Tropical diseases today constitute a major health problem and a big challenge for drug discovery. Because of the limited arsenal of effective antiparasitic agents and the frequent appearance of chemoresistance, there is an urgent and continuous need to develop new drugs against these ailments. Metal compounds still offer excellent opportunities to find new ‘leads’ against the major protozoan diseases such as malaria, leishmaniasis and trypanosomiasis. A few metal-based drugs are already available in this therapeutic area, and others are currently being developed. Recent progress in parasite genomics and the identification of a few biomolecular targets hold great promise for the discovery of new ‘mechanism-based’ antiparasitic metallodrugs. The trends and perspectives for this exciting research field are outlined here.

Different extraction procedures and clean-up methods were compared in order to develop a sample preparation<br />procedure for the multi-residue analysis of six post-emergence herbicides (metsulfuron methyl, bensulfuron<br />methyl, pyrazosulfuron ethyl, bentazone, bispyribac sodium and cyhalofop butyl) in rice grains followed by<br />liquid chromatography-diode array detection (LC-DAD). Optimum results were obtained dispersing milled rice<br />grain in water, followed by the addition of 1% acetic acid in acetonitrile, MgSO4 and sodium acetate as a<br />modification of the quick, easy, cheap, effective, rugged and safe (QuEChERS) method but no primary and<br />secondary amine (PSA) sorbent was added due to the acidic nature of the herbicides. The method was further<br />expanded to other post-emergence herbicides (quinclorac, clomazone and propanil). Except for quinclorac, which<br />cannot be analysed with this method, the recoveries of the other eight herbicides were in the range 73–111%, with<br />relative standard deviations lower than 12%. Limits of detection (LODs) ranged from 0.03 to 0.08mg kg�1.<br />A single analyst can extract twelve samples in 4 h. The method presented here allows the simultaneous residue<br />determination of the most common post-emergence herbicides employed in cultivating rice. It is simple, rapid,<br />sensitive, and can be applied routinely to polished rice grain herbicide residue analysis.

Two simple and straightforward sample preparation methods were developed for the multiresidue analysis of post-emergence herbicides in loam soil that are commonly used in rice crop cultivation. A number of strategic soil extraction and cleanup methods were evaluated. The instrumental analysis was performed by HPLC with a diode array detector. The best compromise between the recoveries (69–98%) and good repeatability (RSD <15%) was obtained using either methanol or methanol–ethyl acetate (70 + 30, v/v) as extraction solvents without any further cleanup. For the first time, residues of metsulfuron methyl, bensulfuron ethyl, bentazone, cyhalofop butyl, pyrazosulfuron ethyl, propanil, and clomazone were analyzed simultaneously. Quinclorac and bispyribac sodium were also assayed, but their recoveries were below 50%. Both methods had an LOD of 0.7 mg/kg and could accurately determine the residues at the 2 mg/kg level. These two methods could not be applied directly to other soil types as the recoveries strongly depended on the soil composition. The developed methodologies were successfully applied in monitoring 87 real-world soil samples, in which only propanil (6 to 12 mg/kg) and clomazone (15 to 20 mg/kg) residues could be detected.

The thioredoxin and/or glutathione pathways occur in all organisms. They provide electrons for deoxyribonucleotide synthesis, function as antioxidant defenses, in detoxification, Fe/S biogenesis and participate in a variety of cellular processes. In contrast to their mammalian hosts, platyhelminth (flatworm) parasites studied so far, lack conventional thioredoxin and glutathione systems. Instead, they possess a linked thioredoxin-glutathione system with the selenocysteine-containing enzyme thioredoxin glutathione reductase (TGR) as the single redox hub that controls the overall redox homeostasis. TGR has been recently validated as a drug target for schistosomiasis and new drug leads targeting TGR have recently been identified for these platyhelminth infections that affect more than 200 million people and for which a single drug is currently available. Little is known regarding the genomic structure of flatworm TGRs, the expression of TGR variants and whether the absence of conventional thioredoxin and glutathione systems is a signature of the entire platyhelminth phylum. Results: We examine platyhelminth genomes and transcriptomes and find that all platyhelminth parasites (from classes Cestoda and Trematoda) conform to a biochemical scenario involving, exclusively, a selenium-dependent linked thioredoxin-glutathione system having TGR as a central redox hub. In contrast, the free-living platyhelminth Schmidtea mediterranea (Class Turbellaria) possesses conventional and linked thioredoxin and glutathione systems. We identify TGR variants in Schistosoma spp. derived from a single gene, and demonstrate their expression. We also provide experimental evidence that alternative initiation of transcription and alternative transcript processing contribute to the generation of TGR variants in platyhelminth parasites. Conclusions: Our results indicate that thioredoxin and glutathione pathways differ in parasitic and free-living flatworms and that canonical enzymes were specifically lost in the parasitic lineage. Platyhelminth parasites possess a unique and simplified redox system for diverse essential processes, and thus TGR is an excellent drug target for platyhelminth infections. Inhibition of the central redox wire hub would lead to overall disruption of redox homeostasis and disable DNA synthesis.

Conformational diseases are a group of diverse disorders that have been associated with misfolding of specific proteins, leading to their aggregation in particular cell tissues. Despite their relevance, the mechanisms involved in neurodegenerative processes remains poorly understood. Mutations in Cu,Zn superoxide dismutase (SOD1) are implicated in death of motor neurons in amyotrophic lateral sclerosis. Among others, the SOD1G93A mutation is known to weaken the structure and this could lead to conformational variations of the protein. As an approach to understand the tissue-specific propensity of protein aggregation, we developed an experimental procedure allowing rapid extraction of variants of human SOD1 (hSOD1) produced in different tissues. Using an antibody-based affinity chromatography procedure enzymatically active hSOD was extracted, indicating preservation of its native conformation. Analysis of the eluted fractions of hSOD extracted from the brain and liver of transgenic hSODG93A rats provided evidence about heterodimers rSOD–hSODG93A formation in both extracts. Moreover, when characterized by 2-DE and MALDI-TOF/TOF MS, the extracted hSODG93A showed a complex profile suggesting the existence of various covalent modifications of the enzyme in both tissues. Thus, this method should allow following post-translational modifications of hSOD1 produced in various tissues.

The sensory characteristics of cosmetic products are commonly used for attracting consumers’ attention in advertising campaigns and packaging. Thus, in order to appropriately satisfy consumers’ sensory expectations, it is important to gather information about their perception of the sensory characteristics. In this context, the aim of the present work was to apply two consumer methodologies (intensity scales and a check-all-that-apply [CATA] question) to gather information about the sensory profile of cosmetic emulsions. Six cosmetic emulsions with different formulation, widely differing in their sensory characteristics, were evaluated by two groups of 50 female consumers. One group used intensity scales to evaluate eight attributes of the emulsions and the other completed a CATA question comprising 20 terms related to sensory characteristics of the emulsions, cosmetic applications and effects on the skin. Both intensity scales and CATA question were able to detect differences in consumers’ perception of the sensory characteristics of the evaluated emulsions. These differences could be explained considering the samples’ formulation, which suggests their validity and indicates that they could consist on interesting alternatives to traditional sensory profiles obtained with trained assessors’ panels. 3

A summary of the resource.
A study on the phenolic content and antioxidant properties of Uruguayan propolis is presented. Identification and quantification was carried out by Reverse Phase High Performance Liquid Chromatography (RP-HPLC). Antioxidant activity was determined by Antiradical Activity Determination (AAD) measured with DPPH and Microsomal Lipid Peroxidation (MLP) assays. Two phenolic acids (gentistic and p-coumaric) and eight flavonoids (fisetin, myricetin, luteolin, quercetin, kaempferol, pinocembrin, chrysin and tectochrysin) were identified. The existence of five other phenols is postulated. The large difference between range of values of scavenger activity AAD (between 60% and 100%) as compared with lipoperoxidation inhibition (between 15% and 92%), suggests that the complexity of the biologic matrix could modulate the antioxidant effect. From a commercial standpoint, research such as the one presented here endows the product with added value, making it more appealing to the interest of industry and entrepreneurs.

Bulk SrMnOx samples with oxygen contents 2.5pxo2.7 have been studied using a combination of neutron time-of-flight and highenergy high-resolution synchrotron X-ray diffraction measurements along with thermogravimetric analysis. We report the identification and characterization of two new oxygen-vacancy ordered phases, Sr5Mn5O13 (SrMnO2.6-tetragonal P4/m a¼ 8.6127(3)A ˚ , c ¼ 3.8102(2)A ˚ ) and Sr7Mn7O19 (SrMnO2.714-monoclinic P2/m a¼ 8.6076(4)A ˚ , b ¼ 12.1284(4)A ˚ , c ¼ 3.8076(2)A ˚ , g ¼ 98.203(2)1). The nuclear and magnetic structures of Sr2Mn2O5 are also reported (SrMnO2.5 nuclear: orthorhombic Pbam, magnetic: Orthorhombic Ay type Pcbam with cM ¼ 2c). In the three phases, oxygen-vacancies are ordered in lines running along one of the (100) directions of the parent cubic perovskite system. Oxygen-vacancy ordering allows the charge and orbital ordering of the Mn3+ and Mn4+ cations in the new phases.

Dietary PUFAs (polyunsaturated fatty acids) co-ordinately suppress transcription of a group of hepatic genes encoding glycolytic and lipogenic enzymes. Suppression of Fasn (fatty acid synthase) transcription involves two PUFA-responsive regions, but the majority of PUFA sensitivity maps to a region within the proximal promoter containing binding sites for NF-Y (nuclear factor-Y), Sp1 (stimulatory protein 1), SREBP (sterol-regulatory-elementbinding protein), and USF (upstream stimulatory factor). Promoter activation assays indicate that altered NF-Y is the key component in regulation of Fasn promoter activity by PUFA. Using electrophoretic mobility-shift assay and chromatin immunoprecipitation analysis, we demonstrate for the first time that PUFAs decrease in vivo binding of NF-Y and SREBP-1c to the proximal promoter of the hepatic Fasn gene and the promoters of three additional genes, spot 14, stearoyl-CoA desaturase and farnesyl diphosphate synthase that are also down-regulated by PUFA. The comparable 50% decrease in NF-Y and SREBP-1c binding to the promoters of the respective PUFA-sensitive genes occurred despite no change in nuclear NF-Y content and a 4-fold decrease in SREBP-1c. Together, these findings support a mechanism whereby PUFA reciprocally regulates the binding of NF-Y and SREBP-1c to a subset of genes which share similar contiguous arrangements of sterol regulatory elements and NF-Y response elements within their promoters. PUFA-dependent regulation of SREBP-1c and NF-Y binding to this unique configuration of response elements may represent a nutrient-sensitive motif through which PUFA selectively and co-ordinately targets subsets of hepatic genes involved in lipid metabolism. Se estudió la interacción entre diversos extractos acuosos y alcohólicos de C. pareira con el veneno de B. neuwiedi diporus COPE, por electroforesis SDS-PAGE. Los extractos activos fueron testeados in vitro para determinar su actividad antihemolítica en placas de agar sangre- fosfatidilcolina, llegando a la conclusión de que el extracto alcohólico de hojas inhibe la actividad hemolítica del veneno en forma concentración dependiente lo que avalaría su uso, en la medicina popular, como alexítera.

Nitrated fatty acids (nitroalkenes) have been recently detected and quantified in cell membranes and human plasma. However, nitration of arachidonate (AA), that could redirect AA-dependent cell signaling pathways, has not been studied in detail. Herein, we synthesized and determined for the first time the isomer distribution of nitroarachidonate (AANO2) and demonstrate its ability to modulate inflammation. Synthesis of AANO2 was achieved by AA treatment with sodium nitrite in acidic conditions following HPLC separation. Mass spectrometry (MS) analysis showed the characteristic MS/MS transition of AANO2 (m/z 348/301). Moreover, the IR signal at 1378.3 cm-1 and NMR studies confirmed the presence of mononitrated nitroalkenes. Positional isomer distribution was determined by NMR and MS fragmentation with lithium; four major isomers (9-, 12-, 14-, and 15-AANO2) were identified, which exhibited key anti-inflammatory properties. These include their ability to release biologically relevant amounts of nitric oxide, induce cGMP-dependent vasorelaxation, and down-regulate inducible nitric oxide synthase (NOS2) expression during macrophage activation, providing unique structural evidence and novel regulatory signaling properties of AANO2.

An efficient, fast, and accurate matrix solid-phase dispersion sample cleanup procedure was developed specifically for the determination of pesticide residues in lanolin. The scope of the method for organophosphorus, organochlorine, and pyrethroid insecticides is the same as that of official methods from various pharmacopeias. After lanolin dispersion on C18 bonded silica, pesticides are eluted with acetonitrile saturated with n-hexane. Recoveries ranged from 83 to 118% with RSD values of <20% for most pesticides listed, in compliance with the requirements of European and U.S. pharmacopeias.

Abstract
A summary of the resource.
The azine bridged dicatechol ligand (E,E)-benzaldehyde azine (H4L) was fully characterized by X-ray analysis. The reaction of [ReCl6]2� with this compound was studied and the novel Re(IV) complex (HNEt3)(NBu4)[ ReCl4(H2L)] was prepared and characterized. The structure and spectroscopy of the compound H4L and its Re(IV) complex were studied experimentally and by means of density functional calculations.

Abstract
A summary of the resource.
Acinetobacter baumannii is considered an important pathogen in our hospital environment having a well-known capacity to acquire different mechanisms of antibiotic resistance. Previous studies in our laboratory had exposed the high dispersion of class 2 integrons in this species. In the present study, we analyzed 7 multiresistant intI2 positive A. baumannii isolates, 6 of which were found to harbour the Tn7::In2-8 element. Our results demonstrate the unusually high distribution of Tn7::In2-8 among different A. baumannii clones from Chile, suggesting a particular behavior of these elements at geographical level.

Abstract
A summary of the resource.
Radiometals have become increasingly important because of their use both for diagnostic molecular imaging and therapy in Nuclear Medicine. The focus is on the study of biochemical processes at cellular and sub-cellular level in order to detect metabolic abnormalities associated with various diseases. For that purpose, molecules that selectively accumulate in the organ or tissue of interest by a specific mechanism such as receptor binding or interaction with biomolecules are labeled with 99mTc, 68Ga, 153Sm, 186/188Re, 177Lu, among others and used as radiopharmaceuticals. However, considerable effort is necessary to combine these radionuclides with biomolecules relevant to different pathological conditions. Intensive research on the coordination chemistry of these metals has led to novel labeling methods that yield stable compounds which retained the original biological activity of the ligand. Chemical aspects and clinical applications will be reviewed in this paper.

Complexes [RuCl(H4NO2Fo4M)(bipy)(dppb)]PF6 (1), [RuCl(H4NO2Fo4M)(Mebipy)(dppb)]PF6 (2), [RuCl (H4NO2Fo4M)(phen)(dppb)]PF6 (3), [RuCl(H4NO2Ac4M)(bipy)(dppb)]PF6 (4), [RuCl(H4NO2Ac4M) (Mebipy)(dppb)]PF6 (5) and [RuCl(H4NO2Ac4M)(phen)(dppb)]PF6 (6) with N4-methyl-4-nitrobenzalde hyde thiosemicarbazone (H4NO2Fo4M) and N4-methyl-4-nitroacetophenone thiosemicarbazone (H4NO2 Ac4M) were obtained from [RuCl2(bipy)(dppb)], [RuCl2(Mebipy)(dppb)], and [RuCl2(phen)(dppb)], (dppb ¼ 1,4-bis(diphenylphospine)butane; bipy ¼ 2,20-bipyridine; Mebipy ¼ 4,40-dimethyl-2,20-bipyridine; phen ¼ 1,10-phenanthroline). In all cases the thiosemicarbazone is attached to the metal center through the sulfur atom. Complexes (1e6), together with the corresponding ligands and the Ru precursors were evaluated for their ability to in vitro suppress the growth of Trypanosoma cruzi. All complexes were more active than their corresponding ligands and precursors. Complexes (1e3) and (5) revealed to be the most active among all studied compounds with ID50 ¼ 0.6e0.8 mM. In all cases the association of the thiosemicarbazone with ruthenium, dppb and bipyridine or phenanthroline in one same complex proved to be an excellent strategy for activity improvement.

A DFT study on 5-NI derivatives was carried out at different levels of theory, in order to seek for the best methodology that helps us to calculate hyperfine coupling constants (hfccs), spin density distributions and electronic properties, since it is difficult to obtain a good agreement between calculated and experimental hfccs for nitrogen center radical compounds. Different functional hybrids namely B1LYP, B3LYP and PBE0 were used in conjunction with various medium to large size basis sets. C-PCM model was considered for solute-solvent interactions. Comparisons of our calculations with previous experimental data obtained from EPR and X-ray techniques confirmed that this computational approach represents an important aid. Our results indicated that solvent effects and consideration of diffuse functions are the most important factors on the calculation of hyperfine coupling constants for this nitro-heterocyclic system. Also, we found that some electronic properties calculated (LUMO orbital, condensed Fukui functions for nucleophilic attacks and electron affinity) do not correlate as expected with the biological activity.

The impact of the use of herbicides in agriculture can be minimized by compliance with good management practices that reduce the amount used and their release into the environment. Simple tests that provide real time on-site information about these chemicals are a major aid for these programs. In this work, we show that phage anti-immunocomplex assay (PHAIA), a method that uses phage-borne peptides to detect the formation of antibody-analyte immunocomplexes, is an advantageous technology to produce such field tests. A monoclonal antibody to the herbicide clomazone was raised and used in the development of conventional competitive and noncompetitive PHAIA immunoassays. The sensitivity attained with the PHAIA format was over 10 times higher than that of the competitive format. The cross-reactivity of the two methods was also compared using structurally related compounds, and we observed that the two-site binding of PHAIA "double-checks" the recognition of the analyte, thereby increasing the assay specificity. The positive readout of the noncompetitive PHAIA method allowed adaptation of the assay into a rapid and simple format where as little as 0.4 ng/mL clomazone (more than 10-fold lower than the proposed standard) in water samples from a rice field could be easily detected by simple visual inspection.
