A summary of the resource.
In this work different samples of Brazilian macauba oil obtained from mechanical pressing were characterized and production of esters of fatty acids using a catalyst-free continuous process under supercritical alcohols was assessed. Analysis of oil samples showed that the major fatty acid on pulp oil was oleic acid (mean value 62.8%), the amount of free fatty acid (FFA) was very high (37.4–65.4%), samples contained glycerides (7.4–16.5% TAG, 14.2–16.8% DAG and 1.0–3.4% MAG) and moisture was around 1.0%. Oil was processed in a continuous reactor using supercritical methanol or ethanol and the effects of temperature (573, 598, 623 and 648 K), pressure (10, 15 and 20 MPa), oil to alcohol molar ratio (1:20, 1:30 and 1:40), water concentration (0, 5 and 10 wt% added) and the flow rate of reaction mixture (1.0, 1.5, 2.0, 2.5 and 3.0 mL/min) on process efficiency were evaluated. The highest ester content achieved in reactions with supercritical methanol was 78.5% (648 K, 15 MPa, 1:30 oil:methanol molar ratio, 5 wt% water and 2.5 mL/min flow rate), while with supercritical ethanol was 69.6% (598 K, 15 MPa, 1:30 oil:ethanol molar ratio, 5 wt% water and 2.0 mL/min flow rate). The extent of the reaction was explored using a novel parameter, convertibility, which corresponds to the maximum ester content attainable from the feedstock. According to the convertibility of macauba pulp oil, the highest ester content corresponded to efficiencies of 98.0% and 86.9%, respectively. Results demonstrate that macauba oil might be a potential alternative for biodiesel production, though purification steps should be taken into account to achieve biodiesel specifications.

We report the synthesis, characterization, antibacterial and antifungal activities, phytotoxicity, and genotoxicity of two new complexes of silver(I) with sulfachloropyridazine (SCP), one of which is heteroleptic with SCP and SCN− ligands (Ag–SCP–SCN), the other of which is homoleptic (Ag–SCP); furthermore, the crystal structure of the homoleptic complex is disclosed. The heterocyclic N atom nearest to the Cl atom and the Nsulfonamide atom could be coordination sites for the silver ion in the Ag–SCP–SCN complex. The Ag–SCP complex is a polymeric compound with metal–metal bonds, and the heterocyclic and sulfonamide N atoms are points of coordination for AgI. Both complexes showed activity against all the tested bacteria, and in the cases of Escherichia coli and Pseudomonas aeruginosa, the action was better than that of SCP. In all cases, both silver–SCP complexes showed better antifungal activity than SCP, which was inactive against the tested fungi. Notably, the activity against P. aeruginosa, a nosocomial multidrug-resistant pathogen, was better than that of the reference antibiotic cefotaxim. Both silver–sulfa complexes displayed moderate activity against the tested yeast, especially for C. neoformans, which is an important fact considering the incidence of cryptococcosis, mainly in immune-deficient patients. No chromosomal aberrations were observed with the Allium cepa test, which is auspicious for further study of these complexes as potential drugs.

Leishmania major and Trypanosoma cruzi are the main causes of leishmaniasis and Chagas disease, two endemic parasitosis identified as neglected diseases by the World Health Organization. Fumarate reductase (FR) is a central enzyme in the conversion of fumarate to succinate, an energy releasing path essential for the survival of these protozoans which is also absent in their mammalian hosts. FR can thus be considered as a good candidate for targeting specific inhibition by new drugs designed against L. major and T. cruzi. The lack of tertiary structures available for LmFR and TcFR has limited until now the possibility of performing structure-based drug design. Here we used homology modeling combined with enzyme-cofactor docking to propose tertiary structures for NADH-dependent LmFR and TcFR using an homologous X-ray crystallographic structure of flavine-adenine dinucleotide (FAD) dependent FR from Shewanella frigidimarina (PDB ID: 1QO8) as template. These models were refined and stabilized with/without substrate in the active site using classical molecular dynamics simulations under quasi-physiological conditions. Structural features relevant for understanding the mechanism of action of the enzyme were also analyzed, with special attention to the hydrogen bond network involving the cofactor and water molecules present at the binding sites. A small set of compounds previously synthesized and assayed for their inhibitory capacity against TcFR ([M(mpo)₂] metal complexes with M=Pt(II), Pd(II) and V(IV)O and mpo=2-mercaptopyridine N-oxide) and LmFR (licochalcone A) were screened by protein-ligand docking using the NADH-LmFR and NADH-TcFR models here proposed and validated, gaining insight into their binding modes in each enzyme.

Se estudió la distribución enantiomérica de componentes quirales relevantes del aceite esencial proveniente de ejemplares de A. gratissima var. gratissima de la zona de Río Empedrado, Corrientes, Argentina, con el objeto de correlacionar variaciones detectadas en la rotación óptica de la esencia con su composición química. El aceite esencial, separado de ejemplares colectados en verano, caracterizado por un alto contenido en linalol (15%) y germacreno D (10%) mostró un exceso enantiomérico para enantiómeros levo, lo que está de acuerdo con la actividad óptica del aceite en este período vegetativo. Se evalúan implicancias en la valorización de este aceite.

Four strains of endophytic microorganisms isolated from carrot root were able to carry out the reduction of the carbonyl group with diverse degree of enantio-, and diasteroselectivity. Furthermore, biotransformation in the presence of bacterial inhibitor affects the stereochemical outcome of the reaction, and the concomitant addition of a yeast inhibitor results in a large decrease in the conversion percentage. These results indicate that endophytic microorganisms might be involved in the enantioselective reduction of ketones and ketoesters with fresh carrot root pieces.

A recent study on the Raman spectrum of the cysteine zwitterion and anion, and the 2:1 (Cys)2Zn complex was reanalyzed employing B3LYP/6-311??G (3df,2pd) calculations in a simulated water environment. The spectra were rediscussed in light of the apparent incorrect structure determined in the original paper for this complex. The complex turns out to be tetrahedral and tetracoordinated instead of octahedral hexacoordinated, as initially proposed. The calculated Raman spectrum of the complex agrees very well with the experimental data, showing that both the geometrical and electronic structures are well represented. Three metal–ligand bands are found, two of them involving mostly the symmetrical and asymmetrical stretching of the Zn–N and Zn–S bonds. They were measured at 334 and 296 cm-1 and calculated at 319 and 249 cm-1, respectively. The third band involves the stretching of Zn–S bonds but also skeletal vibrations of the ligand. This band, measured at 399 cm-1 and calculated at 444 cm-1, has been previously assigned incorrectly to a Zn–O bond which does not actually exists since the CO2 -1 fragments are located away from the Zn ion.

A summary of the resource.
We present a new application of the noncompetitive phage anti-immunocomplex assay (PHAIA) by converting an existing competitive assay to a versatile noncompetitive sandwich-type format using immunocomplex binding phage-borne peptides to detect the brominated flame retardant, brominated diphenyl ether 47 (BDE 47). Three phage-displayed 9-mer disulfide-constrained peptides that recognize the BDE 47–polyclonal antibody immunocomplex were isolated. The resulting PHAIAs showed variable sensitivities, and the most sensitive peptide had a dose–response curve with an SC50 (concentration of analyte producing 50% saturation of the signal) of 0.7 ng/ml BDE 47 and a linear range of 0.3–2 ng/ml, which was nearly identical to the best heterologous competitive format (IC50 of 1.8 ng/ml, linear range of 0.4–8.5/ml). However, the PHAIA was 1400-fold better than homologous competitive assay. The validation of the PHAIA with extracts of house furniture foam as well as human and calf sera spiked with BDE 47 showed overall recovery of 80–113%. The PHAIA was adapted to a dipstick format (limit of detection of 3.0 ng/ml), and a blind test with six random extracts of local house furniture foams showed that the results of the PHAIA and dipstick assay were consistent, giving the same positive and negative detection.

Abstract
A summary of the resource.
The main objective of present study was to investigate the influence of geographical region on the flavour compounds of various cold pressed lemon oils i.e. Argentinean, Ivory Cost, Italian, Spanish and Uruguayan. Among the flavor compounds, the major volatile flavour compounds from different chemical classes namely monoterpene hydrocarbons (i.e. limonene, sabinene plus β-pinene, γ-terpinene, myrcene, α-pinene and α-terpinolene), esters (i.e. neryl acetate and geranyl acetate), alcohol (i.e. α-Terpineol) and aldehyde compounds (i.e. geranial and neral) were composed of > 90% of total volatile flavor compounds of target cold pressed lemon oils. Uruguayan essential oil had the least concentration level of all major classes of flavor compounds except for alcohol compounds; while the highest content of all chemical classes of flavor compounds identified in this study. The principle component analysis (PCA) allowed us to discriminate various cold pressed lemon oil based on their origin resource thereby classify their sensorial attributes.

The total synthesis of (+)-bromoxone and a formal synthesis of (�)-bromoxone were performed in a simple chemoenzymatic manner, starting from bromobenzene, and using an enantiodivergent strategy. The usefulness of chiral cyclohexadienediols derived from the biotransformations of monosubstituted-aromatic compounds as building blocks for the preparation of natural epoxyenones was confirmed.

BACKGROUND: Traditionally, breeding research has been focused on increasing yield and fruit size, improving resistance to diseases, behaviour during transport and increasing postharvest shelf-life. However, consumers’ liking is one of the biggest challenges for strawberry breeding programs. In this context, the aim of the present work was to evaluate the application of a check-all-that-apply (CATA) question to study consumers’ perception of new strawberry cultivars. RESULTS: Average liking scores were low for a highly appreciated product like strawberries, which could be explained considering unfavourable weather conditions prior to harvest. However, despite the small differences in overall liking scores, significant differences were found in the frequency in which 14 out of the 22 terms of the CATA question were used to describe samples. This suggests that the evaluated CATA questionwas able to detect differences in consumers’ perception of the sensory characteristics of the evaluated cultivars.Moreover, a significant correlation was found between laboratory analyses (firmness, colour, soluble solids and acidity) and consumers’ responses to the CATA question, which indicates their validity. CONCLUSIONS: Considering results from the present study, the use of CATA questions could be a simple and interesting methodology to gain insight into consumers’ perception of different fruit cultivars.

In the search for new compounds with antitumor activity, coordination complexes with different metals are being studied by our group. This work presents the synthesis and characterization of six copper complexes with general stoichiometry [Cu(L-dipeptide)(phen)]·nH2O (were phen= 1,10-phenanthroline) and their cytotoxic activities against tumor cell lines. To characterize these systems, analytical and spectroscopic studies were performed in solid state (by UV–visible, IR, X-ray diffraction) including the crystal structure of four new complexes (of the six complexes studied): [Cu(Ala-Phe)(phen)]·4H2O, [Cu(Phe-Ala)(phen)]·4H2O, [Cu(Phe-Val)(phen)]·4.5H2O and [Cu(Phe-Phe)(phen)]·3H2O. In all of them, the copper ion is situated in a distorted squared pyramidal environment. The phen ligand is perpendicular to the dipeptide, therefore exposed and potentially available for interaction with biological molecules. In addition, for all the studied complexes, structural information in solution using EPR andUV– visible spectroscopieswere obtained, showing that the coordination observed in solid state ismaintained. The lipophilicity, DNA binding and albumin interaction were also studied. Biological experiments showed that all the complexes induce cell death in the cell lines: HeLa (human cervical adenocarcinoma),MCF-7 (human metastatic breast adenocarcinoma) and A549 (human lung epithelial carcinoma). Among the six complexes, [Cu(Ala-Phe)(phen)] presents the lowest IC50 values. Taken together all these data we hypothesize that [Cu(Ala-Phe)(phen)] may be a good candidate for further studies in vivo.

Geogenic As in groundwater has been recently studied in different aquifers of Uruguay. However, no background epidemiology studies are available regarding environmental population exposure. As a consequence, it was necessary to conduct scientific research in order to study arsenic as an environmental health issue in the country. This research needs have resulted in joint studies of experts from both the geosciences and biosciences. The groups have coordinated studies to assess the risks of exposure to environmental arsenic in Uruguay with common research objectives and, now, this is a matter of common interest through a multidisciplinary approach. The aim of this work is to present a review of these studies conducted in Uruguay.

Abstract
A summary of the resource.
We have identified phenazine 5,10-dioxides as prodrugs for antitumour therapy that undergo hypoxicselective bioreduction to form cytotoxic species. Here, we investigated some structural modifications in order to find new selective hypoxic cytotoxins and to establish the structural requirements for adequate activity. Three different chemical-series were prepared and the clonogenic survival of V79 cells on aerobic and anaerobic conditions was determined. Electrochemical- and DNA-interaction studies were done for the most relevant derivatives. The new fluoro-derivative 7-fluoro-2-aminophenazine 5,10- dioxide displayed selective toxicity towards hypoxic V79 cells having adequate hypoxic cytotoxicity ratio (HCR ¼ 6.8) and being the most potent hypoxic cytotoxins (P¼ 2.5 mM) described for this family of bioreductive agents. The reduction potential of the N-oxide moiety in this new fluoro-derivative was in the range for adequate bioreduction property. According to the fluorescence studies, the DNA-interaction mechanism was especially operative in the phenazine drugs more than in the corresponding prodrugs, phenazine dioxides.

Phenazine 5,10-dioxides are prodrugs for antitumor therapy that undergo hypoxic-selective bioreduction to form cytotoxic species. Here we investigate the expanded system benzo[a]phenazine 7,12-dioxides as selective hypoxic cytotoxin-scaffold. The clonogenic survival of V79 cells on aerobic and anaerobic conditions, conduct us to study antiproliferative activity on Caco-2 tumoral cells in normoxia. Electrochemical, DNA-interaction and DNA-damage studies were performed to establish the mode of action. The results demonstrated the potential biological properties of the studied scaffold being derivatives 6–10 structural hits for further chemical-modifications to become into therapeutics for solid tumors. Compounds 6 and 8 with cytotoxicity against V79 cells in both conditions (aerobia and anaerobia) were also cytotoxic against Caco-2 tumoral cells in aerobiosis.

En este trabajo se utilizaron diferentes métodos para la obtención de un concentrado de orizanoles a partir de las borras de neutralización del aceite de salvado de arroz. El contenido de orizanoles de las borras crudas fue de 2.4 %, el cual se incrementó a 3.8 % al eliminar el agua de las mismas mediante liofilización. Partiendo de las borras previamente liofilizadas y mediante diferentes métodos de extracción convencionales como Soxhlet y partición con diferentes mezclas de solventes fue posible alcanzar concentrados con un contenido de orizanoles de hasta un 17.6 %, conteniendo además una proporción importante de ácidos grasos libres. Cuando las borras de neutralización fueron sometidas previamente a una etapa de etanólisis, esterificándose completamente los ácidos grasos libres, fue posible obtener un concentrado con un contenido de orizanoles de 31.3 % (lo que implica una concentración en un factor de 13) y un porcentaje de recuperación del 93% . La modificación química de las borras mediante etanólisis y la posterior extracción con solventes ofrece un método relativamente simple para la preparación de un concentrado de orizanoles atractivo para su utilización como tal en el área farmacéutica o como aditivo de potencial antioxidante en la industria de los alimentos.

A simulated therapeutic equivalence scenario was created between two multisource drug products (tablets marketed in Uruguay) containing a poorly water-soluble compound, flunitrazepam, and the reference product (Rohypnol®). The simulated study was built from an in vitro - in vivo dissolution-pharmacokinetic C-level (IVIV) correlation and from an in vivo pharmacokinetic-pharmacodynamic (PKPD) correlation, using experimental and bibliographic data. In vitro parameters were experimentally obtained from the dissolution profile for each commercial brand at the three biorelevant dissolution media (pH 1.2, 4.5 and 6.8) whereas in vivo parameters were acquired from the bibliography. From IVIV correlation plasma drug concentration-time profiles were simulated. These concentrations were used to simulate the intensity of drug effect throughout time in order to predict relative clinical responses of each brand. It could be concluded that from the three brands of flunitrazepam marketed in Uruguay, one was revealed as having a very poor sedative effect based on its simulated low pharmacodynamic-time profile, reinforcing hence the ineffective clinical experience claimed by physicians.

Context can be regarded as the situational and temporal conditions in which food choice occurs. Although context has a large effect on consumer perception and choice of food products, it has been mostly overlooked in research exploring the determinants of consumers' food choice. The aims of the present work were to study the influence of purchase and consumption context on motives underlying food choice and to evaluate if gender and age modulate this effect. One hundred and ninety-five consumers from Montevideo (Uruguay) participated in an online survey. They were asked to complete a free-listing task about all the things they take into account when selecting foods in four contexts: purchase, lunch, snacking and dinner. Consumer responses were qualitatively analyzed and grouped into categories and their cognitive salience was determined. Categories were grouped into 10 dimensions: price, health/nutrition, perceived quality, freshness, sensory characteristics, hedonics, convenience, context, variety and satiety. Context had a strong influence on the motives underlying consumer food choices. The dimensions with the highest cognitive salience in the purchase context were price, perceived quality and freshness, while convenience and variety were the most relevant dimensions for the consumption contexts. The effect of context on the salience of the elicited categories was not markedly affected by gender and age.

In the search for new therapeutic tools against tuberculosis and to further address the therapeutic potential of pyridine-2-thiol 1-oxide (Hmpo) metal complexes, two new octahedral [M(III)(mpo)3] complexes, with M = Ga or Bi, were synthesized and characterized in the solid state and in solution. Attempts to crystallize [Ga(III)(mpo)3] in CH2Cl2 led to single crystals of the reaction product [GaCl(mpo)2], where the gallium(III) ion is in a square basis pyramidal environment, trans-coordinated at the basis to two pyridine-2-thiolato 1-oxide anions acting as bidentate ligands through their oxygen and sulfur atoms. The biological activity of the new [M(III)(mpo)3] complexes together with that of the previously reported Fe(III) analogous compound and the pyridine-2-thiol 1-oxide sodium salt (Na mpo) was evaluated on Mycobacterium tuberculosis. The compounds showed excellent activity, both in the standard strain H37Rv ATCC 27294 (pan-susceptible) and in five clinical isolates that are resistant to the standard first-line anti-tuberculosis drugs isoniazid and rifampicin. These pyridine-2-thiol 1-oxide derivatives are promising compounds for the treatment of resistant tuberculosis.

Expanding our previous work on salicylaldehyde semicarbazone metal complexes as prospective anti-trypanosomal agents, five new fac-ReI(CO)3-containing complexes with ligands of this semicarbazone series were synthesized and characterized. An atypical coordination mode of these potentially tridentate ligands through only the carbonylic oxygen and the azomethine nitrogen (the so-called N,O fashion) was demonstrated by IR spectroscopy and supported by theoretical calculations. Three of the compounds showed moderate in vitro anti-Trypanosoma cruzi activity and increased activity with respect to the corresponding free ligands. The brominated ligands, 5-bromo-2-hydroxybenzaldehyde semicarbazone (L2) and 5-bromo-2-hydroxy-3-methoxybenzaldehyde semicarbazone (L5), led to the most active rhenium(I) complexes. These compounds are among the few reported examples of rhenium complexes bearing in vitro activity against T. cruzi.

In the search for new agents against Trypanosoma cruzi and cancer the effect of the nature of the metal center and of the presence of a polypiridyl coligand on the antitrypanosomal and antitumoral activities of selected N,N,O ligands [2-(benzothiazol-2-yl-hydrazonomethyl phenol (HL1) and 2-(benzothiazol-2-yl-hydrazonomethyl)-6-methoxyphenol (HL2)] is explored. The new complexes [VVO2(L1)], [VVO2(L2)], [VIVO(L1-H)(phen))] and [GaIII(L2)2](NO3) were synthesized and characterized by using different techniques. The stability of the vanadium complexes in solution was investigated by EPR and 51V NMR spectroscopies and that of the gallium compound by UV–Vis spectroscopy, 1H NMR and conductivity measurements. While the vanadium complexes show high stability in DMSO, the gallium complex shows very good stability in DMSO and moderate stability in aqueous – DMSO medium. <br /><br />The cytotoxicity on human tumor cell lines (ovarian A2780, breast MCF7 and prostate PC3 cell lines) that show different sensitivity to cisplatin was evaluated. All the compounds evidenced antiproliferative activity in the micromolar range. The highest cytotoxic activity, in molar units, is shown by [GaIII(L2)2](NO3) (IC50: 1.7 μM) and [VIVO(L1-H)(phen)] (IC50: 2.7 μM) against the ovarian cancer cells. With the exception of [VVO2(L1)], the cytotoxic activity of the ligands and complexes is similar to that of cisplatin in A2780 cells and surpass cisplatin in the other tumor cells. Regarding the activity on T. cruzi, [VIVO(L1-H)(phen)] showed a 10-fold decrease of IC50 with respect to HL1 and an IC50 value (10.7 μM) in the same order of that of the antitrypanosomal drug Nifurtimox (IC50: 6.0 μM). HL2 showed significant growth inhibitory effect on the parasite (IC50: 23.5 μM) and its coordination to Ga(III) lead to a 2-fold increase in activity in molar units (IC50: 14.2 μM). In order to explain the high inhibitory activity of [VIVO(L1-H)(phen)] against the parasite and the tumor cells, the interaction of this compound with plasmid DNA was preliminarily evaluated by AFM. The corresponding images suggest that DNA may be considered as a potential target.

Mate (MT) is a popular South American beverage that has been used as a traditional medicine for centuries, spurring recent interest in its nutraceutical properties.MT is prepared as an infusion of leaves fromthe Yerba Mate (llex paraguriensis) tree. MT has been reported to have antioxidant properties in vitro and in vivo, but these have not been fully characterized in terms of effects against specific radicals. Accordingly, we examined the antioxidant effects of an MT infusion against hydroxyl and superoxide radicals in both chemical and cell culture assays. MT infusions were prepared at 3.10 g/L in boiling water and diluted to experimental dilutions from this stock. Electron spin resonance (ESR) experiments indicated that MT scavenged hydroxyl radicals (produced via the Fenton reaction) and superoxide radicals (produced via the xanthine/xanthine oxidase enzymatic reaction) at all concentrations tested (P <0.05). Further controls indicated that superoxide radical scavengingwas not due to xanthine oxidase inhibition. MT scavenged hydroxyl radicals and decreased cellular oxygen consumption in a dose-dependent manner in Cr(VI)-stimulated RAW 264.7 cells, based on ESR and oxygraph measurements (P < 0.05). Similarly, MT also inhibited hydroxyl-radical-induced lipid peroxidation and DNA damage in a dose-dependent manner in RAW 264.7 cells, based on malondialdehyde and Comet assay data (P < 0.05). This study indicates thatMT possesses potent antioxidant effects against hydroxyl and superoxide radicals in both chemical and cell culture systems, as well as DNA-protective properties. These data further clarify the reported antioxidant effects of YerbaMate infusions. Mate (MT) is an infusion brewed from dried leaves and stemlets of the Yerba Mate (llex paraguriensis) holly tree that grows in the southern portion of the Paraguay and Parana River basins (Filip and others 2001). The use of MT as a traditional medicine by the native peoples of Paraguay and Brazil predates the arrival of Spanish and Portuguese explorers. MT is used as a traditional medicine to treat conditions ranging from arthritis to digestive disorders (Bixby and others 2005; Bastos and others 2007). More recent studies have reported that MT has antioxidant and antiobesity properties in humans, while DNA-protective and cholesterol-lowering properties have been reported in animal studies. (Heck and de Mejia 2007; Moraes-de-Souza and others 2008; Matsumoto and others 2009). Microencapsulation of the Schinus molle Rev L. (Anacardiaceae) leaves essential oil (EO) has been employed to control the release of active ingredients, protecting them from the external environment, with the concurrent improvement of its insecticidal potential on Haematobia irritans. Four microcapsule formulations (EEO1 to EEO4) of S. molle EO were prepared by spray-drying, using a mini spray dryer and gum Arabic/maltodextrin (AG/MDX) as the carrier in different proportions, at a ratio of 4:1 (MDX/AG:EO). Encapsulation efficiency (EE: 96–100%), powder morphology and particle size distribution were analyzed as responses. An interesting correlation was found between EO free and microcapsules (EEO) in the preliminary and comparative studies of stability (at 45 °C) and in the insecticide activity on H. irritans. In fact, a very slow liberation profile of the microencapsulated EO (EEO4) was observed over a period of 366 h (71% of EO retention), as well as a slower time-dependent insecticide effect (32 and 73% of dead flies at 2 and 4 h of exposure time) compared to the free EO (96% of dead flies at 2 h).
