Polychlorinated dibenzo-p-dioxins (PCDD) and dibenzofurans (PCDF) are persistent, toxic, and bioaccumulate in the environment. Due to their high analytical costs, these compounds are hardly regulated and mostly not monitored in the Third World. To overcome this, bioassays have been proposed as low-cost alternative methods. Two of the most established bioanalytical tools, the dioxin antibody-based enzyme-linked immunosorbent assay ELISA and the micro-EROD bioassay are evaluated and compared to high resolution gas chromatography and high resolution mass spectrometry (HRGC/HRMS) analytical methodology. The methods were tested using thirteen soils and sediment samples selected from diverse sites in Montevideo, Uruguay. The WHO₂₀₀₅ total toxic equivalent (WHO₂₀₀₅-TEQ) of soils ranged from 2.4 to 2212 (ng WHO₂₀₀₅-TEQ/kg dry sample) and from 0.14 to 9.4 (ng WHO₂₀₀₅-TEQ/kg dry sample) in sediments. This study shows significant contamination related to dioxin-like compounds, particularly in sites where uncontrolled burnings were carried out. ELISA and micro-EROD bioassay correlated well with HRGC/HRMS, R Spearman 0.773 and 0.913, respectively and were highly correlated to each other, R Spearman 0.879. Preliminary threshold values of bioassay toxic equivalents of 330 (ng/kg dry sample) for the micro-EROD bioassay and 220 (ng/kg dry sample) for ELISA are proposed.

Con un extracto crudo (EC) de importante actividad proteolítica, obtenido a partir de frutos maduros de Bromelia antiacantha se trataron epiginos de Apopyllus iheringi a fin de remover, mediante hidrólisis, los tejidos blandos presentes en estas estructuras. La acción enzimática sobre las muestras fue evaluada en distintas condiciones de: pH, actividad y tiempo de biocatálisis. Luego de la acción biocatalítica, se observaron las muestras bajo aumento, tomándose registros fotográficos. De todas las condiciones evaluadas, los mejores resultados de biocatálisis fueron obtenidos a pH 6.0, condición óptima de actividad de estos biocatalizadores con sustratos solubles. La digestión tisular completa, observada a través del Microscopio Electrónico de Barrido, se obtuvo a pH 6.0, 42ºC, actividad específica de 308.3 UE/ mg y 8 h de incubación. Utilizando las mismas condiciones de catálisis fueron tratados epiginos de otras dos especies de arañas obteniendo muy buenos resultados.

In this work, highly reactive carbon–iron composites were prepared using a waste, i.e. tar, as carbon precursor and a simple iron oxide, i.e. hematite. Tar was impregnated on Fe2O3 with different tar/hematite weight ratios of 1:1; 2:1 and 4:1, and thermally treated under N2 atmosphere (400◦C, 600◦C and 800◦C). Mössbauer, XRD andmagnetizationmeasurements suggested that treatment at 400◦C and 600◦C produces Fe3O4 but treatment at 800◦C produced mainly Fe◦. Raman and TG analyses of the different composites suggested the formation of carbon contents of 18, 24 and 32 wt.% as amorphous and graphitic highly dispersed on the Fe surface. The composites obtained at 800◦C showed high efficiency to reduce Cr(VI) as CrO2− 4 in aqueous medium with much better results compared to finely ground commercial Fe◦.

Direct ethanol fuel cells are based in ethanol electro-oxidation at low temperature and they constitute an alternative energy conversion system. However, they need catalysts in order to improve their efficiency, given that ethanol electro-oxidation is a slow-kinetic reaction. Among those catalysts platinum and platinum alloys play an important role in the increase of the ethanol cleavage kinetics for fuel cell application. However, to maximize the catalyst performance, support materials are needed in order to reduce the catalyst load considering its high cost. One of the more versatile catalyst supports is activated carbon. Recently, attention has focused on wood as carbon material precursor, because of its sustainability and also because the obtained carbons have excellent final properties. In the present work, activated carbon obtained by physical activation of Eucalyptus grandis wood (biocarbon) was tested as Pt and PtSn catalyst support, for ethanol electro-oxidation reaction. For comparison purposes, commercial activated carbon Vulcan XC72 was also tested. The catalyst supports were characterized by textural analysis, elemental analysis and infrared spectroscopy. The obtained catalysts were characterized regarding structure by XRD and their electrochemical behavior was evaluated by cyclic voltammetry. Biocarbon-supported PtSn electrocatalysts showed better electrochemical performance related to the commercial activated carbon (Vulcan XC72)-supported ones, since its developed current density and potential were the highest and its onset potential was the lowest. However, pure platinum showed better values for current density, potential and onset potential in Vulcan XC72-based activated carbons, being the untreated one the best support in this case.

Small compounds cannot bind simultaneously to two antibodies, and thus, their immunodetection is limited to competitive formats in which the analyte is indirectly quantitated by measuring the unoccupied antibody binding sites using a competing reporter. This limitation can be circumvented by using phage-borne peptides selected for their ability to specifically react with the analyte–antibody immunocomplex, which allows the detection of these small molecules in a noncompetitive format (PHAIA) with increased sensitivity and a positive readout. In an effort to find substitutes for the phage particles in PHAIA, we explore the use of the B subunit of the Shiga-like toxin of Escherichia coli, also known as verotoxin (VTX), as a scaffold for multivalent display of anti-immunocomplex peptides. Using the herbicides molinate and clomazone as model compounds, we built peptide–VTX recombinant chimeras that were produced in the periplasmic space of E. coli as soluble pentamers, as confirmed by multiangle light scattering analysis. These multivalent constructs, which we termed nanopeptamers, were conjugated to a tracer enzyme and used to detect the herbicide–antibody complex in an ELISA format. The VTX–nanopeptamer assays performed with over a 10-fold increased sensitivity and excellent recovery from spiked surface and mineral water samples. The carbon black-labeled peptide–VTX nanopeptamers showed great potential for the development of a lateral-flow test for small molecules with a visual positive readout that allowed the detection of up to 2.5 ng/mL of clomazone.

Stereoselective hydrogenation of methylcyclohex-2-ene-1,4-diols used as important intermediates for the preparation of ampelomins and deoxy-carbasugars was studied. These olefins were obtained in few steps from a chiral cis-diol resulting from microbial oxidation of toluene. Although the stereoselective hydrogenation of this type of substrates is difficult, high yields were obtained for heterogeneous hydrogenation using Adam’s catalyst, where steric hindrance controlled the stereochemical outcome of the process. On the other hand, for homogeneous hydrogenation of similar olefins using Crabtree’s catalyst, coordination with the allylic alcohols allowed for a controlled hydrogen addition from the more hindered face. In this manner two protocols for the hydrogenation of these types of substrates resulting in complementary stereoselectivities are described.

The application of check-all-that-apply (CATA) questions for sensory product characterisation is gaining acceptance and popularity. This question format has been reported to be a quick and reliable means of gathering sensory profiles from consumers, concurrently with hedonic assessment. However, a limitation of CATA questions is that they do not encourage deep processing by respondents. Forced-choice questions, where respondents answer “yes” or “no” for each term, may encourage systematic processing and be useful when consumers undertake sensory profiling tasks. This research compared sensory profiles elicited by consumers using CATA questions or forced-choice Yes/No questions and contribute to ongoing investigations of CATA questions and related question formats with a view to developing guidelines for best practise. Across seven consumer studies with 600+ consumers and multiple product categories, consistent evidence was obtained that forced-choice Yes/No questions are associated with higher term citation frequencies. However, this did not consistently translate into greater product discrimination. Conclusions regarding similarities and differences amongst samples and the stability of sample and term configurations were generally independent of question format (i.e., whether the sensory data were elicited by CATA or forced-choice Yes/No questions). Overall, the comparison of CATA and forced-choice Yes/No questions for sensory characterisation suggested parity of the two question formats. This extended to consumers’ perceived difficulty and tediousness for completing the test. Regardless of question format, consumers, on average, perceived the tests as easy and not tedious.

In response to the growing use of consumers for sensory product characterisation, methodological research contributing to development of best practise guidelines is ongoing. We focus here on concurrent elicitation of hedonic and sensory product characterisation by check-all-that-apply (CATA) questions. Jaeger et al. (2013b) reported that CATA questions only caused weak and transient bias of co-elicited hedonic scores. In the current research six studies were conducted, in which more than 700 consumers took part. Five product categories were tested (rice crackers, lite bread, cheese, kiwifruit, black currant drinks) with 4–7 samples per study. In none of these studies was evidence obtained suggesting bias of hedonic scores and it is now possible to conclude with greater certainty that co-elicitation of hedonic scores and product attribute information using CATA questions is unlikely to bias hedonic scores. A second result of the current research was that the use of designs that rotate presentation order of CATA terms was not associated with hedonic bias, and neither was the use of the forced Yes–No CATA question format. In future research, in light of a strong dominance of positive CATA terms used in these studies, we recommend studying more thoroughly the influence of positive/negative/neutral words in CATA lists as a possible source of hedonic bias. An exploratory component to this research suggested that consumers perceive the concurrent elicitation of hedonic and CATA responses as easy, but that too many samples may make the task tedious.

La selenocisteína (Sec) es incorporada a la cadena polipeptídica mediante un mecanismo no canónico: una secuencia de incorporación de Sec (SECIS) presente en el ARNm de la selenoproteína reprograma al codón UGA para incorporar Sec. Si bien este mecanismo ha sido completamente elucidado en bacterias, en eucariotas aún restan aspectos por comprender. La proteína de unión al elemento SECIS (SBP2) media la incorporación de Sec, sin embargo los roles que cumplen sus dominios y el impacto de su deficiencia en la inserción de Sec no se conoce completamente. Por otra parte, existen genes que se han reportado como involucrados o presuntamente involucrados en la decodificación e incorporación de Sec cuyas funciones no han sido esclarecidas. Para estudiar algunos de estos aspectos, utilizamos Caenorhabditis elegans como modelo ya que se trata de un organismo eucariota simple que posee toda la maquinaria de incorporación de Sec a disposición de una única selenoproteína: la tiorredoxina reductasa citosólica (TRXR-1). Todas las SBP2 descritas hasta el inicio de esta tesis poseían un dominio de unión al ARN (RBD) y uno de incorporación de Sec (SID). Mediante análisis in silico identificamos la SBP2 de C. elegans y, sorprendentemente, encontramos que carece de SID y que consiste únicamente en un RBD. Generamos una estirpe mutante en sbp2 que resultó incapaz de incorporar Sec, demostrando así que este gen es esencial para este fin. El análisis de genomas de otros nematodos reveló que la ausencia de SID es característica del linaje y que el mantenimiento de la incorporación de Sec se encuentra ligado a la TRXR-1. Se deduce entonces que el mecanismo de incorporación de Sec en este filo difiere, al menos en parte, del ya reportado para otros eucariotas; SID podría ser completamente prescindible, u otra proteína, no identificada aún, podría suplir sus funciones. Encontramos también que los nematodos del clado Tylenchina han perdido la capacidad de incorporar Sec. Estos son los primeros animales no artrópodos que se reportan que perdieron esta característica; a nivel genético perdieron todos los genes involucrados en su decodificación, excepto SecP43, que se presume tendría un rol adicional a la metilación de una base del anticodón del ARNtSec y el transporte de complejos supramoleculares asociados a los ARNm de selenoproteínas. Mediante ensayos con una estirpe mutante en SecP43 encontramos que si bien no este gen no es esencial para la incorporación de Sec en condiciones óptimas de laboratorio, es necesario para la correcta expresión de TRXR-1 en condiciones desafiantes. 4 Análogamente, demostramos que R13A5.9 , un gen asociado a la incorporación de Sec, pero de función desconocida, es requerido para la incorporación de Sec en condiciones desafiantes. Así, es probable que R13A5.9, como SecP43, tenga funciones regulatorias en el metabolismo de selenio y/o la incorporación de Sec. Globalmente, nuestros resultados indican que C. elegans y el linaje nematodo proveen indicios claves para comprender el mecanismo de incorporación de Sec y la evolución de la utilización de Sec y de los genes asociados a su incorporación, así como de las selenoproteínas y los selenoproteomas. Finalmente nuestros estudios mostraron evidencia de inicio no canónico de la traducción en C. elegans , mecanismo que no había sido previamente descrito en este organismo modelo. Se identificaron dos genes (uno de ellos sbp2 ) cuya traducción no iniciaría en AUG sino en AUU. Se prevé que el número de transcriptos que tengan inicio no canónico de la traducción sea aún mayor.

Several disciplines like Environmental Toxicology, Epidemiology, Public Health and Geology have been the basis of the development of Medical Geology in Uruguay during the last decade. The knowledge and performance in environmental and health issues have been improved by joining similar aims research teams and experts from different institutions to face environmental problems dealing with the population’s exposure to metals and metalloids and their health impacts. Some of the Uruguayan Medical Geology examples are reviewed focusing on their multidisciplinary approach: Lead pollution and exposed children, selenium in critically ill patients, copper deficiency in cattle and arsenic risk assessment in ground water. Future actions are also presented.

Psoralen interaction with two models of DNA was investigated using molecular mechanics and molecular dynamics methods. Calculated energies of minor groove binding and intercalation were compared in order to define a preferred binding mode for the ligand.We found that both binding modes are possible, explaining the low efficiency for monoadduct formation from intercalated ligands. A comparison between the interaction energy for intercalation between different base pairs suggests that the observed sequence selectivity is due to favorable intercalation in 5�-TpA in (AT)n sequences.

myo-Inositol phosphates possessing the 1,2,3-trisphosphate motif share the remarkable ability to completely inhibit iron-catalysed hydroxyl radical formation. The simplest derivative, myo-inositol 1,2,3-trisphosphate [Ins(1,2,3)P3], has been proposed as an intracellular iron chelator involved in iron transport. The binding conformation of Ins(1,2,3)P3 is considered to be important to complex Fe3+ in a ‘safe’ manner. Here, a pyrene-based fluorescent probe, 4,6-bispyrenoyl-myo-inositol 1,2,3,5-tetrakisphosphate [4,6-bispyrenoyl Ins(1,2,3,5)P4], has been synthesised and used to monitor the conformation of the 1,2,3-trisphosphate motif using excimer fluorescence emission. Ring-flip of the cyclohexane chair to the penta-axial conformation occurs upon association with Fe3+, evident from excimer fluorescence induced by p–p stacking of the pyrene reporter groups, accompanied by excimer formation by excitation at 351 nm. This effect is unique amongst biologically relevant metal cations, except for Ca2+ cations exceeding a 1 : 1 molar ratio. In addition, the thermodynamic constants for the interaction of the fluorescent probe with Fe3+ have been determined. The complexes formed between Fe3+ and 4,6-bispyrenoyl Ins(1,2,3,5)P4 display similar stability to those formed with Ins(1,2,3)P3, indicating that the fluorescent probe acts as a good model for the 1,2,3-trisphosphate motif. This is further supported by the antioxidant properties of 4,6-bispyrenoyl Ins(1,2,3,5)P4, which closely resemble those obtained for Ins(1,2,3)P3. The data presented confirms that Fe3+ binds tightly to the unstable penta-axial conformation of myo-inositol phosphates possessing the 1,2,3-trisphosphate motif.

Objective: Systemic inflammatory response syndrome is characterized by increased urinary excretion of selenium and low serum concentration. Repletion by parenteral selenite is the most efficacious form of supplementation. However, the optimum safe dose and mode of administration remain controversial. We aimed to determine pharmacokinetic and pharmacodynamic profiles of selenite and estimate a safe dose to optimize selenium status. Methods: A prospective, randomized, pilot study in 20 patients with systemic inflammatory response syndrome compared a high-dose (HD) group that received a loading dose of selenium as selenite 15.18 mmol over 2 h and thereafter 10.12 mmol/d as a continuous intravenous infusion (CIV) for 10 d with a very–high-dose (VHD) group that received a loading dose of 25.30 mmol over 2 h and thereafter 20.24 mmol as a CIV for 10 d. Clinical outcome was evaluated by length of stay in the intensive care unit, incidence of ventilator-associated pneumonia, and Sequential Organ Failure Assessment score. Results: Patients in group HD (n ¼ 10, age 54 � 23 y) had an Acute Physiology and Chronic Health Evaluation II score of 23 � 5 and a Sequential Organ Function Assessment score of 10 � 2. Those in group VHD (n ¼ 10, age 41 � 19 y) had scores of 21 � 7 and 8 � 3, respectively. Pharmacokinetic concentration/time curves for serum selenium overlapped but were independent of dose, whereas the pharmacodynamics were different, showing maximum glutathione peroxidase activity only with VHD. Glutathione peroxidase decreased after day 7 independently of the selenium dose. Clinical outcomes were similar in both groups. A summary of the resource.
By means of coupled cluster and density functional M06-2X calculations we have studied the directionality and energetics of the hydrogen bond (HB) between halides and the following H donors: methane, ethylene, and acetylene. The largest interaction energy was obtained for acetylene, the molecule in which the hydrogen atoms present the largest positive charge. For the complex [X���HCCH]-, fluoride presented a different behavior since acetylene donates a hydrogen to fluoride and the [HF���CCH]- complex was formed. We found that for ethylene there is a strong competition between the monodentate- and bidentate-like structures. Because of its small size, fluoride leads to a monodentate complex with C2H4. However, as we move down in the periodic table, the bidentate-like structure becomes more stable. In effect, for chloride and bromide they are nearly isoenergetic, while for iodide the bidentate-like complex is formed. These present results question previous investigations about the directionality of the HB in halide–ethylene complexes

A summary of the resource.
The objective of this work was to obtain water-in-oil (w/o) emulsions with polyglycerol polyricinoleate (PGPR) as emulsifier and to study the effect of the addition of calcium in the dispersed aqueous phase on the stability of these systems. Emulsions were formulated with 0.2, 0.5 and 1.0% w/w PGPR and 10% w/w water containing calcium chloride at varied concentrations or other salts (calcium lactate or carbonate; sodium, magnesium or potassium chloride). The stability of these systems was studied with a vertical scan analyzer during 15 days; coalescence and sedimentation were observed as simultaneous destabilization processes. The increase of PGPR concentration and/or calcium chloride content gave more stable emulsions. The stabilizing effect of calcium salt was attributed to the diminution of the water droplets size, the decrease of the attractive force between water droplets and the increase of the adsorption density of the emulsifier. The viscoelastic parameters of the interfacial film were decreased with increasing calcium and PGPR concentrations. Calcium chloride produced a higher increase of stability than calcium salts with lower dissociation degree. The presence of any assayed salt in the aqueous phase also allowed the stabilization of w/o emulsions with higher water contents.

Biological monitoring of benzene exposure is indicated for petrochemical workers, gas stations, underground car parks, garages and in the general population affected by environmental pollution. Urinary metabolites currently referred as biomarkers are phenylmercapturic acid and trans trans mucónico (tt-mucónico) acid. The aim of this work is to develop an analytical methodology to determine urinary tt- mucónico acid by gas chromatography (GC) with FID detector and make systematic use of a technique accessible by GC as an alternative to High Performance Liquid Chromatography (HPLC) widely released for this analysis in the literature. It sets guidelines for sampling of urine, which is subjected to a pretreatment with liquid-liquid extraction for subsequent chemical transformation or derivatization obtaining a volatile compound that is then put to the quantification on the GC. Analytical conditions are established to achieve the levels of acceptance of the validation parameters of the technique. The value recommended by the ACGIH 2009 for the exposed population is 500 µg/g creatinine in urine. Urine samples from workers and people belonging to non-exposed population (controls) are analyzed. Results: The results show that the developed method is suitable for monitoring exposure of workers exposed to benzene. It was noted that 10% of the cases studied the value found tt-mucónico acid in urine exceeds the value recommended by ACGIH.

Abstract
A summary of the resource.
The crystal structure and the vibrational spectrum of a potential drug for Chagas‘s disease treatment, the (E)-isomer of phenylethenylbenzofuroxan 1 (5(6)(E)- [(2-phenylethenyl)]benzo[1,2-c]1,2,5-oxadiazole N-oxide), are reported. In order to provide insights into structural relationships, quantum mechanical calculations were employed starting from crystal structure. These results have given theoretical support to state interesting structural features, such as the effect of some intermolecular contacts on the molecule conformation and the electronic delocalization decreasing through atoms of the benzofuroxan moiety. Furthermore, the MOGUL comparative analysis in the Cambridge Structural Database provided additional evidences on these structural behaviors of compound 1. Intermolecular contacts interfere on the intramolecular geometry, as, for instance, on the phenyl group orientation, which is twisted by 12.32(6)◦ from the ethenylbenzofuroxan plane. The experimental Raman spectrum of compound 1 presents unexpected frequency shift and also anomalous Raman activities. At last, the molecule skeleton deformation and the characteristic vibrational modes were correlated by matching the experimental Raman spectrum to the calculated one.

<strong>Development of second generation amidinohydrazones, thio- and semicarbazones as Trypanosoma cruzi-inhibitors bearing benzofuroxan and benzimidazole 1,3-dioxide core scaffolds.</strong>
Abstract
A summary of the resource.
Trypanosoma cruzi is the causative agent of Chagas’ disease. The thiosemicarbazone moiety as a pharmacophore has been described for inhibition of the essential cysteine protease, cruzipain, of this parasite. Our recent study identified an amidinohydrazone containing benzofuroxan as a hit compound for cruzipain inhibition with trypanosomicidal activity. Structural modification of the amidinohydrazone, thio- and semicarbazone motifs, using benzofuroxan and including a benzimidazole 1,3-dioxide system as new core scaffolds are described. These changes allowed for the identification of new structural motifs with desired antitrypanosomal activity. The new amidinohydrazone, thio-, and semicarbazone derivatives had excellent anti-trypanosomal activity without improved cruzipain-inhibitory activity compared with the parent compounds. Relevant structural features of these derivatives for further modification have also been determined.

Details are presented for both programs, as well as primary results since their beginning in 2004. The general impact on staff members is discussed.

A summary of the resource.
An increasing share of fine wines is marketed through retail stores. In this context, packaging plays a major role in attracting consumers’ attention and influencing their purchase decisions. The aim of the present work was to identify a specific bottle for Tannat red wine, the typical Vitis vinifera grape of Uruguay, by studying the effect of bottle shape and colour on consumers’ sensory expectations and willingness to purchase. A consumer study was carried out with 64 fine wine consumers, who were first introduced a list of bottle colours and shapes and were asked to answer a check-all-that-apply question. Then, participants were asked to complete a conjoint analysis task about Tannat wine bottles. Results from the check-all-that-apply question showed that consumers associated the shape and colour of bottles with specific types and styles of wine. Conjoint analysis suggested that Bordeaux dark green or dark gray bottles might be the most appropriate for Tannat wine. Interestingly, results shown here may indicate the existence of risks if deep innovation is made in bottle shape designs, since consumers clearly preferred Bordeaux bottles for red wines.
