The horn fly, Haematobia irritans (L.), is a blood-sucking ectoparasite that is responsible for sizeable economic losses in livestock. The salivary gland products facilitate blood intake. Taking advantage of the identification of novel H. irritans salivary antigens (Hematobin, HTB and Irritans 5, IT5), we investigated the parasite loads, H. irritans blood intake, and antibody response of naturally infected bovines during the fly season. Fly loads and fly hemoglobin content fluctuated during the trial. Each time horn fly loads exceeded 200 flies per cattle, a reduction in horn fly blood intake was observed three weeks later. All of the cattle elicited an antibody response against HTB and IT5 that declined once the fly season was over. Cattle anti-IT5 titers were positively correlated with parasite loads and negatively correlated with fly blood intake. These results suggest that the natural changes in the H. irritans blood intake observed in this study were associated with a natural host response against horn fly salivary antigens. A summary of the resource.
The 14-3-3 protein, already described in the metacestode of Echinococcus multilocularis, has been characterized in the Echinococcus granulosus adult worm. Immunolocalization studies show the presence of the 14-3-3 protein in the periphery of testes and externally associated with the apical rostellum and adjacent worm tegument. The alcian blue staining in consecutive parasite sections gave similar reactivity patterns, suggesting that the 14-3-3 protein is produced and secreted by rostellar glands. Immunoblot analysis showed the presence of the 14-3-3 protein in somatic and excretory-secretory worm products with higher and smaller apparent molecular masses, respectively, than those detected in E. multilocularis or E. granulosus metacestode tissues. Conversely, the 14-3-3 protein was not detected in metacestode secretory products. Detection of anti-E. granulosus 14-3-3 reactivity in sera of experimentally infected dogs was achieved at early stages of infection. Specific antibody titres decreased during the course of infection. The possible origin and functions of the 14-3-3 protein produced by the adult worm are discussed.

A summary of the resource.
The preparation and properties of Escherichia coli β-galactosidase conjugates based on a recently developed immobilization method are presented. Enzyme immobilization is performed by reaction of its native thiol groups with agarose-bound thiolsulfonates under mild conditions. Depending on the amount of enzyme applied, 8–114 mg of protein per gram of dried gel derivative was immobilized on the thiolsulfonate agarose. With low protein loadings, the immobilization yield reached 100%. The thiolsulfonate-agarose gels exhibited high selectivity toward active enzyme. Because of this, the specific activity of the immobilized β-galactosidase was up to 50% higher than that of the applied, native enzyme. The method also provides the possibility to design the microenvironment of the immobilized enzyme by blocking residual thiolsulfonate groups with different reagents. The low-load derivatives blocked with glutathione, as well as the high-load derivatives without blocking, had better thermal stability than the soluble enzyme, and also showed excellent long-term stability at low temperatures. Thus, no decrease in enzymic activity was observed after storage of the derivatives for 18 months at +4°C as suspensions in 0.1 m potassium phosphate, pH 7.5.

The development of sensitive and easy-to-use biosensors that allow an adequate characterization of specific weak biological interactions like carbohydrate–lectin interactions still remains challenging today. Nanoparticles functionalized with carbohydrates are one of the most powerful systems for studying carbohydrate–lectin interactions, because they mimic the multivalent presentation of carbohydrates encountered in nature, for example when viruses and bacteria bind to cells. On the basis of the model system glucose–Concanavalin A (ConA), we explore the application of Transient Magnetic Birefringence (TMB) to study these weak interactions, using glucose-functionalized colloidal magnetite nanoparticles (NPs) as probes. We demonstrate that the binding dynamics can be monitored and derive a model to obtain the apparent cooperativity. For our studies, we use nanoparticles of 6 and 8 nm in diameter. The ConA-generated response shows apparent cooperativity, due to the cross-linking of nanoparticles by the ConA tetramer which has four binding sites. Cooperativity is higher for 6 nm NPs, possibly due to a better accessibility of all four ConA binding sites on smaller NPs, enhancing cross-linking. For this system, we find a detection limit of 3–23 nM.

Logos on environmental sustainability could consist of an effective strategy to provide consumers with accurate, understandable and trustworthy information to encourage them to buy environmentally sustainable wines. However, the large number of different logos indicating environmental sustainability available in the market raises the question ofwhether their associated messages are successfully conveyed to consumers. In this context, the aim of the present exploratory study was to investigate how Burgundy wine consumers perceive a series of logos indicating environmental sustainability in wine production. Fourteen logos available in the French market were selected: three logos being specific to wine and eleven nonspecific. The logos were presented to 127 wine consumers from Dijon area (France), following an incomplete balanced block design. For each logo, participants had to answer the question: “What does a bottle of wine with this logo suggest to you?”. Responses were qualitatively analyzed and grouped into different categories. Chi-square tests and Correspondence analysiswere used to identify the relationship among logos and categories. Results showed large differences in how consumers perceived the logos. Biodyvin, the former European AB and the French AB organic logos were the logos that most successfully conveyed their messages, being strongly associated to organic wine. Most logos did not communicate a message related to environmental sustainability, which reaffirms the need to provide consumers with adequate information on environmental sustainability and to conduct further research on this subject.

The action of HCS and HCG on cell-mediated immunity has been investigated. Full-thickness skin homografts were performed in 40 whole adult female Wistar rats. Brown rats of the A X C strain were selected as donors. The animals were divided into four groups injected with HCS, HCG, HCS + HCG, and saline. The graft rejection time and the wet and dry weight of thymus and spleen were evaluated. No hormonal treatment showed any effect on skin graft survival. Thymus weight, both wet and dry, decreased significantly by treatment with HCP or HCS + HCG. No modification was observed in spleen weight. These results do not agree with the theory that HCS and HCG modify immunological competence of maternal lymphocytes and thus may contribute to prevent rejection of the fetus.<br /><br />

In recent years major outbreaks of prion disease linked to oral exposure of the prion agent have occurred in animal and human populations. These disorders are associated with a conformational change of a normal protein, PrPC (prion protein cellular), to a toxic and infectious form, PrPSc (prion protein scrapie). None of the prionoses currently have an effective treatment. A limited number of active immunization approaches have been shown to slightly prolong the incubation period of prion infection. Active immunization in wild-type animals is hampered by auto-tolerance to PrP and potential toxicity. Here we report that mucosal vaccination with an attenuated Salmonella vaccine strain expressing the mouse PrP, is effective at overcoming tolerance to PrP and leads to a significant delay or prevention of prion disease in mice later exposed orally to the 139A scrapie strain. This mucosal vaccine induced gut anti-PrP immunoglobulin (Ig)A and systemic anti-PrP IgG. No toxicity was evident with this vaccination approach. This promising finding suggests that mucosal vaccination may be a useful method for overcoming tolerance to PrP and preventing prion infection among animal and potentially human populations at risk.

During the “El Niño”- Southern Oscillation (ENSO) event 2009–2010, trophic state uni (TSI’s) and multimetric (TRIX’s) indexes were determined in the north coast of the Río de la Plata (RdlP). The relationships of such indexes with the hydrological, physical and chemical parameters were explored to identify climatic variability or anthropogenic effects on the eutrophication process. Eleven oceanographic sampling campaigns at 25 stations were conducted along the north coast of RdlP from 2009 to 2011. Temperature, salinity and oxygen saturation, water transparency (Secchi depth), chlorophyll (Chl a) content and total nutrient concentrations (total nitrogen and phosphorous) were determined. Trophic indexes (TSI Chl a, TSI TP, TRIX NP-Voll. and TRIX NP-Mvdeo.) were quantified and the RdlP flow was measured. RdlP flow showed significant differences between ENSO phases, with higher values during “El Niño” (EN), negative and significant correlations with salinity, Secchi depth and positive with total nutrients. The trophic status indexes showed spatial and temporal variability and associations with flow, physical and chemical parameters. Trophic state indexes showed spatial and temporal heterogeneity and were strongly associated to environmental conditions (hydrological and seasonal oscillations) during the study period but also to the anthropogenic factors affecting the north coast of RdlP. The system has been classified as mesotrophic to hypertrophic, depending on the selected indicator. According to these results, we suggest the use of a multimetric trophic state indicator, i.e., the TRIX index, as explains better the causes and effects of the eutrophication processes. To implement the TRIX index, (TRIX NP-Mvdeo.) we suggest researchers to consider a large number of local records (Chl a, nutrients, oxygen) at seasonal scales. This index is adapted to the environmental conditions of the study zone and complemented with other community structure indexes, and should be used for eutrophication assessment programs in the north coast of RdlP.

Food insecurity has been associated with negative short, medium, and long-term health consequences, which are more detrimental for children and adolescents. These effects may depend on the coping strategies developed to deal with food shortages. The present research aimed at exploring coping strategies in food insecure households with children and adolescents in Uruguay, incorporating sociological theoretical insights from Bourdieu. A qualitative approach based on individual semi-structured interviews was used. A total of 40 interviews were conducted with adults who had parental responsibilities of children and adolescents and who received different types of food assistance, between July and December 2022, in four cities. Results showed that adults tend to develop a wide range of coping strategies aimed at: reducing food expenditure, increasing the availability of money for purchasing food, increasing food availability and/or rationing the food available in the household. Some of the strategies were implemented regardless of the severity of food insecurity, whereas others were characteristic of the moderate and severe levels of the construct. Evidence to support the mediation effect of coping strategies on health outcomes was found. Discourses suggested that lower accumulation of economic and cultural capital may be aligned with the adoption of less socially accepted mechanisms to access to food. Expressions of a specific habitus aimed at securing food were identified among participants with more deprivations. Taken together, the findings suggest that coping strategies may not be a universal or invariant sequence according to the severity of food insecurity and stress the importance of considering households’ resources and local context for the development of strategies to improve access to food.

Monitoring economic access to food is fundamental to implement strategies to reduce food insecurity and enhance the affordability of healthy diets. The aims of the present study were to develop a healthy food basket (HFB) and analyze its cost from 2011 to 2024, in Uruguay, a highincome Latin American country. The HFB was conceptualized as a set of foods that allows the achievement of the nutritional needs of a person at the lowest possible cost, considering the dietary patterns of the local population. Its construction was based on national and international dietary intake recommendations, local food acquisition patterns, and food prices. The daily cost of the HFB in May 2024 was USD 7.5, considering purchasing power parity (PPP) conversion factors. The cost of the basket was higher than the Cost and Affordability of a Healthy Diet elaborated by FAO for the country and the region and higher than the cost of the official food baskets of the country. The cost of the HFB increased 202.3% from January 2011 to May 2024, while inflation increased by 178.4%. Fruits and vegetables, meat, dairy products, and processed cereals made the largest contribution to the cost of the HFB. Results highlight the need to reassess the country’s poverty estimation criteria and implement strategies to improve the affordability of healthy foods, such as adjusting cash transfer amounts and introducing targeted subsidies. The proposed methodological approach can contribute to the standardization of approaches to measure and monitor the cost of healthy diets in other countries.

A series of novel benzoimidazo and N-aryl-5-oxo-imidazo[1,2-b]isoquinoline-10-carbothioamides was developed. All the compounds were evaluated for their in vitro action against the epimastigote form of Trypanosoma cruzi. Four of them showed higher activity than Nifurtimox. Their unspecific cytotoxicity was evaluated using HeLa and L6 cells, being non-toxic at concentrations at least 15 and 200 times higher than that of T. cruzi IC50. To gain insight into the mechanism of action, their DNA binding properties and reactivity with glutathione were studied, and QSAR study was performed

Phenazine 5,10-dioxides are prodrugs for antitumor therapy that undergo hypoxic-selective bioreduction to form cytotoxic species. Here we investigate the expanded system benzo[a]phenazine 7,12-dioxides as selective hypoxic cytotoxin-scaffold. The clonogenic survival of V79 cells on aerobic and anaerobic conditions, conduct us to study antiproliferative activity on Caco-2 tumoral cells in normoxia. Electrochemical, DNA-interaction and DNA-damage studies were performed to establish the mode of action. The results demonstrated the potential biological properties of the studied scaffold being derivatives 6–10 structural hits for further chemical-modifications to become into therapeutics for solid tumors. Compounds 6 and 8 with cytotoxicity against V79 cells in both conditions (aerobia and anaerobia) were also cytotoxic against Caco-2 tumoral cells in aerobiosis.

The catalytic gasification (900 �C) of forestry industry residue (Eucalyptus saligna) was laboratory-studied. Biomass feed rate and type and amount of catalyst were assayed for their effect on the gasified product composition and the overall energy yield of the gasification reaction. The use of a calcined dolomite catalyst resulted in a combustible gas mixture of adequate calorific power (10.65 MJ m�3) for use as fuel, but neither the product gas composition nor the energy yield varied significantly with widely different amounts of the catalyst (2 g and 20 g). The use of NiO-loaded calcined dolomite catalysts did not affect the product gas composition significantly but led to a 30% increase in the total product gas volume and to a reduction in the rate of tar and char formation. The catalyst loaded with the smallest amount of NiO studied (0.4 wt%. Ni/Dol) led to the highest energy yield (21.50 MJ kg�1 on a dry-wood basis) based on the use of the gasified product as fuel. The gasified product was found to have an adequate H2/CO molar ratio and H2 content for use as synthesis gas source and partial source of H2.

Temporal dominance of sensations (TDS) methodology was used to study the influence of yoghurt composition on sensory texture and to compare results with those from quantitative descriptive analysis (QDA). Eight yoghurts with different texture characteristics were formulated varying fat, modified starch and gelatine concentration. A trained assessor panel evaluated yoghurt texture using QDA and TDS. TDS provided information about the sequence of attributes that were dominant during consumption. This methodology was useful to characterise yoghurt texture, providing information about the influence of the evaluated formulation variables that was not gathered by QDA; starch did not significantly increase creaminess intensity but it led to an increase in its total dominance. Differences between attribute intensity and dominance were higher for complex texture attributes which were evaluated throughout consumption, such as creaminess and mouth coating.

La reciente búsqueda de fuentes de energía renovables ha motivado la investigación en nuevas tecnologías para la producción de energía eléctrica, siendo un ejemplo de ello las celdas de combustible microbianas (MFC). En estos dispositivos se produce energía eléctrica por acción del metabolismo microbiano. Estas celdas constan de una cámara anódica anaerobia, la cual contiene al ánodo, materia orgánica y microorganismos y cámara catódica aerobia, en la cual se encuentra el cátodo. El ánodo y el cátodo se encuentran conectados por una resistencia. Los electrones generados por el metabolismo microbiano son transferidos al ánodo y viajan a través de un circuito al cátodo, donde ocurre la reducción de oxígeno a agua. Este movimiento de electrones a través del circuito genera una corriente eléctrica. Resulta esencial para la producción de corriente eléctrica que la comunidad microbiana desarrollada en el ánodo se encuentre enriquecida en bacterias capaces de utilizar al ánodo como aceptor final en su cadena de transporte de electrones. Este trabajo se enfoca en un tipo novedoso de MFC, denominado celda de combustible microbiana de sedimento (SMFC), en el cual la energía eléctrica se produce a partir de la actividad metabólica de los microorganismos presentes en suelos o sedimentos. La energía en las SMFC se produce gracias a la capacidad de algunas bacterias (bacterias electroactivas) de transferir los electrones producidos a partir de la oxidación de los compuestos orgánicos del suelo a un ánodo. Hasta la fecha, pese a que los suelos o sedimentos son reservorios de biodiversidad muy importantes, pocos trabajos han estudiado las comunidades microbianas anódicas de este tipo de MFC y el aislamiento de microorganismos electroactivos a partir de estas MFC es casi nulo. En el presente trabajo se construyeron SMFC con dos suelos de arroz y un sedimento de laguna de nuestro país. Se estudió la comunidad microbiana presente en los ánodos por técnicas independientes de cultivo y se aislaron bacterias electroactivas de los ánodos. Se logró producir energía eléctrica a partir de los dos suelos de arroz y el sedimento de laguna, sin embargo, se observó una mejor performance para las SMFCs construidas con sedimento de laguna. La producción energética observada, se asoció a un cambio en la comunidad microbiana anódica, en la cual se detectó un enriquecimiento en microorganismos pertenecientes a la clase Deltaproteobacteria. Se lograron aislar 11 5 cepas electroactivas a partir del biofilm anódico. La producción de corriente de las cepas aisladas ensayada por cronoamperometría fue variable, observándose una mejor performance para las cepas DE5 (Shewanella sp.), DE3 (Pseudomonas sp.) y DE2 (Lysinibacillus sp.). El análisis por voltamperometría cíclica de estas cepas reflejó la presencia de un pico redox asociado a la transferencia electrones al ánodo. En base a los resultados obtenidos se propone profundizar el estudio de los mecanismos de transferencia electrónica al ánodo que presentan estas cepas, con el fin de lograr un mejor rendimiento energético.

El estudio de nuevos radiotrazadores para PET es un área mundialmente muy activa que requiere de un trabajo multidisciplinario y que implica desde la selección de los blancos moleculares, la síntesis orgánica, la marcación utilizando metodologías automatizadas que minimicen la irradiación del operador y el desarrollo de metodologías de evaluación fisicoquímica y biológicas compatibles con los cortos períodos de semidesintegración de los radionucleidos. Dentro de éstos últimos, el 11C ocupa un lugar fundamental por sus ventajas, pero también por sus limitaciones. El 11C permite realizar marcaciones isotópicas, con lo cual se logran radiotrazadores sin modificaciones fisicoquímicas significativas respecto a las moléculas endógenas cuyo metabolismo se desea estudiar. Por otra parte, el 11C puede ser obtenido con buenos rendimientos en los ciclotrones comercialmente disponibles, y su radioquímica ha sido bien estudiada y desarrollada a lo largo de las cuatro últimas décadas. La principal limitante de su uso radica fundamentalmente en el corto período de semidesintegración (20.4 minutos), y es lo que supone un desafío para el radioquímico que desarrolla métodos de marcación con 11C. La metodología más utilizada para introducir 11C en moléculas orgánicas a ser utilizadas como radiotrazadores PET, es a través de reacciones de 11C-metilación con los precursores radiactivos secundarios [11C]CH3I o [11C]CH3OTf. Estos precursores están disponibles en plataformas comerciales, su radioquímica ha sido bien estudiada y por ello su utilización es mundialmente muy amplia. Un gran número de radiotrazadores PET se sintetizan utilizando estos dos precursores radioactivos, y a la fecha se utilizan para la producción de todos los radiofármacos de 11C en uso preclínico y clínico en el Centro Uruguayo de Imagenología Molecular (CUDIM). Sin embargo, su aplicabilidad está limitada a moléculas nucleofílicas conteniendo los heteroátomos O, N o S. Otros precursores tales como el [11]CO2, [11C]CO, o el [11C]CN- permiten la obtención de moléculas marcadas en otros grupos funcionales, con lo que se amplía el espectro de estructuras conteniendo 11C y las aplicaciones de las mismas. Teniendo en cuenta lo anterior, es que en el presente trabajo se plantea la posibilidad de trabajar en el CUDIM con precursores de 11C alternativos a [11C]CH3I o [11C]CH3OTf. Como punto de partida del trabajo se propuso al [11C]CN- (comúnmente referido como [11C]HCN o [11C]NH4CN) como precursor radiactivo. En la Química Orgánica tradicional, el anión CN- es un sintón atractivo, pues permite la formación de un enlace C-C por sustitución nucleofílica sobre estructuras que presenten la función C-X (X = halógeno u otros grupos salientes). El grupo nitrilo también está presente en un importante número de fármacos y compuestos de interés biológico. El uso del precursor [11C]CN- permite obtener radiotrazadores atractivos para el diagnóstico mediante PET de alteraciones metabólicas y otros fenómenos patológicos. La síntesis de moléculas de importancia bioquímica tales como 1-[11C]aminoácidos 1-[11C]glucosa, 1-[11C]ácidos grasos, entre otros ha sido descrita en la literatura. CUDIM cuenta con una plataforma automatizada (módulo ProCab®) capaz de convertir químicamente el [11C]CO2 en [11C]CN-, por lo que el trabajo comenzó realizando un estudio de algunas variables de la producción de este precursor en nuestra plataforma. Se 20 determinó que la robustez de la misma depende fundamentalmente de su frecuencia de uso, y de finos ajustes en parámetros físicos tales como el flujo de los gases utilizados. En condiciones optimizadas, el módulo ProCab® es capaz de transformar [11C]CO2 en [11C]CN- con un rendimiento radioquímico de aproximadamente 30% (corregido) en no más de 10 minutos. El [11C]CN- así producido, se utilizó en un sistema modelo de reactividad, lográndose obtener D,L-1-[11C]Fenilalanina, utilizando para la misma una plataforma automatizada TRACERlab® FX C Pro. En una segunda etapa, se trabajó con [11C]CO2 como precursor radiactivo, de acuerdo a la metodología comúnmente conocida como “fijación-reducción de [11C]CO2”. La radioquímica de fijación o incorporación de [11C]CO2 es un terreno fértil para la obtención eficiente de radiotrazadores PET, al punto que es referida por algunos reconocidos investigadores como el “Renacimiento de la Química PET”. Tradicionalmente, el uso de [11C]CO2 se encontraba limitado a la reacción de Grignard para la síntesis de ácidos 11C-carboxílicos. Aun cuando esta técnica ha sido y es extensamente empleada, posee desventajas en cuanto a la inestabilidad inherente de los precursores orgánicos empleados, y la escasa variabilidad estructural de los radiotrazadores obtenidos. Recientemente ha habido avances significativos en la incorporación de 11C en moléculas orgánicas utilizando [11C]CO2, para dar estructuras moleculares tales como ureas, carbamatos, oxazolidinonas y ácidos carboxílicos, o su incorporación y posterior reducción para obtener especies N-11C-metiladas. Dado el alto número de radiofármacos de uso clínico que poseen el grupo 11C-metilo, se considera muy ventajoso sintetizarlos a partir de [11C]CO2 en lugar de los agentes 11C-metilantes tradicionales ([11C]MeI o [11C]MeOTf). El uso directo de [11C]CO2 y de modalidades de reacción “one-pot” minimiza el número de pasos sintéticos, y presupone un aumento de los rendimientos de síntesis, lo que se traduce en una mayor actividad final disponible para estudios preclínicos o clínicos. En tal sentido, se propuso estudiar condiciones para llevar a cabo y optimizar reacciones de incorporación de [11C]CO2 y su posterior reducción con el fin de contar con metodologías eficientes aplicables a la síntesis de 11C-radiotrazadores. Para el trabajo con [11C]CO2 se utilizó también la plataforma automatizada TRACERlab®, con mínimas modificaciones de tal manera de dirigir el [11C]CO2 purificado a un reactor de marcación. Se estudió la fisicoquímica de su captura en solución. Se evaluaron tres sistemas radioquímicos para promover la incorporación reductiva de [11C]CO2 en un sistema modelo de reactividad compuesto por 4-toluidina: i) DBU / PhSiH3, ii) IPr carbeno / PhSiH3 y iii) TBAF / PhSiH3. Para cada uno de ellos se estudió la influencia de factores físicos y químicos, tanto en la pureza radioquímica de la amina 11C-metilada como en su rendimiento radioquímico. Dos de ellos (IPr / PhSiH3 y TBAF / PhSiH3) se aplicaron a la síntesis del radiotrazador PET [11C]PiB. Mediante el uso de [11C]CO2 / TBAF / PhSiH3 en diglyme, a 150 ºC por 1 min, se logró marcar [11C]PiB en un 14.8% de rendimiento radioquímico, con actividad molar de 61 GBq/μmol. Si bien este valor es aparentemente bajo por el uso directo de [11C]CO2, el rendimiento radioquímico del [11C]PiB producido de esta forma duplica al del método de 11C-metilación con [11C]CH3OTf actualmente utilizado en CUDIM.
