Comparative studies of the defensive chemistry of eggs, larvae, pupae, and adults showed differences in alkaloid composition and concentration among life stages. While adults contained mainly the homotropane 1-methyl-9-azabicyclo[3.3.1]nonan-3-one (3), eggs showed the highest concentration of the piperidine 2- (2′-oxopropyl)-6-methylpiperidine (1). We studied the origin of this alkaloid in the eggs by feeding newly emerged, virgin adult beetles with [2-13C]-labeled acetate, and by performing crosses between 13C-fed and unlabeled males and females. GC-MS analysis of alkaloids from 13C-fed males and females showed high incorporation of 13C into the alkaloids, as evidenced from a 20–30% increase of isotopic peaks in diagnostic fragment ions, confirming the expected endogenous origin of these alkaloids. In addition, analyses of eggs from different crosses showed that labeled alkaloids from both parents are incorporated into eggs, indicating that E. paenulata males transfer alkaloids to the females at mating. Biparental endowment of chemical defenses into eggs has been shown previously in insects that acquire defensive compounds from dietary sources. To our knowledge, this is the first report of biparental egg endowment of endogenous defenses. Hanseniaspora is the main genus of the apiculate yeast group that represents approximately 70% of the grape-associated microflora. Hanseniaspora vineae is emerging as a promising species for quality wine production compared to other non-Saccharomyces species. Wines produced by H. vineae with Saccharomyces cerevisiae consistently exhibit more intense fruity flavors and complexity than wines produced by S. cerevisiae alone. In this work, genome sequencing, assembling, and phylogenetic analysis of two strains of H. vineae showed that it is a member of the Saccharomyces complex and it diverged before the whole-genome duplication (WGD) event from this clade. Specific flavor gene duplications and absences were identified in the H. vineae genome compared to 14 fully sequenced industrial S. cerevisiae genomes. The increased formation of 2-phenylethyl acetate and phenylpropanoids such as 2-phenylethyl and benzyl alcohols might be explained by gene duplications of H. vineae aromatic amino acid aminotransferases (ARO8 and ARO9) and phenylpyruvate decarboxylases (ARO10). Transcriptome and aroma profiles under fermentation conditions confirmed these genes were highly expressed at the beginning of stationary phase coupled to the production of their related compounds. The extremely high level of acetate esters produced by H. vineae compared to that by S. cerevisiae is consistent with the identification of six novel proteins with alcohol acetyltransferase (AATase) domains. The absence of the branched-chain amino acid transaminases (BAT2) and acyl coenzyme A (acyl-CoA)/ethanol O-acyltransferases (EEB1) genes correlates with H. vineae's reduced production of branched-chain higher alcohols, fatty acids, and ethyl esters, respectively. Our study provides sustenance for understanding and potentially utilizing genes that determine fermentation aromas.IMPORTANCE The huge diversity of non-Saccharomyces yeasts in grapes is dominated by the apiculate genus Hanseniaspora Two native strains of Hanseniaspora vineae applied to winemaking because of their high oenological potential in aroma and fermentation performance were selected to obtain high-quality genomes. Here, we present a phylogenetic analysis and the complete transcriptome and aroma metabolome of H. vineae during three fermentation steps. This species produced significantly richer flavor compound diversity than Saccharomyces, including benzenoids, phenylpropanoids, and acetate-derived compounds. The identification of six proteins, different from S. cerevisiae ATF, with diverse acetyltransferase domains in H. vineae offers a relevant source of native genetic variants for this enzymatic activity. The discovery of benzenoid synthesis capacity in H. vineae provides a new eukaryotic model to dilucidate an alternative pathway to that catalyzed by plants' phenylalanine lyases. In this work we present the synthesis, optical and electrical characterization of nanoparticles of iron doped anatase and iron doped sodium titanate high aspect ratio nanostructures (Na2Ti2O5⋅H2O). Our study focused mainly in the study of the electronical structure and the derived properties of the dielectric function by DFT (Density Functional Theory) means and their relationship with experimental measurements of optical band gap and electric impedance spectroscopy. Also, structural and morphological characterizations of the systems were carried on by X-ray powder diffraction, high resolution transmission electron microscopy and Raman spectroscopy. It was possible for the doped nanostructures to observe a slight red shift in the band gap of about 0.2 eV and an order of magnitude decrease in the global resistance in the doped system. In this report, we present the study on the microstructure and interparticle interactions of manganitepolyaniline nanocomposites using grazing incidence small angle X-ray scattering (SAXS). In order to determine the nanoparticles mean diameter and correlation distances, data analysis was performed using the Guinier and Beaucage fits, in good agreement with transmission electron microscopy and X-ray diffraction analysis. The analysis of the interference functions revealed the existence of attractive interactions between nanoparticles. The nanocomposites with higher manganite concentration showed best fitting using the sticky hard sphere approximation. A weakening in the attractive interaction with increasing the dilution of nanoparticles in the polymer matrix was observed until a critical volume fraction (fc ~ 0.4) is reached, upon which the hard sphere approximation showed best fitting. The interaction potentials were estimated at room temperature revealing a decrease in the depth and width of the square well with increasing nanoparticle dilution. Coercive field and remanent magnetization showed a decrease with increasing polymer addition suggesting the declining of dipoleedipole interactions, in agreement with SAXS analysis. The compounds were tested against a standard strain of M. tuberculosis H37Rv (ATCC 27294), with minimal inhibitory concentrations (MIC90) of 7.20 and 1.07 μM to Na mpo and [Fe(mpo)3], respectively, and against three clinical isolates with different genotypic profiles, with MIC values ranging from 0.74 to 6.52 and 0.30 to 2.25 μM to Na mpo and [Fe(mpo)3], respectively. [Fe(mpo)3] was more effective against susceptible strains but both compounds were effective in inhibiting MDR and XDR-TB clinical strains. The profile activity was determined through the methodology of a time-kill curve against standard and clinical strains of M. tuberculosis. Time-kill studies indicated that Na mpo had an early bactericidal activity against H37Rv and clinical isolates, with sterilizing effects observed in 5 and 7 days, respectively, at its MIC90. The anti MDR and XDR-M. tuberculosis activity and bactericidal effect of Na mpo and [Fe(mpo)3] demonstrate their potential as new compounds for the treatment of tuberculosis. To assess the reliability of salivary phenytoin (PHT) concentrations and predicted free PHT levels by Sheiner-Tozer equation in order to substitute measured free PHT concentrations in critically ill patients. Methodology: Twenty-four neurocritically ill adult patients receiving intravenous PHT were included in the study. Analyses of total, free plasma and saliva PHT concentrations were performed by fluorescence polarization immunoassay. Plasma albumin levels were also determined. Results: Free PHT concentrations as well as salivary levels better correlate to clinical effect than total drug concentrations. Linear regression analysis showed a strong correlation between estimated free PHT concentrations by Sheiner-Tozer and measured free PHT levels (r=0.835; p<0.001) and salivary PHT concentrations and measured free PHT concentrations (r=0.964; p<0.001). Sheiner-Tozer equation could be misleading in the presence of displacing drugs. Conclusions: Saliva may serve as a feasible fluid to plasma in order to be used as a surrogate for free concentration monitoring of PHT in this population. Paramyosin, a vaccine candidate in different helminthiases, was purified from the adult liver fluke Fasciola hepatica using two different procedures. The first started with a crude extraction of paramyosin in high-salt buffer followed by gel filtration chromatography and two precipitation-solubilization cycles; in the second, anion exchange chromatography replaced the gel filtration step. In both cases, the apparent molecular weight of the purified protein determined by sodium dodecyl sulfate gel electrophoresis under reducing and non-reducing conditions was 97 kDa and 200 kDa, respectively. The molecular weights were consistent with the presence of a dimeric protein linked by disulfide bridges. Western blot analysis showed that the dimeric and monomeric forms were both recognized by an antiserum raised against the F. hepatica 97 kDa band (α-FhPmy), and by an anti-Schistosoma mansoni paramyosin immune serum. Immunohistochemistry using α-FhPmy demonstrated the localization of paramyosin within the subtegumental muscle and in muscle cells surrounding the gut of adult parasites. We also observed labeling of extramuscular structures like testes, surface lamellae of the gut and the tegument of adult flukes. Cystatins are small, phylogenetically conserved proteins that are tight-binding inhibitors of cysteine proteinases. The liver fluke Fasciola hepatica uses a diverse set of cysteine proteinases of the papain superfamily for host invasion, immune evasion and nutrition, but little is known about the regulation of these enzymes. The aim of this work is to characterize the cystatin repertoire of F. hepatica. For this purpose, we first surveyed the available sequence databases, identifying three different F. hepatica single-domain cystatins. In agreement with the in silico predictions, at least three small proteins with cysteine proteinase binding activity were identified. Phylogenetic analyses showed that the three cystatins (named FhStf-1, -2 and -3) are members of the I25A subfamily (stefins). Whereas FhStf-1 grouped with classical stefins, FhStf-2 and 3 fell in a divergent stefin subgroup unusually featuring signal peptides. Recombinant rFhStf-1, -2 and -3 had potent inhibitory activity against F. hepatica cathepsin L cysteine proteinases but differed in their capacity to inhibit mammalian cathepsin B, L and C. FhStf-1 was localized in the F. hepatica reproductive organs (testes and ovary), and at the surface lamella of the adult gut, where it may regulate cysteine proteinases related with reproduction and digestion, respectively. FhStf-1 was also detected among F. hepatica excretion–secretion (E/S) products of adult flukes. This suggests that it is secreted by non-classical secretory pathway and that it may interact with host lysosomal cysteine proteinases. La enfermedad de Alzheimer (EA) es la forma más común de demencia en el mundo, con alrededor de 47.5 millones de personas afectadas. Actualmente no existe cura y los únicos fármacos aprobados para su tratamiento tienen como efecto una mejora transitoria de los síntomas de la enfermedad, pero no son capaces de revertir o detener el daño neuronal asociado. Esto se debe a que los procesos bioquímicos que desencadenan la enfermedad aún se desconocen. Un rasgo distintivo y común en los procesos neurodegenerativos es el aumento intracelular de especies reactivas del oxígeno y del nitrógeno, lo que desencadena una serie de eventos que conducen a la apoptosis. La enzima caspasa-3 ha sido identificada como un blanco terapéutico interesante, ya que la misma no sólo actúa como un mediador clave en la apoptosis neuronal, sino que también participa en procesos no apoptóticos mediante la formación de fragmentos tóxicos de proteínas específicas que afectan la sinapsis y la plasticidad neuronal en etapas tempranas de la EA. Se ha observado que la caspasa-3 no sólo se activa selectivamente en células de hipocampo de ratones transgénicos modelo de EA alterando la función sináptica que da inicio a la pérdida de memoria, sino que también en cerebros de pacientes con EA existen niveles elevados de esta enzima. En la búsqueda de nuevos agentes terapéuticos capaces de modular la sobreactivación de caspasa-3, nuestro grupo de investigación se ha enfocado en el desarrollo de inhibidores reversibles y selectivos de caspasa-3 evidenciando que aquellos compuestos derivados de tiazol y nitrona (imina N-óxido) son buenos inhibidores de caspasa-3 y presentan elevada capacidad antioxidante. En base a estos antecedentes, en el presente trabajo se realizó la síntesis de nuevas moléculas híbridas conteniendo agrupamientos nitrona y tiazol diseñadas para potenciar la selectividad y actividad inhibitoria frente a caspasa-3. Se determinó el efecto neuroprotector antioxidante, antiapoptótico, la capacidad inhibitoria frente a caspasa-3 y caspasa-7 in vitro y se realizaron estudios teóricos de docking y dinámica molecular para estudiar la interacción a nivel atómico de los compuestos en la caspasa-3 y caspasa-7. Los resultados obtenidos muestran que los compuestos evaluados son excelentes agentes antioxidantes y antiapoptóticos, y moderados inhibidores de caspasa-3. El análisis teórico indica que los derivados se unen en la zona de la interfase cercano al sitio alostérico interaccionando con residuos clave para mantener la conformación activa de la enzima caspasa-3 y con menor afinidad en caspasa-7. Las nitronas a4 y a6 presentan muy buena actividad neuroprotectora e inhiben al menos un 20% la actividad caspasa-3. Esto las posiciona como nuevos hits para el rediseño y desarrollo de futuros potenciales fármacos contra EA y otras enfermedades neurodegenerativas. Neurodegenerative diseases affect millions worldwide, posing significant challenges to public health. Oxidative stress and increased caspase-3 activity are key features in the development and progression of neurological disorders leading to neuronal damage and cell loss. As part of our ongoing efforts to identify new molecules with enhanced neuroprotective potential, we have synthesized six novel thiazole-nitrone hybrid compounds, designed as dual-action antiapoptotic and antioxidant agents. The nitrone moiety is known for its spin-trapping ability, which confers antioxidant and neuroprotective properties, while thiazole-containing compounds have been described as antioxidants and potent caspase-3 inhibitors. Our findings demonstrate that these hybrid compounds effectively reduce intracellular reactive oxygen species (ROS) levels and mitigate apoptosis rates in a hippocampal cell line by decreasing active caspase-3 activity, confirming their antioxidant and antiapoptotic properties. Notably, two of the six evaluated compounds exhibited the most promising neuroprotective profiles, including favorable blood-brain barrier permeability. Altogether, these results position the identified molecules as excellent candidates for further structure-based optimization, contributing to the development of more efficient therapeutic strategies against neurodegenerative disorders.
