In the present study a systematic approach was used to model the anti-T. cruzi activity of a series of N-oxide containing heterocycles belonging to four chemical families with a wide structural diversity. The proposed mode of action implies the reduction of the N-oxide moiety; however, the biochemical mechanism underlying the anti-T. cruzi activity is still unkown. For structural representation two types of descriptors were analyzed: quantum chemical (AM1) global descriptors and properties coded by radial distribution function (RDF). Both types of descriptors point to the relevance of electronic properties. The local-RDF (LRDF) identified an electrophilic center at 4.1-4.9 Å from the oxygen atom of the N-oxide moiety, although other properties are required to explain the biological activity. While the mode of action of N-oxide containing heterocycles is still unknown, the results obtained here strengthen the importance of the electrophilic character of the molecule and the possible participation of the heterocycle in a reduction process. The ability of these descriptors to distinguish among activity classes was assessed using Kohonen neural networks, and the best clustering descriptors were later used for model building. Different learning algorithms were used for model development, and stratified 10-fold cross-validation was used to evaluate the performance of each classifier. The best results were obtained using k-nearest neighbors (k-NN) and decision tree (J48) methods combined with global descriptors. Since tree-based methods are easily translated into classification rules, the J48 model is a useful tool in the de novo construction of new N-oxide containing heterocycle lead structures. It is well understood that fungi diversity is still underestimated and that we are far from understanding its importance and potential impact in biotechnology. Flavor compounds as “secondary metabolism” are very sensitive to fermentation conditions and mixed cultures, and although we had experience an exponential development of molecular biology in the last 30 years, metabolomics is still in its infancy. Benzenoid-derived metabolites act as precursors for a wide variety of products  involved in essential metabolic roles in eukaryotic cells. They are synthesised in  plants and some fungi through the phenylalanine ammonia lyase (PAL) or tyrosine  ammonia lyase (TAL) pathways. Ascomycete yeasts and animals both lack the  capacity for PAL/TAL pathways, and metabolic reactions leading to benzenoid  synthesis in these organisms have remained incompletely known for decades. Here  we show genomic, transcriptomic and metabolomic evidence that yeasts use a  mandelate pathway to synthesise benzenoids, with some similarities to pathways  used by bacteria. We conducted feeding experiments using a synthetic  fermentation medium that contained either 13C-phenylalanine or 13C-tyrosine and,  using methylbenzoylphosphonate (MBP) to inhibit benzoylformate decarboxylase,  we were able to accumulate intracellular intermediates in the yeast Hanseniaspora  vineae. To further confirm this pathway, we tested three mutants in separate  fermentation experiments with deletions in the key genes putatively proposed to  form benzenoids (Saccharomyces cerevisiae aro10Δ, dld1Δ and dld2Δ). Our results  elucidate the mechanism of benzenoid synthesis in yeast through phenylpyruvate  linked with the mandelate pathway to produce benzyl alcohol and 4- hydroxybenzaldehyde from the aromatic amino acids, phenylalanine and tyrosine,  as well as sugars. The influence of olive oil concentration and sensory profile on the odor of virgin olive oil-based cosmetic creams was studied. Four olive oils were selected on the basis of different intensities of positive and defective odor attributes: two extra virgin olive oils, one virgin olive oil, and one ordinary virgin olive oil. Thirty cosmetic creams were prepared, by both cold and hot processing methods, using each of the above oils at concentrations of 3%, 5%, and 10%, in addition to mineral oil controls. A trained sensory panel evaluated the fruitiness and defectiveness intensities in the odor of creams, using unstructured 10-cm scales ranging from "none at all" to "much." The fruity and defective attributes perceived in the odor of creams were significantly influenced by the sensory profile of the starting olive oil, oil concentration, and preparation method. Overall, these findings suggest that virgin olive oils of only slightly fruity odor may be conveniently used for the preparation of cold-processed cosmetic creams, whereas ordinary virgin olive oils appear to be suitable for the preparation of cosmetic creams only by hot processing of the emulsion at a low oil concentration. Perceptions of essences for potential use in the development of a line of cosmetic emulsions containing olive oil were studied. Six cream samples prepared with six essences selected in a preliminary study were evaluated for overall liking and intention to purchase by a 63-women sample. A check-all-that-apply (CATA) question consisting of 32 terms was used to gather information about consumer perceptions of fragrance, affective associations, effects on the skin, price, target market, zones of application, and occasions of use. Hierarchical cluster analysis led to the identification of two consumer clusters with different frequency of use of face creams. The two clusters assigned different overall liking scores to the samples and used the CATA terms differently to describe them. A fragrance with jasmine as its principal note was selected for further development of cosmetic creams, as it was awarded the highest overall liking scores by respondents of the two clusters, and was significantly associated with cosmetic features including nourishing, moisturizing, softening, with a delicious and mild smell, and with a natural image, as well as being considered suitable for face and body creams. The use of CATA questions enabled the rapid identification of attributes associated by respondents with a cosmetic cream's fragrance, in addition to contributing relevant information for the definition of marketing and communication strategies. Word association, a projective qualitative technique, was applied to the development of olive oil-based and olive leaf extract-supplemented cosmetic creams. One hundred twenty female respondents presented with five stimulus terms – namely, “nourishing cream,” “antiaging cream,” “olive oil-based cream,” “olive leaf extract-supplemented cream” and “antioxidant-rich cream” – were asked to write down four images, associations, thoughts or feelings evoked by each of the different stimuli. Word associations made by the surveyed sample were then grouped into 14 categories. For each of the proposed stimuli, the frequency of mention of elicited words was analyzed according to and within each category. The use of word association enabled the identification of expected or desired sensory properties and efficacy of the studied cosmetic creams, as well as the concepts associated – in the minds of respondents – with each of the creams. The resulting information may be used to optimize the selection of new products and their denominations. The biomass industrialization valorisation of grains and cereals is considered an opportunity for the countries where agro-industrial activity is one of its main economic activities, rendering new higher-value products with a concomitant solution to waste accumulation issues. To that end, in this work we describe and characterise bioactive compounds generation from rice husk by semi-solid fermentation, obtained from 500 g of the material at room temperature and 60% humidity with mixed cultures of Phanerochaete chrysosporium and Gloeophyllum trabeum. The extract was evaluated in different situations: murine mammary tumour cells (4T1), normal cells (NIH 3T3) and in Lactobacillus acidophilus, and Trichoderma harzianum. The results exhibited that the extract inhibited 4T1 cells at concentrations higher than 20μg/mL, but did not inhibit normal cells, and displayed germicide activity after 3 days incubations. We propose that these functionalized compounds have a potential application in industry/agriculture/medicine obtained from rice husk waste. La albúmina sérica humana (HSA) es la proteína mayoritaria en el plasma, y cumple varias funciones, entre ellas atrapar especies oxidantes y electrófilas. Esta función se asocia a su residuo de cisteína libre, Cys34, el cual representa más del 80 % de los tioles libres en plasma. Este tiol puede reaccionar con peróxidos (i.e. oxidarse) para dar un ácido sulfénico de vida media larga, el cual puede seguir tres destinos: reducirse por reacción con un tiol para formar un disulfuro mixto, oxidarse irreversiblemente a ácido sulfínico, o decaer espontáneamente a un producto aún no caracterizado. Por otro lado, dada su prolongada vida media, la HSA es susceptible a ser modificada (glicada) por compuestos carbonílicos reductores presentes en el plasma, generando productos de glicación. Dichas modificaciones alteran la estructura y función de la HSA. En particular, se ha sugerido que habría una interacción sinérgica entre la glicación de la HSA y la oxidación del tiol. En este trabajo de tesis se usó un conjunto de técnicas experimentales y de modelado computacional para estudiar las propiedades del tiol de la HSA y de su derivado ácido sulfénico, y también, la interacción entre la glicación de la HSA y la oxidación del Cys34. Seis aspectos principales fueron tratados: (1) la estructura y dinámica del entorno del residuo Cys34 en las formas tiol y tiolato; (2) los mecanismos de las reacciones del tiolato de la HSA con peróxidos; (3) la estructura y dinámica del entorno de Cys34 en las formas ácido sulfénico y sulfenato; (4) el mecanismo de la reacción del ácido sulfénico de la HSA con H2O2; (5) la interacción entre la glicación de la HSA y la oxidación del tiol; y (6) la reacción del tiol de la HSA con metilglioxal. Lateralmente, también se validó, metodología tanto experimental como computacional, para describir este tipo de problemas. En primer lugar, se determinó que los residuos Tyr84 y Asp38 cumplen un papel importante modulando propiedades del tiol tales como la acidez y la nucleofilia, y se asignó un rol estructural a His39. Otro hallazgo importante es que el tiol(ato) interacciona con moléculas de agua que hacen de puente con el carboxilato de Asp38. En segundo lugar, se confirmó el carácter SN2 del mecanismo de las reacciones del tiolato de la HSA con peróxidos, obteniendo energías libres de activación de respectivamente 14.2 y 10.4 kcal mol−1 para las reacciones con H2O2 y HOONO. Como hallazgo mecanístico importante, se aprecia que, mientras en la reacción con H2O2 se da una transferencia protónica “post-estado de transición”, 7 esto no ocurre en su análoga con HOONO, mostrando que la naturaleza del grupo saliente determina la naturaleza del producto de reacción. En paralelo, se determinó experimentalmente un valor de entalpía de activación de 13.7 kcal mol−1, próximo al valor resultante del modelado de 12.9 kcal mol−1. En tercer lugar, se llegó a dos hallazgos importantes respecto al entorno del ácido sulfénico de la HSA: (a) la presencia de un enlace de hidrógeno persistente entre el ácido sulfénico y el carboxilato de Asp38; y (b) la cercanía del NH peptídico de Cys34 al Sγ del ácido sulfénico, que además se encuentra formando enlace de hidrógeno (puenteado por agua) con el imidazol de His39. A fraction of rice bran (RB), generated during the brown rice polishing, is utilized to extract oil, resulting in defatted RB (DRB). The aim of this study was to optimize the emulsification conditions to enhance the value of this byproduct by formulating potential vegan dressings and characterizing them. Enzymatic hydrolysis of the starch present in DRB yields the DRB concentrate (DRBC). A central composite design was applied and the results were analyzed using response surface methodology to select optimal conditions for an oil-in-water emulsion formula. Two formulations were chosen: one corresponds to the optimal conditions, with 26.5% of oil and 73.5% of DRBC dispersion (eoptimal), and the other one with 21.7% of oil and 78.3% of dispersion (eED8). The eoptimal formulation exhibited significantly lowermean De Brouckere diameter (D4,3) value and higher viscosity when compared with eED8. For both emulsions, the particle size distribution and D4,3 remained unchanged during storage, whereas viscosity decreased, and backscattering (BS) increased. Initially, both emulsions exhibited solid viscoelastic behavior, whichwas partially lost during quiescent storage. The increase in BS was attributed to particle disaggregation, ultimately leading to the aforementioned change in rheological behavior. In conclusion, although the designed emulsions underwent microstructural changes, they were stable against gravitational separation. To improve stability during quiescent storage, it is suggested to incorporate a thickening agent. Hence, it is propose to procced with the development of a vegan dressing based on the eoptimal emulsion, as it exhibits superior physicochemical properties. El óxido nítrico (•NO) cumple potentes acciones reguladoras en la función celular y viabilidad tisular. La disminución en su biodisponibilidad observada durante procesos inflamatorios crónicos y agudos responde a su transformación en especies reactivas del nitrógeno (ERN) y tiene un rol crítico en la regulación de la oxidación lipídica y en la consecuente formación de derivados nitrados de lípidos (nitroalquenos) con efectos anti-‐ inflamatorios. Los nitrolípidos se forman in vivo y se han convertido en los últimos 20 años en uno de los principales objetos de estudio en el área de la química oxidativa de macromoléculas. Nuestro grupo ha caracterizado químicamente la síntesis y purificación de ácido nitroaraquidónico (NO2AA), un nitroalqueno con propiedades biológicas asociadas a su capacidad anti-‐inflamatoria. El ácido araquidónico (AA•) es precursor de una amplia variedad de lípidos bioactivos, entre ellos los eicosanoides derivados de la vía Prostaglandina G/H sintasa (PGHS), y postulamos que su nitración para formar NO2AA podría representar un mecanismo fundamental de modulación de los procesos inflamatorios vinculados al AA. En ese contexto integramos técnicas bioquímicas, proteómicas y computacionales para caracterizar cinética y estructuralmente la interacción de NO2AA con PGHS-‐1 y PGHS-‐2. Demostramos que el NO2AA es capaz de inactivar estas enzimas en forma irreversible, exhibiendo una potencia comparable a la de las drogas antiinflamatorias no esteroideas (AINES). Trasladamos este resultado a modelos celulares de relevancia biológica en el metabolismo de AA: plaquetas humanas y macrófagos murinos. El NO2AA inhibió la actividad PGHS-‐1 y PGHS-‐2 in vivo, ejerciendo un potente efecto anti-‐agregante en plaquetas modulando el metabolismo normal del AA sumado a su acción sobre blancos adicionales a PGHS-‐1. Finalmente, ponemos a punto un sistema de reacciones para evaluar la síntesis catalítica de NO2AA por la actividad PGHS y proponemos un posible mecanismo de regulación plaquetaria in vivo. Nitro-fatty acids represent endogenously occurring products of oxidant-induced nitration reactions. We have previously synthesized amixture of four isomers of nitroarachidonic acid, a novel anti-inflammatory signaling mediator. In this study, we synthesized and chemically and biologically characterized for the first time an esterified nitroalkene derived from the nitration of methylarachidonate (AAMet): 6-methylnitroarachidonate (6-AAMetNO2). Synthesis was performed by reacting AAMet with sodium nitrite under acidic conditions. Analysis by mass spectrometry (positive-ion ESI-MS) showed an [M+H]+ ion of m/z 364, characteristic of AAMetNO2. Fragmentation of this ion yielded a daughter ion at m/z 317, corresponding to the neutral loss of the nitro group ([M+H−HNO2]+). Furthermore, IR signal at 1378 cm−1 and NMR data confirmed the structure of a 6-nitro-positional isomer. This novel esterified nitroalkene was capable of promoting vascular protective actions including: (a) the induction of vasorelaxation via endothelium-independent mechanisms, associated with an increase in smooth muscle cell cGMP levels, and (b) a potent dose-dependent inhibition of human platelet aggregation. We postulate that 6-AAMetNO2 could be a potential drug for the prevention of vascular and inflammatory diseases, and the presence of the methyl group may increase its pharmacological potential. Aims: To characterize the bioemulsifier produced by a nonfluorescent strain of Pseudomonas putida isolated from a polluted sediment and to determine the influence of pH, temperature, media composition, and carbon and nitrogen source on growth and emulsifying activity. Methods and Results: Different indexes were employed to determine the emulsifying properties of culture supernatants of P. putida ML2 in defined and complex media. Surface tension of cell-free supernatants was measured. Purification and chemical analysis of the emulsifier was performed. Confirmed results indicate that a polysaccharide with hexasaccharide repeating units is responsible for the emulsifying activity in a mineral medium with glucose as sole carbon source. Moreover, an emulsifier is produced when growing on naphthalene. Conclusions: Culture media composition influences the amount and the properties of the emulsifier produced by this P. putida strain. Under nitrogen limiting conditions, a polysaccharide is responsible for the emulsifying activity in defined mineral media. In complex nitrogen rich medium, a different kind of emulsifier is produced. The exopolymer may contribute to hydrocarbons solubilization. Significance and Impact of the Study: This is the first exopolysaccharide with emulsifying properties produced by a Pseudomonas strain reported to the present. Also chemical composition is significantly different from previous reports. This strain has potential use in bioremediation and the purified polysaccharide may be used in food and cosmetic industry. Moreover, the production of the exopolymer may play a role on biofilm formation. In most organisms, thioredoxin (Trx) and/or glutathione (GSH) systems are essential for redox homeostasis and deoxyribonucleotide synthesis. Platyhelminth parasites have a unique and simplified thiol-based redox system, in which the selenoprotein thioredoxin-glutathione reductase (TGR), a fusion of a glutaredoxin (Grx) domain to canonical thioredoxin reductase domains, is the sole enzyme supplying electrons to oxidized glutathione (GSSG) and Trx. This enzyme has recently been validated as a key drug target for flatworm infections. In this study, we show that TGR possesses GSH-independent deglutathionylase activity on a glutathionylated peptide. Furthermore, we demonstrate that deglutathionylation and GSSG reduction are mediated by the Grx domain by a monothiolic mechanism and that the glutathionylated TGR intermediate is resolved by selenocysteine. Deglutathionylation and GSSG reduction via Grx domain, but not Trx reduction, are inhibited at high [GSSG]/[GSH] ratios. We found that Trxs (cytosolic and mitochondrial) provide alternative pathways for deglutathionylation and GSSG reduction. These pathways are operative at high [GSSG]/[GSH] and function in a complementary manner to the Grx domain-dependent one. Despite the existence of alternative pathways, the thioredoxin reductase domains of TGR are an obligate electron route for both the Grx domain- and the Trx-dependent pathways. Overall, our results provide an explanation for the unique array of thiol-dependent redox pathways present in parasitic platyhelminths. Finally, we found that TGR is inhibited by 1-hydroxy-2-oxo-3-(N-3-methyl-aminopropyl)-3-methyl-1-triazene (NOC-7), giving further evidence for NO donation as a mechanism of action for oxadiazole N-oxide TGR inhibitors. Thus, NO donors aimed at TGR could disrupt the entire redox homeostasis of parasitic flatworms. A Trypanosoma brucei selenocysteine synthase(SepSecS) null-mutant, in which selenoprotein synthesis is abolished, displayed similar sensitivity tooxidative stress induced by a short-term exposure to high concentrations of methylglyoxal or H2O2tothat of the parental wild-type cell line. Importantly, the infectivity of the SepSecS knockout cell line wasnot impaired when tested in a mouse infection model and compensatory effects via up-regulation ofproteins involved in thiol-redox metabolism were not observed. African trypanosomiasis is a major problem for human and animal health in endemic countries, where it threatens millions of people and affects economic development. New drugs are needed to overcome the toxicity, administration, low efficacy, and resistance issues of the current chemotherapy. Robust, simple, and economical high‐throughput, whole‐cell‐based assays are required to accelerate the identification of novel chemical entities. With this aim, we generated a bioluminescent cell line of the bloodstream stage of Trypanosoma brucei brucei and established a screening assay. Trypanosomes were stably transfected to constitutively express a thermostable red‐shifted luciferase. The growth phenotype and drug sensitivity of the reporter cell line were essentially identical to that of the parental cell line. The endogenous luciferase activity, measured by a simple bioluminescence assay, proved to be proportional to parasite number and metabolic status. The assay, optimized to detect highly potent compounds in a 96‐well‐plate format, was validated by screening a small compound library (inter‐assay values for Z' factor and coefficient variation were 0.77 and 5.8%, respectively). With a hit‐confirmation ratio of ~97%, the assay was potent enough to identify several hits with EC50 ≤ 10 μM. Preliminary tests indicated that the assay can be scaled up to a 384‐well‐plate format without compromising its robustness. In summary, we have generated reporter trypanosomes and a simple, robust, and affordable bioluminescence screening assay with great potential to speed up the early‐phase drug discovery against African trypanosomes. This study describes the microbial community richness, -dynamics, and -organization of four full-scale anaerobic digesters during a time-course study of 45 days. The microbial community was analyzed using a Bacteria- and Archaea-targeting 16S rRNA gene-based Terminal-Restriction Fragment Length Polymorphism approach. Clustering analysis separated meso- and thermophilic reactors for both archaeal and bacterial communities. Regardless of the operating temperature, each installation possessed a distinct community profile. For both microbial domains, about 8 dominant terminal-restriction fragments could be observed, with a minimum of 4 and a maximum of 14. The bacterial community organization (a coefficient which describes the specific degree of evenness) showed a factor 2 more variation in the mesophilic reactors, compared with the thermophilic ones. The archaeal community structure of the mesophilic UASB reactor was found to be more stable. The community composition was highly dynamic for Bacteria and Archaea, with a rate of change between 20–50% per 15 days. This study illustrated that microbial communities in full-scale anaerobic digesters are unique to the installation and that community properties are dynamic. Converging complex microbial processes such as anaerobic digestion which rely on a multitude of microbial teams apparently can be highly dynamic. In this study, the microbial community characteristics in continuous lab-scale anaerobic reactors were correlated to reactor functionality using the microbial resource management (MRM) approach. Two molecular techniques, denaturing gradient gel electrophoresis (DGGE) and terminal-restriction fragment length polymorphism (TRFLP), were applied to analyze the bacterial and archaeal communities, and the results obtained have been compared. Clustering analyses showed a similar discrimination of samples with DGGE and T-RFLP data, with a clear separation between the meso- and thermophilic communities. Both techniques indicate that bacterial and mesophilic communities were richer and more even than archaeal and thermophilic communities, respectively. Remarkably, the community composition was highly dynamic for both Bacteria and Archaea, with a rate of change between 30% and 75% per 18 days, also in stable performing periods. A hypothesis to explain the latter in the context of the converging metabolism in anaerobic processes is proposed. Finally, a more even and diverse bacterial community was found to be statistically representative for a wellfunctioning reactor as evidenced by a low Ripley index and high biogas production.
