Cannabis use during pregnancy has recently become an important concern due to its increasing trend and association with neurodevelopmental alterations of exposed children. Although inhalation is the primary route of cannabis consumption in humans, few studies replicated this route in preclinical models of prenatal exposure. This study aimed to analyze the effects of chronic prenatal exposure to vaporized cannabis (PEVC) on hippocampal neurodevelopment, functionality, and learning and memory processes. Using a commercially available high-potency cannabis strain (THC 14.7%), pregnant rats were exposed to cannabis vapor from gestational day 8 to 21. A combination of primary hippocampal cultures, electrophysiological studies in slices, and behavioral tests was employed to assess the impact of PEVC. We found that PEVC induced presynaptic remodeling of hippocampal neurons in offspring under both basal and activity-dependent conditions by increasing synaptic vGlut1 abundance and the total recycling pool of synaptic vesicles. Chronic PEVC also induced a downregulation of CB1R at glutamatergic and GABAergic synapses. Additionally, an increase in axonal recruitment and synaptic efficacy was observed at hippocampal CA1 synapses of juvenile offspring. Moreover, long-lasting cognitive impairments were identified during adolescence, including deficits in spatial memory in male and female offspring. In summary, these findings demonstrate that exposure to high-potency cannabis vapor during pregnancy leads to significant changes in synaptic remodeling and efficacy, axonal recruitment, and long-term cognitive consequences in offspring.

Se estima que se conocen más de un millón y medio de especies de insectos, siendo el grupo más diverso de organismos eucariotas, y que un 10% de estas especies constituyen plagas agrícolas. Los efectos negativos del uso de insecticidas sintéticos en la salud, el medio ambiente y los organismos que viven en él, fomentan la búsqueda y desarrollo de estrategias alternativas para el control de plagas. El uso de semioquímicos (feromonas y kairomonas) en estrategias de manejo de insectos plaga data de varias décadas; su modo de acción involucra la modificación del comportamiento de la especie diana y pueden ser utilizados solos o en conjunto con otros compuestos que sean tóxicos o adhesivos. Los beneficios del uso de feromonas incluyen su inocuidad, ser especie-específicas y por ende, constituyen una herramienta muy sensible a la detección de la plaga en bajas poblaciones. En Uruguay, la utilización de feromonas en programas de manejo integrado de plagas se inició hace ya varias décadas (finales de la década de 1970), aunque se ha visto limitada a especies de amplia distribución e importancia económica internacional. Existen, sin embargo, plagas locales o regionales con alta incidencia económica, como es el caso de Pseudaletia adultera, para las cuales no se han identificado sus feromonas, dificultando el desarrollo de estrategias alternativas de control. Pseudaletia adultera (Lepidoptera: Noctuidae) es un insecto polífago cuyas preferencias alimenticias son las gramíneas cultivadas y gramíneas de praderas naturales; se ha reportado la presencia de esta plaga a nivel local (Norte de Argentina, Sur de Brasil, Paraguay y Uruguay). Su importancia económica radica en la frecuencia de los ataques (en Uruguay pueden presentar de 3 a 4 generaciones al año) y la magnitud de los mismos, pudiendo ocasionar daños a hojas, tallos, espigas y granos. En la actualidad, para el muestreo de larvas de esta especie se recomienda el uso de red entomológica; el monitoreo de adultos se realiza mediante trampas de luz. El uso de feromonas sexuales en trampas de monitoreo es un método sensible y específico para lepidópteros, permitiendo detectar adultos incluso en muy bajas densidades, y predecir explosiones poblacionales. De esta manera, se pueden establecer umbrales de capturas para la adopción de medidas de control químico. 2 De acuerdo a lo antedicho, el desarrollo de esta tesis se enmarca en la identificación de la feromona sexual de P. adultera, a través del conocimiento de la biología del insecto, la extracción de glándulas productoras de feromona, identificación química de los compuestos activos y formulación y evaluación de una mezcla sintética que simule la feromona sexual. El presente informe de tesis de maestría consta de una introducción que incluye los antecedentes y los objetivos, dos capítulos centrales que describen el trabajo experimental realizado. En el primero se describe la aproximación a la identificación de los componentes de la feromona sexual de Pseudaletia adultera, y el segundo describe los estudios comportamentales en laboratorio y ensayos a campo.

<p>Temperature variation can promote physico-chemical and microbial changes in the water transported through distribution systems and influence the dynamics of biofilms attached to pipes, thus contributing to the release of pathogens into the bulk drinking water. An experimental real-scale chlorinated DWDS was used to study the effect of increasing temperature from 16 to 24�C on specific pathogens, bacterialfungal<br />communities (biofilm and water samples) and determine the risk of material accumulation and mobilisation from the pipes into the bulk water. Biofilm was developed for 30 days at both temperatures in the pipe walls, and after this growth phase, a<br />flushing was performed applying 4 gradual steps by increasing the shear stress. The fungal-bacterial community characterised by Illumina MiSeq sequencing, and specific pathogens were studied using qPCR: Mycobacterium spp., Mycobacterium avium<br />complex, Acanthamoeba spp., Pseudomonas aeruginosa, Legionella pneumophilia, and Stenotrophomonas maltophilia. Sequencing data showed that temperature variation significantly modified the structure of biofilm microbial communities from the early stages of biofilm development. Regarding bacteria, Pseudomonas increased its relative abundance in biofilms developed at 24�C, while fungal communities showed loss of diversity and richness, and the increase in dominance of Fusarium genus.<br />After the mobilisation phase, Pseudomonas continued being the most abundant genus at 24° C, followed by Sphingobium and Sphingomonas. For biofilm fungal communities after the mobilisation phase, Helotiales incertae sedis and Fusarium were<br />the most abundant taxa. Results from qPCR showed a higher relative abundance of Mycobacterium spp. on day 30 and M. avium complex throughout the growth phase within the biofilms at higher temperatures. The temperature impacts were not<br />only microbial, with physical mobilisation showing higher discolouration response and metals release due to the increased temperature. While material accumulation was<br />accelerated by temperature, it was not preferentially to either stronger or weaker biofilm layers, as turbidity results during the flushing steps showed. This research yields new understanding on microbial challenges that chlorinated DWDS will undergo as global temperature rises, this information is needed in order to protect drinking water quality and safety while travelling through distribution systems.</p>

La digestión anaerobia (DA) es una biotecnología que tiene por objetivo la degradación de materia orgánica y la producción de metano. El proceso es llevado a cabo por un consorcio de microorganismos anaerobios. La DA ha tenido un gran impacto en el tratamiento de aguas residuales con alto contenido en materia orgánica y residuos urbanos, ya que además de tratar el efluente se obtiene energía. En nuestro país se está aplicando esta tecnología en algunas industrias dentro del sector lácteo y de bebidas, entre otros. Si bien la DA es una tecnología consolidada y con gran aplicación, aún se conoce poco la microbiología de estos sistemas en escala real y en particular hay muy pocos estudios de la microbiología de reactores durante su período de arranque. Aprovechando la instalación y puesta en marcha de dos reactores anaerobios en industrias de nuestro país surge esta tesis, en la cual se planteó entre otros objetivos, conocer la dinámica de las comunidades microbianas presentes en dos reactores UASB a escala real durante el período de “arranque”. Ambos reactores fueron inoculados con lodos provenientes de una laguna anaerobia, uno de ellos trató vinaza, generada en la producción de bioetanol a partir de caña de azúcar y el otro efluente lácteo, rico en grasas. Se estudió la comunidad microbiana en muestras de biomasa colectadas durante aproximadamente un año. Se realizaron análisis fisicoquímicos y se analizó la comunidad microbiana por secuenciación masiva el gen del rARN de 16S procariota. Para estudiar la evolución de la población metanogénica se realizaron actividades metanogénicas y se cuantificó por PCR cuantitativo el gen del rARN de 16S de arqueas y el gen mcrA, un gen funcional marcador de la metanogénesis. Los resultados muestran que en términos generales el efecto del tipo de efluente sobre la selección de los grupos de microorganismos -que luego se volvieron dominantes- fue el fenómeno más evidente. En lo que respecta a los consorcios bacterianos, en ambos reactores dominaron los mismos filos, aunque en distintas proporciones. Los filos Chloroflexi, Firmicutes, Proteobacteria, Synergistetes y Verrucomicrobia, junto con el cand. phyl, Hyd 24-12 fueron los grupos dominantes luego de la adaptación al efluente lácteo. Los Clostridiales fueron un grupo resistente y los Synergistetes fueron un grupo resiliente frente a una perturbación fuerte en el pH del reactor que trató efluente lácteo. En el reactor de vinaza los filos Firmicutes, cand. phyl. Hyd 24-12 y Verrucomicrobia demostraron ser resistentes a la vinaza y a las cargas orgánicas aplicadas. Por otra parte, la detención de la alimentación del reactor durante dos períodos de aproximadamente cinco meses no afectó a la comunidad microbiana que permaneció en un estado de latencia. Otro objetivo de esta tesis fue determinar cuáles son los grupos de metanogénicos predominantes en reactores a escala real. Este objetivo se fijó para revisar un postulado en la DA en el cual se estableció que el 70 % de la metanogénesis ocurre por la vía acetoclástica y el 30 % por el consumo de H2 y CO2. Aunque, datos preliminares de actividades viii metanogénicas realizadas en algunos reactores nacionales sugerían lo contrario. En esta parte, se analizaron dos reactores UASB a escala real de la industria láctea, uno de los cuales se encontraba durante su etapa de arranque y el otro durante su 8vo año de operación estable. Se colectaron muestras de biomasa durante aproximadamente un año y se cuantificaron los diferentes grupos de metanogénicos mediante PCR cuantitativo utilizando primers y sondas específicos para el gen del rARN de 16S de los diferentes grupos. Además se realizaron ensayos de actividad metanogénica a partir de acetato y a partir de H2 y CO2. Los resultados moleculares mostraron que las arqueas hidrogenotróficas fueron las dominantes. Estos resultados fueron concordantes con los resultados de las actividades metanogéncias que muestran una mayor actividad hidrogenotrófica. Los Methanobacteriales fueron el orden hidrogenotrófico dominante, seguido del orden Methanosarcinales. Si bien esto no es una demostración que pueda generalizarse a todos los reactores a escala real, es evidencia de que la dominancia de los acetoclásticos no sería un paradigma sino que se cumpliría en determinadas condiciones ambientales. Conocer las dinámicas de las comunidades microbianas durante períodos tan particulares como la etapa arranque brinda herramientas para poder mejorar dichos tiempos. Esto a su vez impacta en la reducción de costos y por lo tanto en la competitividad económica de la digestión anaerobia frente a otras tecnologías de tratamiento de residuos. Finalmente, los hallazgos de este trabajo fueron fruto del estudio integrando de la Ingeniería y la Microbiología, aplicado a los reactores. El conocido efecto adverso sobre la salud de los acidos grasos trans y las recomendaciones relativas a la necesidad de eliminacion de los mismos en las grasas destinadas a la elaboracion de alimentos, ha obligado a la industria alimentaria a buscar alternativas que permitan sustituir las grasas parcialmente hidrogenadas por materiales grasos de funcionalidad similar. Si bien no se trata de una problematica reciente, aun la industria exige soluciones frente a disposiciones legales cada vez mas exigentes y la demanda de productos mas saludables por parte del consumidor. En este escenario, el aceite de salvado de arroz (RBO) aparece como una materia prima particularmente atractiva, ademas de ser un derivado obtenido de una actividad agropecuaria de gran importancia en nuestro pais, presenta propiedades particulares que lo diferencian del resto de los aceites comunes producidos en nuestro medio. Su elevado contenido de acido palmitico (~ 18 %) le confiere una inusual heterogeneidad en relacion al largo de cadena de sus acidos grasos. Adicionalmente, el alto porcentaje de insaponificables ricos en compuestos bioactivos le confieren capacidad antioxidante y efectos beneficos sobre la salud humana. Pese a estas caracteristicas su uso no se ha diversificado y actualmente se lo utiliza como tal, destacandose su uso como aceite para fritura, dada su elevada estabilidad oxidativa. En este trabajo se propone el diseno de fases grasas cero-trans con propiedades atractivas para destinar a alimentos que requieren de materiales lipidicos con estructura, a partir exclusivamente de este aceite. Para ello se lo hidrogeno completamente, se lo mezclo en diferentes proporciones con el aceite nativo sin hidrogenar y finalmente las mezclas se sometieron a una etapa de interesterificacion enzimatica. Los resultados mostraron que la interesterificacion genero productos mas pobres en los triacilgliceroles (TAG) caracteristicos tanto del FHRBO (tri-saturados) como del RBO (tri-insaturados), generandose nuevas especies de TAG de grado de saturacion intermedia (di-saturados mono-insaturados y di-insaturados mono-saturados). Estos cambios generaron productos compuestos por TAG mas compatibles que los de las mezclas de partida. 3 El analisis por calorimetria diferencial de barrido mostro que los productos presentaron menor temperatura de fusion completa que las mezclas correspondientes, asi como menores contenidos de solidos (SFC), determinados por analisis de resonancia magnetica nuclear de pulsos (p-NMR). Como consecuencia de estos cambios, los productos mostraron rangos de fusion y contenido de solidos que sugieren una plasticidad mejorada con respecto a la de las mezclas. Adicionalmente, el analisis por difraccion de Rayos-X (XRD) mostro una fuerte tendencia del FHRBO a cristalizar bajo la forma ƒÀf, lo que se puede atribuir a la heterogeneidad aportada por su relativamente alto contenido de acido palmitico. Si bien tal tendencia disminuyo a medida que se redujo el porcentaje de FHRBO en las mezclas, el efecto se revirtio en los productos de interesterificacion, los que presentaron tambien una marcada tendencia ƒÀf. En linea con lo anterior la morfologia cristalina determinada por microscopia de luz polarizada (PLM) tambien mostro cambios en el tipo de agregados cristalinos, prevaleciendo en los productos esferulitos mas regulares y homogeneos, a diferencia de los agregados irregulares presentados por las muestras. Se ensayo la formulacion de margarinas a partir de algunos productos seleccionados, encontrandose que aquella elaborada con el producto proveniente de la mezcla conteniendo 30% de FHRBO resulto con propiedades muy similares a la de una margarina de mesa comercial. Los resultados obtenidos demostraron que la hidrogenacion completa del aceite de salvado de arroz, su mezcla con aceite sin hidrogenar y la posterior interesterificacion enzimatica de la mezcla, ofrece una metodologia versatil para la obtencion de fases grasas cero-trans con diferentes propiedades fisicoquimicas, aptas para diferentes aplicaciones en el area alimentaria. Estos materiales, ademas de ofrecer una alternativa a las grasas parcialmente hidrogenadas tradicionales, contienen compuestos bioactivos beneficiosos para la salud, ofreciendo ademas la posibilidad de potenciar el uso de un aceite nacional de mayor interes.

The protein DBC1 is the main SIRT1 regulator known so far, and by doing so, it is involved in the regulation of energy metabolism, especially in liver and fat adipose tissue. DBC1 also has an important function in cell cycle progression and regulation in cancer cells, affecting tumorigenesis. We recently showed that during quiescence, non-transformed cells need DBC1 in order to re-enter and progress through the cell cycle. Moreover, we showed that deletion of DBC1 affects cell cycle progression during liver regeneration. This novel concept prompted us to evaluate the role of DBC1 during adult neurogenesis, where transition from quiescence to proliferation in neuronal progenitors is key and tightly regulated. Herein, we analyzed several markers of cell cycle expressed in the dentate gyrus of the hippocampus of controls and DBC1 KO adult mice. Our results suggest a reduced number of neuroblasts therein present, probably due to a decline of neuroblast generation or an impairment in neural differentiation. In agreement with this, we also found that adult DBC1 KO mice had a reduction in the volume of the granule cell layer of the dentate gyrus. Interestingly, behavioral analysis of KO and control mice revealed that deletion of DBC1 parallels to specific cognitive impairments, concerning learning and possibly memory forma tion. Our results show, for the first time, that DBC1 plays an active role in the nervous system. In particular, specific anatomical and behavioral changes are observed when is absent.

<p>Se presenta un esquema analítico sencillo para dosificar Prednisona y Prednisolona en un gran número de asociaciones con otros fármacos. Sin embargo, en presencia de ciertos analgésicos la sistemática propuesta pierde validez. Se expone entonces, una puesta a punto de la dosificación de Prednisona y un tranquilizante y/o relajante muscular (Meprobamato o Carisoprodol).</p>
<p>1) Criterios generales para dosificar prednisona y prednisolona en preparaciones farmacéuticas de polifármacos.</p>
<p>El análisis de preparaciones farmacéuticas de polifármacos es siempre para el Químico Analista un trabajo de búsqueda.</p>
<p>La bibliografía disponible, Farmacopeas en primer lugar y publicaciones especializaddas en segundo lugar, es muy pobre en análisis de asociaciones medicamentosas.</p>
<p>Este hecho conocido, se debe principalmente, a que son muy pocas las asociaciones que tienen realmente significado farmacoclínico y además las técnicas analíticas de las especialidades (Marcas de fábrica)con polifármacos, no son publicitadas.</p>
<p>El trabajo dividirá el enfoque de la siguiente forma:</p>
<p>A) Resumen de métodos dados por diferentes farmacopeas para Prednisona, Prednisolona y algunos preparados y derivados.</p>
<p>B) Características de las diferentes reacciones utilizadas para dosificar Prednisona y Prednisolona (Ampliando a otros corticoides).</p>
<p>C) Generalizaciones de los métodos oficiales para preparacionesde mono y polifármacos.</p>
<p>D) Validez experimental de los esquemas propuestos.</p>

Vascular endothelial growth factor (VEGF) is one of the classic factors involved in tumor-induced angiognesis in several solid tumors. Bevacizumab, a monoclonal antibody against VEGF, can be used as an imaging tool in preclinical studies. The aim of this study was to radiolabel Bevacizumab with (99m)Tc and to evaluate in vivo its imaging properties in an adenocarcinoma animal model. For this purpose, Bevacizumab was derivatized with Suc-HYNIC as a bifunctional coupling agent. A mixture of Tricine/SnCl(2).2H(2)O was added to Bevacizumab-HYNIC and radiolabeled with (99m)TcO(4)(-). The radiochemical stability of the radiolabeled antibody was assessed. Biodistribution and scintigraphy imaging were performed in normal CD1 female mice and in spontaneous adenocarcinoma tumor bearing CD1 mice (n = 5). We demonstrated that 99mTc-HYNIC-Bevacizumab was stable. In vivo biodistribution studies revealed that tumor uptake of (99m)Tc-HYNIC-Bevacizumab was 1.37 ± 0.51% and 5.33 ± 2.13% at 4 and 24 h postinjection, respectively. Scintigraphy image studies showed tumor selective uptake of (99m)Tc-HYNIC-Bevacizumab in the tumor-bearing mice. We conclude that (99m)Tc-HYNIC-Bevacizumb has the potential to be used as a tracer for tumor imaging in preclinical studies.

El pronóstico del cancer es dependiente de vanos factores, y varia considerablemente segun de que tipo de cancer se trate. Muchos tipos de canceres involucran celulas que sobreexpresan receptores de factores de crecimiento epidermico (EGFRs) y se ha estudiado que el crecimiento de estas celulas se regula, en parte, por el estimulo aut6crino 0 paracrino de dichos receptores. La familia de receptores EGFR esta formada por cuatro miembros, HERl, HER2, HER3, HER4, todos ellos estrechan1ente relacionados entre si y que estan localizados en la membrana celular. Dichos receptores pueden encontrarse en su forma monomerica 0 dimerizados y, tras su uni6n alligando, se produce una cascada de senales que resulta en la divisi6n celular. Se ha demostrado que la sobreexpresi6n de este tipo de receptores esta con'elacionada con la recurrencia y un peor pron6stico de la enfermedad. Estos hechos llevaron al desarrollo de biomoleculas dirigidas especificamente contra el dominio extracelular de los receptores EGFRs y que actual mente se aplican al diagn6stico y la terapia mediante radiofarmacos en Medicina Nuclear. El objetivo general de este trabajo consisti6 en estudiar la marcaci6n de moleculas blanco-especificas con 99mTc y 177Lu aplicadas a desarrollar potenciales radiofarmacos de diagn6stico y terapia de cancer. Nimotuzumab, anticuerpo que reconoce especificamente el HERl, se encuentra en camino de ser aprobado como agente para el tratamiento de gliomas, cancer pancreMico, cabeza y cuello, entre otros.Nuestros resultados indican que 177Lu-DOTA-Nimotuzumab tiene una excelente captaci6n tumoral por un tiempo prolongado indicando su potencial aplicaci6n como agente terapeutico en tumores EGFR positivos. Los estudios bio16gicos de 99mTc-HYNIC-Fab(Nimotuzumab) mostraron una eliminaci6n diferente a la del anticuerpo entero. La misma se dio principalmente por via renal y fue posible observar leve captaci6n a nivel tumoral que nos alientan a continuar la linea de investigaci6n como po sible agente de imagenologia molecular en tun10res que sobreexpresan EGFR. Por otro lado se estudi6 un hexapeptido, KCCYSL que reconoce especificamente el HER2, marcado con 177Lu. En los estudios bio16gicos se observ6 una rapida eliminaci6n por via renal y una buena captaci6n tumoral a tiempos cortos, en relaci6n al tejido sano, indicando que podria ser un agente adecuado para aplicaciones teran6sticas de tumores HER2 positivos. Asimismo los estudios del 99mTc-HYNIC-KCCYSL muestraron una leve capaci6n tumoral pero suficiente para observar el tumor y que nos permiten avanzar en la investigaci6n del como posible agente de imagenologia molecular en tumores con sobreexpresi6n de HER2. El desarrollo de este tipo de agentes es de gran relevancia clinica debido a la creciente necesidad de brindar analisis personalizados a los pacientes con cancer y proporcionar a estos una adecuada selecci6n y monitoreo de su tratamiento.

La imagenología molecular es una disciplina que abarca innumerables herramientas. Entre sus aplicaciones se encuentra el diagnóstico in vivo del cáncer con agentes que reconocen marcadores tumorales y que a su vez poseen óptimas características para obtención de imágenes. Asimismo, proporciona información clínicamente fundamental, permitiendo una correcta selección del tratamiento a seguir y el monitoreo de sus efectos. Detalles como la localización, tamaño, morfología y cambios estructurales de un área afectada pueden ser detectados a través de los métodos convencionales de imagen. Sin embargo, técnicas de imagenología molecular como la SPECT, PET y FMT, entre otras, son cada vez más utilizadas. Debido a su característica no invasiva la imagen molecular permite evaluar la patología en su contexto, siendo clave para entender el proceso tumoral, sin perturbar el ambiente y proporcionando información adicional a los métodos convencionales. La estadificación tumoral, detección de metástasis, cirugía guiada, así como la cuantificación de una lesión son diferentes aplicaciones en este área. En este contexto, es muy importante el conocimiento de marcadores tumorales y el desarrollo de nuevos agentes de imagenología molecular específicos de estos. Los aptámeros son oligonucleótidos (DNA o RNA) que tienen la característica de reconocer su blanco con alta afinidad y especificidad. Similar a los anticuerpos en estas características, los aptámeros presentan ventajas que los hacen interesantes para su aplicación como agentes de imagen o terapia. El Sgc8-c es una secuencia truncada del aptámero sgc8, el cual muestra una unión específica y con alta afinidad al receptor PTK7. Sgc8-c tiene solamente 41 bases y una Kd de 0.78 nM para este receptor. Si bien este receptor está presente en células normales, su sobreexpresión ha sido observada en cáncer de colon, tumores gástricos, cáncer de pulmón, próstata, mama e incluso en metástasis. Si bien, su participación en la oncogénesis parece ser clara, aun no es un receptor extensamente caracterizado. El presente trabajo consistió en la investigación y desarrollo del aptámero Sgc8-c como agente de imagenología molecular en cáncer. Para ello Sgc8-c fue modificado en su extremo 5’ con HYNIC, DOTA y se marcó con Tecnecio-99m y Galio-67 respectivamente. Asimismo el aptámero Sgc8-c fue marcado con Alexa647, para su evaluación como agente de imagen óptica en el infrarrojo cercano. Todos los agentes desarrollados fueron evaluados fisicoquímicamente y frente a diferentes líneas celulares tumorales. Finalmente, las características de biodistribución y farmacocinética fueron estudiadas y tres modelos tumorales en ratón fueron seleccionados para realizar imágenes. Resumen 3 Molecular imaging is a discipline that involves many tools. One of its major applications is the in vivo diagnosis of cancer, using molecular probes that recognize tumor markers and have optimal characteristics for imaging. It also provides clinically important information, allowing the correct choice of treatment options and monitoring their effects. Details such as the location, size, morphology and structural changes of an affected area can be detected by conventional imaging methods. Moreover, molecular imaging techniques such as SPECT, PET and FMT, among others, are increasingly used. Due to its minimal invasive characteristics, molecular imaging allows the evaluation of the pathology in context, being key to understanding tumor process without disturbing the environment and providing additional information to conventional methods. Tumor staging, detection of metastasis, guided surgery, as well as quantification of disease are different applications in this area. In this context, knowledge of tumor markers and the development of new targeted molecular imaging agents is very important. Aptamers are oligonucleotides (DNA or RNA), which have the characteristics to recognize their target with high affinity and specificity. Similar to antibodies in their molecular recognition properties, aptamers offer advantages that make them interesting for use as imaging agents or therapy. Sgc8-c is a truncated aptamer sequence, which exhibits specific binding and high affinity for the receptor PTK7. The Sgc8-c aptamer has only 41 bases, but its Kd for its receptor is 0.78 nM. Although this receptor is present in normal cells, its overexpression has been observed in colon, gastric tumors, lung, prostate, breast cancer and even their metastases. While participation in oncogenesis seems clear, the receptor needs to be characterized more extensively in vivo as a valid biomarker. The work presented describes the research and development of aptamer Sgc8-c as a molecular imaging agent for cancer. Sgc8-c was modified at its 5 'end with radiometal chelators HYNIC and DOTA and labeled with technetium-99m and gallium-67 respectively. Also, the Sgc8-c aptamer was labeled with the near infrared dye Alexa647 for evaluation as an optical imaging agent. All agents were developed and evaluated physicochemically against different tumor cell lines. Finally, the pharmacokinetics and biodistribution characteristics of the labeled Sgc8-c aptamer were studied and three mouse tumor models were selected for imaging.

Aptamers are oligonucleotides that have the characteristic of recognizing a target with high affinity and specificity. Based on our previous studies, the aptamer probe Sgc8-c-Alexa647 is a promising tool for molecular imaging of PTK7, which is an interesting biomarker in cancer. In order to improve the delivery of this probe as well as create a novel drug delivery nanosystem targeted to the PTK7 receptor, we evaluate the co-association between the probe and preformed nanostructures. In this work, preformed pegylated liposomes (PPL) and linear and branched pristine polymeric micelles (PMs), based on PEO–PPO–PEO triblock copolymers were used: poloxamer F127® and poloxamines T1307® and T908®. For it, Sgc8-c-Alexa647 and its co-association with the different nanostructures was exhaustively analyzed. DLS analysis showed nanometric sizes, and TEM and AFM showed notable differences between free- and co-associated probe. Likewise, all nanosystems were evaluated on A20 lymphoma cell line overexpressing PTK7, and the confocal microscopy images showed distinctness in cellular uptake. Finally, the biodistribution in BALB/c mice bearing lymphoma-tumor and pharmacokinetic study revealed an encouraging profile for T908-probe. All data obtained from this work suggested that PMs and, more specifically T908 ones, are good candidates to improve the pharmacokinetics and the tumor uptake of aptamer-based probes.

Para extender y fortalecer la Mejora Continua en la “Unidad de Educación Permanente” (UNEP) de la Facultad de Química (FQ) de la Universidad de la República de Uruguay (UdelaR) se emplearon herramientas clásicas de ”Gestión de Calidad” que han demostrado su eficacia en distintas ramas de las actividades Empresarial y Educativa. La experiencia permitió identificar “oportunidades de mejora” e implementar las acciones de seguimiento correspondientes, La experiencia realizada es considerada una muy buena práctica de trabajo en consideración a los resultados obtenidos. Además, su ejecución implicó un uso mínimo de recursos disponibles, se aplicó fácilmente la metodología de trabajo, se logró una comprensión rápida de los participantes y gran empatía por parte de los actores involucrados. En resumen, se consiguió un excelente cumplimiento de los objetivos propios establecidos por el Consejo de Facultad de Química (CFQ) para la función de “Educación Permanente” (EP) de la FQ. La divulgación de esta experiencia puede servir como ejemplo para su aplicación en múltiples áreas de la actividad docente y no docente de la UdelaR.
