A summary of the resource.
Four new lead(II) thiosaccharinate complexes: [Pb(tsac)2H2O] (1) (tsac: thiosaccharinate anion), [Pb2(tsac)4(py)4] (2) (py: pyridine), [Pb(tsac)(o-phen)2](tsac)·CH3CN (3) (o-phen: 1,10-phenantroline), and [Pb(tsac)2(bipy)] (4) (bipy: 2,2�-bipyridine) were prepared. The infrared and electronic spectra as well as the thermal analysis of all the compounds were recorded and discussed. The thiosaccharinate anion acts in three different coordination forms, one of then reported for the first time. The crystal structures of complexes 2 and 3 have been determined by single crystal X-ray diffractometry. In complex 2, two monomeric moieties are joined together forming a symmetric bis-μ-sulphur bridged dimer by interaction of two lead(II) atoms through the exocyclic sulphur atoms of two thiosaccharinate ligands. The seven-fold coordination sphere of each lead atom is completed by two pyridine nitrogen atoms and by another sulfur and two nitrogen atoms of the thiosaccharinate anions. In complex 3, the lead(II) atom is coordinated by four nitrogen atoms of two 1,10-phenantroline molecules and by the sulfur and nitrogen atoms of one thiosaccharinate ion. The second anion has an electrostatic interaction with the nucleus.

A summary of the resource.
A flow injection system is presented for the determination of the insolubles content in used lubricating oil samples. The system is based on the injection of an aliquot of the sample in a stream of organic solvent where it is dispersed, and measurement of the scattered radiation (measured as apparent absorbance) in the visible range (λ = 640 nm). An LED-based photometer was used for this purpose. The whole system including sample injection and data acquisitionwas controlled by a personal computer. Calibration curves exhibited good linearity (h = 0.415±0.016C+ 0.00±0.03, r2 = 0.9995, confidence level of 95%) in the range up to 2.68% (insolubles in pentane). Detection and quantification limits were respectively 0.07% and 0.16% (w/w). The method was validated by analysis of 25 real samples by the proposed method and the FTIR method finding high correlation. Waste generation and reactive consumption is much less than in the official method (ASTM D-893). The proposed method employs 25mL of kerosene per sample while the official method employs 200mL of pentane.

In vitro growth inhibitory activity of 21 new 5-ethenylbenzofuroxan derivatives against the protozoan parasite Trypanosoma cruzi, the causative agent of American trypanosomiasis, was studied. The designed compounds possess the previously described exigencies for optimal anti-parasite activity, the 5-ethenylbenzofuroxanyl moiety with different substituents. The synthetic key for preparing the derivatives was the Wittig procedure, that when 5-formylbenzofuroxan was used as the electrophile the corresponding deoxygenated products were marginally generated. Four of the new derivatives displayed remarkable in vitro activities against the epimastigote form of three strains of T. cruzi, Tulahuen 2, CL Brener, and Y. While the three deoxygenated analogues biologically assayed resulted inactives. Unspecific cytotoxicity was evaluated using human macrophages and active derivatives were not toxic at a concentration at least 13 times that of its IC50 against T. cruzi (CL Brener strain). From the preliminary structure–activity relationship studies lipophilicity and electronic requirements were found relevant to anti-T. cruzi activity. Active compounds are more lipophilic than inactive ones and it was also identified that an optimum value of R Swain-Lupton’s descriptor is required for optimal activity

New benzofuroxans were developed and studied as antiproliferative Trypanosoma cruzi agents. Compounds displayed remarkable in Vitro activities against different strains, Tulahuen 2, CL Brener and Y. Its unspecific cytotoxicity was evaluated using human macrophages being not toxic at a concentration at least 8 times, and until 250 times, that of its T. cruzi IC50. Some biochemical pathways were studied, namely parasite respiration, cysteinyl active site enzymes and reaction with glutathione, as target for the mechanism of action. Not only T. cruzi respiration but also Cruzipain or trypanothione reductase were not affected, however the most active derivatives, the vinylsulfinyl- and vinylsulfonyl-containing benzofuroxans, react with glutathione in a redox pathway. Furthermore, the compounds showed good in ViVo activities when they were studied in an acute murine model of Chagas’ disease. The compounds were able to reduce the parasite loads of animals with fully established T. cruzi infections.

Logos on environmental sustainability could consist of an effective strategy to provide consumers with accurate, understandable and trustworthy information to encourage them to buy environmentally sustainable wines. However, the large number of different logos indicating environmental sustainability available in the market raises the question ofwhether their associated messages are successfully conveyed to consumers. In this context, the aim of the present exploratory study was to investigate how Burgundy wine consumers perceive a series of logos indicating environmental sustainability in wine production. Fourteen logos available in the French market were selected: three logos being specific to wine and eleven nonspecific. The logos were presented to 127 wine consumers from Dijon area (France), following an incomplete balanced block design. For each logo, participants had to answer the question: “What does a bottle of wine with this logo suggest to you?”. Responses were qualitatively analyzed and grouped into different categories. Chi-square tests and Correspondence analysiswere used to identify the relationship among logos and categories. Results showed large differences in how consumers perceived the logos. Biodyvin, the former European AB and the French AB organic logos were the logos that most successfully conveyed their messages, being strongly associated to organic wine. Most logos did not communicate a message related to environmental sustainability, which reaffirms the need to provide consumers with adequate information on environmental sustainability and to conduct further research on this subject.

A summary of the resource.
Response surface methodology (RSM) was applied to optimise laccase production by a new strain of Pycnoporus sanguineus, isolated from the Uruguayan forests, grown on bark shavings under semi-solid-state fermentation conditions. Enhancement of laccase production was studied, with a full 24 factorial design with two replicates and four central points. Operating under the optimal conditions obtained (pH 3.0, yeast extract 10 g L−1, glucose 5 g L−1 and bark shavings 3 g), a laccase activity of nearly 3000 U L−1 was achieved. Subsequently, the effect of different potential inducers on laccase production (Cu+2, Mn+2 and lignin) was tested using the one-factor-at-a-time approach. It was found that the addition of 0.5 mM Cu+2 into the cultures considerably increased laccase production and, thus, maximum activity values of about 16 000 U L−1 were achieved. Further, the optimised culture broth (containing mainly laccase) showed high capacity to decolourise different recalcitrant synthetic dyes such as Acid Red 88 (AR88), Reactive Black 5 (RB5) and Lanaset Grey G (LG). After selecting enzyme concentration and time of reaction, almost 70% of AR88 could be decolourised in the absence of redox mediators. However, mediators were necessary to bleach RB5 and LG. Methyl syringate was the most effective mediator for AR88 (reaching a decolouration degree over 90%) and RB5 (nearly 70%). Around 80% of LG could be decolourised in the presence of violuric acid. These promising results make this enzyme a very useful tool for developing wastewater treatments.

Aplicando leyes fisicoquímicas sencillas y la estequiometría de reacciones se pueden hacer los cálculos que permiten confirmar o rechazar las propuestas efectuadas en la novela “Cinco semanas en globo” de Jules Verne. De ellos se deduce que es factible la flotación y movimientos verticales del aerostato ideado por el autor pero que no son factibles ni su manera de generar corriente continua ni el mecanismo de calentamiento del hidrógeno del globo. Se trata de especulaciones literarias con algo de ciencia incluida. De esta manera, el docente de Química puede ilustrar sus clases con ejemplos tomados de una novela de aventuras, aprovechando para efectuar una aplicación de los conocimientos de una época a través de una integración ciencia/arte.

Abstract

Appling basic Physicochemical Laws and considering the stoichiometry of the reactions involved it is possible to verify the validity of the considerations made by Jules Verne on his novel entitled “Five weeks in a balloon”. By this simple methodology it can be demonstrated that vertical thrust and movement of the balloon are feasible as proposed by author, but situation is different concerning the methods proposed for the generation of direct current and for the heating of the hydrogen gas. This is literary speculation with some included science. In this way, Chemistry teachers can illustrate their lectures with examples from an adventure novel, building to make an application of the knowledge at that time through an integration science/art.

Abstract
A summary of the resource.
The objective of this study was to predict chloroform formation resulting from the process of disinfecting water, particularly trihalomethane which is most frequently produced. A statistical model was used which included repeated measurements of water parameters used for monitoring water quality at 51 sites covering the municipal water system of Montevideo. Samples were taken considering different seasons from June 2009 to July 2011 in Montevideo. Total samples (n = 330) were analytically studied using the headspace-gas chromatography method coupled with mass spectrometry. Chloroform was the dependent variable and the covariables were pH, temperature, free chlorine, and total chlorine. A Tobit analysis with an unstructured correlation matrix was performed, and a significant interaction was found between pH and free chlorine for the prediction of chloroform formation. We concluded that parameters for the continuous control of water quality for consumption can be used to predict the levels of chloroform that may be present. Given the large measurement to variability found in the repeated measurements, the use of averages that include more than one season is not recommended to determine the degree of compliance with acceptable levels established by norms.

Glutathione transferases (GSTs) are believed to be a major detoxification system in helminths. We describe the expression and functional analysis of EgGST, a cytosolic GST from Echinococcus granulosus, related to the Mu-class of mammalian enzymes. EgGST was produced as an enzymatically active dimeric protein (rEgGST), with highest specific activity towards the standard substrate 1-chloro- 2,4-dinitrobenzene (CDNB; 2.5�mol min−1 mg−1), followed by ethacrynic acid. Interestingly, rEgGST displayed glutathione peroxidase activity (towards cumene hydroperoxide), and conjugated reactive carbonyls (trans-2-nonenal and trans,trans-2,4-decadienal), indicating that it may intercept damaging products of lipid peroxidation. In addition, classical GST inhibitors (cybacron blue, triphenylthin chloride and ellagic acid) and a number of anthelmintic drugs (mainly, hexachlorophene and rafoxanide) were found to interfere with glutathione-conjugation to CDNB; suggesting that they may bind to EgGST. Considered globally, the functional properties of rEgGST are similar to those of putative orthologs from Echinococcus multilcularis and Taenia solium, the other medically important cestodes. Interestingly, our results also indicate that differences exist between these closely related cestode GSTs, which probably reflect specific biological functions of the molecules in each parasitic organism.

A major fraction of monoterpenes and norisoprenoids in young wines is conjugated to sugars representing a significant reservoir of aromatic precursors. To promote their release, b-glucosidase, a-arabinosidase, and a-rhamnosidase from a commercial Aspergillus niger preparation, were immobilized onto acrylic beads. The aim of this work was the development and application of an immobilized biocatalyst, due to the well-known advantages over soluble enzyme preparations: control of the reaction progress and preparation of enzyme-free products. In addition, the obtained derivative showed increased stability in simile wine conditions. After the treatment of Muscat wine with the biocatalyst for 20 days, free monoterpenes increased significantly (from 1119 to 2132 lg/L, p < 0.01) with respect to the control wine. Geraniol was increased 3,4-fold over its flavor thresholds, and accordingly its impact on sensorial properties was very relevant: nine of ten judges considered treated wine more intense in fruit and floral notes

A novel epoxyquinol analog was prepared by molecular simplification of monomeric and dimeric scaffolds. A feasible methodology for the insertion of side chains into cyclohexenone skeleton was developed. Insertion of the hydroxymethyl side chain was achieved through a-sulfonylcarbanion chemistry. The alkenyl chain was inserted through palladium cross-coupling strategy.

Nevirapine (NVP) extensive data obtained after oral single dose administration of 200 mg in a crossover study involving 16 healthy subjects was used to develop a descriptive population pharmacokinetic model including drug recirculation, since secondary peaks were observed in plasma concentration-time profiles for all subjects. Through implementation of model event time feature in NONMEM 7.3.0, a simple mechanistic model physiologically consistent with the process of drug cycling was able to describe multiple peaks phenomena and quantify its pharmacokinetic parameters, achieving a better performance than its analogue conventional one. Absorption process, between subject and-between occasion variability of pharmacokinetic parameters was also assessed. Estimated mean fraction of NVP bioavailable dose undergoing recirculation process was 51.6 %, a magnitude which could hardly be explained by enterohepatic cycling. In this work we propose an alternative disposition process to explain NVP drug recirculation: gastric secretion with posterior intestinal reabsorption. Due to physicochemical and pharmacokinetic properties of the drug, this neglected phenomenon is more likely to explain the results obtained, and might be present in disposition of several basic drugs.

Abstract

Twelve yeast Debaryomyces hansenii strains were characterized for their ability to inhibit in vitro growth of Penicillium italicum, production of lytic enzymes (b-1.3-glucanase, chitinase and protease) and postharvest biological control of blue mold decay (caused by P. italicum) on Mexican lemon fruits. The D. hansenii strains inhibited in vitro the growth of P. italicum in different pHs (4.2, 4.6 y 5.0), effect related to killer toxins. The enzyme activity (of b-1.3- glucanase, chitinase and protease) was detected on Yeast Nitrogen Base (YNB) medium inoculated with cell wall fragments of P. italicum, the range enzymatic activity of all strains was between 9 and 13 days after inoculated culture medium. All D. hansenii strains significantly reduced blue mold on Mexican lemon during fifteen days of storage at 13 8C, but the LL1 strain was distinctly more effective under these conditions. These results showed that D. hansenii is an alternative for biological control of blue mold decay of Mexican lemon fruits.

Toxicology is nowadays an interdisciplinary science, including various areas such as Clinic, Experimental, Forensic, Analytic, Occupational, Environmental and Social Toxicology. Apart from the inter- and multidisciplinary approach, there is now growing conscience on the importance of the transdisciplinary view,which transcends and goes through the various disciplines, collecting their common aspects. The Toxicology and Environmental Hygiene Department of the Faculty of Chemistry, University of the Republic, gives a course called “Analytic Toxicology and Legal Chemistry”, in which the students write and present orally a monography. The themes of the course and their treatment may constitute an exercise of acquisition of the transdisciplinary approach. The aim of the present study is to propose a methodology to evaluate this competence in the students, given the analysis of their performance in previous instances of teaching of this course. Previous result,s through a period of five years, show a satisfactory performance of the students, with average marks higher than 80% of the possible maximum for the written and above 75%. for the oral part. This antecedent points to an incipient acquisition of the transdisciplinary approach, which must be evaluated in future instances of this course, through the requirements for the students monography and a suiting questionnaire.

A summary of the resource.
To assess the reliability of salivary phenytoin (PHT) concentrations and predicted free PHT levels by Sheiner-Tozer equation in order to substitute measured free PHT concentrations in critically ill patients. Methodology: Twenty-four neurocritically ill adult patients receiving intravenous PHT were included in the study. Analyses of total, free plasma and saliva PHT concentrations were performed by fluorescence polarization immunoassay. Plasma albumin levels were also determined. Results: Free PHT concentrations as well as salivary levels better correlate to clinical effect than total drug concentrations. Linear regression analysis showed a strong correlation between estimated free PHT concentrations by Sheiner-Tozer and measured free PHT levels (r=0.835; p<0.001) and salivary PHT concentrations and measured free PHT concentrations (r=0.964; p<0.001). Sheiner-Tozer equation could be misleading in the presence of displacing drugs. Conclusions: Saliva may serve as a feasible fluid to plasma in order to be used as a surrogate for free concentration monitoring of PHT in this population.

Phase-equilibrium diagrams of biodiesel synthesis substrate (oil/alcohol/alkyl esters) and product (alkyl esters/glycerol/alcohol) mixtures containing methanol, ethanol and propanol, respectively, were used for the identification of those composition ratios leading to mixture homogeneity. Based on the diagrams, complete conversion of the substrate oil contained in a sunflower oil/ ethanol/ethyl ester mixture at a weight ratio of 7.6:11.4:81.0 resulted in the generation of an amount of glycerol capable of remaining in solution in the product mixture. In contrast, complete conversion using a mixture of a lower alcohol and ethyl ester content (19.0:6.0:75.0) leads to the separation of phases. A continuous reactor charged with Lypozyme TL-IM and fed with the former mixture showed constant conversion and productivity with time, whereas process efficiency was found to decrease drastically when the latter mixture was fed. When a sunflower oil/isopropanol/isopropyl ester mixture at a weight ratio of 35:35:30 was fed in the reactor charged with Novozym 435, the output glycerol concentration was equivalent to that theoretically expected. In contrast, feeding a sunflower oil/ethanol/ethyl ester mixture of equal proportions as above, the glycerol concentration was lower than expected, suggesting the occurrence of glycerol adsorption on the enzyme support.

Herein, we describe an approach toward selenazole preparation based on the cycloisomerization of propargyl selenoamides. The selenoamides were synthesized in situ using the Ishihara reagent with spontaneous cyclization to form the 2,5-disubstituted selenazoles. Heterocylcles 9a-j were prepared using readily available starting materials, and yields ranged from moderate to good (20-80%). Methylselenazole 9a could be transformed into a bromomethyl derivative 13 using NBS. The intermediate 13 would provide a more versatile building block for further derivatizations, e.g., the cyanide 14.

The globe artichoke (Cynara cardunculus L. subsp. cardunculus) is a perennial plant cultivated in the Mediterranean region and Americas for its edible young flower heads and as an interesting source of bioactive compounds. The present study was undertaken to evaluate scavenging capacity against the most physiologically relevant reactive oxygen species (ROS) and reactive nitrogen species (RNS) of three different extracts from artichoke leaves (infusion, decoction and hydroalcoholic) using different solvents, commonly accepted for human consumption (water and a mixture of ethanol/water). Additionally, the phenolic compounds in each extract were identified and quantified by high performance liquid chromatography coupled to diode array and mass spectrometer detectors (HPLC–DAD–MS/MS). Chlorogenic acid was the major phenolic compound identified in all extracts, followed by 1,3-dicaffeoylquinic acid (cynarin), luteolin-7-rutinoside and the infusion extract presented the highest phenolic content (108 mg/g extract, dry basis). In general, the extracts of artichoke leaves presented a remarkable capacity to scavenge ROS and RNS with IC50 values in a low μg/mL range (3.4–43 μg/mL). These findings suggest that artichoke could be a potential source of natural antioxidants and has an undeniable nutraceutical value.

Perceptions of essences for potential use in the development of a line of cosmetic emulsions containing olive oil were studied. Six cream samples prepared with six essences selected in a preliminary study were evaluated for overall liking and intention to purchase by a 63-women sample. A check-all-that-apply (CATA) question consisting of 32 terms was used to gather information about consumer perceptions of fragrance, affective associations, effects on the skin, price, target market, zones of application, and occasions of use. Hierarchical cluster analysis led to the identification of two consumer clusters with different frequency of use of face creams. The two clusters assigned different overall liking scores to the samples and used the CATA terms differently to describe them. A fragrance with jasmine as its principal note was selected for further development of cosmetic creams, as it was awarded the highest overall liking scores by respondents of the two clusters, and was significantly associated with cosmetic features including nourishing, moisturizing, softening, with a delicious and mild smell, and with a natural image, as well as being considered suitable for face and body creams. The use of CATA questions enabled the rapid identification of attributes associated by respondents with a cosmetic cream's fragrance, in addition to contributing relevant information for the definition of marketing and communication strategies.

A novel Re(V)-Re(II) nitrosyl complex, [Re(O)2(pyz)4][Re(NO)Br4(pyz)] (pyz = pyrazine) was prepared and characterized by X-ray diffraction, elemental analysis, infrared and ultraviolet-visible absorption spectra. This product is obtained in the initial steps of the reaction of (NBu4)[Re(NO)Br4(EtOH)] with pyrazine. Both, the cation and the anion are mononuclear complexes. The Re(V) atom in the cation is six-coordinate with four nitrogen atoms from pyrazine ligands, and two oxo ligands. The Re(II) anion is also six-coordinate, with four bromide ligands, a linear nitrosyl group and one nitrogen from pyrazine. The spectroscopic studies are discussed and compared with those already reported separately for the cation and the anion.

Selenocysteine (Sec) is encoded by an UGA codon with the help of a SECIS element present in selenoprotein mRNAs. SECIS-binding protein (SBP2/SCBP-2) mediates Sec insertion, but the roles of its domains and the impact of its deficiency on Sec insertion are not fully understood. We used Caenorhabditis elegans to examine SBP2 function since it possesses a single selenoprotein, thioredoxin reductase-1 (TRXR-1). All SBP2 described so far have an RNA-binding domain (RBD) and a Sec-incorporation domain (SID). Surprisingly, C. elegans SBP2 lacks SID and consists only of an RBD. An sbp2 deletion mutant strain ablated Sec incorporation demonstrating SBP2 essentiality for Sec incorporation. Further in silico analyses of nematode genomes revealed conservation of SBP2 lacking SID and maintenance of Sec incorporation linked to TRXR-1. Remarkably, parasitic plant nematodes lost the ability to incorporate Sec, but retained SecP43, a gene associated with Sec incorporation. Interestingly, both selenophosphate synthetase (SPS) genes are absent in plant parasitic nematodes, while only Cys-containing SPS2 is present in Sec-incorporating nematodes. Our results indicate that C. elegans and the nematode lineage provide key insights into Sec incorporation and the evolution of Sec utilization trait, selenoproteomes, selenoproteins, and Sec residues. Finally, our study provides evidence of noncanonical translation initiation in C. elegans, not previously known for this well-established animal model.

A summary of the resource.
The validation of an analytical procedure for the determination of pesticide residues in beeswax, an interesting matrix for environmental pollution monitoring, is presented. Using the QuEChERS template, the impacts of wax particle size, sample amount, and cleanup procedure (water addition, dispersive solid phase extraction, freeze-out, and combinations thereof) on extraction yield and coextractive load were studied. Sample preparation through liquid-liquid partitioning between acetonitrile and melted wax (∼80 °C), followed by freeze-out and primary-secondary amine dispersive cleanup, was performed on incurred and pesticide-free samples for 51 residues. Determinations were made through LC-MS/MS and GC×GC-TOF, and the whole procedure was validated. Matrix effects were evaluated, with recoveries between 70 and 120% and RSDs below 20% in almost all cases. LC-MS/MS LOQs ranged from 0.01 to 0.1 mg/kg for most pesticides, but for GC-amenable pesticides, GC×GC-TOF sensitivity was lower (0.1-0.2 mg/kg). This methodology can be applied for routine analysis of pesticide residues in beeswax.
