Herein we report the synthesis, characterization, and photophysical and biological evaluation of the complexes Ln(DBM)3(RPhen) (Ln = Sm, R = H; Ln = Sm, Eu, Tb, R = 5-NH2) stabilized by three β-diketonate units (DBM) and a phenanthroline (RPhen) derivative, with the aim of contributing to the development of lanthanide-based compounds with potential application as anticancer agents. The UV-vis spectra of [Sm(DBM)3(Phen)], [Sm(DBM)3(NH2Phen)], [Eu(DBM)3(NH2Phen)] and [Tb(DBM)3(NH2Phen)] measured in DMSO and PBS showed a strong absorption band centered at ca. 350 nm in both solvents. In DMSO, all lanthanide compounds except [Sm(DBM)3(Phen)] show a ligand centered emission band at ca. 520 nm. In PBS only sharp emission peaks are detected. The complexes show similar cytotoxic effects in A2780 ovarian cancer cells, presenting IC50 values at 24 h in the range 16–27 μM. The measurement of the cellular uptake of the complexes in the A2780 cells by inductively coupled plasma mass spectrometry (ICP-MS) revealed preferential accumulation at the membrane and cytoskeleton, with the exception of [Sm(DBM)3(Phen)] that presented higher accumulation in the cytosol than in the cell membranes. All the evaluated lanthanide complexes showed low nuclear uptake, although not negligible. Spectroscopic studies on the interaction of the complexes with calf thymus DNA (ctDNA) revealed a moderate affinity with apparent binding constants in the 104 M−1 range. Complexes bind DNA not by intercalation but probably by electrostatic interactions. A morphological evaluation of the cells treated with the different complexes by electron microscopy (TEM/SEM) proved that all of them induce mitochondrial alterations, which seemed more pronounced for the NH2Phen complexes. In addition, the complex [Eu(DBM)3(NH2Phen)] presented lysosomal uptake that might explain its augmented cytotoxicity.

Es de particular relevancia poder identificar, en el cancer, los mecanlsmos de estimulo y proliferación celular, con el fin de crear estrategias diagn6sticas y terapeuticas especificamente dirigidas contra dianas ·celulares que participan en estos procesos. El diagnostico in vivo del cancer con agentes que reconocen marcadores tumorales, y que a su vez poseen 6ptimas caracteristicas para obtenci6n de imagenes, proporciona informaci6n fundamental desde el punta de vista clinico, permitiendo asi una correcta selecci6n del tratamiento a seguir y el monitoreo de sus efectos. Anticuerpos y sus fragmentos, entre otros, han sido utilizados para identificar la sobreexpresi6n de marcadores tumorales. Se ha descrito amplificaci6n del gen HER2 en varios tipos de canceres, particularmente en el cancer de mama, con el consiguiente aumento en la expresión de la proteina HER2 en la superficie de las celulas tumorales, que se traduce en un receptor HER2 constitutivamente activado. Los estudios indican que los pacientes cuyos tumores presentan una amplificación o sobreexpresión de HER2 presentan una menor expectativa de vida que la de aquellos con tumores sin amplificaci6n 0 sobreexpresi6n de HER2. Trastuzumab (HERCEPTIN®) es un anticuerpo monoclonal humanizado que reconoce de forma selectiva el dominio extracelular de HER2 y actualmente es utilizado como agente quimioterapeutico al inhibir la proliferaci6n de las celulas tumorales humanas que sobreexpresan HER2. Basandose en la alta especificidad de Trastuzumab por HER2, el objetivo del presente trabajo es desarrollar el radiofarmaco 99mTccHYNIC- Trastuzumab como marcador molecular in vivo de la expresión de HER2 en cancer de mama. En el presente estudio se desarrollo y optimizo la marcación de Trastuzumab con 99mTc, para ello se derivatizó el anticuerpo con el ligando bifuncional HYNIC. Se desarrollaron metodos de control basados en sistemas cromatograficos tanto en ITLC como HPLC, tambien se desarrollo un sistema para estudiar la inmunoafinidad in vitro del anticuerpo marcado. Se realizaron estudios de estabilidad in vivo e invitro, se caracterizó biológicamente la molecula marcada a traves de estudios de biodistribuci6n en ratones CD 1 normales y en portadores de adenocarcinoma mamano espontaneo.

A summary of the resource.
We described herein a simple and efficient microwave assisted synthesis of HYNIC analogues. Two different activated esters of HYNIC, the hydrazine protected with a trifluoroacetyl group (5) and the free hydrazine (6) were conjugated to the monoclonal antibody Nimotuzumab. Technetium-99m radiolabeling of Nimotuzumab was achieved with high efficiency using 5 and 6 derivates. The NHS-HYNIC-Tfa derivate allowed better labeling yields during longer times of preservation of the conjugated antibody.

<p>Nanomedicine is a highly demanded discipline. Liposomes have seen an increased attention due to their physicochemical properties that allow them to act as nanocarriers of drugs and also of radioisotopes that can be used to diagnose and treat cancer. In order to obtain a novel permeability cancer imaging agent based on 99mTc-labeled liposomes, we describe microwave-assisted synthesis of stearyl 6 (benzylidenehydrazinyl) nicotinamide lipid, which was included in two formulations: nanometric hydrazinonicotinic acid (HYNIC) liposome and its PEGylated coated analogue, HYNIC-PEG liposome. Radiolabeling with 99mTc via stearyl 6-(benzylidenehydrazinyl) nicotinamide was found to<br />be easy, reproducible, and stable, revealing high radiochemical purity (94 ± 1.7%) for both liposomal formulations. Biodistribution at 4 h and 24 h and scintigraphic images at 4 h were performed in normal and melanoma-bearing C57BL/6 mice. Biodistribution studies at 4 h showed tumor uptake of 99mTc-HYNIC liposome and 99mTc-HYNIC-PEG liposome (1.1 ±<br />0.6 and 2.5 ± 0.4, respectively) and also at 24 h p.i. (1.8 ± 0.5 and 3.0 ± 1.1, respectively). Scintigraphic images showed appreciable tumor uptake in melanoma tumor–bearing mice<br />with both liposomal formulations. Our results show that 99mTc stearyl 6-(benzylidenehydrazinyl) nicotinamide liposomes can be used as diagnostic noninvasive in vivo tumor-targeting agents capable of evaluating tumor permeability and development who can be used in personalized chemotherapy planning.</p>

Process optimization for producing diacylglycerol-enriched oils has received great attention due the importance of these products for healthier and more balanced diets. This work reports experimental data of liquid-liquid phase equilibrium of hydrolyzed olive oil (which comprises on its composition triacylglycerols, diacylglycerols, monoacylglycerols and free fatty acids) with ethanol and water, and subsequent analysis of oxidative stability of the oil obtained from the liquid phases. Liquid-liquid equilibrium experimental data obtained were modeled using the UNIQUAC thermodynamic model, which was well fitted to experimental data with a root mean square deviation of 1.76 wt%. The results obtained demonstrated the potentiality of liquid-liquid extraction as a suitable procedure for olive oil enrichment in diacylglycerol, after a hydrolysis procedure. It was also observed that liquid-liquid phase separation has a significant effect on the content of important antioxidants present in the olive oil, what affects the oxidative stability of the samples submitted to the liquid phase extraction. From the results obtained in this study, it has been also noticed that the acylglycerols and free fatty acids profile in of the oil samples presented a major rule on their stabilities.

Complexes of the type [Ru(II)Cl2(DMSO)2L], where L are 5-nitrofurylsemicarbazone derivatives, were prepared in an effort to combine the potential anti-tumor activity of the metal and the free ligands. The new complexes are excellent DNA binding agents for calf thymus DNA. So, their in vitro anti-tumor activity was tested in cellular models and the complexes were found to be non-cytotoxic on the tumor cell lines assayed, neither in aerobic conditions nor in the bio-reductive assay performed. Redox behavior, lipophilicity and stability were studied in order to explain the lack of cellular cytotoxic effects. The complexes resulted 10–100 times more hydrophilic than the parent ligands thus the bio-activity of these compounds would be compromised by their inadequate lipophilic properties.

La fusariosis de la espiga (FE) es una enfermedad que causa grandes pérdidas de rendimiento y calidad en granos de trigo a nivel mundial. Es producida principalmente por hongos del complejo Fusarium graminearum, los cuales en su mayoría producen micotoxinas que contaminan los granos afectados. El inóculo primario de la enfermedad proviene de los rastrojos del cultivo del año anterior. En condiciones ambientales adecuadas ascosporas o conidias del patógeno infectan las flores que constituyen el órgano susceptible de la planta. Las medidas de control incluyen prácticas culturales y aplicación de fungicidas. Sin embargo, ninguna práctica es por sí sola totalmente efectiva. En este contexto el uso de control biológico surge como una medida complementaria. El objetivo de este trabajo consistió en aislar y seleccionar cepas nativas de Trichoderma spp. para el control del inóculo primario de FE en trigo. Se trabajó con 16 cepas de Trichoderma identif icadas a nivel de especie las cuales exhibieron una o varias actividades que podrían relacionarse con su potencial como agente de biocontrol. Una de las cepas identif icada como T. atroviride, fue seleccionada con base en su habilidad para inhibir la formación de peritecios por parte del patógeno sobre rastrojo de trigo y su capacidad de producción de enzimas asociadas a la degradación del rastrojo que sirve de soporte al patógeno

OBJECTIVE: Warnings are a new directive front-of-pack (FOP) nutrition labelling scheme that highlights products with high content of key nutrients. The design of warnings influences their ability to catch consumers' attention and to clearly communicate their intended meaning, which are key determinants of their effectiveness. The aim of the present work was to evaluate the influence of design features of warnings as a FOP nutrition labelling scheme on perceived healthfulness and attentional capture. DESIGN: Five studies with a total of 496 people were carried out. In the first study, the association of colour and perceived healthfulness was evaluated in an online survey in which participants had to rate their perceived healthfulness of eight colours. In the second study, the influence of colour, shape and textual information on perceived healthfulness was evaluated using choice-conjoint analysis. The third study focused on implicit associations between two design features (shape and colour) on perceived healthfulness. The fourth and fifth studies used visual search to evaluate the influence of colour, size and position of the warnings on attentional capture. RESULTS: Perceived healthfulness was significantly influenced by shape, colour and textual information. Colour was the variable with the largest contribution to perceived healthfulness. Colour, size and position of the warnings on the labels affected attentional capture. CONCLUSIONS: Results from the experiments provide recommendations for the design of warnings to identify products with unfavourable nutrient profile.

5-Phenylethenylbenzofuroxans have displayed in vitro and in vivo activity against Trypanosoma cruzi, the etiologic agent of American Trypanosomiasis. On the basis of benzofuroxans pre-clinical studies we evaluated the potential of six 5-phenylethenyl derivatives to induce cytotoxicity, mutagenicity and genotoxicity using different in vitro models. Cytotoxic effects were evaluated using a set of cells, mammal pre-monocytic macrophages, V-79 lung fibroblast fromChinese hamster, and colorectal adenocarcinoma Caco-2 cells, in the MTT viability assay. Mutagenicity was tested in the Ames assay using Salmonella typhimurium TA98 strain with and without metabolic activation by S9-rat liver homogenate. The genotoxic potentials were evaluated with the alkaline single cell gel electrophoresis (comet assay) in V-79 cells. In view of the Ames test results we study whether the main mammals’ phase I metabolites, the corresponding o-nitroanilines, are involved in the mechanism of mutagenicity. These metabolites are produced by NADPH-dependent enzymes in cytosol and by xanthine oxidase and cytochrome P450 in microsomes fromrat liver.Amongthem, the electronic property of phenyl substituent seems to be responsible for this effect. It could be pointed out that the equimolecular mixture of compounds 1 and 2 (5Eand 5Z-(2-phenylethenyl)benzofuroxan, respectively) could be used in further clinical studies as anti-T. cruzi drug.

A summary of the resource.
In the course of our studies on 3H-1,2-dithiole-3-thione synthesis, a serendipitous reactivity with α-haloketones, in the presence of excess of potassium iodide, has been observed. Instead of the expected reaction of the nucleophile in a remote point of the molecule, we have obtained a product resulted from the electrophile character of the thiocarbonyl moiety on the 3-position of the 1,2-dithiole. In order to obtain an efficient protocol in terms of energy efficiency, this methodology was studied under conventional and microwave heating with similar or better results in the latter conditions. Simplicity and great efficiency in this one-step transformation are some of the advantages of this reaction. Moreover, the results can be explained according to the Pearson's hard and soft acid base theory.

A summary of the resource.
The goal of this work was to include an interdisciplinary research-oriented laboratory experiment taken from medicinal chemistry into the undergraduate biochemistry or organic studies. Phenazine 5,10-dioxides with selective cytotoxicity under hypoxia and with relevance in solid tumor treatment were selected to simulate different metabolism states. In this laboratory experiment, metabolism of 7-bromo-2-hydroxyphenazine-5,10-dioxide under simulated hypoxia and normoxia was studied using rat liver cytosol. Differential hypoxic and normoxic metabolism were monitored by thin-layer chromatography. Specific enzyme inhibitors, dicoumarol, ketoconazole, and menadione, were employed to identify the enzymatic system involved. This experiment introduced students to the concept of drug metabolism, activation, and mechanism of action. Furthermore, basic biochemical techniques of enzyme isolation and study were also discussed.<br /><br />

En esta tesis se muestran tanto la caracterización in vitro como in vivo de péptidos y fagos expresando péptidos en su cápside contra el receptor ErbB3. La tecnología aquí presentada permite un rápido cribado a partir de grandes bibliotecas de fagos en una metodología clásica y funcional desde su descubrimiento como lo es la técnica de phage display. La importancia de esta diana celular radica en que la expresión de ErbB3 posee un valor pronóstico, dado que altos niveles de este receptor están asociados con una sobrevida más corta en pacientes que sobrexpresan ErbB2. A su vez, ErbB3 juega un rol compensatorio en la señalización asociada a la resistencia al tratamiento en cáncer de mama en tumores que sobrexpresan ErbB2. Por ello es de gran importancia monitorear la expresión de ErbB3 y diagnosticarlo in vivo, por lo que sería deseable el desarrollo de una agente de imagen que lo reconociese. El principal objetivo planteado en este trabajo fue el descubrimiento de un péptido anti-ErbB3 obtenido a partir de fagos seleccionados por phage display para imagenología molecular de tumores mamarios. En esta tesis se seleccionaron y desarrollaron dos fagos (MSPC4 y MSPC3) y dos péptidos anti-ErbB3 respectivamente. Se investigó la marcación y caracterización de los distintos péptidos y fagos, obteniéndose imágenes SPECT e imágenes con sistemas ópticos. Se desarrollaron tres métodos de marcación a través de sistemas bifuncionales como lo son DOTA y HYNIC. Se obtuvo para todos los complejos marcados con 99mTc, 111In y 177Lu, pureza radioquímica adecuada y estabilidad tanto in vitro como in vivo, así se mantuvo la selectividad y afinidad por el receptor a nivel in vitro. Los sistemas desarrollados en base a los fagos MSPC4 y MSPC3, demostraron tener captación selectiva por el modelo tumoral utilizado. Constituyéndose así como potenciales candidatos a radiofármacos para la detección de tumor de mama a través de imagenología molecular óptica o SPECT.

A summary of the resource.
Nowadays, new strategies for pest and disease control to be used in rotation with or replacement of conventional pesticides are required. Essential oils (EOs), as botanical pesticides, provide a potential resource to develop more environmentally friendly and less toxic means of control to be applied in different produces. Tomato crop is affected by many insects and fungal diseases, among which, the insects Trialeurodes vaporariorum and Tuta absoluta, and the fungi Alternaria spp. and Botrytis cinerea are of great incidence. In this work two EOs from Uruguayan specimens of the local species Eupatorium buniifolium and the world-wide distributed Artemisia absinthium (Asteraceae) were characterized in their chemical composition and insecticidal and antifungal activities. We found that the EO from local A. absinthium is rich in oxygenated monoterpenes and belongs to the thujone chemotype (β-Thujone abundance is 56 ± 2 %, and α-Thujone, 1.67 ± 0.07 %). On the other hand, monoterpene hydrocarbons (α-Pinene, 22 ± 2 %) and sesquiterpene hydrocarbons [(E)-β-Guaiene, 10 ± 1 %] are the most abundant components of E. buniifolium EO. Eventhough both EOs chemically differ, they exhibit insecticidal and antifungal activity not only by direct contact but also by contact with their vapors against the tested organisms. These results may indicate that these EOs could be raw material to develop control agents to manage some of the main pests and fungal diseases of tomato crops with only one kind of treatment.

A summary of the resource.
Modem molecular biology techniques allow the use of new approaches for the 3D mapping of 1D information. Molecular biology techniques are capable of producing large amounts of 1D information (sequences) from a number of different sources (phage displays, ESTs, etc.). In this work, we present a technique that takes advantage of large sets of 1D information and increasingly available computer power to create 3D models. For the purpose of validation the technique is first applied to the modeling of an erythropoietin analog of known 3D structure from 1D information only. The technique is then used to model the immunoreactive region of echinococcus granulosus AgB based on phage raised mimotopes for which there is no previous structural information. The technique here presented is of general application to similar problems where 1D information is available and structure activity relationships (SAR) is needed.

The standard singles and doubles coupled-cluster method including perturbational treatment of connected triple excitations, CCSD(T), and density functional methods, DFT, using a large, uncontracted, atomic natural orbital (ANO) basis set were employed for calculating the enthalpies of formation of some first-row atoms (H, N, O, F) as well as diatomic and triatomic molecules formed with them. Molecular enthalpies of formation at 298.15 K were obtained from enthalpies of reaction of the atoms, homonuclear diatomic molecules, and isodesmic reactions. It is shown that the errors are the minimum when isodesmic reactions are used. However, contrary to accepted belief, CCSD(T) gives a larger deviation from experiment than DFT in the latter case. DFT exhibits similar accuracy when using a very small basis set (6-31G*) to that with the extended basis set, while the errors obtained with CCSD(T) are much larger. It is concluded that DFT is a more accurate and convenient computational tool than CCSD(T) for the thermochemical study of species containing the F−O bond.

We report herein the antitumor actions of three copper(II) complexes on MG-63 human osteosarcoma cells. The three complexes: Cu-sac, Cu-gln and Cu-sac-gln (sac= saccharinate, gln= glutamine) caused a decline in cell viability. The half-maximal inhibitory concentration in MG-63 cells for Cu-sac-gln is 170 µM, showing the strongest antiproliferative effect. Moreover, only Cu-sac-gln caused a decrease of the mitochondrial activity from 100 μM. Our results indicate that the copper(II) complexes studied here produce DNA damage and suggest that the rise of reactive oxygen species (ROS) is the central mechanism action. Genotoxicity studied by the Cytokinesis-block micronucleus (MN) assay and the Single cell gel electrophoresis (comet assay) could be observed in MG-63 cells treated with Cu-sac-gln from 100 and 50 μM, respectively. Cu-sac and Cu-gln also induced DNA damage; however their effect was definitively weaker. The generation of reactive oxygen species increased from 50 μM of Cu-sac-gln and Cu-sac and only from 250 μM of Cu-gln, as well as a reduction of the GSH/GSSG ratio from 50 μM. When cells were treated with several concentrations of the complexes in addition to a combination of 50 μM of vitamin C plus 50 μM of vitamin E, a total recovery in cell survival was obtained for Cu-gln in the whole range of tested concentrations while only a partial viability recovery was obtained from 250 μM of Cu-sac and Cu-sac-gln. Overall, our results point to a differential cyto- and genotoxicity of the three copper(II) complexes and demonstrate that the complexation with both ligands confers the most potent antitumor action in human osteosarcoma cells.

Pd nanoparticles anchored on Nb-doped TiO2 with functionalized carbon support (denoted as Pd/Nb–TiO2–C) is synthesized through a controllable hydrolysis and impregnation method. The as-synthesized catalyst is characterized by X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and transmission electron microscopy (TEM). Pd nanoparticles exhibit a uniform distribution with an average particle size of 3 nm. The electrochemical performance is tested by cyclic voltammetry (CV) and chronoamperometry (CA). Compared with Pd supported by functionalized carbon (Pd/C), Pd/Nb–TiO2–C demonstrates 15.7% higher metallic Pd content, 23% higher electrochemical active surface area, 75% higher current density in ethanol electro-oxidation, 5% higher durability, and better tolerance of carbonaceous species. The performance enhancement is attributed to the increased conductivity from Nb-doping and the synergistic effect between Pd and TiO2.

In the present work, an activated biocarbon (aBC) was studied as a support in Pd electrocatalyst for ethanol oxidation reaction (EOR) in alkaline medium. Pd/aBC sample was compared with a commercial Vulcan carbon supported Pd electrocatalyst. The Pd particles morphology were influenced by the high content of oxygenated surface functional groups of aBC support, which can be relevant for the synthesis of Pd based electrocatalysts, because the Pd particles formation mechanism involves the adsorption of Pd2+ on negative charged sites of aBC support. This adsorption occurs preferentially on electron donor sites such as oxygenated functional groups. Therefore, the higher concentration of these groups in aBC surface can determine a higher concentration of initial nuclei sites for Pd particles formation contributing to the particles agglomeration on aBC support, compromising the Pd/aBC electrocatalytic performance. For both prepared electrocatalysts, the spectroelectrochemical analysis demonstrates that the acetate ion is the main product of the EOR in the alkaline medium

Abstract Qualitative and quantitative determination of volatile markers in aqueous based fragrances assumes ever-increasing importance, because of both the need for quality control and the safety-regulatory limitations introduced for several compounds. This study reports and critically discusses the results of applying new water-compatible ionic-liquid (IL) GC stationary phases, based on phosphonium and imidazolium derivative cations combined with trifluoromethanesulphonate (Watercol™) to the direct quantitative analysis of aqueous samples in the perfume field with GC–MS. Narrow-bore columns of different lengths, especially prepared for this study, were adopted to minimize the amount of water reaching the MS detector after GC separation. All GC–MS analysis steps were investigated, to achieve results compatible with quality control requirements for the volatiles of interest in this field, in terms of LODs, LOQs, and repeatability. Reliability of the GC–MS results was demonstrated by determining volatile allergens in two commercial perfumes, as per EU regulations concerning no-declaration limits for leave-on (0.001%) and rinse-off (0.01%) cosmetic products.

Quality control of essential oils is fundamental for verifying their authenticity and confor mity with quality standards, ensuring their safety and regulatory compliance, and monitoring their consistency. Companies that produce or market essential oils routinely evaluate the quality and au thenticity of their products. However, they also must deal with increasing attention to environmental sustainability as well as practical considerations such as productivity, cost, and simplicity of methods. In this study, enantioselective gas chromatography (GC) was adopted to evaluate the quality of sweet and bitter orange essential oils, used as a case study. The analytical conditions were optimized and translated to fast GC to evaluate the impact of this approach on the environmental footprint of the analyses. The greenness of fast GC, compared with conventional GC, was quantitatively evaluated using a dedicated metric tool (AGREE), and important improvements have been calculated. The developed methods were applied to a set of commercial essential oils, and the data about the enantiomeric composition and relative percentage abundance were elaborated through multivariate statistics (principal component analysis). The results showed that fast chiral gas chromatography enables the classification of citrus essential oil samples and can be considered an environmentally friendly and sustainable approach for evaluating their quality.
