determinación de zinc en leche en polvo y fórmulas infantiles. La determinación se basa en la reacción de Zn(II) con zincón a pH 9 en presencia de citrato de sodio para enmascarar interferentes de la matriz con detección espectrofotométrica a 612 nm. El sistema fue optimizado mediante experimentos multivariantes. El sistema validado presentó un rango lineal de 0.4 a 10 mg L-1, la precisión del método fue mejor que 5% (sr,%; n=10). El límite de detección fue 0.13 mg L-1. Los resultados obtenidos utilizando el método propuesto y el de referencia fueron estadísticamente equivalentes. La frecuencia de muestreo fue de 75 muestras/hora y el consumo de muestra y reactivo 0.24 mL por determinación. El método propuesto resultó veraz, simple, rápido y está de acuerdo a los principios de la química verde, generando apenas 0.24 mL de residuos químicos

Abstract

Las frutas y hortalizas pueden contener contaminantes que llegan al producto en cualquier punto de la cadena productiva. En este sentido, se debe controlar la acumulación de elementos potencialmente tóxicos como arsénico (As), cadmio (Cd) y plomo (Pb) para asegurar y controlar la inocuidad de estos alimentos. Con el objetivo de monitorear los contaminantes inorgánicos, de forma rápida, confiable y con métodos analíticos en acuerdo con los principios de la química verde, se desarrolló y validó una metodología para la extracción simultánea de estos elementos asistida con microondas, en un solo paso y utilizando ácido diluido.

En el presente trabajo se realiza el diseño de un agente de revelado o revenido químico basado en una pasta conteniendo el reactivo de Fry (una solución de CuCl2 en HCl) y bentonita. Esta pasta de revelado, puede aplicarse a diversas muestras de hierro y aceros similares a las encontradas en muchos motores de vehículos. La eficiencia del revelado es similar a la que presenta el uso del reactivo líquido. Además, esta pasta de revelado presenta ventajas para su aplicación, pues evita los problemas de derrame de soluciones ácidas durante su aplicación por técnicos y peritos policiales. Los ensayos por microscopía electrónica muestran asimismo que no se produce una alteración morfológica de la superficie metálica durante el proceso. Actualmente la pasta desarrollada está siendo eficientemente utilizada por personal policial en Uruguay

Abstract
A summary of the resource.
Previamente en nuestro grupo de investigación, en la búsqueda de inhibidores de la Triosafosfato isomerasa de Trypanosoma cruzi (TcTIM) se evaluaron 230 compuestos de nuestra quimioteca, encontrándose que los mejores inhibidores en su mayoría presentan simetría en su estructura. En esa base, se propuso la obtención de moléculas simétricas derivadas de diarilidencetonas de síntesis sencilla, utilizando condiciones de reacción amigables con el ambiente y de bajo costo de producción. Se sintetizaron 25 moléculas de las cuales 8 tuvieron efecto inhibitorio en la TcTIM, 9 presentaron un CI50 frente a T. cruzi menor a 5 μM, destacándose el compuesto (2E,5E)-2,5-di[(E)-3-(2-tienil)aliliden]ciclopentanona (16) con un CI50 de 40 nM, 200 veces más activo que el fármaco de referencia Nfx (CI50 de 8 μM). Además, se obtuvieron índices de selectividad muy superiores con respecto a los fármacos de referencia. Se destacan, además, el compuesto (1E,3E,6E,8E)-1,9-di(2-furanil)nona- 1,3,6,8-tetraen-5-ona (2) (CI50 de 5 μM) y (2E,6E)-2,6-di[(E)-3-(furan-2- il)aliliden]ciclohexanona (11) (CI50 de 0.6 μM) como las más activas y selectivas. El derivado 11 mostró excelentes resultados en la inhibición de la TcTIM con un CI50= 86 nM. Además, el compuesto 11 no fue activo contra la TIM de Trypanosoma brucei, Leishmania mexicana y la humana, demostrando la alta selectividad a la TcTIM. Adicionalmente, estas moléculas no fueron mutagénicas según el test de Ames. Las diarilidencetonas líderes se usaron en dosis combinadas entre ellas o con el fármaco de referencia (Bnz), en busca de efectos sinérgicos. Se encontraron varias combinaciones sinérgicas in vitro. La combinación del compuesto 2+16 in vivo indujo una reducción de más de un 90 % del pico de parasitemia en el modelo murino agudo de la enfermedad de Chagas. En el análisis histopatológico de los diferentes tejidos de los animales tratados con estos compuestos, se observó disminución de la inflamación y la ausencia de amastigotes en el tejido cardíaco. Al estudiar el mecanismo de muerte de los parásitos, por citometría de flujo y por resonancia magnética nuclear de protón, se evidencia que dos de estos compuestos, 2 y 11, producen muerte por apoptosis tardía/necrosis y la combinación de los compuestos 2+16 induce la muerte de los parásitos por apoptosis.

Introducción: la infección por Estreptococo grupo B (EGB) puede afectar gravemente a la madre y al feto durante la gestación y al recién nacido luego del parto. Actualmente, el diagnóstico de colonización durante el embarazo se realiza por métodos microbiológicos a partir de exudados vaginorrectales. Objetivo: desarrollar métodos rápidos y de bajo costo para la detección del antígeno grupo específico de EGB en exudados vaginorrectales. Material y método: se utilizaron dos cepas de EGB, una autóctona (IH23) y la cepa de referencia O90R, que solo expresa el polisacárido específico de grupo. Para cada una se preparó un antisuero policlonal que se utilizó para el desarrollo de un test inmunocromatográfico y uno de aglutinación de látex. Como controles se emplearon cultivos bacterianos, polisacáridos purificados de EGB y muestras vaginorrectales. Resultados: los límites de detección obtenidos para la inmunocromatografía fueron de 210 μg/ ml y 50 μg/ml para los polisacáridos purificados de sobrenadante y pared, respectivamente, no lográndose detectar antígenos de EGB en las muestras clínicas analizadas. El límite de detección del látex fue 65 μg/ml frente al polisacárido purificado de sobrenadante de cultivo de IH23 y 6,5 x 107 UFC/ml de IH23. La sensibilidad y especificidad para el látex fue de 30% y 90%, respectivamente.

<p>El cáncer de mama es la principal neoplasia en mujeres y la segunda causa de muerte por cáncer a nivel mundial (sólo el cáncer de pulmón mata a más mujeres).<br />Aproximadamente dos de cada tres cánceres de mama muestran una sobreexpresión de receptores de hormonas, en particular del receptor de estrógeno (RE) y receptor<br />de progesterona (RP).<br />El presente trabajo plantea contribuir al diagnóstico y caracterización del cáncer de mama receptor de estrógeno positivo (RE+), a través del desarrollo de potenciales radiofármacos para Imagenología molecular dirigidos al RE.<br />La primer etapa del trabajo consistió en la síntesis, purificación y formulación del 16α-[18F]fluoroestradiol ([18F]FES), radiofármaco considerado como de referencia en Imagenología de RE en un módulo automatizado de síntesis Synthra RNplus Research.<br />Una vez puesta a punto la misma y realizados tres lotes pilotos se procedió a efectuar estudios in vitro, en donde se empleó la línea celular MCF-7 portadora de RE, derivadas de un adenocarcinoma mamario humano. Los estudios demostraron una elevada captación celular, siendo la misma de 6.3 ± 0.4 %.<br />Al mismo tiempo, se trabajó en la síntesis de ligandos derivados de estradiol para la formación de compuestos de coordinación con 99mTc. Para la síntesis de los mismos, se tomó como reactivo de partida al etinilestradiol, fármaco agonista de los receptores de estrógenos. Se probaron distintas estrategias sintéticas para la obtención del ligando Cyclam-2-(2-(1H-1,2,3-triazol-1-il)acetamida) estradiol (L1) sin éxito, lo que llevó a proponer un nuevo ligando: Cyclam-3-(2-(2-(3-(1H-1,2,3 triazol)propoxi)etoxi)etoxi) propanamina estradiol (L2) que permite la formación de un complejo de 99mTc. Debido a que no fue posible su adecuada marcación, se diseñó un tercer ligando: 17[4-((2-azido-1-carboxietil)amino)-N-metilidino-4-oxobutano-1-1aminio)-1H-1.2.3-triazol-1-il] estradiol (L3), que permite la formación de un complejo de 99mTc(III) “4+1”, utilizando como coligando a el ácido 2,2',2''nitrilotriethanethiol-2-((2-carboxietoxi)metil)propano-1,3-di(oxo)dipropanoico (NS3(COOH)3). Dicho complejo fue obtenido con una adecuada pureza radioquímica. Los estudios fisicoquímicos demostraron una alta hidrofilia ya que presentó un log P de -2.05 ± 0.20. El complejo resultó ser estable en plasma a 37°C y en el medio de reacción durante al menos 4 horas. La unión a proteínas plasmáticas fue moderada siendo la misma de 32 ± 7 %. Los ensayos in vitro de unión a células MCF-7 indicaron ausencia de captación celular 0.10 ± 0.02 %, por lo cual no se realizaron estudios in vivo. Se planteó como posible hipótesis que la razón de ausencia de unión pudiera ser el bajo valor de lipofilicidad fuera del rango adecuado para atravesar barreras biológicas.<br />Los principales logros en el marco de esta tesis fueron: la obtención del [18F]FES, para su uso en clínica y como estándar de comparación de potenciales radiofármacos desarrollados y a desarrollar por nuestro grupo de investigación; por otro lado, se realizaron casi todas las etapas involucradas en el desarrollo de un potencial radiofármaco, desde la selección del farmacóforo y radionucleido, síntesis orgánica, marcación, estudios fisicoquímicos y estudios in vitro.</p>

Nuestro objetivo fue desarrollar dos métodos de marcación con Tc (mediante complejos Tc(III)-4+1 y Tc(I)- tricarbonilos) con un ligando flexible conteniendo el grupo 5-nitroimidazol como farmacóforo biorreducible y el grupo isonitrilo para coordinar el metal. Los complejos fueron comparados a través de su estabilidad, lipofilicidad y unión a proteínas plasmáticas. Ambos complejos fueron obtenidos con adecuada pureza radioquímica (PR) y resultaron estables al menos 4h post-marcado, al menos 2h post-incubación en plasma humano y frente a L-cys. El complejo Tc(III)4+1 presentó una lipofilicidad (log.P) de 0,083 y una unión a proteínas plasmáticas tras 1 hora de incubación de 30% aproximadamente. El complejo Tc(I)-tricarbonílico mostró una menor lipofilicidad (0,065) y unión a proteínas plasmáticas de aproximadamente 10%. De acuerdo a los resultados se espera que el complejo Tc(I)-tricarbonílico presente rápida depuración sanguínea y excreción preferentemente renal, propiedades deseables para un radiofármaco.

El melanoma cutáneo es un tipo de cáncer de piel muy agresivo, altamente metastásico, con elevada mortalidad. La sobrevida se encuentra relacionada a un diagnóstico temprano. El diagnóstico temprano y preciso del melanoma es determinante en la selección del tratamiento óptimo y en la sobrevida del paciente. En este trabajo se evaluó, como agentes de diagnóstico específicos para melanoma, el desempeño in vitro e in vivo de cuatro nuevos péptidos análogos de la hormona alfa melanocito estimulante (α-MSH), ciclados mediante dos estrategias diferentes (enlace lactama y átomo metálico), marcados con 99mTc. Para ello, sobre modificaciones de los péptidos análogos de α-MSH, preparados principalmente a través de síntesis en fase sólida, se estudiaron nuevas estrategias de marcación con 99mTc, utilizando tres ligandos bifuncionales diferentes: uno derivado del sistema coordinante hidracino y otros dos derivados de un sistema tetramino. Los cuatro radiopéptidos se evaluaron fisicoquímica y biológicamente mostrando estabilidad y un buen perfil para la de detección in vivo de células que sobreexpresan el receptor de melanocortina MC1R. Los estudios in vivo de biodistribución y adquisición de imágenes centellográficas y micro-SPECT-CT en ratones portadores de melanoma mostraron concordancia con los perfiles fisicoquímicos y el comportamiento in vitro. Tres de los radiopéptidos presentan una buena captación tumoral, buena eliminación renal y metabolismo hepatobiliar. En comparación con otros radiopéptidos descritos en la bibliografía los desarrollados en este trabajo de Tesis Doctoral comparten características fundamentales como para ser potencialmente usados como agentes de diagnóstico para melanoma.

La eutrofización de los ambientes acuáticos promueve la proliferación de cianobacterias, que producen una amplia variedad de compuestos altamente tóxicos para animales, las cianotoxinas. En Uruguay, los principales sistemas acuáticos (Río Negro, Río Uruguay y Río de la Plata) están seriamente afectados por floraciones de cianobacterias de géneros productores de microcistinas. Estas cianotoxinas son heptapéptidos cíclicos de elevada toxicidad, que interfieren en los principales usos del agua: producción de agua potable, recreación, producción agropecuaria, pesca, entre otros. Para evaluar y gestionar los riesgos asociados, se requieren métodos sencillos, rápidos y validados, que viabilicen su determinación a bajo costo. En esta tesis se desarrolló y validó un kit de ELISA sencillo y robusto, basado en anticuerpos policlonales, que ha permitido monitorear microcistinas en aguas de embalses y playas. Los resultados evidenciaron en espumas cianobacterianas, concentraciones extremadamente elevadas de microcistinas (>1000 veces el límite para agua recreacional), que impulsaron el uso de la bandera sanitaria y sustentaron la detección visual de colonias cianobacterianas dispersas y espumas, como un criterio simple de alerta para la toma de decisiones rápidas en la gestión de playas. En una segunda etapa se avanzó en la estandarización de la inmunodetección, generando anticuerpos monoclonales de llama (Nanobodies), implementando estrategias novedosas de selección para aislar los clones de mayor desempeño analítico. Esto posibilitó el desarrollo de un ELISA de segunda generación sensible y con elevada selectividad. Ambos ensayos se han validado rigurosamente y comparado con métodos analíticos de referencia como LC-MS/MS. El nuevo anticuerpo viabilizó el desarrollo de un ensayo inmunocromatográfico en tiras, que se realiza en minutos en el sitio de interés y es adecuado para analizar aguas ambientales y potables. Para generar mayor información sobre la presencia de congéneres individuales de microcistinas, se desarrollaron métodos de MALDI-TOF cualitativos, y posteriormente cuantitativos incorporando estándares internos de síntesis sencilla. Finalmente, utilizando uno de los nanobodies biotinilado in vivo se generó un inmunoadsorbente equipado con estándar interno, que permite el análisis directo de estas toxinas por MALDI-TOF posibilitando su cuantificación ultrasensible en muestras de agua y matrices biológicas complejas

Existe una creciente preocupación respecto al contenido de cadmio (Cd) en los productos derivados del cacao, como la cocoa en polvo; lo que ha llevado al establecimiento de valores de ingesta máxima tolerable de este metal y a la disminución de los niveles máximos (NM) aceptados. En este sentido, es necesario realizar un control sobre los niveles de Cd en estos alimentos, contando con métodos analíticos confiables y amigables con el medio ambiente. Con ese objetivo se desarrolló y validó un método analítico para la determinación de Cd en cocoas puras y en su preparación con leche, la forma de consumo más popular en nuestro país. El método resultó adecuado para el fin definido y presentó como ventaja un menor consumo de ácidos concentrados que el método de referencia de la AOAC International (2016b). Se aplicó para un sondeo del contenido de Cd en muestras disponibles en el mercado local de Montevideo, Uruguay. Si bien todas las muestras tuvieron concentraciones detectables de Cd, resultaron inferiores al NM establecido.

La soja es uno de los cultivos que sufre el mayor ataque de plagas , principalmente insecto s. Entre ellos se destac an las orugas defoliadoras, como la oruga medidora Rachiplusia n u , junto a otras especies de lepidópteros. Sin embargo la planta de soja tiene una extraordinaria capacidad para compensar la defoliación provocada por esta clase de insectos. Para evitar que este tipo de plagas ocasionen daños que afecten la producción del cultivo se hace necesario recurrir a la aplicación de productos químicos.

Esta presentación propone un cambio metodológico en los trabajos experimentales de síntesis en Química Orgánica, mediante la aplicación de la energía de microondas. La comparación de los resultados obtenidos con los correspondientes a los métodos convencionales permiten evidenciar las ventajas de esta técnica: menores tiempos de reacción, altos rendimientos y p de los productos, mayor disponibilidad de tiempo para completar las etapas de síntesis, así como las ventajas económicas y las relacionadas al medio ambiente.

Abstract

Se han realizado muchos estudios sobre el comportamiento de nanotubos de dióxido de titanio (TiO2) para aplicaciones fotocatalíticas. Sin embargo, se han reportado pocos estudios involucrando metales como niobio (Nb). Los óxidos de niobio presentan propiedades catalíticas muy similares a las del TiO2, con la ventaja de que Brasil concentra el 96% de las reservas mundiales de Nb. Procesos que utilizan técnicas electroquímicas como anodización han sido empleados para la obtención de sólidos nanoporosos en metales. En este estudio, las muestras de metales de niobio fueron anodizadas a 90V durante 90 minutos. El electrolito utilizado en la anodización fue una mezcla de glicerol, NH4F y agua. Las muestras anodizadas fueron tratadas térmicamente a 450°C en atmósfera de N2 por 60 minutos. La caracterización estructural fue realizada por difracción de rayos-X y microscopía electrónica de barrido. El desempeño fotoelectroquímico fue evaluado mediante ensayos de fotocorriente en solución de KOH 0,1 M con diferentes concentraciones de glicerol: 0,1 M y 0,2 M. Se constató la presencia de nanotubos de óxido de Nb luego de la anodización, lo que puede haber contribuido a mejorar el desempeño fotoelectroquímico de los sistemas evaluados. La adición de glicerol a la solución de KOH 0,1 M contribuye al aumento del desempeño fotoelectroquímico.

En Uruguay, las condiciones climáticas favorecen el desarrollo de Botrytis cinerea Pers, pudiendo afectar más del 50% de la producción de un viñedo. En un contexto donde cada vez son menos los fungicidas permitidos, y B cinerea ha presentado resistencia a Benzimidazoles y Dicarboximidas, nace como necesidad desarrollar estrategias de control no químico. Surge como alternativa el empleo de prácticas culturales como el deshojado, comúnmente utilizada en variedades tintas. Con el objetivo de introducir esta práctica en variedades blancas, durante las temporadas 2005/06 y 2006/07, se probaron sobre el cv. Sauvignon blanc., distintas intensidades y momentos de deshojado en combinación con aplicaciones de Caolinita (Sorround®), en un diseño en bloques al azar, con cuatro repeticiones. Todos los deshojados mostraron una incidencia significativamente menor de Botrytis sp, que el testigo sin deshojar. El porcentaje de quemado de sol en racimos, fue significativamente mayor en los tratamientos deshojados sin aplicación de caolinita.

A summary of the resource.
A series of compounds bearing an endocyclic –N–O– moiety with potential antimalarial activity based on simple derivatives of the tropolone purpurogallin was prepared by means of a hetero Diels–Alder reaction using nitrosobenzene as a dienophile. The rationale behind the design of these compounds is presented, together with the synthetic route to derivatives bearing aromatic and aliphatic esters of the C4′-position hydroxyl group of the purpurogallin framework, as well as biological data obtained from in vitro assays against Plasmodium falciparum and Trypanosoma cruzi. Several of the new compounds have activities in the 3–9 μM range, and provide leads for the development of a novel class of antiparasitic drugs with improved biological and pharmacological properties.
