The mixed-chelate copper(II) complexes Casiope&iacute;nas&reg; have been tested in several models in vitro and in vivo, showing promising antitumoral results. However, their mechanism of action remains to be defined. Trying to get a deeper insight into their molecular mode of action, further analyses, including gel electrophoresis, atomic force microscopy and circular dichroism were carried out to study their interaction with DNA and some cytoskeleton proteins. Our results revealed that the interaction of Casiope&iacute;nas triggers DNA cleavage by a free radical mechanism. The tested complexes showed a differential response to reducing and scavenger agents. Differences on target preference were also evident using double stranded oligonucleotides as sequence competitors. Surprisingly, distamycin A, a minor groove binder, enhanced the Casiope&iacute;nas' action on DNA. On the other hand, the tested Casiope&iacute;nas produce strong changes in protein structure of tubulin, integrin and fibronectin. All together these results suggest a multiple mode of action for these metal-based drugs. In addition, since it has been proposed that antitumor drugs efficiently interacting with DNA could also show activity against Trypanosoma cruzi, etiologic agent of Chagas disease, we evaluated the activity of these compounds on this protozoan parasite. Background: Prenylated flavanones represent a structurally diverse class of natural compounds with significant biological potential. Among them, chromene flavanones (CFs) constitute a rare and specialized subgroup with promising therapeutic applications. These molecules have gained attention due to their potential to inhibit xanthine oxidase (XO), a key enzyme involved in oxidative stress-related disorders such as gout and hyperuricemia. Their distinctive structural features, combined with notable bioactivity, make them compelling candidates for further pharmacological exploration. Given their potential relevance, this study focuses on the in vitro and in silico evaluation of three CFs isolated from Dalea boliviana Britton [Fabacea], assessing their capacity to inhibit XO and elucidating key structure–activity relationships (SARs) that contribute to their biological effectiveness. Purpose: This study aims to investigate the in vitro and in silico interactions of the chromene flavanones, namely, (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno- (7,8:2″,3″)-flavanone (1), (2S) 5,2′-dihydroxy-6″,6″-dimethylchromeno-(7,8: 2″,3″)-3′-prenylflavanone (2), and obovatin (3), obtained from D. boliviana, with XO, in order to explore their potential as XO inhibitors and their potential therapeutic applications for hyperuricemic diseases. Material and Methods: XO inhibition by the three chromene flavanones was measured spectroscopically. The relationships between their structures and inhibitory activities were evaluated. Moreover, molecular docking studies were performed to propose the binding modes of the most active natural compounds. Results and discussion: Compounds 1 and 2 exhibited potent inhibition, with IC50 values in the nanomolar range (0.5 ± 0.01 nM and 1.7 ± 0.46 nM, respectively), demonstrating significantly higher activity than allopurinol (AL), the reference inhibitor (IC50 = 247 ± 4 nM). In contrast, compound 3 displayed only weak inhibition. SAR analysis revealed that the presence of a chromene moiety in the A-ring, combined with hydroxyl and prenyl groups in the B-ring, played a crucial role in enhancing inhibitory activity. Molecular docking studies confirmed the strong binding affinities of compounds 1 and 2 within the active site of XO (PDB ID: 3NVY), with binding energies of −6.1687 kcal/mol and −6.7820 kcal/mol, respectively. Key stabilizing interactions involved π–π interactions with Phe914 and hydrogen bonding with residues such as Leu873 and Leu1014. These findings highlight the structural features essential for potent XO inhibition and suggest that chromene flavanones represent a valuable scaffold for the development of novel inhibitors. Further molecular dynamics simulations could provide deeper insights into their stability and interaction dynamics, aiding in the rational design of more effective XO inhibitors. Conclusion: Our findings lead us to propose these chromene flavanones as lead compounds for the design and development of novel XO inhibitors for treating diseases in which exacerbated activity of this enzyme is involved. Aquifers are among the main freshwater sources. The Raig&oacute;n aquifer is susceptible to contamination, mainly by nitrate and pesticides, such as atrazine, due to increasing agricultural activities in the area. The capacity of indigenous bacteria to attenuate nitrate contamination in different wells of this aquifer was assessed by measuring denitrification rates with either acetate plus succinate or nitrate amendments. Denitrification activity in nitrate-amended assays was significantly higher than in unamended assays, particularly in groundwater from wells where nitrate concentration was 33.5 mg L(-1) or lower. Furthermore, groundwater denitrifiers capable of using acetate or succinate as electron donors were isolated, identified by 16S rRNA gene sequencing and evaluated for functional denitrification genes (nirS, nirK and nosZ). Phylogenetic affiliation of 54 isolates showed that all members belonged to nine different genera within the Proteobacteria (Bosea, Ochrobactrum, Azospira, Zoogloea, Acidovorax, Achromobacter, Vogesella, Stenotrophomonas and Pseudomonas). In addition, isolate AR28 that clustered separately from validly described species could potentially belong to a new genus. The majority of the isolates were related to species belonging to previously reported denitrifying genera. However, the phylogeny of the nirS and nosZ genes revealed new sequences of these functional genes. To our knowledge, this is the first isolation and sequencing of the nirS gene from the genus Vogesella, as well as the nosZ gene from the genera Acidovorax and Zoogloea. The results indicated that indigenous bacteria in the Raig&oacute;n aquifer had the capacity to overcome high nitrate contamination and exhibited functional gene diversity. In order to determine the occurrence of AbaR- type genomic island in multidrug resistant Acinetobacter baumannii (MDRAb) strains circulating in Argentina, Uruguay, and Chile, we studied 51 MDRAb isolates recovered from several hospitals over 30 years. AbaR-type genomic resistance islands were found in 36 MDRAb iso- lates since 1986 till now. MLST technique allowed us to identify the presence of four different Clonal Complexes (109, 104, 119, 113) among the positive AbaR-type island positive strains. This is the first description of AbaR-type islands in the CC104 and CC113 that are the most wide- spread Clonal Complexes in Argentina. In addition, PCR mapping exposed different arrays to those previously described, evidencing the plasticity of this island. Our results evidence a widespread distribution of the AbaR-type genomic islands along the time in the MDRAb population, including the epidemic global clone 1 (GC1) as well as different clonal complexes to those already described in the literature. Introducci&oacute;n: la apolipoprote&iacute;na E (ApoE) es un constituyente de las lipoprote&iacute;nas plasm&aacute;ticas con un importante rol en el metabolismo como ligando de receptores de lipoprote&iacute;nas. Las variaciones en el locus de ApoE se vinculan a variaciones en las concentraciones de lipoprote&iacute;nas de baja densidad (LDL) plasm&aacute;ticas. El alelo E4 se ha vinculado a aumento del riesgo cardiovascular, a progresi&oacute;n de la nefropat&iacute;a diab&eacute;tica as&iacute; como a severidad y progresi&oacute;n de neuropat&iacute;a y retinopat&iacute;a diab&eacute;ticas. El alelo E2 se asocia a un efecto final protector para el desarrollo de enfermedad cardiovascular. Objetivos: estudiar la asociaci&oacute;n entre este polimorfismo y las repercusiones macroangiop&aacute;ticas, en especial cardiopat&iacute;a isqu&eacute;mica, y el lipidograma en los pacientes diab&eacute;ticos. Material y m&eacute;todo: se estudiaron 78 pacientes diab&eacute;ticos tipo 2, menores de 85 a&ntilde;os de edad, valorando la asociaci&oacute;n entre el polimorfismo de ApoE y las repercusiones macroangiop&aacute;ticas y microangiop&aacute;ticas de la diabetes, el lipidograma y la edad al diagn&oacute;stico de la diabetes. La determinaci&oacute;n genot&iacute;pica se realiz&oacute; por m&eacute;todos est&aacute;ndar de amplificaci&oacute;n- restricci&oacute;n previo consentimiento informado. Las variables se analizaron utilizando chi cuadrado y prueba de Fisher. Error alfa menor a 0,05. Resultados: la frecuencia de portadores del alelo E4 de la Apo E entre los individuos con complicaciones macroangiop&aacute;ticas (43,5%) fue mayor que entre los que estaban libres de estas complicaciones (26,2%), si bien las diferencias no fueron estad&iacute;sticamente significativas. La presencia de cardiopat&iacute;a isqu&eacute;mica se asoci&oacute; en forma significativa con el alelo E4, 56% versus 25%. La presencia del alelo E4 se asoci&oacute; significativamente con un diagn&oacute;stico de diabetes antes de los 40 a&ntilde;os. Los pacientes con alelos de riesgo presentaron valores de LDL y triglic&eacute;ridos (TG) aumentados. Conclusiones: el alelo E4 se asoci&oacute; a un mayor riesgo de cardiopat&iacute;a isqu&eacute;mica y una edad al diagn&oacute;stico de DM2 menor a 40 a&ntilde;os. Se evidenci&oacute; un efecto marcado del genotipo de ApoE sobre el perfil lip&iacute;dico de los pacientes diab&eacute;ticos tipo 2. BACKGROUND: Release of mercury from fluorescent lamps must be minimized to avoid its hazardous effects on human beings and other living organisms. Because of this, increasing attention is given to the improvement of existing techniques for its recovery as well as the development of new ones. This paper describes the application of heterogeneous photocatalysis for the selective reduction of mercury from fluorescent lamps. The whole process involves mercury chemical extraction using aqueous solutions of sodium hypochlorite. After pH adjustment of the resulting aqueous solution, the photocatalytic reduction of mercury is performed in the presence of titanium dioxide and citric acid as an auxiliary organic agent. RESULTS: Mercury removal from the aqueous solution is higher than 99% and the final residual concentration is 4 ppb. A mixture of different mercury compounds is obtained in solid form deposited on the titanium oxide photocatalyst which can easily be redissolved into a small volume of sodium hypochlorite. Metallic mercury could be obtained from the residual Hg(II) solution by conventional cementation techniques with iron as reducing agent. CONCLUSION: Results show high efficiencies of the photocatalytic technique for recovery and recycling of mercury from fluorescent lamps. Mercury (Hg) is a global pollutant that is released into the environment from geological and anthropogenic sources and its compounds are highly mobile and toxic at trace level. A simple method based on cold vapour atomic absorption spectrometry (CV AAS) for the determination of total mercury in irrigation waters and extractable mercury in paddy soil was validated, using a low-cost sample introduction -1 device. The validated method presented a linear range from 0.2 to 2.0 &micro;g L ; detection limits were 0.03 -1 -1 &micro;g L and 0.012 mg kg (dry basis) for irrigation water and paddy soil respectively. Trueness was evaluated using spiked samples; the mean recovery was 101% (n=8) and 99.4% (n=6) for irrigation water and paddy soil respectively. Precision, expressed as relative standard deviation (RSD), was 9.6% (n=10) and 24% (n=8) for irrigation water and paddy soil respectively. These figures of merit were adequate for the determination of total Hg levels in irrigation waters and extractable Hg in paddy soils established by the regulations. To assess analytical performance several samples from the Eastern zone of Uruguay were analysed. Sample preparation consisted of an acid digestion using an economical pressurized reactor. Hg levels in waters and paddy soils were all below the maximum allowed and therefore were apt for agricultural use. These results are very important not only locally but also for the world since Uruguay is currently among the eight-largest paddy rice and milled rice exporter in the world. This method can be an alternative instead of the more expensive techniques mostly not available in developing countries and very easy to implement in environmental laboratories. Benzenoids are compounds associated with floral and fruity flavours in flowers, fruits and leaves and present a role in hormonal signalling in plants. These molecules are produced by the phenyl ammonia lyase pathway. However, some yeasts can also synthesize them from aromatic amino acids using an alternative pathway that remains unknown. Hanseniaspora vineae can produce benzenoids at levels up to two orders of magnitude higher than Saccharomyces species, so it is a model microorganism for studying benzenoid biosynthesis pathways in yeast. According to their genomes, several enzymes have been proposed to be involved in a mandelate pathway similar to that described for some prokaryotic cells. Among them, the ARO10 gene product could present benzoylformate decarboxylase activity. This enzyme catalyses the decarboxylation of benzoylformate into benzaldehyde at the end of the mandelate pathway in benzyl alcohol formation. Two homologous genes of ARO10 were found in the two sequenced H. vineae strains. In this study, nine other H. vineae strains were analysed to detect the presence and per cent homology of ARO10 sequences by PCR using specific primers designed for this species. Also, the copy number of the genes was estimated by quantitative PCR. To verify the relation of ARO10 with the production of benzyl alcohol during fermentation, a deletion mutant in the ARO10 gene of Saccharomyces cerevisiae was used. BACKGROUND: The search for novel chemical entities targeting essential and parasite-specific pathways is considered a priority for neglected diseases such as trypanosomiasis and leishmaniasis. The thiol-dependent redox metabolism of trypanosomatids relies on bis-glutathionylspermidine [trypanothione, T(SH)2], a low molecular mass cosubstrate absent in the host. In pathogenic trypanosomatids, a single enzyme, trypanothione synthetase (TryS), catalyzes trypanothione biosynthesis, which is indispensable for parasite survival. Thus, TryS qualifies as an attractive drug target candidate. METHODOLOGY/PRINCIPAL FINDING: A library composed of 144 compounds from 7 different families and several singletons was screened against TryS from three major pathogen species (Trypanosoma brucei, Trypanosoma cruzi and Leishmania infantum). The screening conditions were adjusted to the TryS&acute; kinetic parameters and intracellular concentration of substrates corresponding to each trypanosomatid species, and/or to avoid assay interference. The screening assay yielded suitable Z' and signal to noise values (&ge;0.85 and ~3.5, respectively), and high intra-assay reproducibility. Several novel chemical scaffolds were identified as low &mu;M and selective tri-tryp TryS inhibitors. Compounds displaying multi-TryS inhibition (N,N'-bis(3,4-substituted-benzyl) diamine derivatives) and an N5-substituted paullone (MOL2008) halted the proliferation of infective Trypanosoma brucei (EC50 in the nM range) and Leishmania infantum promastigotes (EC50 = 12 &mu;M), respectively. A bis-benzyl diamine derivative and MOL2008 depleted intracellular trypanothione in treated parasites, which confirmed the on-target activity of these compounds. CONCLUSIONS/SIGNIFICANCE: Novel molecular scaffolds with on-target mode of action were identified as hit candidates for TryS inhibition. Due to the remarkable species-specificity exhibited by tri-tryp TryS towards the compounds, future optimization and screening campaigns should aim at designing and detecting, respectively, more potent and broad-range TryS inhibitors. Chagas disease, endemic in 21 countries across Latin America, kills more people in the region each year than any other parasite-borne disease. Therapeutic options have problems ranging from toxicity, poor efficacy, drug resistance and high cost. Thus, cheaper and less toxic treatments are necessary. From our in-house chemical library of agents against Trypanosoma cruzi the most relevant N-oxide-containing heterocycles were selected for mode of action and type of death studies. Also included in these studies were two active nitrofuranes. Epimastigotes of T. cruzi were used as the biological model in this study. The metabolic profile was studied by 1H NMR in association with the MTT assay. Excreted catabolites data, using 1H NMR spectroscopy, showed that most of the studied N-oxides were capable of decreasing both the release of succinate and acetate shedding, the compounds therefore possibly acting on mitochondria. Only quinoxalines and the nitrofurane Nf1 showed significant mitochondrial dehydrogenase inhibitions, but with different dose-time profiles. In the particular case of quinoxaline Qx2 the glucose uptake study revealed that the integrity of some pathways into the glycosome could be affected. Optic, fluorescence (TUNEL and propidium iodide) and transmission electron microscopy (TEM) were employed for type of death studies. These studies were complemented with 1H NMR to visualize mobile lipids. At low concentrations none of the selected compounds showed a positive TUNEL assay. However, both quinoxalines, one furoxan and one benzofuroxan showed a necrotic effect at high concentrations. Curiously, one furoxan, Fx1, one benzofuroxan, Bfx1, and one nitrofurane, Nf1, caused a particular phenotype, with a big cytoplasmatic vacuole being observed while the parasite was still alive. Studies of TEM and employing a protease inhibitor (3-methyladenine) suggested an autophagic phenotype for Bfx1 and Nf1 and a 'BigEye' phenotype for Fx1. African trypanosomiasis is a major problem for human and animal health in endemic countries, where it threatens millions of people and affects economic development. New drugs are needed to overcome the toxicity, administration, low efficacy, and resistance issues of the current chemotherapy. Robust, simple, and economical highthroughput, whole-cell-based assays are required to accelerate the identification of novel chemical entities. With this aim, we generated a bioluminescent cell line of the bloodstream stage of Trypanosoma brucei brucei and established a screening assay. Trypanosomes were stably transfected to constitutively express a thermostable redshifted luciferase. The growth phenotype and drug sensitivity of the reporter cell line were essentially identical to that of the parental cell line. The endogenous luciferase activity, measured by a simple bioluminescence assay, proved to be proportional to parasite number and metabolic status. The assay, optimized to detect highly potent compounds in a 96-well-plate format, was validated by screening a small compound library (inter-assay values for Z' factor and coefficient variation were 0.77 and 5.8%, respectively). With a hit-confirmation ratio of ~97%, the assay was potent enough to identify several hits with EC50 &le; 10 &mu;M. Preliminary tests indicated that the assay can be scaled up to a 384-well-plate format without compromising its robustness. In summary, we have generated reporter trypanosomes and a simple, robust, and affordable bioluminescence screening assay with great potential to speed up the early-phase drug discovery against African trypanosomes. EPR spectroscopy suggests a distorted octahedral geometry for the complex with the dppz ligand acting as bidentate, binding through both nitrogen donor atoms in an axial-equatorial mode. An oxo group, two water molecules and a sulphate donor occupy the remainder coordination positions. The complex, as well as the anti-trypanosomal reference drug Nifurtimox, showed IC(50) values in the muM range against T. cruzi Dm28c strain. In addition the complex exhibited excellent in vitro anti-tumor activity against leukemia (HL-60 cell line) comparable to that of cisplatin, inducing cell death by apoptosis with IC(50) values in the micromolar range. Data from gel electrophoresis and atomic force microscopy indicate that the complex interacts with DNA, suggesting that its mechanism of action may include DNA as a target.
