Significant correlations were established between consumer sandiness and the number of crystals and sandiness as measured by a panel of trained assessors, the latter being the best indicator of sandiness as perceived by the consumers. It could be established that the consumers' and assessors' sandiness perception is clearly influenced by the presence of agglomerates, their size distribution and number.<br /><br /><strong>PRACTICAL APPLICATIONS</strong><br /><br />Sandiness is an important defect for a segment of consumers who rejected samples with high levels of sandiness, suggesting the importance of avoiding the occurrence of this defect. Survival analysis methodology was used to estimate the maximum level of sandiness in dulce de leche before consumers reject it. As sandiness is a sensory defect that often limits the shelf life of dulce de leche, the calculated sensory limits could be used in future studies to estimate the sensory shelf life of dulce de leche.<br /><br />

A new method was developed for analysing 4-ethylguaiacol and 4-ethylphenol in the aroma of red wines using dispersive liquid–liquid microextraction (DLLME) coupled with gas chromatography–mass spectrometry detection (GC–MS). Parameters such as extraction solvent, sample volume and disperser solvent were studied and optimised to obtain the best extraction results with the minimum interference from other substances, thus giving clean chromatograms. The response linearity was studied in the usual concentration ranges of analytes in wines (50–1500 �g/L). Repeatability and reproducibility of this method were lower than 5% for both volatile phenols. Limits of detection and limits of quantification were also determined, and the values found were 28 and 95 �g/L for 4-ethylguaiacol and 44 and 147 �g/L for 4-ethylphenol, respectively. This new method has been used for the determination of the volatile phenols concentration in different samples of Tannat wine affected by Brettanomyces contamination.

In the present study, survival analysis was applied on consumers' acceptability scores, focusing the shelf-life risk on the consumers disliking the product. Shelf life was estimated as the time necessary to reach a fixed percentage of consumers disliking the product, that is, the percentage of consumers scoring the product's overall acceptability below 6 in a 9-point hedonic scale. In the present work, this methodology was applied to estimate the sensory shelf life of whole pan bread and alfajores. Shelf lives estimated considering 50% of the consumers disliking the product were shorter than those estimated considering consumers' rejection to consume. These results suggest that a proportion of consumers might dislike the sample but still answer “yes” when asked if they would consume the product at their homes. This could be attributed to the fact that when consumers are asked whether they would consume the product, they might think about consuming it after being stored at their homes. In this situation, consumers might be more tolerant toward sensory defects because they do not want to discard the product.<br />PRACTICAL APPLICATIONS<br /><br />Sensory shelf life can be estimated using survival analysis considering the percentage of consumers disliking the product. This methodology seems to be a conservative criterion in order to assure the product's quality throughout its storage.<br /><br />
