Aims: To characterize the bioemulsifier produced by a nonfluorescent strain of Pseudomonas putida isolated from a polluted sediment and to determine the influence of pH, temperature, media composition, and carbon and nitrogen source on growth and emulsifying activity. Methods and Results: Different indexes were employed to determine the emulsifying properties of culture supernatants of P. putida ML2 in defined and complex media. Surface tension of cell-free supernatants was measured. Purification and chemical analysis of the emulsifier was performed. Confirmed results indicate that a polysaccharide with hexasaccharide repeating units is responsible for the emulsifying activity in a mineral medium with glucose as sole carbon source. Moreover, an emulsifier is produced when growing on naphthalene. Conclusions: Culture media composition influences the amount and the properties of the emulsifier produced by this P. putida strain. Under nitrogen limiting conditions, a polysaccharide is responsible for the emulsifying activity in defined mineral media. In complex nitrogen rich medium, a different kind of emulsifier is produced. The exopolymer may contribute to hydrocarbons solubilization. Significance and Impact of the Study: This is the first exopolysaccharide with emulsifying properties produced by a Pseudomonas strain reported to the present. Also chemical composition is significantly different from previous reports. This strain has potential use in bioremediation and the purified polysaccharide may be used in food and cosmetic industry. Moreover, the production of the exopolymer may play a role on biofilm formation. This paper reviews the relationship between prehistoric mounds (5000 years BP to 17th century) with main landscape features, particularly wetlands, in the east of Uruguay. Former archeological researchers did recognize this relationship, but it was interpreted by extrapolatingcurrent environments to 5000 years of cultural behavior. In an effort to better understand how environmental dynamics were linked to the cultural record, a set of proxy records (diatoms, opal phytoliths, gastropods, shells) were introduced to take into account local environmental evolution data according to global and regional environmental evolution models. Paleolimnological data are presented on trophic development, climatic change and sea level variations in Negra Lagoon, a 142 km2 coastal lagoon in southern Uruguay. Using a sediment core that extended to the early Holocene, analyses of organic matter, carbonate, diatoms and opal phytoliths allowed us to track changes in trophic state and paleosalinity levels, which were closely related to Holocene sea level variation, temperature and humidity. Diatom Association Zones (DAZ) corresponding to transgressive events were dominated by marine/brackish taxa and relatively low organic matter values, while those DAZ corresponding to regressive events showed increases in brackish/freshwater diatoms and organic matter. Opal Phytoliths Association Zones (OPAZ) also were identified, temperature and humidity indices were calculated, and climatic trends were inferred. During the middle Holocene (i.e., �5000–4000 yr BP), cool–dry climatic conditions were established. After �4000 yr BP, a transition from cool to warm–humid conditions was observed, but fully warm humid conditions established only after �1700 yr BP. Such climatic amelioration led to increases in trophic state. Due to their simplicity, speed, low cost, and specificity, immunoassays have become a useful tool for the analysis of environmental pollutants. Once the anti-hapten antibodies are produced, the same hapten or a related molecule is conjugated to a tracer enzyme or coating protein to set up the assay. Here we report the use of peptides that mimic the analyte as advantageous substitutes of competing haptens. These peptides, which open opportunities for innovation in the development of tracer reagents, can be selected from phage display libraries in a straightforward systematic manner. The concept was proven using assays for the herbicides molinate and atrazine as model systems. Several characteristics of the selection process that may affect the final assay were analyzed, such as the phage coat proteins fused to the peptide, the use of linear or constrained peptide libraries, the effect of the concentration of analyte used during the selection process, and the use of monoclonal or polyclonal antibodies as selector molecules. In all cases we found that the selected peptides performed with improved sensitivity as compared with the chemical hapten conventional assays, showing an analogous cross-reactivity pattern. Interestingly, the phage particles perform as robust and highly standardized assay reagents, and due to their filamentous repetitive structure, they function as sensitive multienzymatic reporters. This paper reports the production of monoterpenes, which elicit a floral aroma in wine, by strains of the yeast Saccharomyces cerevisiae. Terpenes, which are typical components of the essential oils of flowers and fruits, are also present as free and glycosylated conjugates amongst the secondary metabolites of certain wine grape varieties of Vitis vinifera. Hence, when these compounds are present in wine they are considered to originate from grape and not fermentation. However, the biosynthesis of monoterpenes by S. cerevisiae in the absence of grape derived precursors is shown here to be of de novo origin in wine yeast strains. Higher concentration of assimilable nitrogen increased accumulation of linalool and citronellol. Microaerobic compared with anaerobic conditions favored terpene accumulation in the ferment. The amount of linalool produced by some strains of S. cerevisiae could be of sensory importance in wine production. These unexpected results are discussed in relation to the known sterol biosynthetic pathway and to an alternative pathway for terpene biosynthesis not previously described in yeast. Indazole derivatives were extensively studied as bioactive compounds, such as anti-aggregatory and vasorelaxant activity by NO release and increase of cGMP levels and anticancer effects, antimicrobial and antiparasitic properties, among others. In vitro cytotoxicities on V79 cells under hypoxic and aerobic conditions were determined. These systems are widely used in organic chemistry as intermediate compounds for the synthesis of numerous heterocycles. In the other hand, furoxan and benzofuroxan derivatives were extensively studied as bioactive compounds. In some cases, molecular mode of action was proposed. Recently, the research and development in the medicinal chemistry of these systems have produced hybrid compounds in which furoxan or benzofuroxan moieties together with a classical drug moieties are present in a single molecule. So, new anti-ulcer drugs, calcium channel modulators and vasodilator derivatives were described and they are currently in study. In this presentation recent developments in the medicinal chemistry of furoxans and benzofuroxans will be reviewed. We have studied the potential-energy curves and the spectroscopic constants of the ground and low-lying excited states of NbC by employing the complete active space self-consistent field method with relativistic effective core potentials followed by multireference configuration-interaction calculations. We have identified 23 low-lying electronic states of NbC with different spin multiplicities and spatial symmetries within 40,000 cm(-1). At the multireference single and double configuration interaction level of theory the 2sigma+ and 2delta states are nearly degenerated, with the 2delta state located 187 cm(-1) lower than the 2sigma+ state. The estimated spin-orbit splitting for the 2delta state results in a 2delta(3/2) ground state and A 2sigma+ which is placed 650 cm(-1) above the ground state, in reasonable agreement with the experimental result, 831 cm(-1). Our computed spectroscopic constants are in good agreement with experimental values although our results differ from those of a previous density-functional investigation of the excited states of NbC, mainly due to the strong multiconfigurational character of NbC. In the present work we have not only suggested assignments for the observed states but also computed more electronic states that are yet to be observed experimentally. We have investigated the SX (X ) first- or second-row atom), SO2, and SO3 molecules employing the correlation-consistent (cc), the recently developed polarization-consistent (pc), and three Pople-type basis sets, in conjunction with the B3LYP functional. The results confirmed that the aug-pc basis sets represent a great contribution in terms of cost-benefits. In the case of the B3LYP functional, when employing the aug-pc-3 and aug-pc-4 basis sets, it is possible to obtain results that are of aug-cc-pV(5+d)Z and aug-cc-pV(6+d)Z quality, respectively, at a much lower cost. The estimations obtained employing smaller members of the family are of nearly double-ú quality and do not provide reliable results. There is no basis set of quadruple-ú quality among the polarized-consistent basis sets, although in terms of composition, the augpc- 3 basis set is a QZ basis set. A precise estimation of the Kohn-Sham complete basis set (CBS) limit with the aug-pc-X basis sets is too difficult for the B3LYP functional because the ¥(aug-pc-4, aug-pc-3, aug-pc-2) extrapolation gives the same results as those of the aug-pc-4 basis set. This is in contrast with the results observed for ab initio methodologies for which the largest basis sets provided the best estimation of the CBS limit. In our opinion, the closest results to the B3LYP/CBS limit are expected to be those obtained with a two-point extrapolation employing the aug-cc-pV(X+d)Z (X ) 5, 6) basis sets. The results obtained with this extrapolation are very close to those predicted by the ¥(aug-pc-3, aug-pc-2, aug-pc-1) extrapolation, and that provides a cheaper but more inaccurate alternative to estimate the CBS limit. Minor problems were found for the aug-pc-X basis sets and the B3LYP functional for molecules in which sulfur is bound to a very electronegative element, such as SO, SF, SO2, and SO3. For these molecules, the cc basis sets were demonstrated to be more useful. The importance of tight d functions was observed; the total atomization energy of SO2 employing the cc-pV6Z basis set is 1 kcal/ mol lower than that obtained with the aug-cc-pV(6+d)Z basis set. The relationship between the herbicidal activity of a number of novel 1,2,5- oxadiazole N-oxides and some physicochemical properties potentially related with this bioactivity, such as polarity, molecular volume, proton acceptor ability, lipophilicity, and reduction potential were studied. The semiempirical molecular orbital method AM1 was used to calculate theoretical descriptors such as dipolar moment, molecular volume, Mulliken´s charge and the octanol/water partition coefficients (log Po/w). The values of the reduction potentials (Er) were obtained by cyclic voltammetry. In addition, the retention factors (log k’w) on a reversed-phase high-performance liquid chromatography (RP-HPLC) column in pure aqueous mobile phases were measured for several N-oxide derivatives. The log k’w values show good correlation with the calculated values of log Po/w, showing that the chromatographic parameter can be used as lipophilicity descriptor for these compounds. The multiple regression analysis between the descriptors for the Noxide derivatives and the herbicide activity indicate that the variance in the biological activity can be explained by changes in the lipophilicity and in the reduction potential. They transfer via the reversible oxidoreduction of thiols the reducing equivalents of NADPH to numerous substrates and substrate reductases and constitute major defenses against oxidative stress. In this study, we cloned from the helminth parasite Echinococcus granulosus two trans-spliced mRNA variants that encode thioredoxin glutathione reductases (TGR). These variants code for mitochondrial and cytosolic selenocysteine-containing isoforms that possess identical glutaredoxin (Grx) and thioredoxin reductase (TR) domains and differ exclusively in their N termini. Western blot analysis of subcellular fractions with specific anti-TGR antibodies showed that TGR is present in both compartments. The biochemical characterization of the native purified TGR suggests that the Grx and TR domains of the enzyme can function either coupled or independently of each other, because the Grx domain can accept electrons from either TR domains or the glutathione system and the TR domains can transfer electrons to either the fused Grx domain or to E. granulosus thioredoxin. The reversible thiol-disulfide reaction is a central theme in biology. On the one hand, this redox exchange is an efficient mechanism of electron transport. On the other hand, this reaction is a molecular device used by nature as a switch to control protein function and localization through the redox state of critical thiol groups (1). The central role in thiol-disulfide exchange in animal cells is played by the thioredoxin (Trx)1 and glutathione (GSH) systems. Both systems have overlapping and distinct properties and targets but function in a similar way. They transfer via the reversible oxidoreduction of thiols the reducing equivalents of NADPH to numerous substrates and substrate reductases (2). They maintain the cellular redox homeostasis and constitute major defenses against oxidative stress, acting directly by reducing oxidized compounds and proteins or providing reducing equivalents to the hydrogen peroxide reductases present in animal cells, namely thioredoxin peroxidase and glutathione peroxidase (3). Both systems provide electrons to ribonucleotide reductase and control vital cellular processes such as transcription and signal transduction through the redox regulation of kinases, phosphatases, and transcription factors (4-6). The animal thioredoxin system comprises the thioredoxin reductase (TR) and Trx, whereas the glutathione system consists of glutathione reductase (GR), glutathione (GSH), and glutaredoxin (Grx). Trx and Grx are thiol-disulfide oxidoreductases that transfer electrons to various substrates and substrates reductases. GSH, another thiol-based reductant, recycles Grx to its reduced state and reduces other protein and non-protein substrates. TR and GR are pyridine nucleotide-disulfide oxidoreductases that transfer the reducing equivalents of NADPH to oxidized Trx and other substrates and to GSH, respectively. Mammalian and Caenorhabditis elegans TR possess C-terminal electron transfer centers containing redox active cysteine and selenocysteine (Sec) residues (7-10). The selenol group of Sec is a strong nucleophile (11) that confers a profound reductive capacity to the enzyme. The C-terminal redox center is not present in GR. The equivalent redox link in the GSH system is thought to be provided by GSH (10). In mammals, both systems are present in mitochondria and the cytosol. This is because of the existence of genes encoding mitochondrial and cytosolic variants for the proteins from both systems and also because of alternative splicing of single genes (12-17). Recently, mitochondrial and cytosolic variants of DrosophilaTR have also been described to be derived from alternative splicing of a single gene (18). Recently, a more complex machine exhibiting specificity for both thioredoxin and glutathione systems has been characterized in mammals (19). This enzyme termed thioredoxin glutathione reductase (TGR) is a selenoprotein oxidoreductase that possesses thioredoxin reductase, glutathione reductase, and glutaredoxin activities, achieving this broad substrate specificity by a fusion of TR and Grx domains. This enzyme has been reported to be present in the microsomal fraction of testis cells from mice. A multifunctional TGR has also been reported inSchistosoma mansoni, a platyhelminth organism (20), indicating that this fusion domain has been evolutionarily conserved. In this article, we report the cloning and characterization of a thioredoxin glutathione reductase of the larval stage ofEchinococcus granulosus, a cestode parasite that dwells in the liver and lungs of human and cattle hosts. The results indicate that the enzyme possesses glutathione reductase, thioredoxin reductase, and glutaredoxin activities and suggest that there is domain communication between the C-terminal Cys-Sec redox center and the N-terminal glutaredoxin domain. Cruzipain, the major cysteine proteinase of Trypanosoma cruzi, might have other biological roles than its metabolic functions. In this report, we have explored the interaction of cruzipain with molecules of the immune system. The enzyme was used to digest all human IgG subclasses at different pH values and lengths of time. At pH 7.3, all subclasses were readily split at the hinge region. Immunoblot and amino acid sequence analysis showed fragments of IgG1 and IgG3 to be compatible with Fab and Fc, whereas IgG2 and IgG4 rendered Fab2 and Fc. In all cases the fragments produced might impair the binding capacities and the effector functions of specific IgG. At these cleavage sites cruzipain displays cathepsin L and/or cathepsin B activities and shows a clear preference for Pro at the P�2 position and polar residues at P1. Despite the activity of cruzipain within the hinge, the enzyme also cleaved all heavy chains between the CH2 and CH3 domains; producing Fc�-like-fragments of 14 kDa. These fragments are potential candidates to block or saturate Fc receptors on immunocompetent cells. At mild acidic pH cruzipain produced further degradation of the Fc of all subclasses, the Fd of IgG4 and partially the Fd of IgG1, with the consistent loss of any antibody activity. The L chains apparently were not affected. The outer-sphere redox behaviour of a series of [LnCoIIINCFeII( CN)5]− (Ln = n-membered pentadentate aza-macrocycle) complexes have been studied as a function of pH and oxidising agent. All the dinuclear complexes show a double protonation process at pH � 2 that produces a shift in their UV/Vis spectra. Oxidation of the different non-protonated and diprotonated complexes has been carried out with peroxodisulfate, and of the non-protonated complexes also with trisoxalatocobaltate(III). The results are in agreement with predictions from the Marcus theory. The oxidation of [Fe(phen)3]3+ and [IrCl6]2− is too fast to be measured, although for the latter the transient observation of the process has been achieved at pH = 0. The study of the kinetics of the outer-sphere redox process, with the S2O8 2− and [Co(ox)3]3− oxidants, has been carried out as a function of pH, temperature, and pressure. As a whole, the values found for the activation volumes, entropies, and enthalpies are in the following margins, for the diprotonated and non-protonated dinuclear complexes, respectively: ΔV� from 11 to 13 and 15 to 20 cm3 mol−1; ΔS� from 110 to 30 and −60 to −90 J K−1 mol−1; ΔH� from 115 to 80 and 50 to 65 kJ·mol−1. The thermal activation parameters are clearly dominated by the electrostriction occurring on outer-sphere precursor formation, while the trends found for the values of the volume of activation indicate an important degree of tuning due to the charge distribution during the electron transfer process. The special arrangement on the amine ligands in the isomer trans- [L14CoIIINCFeII(CN)5]− accounts for important differences in solvent-assisted hydrogen bonding occurring within the outer-sphere redox process, as has been established in redox reactions of similar compounds.
