A summary of the resource.
We present an ab initiosDFT/GGAsstudy on the structural and electronic distortions of modified graphene by the creation of vacancies, the inclusion of boron atoms, and the coexistence of both, by means of total energy and band structure calculations. In the case of coexistence of boron atoms and vacancy, the modified graphene presents spin polarization only when B atoms locate far from vacancy. Thus, when a boron atom fills single and divacancies, it suppresses the spin polarization of the charge density. In particular, when B atoms fill a divacancy, a new type of rearrangement occurs, where a stable BC4 unit is formed inducing important out-of-plane distortions to graphene. All these findings suggest that new chemical modifications to graphene and new types of vacancies can be used to modify its electronic properties.

A summary of the resource.
The carotenoid content and profile evolution of Uruguayan Tannat grapevine fruit was studied. Two differently managed vineyards exposed to different sunlight and temperature conditions were sampled over two successive ripening periods. Seven carotenoids were identified by HPLC-DAD: neoxanthin, violaxanthin, luteoxanthin, lutein 5,6-epoxide, flavoxanthin, lutein, and β-carotene. Irrespective of species, the highest carotenoid concentrations were found at initial sampling, immediately decreasing sharply to a minimum at harvest. However, a different variation was found for each carotenoid. Whereas significant carotenoid degradation was observed in both vineyards, the effect was greater in grapes of vines growing in mulched soil. The evolution of carotenoid breakdown products was also studied by GC–MS–SIM. In fruit from the mulched vineyard, the norisoprenoid content increased noticeably (by 85% of the initial concentration) over the ripening season, whereas no such increase was found in fruit from the conventional vineyard.

An NADPH-dependent enzyme that reduces ethyl 2-methylacetoacetate stereoselectively to ethyl (2 R)-methyl-(3 S)-hydroxybutanoate was purified 730-fold from Escherichia coli. The N-terminal amino acid sequence data obtained from the purified reductase were used to search the E. coli genome, and a single match was found at the start of the yqhE open reading frame. The YqhE protein had been identified previously by Yum et al. as a 2,5-diketo-d-gluconate reductase on the basis of sequence similarity to other bacterial homologues [Yum, D.-Y.; Lee, B.-Y.; Pan, J.-G. Appl.Environ. Microbiol.1999, 65, 3341–3346]; however, it had not been examined for β-keto ester reductions. Our results thus link a key enzyme in the microbial production of ascorbate with stereoselective β-keto ester reductions, two important fields in biocatalysis. The purified YqhE reductase accepts ethyl acetoacetate and a variety of 2-substituted derivatives, and its sequence is similar to other aldose reductase superfamily members that also reduce α-substituted β-keto esters to syn-(2 R,3 S) alcohols.<br /><br />

A summary of the resource.
The volatile components isolated from the root of Angelica (Angelica archangelica L.) cultivated in southern Brazil have been studied. The essential oil was obtained by hydrodistillation; the volatile concentrate was obtained by supercritical fluid extraction (SFE). SFE was conducted at 40°C, 50°C and 60°C, at pressures ranging from 90–120 bar. The identification of the components was completed using capillary gas chromatography and mass spectrometry based on the retention time on methyl silicone phases. Chemical analysis revealed that although roughly the same compounds were extracted, the two oils had a widely different percentage composition. The supercritical CO2 extraction exhibited a higher number and concentration of oxygenated compounds.<br /><br />

Several breeding programs are under way to introduce resistance to bacterial wilt caused by Ralstonia solanacearum in solanaceous crops. The lack of screening methods allowing easy measurement of pathogen colonization and the inability to detect latent (i.e. symptomless) infections are major limitations when evaluating resistance to this disease in plant germplasm. We describe a new method to study the interaction between R. solanacearum and potato germplasm that overcomes these restrictions. The R. solanacearum strain UY031 was genetically modified to constitutively generate light from a synthetic luxCDABE operon stably inserted in its chromosome. Colonization of this reporter strain on different potato accessions was followed using life imaging. Bacterial detection in planta by this non-disruptive system correlated with the development of wilting symptoms. In addition, we demonstrated that quantitative detection of the recombinant strain using a luminometer can identify latent infections on symptomless potato plants. We have developed a novel, unsophisticated and accurate method for high-throughput evaluation of pathogen colonization in plant populations. We applied this method to compare the behavior of potato accessions with contrasting resistance to R. solanacearum. This new system will be especially useful to detect latency in symptomless parental lines before their inclusion in long-term breeding programs for disease resistance.

The dynamic sensory profile of sweeteners is of great importance for the industry during the development of low-calorie products. In the present work the influence of sucrose replacement by low-calorie sweeteners on the dynamic sensory profile of orange juice was evaluated using multiple-sip Temporal Dominance of Sensations (TDS). Seven orange juices with equivalent sweet concentrations of different sweeteners (sucrose, sucralose, thaumatin, and four samples of stevia) were formulated. A sensory panel of 12 trained assessors evaluated the samples using TDS over three consecutive sips, each lasting 20 s. TDS enabled the identification of differences in the dynamics of the sensory characteristics of the juices formulated with different sweeteners, which had not been identified using static measurements. Considering the dominance of the evaluated sensory characteristics over the three sips, the juice with sucralose showed the most similar sensory characteristics to the juice sweetened with sucrose, providing similar sweetness dominance over time without providing negative sensory characteristics. On the other hand, samples of stevia were characterized by a high dominance of bitterness and off-flavor. The consideration of multiple sips enabled the identification of changes in the temporal profile of the juices with repeated rapid ingestions. Results from the present work suggest that multiple sip TDS seems to be an interesting tool for the evaluation of sweeteners, the design of mixtures of sweeteners and the development of low-calorie products.

The cost of starting materials for the production of biodiesel is typically 75 % of the final retail price. Oils previously used for frying, waste frying oils (WFO), are a very suitable resource. Repetitive use of oil for frying foods involves high temperature, moisture and aeration for extended periods. The most important deterioration processes triggered by these conditions are hydrolysis and oxidation. In this study, 24 WFO samples of different origins were analyzed and classified as potential starting materials for biodiesel production using three quality parameters representing the main factors that affect the conversion of WFO. These parameters were: acid value, content of polar compounds and content of polymers, which varied in the ranges from 0.2 to 7.6, 14.9 to 43.2 and 0.9 to 15.2 %, respectively. Ester content obtained using conventional transesterification (TE) for WFO conversion decreased with increased levels of WFO deterioration determined by any of the three parameters noted above. TE products obtained had ester content between 81.4 and 95.7 %. Total ester content of a WFO sample with relatively low %AV could be increased to 96.5 % using a two-stage base catalysis TE. Finally, conversion of WFO samples resulted in ester contents of 89.0 and 91.3 %, respectively, when transesterified by conventional TE. After blending up to 10 % with refined oil, the ester content achieved was near 96.5 %. Thus, the blending represents an alternative for obtaining a product with suitable ester content.

The word association technique was used to study the perception of Uruguayan consumers with regards to goat cheese and compare it with the perception of other cheeses sold in this market. For this, 149 subjects were presented with five different questionnaires to complete with their perceptions of the following cheeses Colonia, Parmesan, Camembert, goat and sheep cheese. The instructions were as follows: “Please, write down all the words, descriptions, associations, thoughts or feelings that come to your mind in connection with these products”. Afterwards, these words were grouped into 16 categories: animal, color, like, dislike, eyes (of the cheese), feelings, flavor, knowledge/consumption, meals and food, occasions, origin, price, quality, rind, smell and texture. Goat cheese was highly associated with factors related to flavor (51.7%) and texture (34.2%). Participants used a wide range of terms, many contradictory (sour, bitter, strong, tasty, mild, bland, creamy, pasty, soft and hard) to describe this cheese. Such diversity in responses may reflect the difference among brands in Uruguay or lack of knowledge on goat cheese. There was also variation in perception in color categories (27.5%), dislike (17.4%) and smell (21.5%). The dislike for this cheese reflected in the choice of words such as “stinky” and “smelly” is mainly associated with negative connotations for smell (fetid and malodorous). Three groups of consumers were identified based on the frequency of consumption of goat cheese. When frequency of consumption decreased, the percentage of mention of the categories like, meals and food and flavor also decreased, whereas the frequency of mention of the categories animal, knowledge/consumption and dislike increased. These results suggest that word association might be an interesting, easy and quick qualitative technique to gather useful information about consumer perceptions of cheese products.

The popularity of projective mapping with consumers for sensory characterization has markedly increased in the last 5 years. To have confidence in this methodology, it is necessary to ensure that a similar product profile would emerge if the test was repeated. Also, deciding whether the study should be replicated or not is a key issue in test implementation. In this context, the aim of the present work was to evaluate global and individual reproducibility of projective mapping for sensory characterization with consumers and to evaluate the influence of the size of difference among samples. Six consumer studies were conducted using a test–retest paradigm. In each study, responses from the same group of consumers to the same sample set in two different sessions were compared. Across the six studies, individual reproducibility tended to be low. However, the RV coefficients of consensus sample configurations between sessions were higher than 0.75, suggesting that test–retest reproducibility of projective mapping with consumers proved to be relatively high.

Projective mapping for sensory characterisation with consumers has been used for a relatively short period of time, which suggests that the development of guidelines regarding best practices is strongly needed. The present work aims to provide an insight on the minimum number of consumers needed to reach stable sample configurations. Data sets from 21 different consumer studies, differing in product category, number of samples and degree of difference among them, were used to evaluate the influence of the number of consumers on the stability of sample configurations by means of a resampling approach. For each study, 1000 random subsets of different number of consumers were generated from the original data set. For each virtual panel, sample configurations were obtained using Multiple Factor Analysis. The agreement between them and the reference configurations (obtained with all the consumers) was evaluated through the RV coefficient, using the first two and the first four dimensions of the MFA. Results showed that the stability of sample configuration clearly depended on the degree of difference and type of differences among samples and the number of samples in the dataset. Across the 21 data sets analysed, results suggested that when working with widely different samples, 50 consumers seems as a safe recommendation of minimum consumer panel size to obtain reliable results with projective mapping. However, after any characterisation by projective mapping is completed, it is highly recommended to check, a posteriori, the reliability of the sample space configuration using a bootstrapping procedure.

BACKGROUND Strawberry breeding programs should be able to rely on valid and simple methodologies for evaluating sensory quality of new cultivars. In this context, the aim of the present work was to evaluate the applicability of a simple sensory methodology, named projective mapping, to evaluate the sensory characteristics of strawberry cultivars and advanced selections throughout the harvest season. Three strawberry cultivars and two advanced selections were evaluated by an untrained assessor panel using projective mapping in three different moments of the harvest season: June (early season), August (mid-season) and September (peak harvest). Instrumental measurements were also performed. RESULTS Projective mapping enabled the identification of the main sensory characteristics of the strawberry cultivars and advanced selections, as well as the similarities and differences among them. Sensory characteristics of the five evaluated strawberry cultivars and advanced selections largely varied throughout the harvest season. Simple instrumental measurements were not able to predict the sensory characteristics of the strawberry cultivars, suggesting the importance of sensory methodologies for the evaluation of new cultivars in breeding programs. CONCLUSIONS Projective mapping consisted of a quick alternative for the evaluation of new cultivars relative to standard commercial cultivars. Its main advantage is that a large number of cultivars can be screened with minimal investment of time and resources. © 2013 Society of Chemical Industry

Two new isomers trans,mer-[RuIICl2(bpea)(DMSO)], 2a, and cis,fac-[RuIICl2(bpea)(DMSO)], 2b, (bpea ) N,N-bis- (2-pyridylmethyl)ethylamine), as well as the bis-DMSO complex trans,fac-[RuIICl(bpea)(DMSO)2]Cl, 3, have been synthesized and characterized by cyclic voltammetry and UV-vis and 1D and 2D NMR spectroscopy in solution. Their solid-state structure has also been solved by means of single-crystal X-ray diffraction analysis. All the three complexes display a ruthenium metal center possessing a distorted-octahedral type of coordination, where the bpea ligand is coordinated in a meridional fashion in 2a and in a facial fashion in 2b and 3. The isomer 2a is the kinetically favored and thus can be thermally converted into 2b, that is the thermodynamically favored one. A thorough kinetic analysis strongly points toward a dissociative mechanism, where in the first step a chloro ligand is removed from the metal coordination sphere, followed by a geometric rearrangement before the chloro ligand coordinates again, generating the final complex. DFT calculations agree with the experimental data for the proposed mechanism and allow us to further characterize the mechanism of the 2a f 2b rearrangement by obtaining the intermediates and transition state.

Metal complexes that establish interactions with DNA are being studied not only because of their potential use as therapeutic agents but also as tools for biochemistry and molecular biology. Searching for drugs with anti-trypanosome activity, we previously synthesized a series of ruthenium mixed ligand dimethyl sulfoxide complexes of the type [RuIICl2(DMSO)2L], where L is 5-nitrofurylsemicarbazone derivatives and DMSO is dimethyl sulfoxide. Though they present the ability to bind DNA, no activity against parasites in cell culture was observed. Considering their potential application as molecular tools we further analyzed the interactions with DNA through an electrophoretic approach. Non covalent withdrawal of superhelicity and a rapid nicking activity upon covalent interaction was observed. Inhibition of both effects was observed in the presence of distamycin suggesting the involvement of the DNA minor groove in the interaction with the nitrofurylsemicarbazone ruthenium complexes. In addition cleavage inhibition by dimethyl sulfoxide suggests an oxidative mechanism of action.

A recombinant Escherichia coli expressing an a-alkyl-b-ketoester reductase that yields the anti (2R,3R) a-alkyl-bhydroxyester as the major product was constructed. The enzyme was identified from genomic libraries of Paucimonas lemoignei, cloned and expressed in E. coli providing a whole cell biocatalyst with novel stereoselectivity. This constitutes the first report on a whole cell biocatalyst for the synthesis of (2R,3R) a-alkyl-b-hydroxy-esters.

Adequate methods for skin diagnosis are necessary to determine the characteristics state of the skin of cosmetic users in order to properly formulate cosmetics products, and could provide a useful tool for supporting cosmetics efficacy claims. In the present work sensory methods of clinical evaluation and selfevaluation were developed for the diagnosis of the state of the facial skin of fifty nine volunteers. A Cosmetologist observed the facial skin of the volunteers and scored six attributes using a structured 9-point quality scale. Besides, volunteers were asked to evaluate the state of their facial skin by scoring the same attributes than the Cosmetologist, using the same quality scale. Both clinical evaluation and selfevaluation methods provided a simple an efficient tool to characterize the state of the volunteers’ skin. Clinical evaluation provided information regarding the objective state of the skin, whereas selfevaluation provided a measure of the perception that each volunteer has of her skin. Cluster analysis allowed the identification of groups of volunteers with similar state of their skin, according to clinical evaluation and selfevaluation. Considering clinical evaluation data, two groups of volunteers were identified, which differed in the type and frequency of the skin care treatment they used. Volunteers that frequently used antiaging and moisturizing products showed higher clinical evaluation scores, showing the positive effect of cosmetic treatments on the state of the skin. Regarding selfevaluation, three clusters were identified, that mainly differed in their age. Older volunteers tended to be stricter with themselves and gave lower scores to the state of their skin than younger volunteers, probably due to different expectations. Although clinical evaluation and selfevaluation data were not correlated, they provided complementary information.

A summary of the resource.
The aim of the present work was to study the influence of modified-atmosphere packaging on the microbiological and sensory quality of shiitake mushrooms (Lentinula edodes). Mushrooms were packaged under atmospheric air (passive modified atmosphere) and an initial gas mixture of 5% O2 and 2.5% CO2 (active modified atmosphere), in bags of two different films: low-density polyethylene (PE) and polypropylene (PP). As control, mushrooms were packaged inmacroperforated PP films. Bags were stored at 5 ◦C for 20 days. Package atmosphere composition, mushroom respiration rate, weight loss, microbiological counts and sensory quality were determined during storage. Risk assays were also performed. Under the studied conditions, shiitake mushroom deterioration was not due to microorganism growth, and therefore the shelf-life of this product might be defined by changes in its sensory characteristics. Sensory analysis showed that mushrooms stored under modified atmosphere (active and passive) had a higher deterioration rate than those stored in PP macroperforated films, and lower sensory quality values during the entire storage time. These results suggest that mushroom deterioration was probably due to shiitake mushrooms’ sensitivity to high CO2 concentrations.

The bacterial communities in floodwater, from a rice-planted and an unplanted field were characterized at the beginning (flooding stage) and at the end (harvest stage) of the rice cropping cycle. Most probable number estimations and plate counts of aerobic and anaerobic heterotrophic bacteria and of several metabolic bacterial groups (methanogens, sulfate-reducers, anaerobic sulfur and nonsulfur phototrophs, denitrifiers and ammonifiers) were similar in rice and unplanted floodwater at both sampling times. The analysis of denitrifiers and methanogens by fluorescent in situ hybridization revealed a shift in the phylogenetic affiliation only of the former group in the rice-planted floodwater. Terminal restriction fragment length polymorphism of 16S rRNA gene amplicons indicated that the bacterial communities of the rice-planted and unplanted soils were consistently diverse and strongly influenced by the season.
