A summary of the resource.
The aim of the present study was to present new evidence supporting the use of saliva as a biological fluid in relative bioavailability studies. Carbamazepine was chosen as a model drug because of its suitability for salivary therapeutic drug monitoring and its well-documented plasma bioavailability. A relative bioavailability study of four different immediate release carbamazepine products was performed. Stimulated saliva samples were collected by chewing on parafilm wax and by the spitting method. In vitro dissolution testing of formulations, using 900 ml of 1% sodium lauryl sulphate in water, was also carried out. The in vitro– in vivo correlations obtained in this salivary study were consistent with previous correlations assessed using plasma. These results support the suitability of saliva as the biological fluid in relative bioavailability studies

Abstract
A summary of the resource.
The synthesis of natural 3¢,3¢¢¢-binaringenin and four related biflavonoids was performed in good overall yield (15–35%) starting from readily available phloroglucinol and 4-hydroxy- or 4-methoxybenzaldehyde. Preliminary results indicate that some of these compounds have an interesting activity against S. aureus.

A summary of the resource.
Selenium is an essential trace element for humans. Its importance is due to selenoproteins, a discrete set of proteins possessing the rare amino acid selenocysteine, an analog of cysteine, which contains selenium instead of sulfur. Selenocysteine is incorporated into proteins by a specific mechanism, superimpose on the canonical translational apparatus, which allows UGA codons to be decoded as selenocysteine. Selenocysteine is more reactive than cysteine, and in selenoproteins of known function is always involved in redox catalysis. The human selenoproteome consists of 25 proteins. In recent years important advances have been made in elucidating the function of many selenoproteins, although specific functions have not been ascribed to all of them. Selenoproteins are important in maintaining redox homeostasis (due to selenoproteins of the thioredoxin and glutathione systems), metabolism of thyroid hormones (due to thyronine deiodinases), sperm cells maturation (mainly due to one of the glutathione peroxidases), and muscle function (mainly due to selenoprotein N), among other physiological processes. Finally, studies on thioredoxin glutathione reductase from flatworm parasites are summarized. In the last years, we have contributed to elucidate the catalytic mechanisms underlying the different activities of this unique multifunctional selenoprotein, which controls the overall flatworm parasites redox homeostasis.

Environmental water chemistry analysis and microcosm toxicity bioassays (MTB) were performed to assess lethality of herbicides on the mole crab Emerita brasiliensis in a sandy beach affected by a freshwater discharge (Andreoni canal) from rice crops. A 5-yr macrocosm field sampling (MFS) was conducted to evaluate freshwater effects on population abundance. Propanil was only detected at the inner portion of the Andreoni canal (IAC), whereas quinclorac and clomazone were found at the IAC and at the canal mouth (CM). A major propanil metabolite was detected at the CM. Herbicides were undetectable at 13 km from the CM. MTB showed an increased susceptibility to propanil with decreasing crab sizes. The MFS showed a drastic decrease in abundance towards the freshwater discharge, concurrently with decreasing salinities. The triad approach that included water analyses, toxicological experiments and long-term field sampling allowed rejecting relationships between herbicide exposure and mole crab lethal effects.

Blends of beef tallow and high-oleic sunflower oil at different proportions were interesterified using enzyme catalysis (Lipozyme TL IM) and the products were analyzed by DSC and HPLC. According to fatty acid composition, the nutritional properties of the products were improved in comparison with the original beef tallow, due to the reduction in the levels of saturated fatty acids and the increase of monounsaturated ones. The melting temperature of the blend with 40% of higholeic sunflower oil decreased from 45 to 37ºC after interesterification, which, from the standpoint of a fatty material to be used in foods, represents a considerable advantage with respect to beef tallow. DSC analysis of the products from interesterification showed the disappearance of polymorphs with high melting points found in the starting blends, possibly corresponding to β-type crystals, something that is also a beneficial aspect of the process under study.

Cryptoblabes gnidiella Milliére (Lepidoptera: Pyralidae) is an economically important exotic pest of vineyards in Southern Brazil and Uruguay. The sex pheromone of C. gnidiella was identified as a mixture of (Z)-11-hexadecenal and (Z)-13-octadecenal, and has been used to monitor populations of this pest in Israel. The development of mating disruption for this species may be hampered by the chemical instability of the natural pheromone components. Therefore, studies on more stable pheromone analogs may provide tools for a control strategy based on behavior-modifying chemicals. We report here the electrophysiological and behavioral responses of C. gnidiella males to (Z)-9-tetradecenyl formate and (Z)-11-hexadecenyl formate, structural analogs of the pheromone components. In gas chromatography-electroantennogramdetection (GC-EAD) studies, both analogs elicited responses from C. gnidiella male antennae. Pre-exposure to the formates did not affect the subsequent EAD responses to the natural pheromone components. The formates acted as pheromone antagonists in wind tunnel tests, inhibiting the responses of males toward both synthetic pheromone and calling females. In the field, captures of males in pheromone-baited traps decreased, in a dose-response pattern, when different amount of formates were added to the pheromone. These pheromone antagonists, thus, are potentially useful as mating disruptants for C. gnidiella in commercial vineyards.

El presente trabajo de tesis se enmarca en una línea de investigación vinculada a la enfermedad de Chagas y desarrollada por muchos años en el actual Centro de Bioinformática Estructural del DETEMA, de la Facultad de Química, Universidad de la República. Esta línea, iniciada con motivo de la tesis doctoral de la patrocinante, la Dra. Paulino, ha dado múltiples trabajos publicados, entre ellos “Modeling and study of cyclosporyn A and related compounds in complexes with Trypanosoma cruzi cyclophilin”, como producto de la primer parte del trabajo presentado en esta tesis. También la línea de investigación ha originado otras tesis de Maestría y Doctorado, entre ellas la de uno de los copatrocinantes, el Dr. Iribarne. Por otro lado no es ajena la mencionada línea al Laboratorio de Inmunotecnología dirigido por el otro copatrocinante, MsC. Battistoni. Mucho se ha investigado con el objetivo de obtener una cura para el Mal de Chagas. Esta Tesis es un aporte más, centrándose en el estudio de la molécula de ciclosporina (CsA), y compuestos estructuralmente relacionados. Estos compuestos han mostrado actividad inhibitoria sobre Trypanosoma cruzi. Si bien la CsA es una droga con actividad inmunosupresora, algunos de los compuestos estructuralmente relacionados activos contra T. cruzi no presentan efectos inmunosupresores. Se ha propuesto a CsA como compuesto líder para el desarrollo de nuevas drogas útiles en la farmacoterapias de la enfermedad de Chagas. Se conocen con cierto grado de detalle las bases bioquímicas de las actividades inmunosupresoras e inhibitorias sobre protozoarios de CsA. En ambas están involucradas una familia de proteínas, las ciclofilinas. Se han publicado datos de las actividades inhibitorias de CsA y compuestos relacionados sobre ciclofilinas presentes en T. cruzi, en particular sobre la ciclofilina mas expresada en T. cruzi, la TcCyP19. Las bases moleculares de la diferencia de actividad inmunosupresora y de inhibición sobre T. cruzi de CsA y de una serie análogos no han sido dilucidadas. Esta tesis pretende, mediante el uso de diversas herramientas de modelado molecular, docking y de mecánica molecular, estudiar el comportamiento de los complejos binarios de TcCyP19 con una serie de compuestos análogos a CsA para obtener información a nivel molecular que aporte a la comprensión de las diferencias encontradas. En forma complementaria, se estudian complejos de CsA y dos de sus análogos con una ciclofilina humana, así como el comportamiento de una proteína mutante de la ciclofilina humana con baja afinidad por CsA. Una vez elaborados los modelos de los sistemas a estudiar se analizaron los posibles sitios de unión de estos compuestos con las ciclofilina en estudio, así como los modos de unión a los mismos. Utilizando simulaciones de dinámica molecular, se obtuvo un muestreo del espacio de fases de los sistemas en estudio que permitió analizar estructuralmente y energéticamente dichos sistemas. Varios términos energéticos se calcularon, que incluyeron energías de interacción ligando-proteína y cálculos de energías libres de asociación por medio del método LIE (Linear Interacction Energy). Se discuten algunos aspectos metodológicos, como la necesidad de hacer varias simulaciones de dinámica molecular para obtener un muestreo mas apropiado del comportamiento del sistema y el cumplimiento de las premisas para usar el método LIE. Los resultados obtenidos muestran que se trata de sistemas con comportamiento muy similar, para los cuales los métodos de cálculo usados no son lo suficientemente sensibles para diferenciar energías libres de unión. Se demostró la posible existencia de lugares de unión de Modelado y estudio de complejos de Ciclosporina A y compuestos relacionados ….. 5 los compuestos estudiados diferentes al sitio catalítico, cuya relevancia deberá ser estudiada in vitro. Todo lo aprendido con estos sistemas permitirá abordar con mejores perspectivas ulteriores etapas del estudio de esta familia de compuestos que involucran complejos ternarios y sus potenciales actividades inmunosupresoras en el huésped o inhibitorias del crecimiento de T. cruzi.

In the search for new therapeutic tools against tuberculosis two novel iron complexes, [Fe(L–H)3], with 3- aminoquinoxaline-2-carbonitrile N1,N4-dioxide derivatives (L) as ligands, were synthesized, characterized by a combination of techniques, and in vitro evaluated. Results were compared with those previously reported for two analogous iron complexes of other ligands of the same family of quinoxaline derivatives. In addition, the complexes were studied by cyclic voltammetry and EPR spectroscopy. Cyclic voltammograms of the iron compounds showed several cathodic processes which were attributed to the reduction of the metal center (Fe(III)/Fe(II)) and the coordinated ligand. EPR signals were characteristic of magnetically isolated high-spin Fe(III) in a rhombic environment and arise from transitions between mS=±1/2 (geff~9) or mS=±3/2 (geff~4.3) states. Mössbauer experiments showed hyperfine parameters that are typical of high-spin Fe(III) ions in a not too distorted environment. The novel complexes showed in vitro growth inhibitory activity on Mycobacterium tuberculosis H37Rv (ATCC 27294), together with very low unspecific cytotoxicity on eukaryotic cells (cultured murine cell line J774). Both complexes showed higher inhibitory effects on M. tuberculosis than the “second-line” therapeutic drugs.

<p>Selenium is a trace element of environmental relevance. Studies on its solution chemistry are scarce and were mostly carried out under experimental conditions of little relevance to environmental research. Thus, we have performed new studies of selenium speciation in solutions of low ionic strength, in contrast to those prevailing in the literature data. In this work, potentiometric titrations (at 20.0 °C, and I = 0.15 mol·L−1 NaClO4) were carried out for systems containing Se(VI) or Se(IV) oxyanions and divalent metal ions (Mg, Ca, Sr, Ba, Mn, Fe, Co, Ni, Cu, Zn, Cd, Hg, and Pb). Ion pairs such as [M(SeO4)] and [M(HSeO4)2], or [M(HSeO3)]+ and [M(SeO3)], exist in solutions. The data reported here provide the basis for determining selenium speciation in natural aquatic systems, on which the bioavailability and toxicity of this element depends.</p>

The aim of the present work was to study the influence of brand on consumers’ expectations and liking of orange-flavoured powdered drinks. Ten samples of orange-flavoured powdered drinks were evaluated by 108 consumers who scored their expected liking (looking at the pack only) and the overall liking under blind and informed-testing conditions (tasting the products). Consumers were also asked to answer a check-all-that-apply question with eight statements related to some products’ characteristics and usage occasions. Results showed that brand and package information had a large impact on consumers’ liking scores only for well-recognized brands. Hierarchical multiple factor analysis performed on liking scores for the three evaluation conditions showed that informed liking scores were closer to expected liking scores than to blind liking scores, suggesting that brand information was more important than hedonic reaction to the sensory characteristics in determining consumers’ liking scores for the samples on the informed condition

Abstract
A summary of the resource.
In a continuing effort to identify new active compounds for combating Chagas disease and other neglected diseases, our research group synthesized and evaluated 23 3-arylquinoxaline-2-carbonitrile di-N-oxides against Trypanosoma cruzi. Five of them presented IC50 values of the same magnitude as the standard drug Nifurtimox, making them valid as new lead compounds. The optimized molecular structures of 23 derivatives represented by 1497 types of DRAGON descriptors were subjected to linear regression analysis, and the derived QSAR was shown to be predictive. In this way, we achieved a rational guide for the proposal of new candidate structures whose activities still remain unknown.

Abstract
A summary of the resource.
This work investigates and compares the reaction performance of soybean oil transesterification under supercritical methanol and ethanol, in a continuous catalyst-free process, as a cleaner alternative to conventional chemically catalyzed process. Reactions were performed in a tubular reactor, at 20 MPa, with oil to alcohol ratio of 1:40, varying the temperature in the range from 250 �C to 350 �C, and at two levels of water concentrations, 0 and 10 wt%. Although both processes proceeded with a relatively high reaction rate, conversion achieved by methanolysis was higher than that obtained by ethanolysis. Water positively affected both process: higher ester content and triacylglycerols depletion occurred when 10 wt% water was used compared with anhydrous conditions. Temperature increase favored the conversion of soybean oil to the corresponding methyl or ethyl esters, although temperatures above 300 �C increased the fatty acid degradation degree, a phenomenon responsible for the low ester contents obtained at the highest temperatures and lowest flow rates studied.

La formación de peroxinitrito en la mitocondria está favorecida por el suministro constante de radical superóxido a partir de la cadena de transporte electrónico mitocondrial y la difusión de óxido nítrico que alcanza a este organelo. La mayor parte de este trabajo fue realizado en un modelo de partículas submitocondriales (PSM), en presencia de succinato y un dador de óxido nítrico, el NOC-­‐7, de forma tal de estudiar la formación y cuantificar al peroxinitrito formado por una multiplicidad de métodos incluyendo fluorescencia, quimioluminiscencia y análisis inmunoquímicos. Mientras que todas las sondas que fueron evaluadas revelaron niveles casi estequiométricos de peroxinitrito respecto a sus radicales precursores, el ácido cumarin borónico (un compuesto que reacciona directamente con peroxinitrito) fue el que mostró un perfil de oxidación más consistente en las PSM formando radical superóxido, en función del flujo de óxido nítrico. Un resultado interesante surgió de los estudios inmunoquímicos que verificaron la generación de radicales proteicos en PSM inducida por peroxinitrito. En otro tipo de enfoque, utilizamos un análogo hidrofóbico de tirosina (N-­‐t-­‐BOC L-­‐tirosina tert-­‐ butil éster, BTBE) pre-­‐incorporado en mitocondrias y PSM, el cual se nitra y oxida por peroxinitrito. En las PSM conteniendo BTBE y formando radical superóxido, la exposición a flujos de óxido nítrico, causó la oxidación y posterior dimerización de la sonda en conexión con procesos de lipoperoxidación. Las PSM suplementadas con sustratos respiratorios redujeron a las Mn(III)porfirinas (MnP) a Mn(II)P, a niveles de oxígeno fisiológicamente relevantes (3-­‐30 μM); a continuación, las Mn(II)P fueron capaces de reducir al peroxinitrito a nitrito y proteger las PSM de la inhibición del consumo de oxígeno por complejo I, de la formación de radicales proteicos y de la nitración de proteínas de membrana. La pre-­‐ incubación de MnP en células endoteliales en cultivo protegió de la nitración de tirosinas proteicas mediada por peroxinitrito a partir de exposición a SIN-­‐1. A través de miscroscopía confocal Raman Resonante detectamos directamente a las MnP dentro de las células, tanto en forma reducida como oxidada; el predominio observado de la forma reducida es totalmente consistente con su rol citoprotector en procesos nitroxidativos. Globalmente, los datos apoyan la formación de peroxinitrito en mitocondrias sometidas a flujos óxido nítrico y demuestran que las MnP pueden participar de un ciclo catalítico redox a nivel mitocondrial para neutralizar el daño oxidativo dependiente de peroxinitrito.

Al administrar un medicamento a un individuo se busca producir una respuesta clínica específica a través de un efecto farmacológico determinado en gran parte por la respuesta farmacocinética, es decir, por las cantidades de fármaco presentes a lo largo del tiempo en los diferentes tejidos y fluidos del cuerpo humano. Medicamento y organismo conforman un complejo sistema que debe ser correctamente interpretado para cumplir con los objetivos terapéuticos de eficacia y seguridad al diseñar un tratamiento farmacológico. La natural variabilidad fisiológica que presentan los individuos a lo largo de una población exige racionalizar la instauración de los mismos, ya que posee un gran impacto en el destino de un fármaco administrado. El sexo como característica fisiológica divide a la población en dos mitades, su relevancia en el campo de la medicina es altamente notoria, hombre y mujer al interaccionar con un medicamento componen sistemas diferentes. Es de esperar entonces que ciertas características específicas al sexo del paciente puedan condicionar el éxito de un tratamiento farmacológico. Durante esta tesis doctoral se analizó la influencia del sexo en la respuesta farmacocinética de principios activos correspondientes a diversos grupos farmacológicos dividiendo el análisis en dos bloques: su efecto sobre la evaluación de biodisponibilidad de medicamentos y su efecto sobre la disposición de fármacos. La diferente discriminación de marcas comerciales de medicamentos que pueden hacer hombres y mujeres, y sus consecuencias sobre las conclusiones de un ensayo de bioequivalencia promedio no habían sido analizadas hasta el momento en profundidad. El análisis realizado demuestra la importancia de tener en cuenta el sexo en la evaluación de bioequivalencia y de diseñar ensayos clínicos a modo de obtener conclusiones sobre ambos grupos. Los resultados hallados aumentan la evidencia respecto a diferentes respuestas farmacocinéticas en hombres y mujeres. Se incorpora la recirculación de fármacos como proceso de disposición relevante, y se introducen modelos no lineales de efectos mixtos para describir y cuantificar su contribución. Al indicar un medicamento, el sexo del paciente debe ser considerado ya que de acuerdo a las características del mismo podría tener un significativo efecto sobre el resultado clínico del tratamiento.

Introducción: las reacciones adversas a medicamentos constituyen un problema fundamental para los servicios de salud, la industria farmacéutica y los organismos reguladores. Gran parte de estas reacciones son relativamente leves y desaparecen al modificar la dosis, pero otras son más graves, pudiendo llegar a la muerte del individuo. El irinotecán es un agente citotóxico activo en cáncer colorrectal y pulmonar. Está asociado a una severa toxicidad hematológica y gastrointestinal, impredecibles en la práctica. Su metabolito activo (SN-38) es detoxificado mediante la enzima UDP-glucuronosiltransferasa-1A1 (UGT1A1). La variación en la actividad de esta enzima ha sido relacionada con polimorfismos en el gen UGT1A1, produciendo los efectos adversos observados. El polimorfismo más importante se presenta en la región promotora y consiste en un número variable de repeticiones timinaadenina. El alelo mayoritario tiene seis repeticiones (TA)6, siendo el polimorfismo de siete repeticiones la variante alélica más frecuente. Objetivo: diseño de un ensayo molecular para el estudio de las variantes alélicas de la región promotora del gen UGT1A1. Material y método: se seleccionó un grupo de 50 voluntarios sin vínculo familiar. Las determinaciones moleculares se realizaron mediante amplificación y posterior secuenciación de la región promotora. * Magister

Prion diseases are a unique category of illness, affecting both animals and humans, where the underlying pathogenesis is related to a conformational change of a normal self protein called cellular prion protein to a pathological and infectious conformer known as scrapie prion protein (PrPSc). Currently, all prion diseases lack effective treatment and are universally fatal. Past experiences with bovine spongiform encephalopathy and variant Creutzfeldt–Jakob disease mainly in Europe, as well as the current epidemic of chronic wasting disease in North America, have highlighted the need to develop prophylactic and/or therapeutic approaches. In Alzheimer’s disease that, like prion disease, is a conformational neurodegenerative disorder, both passive and active immunization has been shown to be highly effective in model animals at preventing disease and cognitive deficits, with emerging data from human trials suggesting that this approach is able to reduce amyloid-related pathology. However, any immunomodulatory approach aimed at a self-antigen has to finely balance an effective humoral immune response with potential autoimmune toxicity. The prion diseases most commonly acquired by infection typically have the alimentary tract as a portal of infectious agent entry. This makes mucosal immunization a potentially attractive method to produce a local immune response that partially or completely prevents prion entry across the gut barrier, while at the same time producing modulated systemic immunity that is unlikely to be associated with toxicity. Our results using an attenuated Salmonella vaccine strain expressing the prion protein showed that mucosal vaccination can protect against prion infection from a peripheral source, suggesting the feasibility of this approach. It is also possible to develop active and/or passive immunomodulatory approaches that more specifically target PrPSc or target the shared pathological conformer found in numerous conformational disorders. Such approaches could have a significant impact on many of the common age-associated dementias.
