Las preferencias de aprendizaje de los estudiantes están cambiando, acorde a los múltiples cambios que transforman el mundo y las sociedades que lo integran. Esto constituye un desafío para los docentes, debido a que buscan hacer propuestas que interesen y motiven al estudiante y que a su vez aporten conocimiento riguroso de los contenidos de las asignaturas. En este trabajo se plantea una actividad experimental, en donde se determina in vitro la capacidad neutralizante de ácido, en distintos productos antiácidos de amplio uso, empleando pHmetro y datalogger con sensor de pH. Capacidad neutralizante de antiácidos, una propuesta experimental.

Free simple phenols have a significant role in defining the sensory and nutritional characteristics of wines, affecting the organoleptic profile and having positive effects on health, but glycosidically bound phenols can also be hydrolysed during the winemaking process, releasing the corresponding volatile compounds and making a possible contribution to the final sensory profile. In this work, application of on-line SPE liquid chromatography–high resolution mass spectrometry, operating in negative polarity with heated electrospray, allowed to detect over eighty free and glycosylated simple phenols in Primitivo di Manduria and Negroamaro wines. Sixty-one phenols, four of which phenolic glucosidic precursors, were quantified as having quantification limits ranging from 0.001 to 0.1 lgmL�1, calibration R2 of 0.99 for over 92% of compounds, and precision (R.S.D.%) always lower than 12%. Twenty-four simple phenolic precursors were tentatively identified as hexoside, pentoside and hexoside– hexoside derivatives, on the basis of accurate mass, isotopic pattern and MS/MS fragmentation

Vitis vinifera is one of the most widespread grapevines around the world representing the raw material for high quality wine production. The availability of more resistant interspecific hybrid vine varieties, developed from crosses between Vitis vinifera and other Vitis species, has generated much interest, also due to the low environmental effect of production. However, hybrid grape wine composition and varietal differences between interspecific hybrids have not been well defined, particularly for the simple phenols profile. The dynamic of these phenols in wines, where the glycosylated forms can be transformed into the free ones during winemaking, also raises an increasing health interest by their role as antoxidants in wine consumers. In this work an on-line SPE clean-up device, to reduce matrix interference, was combined with ultra-high liquid chromatography-high resolution mass spectrometry in order to increase understanding of the phenolic composition of hybrid grape varieties. Specifically, the phenolic composition of 4 hybrid grape varieties (red, Cabernet Cantor and Prior; white, Muscaris and Solaris) and 2 European grape varieties (red, Merlot; white, Chardonnay) was investigated, focusing on free and glycosidically bound simple phenols and considering compound distribution in pulp, skin, seeds and wine. Using a targeted approach 53 free simple phenols and 7 glycosidic precursors were quantified with quantification limits ranging from 0.001 to 2 mg Kg− 1 and calibration R2 of 0.99 for over 86% of compounds. The untargeted approach made it possible to tentatively identify 79 glycosylated precursors of selected free simple phenols in the form of -hexoside (N = 30), -pentoside (21), -hexoside-hexoside (17), -hexoside-pentoside (4), -pentoside-hexoside (5) and -pentoside-pentoside (2) derivatives on the basis of accurate mass, isotopic pattern and MS/MS fragmentation.

A pesar de los grandes avances en la quimioterapia de las enfermedades infecciosas estas son aún una de las principales causas de muerte en el mundo. Aunque parece existir una gran gama de fármacos antibacterianos y antifúngicos de uso clínico, la aparición de microorganismos resistentes los hace a veces ineficaces. Entre los patógenos más problemáticos clínicamente relevantes en la actualidad el Staphylococcus aureus meticilino resistente (MRSA) se ubica como una de las bacterias más difíciles de tratar. La importancia de las enfermedades de plantas en la agricultura moderna es un hecho ampliamente documentado y reconocido. Las enfermedades tienen el potencial de destruir enteramente las cosechas o reducir en forma crónica el rendimiento de la mayoría de los cultivos, obligan a tomar medidas de combate que aumentan los costos de producción, y afectan la calidad y la durabilidad de los productos cosechados. Alrededor de dos tercios de las enfermedades de las plantas son causadas por hongos. La utilización intensiva de compuestos antimicrobianos en la agricultura industrial favorece la aparición de resistencias. Esto, unido a cambios frecuentes en los límites de aplicación, límites de residuos en el ambiente y en el producto, lleva a que este sea un sector altamente dinámico con una rápida rotación de los compuestos activos. Los hongos comparten con las plantas y los animales ciertos agentes infecciosos tales como bacterias y hongos patógenos y por lo tanto tienen el potencial de producir metabolitos con actividad farmacológica de uso en humanos y cultivares. Los “microhongos” son una fuente ya establecida como importante de compuestos antimicrobianos. Sin embargo, para los “macrohongos” (aquellos cuyo cuerpo de fructificación es visible a simple vista, la gran mayoría perteneciente al filo Basidiomycota) sólo unos pocos compuestos aislados se encuentran en la etapa de ensayos clínicos y de campo. 2 El presente trabajo apuntó a la enorme diversidad metabólica de los microorganismos, utilizando basidiomicetes como fuente original. Para eso se abordaron dos estrategias diferentes, el trabajo con los micelios y caldos extracelulares de cultivos fúngicos y el trabajo con los cuerpos de fructificación. A partir de cultivos de diecinueve hongos basidiomicetes se prepararon cincuenta y tres extractos que posteriormente fueron sometidos a ensayos biológicos. Cuarenta por ciento de los extractos presentaron actividad frente a los patógenos humanos evaluados y diecisiete por ciento de los extractos presentaron actividad frente a los fitopatógenos evaluados. Para la cepa más prometedora, se estudió la influencia de variables en las condiciones de cultivo sobre la producción de metabolitos de interés para obtener cantidades de extracto suficientes para avanzar en el aislamiento de los compuestos responsables de la actividad antimicrobiana. Considerando que los resultados obtenidos no fueron los esperados se recorrió el camino inverso, aislando compuestos con actividad antimicrobiana a partir de cuerpos de fructificación de Gymnopilus junonius y posteriormente induciendo la producción de los mismos en cultivos fúngicos de dicho hongo. El agregado de agentes epigenéticos influyó positivamente en la riqueza química de los extractos, no incidiendo en la producción de bisnoryangonina, compuesto aislado con promisoria actividad antimicrobiana frente a S. aureus meticilino resistente.

Despite the great advances in chemotherapeutics, infectious diseases are still one of the leading causes of death worldwide. Among some of the clinically relevant pathogens, methicillin-resistant Staphylococcus aureus (MRSA) ranks as one of the most difficult bacteria to treat. It is a common cause of skin, soft-tissue, and endovascular infections, as well as pneumonia, septic arthritis, endocarditis, osteomyelitis, and sepsis. The research on Basidiomycota is extensive; many species show a broad spectrum of pharmacological activities, including antimicrobial activity. The vast majority of the literature to date generally focuses on screening the antibacterial properties of mushroom extracts. A gap still exists in the identification of the individual compounds responsible for these properties, and few low molecular weight compounds have been described. Gymnopilus junonius, the big laughter mushroom, grows wild in Uruguay, especially on Eucalyptus spp. plantations; it is known as the "eucalyptus fungus." In this work, we report the bioguided isolation, structural elucidation, and antistaphylococcal activity of the main antimicrobial components of fresh basidiocarps of G. junonius.

Crystal structures can strongly deviate from bulk states when confined into nanodomains. These deviations may deeply affect properties and reactivity and then call for a close examination. In this work, we address the case where extended crystal defects spread through a whole solid and then yield an aperiodic structure and specific reactivity. We focus on iron boride, α-FeB, whose structure has not been elucidated yet, thus hindering the understanding of its properties. We synthesize the two known phases, α-FeB and β-FeB, in molten salts at 600 and 1100 °C, respectively. The experimental X-ray diffraction (XRD) data cannot be satisfactorily accounted for by a periodic crystal structure. We then model the compound as a stochastic assembly of layers of two structure types. Refinement of the powder XRD pattern by considering the explicit scattering interference of the different layers allows quantitative evaluation of the size of these domains and of the stacking faults between them. We, therefore, demonstrate that α-FeB is an intergrowth of nanometer-thick slabs of two structure types, β-FeB and CrB-type structures, in similar proportions. We finally discuss the implications of this novel structure on the reactivity of the material and its ability to perform insertion reactions by comparing the reactivities of α-FeB and β FeB as reagents in the synthesis of a model layered material: Fe2AlB2. Using synchrotron-based in situ X-ray diffraction, we elucidate the mechanisms of the formation of Fe2AlB2. We highlight the higher reactivity of the intergrowth α-FeB in agreement with structural relationships.

The dehalogenation of 2-chloroethanol (2ClEtOH) in the gas phase with and without the participation of catalytic water molecules has been investigated using methods rooted into the density functional theory. The well-known HCl elimination leading to vinyl alcohol (VA) was compared to the alternative elimination route toward oxirane and shown to be kinetically and thermodynamically more favorable. However, the isomerization of VA to acetaldehyde in the gas phase, in the absence of water, was shown to be kinetically and thermodynamically less favorable than the recombination of VA and HCl to form the isomeric 1-chloroethanol (1ClEtOH) species. At the ωB97X-D/cc-pVTZ level of calculation, this species is more stable than 2ClEtOH by about 6 kcal mol–1 at 298 K, and the reaction barrier for VA to 1ClEtOH is 23 kcal mol–1 versus 55 kcal mol–1 for the direct transformation of VA to acetaldehyde. In a successive step, 1ClEtOH can decompose directly to acetaldehyde and HCl with a lower barrier (29 kcal mol–1) than that of VA to the same products (55 kcal mol–1). The calculations were repeated using a single ancillary water molecule (W) in the complexes 2ClEtOH_W and 1ClEtOH_W. The latter adduct is now more stable than 2ClEtOH_W by about 8 kcal mol–1 at 298 K, implying that the water molecule increased the already higher stability of 1ClEtOH in the gas phase. However, this catalytic water molecule lowers dramatically the barrier for the interconversion of VA to acetaldehyde (from 55 to 7 kcal mol–1). This barrier is now smaller than the one for the conversion to 1ClEtOH (which also decreases, but not so much, from 23 to 13 kcal mol–1). Thus, it is concluded that while 1ClEtOH may be a plausible intermediate in the gas phase dehalogenation of 2ClEtOH, it is unlikely that it plays a major role in water complexes (or, by inference, aqueous solution). It is also shown that neither in the gas phase nor in the cluster with one water molecule, the oxirane path is more favorable than the VA alcohol path. Additionally, a direct conversion of 2ClEtOH to 1ClEtOH through a transition state which resembles a VA molecule in a complex with a chlorine atom and a hydrogen atom on both sides of this planar species was found. This reaction path has also lower activation energy than the conversion to oxirane but not as low as the conversion to VA.

Abstract
A summary of the resource.
In a previous work, we have investigated the initial steps of the reaction of toluene with the hydroxyl radical using several quantum chemical approaches including density functional and composite post-Hartree-Fock models. Comparison of H-abstraction from the methyl group and additions at different positions of the phenyl showed that the former reaction channel is favored at room temperature. This conclusion appears at first sight incompatible with the experimental observation of a lower abundance of the product issuing from abstraction (benzaldehyde) with respect to those originating from addition (cresols). Further reactions of the intermediate radicals with oxygen, water and additional OH radicals are explored in this paper through theoretical calculations on more than 120 species on the corresponding potential energy surface. The study of the addition reactions, to obtain the cresols through hydroxy methylcyclodienyl intermediate radicals, showed that only in the case of o-cresol the reaction proceeds by addition of O2 to the ring, internal H-transfer and hydroperoxyl abstraction, and not through direct H abstraction. Both for p- and m-cresol instead the reaction occurs through a higher energy direct H-abstraction, thus explaining in part the observed larger concentration of the ortho isomer in the final products. It was also found that the benzyl radical, formed by H-abstraction from the methyl group, is able to react further if additional OH is present. Two reaction paths leading to o-cresol, two leading to pcresol and one leading to m-cresol were determined. Moreover, in this situation, benzyl radical is predicted to produce benzyl alcohol, as was found in some experiments. The commonly accepted route to benzaldehyde was found to be not the energetically favored one. Instead, a route leading to benzoyl radical (and ultimately to benzoic acid) with the participation of one water molecule was clearly more favorable, both thermodynamically and kinetically.

This work reports density functional and composite model chemistry calculations performed on the reactions of toluene with the hydroxyl radical. Both the experimentally observed H-abstraction from the methyl group and possible OH-additions to the phenyl ring were investigated. Reaction enthalpies and barrier heights suggest that H-abstraction is more favorable than OH-addition to the ring. The calculated reaction rates at room temperature and the radical-intermediate product fractions support this view. At first sight, this might seem to disagree with the fact that, under most experimental conditions, cresols are observed in a larger concentration than benzaldehyde. Since the accepted mechanism for benzaldehyde formation involves H-abstraction, a contradiction arises that calls for a more elaborate explanation. In this first exploratory study, we provide evidence that support the preference of H-abstraction over OH-addition and present an alternative mechanism which shows that cresols can be actually produced also through H-abstraction and not only from OH-addition, thus justifying the larger proportion of cresols than benzaldehyde among the products.

Purine transporters as substrate entry points in organisms, are involved in a number of cellular processes such as nitrogen source uptake, energy metabolism and synthesis of nucleic acids. In this study, two nucleobase transporter genes (phZ, phU) from Phanerochaete chrysosporium were cloned, identified, and functionally characterized. Our results show that PhZ is a transporter of adenine and hypoxanthine, and a protein belonging to the AzgA-like family, whilst PhU belongs to the NAT/NCS2 family, transporting xanthine and uric acid. No other sequences belonging to these families were detected in P. chrysosporium's genome. Phylogenetic analyses show that AzgA-like sequences form monophyletic groups for each major lineage (Ascomycota, Basidiomycota and Zygomycota). In contrast, Ascomycota and Basidiomycota NAT/NCS2 sequences do not form monophyletic groups and several copies of this protein are distributed across the tree. Expression of phU was significantly downregulated in the presence of a primary source like ammonium, and enhanced if purines were present or if the mycelium was nitrogen starved. phZ was clearly induced by its substrates (hypoxanthine, adenine), very lightly induced by xanthine, suppressed by urea and amino acids and expressed at a basal level when uric acid or ammonium was the nitrogen source or when the mycelium was starved for nitrogen. In order to perform substrate analyses, both P. chrysosporium proteins (PhZ, PhU) were expressed in Aspergillus nidulans. Epifluorescent microscopy showed that under inducing conditions, PhZ-GFP and PhU-GFP were present at the plasma membrane of A. nidulans transformed strains, and were internalized in repressed conditions. Our results suggest that in the white-rot fungus P. chrysosporium, phU has a catabolic role and phZ, (less dependent of the nitrogen source), plays a key role in purine acquisition to provide biosynthetic components. These are the first purine transporters characterized in Basidiomycota.

Sterols (e.g. cholesterol) present in wool scouring effluent represent the most recalcitrant fraction in anaerobic treatment. This study was conducted to examine the feasibility of removal of this organic load through a denitrifying post-treatment stage. A stable cholesterol-denitrifying enrichment (CHOL-1) was obtained from sludge of a bench-scale upflow sludge bed (USB) denitrifying reactor integrated to a carbon and nitrogen removal system for sanitary landfill leachate. According to the amounts of cholesterol degraded and of nitrite and nitrogen gas formed, the capacity for complete cholesterol oxidation under anaerobic conditions by CHOL-1 can be assumed. Nitrite accumulation observed at a low C/N ratio outlines the importance of determining the optimal C/N ratio for adequate denitrifying reactor performance. The enrichment was partly identified with molecular analysis of cloned 16S rDNA sequences revealing the presence of two groups of bacteria belonging to the b subclass of the Proteobacteria. According to analysis of sequences, it can be inferred that a yet uncultivated new bacterium is the one responsible for cholesterol oxidation. Results of this study suggest that sludge from a denitrifying reactor treating leachate is potentially useful in a combined anaerobic-anoxic system for degradation of cholesterol that remains after methanogenic treatment.

The study of the adaptation mechanisms that allow microorganisms to live and proliferate in an extreme habitat is a growing research field. Directly exposed to the external environment, lipopolysaccharides (LPS) from Gram-negative bacteria are of great appeal as they can present particular structural features that may aid the understanding of the adaptation processes. Moreover, through being involved in modulating the mammalian immune system response in a structure-dependent fashion, the elucidation of the LPS structure can also be seen as a fundamental step from a biomedical point of view. In this paper, the lipid A structure of the LPS from Spiribacter salinus M19-40T, a halophilic gamma-proteobacteria, was characterized through chemical analyses and matrix-assisted laser desorption ionization (MALDI) mass spectrometry. This revealed a mixture of mono- and bisphosphorylated penta- to tri-acylated species with the uncommon 2 + 3 symmetry and bearing an unusual 3-oxotetradecaonic acid.

A summary of the resource.
In the present study the use of an open-ended question to identify drivers of liking of milk desserts was evaluated. Eight samples of vanilla milk desserts were presented to 80 consumers, who were asked to score their overall liking using a 9-point hedonic scale and to provide up to four words to describe each of the samples. Also, the samples were evaluated by a panel of trained assessors. Consumers’ descriptions of the samples were mainly related to hedonic and sensory characteristics of the milk desserts. The elicited terms provided information about which are the attributes that drive consumers’ preferences using their own language. Multiple factor analysis was used to obtain a synthetic map of the samples based on consumers’ descriptions, sensory data and overall liking scores. This methodology could be an interesting technique to select the most relevant attributes for consumers and to study the relationship between sensory data and consumers descriptions.

Benzofuroxan derivatives have been shown to inhibit the growth of Trypanosoma cruzi, the etiological agent of Chagas� disease. Therefore, 2D- and 3D-QSAR models of their in vitro antichagasic activity were developed. Six new derivatives were synthesized to complete a final set of 26 structurally diverse benzofuroxans. The 2D-QSAR model (r = 0.939, r2 adj ¼ 0:849) was generated using multiple regression analysis of tabulated substituents� physicochemical properties and indicator variables. In addition, a 3D-QSAR model (r2 = 0.997, q2 = 0.802) was obtained using a comparative molecular field analysis (CoMFA). Due to the wellknown benzofuroxan tautomerism, in both approaches (2D- and 3D-QSAR) it was necessary to include an indicator variable to consider the N-oxide position (I6). This parameter was established using low-temperature NMR experiments. Both QSAR models identified the electrophilic character of the substituent a-atom as a requirement for activity. Further support was found using a density functional theory (DFT) approach.

Sensory analysis of olive oils is one of the most important evaluations for determining the quality of virgin olive oils. Sensory evaluation is carried out by a panel of trained tasters, and the median values of the positive and negative attributes are used for classification based on the guidelines set by the International Olive Council. The main purpose of this quality assessment is to ensure that consumers have access to high-quality olive oils. However, consumer perception of the sensory characteristics of high-quality olive oils does not always agree with the panel evaluation. This review aims to discuss the relationship between the two approaches, the official method, and consumer evaluation.

<p>The challenges posed by intrinsically disordered proteins (IDPs)<br />to atomistic and coarse-grained (CG) simulations are boosting efforts to develop and reparametrize current force fields. An assessment of the dynamical behavior of IDPs’ and unstructured peptides with the CG SIRAH force field suggests that the current version achieves a fair description of IDPs’ conformational flexibility. Moreover, we found a remarkable capability to capture the effect of point mutations in loosely structured peptides.</p>

Electron spin resonance (ESR), called also electron paramagnetic resonance (EPR) together with the spin trapping technique, has allowed us to study and understand how free radicals are involved in various pathologies. In this review, the importance of spin trapping technique in the study of diseases such as cancer, diabetes, hypertension and parasitic diseases is discussed. In addition, advances in the use of this technique as therapeutic agents and other interesting applications as the immuno-spin trapping technique are reviewed.

Abstract
A summary of the resource.
A new series of heteroaryl nitrones were synthesized and evaluated as free radical traps due to the results showed in our previous report. The physicochemical characterization of these new nitrones by electron spin resonance (ESR) demonstrated their high capability to trap and stabilize different atom centered free radicals generated by the Fenton reaction. Additionally, we intensely studied them in terms of their physicochemical properties. Kinetic studies, including the use of a method based on competition and the hydroxyl adduct decay, gave the corresponding rate constants and half-lives at the physiological pH of these newly synthesized nitrones. New nitrones derived from quinoxaline 1,4-dioxide heterocycles were more suitable than DMPO to trap hydroxyl free radicals with a half-life longer than two hours. We explain some of the results using computational chemistry through density functional theory (DFT).

A summary of the resource.
In this work the study of Calf Thymus DNA interaction with several Cu(L-dipeptide) complexes was reported. The binding stoichiometry (Cu(mmol)/DNAmol base) was determined and in an attempt to clarify the binding mode, EPR and CD experiments were performed. All the studied complexes interacted with DNA, in a more selective way than [Cu(H2O)6]2+, being the [Cu(ala-phe)] the complex with the highest interaction. The EPR experiments suggested that the monomeric species formed in solution were coordinated through a nitrogen atom of the DNA bases (inner-sphere binding) and the CD studies showed structural changes upon the DNA–complex interaction. Besides, the ratio Cu(mmol)/DNAmol base obtained by the binding stoichiometry experiments was close to that found by EPR and CD determinations. The effect on cell proliferation determined by the crystal violet bioassay on UMR106 rat osteosarcoma- derived cells showed that the [Cu(ala-phe)] complex exerted an antiproliferative action against this tumor line.

<p>Oxidizing agents of sulfhydryl groups such as iodosobenzoate, alkylating agents such as iodoacetamide, and mercaptide-forming agents such as cadmium chloride, mercuric chloride, <em>p</em>-chloromercuribenzoate, sodium arsenite, and <em>p</em>-carboxyphenylarsine oxide, added in small concentrations to a suspension of sea urchin sperm produced an increase in respiration. When the concentration was increased there was an inhibition. These effects are explained by postulating the presence in the cells of two kinds of sulfhydryl groups: <em>soluble sulfhydryl groups</em>, which regulate cellular respiration, and <em>fixed sulfhydryl groups</em>, present in the protein moiety of enzymes. Small concentrations of sulfhydryl reagents combine only with the first, <em>thus</em> producing an increase in respiration; when the concentration is increased, the fixed sulfhydryl groups are also attacked and inhibition of respiration is the consequence.</p>
Other inhibitors of cell respiration, such as cyanide and urethanes, which do not combine with —SH groups, did not stimulate respiration in small concentration.
