Exposure to environmental lead (Pb) is a mild risk factor for amyotrophic lateral sclerosis (ALS), a paralytic disease characterized by progressive degeneration of motor neurons. However, recent evidence has paradoxically linked higher Pb levels in ALS patients with longer survival. We investigated the effects of lowlevel Pb exposure on survival of mice expressing the ALS-linked superoxide dismutase-1 G93A mutation (SOD1G93A). SOD1G93A mice exposed to Pb showed longer survival and increased expression of VEGF in the ventral horn associated with reduced astrocytosis. Pretreatment of cultured SOD1G93A astrocytes with low, non toxic Pb concentrations upregulated VEGF expression and significantly abrogated motor neuron loss in coculture, an effect prevented by neutralizing antibodies to VEGF. The actions of Pb on astrocytes might explain its paradoxical slowing of disease progression in SOD1G93A mice and the improved survival of ALS patients. Understanding how Pb stimulates astrocytic VEGF production and reduces neuroinflammation may yield a new therapeutic approach for treating ALS.

“Chemical and microbiological prospecting woodland along the Uruguay River”. Bioprospecting is the search for information from biological species for later use in production processes in various sectors. Examples of this information is contained in the genetic material of all living beings in the chemicals they produce, in their interactions or the knowledge of people who in one way or another, have studied these living beings. This paper presents the results of botanical, chemical and microbiological study of the gallery forest of the Uruguay River in the area of the Queguay river. This ecosystem presents great biodiversity (comprising about 200 species) and unusual features in the country given its subtropical character. We studied 75 extracts from 31 species belonging to fifteen families covering therefore a large biodiversity

A summary of the resource.
The purpose of the present research was further investigate the reproducibility of check-all-that-apply (CATA) questions for sensory product characterization. Evaluations obtained when such questions are used by consumers are rarely replicated and therefore reproducibility of the data may be at risk. Results from the present work, which included five studies, each with 100–200 consumers across a range of product categories, revealed that sensory product characterizations obtained using CATA questions with consumers are highly reproducible. Hence, the research confirms previous research by Jaeger, Chheang, et al. (2013) and extends it to the use of CATA terms in randomised presentation order as has been recommended to avoid satisficing response behavior. In the future, if CATA studies are conducted without replication and researches seek to examine the reliability of CATA data, the use of a posterior bootstrapping re-sampling approach is suggested.

their free radical-trapping properties. The physicochemical characterization by electron paramagnetic resonance (EPR) demonstrated its capability to trap and stabilize oxygen-, carbon-, sulfur-, and nitrogen- centered free radicals. The 4-furoxanyl nitrone 3 (FxBN), a(Z)-(3-methylfuroxan-4-yl)-N-tert-butylnitrone, showed appropriate solubility in aqueous solution and taking into account that this physicochemical property is very important for biological applications, we studied it deeply in terms of its trapping and kinetic behaviors. For this, kinetic studies of the hydroxyl adduct decay gave rate constants kST of 1.22 � 1010 dm3 mol�1 s�1 and half-live up to 7200 s at physiological pH, without any artifactual signals. The ability of FxBN to directly traps and stabilizes superoxide free radical, with a half-life of 1620 s at physiological pH, was also demonstrated. Besides, FxBN-hydroxyl and -superoxide adducts exhibited distinct and characteristic EPR spectral patterns. Finally, we confirmed the ability of FxBN to act as spin trap in a specific biological system, that is, in the free radical production of experimental anti-trypanosomatid drugs using Trypanosoma cruzi microsomes as biological system. Moreover, previous observations of low FxBN toxicity transform it in a good candidate for in vivo spin trapping

The current research explored the possibility of using attribute ratings as a variant (RATA: rate-allthat- apply) to CATA questions (check-all-that-apply), in order to improve sample description and discrimination and to engage participants in greater cognitive processing. The RATA question variant was implemented by asking participants, for the terms they ticked as ‘‘apply’’, to indicate intensity (using a 3-pt scale with anchors ‘low’, ‘medium’ or ‘high’) or rate applicability (using a 5-pt scale anchored at ‘slightly applicable’ and ‘very applicable’). A total of four studies with 328 consumers were conducted. Studies 1–3 involved the consumption of products (milk desserts, bread and gummy lollies), whereas Study 4 was performed with yogurt labels. A between-subjects design was used in all studies to compare product characterizations from CATA and RATA questions. Across the four studies, compared to the simple CATA questions, the RATA variant led to an increase in the total number of selected terms and a small increase in the percentage of terms for which significant differences among samples were identified. Although the stability of sample and term configurations from CATA and RATA questions, calculated using a bootstrapping re-sampling approach, were similar, for two of the four studies RATA questions provided more stable sample and term configurations. Results from the present work reveal the potential of intensity-based CATA variants with consumers for sensory product characterization, but also suggest that these may be study and sample specific.

A summary of the resource.
Interest in understanding how foods affect consumers’ perceived wellbeing has grown in the last decade due to the increasing need to modify dietary patterns. Considering that wellbeing is a broad concept that lacks of a unique definition, in order to use and measure this concept it is necessary to explore how consumers understand it, particularly in the context of food consumption. The aim of the present work was to investigate consumers’ perception of wellbeing in a food-related context using an exploratory qualitative approach. A study was carried out with 120 Uruguayan participants using three qualitative techniques: word association, open-ended questions and free listing. Wellbeing in a food-related context was strongly associated with physical health. The expected effects of foods on wellbeing were mainly related to non-communicable diseases such as high cholesterol levels, hypertension, and cardiovascular diseases. However, hedonic and emotional aspects of food consumption were also salient for consumers perceived wellbeing. The information gathered in this study can contribute to the development of scales for measuring consumer perceived wellbeing when consuming foods.

A summary of the resource.
Sensory characterization provides a complete description of the sensory characteristics of food products. Descriptive Analysis is the most common methodology for this purpose. However, due to the cost and time associated with its application, several alternative methods have been recently developed. These methods do not require training, can be performed by trained assessors or consumers, and have been reported to be a good option when quick information about the sensory characteristics of a set of products is needed. There are basically four types of methodologies: methodologies based on the evaluation of specifi c attributes, on global differences among products, on the comparison with references, and methodologies that provide a verbal description of the products. In the present chapter these novel methodologies for sensory characterization of food products are described. Advantages and disadvantages of each methodology are discussed and recommendations for their application are provided.

Las levaduras juegan un papel fundamental en la transformación del jugo de uva en vino resaltando principalmente la expresión de los aromas de las variedades utilizadas y aportando aromas de fermentación que pueden enriquecer y resaltar el vino obtenido en concentración y complejidad sensorial. Este trabajo propone la aplicación de nuevas cepas no-Saccharomyces (no-Sac) nativas, aisladas en Uruguay, como pretratamiento de uvas tintas previo a la inoculación de una cepa comercial, con el objetivo de enriquecer con mayor complejidad aromática vinos de alta gama. Con este objetivo, se realizaron aislamientos de cepas nativas de diferentes uvas y mostos tintos. Como método de selección se evaluó la capacidad fermentativa y el aporte aromático de cada cepa en medios de bajo nitrógeno (menos de 150mgN/L). El resultado de esta selección fue la elección de 4 cepas no-Sac que presentaban además de buenas condiciones de vinificación, ausencia de defectos aromáticos pero con un aporte destacable sobre las características sensoriales. Estas cepas fueron evaluadas en cultivos mixtos con una cepa comercial de Saccharomyces (L254) para simular las condiciones de fermentación a nivel de bodega. Las fermentaciones fueron analizadas por GC-MS para caracterizar las cepas utilizadas en cultivos mixtos en comparación con la cepa Saccharomyces pura. Finalmente se seleccionó una cepa no-Sac (02/5A) como la más propicia para realizar un escalado a nivel industrial que fue clasificada como Hanseniaspora vineae por análisis del ADN ribosomal en las regiones D1/D2. El secado a nivel piloto de la cepa y su aplicación a nivel de bodega fueron considerados óptimos en cuanto a su viabilidad celular al momento de la inoculación. La aplicación de levaduras de este tipo asegura el desarrollo de fermentaciones con mayor biodiversidad sin riesgos de la aparición de vinos con defectos, uno de los objetivos de la Enología de Mínima Intervención (EMI).

Mate finding in many spider species is mediated, at least in part, by chemical cues. Although few have been characterized, most spider sex pheromones seem to be associated with the silk threads of adult females, attracting and/or triggering sexual behaviors in males. Schizocosa malitiosa (Araneae: Lycosidae) is a wolf spider common in dry environments in Southern Uruguay. Here, we report evidence for the occurrence of a female sex pheromone in the silk of virgin S. malitiosa females. The pheromone elicits typical courtship displays by conspecific males (palpal drumming and leg shaking), when it is applied to an artificial substrate. We also showed that this pheromone is quickly inactivated under field conditions, possibly due to the effect of dew, and that it is readily extracted from the silk by water, which renders the silk threads inactive. Preliminary chemical analyses by mass spectrometry suggest that the pheromone is a low molecular weight, highly oxygenated polar compound, present in a high concentration only in older virgin females. Quick inactivation in the field of the pheromone would allow males to discriminate between recent and old cues, thus facilitating mate searching and sexual meeting, by preventing males from following old cues for wandering females that could be distant in space and/or time.

We have investigated important intermediates and key transition states of the organocatalyzed Knoevenagel condensation using density functional theory and two different basis sets (6-31 G(d,p) and 6-311++G(2df,2pd)), both in gas phase and simulating the bulk solvent (pyridine) using the PCM method. Calculated structures for reactants, intermediates, and key transition states suggest that the secondary amine catalyst is essential, both for activating the aldehyde for nucleophilic attack, and in the possible decarboxylation pathways. The calculated results are shown to agree with available experimental information. On the basis of the results obtained, the studied mechanism may be important in the understanding of vinylphenol production during malting and brewing of wheat and barley grains.

Chagas disease is caused by the trypanosomatid parasite Trypanosoma cruzi and threatens millions of lives in South America. As other neglected diseases there is almost no research and development effort by the pharmaceutical industry and the treatment relies on two drugs, Nifurtimox and Benznidazole, discovered empirically more than three decades ago. Nifurtimox, a nitrofurane derivative, is believed to exert its biological activity through the bioreduction of the nitro-group to a nitro-anion radical which undergoes redox-cycling with molecular oxygen. This hypothesis is generally accepted, although arguments against it have been presented. In the present work we studied the ability of Nifurtimox and five N-oxide-containing heterocycles to induce oxidative stress in T. cruzi. N-Oxide-containing heterocycles represent a promising group of new trypanosomicidal agents and their mode of action is not completely elucidated. The results here obtained argue against the oxidative stress hypothesis almost for all the studied compounds, including Nifurtimox. A significant reduction in the level of parasitic low-molecular-weight thiols was observed after Nifurtimox treatment; however, it was not linked to the production of reactive oxidant species. Besides, redox-cycling is only observed at high Nifurtimox concentrations (>400 mM), two orders ofmagnitude higher than the concentration required for anti-proliferative activity (5 mM). Our results indicate that an increase in oxidative stress is not the main mechanism of action of Nifurtimox. Among the studied N-oxide-containing heterocycles, benzofuroxan derivatives strongly inhibited parasite dehydrogenase activity and affected mitochondrial membrane potential. The indazole derivative raised intracellular oxidants production, but it was the least effective as anti-T. cruzi.

BACKGROUND: Release of mercury from fluorescent lamps must be minimized to avoid its hazardous effects on human beings and other living organisms. Because of this, increasing attention is given to the improvement of existing techniques for its recovery as well as the development of new ones. This paper describes the application of heterogeneous photocatalysis for the selective reduction of mercury from fluorescent lamps. The whole process involves mercury chemical extraction using aqueous solutions of sodium hypochlorite. After pH adjustment of the resulting aqueous solution, the photocatalytic reduction of mercury is performed in the presence of titanium dioxide and citric acid as an auxiliary organic agent. RESULTS: Mercury removal from the aqueous solution is higher than 99% and the final residual concentration is 4 ppb. A mixture of different mercury compounds is obtained in solid form deposited on the titanium oxide photocatalyst which can easily be redissolved into a small volume of sodium hypochlorite. Metallic mercury could be obtained from the residual Hg(II) solution by conventional cementation techniques with iron as reducing agent. CONCLUSION: Results show high efficiencies of the photocatalytic technique for recovery and recycling of mercury from fluorescent lamps.

the formation of ultimate carcinogens. CPA could act through various molecular mechanisms, for example by interfering with the action of procarcinogen. This could be attained by increasing the phase II enzymes levels of quinone reductase (QR) and glutathione S-transferase (GST). New flavonoids, especially chalcones, have been identified as in vivo monofunctional phase II enzymes inducers. Oral administration of chalcone, 4, and both p-methoxy-substituted chalcones, 6 and 14, increased hepatic QR activity with concomitant decrease in CYP1A1 activity, a member of the most important group of phase I enzymes cytochrome P450. Among them, 4 also increased GST activity. While p-bromo-substituted chalcone 8 was the best inducer of QR it decreased hepatic GST expression and cytochrome P450, being the most effective decreasing cytochrome P450-expression. Thienyl-chalcone 20 being the bioisostere of chalcone 4 did not display the same in vivo profile in the phase I level modification. As chalcone 4 its bioisostere, chalcone 20, displayed low DNA strand breakage and absence of mutagenicity. Also, in our preliminary in vivo tumourigenesis/chemopreventive and acute-toxicity studies, chalcones 4, 6 and 8 showed the best behaviours as CPA justifying additional studies that are ongoing

The environmental impact of rice agriculture is poorly studied in developing countries, mainly due to limitations of the analytical capacity. Here, we report the development of a clomazone enzymelinked immunosorbent assay as a fast and cost-effective tool to monitor the dissipation of this herbicide along the harvest. Antibodies were prepared using different strategies of hapten conjugation, and the best hapten/antibody pair was selected. It proved to be a reliable tool to measure the herbicide in the 2.0-20 ng/mL range in field samples, with excellent correlation with highperformance liquid chromatography results. The assay was used to study the dissipation of the herbicide in the floodwater of experimental rice paddies in Uruguay. Large differences in the residual amounts of herbicide were observed depending on the flooding practices. Because of its robustness and simplicity, the assay may be useful to delineate and monitor management practices that can contribute to minimizing the release of the herbicide in the environment.

2-Thiobarbituric acid (TBA) coated gold nanoparticles (average diameter ) 5.90 nm) were produced and studied by several experimental and theoretical methods. As part of this study, the molecular structure of TBA tautomers in the solid, in polar solutions, and adsorbed onto gold nanoparticles was studied. The resolution of this complicated system (10 possible isomers) was accomplished with the aid of experimental (IR, UVvis, and NMR) and theoretical (DFT and MP2) methods. The general conclusion is that there are two preeminent isomers, N1 and N10, with different stabilities in different media. N1, the keto-thione tautomer, is the most stable in gas phase (¢Go 298 � 8-9 kcal/mol lower than the second-most stable isomer, depending on the method of calculation used). However, experimental spectroscopic data supported by the theoretical calculations strongly suggest an equilibrium between the tautomers N1 and N10 in methanol solution, where enolization of one keto group is produced by proton transfer from the methylene group, which is more acidic than the NH groups. With the use of the polarizable continuum method for simulating solvents, N10 is predicted to be even more stable than N1 by ¢Go 298 � 1 kcal/mol in methanol. On the other hand, the IR spectrum of the solid can be best explained by assuming that only N10 is present, a fact also supported by the observation that the IR spectrum of TBA absorbed onto gold nanoparticles can be explained by a larger ratio of [N10]/ [N1] than that present in methanolic solution. Isomerization of N1H N10 can be explained by intervention of the solvent, proceeding faster in methanol solutions than in DMSO, where it is nevertheless observed after a time, according to the 13C NMR spectra. Our experiments support absorption of TBA onto gold nanoparticles through S-Au and N-Au interactions, with the preeminence of a N10-like enol structure. The experiments also demonstrate that the synthesized TBA-coated gold nanoparticles can autoassociate by hydrogen bonding to form larger structures. This same H-bonding capacity also assures that these coated nanoparticles act as thistles toward proteins in solution, binding them strongly, presumably not by chemical reaction but by a network of hydrogen bonds.

The spectroscopic behavior of 2-benzyl-2-methyl-2H-benzimidazole 1,3-dioxide derivatives in solution was studied in terms of electronic and nuclear magnetic resonance (1H and 13C NMR) techniques. The experimental spectra were compared to the theoretical ones, obtained at DFT level, proving that the compounds adopt in solution a bird-like conformational distribution. Also, theoretically this conformational distribution resulted the most stable in gas phase. Infrared spectroscopy was used to study solid state behavior identifying experimentally the N–O stretching near to 1380, 1365 and 1225 cm−1 and the vibrational benzimidazole skeleton near to 1610 and 1590 cm−1. The vibrational spectrum was satisfactorily described by DFT calculations funding the N–O stretching as a coupled vibration near to 1470, 1350 and 1285 cm−1. The fragmentation that takes place in mass spectrometry was assigned for all of the new derivatives

Application of passive modified atmosphere packaging (MAP) for shelf life extension of enzymatically and nonenzymatically peeled orange segments was evaluated. Oranges (Citrus sinensis cv. Valencia) were washed, sanitized and peeled by applying vacuum pulses only (vacuum peeling) and vacuum pulses followed by incubation in enzymatic solution (enzymatic peeling). Segments were packaged in polypropylene film and stored at 7C for 16 days. Package atmosphere composition, weight loss, respiration rate, color, instrumental texture, pH, soluble solids, sensory attributes, and bacterial and fungal growth were evaluated during storage. Microbiological limits (cfu/g > 106) were reached by enzymatically and vacuum-peeled segments on days 12 and 8, respectively. Integrity and characteristic odor had significantly changed by this day. Vacuum-peeled segments presented better texture attributes and integrity (50% higher scores in sensory evaluation). Both types of segments benefited from MAP and are an attractive alternative for commercialization of ready-to-eat oranges. Practical Applications The information presented in this paper can assist fresh-cut producers in developing ready-to-eat oranges. Quality of segments obtained with two peeling methods was evaluated, and potentials and limitations of the segments obtained by each process were pointed out. Passive MAP in polypropylene at a reasonable storage temperature (7C) resulted to be an inexpensive technology that can extend segment shelf life in eventual cold storage of orange segments.

Characterization studies were performed to a proteolytic extract, prepared from latex of Ficus luschnathiana (Miq.) Miq., a Moraceae family tree. The mechanism of action was determined, corresponding to serine proteinases behaviour. Studies of optimum pH and temperature showed a maximum catalytic activity at pH 8.0 and 75°C, respectively. The proteolytic activity exhibited good stability when incubated at different pH and temperature conditions. At 7°C and pH 7.5, the extract kept a 100% of the initial activity for 5 months. When comparing the effect of pH on the stability, maximum values were obtained at pH 6.5, with 98% of the initial activity remaining after 5 h of incubation at 60°C. After 1 min at 100°C total inactivation occurred. Regarding the potential use in the dairy industry, milk clotting activity was determined obtaining a very effective coagulant action. Analysis protocols which included molecular exclusion chromatography, electrophoretic methods and mass spectrometry revealed a complex protein profile. It was found that the molecules responsible for the proteolytic activity of the extract were several proteins of different molecular weight and isoelectric points, most of which still retained the activity in presence of SDS and reducing agents. These proteic components displayed a pH-dependant tendency to form aggregates. Using the proteolytic activity of the crude extract under controlled hydrolysis conditions on whey proteins, yielded peptidic fractions with antimicrobial activity against a S. aureus strain. It was found that fractions of peptide concentration of 0.96 μmol mL-1 inhibited growth by 94%.

Casiopeínas® are mixed-chelate copper complexes with antitumor tested potential. Their activity, both in vitro and in vivo, as antiproliferative, cytotoxic, and genotoxic drugs has been assessed. Biological results of these copper compounds have deserved some of them entering clinical trials. Significant efforts have been devoted to the in-depth identification of their mechanism of action. Using gel electrophoresis analysis, we have previously shown that the interaction of the Casiopeínas® Cas II-gly, [Cu(4,7-dimethyl-1,10- phenanthroline)(glycinate)]NO3 with DNA, triggers the cleavage of the biomolecule by a free radical mechanism. In this work, we further study the behavior of different complexes of the same Casiopeínas® series also including glycinate as co-ligand {Cas VI-gly (5,6 dimethyl-1,10- phenanthroline glycinato copper(II) nitrate), Cas VII-gly (1,10-phenanthroline glycinato copper(II) nitrate), and Cas IX-gly (2,2′-bipyridine glycinato copper(II) nitrate)} and of a Casiopeínas® with a different co-ligand (Cas III-Cs; 4,7-dimethyl- 1,10-phenanthroline salicylaldehydato-copper(II) nitrate). While all of them produce DNA degradation, the performance in the presence of a radical scavenger suggests the existence of differences in their mechanism of interaction with DNA.

Atrazine is one of the most persistent herbicides that contaminate watercourses. However, its biodegradation has been scarcely studied in aquatic ecosystems. The present work studies the potential of bacteria collected from freshwater and sludges of three water potabilization plants to degrade atrazine. Bacteria from all samples, rarely exposed to the herbicide, were able to degrade atrazine as the sole N source in microcosms assays. Five bacterial consortia that degraded consistently 1 or 25 mg L�1 of atrazine within 48 h were established. T-RFLP analysis showed that three ribotypes accounting for more than 50% of the bacterial communities were common to the five consortia and the complete set of atrazine-degrading genes trzN, atzB, atzC and trzD was detected on them. Two consortia from the same origin but incubated with different atrazine concentrations (SAC 1 and SAC 25) contain isolates affiliated to Sinorhizobium sp. and Pseudomonas citronellolis, but Arthrobacter uraefaciens and Achromobacter xylosoxidans were specific of SAC 1 and SAC 25, respectively. These two consortia degraded atrazine in the presence of nitrate and grew in cyanuric acid. This paper describes the great potential for atrazine degradation by bacteria present in aquatic environments which can be exploited to design bioremediation processes.

The antifungal activity of a library of 32 cyclitols and derivatives, including 6 previously unreported cyclitol amino acid conjugates, was studied against the clinically important yeasts Candida albicans, Candida tropicalis and Cryptococcus neoformans along with Saccharomyces cerevisiae. Bioautography followed by standardized microbroth dilution methods were used and allowed to identify an azidoinositol glycoside (11) and an azidoconduritol linked to an aromatic aldehyde (18) as promising compounds. The results suggest the relevance of exploring synthetic cyclitolic structures as potential antifungal leaders.

Chagas disease, endemic in 21 countries across Latin America, kills more people in the region each year than any other parasite-borne disease. Therapeutic options have problems ranging from toxicity, poor efficacy, drug resistance and high cost. Thus, cheaper and less toxic treatments are necessary. From our in-house chemical library of agents against Trypanosoma cruzi the most relevant N-oxide-containing heterocycles were selected for mode of action and type of death studies. Also included in these studies were two active nitrofuranes. Epimastigotes of T. cruzi were used as the biological model in this study. The metabolic profile was studied by 1H NMR in association with the MTT assay. Excreted catabolites data, using 1H NMR spectroscopy, showed that most of the studied N-oxides were capable of decreasing both the release of succinate and acetate shedding, the compounds therefore possibly acting on mitochondria. Only quinoxalines and the nitrofurane Nf1 showed significant mitochondrial dehydrogenase inhibitions, but with different dose-time profiles. In the particular case of quinoxaline Qx2 the glucose uptake study revealed that the integrity of some pathways into the glycosome could be affected. Optic, fluorescence (TUNEL and propidium iodide) and transmission electron microscopy (TEM) were employed for type of death studies. These studies were complemented with 1H NMR to visualize mobile lipids. At low concentrations none of the selected compounds showed a positive TUNEL assay. However, both quinoxalines, one furoxan and one benzofuroxan showed a necrotic effect at high concentrations. Curiously, one furoxan, Fx1, one benzofuroxan, Bfx1, and one nitrofurane, Nf1, caused a particular phenotype, with a big cytoplasmatic vacuole being observed while the parasite was still alive. Studies of TEM and employing a protease inhibitor (3-methyladenine) suggested an autophagic phenotype for Bfx1 and Nf1 and a 'BigEye' phenotype for Fx1.
