We previously reported a multigene family of monodomain Kunitz proteins from Echinococcus granulosus (EgKU-1-EgKU-8), and provided evidence that some EgKUs are secreted by larval worms to the host interface. In addition, functional studies and homology modeling suggested that, similar to monodomain Kunitz families present in animal venoms, the E. granulosus family could include peptidase inhibitors as well as channel blockers. Using enzyme kinetics and whole-cell patch-clamp, we now demonstrate that the EgKUs are indeed functionally diverse. In fact, most of them behaved as high affinity inhibitors of either chymotrypsin (EgKU-2-EgKU-3) or trypsin (EgKU-5-EgKU-8). In contrast, the close paralogs EgKU-1 and EgKU-4 blocked voltage-dependent potassium channels (Kv); and also pH-dependent sodium channels (ASICs), while showing null (EgKU-1) or marginal (EgKU-4) peptidase inhibitory activity. We also confirmed the presence of EgKUs in secretions from other parasite stages, notably from adult worms and metacestodes. Interestingly, data from genome projects reveal that at least eight additional monodomain Kunitz proteins are encoded in the genome; that particular EgKUs are up-regulated in various stages; and that analogous Kunitz families exist in other medically important cestodes, but not in trematodes. Members of this expanded family of secreted cestode proteins thus have the potential to block, through high affinity interactions, the function of host counterparts (either peptidases or cation channels) and contribute to the establishment and persistence of infection. From a more general perspective, our results confirm that multigene families of Kunitz inhibitors from parasite secretions and animal venoms display a similar functional diversity and thus, that host-parasite co-evolution may also drive the emergence of a new function associated with the Kunitz scaffold.

Potato (Solanum tuberosum L.) is one of the main hosts of Ralstonia solanacearum, the causative agent of bacterial wilt. This plant pathogen bacteria produce asymptomatic latent infections that promote its global spread, hindering disease control. A potato breeding program is conducted in Uruguay based on the introgression of resistance from the wild native species S. commersonii Dun. Currently, several backcrosses were generated exploiting the high genetic variability of this wild species resulting in advanced interspecific breeding lines with different levels of bacterial wilt resistance. The overall aim of this work was to characterize the interaction of the improved potato germplasm with R. solanacearum. Potato clones with different responses to R. solanacearum were selected, and colonization, dissemination and multiplication patterns after infection were evaluated. A R. solanacearum strain belonging to the phylotype IIB-sequevar 1, with high aggressiveness on potato was genetically modified to constitutively generate fluorescence and luminescence from either the green fluorescence protein gene or lux operon. These reporter strains were used to allow a direct and precise visualization of fluorescent and luminescent cells in plant tissues by confocal microscopy and luminometry. Based on wilting scoring and detection of latent infections, the selected clones were classified as susceptible or tolerant, while no immune-like resistance response was identified. Typical wilting symptoms in susceptible plants were correlated with high concentrations of bacteria in roots and along the stems. Tolerant clones showed a colonization pattern restricted to roots and a limited number of xylem vessels only in the stem base. Results indicate that resistance in potato is achieved through restriction of bacterial invasion and multiplication inside plant tissues, particularly in stems. Tolerant plants were also characterized by induction of anatomical and biochemical changes after R. solanacearum infection, including hyperplasic activity of conductor tissue, tylose production, callose and lignin deposition, and accumulation of reactive oxygen species. This study highlights the potential of the identified tolerant interspecific potato clones as valuable genetic resources for potato-breeding programs and leads to a better understanding of resistance against R. solanacearum in potato.

Leaf removal is beginning to receive consideration for many growers as an important cultural practice for bunch rots control. It is normally applied after veraison and almost only on red varieties because of the high sunburn susceptibility of white grapes varieties under Uruguayan climatic conditions. The aim of this study was to determine the time and intensity of leaves removal impact on bunch rots and grapes quality on Sauvignon blanc. In parallel, the effects of kaolin (Surround® WP) applications in reducing the negative impact of excessive exposure to high temperatures were evaluated. Different intensities and times of defoliation, in combination with kaolin applications, were evaluated over 2006/07, 2007/08 and 2008/09 seasons. All defoliation treatments and mainly those made after fruit set, significantly reduced the occurrence of bunch rots, having a critical impact on those metabolites responsible for the final taste of wines. Early defoliation was significantly more effective controlling Botrytis incidence and improving berry quality than later in the season. Leaf removal had a determining impact over fruit quality and primary and secondary metabolites contents. An increase in soluble solids and glycosides concentrations was observed, while IBMP grape content was reduced, thus affecting the flavor of Sauvignon blanc wines. Kaolin applications significantly reduced berries temperature, sunburn and other berry damages associated with clusters solar exposure. In sum, leaves removal and exogenous kaolin application in Sauvignon blanc showed great potential as Botrytis mitigation strategy because it positively impacts berry quality as a result of many molecular and biochemical changes in key secondary metabolic pathways. Astringency is one of the most important characteristics that define the quality of red wine, and is of particular relevance for Tannat, Uruguayan emblematic red wine variety. Astringency is a time-dependant and complex sensory characteristic, related to several sensations, or sub-qualities, that can be simultaneously perceived. The aim of the present study was to obtain a sensory characterization of the astringency of commercial Uruguayan Tannat wines. Forty samples with different characteristics in terms of vintage, price segment and aging in oak barrels were assessed by a panel of 9 trained assessors. Total astringency intensity was evaluated using time-intensity (TI), while astringency sub-qualities were described using a check-all-that-apply (CATA) question composed of sixteen terms. TI and the CATA question provided different information on the astringency of Tannat wines. Regarding global astringency, samples mainly differed in intensity-related parameters rather than in the development of astringency over time, although the variability was moderate. A wide range of sub-qualities, from silky and velvety to harsh and aggressive were used to describe the astringency of the evaluated wines. Four groups of samples with different astringency characteristics were identified, but this sorting was not related to vintage, price segment or aging in oak barrels. Further research is necessary to better understand how astringency characteristics are influenced by production variables, and to understand their relationship to consumers' and experts' perceived quality of Tannat wines.

We report a study on the preparation and characterization of polyaniline-graphene oxide (PANI-GO-X) nanocomposites focusing in the study of its structure, microstructure, and correlations with electrical transport properties under low applied electric fields. X-ray diffraction analysis showed evidence of graphene oxide (GO) blending between polyaniline fibers, and according to small angle X-ray scattering, the fractal dimensionality was quasi-one dimensional for all nanocomposites. Confocal Raman spectroscopy revealed that the addition of GO leads to a notorious decrease of the polaron population of polyaniline. A significant increase in resistivity was observed for PANI-GO-X nanocomposites with respect to pure polyaniline, in agreement with the decrease in the polaron population. The electrical transport mechanism could be explained by an Arrhenius behavior at high temperatures (T > 255 K) and a broad transition with a logarithmic dependence of the activation energy with temperature for the low temperature regime (T < 255 K). Additionally, an increase in the hopping transport dimensionality from quasi-1D to quasi-3D was observed for X = 7% GO nanocomposites. The transport properties, studied after the polarization using low applied electric fields, revealed the modification of the electrical transport at low temperatures. In addition, the electrical transport of PANI-GO-X nanocomposites followed a typical ln(σ) vs E1/2 Poole-Frenkel type dependence in the presence of low applied electric fields. At low temperatures, X = 7% GO nanocomposites showed a notorious increase in the Poole-Frenkel slope at low applied electric fields with respect to pure polyaniline. The enhancement on the electric response using low applied electric fields (E ∼ 2 V/cm) could be of great interest for the development of new organic electronic devices.

Here, we report the tuning from the positive to negative magnetoresistance response at room temperature and low applied magnetic fields (H ∼ 200 mT) for polyaniline nancomposites prepared via in situ growth of titanium oxide quantum dots. In addition, we showed experimental Raman evidence revealing that the positive magnetoresistance response in these polyaniline nanocomposites is mediated by the bipolaron mechanism. Confocal Raman spectroscopy under applied magnetic field analysis showed the decrease of the polaron population to form bipolarons of polyaniline when exposed to an applied magnetic field for the TiO2 quantum dot diluted regime. Negative magnetoresistance, observed for the TiO2 quantum dot higher concentration regime, was attributed to the suppression of polyaniline polarons probably associated with its partial chemical functionalization at the interface due to the increasing concentration of TiO2 quantum dots.

A summary of the resource.
In Fischer-Tropsch synthesis of hydrocarbons, the selectivity toward various hydrocarbons is altered by using rare earth oxides or rare earth oxides combined with Pd as isosynthesis catalysts. For example, Dy<sub>2</sub>O<sub>3</sub> and La<sub>2</sub>O<sub>3</sub> catalysts show a high selectivity toward butane [106-97-8] and a high isobutane [75-28-5] butane ratio, but Pd/ThO<sub>2</sub> and Pd/SiO<sub>2</sub>.Al<sub>2</sub>O<sub>3</sub> were more selective toward CH<sub>4</sub> and MeOH [67-56-1]. Dy<sub>2</sub>O<sub>3</sub> and La<sub>2</sub>O<sub>3</sub> gave a product distribution far from the Schulz-Flory theory due to an abnormal max. of C<sub>4</sub> hydrocarbons. As the contact time shortens, the selectivity toward MeOH increases. A mechanism is proposed for the 1st step of the isosynthesis.

A summary of the resource.
Zinc forms part of the active site and/or stabilizes the conformation of several enzymes. The protein metallothionein constitutes a zinc deposit from which the metal is transferred to enzymes. Zinc participates in the synthesis of nucleic acids and hence in the synthesis of proteins, principally through multi-enzyme pir 1-3 and DNA-dependent RNA polymerase, which are metallo-enzymes of zinc. Zinc is therefore necessary for normal growth, development, healing and ossification.

The free sterols of the fungi <em>Ganoderma applanatum</em>, <em>Ganoderma lucidum</em> and <em>Polyporus sulfureus</em> were isolated and characterized by means of GC and GC/MS techniques. 24-Methylcholesta-7,22-dien-3β-ol was the main component of the sterol mixtures while 24-methylcholesta-5,7,22-trien-3β-ol ergosterol) and 24-methylcholest-7-en-3β-ol were also present although in lower amounts. <em>P. sulfureus</em>, besides the mentioned sterols, also contained 24 ethylcholestan-3β-ol.

Herein, we report the preparation and characterization of a novel polymeric blend between linear polyethylene imine (PEI) and polyacrylonitrile (PAN), with the purpose of facilitating the dissociation of lithium perchlorate salt (LiClO4) and thus to enhance Li ion transport. It is a joint theoretical and experimental procedure for evaluating and thus demonstrating the lithium salt dissociation. The procedure implies the correlation between the theoretical pair distribution function (PDF) and conventional X-ray diffraction (XRD) by means of a molecular dynamics (MD) approach. Additionally, we correlated the experimental and theoretical Raman and infrared spectroscopy for vibrational characterization of the lithium salt after dissociation in the polymeric blend. We also performed confocal Raman microscopy analysis to evidence the homogeneity on the distribution of all components and the LiClO4 dissociation in the polymer blend. The electrochemical impedance analysis confirmed that the Li–PAN–PEI blend presents a slightly better lithium conductivity of ∼8 × 10–7 S cm–1. These results suggest that this polymer blend material is promising for the development of novel fluorine-free solid polymer lithium ion electrolytes, and the methodology is suitable for characterizing similar polymeric systems.

A summary of the resource.
A succinate dehydrogenase mutant strain of Rhizobium meliloti was isolated after nitrosoguanidine mutagenesis. It failed to grow on succinate, glutamate, acetate, pyruvate, or arabinose but grew on glucose, sucrose, fructose, and other carbohydrates. The mutant strain showed delayed nodulation of lucerne plants, and the nodules were white and ineffective. A spontaneous revertant strain of normal growth phenotype induced red and effective nodules.

<span class="hit">A</span> double <span class="hit">mutant</span> strain (UR3) of R. <span class="hit">meliloti</span> L5-30 was isolated from <span class="hit">a</span> phosphoglucose isomerase <span class="hit">mutant</span> (UR1) on the basis of its resistance to fructose inhibition when grown on fructose-rich medium. UR3 lacked both phosphoglucose isomerase and <span class="hit">fructokinase</span> activity. <span class="hit">A</span> <span class="hit">mutant</span> strain (UR4) lacking only the <span class="hit">fructokinase</span> activity was derived from UR3; it grew on the same C sources as the parent strain, but not on fructose, mannitol, or sorbitol. <span class="hit">A</span> spontaneous revertant (UR5) of normal growth phenotype contained <span class="hit">fructokinase</span> activity. <span class="hit">A</span> fructose transport system was found in L5-30, UR4, and UR5 grown in arabinose-fructose minimal medium. No fructose uptake activity was detected when L5-30 and UR5 were grown on arabinose minimal medium, but this activity was present in strain UR4. Free fructose was concd. intracellularly by UR4 >200-fold above the external level. <span class="hit">A</span> partial transformation of fructose into mannitol and sorbitol was detected by enzymic anal. of the uptake products. Polyol dehydrogenase activity was detected in UR4 grown in arabinose-fructose minimal medium. The induction pattern of polyol dehydrogenase activities in this strain might be due to slight intracellular fructose accumulation.

There are recent experimental reports on the preparation of edge-modified graphene quantum dots (GQDs) and its loading into conductive polymers for energy applications. Recently, we have studied the influence of the graphene quantum dots (GQDs) addition to polyaniline (PANI) conductive polymer in the electronic transport, finding important modifications in the structural and electrical properties respect to the pristine polymer. In this work, we present a mixed approach, based in first-principles molecular dynamics with time-dependent density functional calculations, for the description of the electronic dynamics, particularly for the determination of the optical properties. We found that edge modification of the GQDs favors the PANI/GQDs interaction, promoting very interesting electronic properties. Particularly, when hydroxyl chemical functional groups are present at the edges of the GQD a very good electron–hole separation during the photon excitation is obtained, demonstrating its potential use for optical applications in general and solar cells in particular

Here, we report a novel strategy for the preparation of TiO2 quantum dots fillers prepared from alkoxide precursor via in situ water vapor flow diffusion into poly(N-vinylcarbazole) host. A detailed characterization by means of infrared and Raman spectroscopy, X-ray powder diffraction, small angle X-ray scattering and differential scanning calorimetry is reported. The growth mechanism of both crystallites and particles was mostly governed by the classical coarsening reaction limited growth and the polymer host showed no detectable chemical modifications at the interface or active participation in the growing process. The main relevance of our strategy respect to the typical sol-gel growth in solution is the possibility of the interruption of the reaction by simple stopping the water vapor flow diffusion into the polymer host thus achieving good control in the nanoparticles size. The thermal stability and fractal behavior of our nanocomposites were also studied by differential scanning calorimetry and in situ small angle X-ray scattering versus temperature. Strong correlations between modifications in the fractal behavior and glass transition or fusion processes were observed for these nanocomposites.

In this report, we studied Raman and impedance spectroscopy under applied dc bias for poly(vinyl carbazole):titania quantum dots (PVK:TiO2-QDs) nanocomposites to understand the electrical transport in this donor:acceptor system as promising active layer materials for polymeric solar cells. The nanocomposites were synthesized using a novel strategy involving the sol–gel method via in situ water vapor flow diffusion in the polymer host leading to ∼3–8 nm TiO2 quantum dots embedded in PVK with excellent homogeneity. Raman spectroscopy was used to monitor the presence of charge carriers in the organic semiconductor for PVK:TiO2-QDs with different TiO2 quantum dots compositions with and without applied dc voltage. The joint study of confocal Raman spectroscopy supported by theoretical simulations together with impedance spectroscopy under applied dc voltage could give us a major approach to understand the electrical transport mechanism of these hybrid donor:acceptor nanocomposites.

The mechanism of CH<sub>4</sub> formation in industrial MeOH synthesis from CO and H using La<sub>2</sub>O<sub>3</sub> catalyst is detd. by the catalyst compn., the reaction medium (H or inert gas), catalyst pretreatment with Pt, and hydration or carbonate or metal deposits on the catalyst. The formation of CH<sub>4</sub> via unstable <span class="hit">methoxy</span> <span class="hit">species</span> has a neg. effect on the MeOH reaction and adequate control of reaction conditions can minimize the side reaction.

Abstract
A summary of the resource.
1. The electrokinetic and thermostability properties of phosphoglucose isomerase (PGI) in five species of tropical fish (Ostariophysi)—<em>Leporinus friderici, L. silvestrii, Schizodon nasuttus, Hypostomus sp</em> and <em>Pimelodus maculatus</em>—have been studied in order to study the adaptative nature of protein heterogeneity found in ectotherms.
<p>2. Unlike most diploid fishes, the PGI of these species seemed to be encoded by three, four or rive loci.</p>
<p>3. The subunits encoded by these loci occurred at different levels in the different tissues and organs analyzed.</p>
<p>4. Genetic variants at the three PGI loci were detected in two species of the family Anostomidae (<em>L. friderici</em> and <em>S. nasuttus</em>). In the family Pimelodidae, <em>P. maculatus</em> showed a genetic variant as the Pgi-1 locus.</p>
<p>5. The product of these loci could be separated in three PGI regions based on their electrophoretic mobility, tissue distribution and thermostability properties.</p>
<p>6. The more anodal region (including isozymes and allozymes) was more thermolabile than the less anodal one, which is predominant in a single tissue-skeletal muscle.</p>

A summary of the resource.
The effects on the hippocampal electroencephalogram (EEG) of intraventricular injections of the nicotinic ligand α-<em>Naja naja</em> toxin, and of <em>d</em>-tubocurarine, were studies in rats immobilized with gallamine or anesthetized with urethane. The EEG recordings were taped and processed off-line to calculate power spectra, autocorrelation functions, and averages. In addition, the times at which spike-and-wave complexes appeared were identified and autocorrelation histograms and cross correlations (with the EEG) were made. <em>Naja</em> toxin and <em>d</em>-tubocurarine provoked a 3.5- to 5-Hz theta rhythm in both hippocampi. Higher doses elicited rhythmic epileptic spike and wave complexes which appeared at a preferred phase of theta rhythm. Atropine and medial septal lesions blocked the rhythm and disrupted the rhythmicity of epileptiform activity. We conclude that different neural subsystems sustain the theta rhythm and epileptiform spikes, and discuss the possible mechanisms involved.

A summary of the resource.
Several mutant strains of Rhizobium meliloti isolated after nitrosoguanidine mutagenesis were selected as unable to grow on mannose. Some of them also failed to grow on glucose, fructose, ribose, and xylose but grew on L-arabinose, galactose, and many other carbon sources. Biochemical analysis demonstrated that the mutants lacked NAD- and NADP-linked glucose-6-phosphate dehydrogenase activities that reside on a single enzyme species. One such mutant was found to accumulate glucose-6-phosphate, and this could partially explain the inhibition of growth observed on mixtures of permissive and nonpermissive carbon sources. Symbiotic properties remained unaffected in all these mutants.

Aim: Noninvasive studies of the acetylcholinesterase (AChE) level in Alzheimer's disease (AD) patients can contribute to a better understanding of the disease and its therapeutic. We propose 3-(benzyloxy)-1-(5-[18F]fluoropentyl)-5-nitro-1H-indazole, [18F]-IND1, structurally related to the AChE-inhibitor CP126,998, as a new positron emission tomography-radiotracer. Experimental: Radiosynthesis, with 18F, stability, lipophilicity and protein binding of [18F]-IND1 were studied. In vivo behavior, in normal mice and on AD mice models, were also analyzed. Results: [18F]-IND1 was obtained in good radiochemical yield, was stable for at least 2 h in different conditions, and had adequate lipophilicity for blood–brain barrier penetration. Biodistribution studies, in normal mice, showed that [18F]-IND1 was retained in the brain after 1 h. In vivo tacrine-blocking experiments indicated this uptake could be specifically due to AChE interaction. Studies in transgenic AD mice showed differential, compared with normal mice, binding in many brain regions. Conclusion: [18F]-IND1 can be used to detect AChE changes in AD patients.

A summary of the resource.
Owing to their potential as single-source precursors for compositionally complex materials, there is growing interest in the rational design of multimetallic compounds containing fluorinated ligands. In this work, we show that chemical and structural principles for a materials-by-design approach to bimetallic trifluoroacetates can be established through a systematic investigation of the crystal-chemistry of their monometallic counterparts. A(CF3COO)2·nH2O (A = Mg, Ca, Sr, Ba, Mn) monometallic trifluoroacetates were employed to demonstrate the feasibility of this approach. The crystal-chemistry of monometallic trifluoroacetates was mapped using variable-temperature single-crystal X-ray diffraction, powder X-ray diffraction, and thermal analysis. The evolution with temperature of the previously unknown crystal structure of Mg(CF3COO)2·4H2O was found to be identical to that of Mn(CF3COO)2·4H2O. More important, the flexibility of Mnx(CF3COO)2x·4H2O (x = 1, 3) to adopt two structures, one isostructural to Mg(CF3COO)2·4H2O, the other isostructural to Ca3(CF3COO)6·4H2O, enabled the synthesis of Mg-Mn and Ca-Mn bimetallic trifluoroacetates. Mg0.45Mn0.55(CF3COO)2·4H2O was found to be isostructural to Mg(CF3COO)2·4H2O and exhibited isolated metal-oxygen octahedra with Mg(2+) and Mn(2+) nearly equally distributed over the metal sites (Mg/Mn: 45/55). Ca1.72Mn1.28(CF3COO)6·4H2O was isostructural to Ca3(CF3COO)6·4H2O and displayed trimers of metal-oxygen corner-sharing octahedra; Ca(2+) and Mn(2+) were unequally distributed over the central (Ca/Mn: 96/4) and terminal (Ca/Mn: 38/62) octahedral sites.
