The efficiency of supercritical carbon dioxide for the selective removal of the more important pesticides from commercial lanoline was studied. Lanoline solubility values from 0.03 to 2.50 mg/g CO2 were obtained, corresponding to CO2 densities from 498.4 to 857.3 kg/m3, respectively. The original lanoline showed the following pesticide profile: diazinon 66 ppm, methyl-pirimifos 4.6 ppm and ethion 140 ppm. By an extraction carried out at 25 MPa and 45 °C, using a mass ratio solvent to substrate of 19.5 g CO2/mg lanoline, only 3.4% of lanoline was co-extracted and the pesticide concentrations were reduced to 0.47 ppm for diazinon, less than 20 ppb for methyl-pirimifos and 0.2 ppm for ethion. These results shows that supercritical CO2 is a very convenient solvent for the selective extraction of the main pesticides from lanoline, without the occurrence of substantial lanoline losses during the process. One hundred four bacterial strains mediating urinary tract infections in separate individuals from a Uruguayan community were isolated. Forty-six strains conferred a multidrug resistance phenotype. All 104 strains were examined for the presence of class 1, 2, and 3 integrons. Class 1 integrons were found in 21 isolates across four distinct bacterial genera. A large class 1 integron in a Klebsiella pneumoniae strain was fully sequenced and was 29,093 bp in length. This integron probably arose by homologous recombination since it was embedded in a hybrid Tn21-like transposon backbone which comprised a Tn5036-like tnp transposition module at the IRi integron end and a Tn21 mer module at the IRt integron end. The parent integron/transposon that contributed the Tn5036 module was not related to Tn1696 since the integron insertion points in the transposon backbones were 16 bases apart. Examination of the other 20 class 1 integron-containing strains revealed further evidence of genetic exchange. This included a strain that possessed a Tn5036 module at the IRt end but not at the IRi end and another that possessed a tnp module beyond IRi that was a hybrid of Tn21 and Tn5051 and that is presumed to have arisen by site-specific recombination. This study highlights the ability of different genetic elements to act cooperatively to spread and rearrange antibiotic resistance in a community. The purpose of the present work was to study the influence of cut (manual and using a sharp knife) and packaging film (bioriented polypropylene and polyolefin PD-961) on the sensory quality of fresh-cut butterhead lettuce stored under passive modified atmosphere at 5C. Results showed that lettuce cut by hand had a lower respiration rate and deterioration rate than those cut with a knife. This difference could be attributed to a difference in the level of wounding, which can be explained by the fact that cutting by hand caused the tear to follow the naturally weaker areas of the lettuce, leading to less tissue damage than that engendered by the use of a sharp knife. Protein glycosylation is an important post-translational modification underlying host–parasite interactions, which may determine the outcome of infection. Although Mesocestoides vogae represents an important model for investigating the various aspects of cestode biology, virtually no information is available about the structure and synthesis of glycans in this parasite. In this work, focused on the initiation pathway of mucin-type O-glycosylation in M. vogae, we characterized O-glycoproteins bearing the simple mucin-type cancer-associated Tn and sialyl-Tn antigens, and the expression and activity of ppGalNAc-T, the key enzyme responsible for the first step of mucin-type O-glycosylation. Using immunohistochemistry, Tn and sialyl-Tn antigens were detected mainly in the tegument (microtriches) and in parenchymal cells. Tn expression was also observed in lateral nerve cords. Both Tn and sialyl-Tn antigens were detected in in vitro cultured parasites. Based on their electrophoretic mobility, Tn- and sialyl-Tn-bearing glycoproteins from M. vogae were separated into several components of 22 to 60 kDa. The observation that Tn and sialyl-Tn glycoproteins remained in the 0.6 N perchloric acid-soluble fraction suggested that they could be good candidates for characterizing mucin-type glycosylation in this parasite. O-glycoproteins were purified and initially characterized using a proteomic approach. Immunohistochemical analysis of the tissue distribution of ppGalNAc-T revealed that this enzyme is expressed in the sub-tegumental region and in the parenchyma of the parasite. In M. vogae cultured in vitro, ppGalNAc-T was mainly detected in the suckers. Using a panel of 8 acceptor substrate synthetic peptides, we found that M. vogae ppGalNAc-T preferentially glycosylate threonine residues, the best substrates being peptides derived from human mucin MUC1 and from Trypanosoma cruzi mucin. These results suggest that M. vogae might represent a useful model to study O-glycosylation, and provide new research avenues for future studies on the glycopathobiology of helminth parasites. Waxes from lime (L) and orange (0) fruits, residues from the citrus industry, were evaluated as possible control methods for Cryptotermes brevis (Walker), the dry-wood termite. This termite is apermanent threat to wooden materials and goods in tropical and subtropical countries. To identify active compounds, orange (0) and lime (L) waxes were partitioned between hexane and methanol. The methanolic extracts of both waxes were further fractionated by vacuum chromatography on silica gel using a polarity gradient of chloroform (01, L1), ethyl acetate (02, L2) and methanol (03, L3). These fractions were "painted" on Pinus sp. blocks. Termite mortality (expressed as % of dead insects) was evaluated over a 60 day period. 01 was active, resulting in an insect mortality of 80% at 45 days. The L2 fraction caused 45% mortality in 28 days The lime wax fraction (L3) resulted in 86% insect mortality at the end of the 60 days. A number of treated wood blocks were stored for 5 months, and exposed to termites after this period. Blocks treated with the O1 fraction were not consumed at all, but 75% of the insects exposed died. The evaluation of the chemical composition of active fractions revealed the presence of oxygenated terpenes and coumarins in O1. Furanocoumarins, flavonoids and limonin, compounds reported as bioactive, were identified in the wax fractions. Azospirillum sp promotes the growth of many important crop plants. We demonstrated lectin binding activity in outer-membrane protein extracts of A. brasilense Sp7 by hemagglutination assays. The lectin specifically recognised the exopolysaccharide (EPS) produced by aggregated cells. Affinity chromatography using EPS-Sepharose was used to identify a 67 kDa outer-membrane lectin (OML) that recognised a binding region in the extracellular polysaccharide. Results show the specific recognition and binding between EPS and OML. The potential relationship between cell-to-cell aggregation and the OML–EPS interaction is discussed. In this article a potentiometric method for reducing sugar quantification is described. Copper(II) ion reacts with the reducing sugar (glucose, fructose, and others), and the excess is quantified using a copper wire indicator electrode. In order to accelerate the kinetics of the reaction, working conditions such as pH and temperature must be adjusted. However, a good relationship of mass of reducing sugar to potential was reached. Some of the advantages of the method are its simplicity, low cost, and that can be carried out even in colored and turbid samples. The analysis is safer than the standard Lane–Eynon method for determination of reducing sugars. To our knowledge, the use of a simple copper wire as electrode has not been reported previously for this type of determination, leading to a simplification of the experimental equipment. Heterogeneous nucleation and further growth of HgI2 onto glass were studied by the physical vapor deposition (PVD) method, with and without an argon atmosphere. Glass substrates 2″×2″ in area were used for nucleation and growth. Supersaturation strongly determined not only the nuclei population and size, but also the HgI2 nucleation phase. A range of nucleation–growth temperatures between 303 and 333 K for the substrate, and a range of initial argon pressure between 1.3 and 2.6×104 Pa were found appropriate for nucleation and growth in the α (red) phase of HgI2. Other conditions, such as an argon pressure higher than 4×104 Pa, determined the nucleation and growth in the β (yellow) metastable phase. After a nucleation growth time of 30 min, clusters of about 0.5 μm for β-HgI2 and 1 μm for α-HgI2 and a minimum distance between the clusters of about 1 and 10 μm were, respectively, obtained. Coalescence was performed by annealing the clusters at a temperature between 303 and 333 K and from 5 min to 24 h, at an initial argon pressure of 104 Pa. They coalesced to form larger clusters, which, in the case of the β-phase also transformed to the α-stable phase. From the experimental nucleation–growth and further coalescence conditions, correlations between supersaturation conditions and phase were established. Also, considerations about interface free energies and nucleation phase were made. Future work will be conducted to diminish nuclei size and enlarge nuclei population, seeking for early stages of nucleation. In order to get insight into the trypanocidal mechanism of action of a series of 5-nitrofuryl containing hiosemicarbazones some studies related to their bioreduction were performed. Electron spin resonance spectra of radicals generated in T. cruzi by compounds bioreduction were analyzed. Three different patterns of ESR signals were observed for the different assayed compounds. These results were in agreement with the changes in the T. cruzi-oxygen uptake promoted by these compounds. Cyclic voltammetry (CV) and electron spin resonance (ESR) techniques were used in the investigation of novel palladium complexes with bioactive thiosemicarbazones derived from 5-nitrofurane or 5-nitrofurylacroleine. Sixteen palladium complexes grouped in two series of the formula [PdCl2HL] or [PdL2] were studied. ESR spectra of the free radicals obtained by electrolytic reduction were characterized and analyzed. The ESR spectra showed two different hyperfine patterns. The stoichiometry of the complexes does not seem to affect significantly the hyperfine constants however we observed great differences between 5-nitrofurane and 5-nitrofurylacroleine derivatives. The scavenger properties of this family of compounds were lower than Trolox. We studied the in vitro antitumoral effect of a series of phenazine di-N-oxide derivatives, named 2-chloroacetylamino-7(8)-nitrophenazine N5,N10-dioxide (1), 2-amino-7(8)-(1,3-dioxol-2-yl)phenazine N5,N10-dioxide (2), 2-chloroacetylamino-7(8)-(1,3-dioxol-2-yl)phenazine N5,N10-dioxide (3), and 2-amino-7(8)-methoxyphenazine N5,N10-dioxide (4), on Caco-2 cells. These phenazine N5,N10-dioxide derivatives belong to our in-house chemical library. The products were selected according to their stereoelectronic characteristics and taking into account their differential cytotoxicity against V79 cells. Human colorectal adenocarcinoma cell line Caco-2 was used to study the cell growth inhibition capacity of these compounds, their capacity of altering the cell cycle and possible induction of apoptosis, DNA fragmentation, and genotoxic damage. The IC50 after 24 h of incubation was lower for 1, 2, and 3 (4.8, 46.8, and 8.2 μM, respectively) than for 4 (474.7 μM). Compound 1 induced arrest in the G2/M phase at 24 and 48 h of treatment and apoptosis at the highest doses at 24 h of treatment. These facts were corroborated with caspase 3, caspase 9, and cytochrome c activation and DNA fragmentation at 24 h of treatment. The derivatives studied induced neither significant single strand breaks nor oxidative damage at the different studied times.We concluded that among the series of N5,N10-dioxide phenazine derivatives analyzed, 1, which contains a nitro moiety and a chloroacetamide group, is the most promising as an antitumoral compound. Homology models of nicotinic acetylcholine receptors (nAChRs) suggest that subtype specificity is due to non-conserved residues in the complementary subunit of the ligand-binding pocket. Cytisine and its derivatives generally show a strong preference for heteromeric a4b2* nAChRs over the homomeric a7 subtype, and the structural modifications studied do not cause large changes in their nAChR subtype selectivity. In the present work we docked cytisine, N-methylcytisine, and several pyridone ring-substituted cytisinoids into the crystallographic structure of the Lymnaea stagnalis acetylcholine binding protein (AChBP) co-crystallized with nicotine (1UW6). The graphical analysis of the best poses showed that cytisinoids have weak interactions with the side chains of the non-conserved amino acids in the complementary subunit justifying the use of PDB 1UWB as a surrogate for nAChR. Furthermore, we found a high correlation (R2 = 0.96) between the experimental pIC50 values at a4b2* nAChR and docking energy (S) of the best cytisinoid poses within the AChBP. Due to the quality of the correlation we suggest that this equation might be used as a predictive model to propose new cytisine-derived nAChRs ligands. Our docking results also suggest that further structural modifications of these cytisinoids will not greatly alter their a4b2*/a7 selectivity. Electronic, lipophilic and steric descriptors included in QSAR-2D and -3D are analyzed for a set of ortho- and para-naphthoquinones that have proved to be powerful oxidative agents with potent trypanocidal activities specially against Leptomonas seymouri and Trypanosoma cruzi. Electronic properties are calculated by means of semiempirical (PM3), ab initio (HF/3-21G) and density functional theory (B3LYP/6-31 + G&lowast;) methodologies. Three different electronic states, neutral quinones, hydroquinones and semiquinones, are studied to investigate if any one of them are statistically related with the biological activities. The best correlations were obtained at the B3LYP level of theory because it includes electronic correlation. The QSAR-2D indicates that the best trypanocidal growth inhibitors are molecules in the semiquinone electronic state, with the following properties: (a) high negative value of EHOMO, (b) high negative charge in the oxygen atoms of the carbonyl groups, (c) high positive charge in the carbon atom of one of carbonyl moieties and (d) high electronegativity (χ). In a complementary way, the QSAR-3D indicates that the electrostatic field correlates with trypanocidal activity and the presence of bulk moieties would increase activity. The idea of comparing the three electronic states may prove to be of most importance in the general strategy to the design of new trypanocidal drugs. In fact, the experimental results showed that semiquinone is the one really statistically relevant indicating a clear connection between biochemical and theoretical aspects. Finally, we demonstrated that to be a good anti-trypanosomatid compound, the molecule must be a good electron acceptor to reach easily the essential semiquinone state. We expect that the present results motivate new experimental as well as theoretical investigations that confirm our findings. Aim We sought to determine variations in fiber organization at the molecular level using x-ray diffraction analyses on human blood vessel specimens after cryopreservation processes. Materials and Methods Diffractometric profiles were performed on aortic and carotid cryopreserved-thawed vessel samples (CVS) versus the same fresh vessel samples (FVS). X-ray diffraction was performed on vascular tissues from 17 cadaveric donors after informed consent. Ten aortic and 8 carotid diffractometric profiles were analyzed, using differential planimetric surfaces measured under x-ray diffraction curve. Diffractographic profiles were analyzed according to intervals based upon the ages of the donors. An ordering profile coefficient (OPC) was obtained as the quotient between the differential planimetric surface (DPS) of FVS versus CVS vessel ordering diffraction. Results There was a decreased ordering profile according to age: older donors showed less ordering than younger ones. Clear peaks at d-spacing of 2.86 Å and 2.15 Å (2-theta = 31.3° and 42.0°, respectively) were always confirmed despite the different profiles of samples. OPC showed a higher ordering profile among the CVS than FVS: 70% aortas and 62.5% carotids. Conclusion The cryopreserved-thawed procedure does not damage the fibrillar organization of vessels. Hybrid compounds containing hydrazones and benzofuroxan pharmacophores were designed as potential Trypanosoma cruzi-enzyme inhibitors. The majority of the designed compounds was successfully synthesized and biologically evaluated displaying remarkable in vitro activity against different strains of T. cruzi. Unspecific cytotoxicity was evaluated using mouse macrophages, displaying isothiosemicarbazone 10 and thiosemicarbazone 12 selectivity indexes (macrophage/parasite) of 21 and 27, respectively. New 1,2,4-thiadiazolylnitrones and furoxanylnitrones were developed and evaluated as neuroprotective agents on a human neuroblastoma (SH-SY5Y) cells model. They inhibited at low micromolar concentrations the oxidative damage and the death induced by exposure to hydrogen peroxide. These heteroarylnitrones showed excellent peroxyl free radical absorbance capacities, analyzed by oxygen radical absorbance capacity (ORAC) assay with fluorescein as the fluorescent probe, ranging from 1.5- to 16.5-fold the value of the reference nitrone, α-phenyl-N-tert-butylnitrone (PBN). The electron spin resonance spectroscopy (ESR) demonstrated the ability of these derivatives to directly trap and stabilize oxygen, carbon, and sulfur-centered free radicals. These results demonstrated the potential use of these heteroarylnitrones as neuroprotective agents in preventing the death of cells exposed to enhanced oxidative stress and damage.
