A summary of the resource.
Characterization studies were performed to a proteolytic extract, prepared from latex of Ficus luschnathiana (Miq.) Miq., a Moraceae family tree. The mechanism of action was determined, corresponding to serine proteinases behaviour. Studies of optimum pH and temperature showed a maximum catalytic activity at pH 8.0 and 75°C, respectively. The proteolytic activity exhibited good stability when incubated at different pH and temperature conditions. At 7°C and pH 7.5, the extract kept a 100% of the initial activity for 5 months. When comparing the effect of pH on the stability, maximum values were obtained at pH 6.5, with 98% of the initial activity remaining after 5 h of incubation at 60°C. After 1 min at 100°C total inactivation occurred. Regarding the potential use in the dairy industry, milk clotting activity was determined obtaining a very effective coagulant action. Analysis protocols which included molecular exclusion chromatography, electrophoretic methods and mass spectrometry revealed a complex protein profile. It was found that the molecules responsible for the proteolytic activity of the extract were several proteins of different molecular weight and isoelectric points, most of which still retained the activity in presence of SDS and reducing agents. These proteic components displayed a pH-dependant tendency to form aggregates. Using the proteolytic activity of the crude extract under controlled hydrolysis conditions on whey proteins, yielded peptidic fractions with antimicrobial activity against a S. aureus strain. It was found that fractions of peptide concentration of 0.96 μmol mL-1 inhibited growth by 94%.

Mate finding in many spider species is mediated, at least in part, by chemical cues. Although few have been characterized, most spider sex pheromones seem to be associated with the silk threads of adult females, attracting and/or triggering sexual behaviors in males. Schizocosa malitiosa (Araneae: Lycosidae) is a wolf spider common in dry environments in Southern Uruguay. Here, we report evidence for the occurrence of a female sex pheromone in the silk of virgin S. malitiosa females. The pheromone elicits typical courtship displays by conspecific males (palpal drumming and leg shaking), when it is applied to an artificial substrate. We also showed that this pheromone is quickly inactivated under field conditions, possibly due to the effect of dew, and that it is readily extracted from the silk by water, which renders the silk threads inactive. Preliminary chemical analyses by mass spectrometry suggest that the pheromone is a low molecular weight, highly oxygenated polar compound, present in a high concentration only in older virgin females. Quick inactivation in the field of the pheromone would allow males to discriminate between recent and old cues, thus facilitating mate searching and sexual meeting, by preventing males from following old cues for wandering females that could be distant in space and/or time.

<em>Quillaja brasiliensis</em> (A. St.-Hil. & Tul.) Mart. is currently one of the only two species of the only genus in the Quillajaceae D. Don (Dicotyledoneae) family. Recently, remarkable developments have been achieved regarding its domestication,<br />propagation, and biochemical study. The first results on the structural study of the complex mixture of saponins produced by different organs of the tree have been published. Analogously to the related species Q. saponaria Molina, its saponins, either alone or in colloidal formulations, have been proved to be very effective as adjuvants in experimental vaccines, being able to elicit an early humoral and cellular response against the co-administered antigens. This paper describes a non-destructive “peak parking” protocol in order to assess the axial heterogeneity of an in situ modified monolithic column for high performance liquid chromatography; a “gradient stationary phase” was designed whereby the ligand density decreases along the length of the rod in the “forward flow” configuration. The results of multi-location peak parking demonstrated a consistent increase in peak variance from the 1 cm position of the column to the 9 cm location. This increase in band broadening supported the theory of a decreasing ligand density along the length of this gradient column. This is consistent with efficiency measurements performed in both the forward and reverse flow directions, with an improved efficiency (15% increase in N m−1) in the reverse direction. These results are consistent with theoretical investigations into stationary phase gradients.

Abstract
A summary of the resource.
This work presents the synthesis and characterization of eight copper complexes [Cu(L-dipeptide)(neo)]·nH2O (neo = neocuproine) and their cytotoxic activities against tumor cell lines. The crystalline structure of [Cu(gly-val)(neo)]·3H2O, [Cu(gly-leu)(neo)]·H2O, [Cu(ala-gly)(neo)]·4H2O, [Cu(val-phe)(neo)]·4.5H2O and [Cu(phe-phe)(neo)]·3H2O were determined by single crystal X-ray diffraction. In all of them, the Cu(II) is pentacoordinated, in a square pyramidal environment. The coordination observed in solid state was retained in the major species in aqueous solution, as suggested by Electronic Paramagnet Resonance and UV–vis spectroscopies. The complexes were shown to have affinity for isolated DNA, as determined by Circular Dichroism experiments. Furthermore, biological experiments showed that all the complexes present high cytotoxic activity against the cell lines: MDA-MB-231, MCF-7 (human metastatic breast adenocarcinomas, the first triple negative), MCF-10A (human normal breast cells), A549 (human lung epithelial carcinoma) and MRC-5 (human lung epithelial cells). Together, these results suggest that these compounds are promising steps towards new effective drugs to treat cancer.

Coordination complexes may act as anticancer agents. Among others, the formation of the complex may facilitate the ligand uptake by the cell. Searching for new copper compounds with cytotoxic activity, the complex Cu-dipicolinate was studied as a neutral scaffold to form ternary complexes with diimines. A series of [Cu(dipicolinate)(diimine)] complexes (where diimine: Phenan throline, phen, 5-NO2 -phenanthroline, 4-methyl-phenanthroline, neocuproine, 3,4,7,8-tetramethyl phenanthroline, tmp, bathophenanthroline, bipyridine, dimethyl-bipyridine, as well as the ligand 2,2-dipyridil-amine, bam) were synthesized and characterized both in the solid state, including a new crystal structure of [Cu2 (dipicolinate)2 (tmp)2 ]·7H2O. Their chemistry in aqueous solution was explored by UV/vis spectroscopy, conductivity, cyclic voltammetry, and electron paramagnetic resonance studies. Their DNA binding was analyzed by electronic spectroscopy (determining Kb values), circular dichroism, and viscosity methods. The cytotoxicity of the complexes was assessed on human cancer cell lines MDA-MB-231, MCF-7 (breast, the first triple negative), A549 (lung epithelial) and A2780cis (ovarian, Cisplatin-resistant), and non-tumor cell lines MRC-5 (lung) and MCF-10A (breast). The major species are ternary, in solution and solid state. Complexes are highly cytotoxic as compared to Cisplatin. Complexes containing bam and phen are interesting candidates to study their in vivo activity in triple-negative breast cancer treatment.

Cyanobacterial 16S ribosomal RNA gene diversity was examined in a benthic mat on Fildes Peninsula of King George Island (62809054.400S, 58857020.900W), maritime Antarctica. Environmental DNA was isolated from the mat, a clone library of PCR-amplified 16S rRNA gene fragments was prepared, and amplified ribosomal DNA restriction analysis (ARDRA) was done to assign clones to seven groups. Low cyanobacterial diversity in the mat was suggested in that 83% of the clones were represented by one ARDRA group. DNA sequences from this group had high similarity with 16S rRNA genes of Tychonema bourrellyi and T. bornetii isolates, whose geographic origins were southern Norway and Northern Ireland. Cyanobacterial morphotypes corresponding to Tychonema have not been reported in Antarctica, however, this morphotype was previously found at Ward Hunt Lake (838N), and in western Europe (528N). DNA sequences of three of the ARDRA groups had highest similarity with 16S rDNA sequences of the Tychonema group accounting for 9.4% of the clones. Sequences of the remaining three groups (7.6%) had highest similarity with 16S rRNA genes of uncultured cyanobacteria clones from benthic mats of Lake Fryxell and fresh meltwater on the McMurdo Ice Shelf.

Estudios previos realizados en el Laboratorio de Ecologfa Microbiana-IIBCE demostraron que diferentes bacterias no patogenas de ani males, entre ellas los rizobios, eran capaces de utilizar compuestos hemfnicos (hemina, hemoglobina y leghemoglobina) como fuentes de hierro.
EI objetivo general del presente trabajo de tesis fue el de caracterizar los mecanismos de adquisicion de hierro mediados por compuestos hemfnicos presentes en Sinorhizobium meliloti 242.
Los objetivos particulares fueron: a) estudiar la funcion de la L-metionina en la adquisicion de hierro a partir de compuestos hemfnicos y b) detectar y purificar las posibles protefnas de membrana externa con capacidad para unirse a hemina, hemoglobina y leghemoglobina.
A partir de ensayos de crecimiento bacteriano en medio Ifquido y bioensayos, se determino que es necesario suplementar al medio mfnimo de cultivo, MV, con L-metionina 300I-lM para que la cepa Sm 242 sea capaz de crecer en medios limitados en hierro con el agregado de diferentes fuentes de este metal (compuestos hemfnicos 0 sideroforos). Cuando se utilizo glutamato en lugar de amonio como fuente de nitrogeno y se agrego L-metionina al medio de cultivo, se obtuvo una mayor produccion de sideroforos y mayores rendimientos celulares. A partir de la informacion disponible, una posible interpretacion de los resultados obtenidos es que en condiciones de baja disponibilidad de hierro, la vfa de asimilacion del azufre a partir del sulfato, este afectada probablemente a nivel de la sulfito reductasa. La L-metionina en estas condiciones serfa una mejor fuente de azufre para la cepa Sm 242.
Se purifico leghemoglobina a partir de nodulos de soja y se obtuvieron anticuerpos de conejo anti-Ieghemoglobina. Estos anticuerpos se caracterizaron por Western blot comprobondose que no presentan reactividad cruzada con otros compuestos hemfnicos ni con las demos protefnas presentes en el macerado nodular. A su vez, por ELISA, se determino la dilucion de trabajo de los anticuerpos, los cuales seron empleados en experimentos futuros.
Por ultimo, mediante anolisis por SDS-PAGE de fracciones enriquecidas en protefnas de membrana externa, se detecto la presencia de 3 protefnas reguladas por hierro de 80, 82 y 90 kDa. De las 3 protefnas identificadas, la de 90 kDa presento la capacidad de unirse a hemina inmovilizada en agarosa. Anolisis por espectrometrfa de masas (EM-MALDI-TOF) determinaron que la protefna de 90 kDa es homologa 0 aun identica a una posible protefna involucrada en el transporte de hierro (Smc02726) deducida a partir del genoma secuenciado de Sm 1021. Anolisis bioinformoticos mostraron que la secuencia aminoaddica de esta protefna presenta dominios altamente conservados con otras protefnas de membrana externa receptoras para la hemina. A partir de los resultados obtenidos podemos concluir que se ha identificado en Sm 242 un receptor de hemina.

Se evaluó la respuesta de cerdos a soja desactivada artesanalmente en dietas isoenergéticas e isoproteicas en base a maíz. Se utilizaron 28 machos castrados entre 40 y 105 kg de peso vivo asignados a cuatro tratamientos variando el suplemento proteico: T1: harina de soja, T2: poroto de soja cocido, T3: poroto de soja tostado, T4: poroto de soja crudo. Se evaluaron: consumo diario (CD) y total (CT) de alimento, ganancia diaria de peso (GP), índice de conversión (IC), rendimiento de carcasa (R), largo de res (L), espesor de grasa dorsal (GD), perfil lipídico de la grasa dorsal (PL). Los animales del T4 presentaron menor CD (P≤0,01) sin diferencias en el CT. La GP fue mayor (P≤0,01) para T1, sin diferencias entre T2 y T3 y menor para T4. T1 tuvo mejor IC que T4 (P≤0,01) y T2 y T3 (P≤0,05) mientras que T4 fue inferior (P≤0,01). No se observaron diferencias en R ni GD, mientras que L fue mayor (P≤0,01) para el T1. Se observaron diferencias en PL, con mayor contenido en PUFA y en los ácidos oleico, linoleico y linolénico en los tratamientos incluyendo poroto de soja integral (P≤0,01). Se concluye que los tratamientos realizados al grano de soja mejoraron su valor nutricional con respecto al poroto crudo, aunque no se igualaron los resultados de la dieta testigo. Las carcasas provenientes de dietas con poroto de soja integral presentan menor valor industrial, pero mejor valor nutricional para consumo fresco que las basadas en harina de soja.

A summary of the resource.
The design of vaccines that protect against intracellular infections or cancer remains a challenge. In many cases, immunity depends on the development of antigen-specific memory CD8+ T-cells that can express cytokines and kill antigen-bearing cells when they encounter the pathogen or tumor. Here, the authors review current understanding of the signals and cells that lead to memory CD8+ T-cell differentiation, the relationship between the primary CD8+ T-cell response and the memory response and the regulation of memory CD8+ T-cell survival and function. The implications of this new knowledge for vaccine design are discussed, and recent progress in the development of lipidated peptide vaccines as a promising approach for vaccination against intracellular infections and cancer is reviewed.

The aims of this work were to investigate the existence of Tindependent antigens in Echinococcus granulosus protoscoleces and to evaluate the relative contribution of T-independent stimulation to the overall antibody response in early infection. Mice depleted of CD4þ-cells were immunized with protoscolex somatic antigens (PSA) or infected with E. granulosus protoscoleces (PSC). Results showed that the response of CD4-depleted immunized mice had the expected characteristics of a T-independent stimulation and that such T-independent stimulation was important mainly during primary response. During infection absence of CD4þ-cells affected mainly the secretion of all IgG subclasses with the exception of IgG3 and IgM. To carry out a preliminary isolation of PSC T-independent antigens we prepared a carbohydrate enriched fraction from protoscolex antigens, using a monoclonal antibody specific for the carbohydrate moiety Gala(1,4)Gal highly expressed in PSC. This fraction was mitogenic for naive mouse splenocytes and was recognized by a high percentage of the specific antibodies secreted by CD4-depleted immunized or infected mice. In summary, these results suggest that E. granulosus protoscoleces contain immunogenic T-independent antigens. Primary antibody responses to protoscolex somatic antigens and the production of IgM and IgG3 in early infection would be mainly stimulated by a T-independent mechanism.

In this work we analysed the modulation of the antibody response to Echinococcus granulosus antigens via anti-idiotype (Ab2) administration. In a first set of experiments, we determined the antibody response to hydatid cyst fluid antigen (HCFA) of mice immunized with Ab2. Results showed that Ab2 elicited anti-HCFA antibodies in mice that had never been exposed to HCFA before. Moreover, that response was characterized by booster effect, avidity maturation and an inverse correlation between avidity and Ab2 dose. We infected these mice and measured the titre and avidity of anti-HCFA antibodies during 250 days of infection. Lack of avidity maturation was the most important feature observed, suggesting that the parasite (either protoscolex or cyst) could modulate the antibody response of its host. In a second set of experiments, we investigated the presence of regulatory anti-idiotypes in the Ab2 preparation. In this context we treated neonates with different doses of Ab2, immunized them with HCFA 12 weeks later and determined their anti-HCFA titres. A specific and Ab2 dose-dependent suppression of the response to HCFA was observed, suggesting the existence of regulatory anti-idiotypes in the Ab2 preparation. Although this Ab2 could be involved in the regulation of the anti-HCFA response, other anti-idiotypes could also be involved. Our results show that it may be possible to improve the avidity of the anti-HCFA response via the administration of the anti-idiotype Ab. However, the live parasite could successfully revert this effect by mechanisms not yet characterized.

Within all the eukaryotic cells there is an important group of biomolecules that has been potentially related to signalling functions: myo-inositol phosphates (InsPs). In nature, the most abundant member of this family is the so called phytate (InsP6, L12−). The importance of the interaction of InsP6 with cations has been recognized during the last few years, and any attempt to know the function of this molecule in eukaryotic cells must include the fact that InsP6 will be strongly associated with simple inorganic or organic ammonium cations. In this work we expand on our earlier findings by studying the systems phen–phytate (phen = 1,10-phenanthroline) and Cu(II)–phen–phytate. The structures of (Hphen)4(H8L)·7.5H2O and [Cu5(H7L)2(H2O)2(phen)5]·23H2O are presented. Solution studies of the binary and ternary systems were also performed by means of potentiometric titrations at 37.0 °C and 0.15 M ionic strength. The work was completed with a molecular modelling study. All this information is essential in the process of gaining reliable structural knowledge about the most important InsP6 species in the in vitro and in vivo experiments, and how these features modulate their probable biological functions.

myo-inositol phosphates are an important group of biomolecules that are present in all eukaryotic cells. The most abundant member of this family in nature is InsP6 (H12L1 ), which interacts strongly with inorganic and organic cations. This interaction is essential for determining the possible functions of this biomolecule. A few crystal structures containing InsP6, and a divalent cation, have been reported showing a great versatility of this bioligand. Ins(1,2,3)P3 (H6L2 ) is another important member of the group, usually thought as a safe cellular iron ligand. No crystal structures showing the interaction of L2 with metal ions can be found in the literature. In this work we characterized by X-ray diffraction two polynuclear complexes, [Cu3(H6L1) (phen)5] (phen = 1,10-phenanthroline) (1) and [Cu2(H2L2 )(H2O)(terpy)2] (terpy = 2,2′:6′,2″-terpyridine) (2). The crystal structure of 2 furnishes, for the first time, a picture of the coordination ability of Ins(1,2,3)P3 against a divalent metal ion. In addition, a PDB survey was performed on all InsPs to frame the coordination modes derived from our small-molecule supramolecular approach within a more realistic biological context.

It is urgent yet challenging to develop new environmentally friendly and cost-effective sources of energy. Molecular solar thermal (MOST) systems for energy capture and storage are a promising option. With this in mind, we have prepared a new water-soluble (pH > 6) norbor nadiene derivative (HNBD1) whose MOST properties are reported here. HNBD1 shows a better matching to the solar spectrum compared to unmodified norbornadiene, with an onset absorbance of λonset = 364 nm. The corresponding quadricyclane photoisomer (HQC1) is quantitatively generated through the light irradiation of HNBD1. In an alkaline aqueous solution, the MOST system consists of the NBD1−/QC1− pair of deprotonated species. QC1− is very stable toward thermal back-conversion to NBD1−; it is absolutely stable at 298 K for three months and shows a marked resistance to temper ature increase (half-life t1/2 = 587 h at 371 K). Yet, it rapidly (t1/2 = 11 min) releases the stored energy in the presence of the Co(II) porphyrin catalyst Co-TPPC (∆Hstorage = 65(2) kJ·mol−1 ). Under the explored conditions, Co-TPPC maintains its catalytic activity for at least 200 turnovers. These results are very promising for the creation of MOST systems that work in water, a very interesting solvent for environmental sustainability, and offer a strong incentive to continue research towards this goal.
